Abstract We introduce a groundbreaking proof-of-concept for a novel glucose monitoring transducing mechanism, marking the first demonstration of a spore-forming microbial whole-cell sensing platform. The approach uses selective and sensitive germination of Bacillus subtilis spores in response to glucose in potassium-rich bodily fluids such as sweat. As the rate of germination and the number of metabolically active germinating cells are directly proportional to glucose concentration, the electrogenic activity of these cells—manifested as electricity—serves as a self-powered transducing signal for glucose detection. Within a microengineered, paper-based microbial fuel cell (MFC), these electrical power outputs are measurable and can be visually displayed through a compact interface, providing real-time alerts. The dormant spores extend shelf-life, and the self-replicating bacteria ensure robustness. The MFC demonstrated a remarkable sensitivity of 2.246 µW·(log mM)−1·cm−2 to glucose concentrations ranging from 0.2 to 10 mM, with a notably lower limit of detection at ~0.07 mM. The sensor exhibited exceptional selectivity, accurately detecting glucose even in the presence of various interferents. Comparative analyses revealed that, unlike conventional enzymatic biosensors whose performance degrades significantly through time even when inactive, the spore-based MFC is stable for extended periods and promptly regains functionality when needed. This preliminary investigation indicates that the spore-forming microbial whole-cell sensing strategy holds considerable promise for efficient diabetes management and can be extended toward noninvasive wearable monitoring, overcoming critical challenges of current technologies and paving the way for advanced biosensing applications.

Background: S100A8 is a member of the S100 protein family and plays a pivotal role in regulating inflammation and tumor progression. This study aimed to comprehensively assess the expression patterns and functional roles of S100A8 in glioma progression. Methods: Glioma tissues were collected from 98 patients who underwent surgical treatment at Fudan University Shanghai Cancer Center. S100A8 expression in glioma tissues was analyzed using immunohistochemistry (IHC) to establish its correlation with clinicopathological features in patients. The expression and prognostic effect of S100A8 in glioma were analyzed using TCGA and CGGA public databases. Then, we investigated the role of S100A8 in glioma through a series of in vivo and in vitro experiments including Transwell, wound healing, CCK8, and intracranial tumor models. Subsequently, bioinformatics analysis, single-cell sequencing and coimmunoprecipitation (Co-IP) were used to explore the underlying mechanism. Results: S100A8 was upregulated in gliomas compared to paracancerous tissues, and this phenotype was significantly correlated with poor prognosis. Subgroup analysis showed that S100A8 expression was higher in the high-grade glioma (HGG) group than that in the low-grade glioma (LGG) group. S100A8 overexpression in glioma cell lines promoted cell proliferation, migration and invasion, while silencing S100A8 reversed these effects. In vivo experiments showed that S100A8 knockdown can significantly reduce the tumor burden of glioma cells. Notably, S100A8 was observed to stimulate microglial M2 polarization by interacting with TLR4, which subsequently induced NF-κB signaling and IL-10 secretion within the tumor microenvironment. Conclusions: S100A8 promotes tumor progression by inducing phenotypic polarization of microglia through the TLR4/IL-10 signaling pathway in glioma. It might represent a therapeutic target for further basic research or clinical management of glioma.

Abstract 53BP1 plays an important role in DNA double-strand break (DSB) repair and this activity is negatively regulated by its interaction with Tudor interacting repair regulator (TIRR). However, how the TIRR-53BP1 repair axis is regulated in response to DNA damage remains elusive. Here, we demonstrate that TIRR is translocated to the cytoplasm and degraded upon DNA damage. Ubiquitination of TIRR at lysine 187 by DTX3L is a critical process that regulates NHEJ pathway activity and PARP inhibitor sensitivity by facilitating XPO1-mediated TIRR nuclear export and degradation after DNA damage. We show that DTX3L is overexpressed in prostate cancers in patients and that decreased expression of TIRR due to DTX3L overexpression impairs the negative regulatory effect of TIRR on 53BP1, which consequently induces HR deficiency and chromosomal instability and sensitizes prostate cancer cells to poly (ADP-ribose) polymerase (PARP) inhibitors. Our work reveals a dual action of DTX3L on TIRR degradation and nuclear exportation and identifies DTX3L as an upstream regulator of the TIRR-53BP1 axis that governs DNA repair pathway choice and PARP inhibitor sensitivity. These findings suggest that TIRR ubiquitination and DTX3L overexpression could be viable biomarkers predicting PARP inhibitor sensitivity in cancers.

