Abstract Purpose This review primarily discusses the current research advance of predictive biomarkers for melanoma immunotherapy. The aim of the present review is to summarize the biomarkers and evaluate the advantages and disadvantages. Methods All reference can be found through Pubmed. This review mainly focuses on three main directions: tumor-related factors, host factors, and the tumor microenvironment. In the end, there exhibits some unusual aspects of predictive biomarkers and forecasts the future model. Results The mainsteam of predictive biomarkers focuses on PD-L1, TMB, gene mutations, immune cells, IDO1, LDH, tertiary lymphoid structures (TLS), HLA-DR, tumor-associated macrophages (TAMs), tumor-infiltrating lymphocytes (TILs), and Extracellular vesicles (EVs). Conclusion The current research advance of predictive biomarkers for melanoma immunotherapy can be mainly divided into three parts: tumor-related factors, host factors, and the tumor microenvironment. The predictive biomarkers include PD-L1, TMB, gene mutations, immune cells, IDO1, LDH, TLS, HLA-DR, TAMs, TILs, and EVs. A model based on multiple biomarkers is expected to become the answer to predicting prognosis.

Abstract Aims This study aimed to describe baseline characteristics and adherence among patients with transthyretin amyloid cardiomyopathy (ATTR‐CM) treated with tafamidis (VYNDAQEL®) in Japan using the Japanese Medical Data Vision (MDV) database. Methods and results This study was a non‐interventional, retrospective cohort study of adult (≥18 years old) patients in the Japanese MDV claims database diagnosed with ATTR‐CM and with at least two tafamidis prescriptions of dose strength 4 × 20 mg/day between 1 March 2019 and 31 August 2021. The date of the first prescription was defined as the index date, with follow‐up time defined as the time between the first and last prescription plus the days' supply from the last refill. Baseline characteristics were assessed during a 12 month pre‐index period. Adherence was measured using two metrics: (i) the modified medication possession ratio (mMPR), calculated by taking the sum of days supplied for all fills within the follow‐up period, divided by the number of days of follow‐up, and reported as a percentage, with patients classified as adherent with an mMPR of ≥80%, and (ii) the proportion of days covered (PDC), calculated by taking the total number of days' supply dispensed during the follow‐up period divided by the number of days of follow‐up, adjusting for any days' supply overlap. A total of 210 patients were identified; the mean (standard deviation) age of the cohort was 77 (5.9) years, and the majority (89%) were male. The most common baseline cardiovascular comorbidities were heart failure (85%), ischaemic heart disease (66%), hypertensive diseases (49%), and diabetes (35%); 75% of patients received heart failure medications in the 12 months prior to index, with the most common being beta‐blockers (49%), diuretics (48%), angiotensin receptor blockers (30%), angiotensin‐converting enzyme inhibitors (22%), and sodium–glucose cotransporter‐2 inhibitors (8.1%). Over an average 14 month follow‐up, mean mMPR was 96% with a median of 100% [inter‐quartile range (IQR): 97–101%]; 93% of patients were adherent (defined as an mMPR ≥ 80%). In the same follow‐up period, mean PDC was 93.6% with a median of 99% (IQR: 93–100%). Persistence was high with 78% of patients having a 0 day gap between prescription refills. Conclusions This study found high adherence rates to tafamidis in this real‐world Japanese patient population. Adherence rates in this study were similar to those reported by the tafamidis clinical trial and a previously published US commercial claims adherence analysis. Further studies should be conducted to assess the impact of real‐world adherence on real‐world outcomes.

Abstract Proton radiotherapy favored over X-ray photon therapy due to its reduced radiation exposure to surrounding healthy tissues, is highly dependent on the accurate positioning of the Bragg peak. Existing methods like PET and prompt gamma imaging to localize Bragg peak face challenges of low precision and high complexity. Here we introduce a 3D protoacoustic imaging with a 2D matrix array of 256 ultrasound transducers compatible with 256 parallel data acquisition channels provides real-time imaging capability (up to 75 frames per second with 10 averages), achieving high precision (5 mm/5% Gamma index shows accuracy better than 95.73%) at depths of tens of centimeters. We have successfully implemented this method in liver treatment with 5 pencil beam scanning and in prostate cancer treatment on a human torso phantom using a clinical proton machine. This demonstrates its capability to accurately identify the Bragg peak in practical clinical scenarios. It paves the way for adaptive radiotherapy with real-time feedback, potentially revolutionizing radiotherapy by enabling closed-loop treatment for improved patient outcomes.

Abstract Rheumatoid arthritis (RA) is a debilitating autoimmune disease characterized by chronic joint inflammation and cartilage damage. Current therapeutic strategies often result in side effects, necessitating the development of targeted and safer treatment options. This study introduces a novel nanotherapeutic system, 2-APB@DGP-MM, which utilizes macrophage membrane (MM)-encapsulated nanoparticles (NPs) for the targeted delivery of 2-Aminoethyl diphenylborinate (2-APB) to inflamed joints more effectively. The NPs are designed with a matrix metalloproteinase (MMP)-cleavable peptide, allowing for MMP-responsive drug release within RA microenvironment. Comprehensive in vitro and in vivo assays confirmed the successful synthesis and loading of 2-APB into the DSPE-GPLGVRGC-PEG (DGP) NPs, as well as their ability to repolarize macrophages from a pro-inflammatory M1 to an anti-inflammatory M2 phenotype. The NPs demonstrated high biocompatibility, low cytotoxicity, and enhanced cellular uptake. In a collagen-induced arthritis (CIA) mouse model, intra-articular injection of 2-APB@DGP-MM significantly reduced synovial inflammation and cartilage destruction. Histological analysis corroborated these findings, demonstrating marked improvements in joint structure and delayed disease progression. Above all, the 2-APB@DGP-MM nanotherapeutic system offers a promising and safe approach for RA treatment by modulating macrophage polarization and delivering effective agents to inflamed joints. Abstract Emulsified asphalt mixtures’ cold mix and paving features facilitate asphalt pavements in fulfilling dual-carbon and energy-saving demands. Anionic emulsifiers can enhance emulsion stability, ensure uniform dispersion of oil and water, possess good decompression viscosity, thickening, and lubricating properties, and maintain good stability under acidic conditions. Nevertheless, anionic emulsified asphalt is restricted in engineering applications due to problems like its storage stability. In this paper, eight anionic emulsifiers and two preparation procedures were chosen for stability tests. Through static tests, storage tests, sieve residue tests, and laser particle size tests, the impacts of emulsifiers on the storage stability and dispersion of asphalt were analyzed. Waterborne epoxy resin exhibits excellent adhesive properties, mechanical properties, chemical resistance, and heat stability. A fluorescence microscope test, static and storage test, laser particle size test, and cementation test were employed to explore the effects of different preparation processes and waterborne epoxy mixing ratios on emulsified asphalt’s storage stability, dispersion stability, and structural stability. The results showed that: (1) the emulsified asphalt prepared with the BWH-02 emulsifier exhibits the best storage stability, and blending with 20% of the waterborne epoxy modifier can notably enhance the bonding properties; (2) the shear strength of the BWH-02 waterborne epoxy emulsified asphalt prepared can reach 1.543 MPa, and the tensile viscosity can reach 0.848 MPa; (3), The emulsified asphalt prepared by the process of modification has better storage stability than that prepared by the side of the emulsification process. Moreover, the storage stability of emulsified asphalt prepared by emulsification and modification is superior to that of the emulsification and modification process. This research provides theoretical and technical support for popularizing and applying cold-mixed cold-paving asphalt mixtures.

Abstract Fruit ripening is accompanied by the development of fruit quality traits; however, this process also increases the fruit’s susceptibility to various environmental stresses, including pathogen attacks and other stress factors. Therefore, modulating the fruit ripening process and defense responses is crucial for maintaining fruit quality and extending shelf life. Membrane proteins play intricate roles in mediating signal transduction, ion transport, and many other important biological processes, thus attracting extensive research interest. This review mainly focuses on the functions of membrane proteins in regulating fruit ripening and defense responses against biotic and abiotic factors, addresses their potential as targets for improving fruit quality and resistance to environmental challenges, and further highlights some open questions to be addressed.

