Context As a well-known traditional Chinese medicine for protecting the liver, the mechanism of Radix Gentianae (RG) remains unclear. Objective The hepatoprotective effect and metabonomics of RG were studied to explore the molecular and metabolic mechanisms of RG protecting the liver. Materials and methods Sprague-Dawley rats were divided into control and model group (n = 10, orally given distilled water), intervention group (4 subgroups, n = 10, prophylactically and orally given 0.63, 2.5 and 5.6 g/kg RG and 0.2 g/kg bifendatatum for 7 d). On day 7 of the intervention, all rats except the control were injected intraperitoneally with 2.5% carbon tetrachloride vegetable oil solution (1.5 mL/kg) to induce liver injury. After 24 h of carbon tetrachloride injection, rat serum and liver tissue were collected for determining AST, ALT, TNF-α, MCP-1, IL-6, SOD, MDA, GSH, and GSH-PX. Abstract Background It is not known what combination of bevacizumab and chemotherapy agents is the best therapeutic regimen. Comparative study results among the efficacies of bevacizumab plus chemotherapy remain controversial in patients with HER2-negative metastatic breast cancer. Methods We searched Pubmed, Embase, and Cochrane Library Central Resister of Controlled Trials through were July 2019 for randomized controlled trials that evaluated the efficacy of bevacizumab plus chemotherapy in HER2-negative metastatic breast cancer. Data on included study characteristics, outcomes, and risk of bias were abstracted by two reviewers. Results A total of 16 RCT studies involving 5689 patients were included. The results showed that bevacizumab (Bev) - taxanes (Tax) - capecitabine (Cap) has highest-ranking and is probably more effective for prolonging progression-free survival (PFS) than Tax, Cap, Bev-Tax and Bev-Cap, which was no convincing differences among Bev-Cap-vinorelbine, Bev-Tax-everolimus, Bev-Tax-trebananib, Bev-exemestane, Bev-Cap-cyclophosphamide in Bev-containing regimens. For overall response rate (ORR), Bev-Tax-Cap is superior to Tax, Cap and Bev-Cap, while Bev-Tax-trebananib is superior to Cap. The cumulative probability ranking showed that Bev-Tax-Cap or Bev-Tax-trebananib may have best pathological response rate in HER2-negative metastatic breast cancer. Conclusion Our results provide moderate quality evidence that bevacizumab-taxanes-capecitabine maybe the most effective bevacizumab plus chemotherapy on PFS and ORR in HER2-negative metastatic breast cancer, however it should be also considered that bevacizumab may add toxicity to chemotherapy and whether improve overall survival (OS) or not.

Background Although dexmedetomidine (Dex) has a significant neuroprotective effect in various nerve-damage models, the exact mechanism of which Dex protects cells from oxidative damage is not fully clear. This article recommended the protective effect of Dex on oxidative damage in PC12 cells.Methods The PC12 cells were incubated by hydrogen peroxide (H2O2) for 24 h and pre-treated by Dex for 30 min. Cell viability, apoptosis, HIF-1α expression and ROS level were detected by CCK-8, apoptosis assay, Western blot and ROS assay, respectively. The miR-199a expression was tested by qRT-PCR. Targeting relationship between miR-199a and HIF-1α was performed by dual luciferase activity assay. The activation of PI3K/AKT/mTOR and Wnt/β-catenin pathways was tested by western blot.Results Dex attenuated H2O2-induced oxidative damage, including the decline of cell viability, the raise of apoptosis and the generation of ROS in PC12 cells by down-regulating miR-199a expression. Moreover, Dex up-regulated HIF-1α expression via decreasing miR-199a level in PC12 cells and miR-199a targeted the 3′-UTR of HIF-1α. In addition, Dex activated PI3K/AKT/mTOR and Wnt/β-catenin pathways by declining miR-199a level.Conclusions This article illustrated the protective effect of Dex on oxidative damage in PC12 cells. Furthermore, Dex prevented PC12 cells from oxidative injury through the regulation of miR-199a/HIF-1α.