The natural fracture system plays a key role in the formation of hydrocarbon reservoirs in the carbonate buried hill of the Nanpu Sag in the Bohai Bay Basin, affecting the distribution of high-quality reservoirs and the migration and accumulation of hydrocarbons. Using data from outcrops, cores, thin sections, and image logs, a quantitative analysis was conducted on the development patterns of fractures both in vertical and horizontal directions, and the main controlling factors for fracture development were identified. On this basis, numerical simulation techniques were applied to quantitatively predict the development patterns of fractures in the carbonate reservoirs of the ancient buried hills in Nanpu Sag. Four types of fractures were identified in the study area: structural fractures, diagenetic fractures, weathering fractures, and dissolution fractures, with structural fractures being the most predominant. The fractures show a low degree of filling, with 59% being effective, indicating good fracture effectiveness. The linear density of structural fractures ranges from 3 to 10 m−1, with an average of 5.6 m−1. The height of structural fractures is generally less than 30 cm, mainly distributed between 5 and 20 cm. The microscopic fracture areal density ranges from 25 to 50 cm/cm2, with an average of 32.3 cm/cm2, and the porosity of micro-fractures ranges from 0.24% to 0.69%, averaging at 0.55%. These micro-fractures provide essential storage space in tight reservoirs and enhance pore connectivity, facilitating hydrocarbon migration and accumulation. Three primary fracture groups were identified in the study area: nearly E–W trending fractures, NE–SW trending fractures, and NW–SE trending fractures, with the first two groups being the most developed. The degree of fracture development in the study area is mainly affected by lithology, rock mechanical layers, and faults. Fractures are most abundant in dolomite and dolomitic limestone, but less developed in mudstone. Different rock mechanical interfaces affect the geometry, scale, and intensity of fracture development. Stratigraphy-bound fractures are generally vertical and terminate at rock mechanical interfaces, while throughgoing fractures usually span multiple mechanical layers and are controlled by more extensive mechanical interfaces. Faults are important factor in fracture heterogeneity, with fracture intensity being highest near fault cores, especially at fault tips, overlaps, intersections, and the hinges of fault-associated folds. The number of fractures decreases as the distance from the fault zone increases.

In terrestrial ecosystems, carbon (C) transportation and C pool transformation processes both occur at the land-river-estuary continuum. Moreover, C budget and C balance processes are generally critical in achieving the C neutrality of terrestrial ecosystems. This study analyzes key C transportation processes at multiple interfaces that collectively constitute the land-river-estuary continuum, discusses C transportation and sedimentation processes at the land-river interface, and reveals aquatic plant C sequestration coupling processes and associated productivity. Transformation mechanisms of inorganic-organic C pools are also investigated here as well as a systematic evaluation of C transport flux within the different interfaces that constitute the land-river-estuary continuum. Results show that the net C sink of terrestrial ecosystems was 1.70 Pg C yr−1, wherein the gross primary productivity (GPP) of global terrestrial vegetation reached 123 Pg C yr−1, while rock weathering also consumed 0.30 Pg C yr−1 of atmospheric carbon dioxide (CO2). Subsequently, the C transported by the land-river-estuary continuum reached 1.70 Pg C yr−1. During this process, 0.20 Pg C is deposited and buried in inland water and 1.00 Pg C escapes from inland water systems each year. Therefore, only 0.85 Pg C is transported to the estuary. Finally, this study clarifies control mechanisms of C transportation and transformation processes at the land-river-estuary continuum. The aim of this study is to provide an important scientific basis for the quantitative analysis of C sources and sinks at the land-river-estuary continuum and C neutrality of the biosphere.

Abstract The structural basis for the allosteric interactions within G protein-coupled receptors (GPCRs) heterodimers remains largely unknown. The metabotropic glutamate (mGlu) receptors are complex dimeric GPCRs important for the fine tuning of many synapses. Heterodimeric mGlu receptors with specific allosteric properties have been identified in the brain. Here we report four cryo-electron microscopy structures of mGlu2-4 heterodimer in different states: an inactive state bound to antagonists, two intermediate states bound to either mGlu2 or mGlu4 agonist only and an active state bound to both glutamate and a mGlu4 positive allosteric modulator (PAM) in complex with Gi protein. In addition to revealing a unique PAM binding pocket among mGlu receptors, our data bring important information for the asymmetric activation of mGlu heterodimers. First, we show that agonist binding to a single subunit in the extracellular domain is not sufficient to stabilize an active dimer conformation. Single-molecule FRET data show that the monoliganded mGlu2-4 can be found in both intermediate states and an active one. Second, we provide a detailed view of the asymmetric interface in seven-transmembrane (7TM) domains and identified key residues within the mGlu2 7TM that limits its activation leaving mGlu4 as the only subunit activating G proteins.

Abstract Using 7.33 fb −1 of e + e − collision data samples collected with the BESIII detector at center-of-mass energies between 4.128 and 4.226 GeV, we search for the radiative decay D s + → γρ 770 + $$ {D}_s^{+}\to \gamma \rho {(770)}^{+} $$ for the first time. A hint of D s + → γρ 770 + $$ {D}_s^{+}\to \gamma \rho {(770)}^{+} $$ is observed with a statistical significance of 2.5σ. The branching fraction of D s + → γρ 770 + $$ {D}_s^{+}\to \gamma \rho {(770)}^{+} $$ is measured to be (2.2 ± 0.9stat. ± 0.2syst. ) × 10 −4, corresponding to an upper limit of 6.1 × 10 −4 at the 90% confidence level.