Objective To employ two dimensional speckle tracking imaging （2D-STI） and non-invasive left ventricular pressure-strain loop for observing and evaluating the changes of left ventricular myocardial function of patients with end-stage renal disease （ESRD） during early establishment of autogenous arteriovenous fistula （AVF）. Methods A total of 55 ESRD patients who were to undergo AVF surgery in the Department of Nephrology, Subei People’s Hospital from October 1, 2022 to March 31, 2023 were selected. Echocardiography was performed at Week 1 pre-operation, Day 1 post-operation and Week 6 post-operation. The changes of global longitudinal strain （GLS） and myocardial work （MW） were compared at different timepoints pre/post-AVF. Sensitivity and correlation of myocardial work （MW） were examined for global work efficiency （GWE）, global work index （GWI）, global wasted work （GWW）, global constructive work （GCW） and global longitudinal strain （GLS）. Results Absolute values of GWE （P = 0.025） and GLS （P＜0.001） rose and GWW （P = 0.041） declined at Day 1 post-AVF. Absolute values of GWI （P = 0.003, P = 0.001）, GWE （P = 0.005, P＜0.001） and GLS （P = 0.026, P＜0.001） at Week 6 post-operation were lower than those at Week 1 pre-operation and Day 1 post-operation while GWW rose （P＜0.001, P＜0.001）. Meanwhile area under ROC curve of GLS was 0.607. It was smaller than that of GWI/GWW/GWE （0.612, 0.657 & 0.618）. Correlation analysis indicated that GLS was correlated negatively with GWI/GCW/GWE （r = −0.629; r = −0.574; r = −0.659; all P＜0.05） and positively with GWW （r = 0.418; P＜0.05）. Conclusion In ESRD patients, left ventricular myocardial function becomes transiently elevated at the start of AVF establishment. After some time, impairment is lessened. As compared with 2D-STI /LVEF, non-invasive LVPSL may effectively evaluate the changes of left ventricular myocardial function during early AVF establishment in ESRD patients.

Abstract Biochar (BC) applications in soil has positive effects on plant performance, particularly for loose soil in agricultural context. However, how biochar types affect plant performance of non-crop species and soil–plant carbon relationships is not clear. We selected five different BC types and three plant species to investigate the responses of plant performance and the soil–plant carbon relationship to BC effects. The result demonstrated that peanut shell BC led to the death of both R. tomentosa and C. edithiae, due to a reduction in nutrient uptake caused by higher soil electricity conductivity (2001.7 and 976.3 µS cm−1). However, the carbon content of S. arboricola increased by 57% in peanut shell BC-amended soil, suggesting that S. arboricola has a higher tolerance for soil salinity. Wood BC-amended soil led to better stomatal conductance (gs) and leaf area index (LAI) of both R. tomentosa and C. edithiae due to the higher water retention in the soil (22.68% and 20.79%). This illustrated that a higher amount of water retention brought by wood BC with a great amount of pore volume might be the limited factor for plant growth. The relationship between gs and LAI suggested that gs would not increase when LAI reached beyond 3. Moreover, wood and peanut shell BC caused a negative relationship between soil organic carbon and plant carbon content, suggesting that plants consume more carbon from the soil to store it in the plant. Overall, wood BC is recommended for plant growth of R. tomentosa and C. edithiae, and peanut shell BC is suggested for S. arboricola carbon storage. Graphical Abstract

Abstract Beige fat activation involves a fuel switch to fatty acid oxidation following chronic cold adaptation. Mitochondrial acyl-CoA synthetase long-chain family member 1 (ACSL1) localizes in the mitochondria and plays a key role in fatty acid oxidation; however, the regulatory mechanism of the subcellular localization remains poorly understood. Here, we identify an endosomal trafficking component sortilin (encoded by Sort1) in adipose tissues that shows dynamic expression during beige fat activation and facilitates the translocation of ACSL1 from the mitochondria to the endolysosomal pathway for degradation. Depletion of sortilin in adipocytes results in an increase of mitochondrial ACSL1 and the activation of AMPK/PGC1α signaling, thereby activating beige fat and preventing high-fat diet (HFD)-induced obesity and insulin resistance. Collectively, our findings indicate that sortilin controls adipose tissue fatty acid oxidation by substrate fuel selection during beige fat activation and provides a potential targeted approach for the treatment of metabolic diseases.

The global food crisis has intensified with climate change, and hybrid rice (Oryza sativa L.) is expected to play a crucial role in ensuring world food security. This study analyzed the parental origins and variety characteristics of 930 hybrid rice varieties bred in Sichuan Province, a typical low-light region and one of the main production areas for hybrid rice, from 2002 to 2022. The results indicate that these varieties were hybridized using 329 direct female and 539 direct male parents. Notably, the female parent Peiai 64 and the male parents Yuehesimiao and Fenghuazhan demonstrated a higher probability of breeding rice varieties with high yield and good quality. An ideal panicle type should possess more grains, a lower 1000-grain weight, a higher seed setting percentage, and greater grain density. It is recommended to increase the utilization of excellent parents in Sichuan Province, such as parents with ideal panicle type to improve resistance to climate change, particularly global dimming.

Oxide-dispersion-strengthened (ODS) steels are candidate materials for application in advanced nuclear reactors. In this study, the low-cycle fatigue performances of 13Cr-ODS ferritic steel pipes were investigated at 600, 700, and 800 °C. Cyclic softening was observed at high strain amplitudes with an increase in the number of fatigue cycles. However, cyclic hardening appeared first, and then cyclic softening occurred at a low strain amplitude with the increase in the number of fatigue cycles. By comparing the cyclic stress–strain curves and the monotonic stress–strain curves, it was found that cyclic softening occurred regardless of the strain amplitude. The Coffin–Manson and Basquin equations were used to predict the fatigue of the pipes. Microstructure analysis indicated that cyclic softening was induced by the dynamic recovery and recrystallization, which reduced the number of low-angle grain boundaries in the deformed grains by promoting dislocation annihilation and reorganization. A complex multi-layer core–shell structure with a large size (∼500 nm) was observed.

Abstract Background China has made remarkable achievements in tuberculosis (TB) prevention and control, but it still takes long way to achieve the End TB goal especially in underdeveloped Southwest China. TB healthcare workers (HCWs) are core forces in TB prevention and control but often face job burnout. This study aimed to explore the burden and associated factors of job burnout among TB HCWs in Southwest China. Methods This cross-sectional study used both survey questionnaires and semi-structured interviews, to assess job burnout among TB HCWs based on Malasch Model and explore the associated factors based on Herzberg’s two-factor theory (different hygiene and motivation factors). Quantitative data analysis adopts multiple linear regression to in SPSS 22.0, and qualitative data were analyzed through a framework approach. Results A total of 1140 TB HCWs were included in questionnaire surveys. The overall job burnout rates of TB HCWs in Centers for Disease Control and Prevention (CDC), designated hospitals and Primary Health Care (PHC) sectors were 55%, 70.1% and 67.5%, respectively. TB HCWs in CDC who scored lower in interpersonal factors had a higher risk of depersonalization (DP) [B(95%CI): -0.89 (-1.71 to -0.80)]. TB HCWs in designated hospitals who scored lower in doctor-patient relationship factors [B (95%CI): 6.63 (-12.06 to -1.20)] were more likely to have emotional exhaustion (EE). TB HCWs who were less satisfied with training, supervision and assessment in PHC sectors [B(95%CI): 0.65 (0.03 to 1.26)] had less personal accomplishment (PA). Interviews with nine TB HCWs showed that poor environment could lead to high infection and heavy workload could lead to work pressure among TB HCWs in Chongqing. It is also found that performance assessment and management of TB HCWs, communication and cooperation and so on are related to job burnout. Conclusions TB HCWs had different levels of job burnout in CDC, designated hospitals, and PHC sectors of Chongqing, which were affected by different hygiene and motivation factors. Governments, organizations and individuals should take cooperative measures such as strengthening communication to deal with job burnout among TB HCWs.