Accurate detection of water holdup in oil-water two-phase flow is crucial for optimizing production and improving crude oil recovery. The transmission lines method is currently one of the few effective methods to measure the water holdup of oil-water two-phase flow. However, variations in temperature and mineralization will alter the dielectric constant and conductivity of the oil-water mixture respectively, posing challenges for precise water holdup measurement. The complex nonlinear relationship between these factors limits the prediction range and accuracy of widely used models, such as the BP neural network and Support Vector Machine (SVM). In order to overcome these issues, this paper establishes a multi-sensor oil-water two-phase flow indoor experiment system and studies the complex relationship between the phase shift of sensor signal and influencing factors. On this basis, this paper proposes a combined water holdup prediction model (BO-XGBoost) of Bayesian optimization (BO) algorithm and extreme gradient boosting (XGBoost). The results demonstrate that the XGBoost model outperforms traditional BP neural network and SVM in predicting water holdup across the full range of 0%-100%. The average absolute error of the BO-XGBoost model is only 1.50%. The above research achieves a full-range, high-precision water holdup prediction, providing a new solution for oilfield development and possessing practical engineering significance.

Summary: The 2022 mpox outbreak led the World Health Organization (WHO) to declare it a public health emergency of international concern (PHEIC). There is a need to develop more effective and safer mpox virus (MPXV)-specific vaccines in response to the mpox epidemic. The mRNA vaccine is a promising platform to protect against MPXV infection. In this study, we construct two bivalent MPXV mRNA vaccines, designated LBA (B6R-A29L) and LAM (A35R-M1R), and a quadrivalent mRNA vaccine, LBAAM (B6R-A35R-A29L-M1R). The immunogenicity and protective efficacy of these vaccines alone or in combination were evaluated in a lethal mouse model. All mRNA vaccine candidates could elicit potential antigen-specific humoral and cellular immune responses and provide protection against vaccinia virus (VACV) infection. The protective effect of the combination of two bivalent mRNA vaccines and the quadrivalent vaccine was superior to that of the individual bivalent mRNA vaccine. Our study provides valuable insights for the development of more efficient and safer mRNA vaccines against mpox.

[Objectives] This study was conducted to establish a method of quantitative analysis of multi-components by single marker (QAMS) for the simultaneous determination of such seven chemical components as gallic acid, epicatechin, catechin, ferulic acid, chlorogenic acid, rutin and caffeic acid in Vidal grape. [Methods] The high performance liquid chromatography was carried out using a COSMOSIL C18-MS-II column (4. 6 mm x 250 mm, 5 µm) with the mobile phase acetonitrile-2% acetic acid aqueous solution (gradient elution) at a flow rate of 1.0 ml/min. The detection wavelength was 280 nm, and the column temperature was 25 °C. Using caffeic acid as an internal reference, the relative correction factors between it and other six to-be-detected components, and the contents of the seven components were calculated using the correction factors. The established was compared the results with the external standard method to verify the feasibility and accuracy of the method. [Results] The seven components had a good linear relationship in the ranges of 1. 060 - 10. 60, 1. 419 - 14. 19, 1. 062 - 10. 62, 0.295 0-2.950, 0.101 9-1.019, 0.2014-2.014, and 0.149 8-1.498 µg, respectively, and the relative correction factors of gallic acid, epicatechin, catechin, ferulic acid, chlorogenic acid and rutin were 0. 976 0,0.780 6, 0. 327 7,1.640, 1.161, 2. 778, respectively. There was no significant difference between the results of the QAMS method and the external standard method. [Conclusions] The QAMS method using caffeic acid as an internal reference is accurate and feasible, and provides a reliable method for the quality evaluation of Vidal ice grape. Pulmonary blastoma (PB) is a rare and invasive malignancy of the lungs with a poor prognosis. Although the mainstay treatment of PB is surgery, and radiotherapy and chemotherapy have been reported, no standard therapy exists for patients inoperable in advanced stages. Moreover, little is known about driver mutation status and immunotherapy efficacy. This paper presents a male patient diagnosed with classic biphasic PB using CT-guided lung biopsy pathology and immunohistochemistry. The patient’s symptoms included cough, chest pain, shortness of breath, hemoptysis, and hypodynamia. The primary focus of this paper is to discuss the impact of anti-PD-1 immunotherapy on PB. The patient experienced progression-free survival (PFS) of over 27 months following sintilimab second-line anti-PD-1 therapy. The patient has currently survived for nearly 40 months with a satisfactory quality of life.