Abstract Background The prognostic value of circulating tumor cells (CTCs) in metastatic breast cancer (MBC) has been extensively studied and verified by the CellSearch® system. Varieties of microfluidic systems have been developed to improve capture efficiency with the lack of standardization and automation. This study systematically verified the positive threshold for prognosis and its guidance value in anti-HER2 therapy based on a novel automated microfluidic system OmiCell®. Methods CTCs isolation, enumeration and labeling were performed using the OmiCell® system. CTCs identification and reporting were performed using the DeepSight® scanning system. Results The capture efficiency and specificity of OmiCell® system was 91.9% and 90%, respectively. Then, 65 MBC patients with known HER2 status of their metastatic tumors were enrolled. Abstract Members of the Metal Tolerance Protein (MTP) family are critical in mediating the transport and tolerance of divalent metal cations. Despite their significance, the understanding of MTP genes in mustard (Brassica juncea) remains limited, especially regarding their response to heavy metal (HM) stress. In our study, we identified MTP gene sets in Brassica rapa (17 genes), Brassica nigra (18 genes), and B. juncea (33 genes) using the HMMER (Cation_efflux; PF01545) and BLAST analysis. For the 33 BjMTPs, a comprehensive bioinformatics analysis covering the physicochemical properties, phylogenetic relationships, conserved motifs, protein structures, collinearity, spatiotemporal RNA-seq expression, GO enrichment, and expression profiling under six HM stresses (Mn2+, Fe2+, Zn2+, Cd2+, Sb3+, and Pb2+) were carried out. According to the findings of physicochemical characteristics, phylogenetic tree, and collinearity, the allopolyploid B. juncea’s MTP genes were inherited from its progenitors, B. rapa and B. nigra, with minimal gene loss during polyploidization. Members of the BjMTP family exhibited conserved motifs, promoter elements, and expression patterns across subgroups, consistent with the seven evolutionary branches (G1, G4–G9, and G12) of the MTPs. Further, spatiotemporal expression profiling under HM stresses successfully identified specific genes and crucial cis-regulatory elements associated with the response of BjMTPs to HM stresses. These findings may contribute to the genetic improvement of B. juncea for enhanced HM tolerance, facilitating the remediation of HM-contaminated areas.

Abstract Background Type 2 diabetes mellitus (T2D) is associated with an increased risk of cognitive dysfunction. Angiopoietin-like protein 8 (ANGPTL8) is an important regulator in T2D, but the role of ANGPTL8 in diabetes-associated cognitive dysfunction remains unknown. Here, we explored the role of ANGPTL8 in diabetes-associated cognitive dysfunction through its interaction with paired immunoglobulin-like receptor B (PirB) in the central nervous system. Methods The levels of ANGPTL8 in type 2 diabetic patients with cognitive dysfunction and control individuals were measured. Mouse models of diabetes-associated cognitive dysfunction were constructed to investigate the role of ANGPTL8 in cognitive function. The cognitive function of the mice was assessed by the Barnes Maze test and the novel object recognition test, and levels of ANGPTL8, synaptic and axonal markers, and pro-inflammatory cytokines were measured. Primary neurons and microglia were treated with recombinant ANGPTL8 protein (rA8), and subsequent changes were examined. In addition, the changes induced by ANGPTL8 were validated after blocking PirB and its downstream pathways. Finally, mice with central nervous system-specific knockout of Angptl8 and PirB −/− mice were generated, and relevant in vivo experiments were performed. Results Here, we demonstrated that in the diabetic brain, ANGPTL8 was secreted by neurons into the hippocampus, resulting in neuroinflammation and impairment of synaptic plasticity. Moreover, neuron-specific Angptl8 knockout prevented diabetes-associated cognitive dysfunction and neuroinflammation. Mechanistically, ANGPTL8 acted in parallel to neurons and microglia via its receptor PirB, manifesting as downregulation of synaptic and axonal markers in neurons and upregulation of proinflammatory cytokine expression in microglia. In vivo, PirB −/− mice exhibited resistance to ANGPTL8-induced neuroinflammation and synaptic damage. Conclusion Taken together, our findings reveal the role of ANGPTL8 in the pathogenesis of diabetes-associated cognitive dysfunction and identify the ANGPTL8-PirB signaling pathway as a potential target for the management of this condition.

Introduction: Penicillium expansum is a harmful plant fungal pathogen that causes blue mold disease and produces mycotoxin patulin, leading to huge economic losses and food safety hazard. Set1 associated complex Set1/COMPASS deposits the methylation at lysine 4 of histone H3, which is associated with gene expression in diverse biological processes of fungi. However, the function and underlying mechanisms of Set1/COMPASS are poorly defined in P. expansum. Objectives: The study aimed to identify Set1/COMPASS and investigate its regulation mechanisms on growth, pathogenicity, and patulin biosynthesis of P. expansum. Methods: Analyses of phylogenetic relationship, conserved structural domain, and gene deletion were used to identify components of Set1/COMPASS. Phenotype analysis and stress tolerance test of gene deletion mutants were conducted to analyze the function of these components. Yeast two-hybrid, Co-Immunoprecipitation (Co-IP), and point mutation were performed to verify the protein interaction. Western blot was conducted for detection of H3K4 methylation levels. Results: P. expansum owns six components of Set1/COMPASS besides PeSet1. Absence of each component resulted in reduction of H3K4 methylation levels and impaired growth, pathogenicity, and patulin biosynthesis, as well as altered stress responses of P. expansum. One component PeBre2p was found to interact with the conserved global regulator PeVelB (VelvetLike protein B) at Asp294 of PeBre2p. This interaction affected fungal growth and utilization of fructose, lactose, glycine, and proline in P. expansum. Conclusion: This study revealed the important roles of Set1/COMPASS in P. expansum and clarified for the first time the combined regulation of PeBre2p and PeVelB in fungal growth and nutrition utilization. These results will provide potential targets for the control of blue mold disease.

ABSTRACTIntroduction The rapid development of COVID-19 vaccines has provided crucial tools for pandemic control, but the occurrence of vaccine-related adverse events (AEs) underscores the need for comprehensive monitoring.Methods This study analyzed the Vaccine Adverse Event Reporting System (VAERS) data from 2020–2022 using statistical methods such as zero-truncated Poisson regression and logistic regression to assess associations with age, gender groups, and vaccine manufacturers.Results Logistic regression identified 26 System Organ Classes (SOCs) significantly associated with age and gender. Females displayed especially higher odds in SOC 19 (Pregnancy, puerperium and perinatal conditions), while males had higher odds in SOC 25 (Surgical and medical procedures). Older adults (>65) were more prone to symptoms like Cardiac disorders, whereas those aged 18–65 showed susceptibility to AEs like Skin and subcutaneous tissue disorders. Moderna and Pfizer vaccines induced fewer SOC symptoms compared to Janssen and Novavax. The zero-truncated Poisson regression model estimated an average of 4.243 symptoms per individual.Conclusion These findings offer vital insights into vaccine safety, guiding evidence-based vaccination strategies and monitoring programs for precise and effective outcomes.

In recent years, the emergence of novel biological threats has brought biosecurity issues to the forefront of societal concerns. The United Kingdom established the UK Biological Security Strategy in 2018, and after revisions, a new version was published in June 2023. The strategy presents the UK's new vision, mission, outcomes, and plans, and provides action plans from four perspectives: understanding, preventing, detecting, and responding to major biological threats to protect the UK's national interests and enhance biosecurity emergency preparedness and response capabilities. This article introduces the main contents and framework of the 2023 UK Biosecurity Strategy in detail, and reflects on its implications to provide a reference for the construction and development of China's biosecurity strategy.

The bacterial communities in rhizosphere soil interact with the roots of plants. This interaction is beneficial for both the bacteria and the plants, which makes it very important to identify the structure of these bacterial communities for plant growth and development. However, the composition characteristics of bacterial communities in rhizosphere soil of 2-year and 5-year Haloxylon ammodendron have not been clearly defined. The purpose of this study was to identify the diverse composition of 2-year and 5-year Haloxylon ammodendron in Turpan, Xinjiang. Thus, rhizosphere soil bacteria were analyzed by isolating, purifying, and identifying the species through high-throughput sequencing technology. The bacterial strains in the rhizosphere soil of Haloxylon ammodendron were isolated with the dilution coating method, resulting in 37 isolated strains. The selective media were used to screen the growth-promoting characteristics of the rhizosphere soil isolates of Haloxylon ammodendron. The results of high-throughput amplification sequencing showed that the rhizosphere bacteria in the 2-year rhizosphere soil belonged to 45 phyla, 109 classes, 288 orders, 451 families, 826 genera, and 404 species, and those in the 5-year rhizosphere soil belonged to 56 phyla, 148 classes, 369 orders, 601 families, 1062 genera, and 671 species. Among them, Firmicutes, Proteobacteria, Actinobacteriota, Bacteroidota, Crenarchaeota, and so on are the dominant bacteria. There were 12206 and 14,053 OTUs in the 2-year-old and 5-year-old rhizosphere soil bacteria, respectively, and 3329 OTUs in the 2-year- and 5-year-old rhizosphere soil, accounting for 16.98% of the total number of OTUs. The results showed that three strains, sg16, sg21, and ss4, had the highest inorganic phosphorus solubility index (1.58). The isolated strain did not have the ability to dissolve organophosphorus and potassium, while the screened strain sg16 had the ability to fix nitrogen. Two strains with a good iron-bearing capacity, Sg9F and Sg1, were screened, among which Sg9F had the highest D/d value and Sg9F had the strongest iron-bearing capacity. The results showed that 37 strains of rhizosphere soil bacteria belonged to six genera. They are Bacillus, Corynebacterium, Phyllobacterium, Lysinibacillus, Sinorhizobium meliloti, and Streptomyces levis. Among them are sg21 (Bacillus sp.), sg1 (Bacillus sp.), sg9F (Streptomyces levis), sg16 (Phyllobacterium phragmitis), and ss4 (Sinorhizobium meliloti). This study provides a particular research basis for the influence of Haloxylon ammodendron rhizosphere bacteria on soil nutrient release and depicts a solution for improving the yield and quality of cistanche deserticola indirectly through isolating, screening, and identifying rhizosphere soil bacteria, including screening strains with growth-promoting functions and analyzing the population structure of rhizosphere bacteria in 2- and 5-year soil in combination with high-throughput sequencing technology.

A micro-tiller vibrates severely during the rotary tillage process, which may cause operators to develop white finger disease. However, for most vibration models, the acting force between the soil and the rotary cutter roll was simplified to only a constant or sine curve, which may not describe the whole dynamic. Rotary tillage processes have been simulated based on the smoothed particle hydrodynamics method in this paper. The acting forces of the soil on the cutter roll have been obtained with the simulation model. Four different working conditions were simulated. The average error between the calculated forces and the simulated mean forces is 10.96%, which proves the SPH model. By introducing simulated acting forces into the vibration model, a new vibration model of the micro-tiller, which includes the soil–blade interaction, has been constructed. Time and frequency characteristics were simulated with the new vibration model. The errors between the simulated and tested RMS values are 4.28%, 5.03%, and 6.35% for the engine, cutter roll, and right handle, respectively. Two domain-dominant frequencies were found with the vibration model, namely 44.7 Hz and 257.0 Hz. It is helpful to reveal the whole dynamic map of micro-tillers.

Introduction Tobacco smoking is a major risk factor for various diseases worldwide, including pancreatic exocrine diseases such as pancreatitis and pancreatic cancer (PC). Currently, few studies have examined the impact of smoking cessation on the likelihood of common pancreatic exocrine diseases. This study sought to determine whether smoking cessation would reduce pancreatitis and PC morbidity. Methods This cohort study used data from the UK Biobank (UKB) to examine the association between smoking status and the likelihood of pancreatitis and PC among 492855 participants. The subjects were divided into never smokers, ex-smokers, and current smokers. Using a multivariate-adjusted binary logistic regression model, we analyzed the relationship between different smoking conditions and the likelihood of pancreatitis and PC. Further, we studied the impact of smoking cessation on pancreatitis and PC compared with current smoking. Results After adjusting for potential confounders, current smokers had higher odds for acute pancreatitis (AP) (AOR=1.38; 95% CI: 1.18–1.61), chronic pancreatitis (CP) (AOR=3.29; 95% CI: 2.35–4.62) and PC (AOR=1.72; 95% CI: 1.42–2.09). People who quit smoking had comparable odds for the diseases as those who never smoked. Compared with current smokers, ex-smokers had reduced odds for AP (AOR=0.76; 95% CI: 0.64–0.89), CP (AOR=0.31; 95% CI: 0.21–0.46), and PC (AOR=0.62; 95% CI: 0.50–0.76). Subgroup analysis revealed reduced odds for these pancreatic diseases in males and females. Conclusions Smokers have an increased odds for pancreatitis and pancreatic cancer. Moreover, smoking cessation can significantly reduce the odds for acute pancreatitis, chronic pancreatitis and pancreatic cancer.

Abstract Objectives Rheumatoid arthritis (RA) is a severe and common autoimmune disease. Conventional diagnostic methods are often subjective, error-prone, and repetitive works. There is an urgent need for a method to detect RA accurately. Therefore, this study aims to develop an automatic diagnostic system based on deep learning for recognizing and staging RA from radiographs to assist physicians in diagnosing RA quickly and accurately. Methods We develop a CNN-based fully automated RA diagnostic model, exploring five popular CNN architectures on two clinical applications. The model is trained on a radiograph dataset containing 240 hand radiographs, of which 39 are normal and 201 are RA with five stages. For evaluation, we use 104 hand radiographs, of which 13 are normal and 91 RA with five stages. Results The CNN model achieves good performance in RA diagnosis based on hand radiographs. For the RA recognition, all models achieve an AUC above 90% with a sensitivity over 98%. In particular, the AUC of the GoogLeNet-based model is 97.80%, and the sensitivity is 100.0%. For the RA staging, all models achieve over 77% AUC with a sensitivity over 80%. Specifically, the VGG16-based model achieves 83.36% AUC with 92.67% sensitivity. Conclusion The presented GoogLeNet-based model and VGG16-based model have the best AUC and sensitivity for RA recognition and staging, respectively. The experimental results demonstrate the feasibility and applicability of CNN in radiograph-based RA diagnosis. Therefore, this model has important clinical significance, especially for resource-limited areas and inexperienced physicians.

Abstract With the advancement of science and technology, coal-washing plants are transitioning to intelligent, information-based, and professional sorting systems. This shift accelerates the construction a modern economic system characterized by green and low-carbon development, thereby promoting the high-quality advancement of the coal industry. Traditional manual gangue picking and multi-axis robotic arm gangue selection currently suffer from low recognition accuracy, slow sorting efficiency, and high worker labor intensity. This paper proposes a deep learning-based, non-contact gangue recognition and pneumatic intelligent sorting system. The system constructs a dynamic database containing key feature information such as the target gangue's contour, quality, and center of mass. The system elucidates the relationships between ejection speed, mass, volume, angle of incidence, and the impact energy matching mechanism. Demonstration experiments using the system prototype for coal gangue sorting reveal that, compared to existing robotic arm sorting methods in coal washing plants, this system achieves a gangue identification accuracy exceeding 97%, a sorting rate above 91%, and a separation time of less than 3 s from identification to separation, thereby effectively enhancing raw coal purity.

Abstract Tafamidis is the only disease-modifying therapy approved to treat patients in the United States with transthyretin amyloid cardiomyopathy (ATTR-CM), which most commonly affects patients aged ≥ 65 years. The manufacturer operates a patient assistance program (PAP) to support access to tafamidis. This study conducted Privacy Preserving Record Linking (PPRL) using Datavant tokens to match patients across Medicare prescription drug plan (PDP) and PAP databases to evaluate the impact of PAPs on treatment exposure classification, adherence, and persistence determined using Medicare PDP data alone. We found 35% of Medicare PDP patients received tafamidis through the PAP only; 14% through both Medicare PDP and the PAP, and 51% through Medicare PDP only. Adherence and persistence were comparable between these cohorts but underestimated among patients who received ≥ 2 prescriptions through Medicare PDP and ≥ 1 through the PAP when solely using Medicare data versus pooled Medicare and PAP data (modified Medication Possession Ratio: 84% [69% ≥ 80% adherent] vs. 96% [93%]; Proportion of Days Covered: 77% [66% ≥ 80% adherent] vs. 88% [88%]; mean days to discontinuation: 186 vs. 252; total discontinuation: 13% vs. 11%). Cross-database PPRL is a valuable method to build more complete treatment journeys and reduce the risk of exposure misclassification in real-world analyses.