Skin cutaneous melanoma (SKCM) is a skin cancer type characterized by a high degree of immune cell infiltration. The potential function of lactate, a main metabolic product in the tumor microenvironment (TME) of SKCM, remains unclear. In this study, we systemically analyzed the predictive value of lactate-related genes (LRGs) for prognosis and response to immune checkpoint inhibitors (ICIs) in SKCM patients included from The Cancer Genome Atlas (TCGA) database. Cluster 3, by consensus clustering for 61 LRGs, manifested a worse clinical outcome, attributed to the overexpression of malignancy marks. In addition, we created a prognostic prediction model for high- and low-risk patients and verified its performance in a validation cohort, GSE65904. Between TME and the risk model, we found a negative relation of the immunocyte infiltration levels with patients’ risk scores. The low-risk cases had higher ICI expression and could benefit better from ICIs relative to the high-risk cases. Thus, the lactate-related prognosis risk signature may comprehensively provide a basis for future investigations on immunotherapeutic treatment for SKCM.

Abstract Litter is essential to promote nutrient cycling and maintain the sustainability of forest resources. However, its variability has not been sufficiently studied at the local scale. The prediction of litter amount using ordinary cokriging with Pearson correlation analysis (COKP) and ordinary cokriging with principal component analysis (COKPCA) was compared with that using ordinary kriging (OK) based on cross‐validation at the local scale of a 1‐ha plot over natural spruce–fir mixed forest in Jilin Province, China. Litter samples in semidecomposed (F) and complete decomposed (H) horizons were collected using an equidistant grid point sampling of 10 m × 10 m. Pearson correlation analysis and principal component analysis (PCA) were used to confirm auxiliary variables. The results showed that the amount of litter was 19.65 t/ha in the F horizon and 10.37 t/ha in the H horizon. The spatial structure variance ratio in the H horizon was smaller than that in the F horizon, indicative of its stronger spatial autocorrelation. Spatial distributions of litter amount in both horizons exhibited a patchy and heterogeneous pattern. Of the selected stand characteristics and litter properties, litter moisture content indicated the strongest relationship with litter amount. Cross‐validation revealed that COKPCA using the comprehensive score as an auxiliary variable produced the most accurate map. The average standard error and root‐mean‐square error between the predicted and measured values were always smaller, the mean error and mean standardized error were much closer to 0, and the root‐mean‐square standardized error was closer to 1 than COKP using litter moisture and OK. Therefore, a clear advantage of cokriging based on principal component analysis was observed and COKPCA was found to be a very useful approach for further interpolation prediction.

Background: Immune thrombocytopenic purpura (ITP) is an autoimmune bleeding disorder and the decreased number and immunosuppressive dysfunction of Treg cells are key promoters of ITP. However, their mechanisms in ITP development have not been fully clarified.Methods: HUWE1 mRNA and protein levels in CD4+ T cells in peripheral blood from ITP patients were assessed by quantitative real-time PCR and Western blot. HUWE1 function in ITP was estimated using flow cytometry, enzyme-linked immunosorbent assay and immunosuppression assay. Besides, the HUWE1 mechanism in reducing the number and function of Treg cells in ITP was investigated by immunoprecipitation, cycloheximide-chase assay, ubiquitin experiment and immunofluorescence assay.Results: HUWE1 expression was elevated in CD4+ T cells in peripheral blood from ITP patients and HUWE1 mRNA level was negatively correlated with platelet counts and Treg cell percentage. Moreover, the interference with HUWE1 increased the number of Treg cells and enhanced its immunosuppressive function, and the HUWE1 overexpression produced the opposite results. For the exploration of mechanism, HUWE1 interacted with E26 transformation-specific-1 (Ets-1) and this binding was dependent on the negative regulation of the phosphorylation level of Ets-1 (Thr38) and HUWE1 facilitated the ubiquitin degradation of Ets-1 protein to restrain Treg cell differentiation and weaken their immunosuppressive functions. The in vivo assay confirmed that the HUWE1 inhibitor alleviated ITP in mice.Conclusion: HUWE1 induced the immune imbalance in ITP by decreasing the number and weakening the function of Treg cells through the ubiquitination degradation of Ets-1.