Abstract The complex tip flow instability and its induced non-synchronous vibration have become significant challenges, especially as aerodynamic loading continues to increase. This study investigates the effects of tip clearance on non-synchronous propagating flow disturbances of compressor rotors under high aerodynamic loading conditions by conducting full-annulus unsteady numerical simulations with three typical tip clearance values for a 1-1/2 stage transonic compressor. The non-synchronous aerodynamic excitation frequency, circumferential mode characteristics, and annular unstable flow structures are analyzed under near stall conditions. The results show that the total pressure ratio and normalized mass flow parameters first increase and then decrease as the tip clearance increases from 0.5%C (where C represents the tip chord length) to 2%C under high aerodynamic loading conditions, instead of constantly decreasing. For the 0.5%C tip clearance case, the traveling large-scale tornado-like separation vortices cause a low non-synchronous aerodynamic excitation frequency and severe pressure fluctuations. The periodic shedding and reattachment processes of the rotor blades separated by 2 – 3 pitches result in 19 dominant mode orders in the circumferential direction. As the tip clearance increases from 1%C to 2%C, the difference of tip flow structures in each blade passage is significantly weakened, and the dominant mode order of the disturbance is equal to the rotor blade-passing number. The pressure fluctuation is mainly caused by cross-channel tip leakage flow, and the aerodynamic excitation frequency exhibits evident broadband hump characteristics, which has been reported as a rotating instability phenomenon.

Traditional separation membranes used for dye removal often suffer from a trade-off between separation efficiency and water permeability. Herein, we propose a facile approach to prepare cyclodextrin-based high-flux nanofiber membranes by electrospinning and vapor-driven crosslinking processes. The application of glutaraldehyde vapor for crosslinking hydroxypropyl-β-cyclodextrin (HP-β-CD)/polyvinyl alcohol (PVA)/laponite electrospun membranes can build interconnected structures and lead to the formation of a porous hierarchical layer. In addition, the incorporation of inorganic salt, laponite, can alter the crosslinking process, resulting in membranes with improved hydrophilicity and highly maintained electrospun nanofibrous morphology, which contributes to an ultrafast water flux of 1.0 × 105 Lh−1m−2bar−1. Due to the synergetic effect of strong host–guest interaction and electrostatic interaction, the membranes exhibit suitable rejection toward anionic dyes with a high removal efficiency of >99% within a short time and achieve accurate separation for cationic against anionic dyes, accompanied by suitable recyclability with >97% separation efficiency after at least four separation–regenerations. The prepared membranes with remarkable separation efficiency and ultrafast permeation properties might be a promising candidate for high-performance membranes in water treatment.

Abstract Background First rib tumors are extremely rare. Its compression of neurovascularity can easily lead to severe complications such as thoracic outlet syndrome, so early surgical resection is crucial. However, there is no standardized approach to surgery. Case presentation A previously healthy 18-year-old Chinese male undergoes a chest computed tomography (CT) scan that incidentally reveals a raised calcified mass on the right first rib, which is most likely an osteochondroma when combined with magnetic resonance imaging (MRI). We achieved excellent results with resection and thoracic reconstruction by adopting an inverse L-shaped incision in the anterior chest and a longitudinal split of the sternum. Conclusions Our practice provides great reference for the surgical management of first rib tumors.

Abstract Background The modified pancreatitis activity scoring system (mPASS) was proposed to assess the activity of acute pancreatitis (AP) while it doesn’t include indicators that directly reflect pathophysiology processes and imaging characteristics. Objectives To determine the threshold of admission mPASS and investigate radiomics and laboratory parameters to construct a model to predict the activity of AP. Methods AP inpatients at institution 1 were randomly divided into training and validation groups based on a 5:5 ratio. AP inpatients at Institution 2 were served as test group. The cutoff value of admission mPASS scores in predicting severe AP was selected to divide patients into high and low level of disease activity group. LASSO was used in screening features. Multivariable logistic regression was used to develop radiomics model. Meaningful laboratory parameters were used to construct combined model. Results There were 234 (48 years ± 10, 155 men) and 101 (48 years ± 11, 69 men) patients in two institutions. The threshold of admission mPASS score was 112.5 in severe AP prediction. The AUC of the radiomics model was 0.79, 0.72, and 0.76 and that of the combined model incorporating rad-score and white blood cell were 0.84, 0.77, and 0.80 in three groups for activity prediction. The AUC of the combined model in predicting disease without remission was 0.74. Conclusions The threshold of admission mPASS was 112.5 in predicting severe AP. The model based on CECT radiomics has the ability to predict AP activity. Its ability to predict disease without remission is comparable to mPASS. Critical relevance statement This work is the first attempt to assess the activity of acute pancreatitis using contrast-enhanced CT radiomics and laboratory parameters. The model provides a new method to predict the activity and prognosis of AP, which could contribute to further management. Key Points Radiomics features and laboratory parameters are associated with the activity of acute pancreatitis. The combined model provides a new method to predict the activity and prognosis of AP. Abstract Reactive jammers select jamming strategies according to the users’ responses; thus, conventional anti‐jamming methods such as frequency hopping are inadequate to defeat the jamming attack. In this article, the authors propose a novel uncoupled deception scheme to trap the reactive jammer into attacking a decoy channel in distributed networks. Specifically, the authors design a multi‐functional network utility for every user to mislead the jammer with a minimum energy consumption while achieving the highest network throughput. Based on the network utility, the anti‐jamming problem is formulated as an exact potential game such that the existence of Nash equilibrium can be guaranteed theoretically. The authors further propose a back‐to‐back coordination‐based learning algorithm to reach the optimal channel selection and power adaption in a non‐cooperative way. To alleviate the lack of mutual information exchange, the back‐to‐back coordination mechanism derives all users to deceive the jammer by inferring others’ strategies based on a shared belief. Simulation results show that the proposed algorithm yields higher network throughput and efficiency‐cost ratio compared to the state‐of‐the‐art cooperative schemes.

Abstract To address the problems of wall collapse, cuttings slurrying, and scaling on the inner wall of the drill pipe, which often occur in small-diameter diamond wireline core drilling, the inhibition properties of polyvinyl alcohol (PVA) was investigated, and the formulation of solids-free drilling fluid with PVA as the inhibitor were completed. PVA has the advantages of fast adsorption and easy regulation of rheological properties Firstly the inhibition effect of PVA was compared with that of common inorganic salts (sodium chloride, NaCl, potassium chloride, KCl) by bentonite dispersion test, linear swelling test, shale rolling recovery test and mud ball immersion test in this study. Then, the inhibition mechanism of PVA was analyzed with potentiometric particle size tests, Fourier transform infrared (FT-IR) and X-ray Diffraction (XRD) measurements. Based on the outstanding inhibition performance of PVA, tackifiers and filtration reducers were preferred through the compatibility test. And finally, the effects of various contaminants on the comprehensive performance of the formulated solids-free drilling fluids were evaluated. The results showed that PVA exhibited better inhibition of clay hydration and dispersion in shale recovery and linear swelling compared to NaCl and KCl, which was particularly evident in the mud ball immersion test. FT-IR and XRD tests revealed that the inorganic salts were used to replace the cations with larger radius and high degree of hydration in the clay layer by ion exchange ti achieve the effect of clay de-watering by reducing the spacing of the clay interlayer and the electrostatic repulsion between the particles. However, PVA is strongly adsorbed on the clay surface in the form of hydrogen bonds due to its unique multi-hydroxyl chain structure, forming a hydrophobic barrier to prevent water molecules from entering the clay layer, thus inhibiting the hydration and swelling of the clay. Using PVA as an inhibitor, compounded with xanthan gum, sulfonated lignite resin and sulfonated gilsonite (FT-1), the solids-free drilling fluid is promising for use in diamond wireline core drilling in complex formations.

Abstract Background Malnutrition is related to impaired oral health and function that causes poor dietary intake, declining the general health of older adults. The role of dietary intake in the association between oral function and nutritional status of Chinese older adults (aged 75 and above) was examined in this cross-sectional study. Methods Through the randomized cluster sampling method, 267 older adults living in rural areas of Qingdao, Shandong (aged 81.4 ± 4.3, 75–94 years) were chosen as the primary research participants. A Mini Nutritional Assessment - Short Form was used to determine nutritional status, and Food Frequency Questionnaire and 24-hour Food Intake Recall were used to assess dietary intake. The oral function was evaluated by analyzing the teeth, oral problems, bite force, tongue pressure, lip sealing pressure, chewing function questionnaire, whole saliva flow rate, 10-Item Eating Assessment Tool, and water swallow test. Results Based on the MNA-SF score, it was divided into a well-nourished group and a malnutrition group, with the malnutrition group comprising 40.6% of participants. The participants in the malnutrition group showed a higher rate of xerostomia, lower bite force, tongue pressure, and lip sealing pressure, and higher Chewing Function Questionnaire and 10-Item Eating Assessment Tool scores. Furthermore, their plant fat, iron, cereals and potatoes, vegetables, fruits, and seafood intake were relatively low. The regression model indicated that exercise frequency, stroke, chewing and swallowing function, intake of vegetables and fruits were risk factors for nutritional status of older adults. Conclusion Malnutrition was relatively common among the Chinese older adults aged 75 and above, and it was significantly correlated with exercise frequency, stroke, chewing and swallowing function, and intake of vegetables and fruits. Therefore, nutrition management should be carried out under the understanding and guidance of the oral function and dietary intake of the older adults.

Abstract Due to the challenge of collecting a substantial amount of production-quality data in real-world industrial settings, the implementation of production quality prediction models based on deep learning is not effective. To achieve the goal of predicting production quality with limited data and address the issue of model degradation in the training process of deep learning networks, we propose Meta-Learning based on Residual Network (MLRN) models for production quality prediction with limited data. Firstly, the MLRN model is trained on a variety of learning tasks to acquire knowledge for predicting production quality. Furthermore, to obtain more features with limited data and avoid the issues of gradient disappearing or exploding in deep network training, the enhanced residual network with the effective channel attention (ECA) mechanism is chosen as the basic network structure of MLRN. Additionally, a multi-batch and multi-task data input approach is implemented to prevent overfitting. Finally, the availability of the MLRN model is demonstrated by comparing it with other models using both numerical and graphical datasets.

Abstract Heterochromatin is generally associated with the nuclear periphery, but how the spatial organization of heterochromatin is regulated to ensure epigenetic silencing remains unclear. Here we found that Sad1, an inner nuclear membrane SUN-family protein in fission yeast, interacts with histone H2A-H2B but not H3-H4. We solved the crystal structure of the histone binding motif (HBM) of Sad1 in complex with H2A-H2B, revealing the intimate contacts between Sad1HBM and H2A-H2B. Structure-based mutagenesis studies revealed that the H2A-H2B-binding activity of Sad1 is required for the dynamic distribution of Sad1 throughout the nuclear envelope (NE). The Sad1-H2A-H2B complex mediates tethering telomeres and the mating-type locus to the NE. This complex is also important for heterochromatin silencing. Mechanistically, H2A-H2B enhances the interaction between Sad1 and HDACs, including Clr3 and Sir2, to maintain epigenetic identity of heterochromatin. Interestingly, our results suggest that Sad1 exhibits the histone-enhanced liquid-liquid phase separation property, which helps recruit heterochromatin factors to the NE. Our results uncover an unexpected role of SUN-family proteins in heterochromatin regulation and suggest a nucleosome-independent role of H2A-H2B in regulating Sad1’s functionality.

In the present work, a novel high-V high-Mn twinning induced plasticity (TWIP) steel for cryogenic storage tank applications was developed. Furthermore, innovative two-stage cooling processes encompassing ultra-fast cooling and the subsequent furnace cooling were proposed. Effects of cooling process on the microstructure and mechanical properties of the experimental steel were investigated. It was observed that with the change in cooling process (the initial temperature of furnace cooling rised gradually), the dwell time of hot-rolled plates above 600 °C was extended. The extension in dwell time facilitated the precipitation of secondary phase particles, leading to the change in average diameter and volume fraction of secondary phase particles. The yield strength increased with the increasing initial temperature of furnace cooling due to the combined effect of small-sized secondary phase particles inside the grains and large-sized ones located at the grain boundaries. The occurrence of intergranular cracking, primarily caused by the presence of large-sized secondary phase particles at the grain boundaries, was the main factor responsible for the significant reduction in total elongation. The small-sized secondary phase particles inside the grains inhibited deformation twinning, while the large-sized ones at the grain boundaries weaken the binding force between grain boundaries, which ultimately resulted in a decrease in cryogenic impact toughness. The implementation of two-stage cooling processes effectively mitigated the trade-off between yield strength and cryogenic impact toughness, offering a novel approach and valuable insights for optimizing the strength and toughness characteristics of cryogenic high-Mn steels.

Clean and efficient recycling of spent lithium-ion batteries (LIBs) has become an urgent need to promote sustainable and rapid development of human society. Therefore, we provide a critical and comprehensive overview of the various technologies for recycling spent LIBs, starting with lithium-ion power batteries. Recent research on raw material collection, metallurgical recovery, separation and purification is highlighted, particularly in terms of all aspects of economic efficiency, energy consumption, technology transformation and policy management. Mechanisms and pathways for transformative full-component recovery of spent LIBs are explored, revealing a clean and efficient closed-loop recovery mechanism. Optimization methods are proposed for future recycling technologies, with a focus on how future research directions can be industrialized. Ultimately, based on life-cycle assessment, the challenges of future recycling are revealed from the LIBs supply chain and stability of the supply chain of the new energy battery industry to provide an outlook on clean and efficient short process recycling technologies. This work is designed to support the sustainable development of the new energy power industry, to help meet the needs of global decarbonization strategies and to respond to the major needs of industrialized recycling.

Background: Stage III and IVA–B hypopharyngeal carcinoma presents a substantial risk of recurrence and metastasis. The treatment strategy remains uncertain. The objective of this observational study was to compare the outcomes of induction chemotherapy followed by radiotherapy (ICRT) and induction chemotherapy followed by chemoradiotherapy (ICCRT) in the treatment of locally advanced hypopharyngeal squamous cell carcinoma. Methods: 58 patients with stage III and IVA–B hypopharyngeal squamous cell carcinoma treated with ICRT (n = 26) or ICCRT (n = 32) were enrolled in the study. Baseline variables and toxicity rates were compared by Chi-squared test. Survival curves were constructed by the Kaplan-Meier method and compared by log-rank test. Multivariate Cox proportional hazard analysis was performed to evaluate the potential survival effects. Results: There were no significant differences in gender, age, smoking, drinking, T category, N category, overall stage, induction chemotherapy schemes and cycles between the two groups. The median follow-up time was 36.3 months (range, 2.3–97.5 months). The 2-year recurrence-free survival (RFS), locoregional recurrence-free survival (LRFS), distant metastasis-free survival (DMFS), and the 1-year, 2-year overall survival (OS) expressed no significant differences between the two groups. Furthermore, induction chemotherapy regimen of TPF achieved better OS than TP or PF (hazard ratio [HR] 0.395, 95 % confidence interval [CI] 0.178–0.879; P = 0.023), OS of patients in N2-3 category was worse than N0-1 (HR 2.594, 95 % CI 1.230–5.471; P = 0.012). In addition, the grade 3–4 therapy-associated toxicities during radiotherapy were higher in the chemoradiotherapy group than in radiotherapy alone group (P = 0.020). Conclusion: Following induction chemotherapy in patients with stage III/IVA-B hypopharyngeal squamous cell carcinoma, the concurrent chemoradiotherapy regimen provided similar survival rates with radiotherapy alone. Meanwhile, the incidence of treatment-related side effects during radiotherapy after induction chemotherapy were lower than that during chemoradiotherapy.