Photoaging is not only the main cause of skin aging caused by exogenous factors, it is also related to a variety of skin diseases and even malignant tumors. Excessive and repeated exposure to ultraviolet radiation, especially UVA induces oxidative stress, DNA damage, inflammation, and collagen and elastin degeneration, ultimately leads to skin photoaging, manifested by skin redness, coarse wrinkles, and pigmentation even skin cancer. There has been a large demand of effective prevention and medications but approaches in the current management of photoaging are very limited. In the previous study, we found that a non-coding circular RNA circ_0011129 acts as a miR-6732-5p adsorption sponge to inhibit the reduction of type I collagen and the denaturation and accumulation of elastin in UVA-induced HDF cells photoaging model. However, in vivo instability and efficient delivery to the target cell of circRNA is a major challenge for its clinical application. Therefore, improving its stability and delivery efficiency are desired. In this study, we proposed a strategy of delivering circ_0011129 with small extracellular vesicles (sEVs) from human adipose-derived stem cells (hADSCs) to intervene in the photoaging process. The results showed that sEVs from hADSCs in 3D bioreactor culture (3D-sEVs) can prevent photoaging. Consequently, by overexpressing circ_0011129 in hADSCs, we successfully loaded it into 3D-sEVs (3D-circ-sEVs) and its protective effect was better. Our studies provide a novel approach to preventing skin photoaging, which has important clinical significance and application value for the development of non-coding RNA drugs to treat skin photoaging. We first screened out hADSCs-derived sEVs with excellent anti-oxidant effects. We then compared the sEVs collected from traditional 2D culture with 3D bioreactor culture. By miRNA-seq and GEO data analysis, we found that miRNAs in 3D-sEVs were enriched in cell activities related to apoptosis, cellular senescence, and inflammation. Subsequently, we prepared circ_0011129-loaded 3D-sEVs (3D-circ-sEVs) by overexpressing it in hADSCs for the treatment of photoaging in vitro. We proved that 3D-circ-sEVs can interfere with the process of cell photoaging and protect cells from UVA radiation damage, as well as in a H2O2-induced oxidative stress model.

Nowadays, we face a series of global challenges, including the growing depletion of fossil energy, environmental pollution, and global warming. The replacement of coal, petroleum, and natural gas by secondary energy resources is vital for sustainable development. Hydrogen (H2) energy is considered the ultimate energy in the 21st century because of its diverse sources, cleanliness, low carbon emission, flexibility, and high efficiency. H2 fuel cell vehicles are commonly the end-point application of H2 energy. Owing to their zero carbon emission, they are gradually replacing traditional vehicles powered by fossil fuel. As the H2 fuel cell vehicle industry rapidly develops, H2 fuel supply, especially H2 quality, attracts increasing attention. Compared with H2 for industrial use, the H2 purity requirements for fuel cells are not high. Still, the impurity content is strictly controlled since even a low amount of some impurities may irreversibly damage fuel cells’ performance and running life. This paper reviews different versions of current standards concerning H2 for fuel cell vehicles in China and abroad. Furthermore, we analyze the causes and developing trends for the changes in these standards in detail. On the other hand, according to characteristics of H2 for fuel cell vehicles, standard H2 purification technologies, such as pressure swing adsorption (PSA), membrane separation and metal hydride separation, were analyzed, and the latest research progress was reviewed.

This paper investigates the utility maximization problem for the downlink of the multi-interface multichannel wireless mesh network with orthogonal frequency division multiple access. A cross-layer joint power and multiple access control algorithm are proposed. Rosen projection matrix is combined with Solodov projection techniques to build a three-memory gradient Rosen projection method, which is applied to solve this optimization problem. The convergence analysis is given and simulations show that the proposed solution achieves significant throughput compared with existing approaches.

With the rapid development of wireless sensor networks, reducing energy consumption is becoming one of the important factors to extend node lifetime, and it is necessary to adjust the launching power of each node because of the limited energy available to the sensor nodes in the networks. This paper proposes a power and rate control model based on the network utility maximization (NUM) framework, where a weighting factor is used to reflect the influence degree of the sending power and transmission rate to the utility function. In real networks, nodes interfere with each other in the procedure of transmitting signal, which may lead to signal transmission failure and may negatively have impacts on networks throughput. Using dual decomposition techniques, the NUM problem is decomposed into two distributed subproblems, and then the conjugate gradient method is applied to solve the optimization problem with the calculation of the Hessian matrix and its inverse in order to guarantee fast convergence of the algorithm. The convergence proof is also provided in this paper. Numerical examples show that the proposed solution achieves significant throughput compared with exiting approaches.