BackgroundThe inflammatory response is the main pathophysiological basis of acute exacerbations of chronic obstructive pulmonary disease (AECOPD) and is a key factor leading to frequent exacerbations and disease progression. Suppressing the inflammatory response can improve pulmonary function, prognosis, and quality of life in AECOPD patients.PurposeTo evaluate the effect of Qingjin Huatan decoction (QHD) on pulmonary function and inflammatory mediators in AECOPD patients.MethodsRandomized controlled trials (RCTs) on the treatment of AECOPD with QHD were retrieved from eight Chinese and English electronic databases up to 31 May 2024. The quality of the studies was assessed using the Cochrane Risk of Bias Tool and the modified Jadad scale. Statistical analysis, sensitivity analysis, and publication bias assessment were performed using Stata 17.0 software.ResultsA total of 40 RCTs involving 3,475 AECOPD patients were included. Compared to conventional treatment, QHD significantly improved pulmonary function, with increases in FEV1 (MD = 0.30, 95% CI: 0.26 to 0.34, p = 0.000), FVC (MD = 0.34, 95% CI: 0.27 to 0.41, p = 0.000), FEV1/FVC (MD = 6.07, 95% CI: 5.55 to 6.58, p = 0.000), and PaO2 (MD = 7.20, 95% CI: 4.94 to 9.47, p = 0.000), and a decrease in PaCO2 (MD = −5.37, 95% CI: 7.99 to −2.74, p = 0.000). QHD also significantly suppressed the expression of inflammatory mediators, including TNF-α (MD = −10.87, 95% CI: 12.51 to −9.23, p = 0.000), IL-1β (MD = −13.63, 95% CI: −16.31 to −10.95, p = 0.000), IL-6 (MD = −7.58, 95% CI: −10.10 to −5.06, p = 0.000), IL-8 (MD = −9.45, 95% CI: −12.05 to −6.85, p = 0.000), CRP (MD = −5.62, 95% CI: −6.60 to −4.65, p = 0.000), and PCT (MD = −0.84, 95% CI: −1.07 to −0.62, p = 0.000). Additionally, QHD improved clinical efficacy (RR = 4.16, 95% CI: 3.26 to 5.30, p = 0.000) without increasing the incidence of adverse reactions (RR = 1.04, 95% CI: 0.68 to 1.61, p = 0.000).ConclusionExisting evidence suggests that QHD can significantly improve pulmonary function, suppress the expression of inflammatory mediators, and enhance clinical efficacy in AECOPD patients, with a good safety profile. Summary: Infrared and visible image fusion aims to produce images that highlight key targets and offer distinct textures, by merging the thermal radiation infrared images with the detailed texture visible images. Traditional auto encoder-decoder-based fusion methods often rely on manually designed fusion strategies, which lack flexibility across different scenarios. Addressing this limitation, we introduce EMAFusion, a fusion approach featuring an enhanced multiscale encoder and a learnable, lightweight fusion network. Our method incorporates skip connections, the convolutional block attention module (CBAM), and nest architecture within the auto encoder-decoder framework to adeptly extract and preserve multiscale features for fusion tasks. Furthermore, a fusion network driven by spatial and channel attention mechanisms is proposed, designed to precisely capture and integrate essential features from both image types. Comprehensive evaluations of the TNO image fusion dataset affirm the proposed method’s superiority over existing state-of-the-art techniques, demonstrating its potential for advancing infrared and visible image fusion.

lysine is an essential amino acid with significant importance, widely used in the food, feed, and pharmaceutical industries. To meet the increasing demand, microbial fermentation has emerged as an effective and sustainable method for L-lysine production. Escherichia coli has become one of the primary microorganisms for industrial L-lysine production due to its rapid growth, ease of genetic manipulation, and high production efficiency. This paper reviews the recent advances in E. coli strain engineering and fermentation process optimization for L-lysine production. Additionally, it discusses potential technological breakthroughs and challenges in E. coli-based L-lysine production, offering directions for future research to support industrial-scale production.

Background and aimsAge-related cognitive impairment impacts a significant portion of the elderly population. Remnant cholesterol (RC) has attracted increased attention in relation to cardiovascular disease, diabetes, hypertension, and fatty liver disease. Nevertheless, its role in cognitive function is still enigmatic, prompting our exploration into the potential associations between them.MethodsA total of 1,331 participants from the NHANES (2011–2014) database, all aged over 60, were included in this investigation. Cognitive function was assessed using four widely applied tests, including the Consortium to Establish a Registry for Alzheimer’s Disease Word Learning (CERAD-WL), CERAD Delayed Recall (CERAD-DR), Animal Fluency Test (AFT), as well as Digit Symbol Substitution test (DSST). Z-score is calculated by scores from the above four tests. The association between RC, total cholesterol (TC) to RC and cognitive performance was assessed by logistic regression analyses. In addition, restricted cubic spline (RCS) regression was performed to assess non-linearity between RC and cognitive function. Subgroup analysis was performed to evaluate the robustness of the results in populations with relevant covariate variables.ResultsThose with Z-scores below the 25% quartile are defined as having cognitive impairment, totaling 498 individuals. Observationally, higher RC levels and a lower TC/RC were associated with an increased risk of cognitive impairment. After adjusting for confounding factors, the impact of RC levels on cognitive performance quartiles was consistent across various subgroups, except in individuals with trouble sleeping, no/unknown alcohol use, and no hypertension. Americans with high RC levels and trouble sleeping are more likely to develop cognitive impairment, with an odds ratio of 2.33 (95% CI: 1.18–4.59).ConclusionThis study suggests that higher RC levels and lower levels of TC/RC are associated with an increased likelihood of cognitive impairment, suggesting that RC can serve as a novel and convenient indicator for predicting the risk of cognitive impairment in the US population.

Pyrolysis technology is a green and efficient method for recycling enameled wires. However, since waste enameled wires are typically recovered from electronic waste, they often contain small amounts of wires and cables. Therefore, during the pyrolysis process of waste enameled wires, it is inevitable for the paint film and the cable sheath to undergo co-pyrolysis. Polyesterimide enameled wires (EPEsI) and polyvinyl chloride (PVC) were chosen as represent enameled wires and cable sheath materials, respectively. Using thermogravimetric analysis with various pyrolysis kinetic analysis methods, the pyrolysis characteristics and kinetics of EPEsI and Mixture (mixture of EPEsI and PVC) were studied. Through synergy analysis and pyrolysis-gas chromatography/mass spectrometry analysis, the influence of PVC on the pyrolysis of EPEsI was elucidated from aspects such as pyrolysis characteristics and product distribution. Based on density functional theory calculations and wave function analysis, the role of endogenous metal Cu in EPEsI on the pyrolysis processes of PEsI and PVC, as well as the mechanism of HCl from PVC on the pyrolysis of PEsI, were clarified.

Sustainable management of marine fish resources requires scientifically informed advice, which is complicated by uncertainty in biological dynamics, difficulty communicating scientific concepts to a myriad of stakeholders, and the challenge of empirically testing management options. Management strategy evaluation (MSE) is a modeling tool that is increasingly applied to simulate and test the robustness and efficacy of long-term management strategies and short-term management regulations. The research team developed a bespoke web-based MSE decision-support tool tailored to the high-profile and highly contentious Gulf of Mexico (GoM) red snapper (Lutjanus campechanus) resource. The MSE tool quantifies the risks and trade-offs among a variety of performance measures when alternative management strategies and regulations, which have been proposed for rebuilding the red snapper resource, are implemented. The operating model mimics the intricate population dynamics, fishery processes, and management procedures of red snapper, and provides users with the ability to manipulate many aspects of the system. Moreover, uncertainty is accounted for through the integration of process, observation, assessment, and implementation error. A user-friendly graphical interface facilitates management scenario input, while sophisticated output visualization tools provide management insights through interactive graphics, allowing users to compare performance across multiple scenarios simultaneously. Many unique facets of the tool, including iterative feedback, beta testing by a variety of stakeholders, as well as technical review by scientists, are essential practices that enhance usability, ensure versatility, and foster user engagement; and we recommend this multifaceted approach for the development of future MSE tools. Ultimately, this free-to-use, convenient, fast-running decision support tool serves as a platform for promoting science-based red snapper fishery management. It paves the way for a complete stakeholder-engaged MSE process, which would enhance and facilitate collaboration among scientists, resource managers, and other stakeholders, thus fostering the sustainable management of the GoM red snapper resource.

The conventional droop method for load sharing control in DC microgrid suffers from poor power sharing and voltage regulation, which affects the State of Charge (SoC) balance of distributed Battery Energy Units (BEUs). In this paper, an adaptive droop method based on superimposed frequency is proposed to balance the SoC among distributed BEUs. Accurate current sharing is achieved by using the introduced AC reactive power to adjust the reference voltage according to differences in line resistance. With the proposed SoC balance control method, the SoC balance among BEUs can be guaranteed by the accurate current sharing regardless of the different line resistances in the DC microgrid. In addition, voltage restoration is performed to maintain the average output voltage of the BEU at the nominal value. The effectiveness of the proposed control system is verified by simulation and HIT experimental tests.

Parametric uncertainties and inverter nonlinearity exist in the permanent magnet synchronous motor (PMSM) drive system, which may lead to performance degradation or failure of model-based control. This paper presents an enhanced model-free synergetic control method for PMSMs, addressing the limitations of relying on accurate motor models in existing speed control strategies. The proposed approach aims to achieve robust control performance in the presence of parameter perturbations without the need for an accurate motor model. Firstly, a new ultra-local model of the speed loop for the PMSM is established based on a newly developed ultra-local theory. Subsequently, a model-free synergetic controller is designed using the principles of synergetic control theory. To address the issue of output chattering in the sliding mode observer used to estimate the unknown part of the ultra-local model, a synergetic observer method is proposed. Simulation results are presented and compared with those obtained using a proportional-integral (PI) controller and a conventional model-free sliding mode controller. The results demonstrate that the model-free synergetic controller exhibits robust performance and provides accurate estimation of the unknown part without output chattering.

Abstract Background Mecapegfilgrastim, a long‐acting granulocyte‐colony stimulating factor has been approved for reducing the incidence of infection, particularly febrile neutropenia (FN), in China. Objective We conducted a multicenter prospective observational study to examine the safety and effectiveness of mecapegfilgrastim in preventing neutropenia in gastrointestinal patients receiving the chemotherapy, including S‐1/capecitabine‐based regimens or the fluorouracil, leucovorin, oxaliplatin, and irinotecan (FOLFOXIRI)/fluorouracil, leucovorin, and oxaliplatin (FOLFOX)/fluorouracil, leucovorin, oxaliplatin, and irinotecan (FOLFIRINOX) regimens. Method Five hundred and sixty‐one gastrointestinal patients from 40 sites across China, between May 2019 and November 2021, were included. The administration of mecapegfilgrastim was prescribed at the discretion of local physicians. Results The most common adverse drug reactions (ADRs) of any grade for all patients was increased white blood cells (2.9%). Grade 3/4 ADRs were observed for anemia (0.2%), decreased white blood cells (0.2%), and decreased neutrophil count (0.2%). Among the 116 patients who received S‐1/capecitabine‐based chemotherapy throughout all cycles, ADRs of any grade included anemia (1.7%), myalgia (0.9%), and increased alanine aminotransferase (0.9%). No grade 3/4 ADRs were observed. In 414 cycles of patients who underwent S‐1/capecitabine‐based regimens, only one (0.2%) cycle experienced grade 4 neutropenia. In the FOLFIRINOX, FOLFOXIRI, and FOLFOX chemotherapy regimens, grade 4 neutropenia occurred in one (2.7%) of 37 cycles, four (4.7%) of 85 cycles, and two (1.2%) of 167 cycles, respectively. Conclusion In a real‐world setting, mecapegfilgrastim has proven effective in preventing severe neutropenia in gastrointestinal patients following chemotherapy. This includes commonly used moderate or high‐risk FN regimens or regimens containing S1/capecitabine, all of which have demonstrated favorable efficacy and safety profiles.

Riparian vegetation always grows on both sides of a natural river during a dam-break event, while the river typically retains some water instead of completely drying out. The RNG k-ϵ turbulence model is employed to examine the evolutionary characteristics of the dam-break flow in the downstream vegetated channel. This model facilitates the analysis of water-level fluctuations, velocity distributions, and the retardation effects of vegetation on the dam-break flow by varying the initial water depth (IWD) in the downstream channel. Results indicates that as the water depth in the downstream channel increases, the dam-break wave tends to form more readily, possesses a larger amplitude, and the duration of rapid congestion in the dam-break current decreases. Hence, the surface velocity gradually declines and shows an intermittent distribution. Vegetation impedes the flow evolution in the vegetated area, an effect that diminishes progressively with increasing IWD. In contrast, vegetation hastens the evolution of water flow in the main channel, which remains minimally affected by the IWD.

To study the effect of melatonin supplementation on the gut microbes of broilers, 160 healthy 3-week-old Ross 308 broilers with similar body weights were selected and randomly divided into four groups (M0, M20, M40, and M80) supplemented with 0, 20, 40, or 80 mg/kg melatonin. The results showed that the abundance-based coverage estimator (ACE) index of cecum microorganisms was significantly lower in the M80 group. The dominant phyla of intestinal contents in the M0, M20, M40, and M80 groups were Bacteroidetes and Firmicutes. The M40 group showed an increase in the relative abundance of Bacteroidetes spp. in the intestine, while the relative abundance of Ruminococcus spp. in the intestine of the M20, M40, and M80 groups was significantly greater than that of the M0 group. Kyoto Encyclopedia of Genes and Genomes (KEGG) functional analyses revealed that the supplementation of melatonin increases the expression of genes related to cellular processes (cell motility, cell growth and death, and cellular community-eukaryotes), environmental information processing (membrane transport and signal transduction), and genetic information processing (transport and transcription), and Cluster of Orthologous Groups (COG) of proteins functional analyses revealed that the supplementation of melatonin resulted in a significant increase in cellular processes and signaling (cell motility, signal transduction mechanisms, intracellular trafficking, secretion, and vesicular transport), information storage and processing (RNA processing and modification, chromatin structure and dynamics, translation, ribosomal structure, and biogenesis), metabolism (energy production and conversion, lipid transportation and metabolism, inorganic ion transport and metabolism, secondary metabolite biosynthesis, transport, and catabolism), and poorly characterized (general function prediction only). In summary, supplementation of feed with melatonin can increase the diversity of intestinal microorganisms and the relative abundance of Bacteroides and Firmicutes in the cecum, improve digestive ability and nutrient absorption ability, and positively regulate the metabolic ability of broilers.

Objective: Osteosarcoma (OS) is the most common primary bone sarcoma with a high propensity for local invasion and metastasis. Although the antitumor effect of apatinib has been well confirmed in advanced OS, the synergistic effect of apatinib and immunotherapies has not yet been elucidated. Methods: In this study, we established tumour-bearing mice and observed tumour size with low and high doses of apatinib treatments. The expression of 17 cytokines, including vascular endothelial growth factor (VEGF), was detected by protein microarray analysis. Moreover, we designed apatinib and antigen-specific dendritic cell (DC)-T combination treatment for tumour-bearing mice. Tumour growth was detected by statistical analysis of tumour size and microvessel density (MVD) counting, the protein expression of VEGF by western blotting, the cytokines interleukin 6 (IL-6), IL-17 and interferon-gamma (IFN-γ) by enzyme-linked immunosorbent assay (ELISA), and the numbers of myeloid-derived suppressor cells (MDSCs) and tumour-infiltration macrophages (TAMs) by flow cytometry. Results: The results showed that apatinib efficiently suppressed tumour growth, and high-dose apatinib achieved a stronger effect. The same was true for DC-T immunotherapy. However, their combination treatment revealed a better oncolytic effect. Meanwhile, apatinib or DC-T treatment inhibited the expression of VEGF and the proangiogenic mediators IL-6 and IL-17 but increased IFN-γ production. Combination therapy further reduced/increased these effects. In addition, the combination treatment reduced MDSC but enhanced TAM-M1 ratios in the OS microenvironment. These findings indicated that apatinib and antigen-specific DC-T combination therapy was more efficient in oncolysis by regulating pro-/anti-angiogenic inducers and improving the immune state in the OS microenvironment. Conclusion: This study proved that it was feasible to employ immunotherapy with therapeutic agents in OS treatment, which may provide a new approach in addition to the combination of surgery with chemotherapy in tumour treatment.

Abstract Numerous single‐cell transcriptomic datasets from identical tissues or cell lines are generated from different laboratories or single‐cell RNA sequencing (scRNA‐seq) protocols. The denoising of these datasets to eliminate batch effects is crucial for data integration, ensuring accurate interpretation and comprehensive analysis of biological questions. Although many scRNA‐seq data integration methods exist, most are inefficient and/or not conducive to downstream analysis. Here, DeepBID, a novel deep learning‐based method for batch effect correction, non‐linear dimensionality reduction, embedding, and cell clustering concurrently, is introduced. DeepBID utilizes a negative binomial‐based autoencoder with dual Kullback–Leibler divergence loss functions, aligning cell points from different batches within a consistent low‐dimensional latent space and progressively mitigating batch effects through iterative clustering. Extensive validation on multiple‐batch scRNA‐seq datasets demonstrates that DeepBID surpasses existing tools in removing batch effects and achieving superior clustering accuracy. When integrating multiple scRNA‐seq datasets from patients with Alzheimer's disease, DeepBID significantly improves cell clustering, effectively annotating unidentified cells, and detecting cell‐specific differentially expressed genes.