Abstract Fructus Choerospondiatis (FC) has been utilized in Mongolian medicine for the treatment of many diseases. This investigation assessed the coloration, five active components, total phenolic and flavonol contents, as well as the antioxidant and antibacterial properties of different parts of FC from eight cultivars, and the optimal cultivar was identified to determine the anti-inflammatory capacity of all parts. The findings revealed that L* and b* values of the dried peel powder were the highest and the a* value of whole fruit powder was the highest. The concentrations of gallic acid, catechin, epicatechin, total phenols, and total flavonoids exhibited a positive correlation with antioxidant activity. A comprehensive analysis revealed that the peel contained a greater abundance of active compounds and exhibited stronger antioxidant and antibacterial properties compared to other parts. RPNO.1 can be used as the source of high-quality FC for clinical application. In vivo anti-inflammatory experiments indicated that both the flesh and peel of RPNO.1 displayed the highest anti-inflammatory effect. This research not only addressed the gap in studies regarding FC’ s anti-inflammatory properties but also proved that the peel of FC, which was often discarded, had utilization value and could be used as a medicinal herb.

The tropospheric delay has a significant impact on high-accuracy positioning of the Global Navigation Satellite System (GNSS). Traditional solutions have their weaknesses. First, the estimation of tropospheric delay as a state parameter slows the positioning filter’s convergence, especially critical for Precise Point Positioning (PPP). Second, correction-based approaches, including empirical model, meteorological model and GNSS network observations, have their corresponding limitations. The empirical model comprises yearly data-based statistics, which ignores high temporal-variation components, leading to decreased correction accuracy. The meteorological model requires real-time local weather observations. One can enable the network method of the expensive regional infrastructure of GNSS stations, of which performance depends on the rover-network geometry. In this study, we enable a real-time tropospheric regional correction service by polynomial coefficients from the Kalman filtering of multisource data, including the Global Pressure and Temperature 2 wet (GPT2w) model, weather observations from the National Oceanic and Atmospheric Administration (NOAA), and GNSS network observations. After discussing the weighting strategy examined by the regional dataset from Zhejiang Province, we evaluate the performance of the proposed fusion approach with post-processed PPP results as references. We obtained the optimal weightings for the corresponding dataset, and the average accuracy for Zenith Tropospheric Delay (ZTD) is 0.43, and 1.20 cm under static, active, and overall weather conditions, respectively. Compared with the real-time GNSS network ZTD solution, our proposed fusion solution is improved by 48.21%, 55.20%, and 41.70%, respectively. In conclusion, the proposed approach makes the best of three traditional correction-based methods to provide optimized real-time tropospheric service.

To solve the problem of poor thermal stability of the current D-allulose 3-epimerase (DAEase), the ancestor sequences of DAEase with different catalytic domains were reconstructed by big data mining, reasonable modification and ancestor sequence reconstruction (ASR) strategy under the guidance of phylogenetic information. The expression vectors of the ancestor sequences were constructed, and DAEase A13 with significantly enhanced thermal stability was screened by recombinant expression and molecular docking, and its enzymatic properties were characterized. In addition, the molecular mechanism of thermal stability enhancement of DAEase A13 was revealed based on structural analysis and molecular dynamics. The results showed that the half-life of A13 constructed based on ASR strategy could reach 8.4 h at 70 ℃, indicating that its thermal stability was significantly enhanced compared with that of wild-type (WT) enzyme. The maximum conversion rate of A13 reached 31%, indicating that the catalytic activity of A13 was slightly higher than that of WT enzyme. The structural and molecular dynamics analysis revealed that the increase in hydrogen bonding and hydrophobic interaction in ASR A13 was the main factor responsible for maintaining the stability of the enzyme's molecular structure at high temperatures. The results showed that ASR strategy could modify DAEases to enhance the stability, activity or hybridity, which could provide superior biocatalyst sources for various industrial applications of functional sugars.

Abstract Using e + e − collision data collected by the BESIII detector at BEPCII corresponding to an integrated luminosity of 30 fb −1, we measure Born cross sections and effective form factors for the process e + e − → Ξ 0 Ξ ¯ 0 $$ {e}^{+}{e}^{-}\to {\Xi}^0{\overline{\Xi}}^0 $$ at forty-five center-of-mass energies between 3.51 and 4.95 GeV. The dressed cross section is fitted, assuming a power-law function plus a charmonium(-like) state, i.e., ψ(3770), ψ(4040), ψ(4160), ψ(4230), ψ(4360), ψ(4415) or ψ(4660). No significant charmonium(-like) state decaying into Ξ 0 Ξ ¯ 0 $$ {\Xi}^0{\overline{\Xi}}^0 $$ is observed. Upper limits at the 90% confidence level on the product of the branching fraction and the electronic partial width are provided for each decay. In addition, ratios of the Born cross sections and the effective form factors for e + e − → Ξ 0 Ξ ¯ 0 $$ {e}^{+}{e}^{-}\to {\Xi}^0{\overline{\Xi}}^0 $$ and e + e − → Ξ − Ξ ¯ + $$ {e}^{+}{e}^{-}\to {\Xi}^{-}{\overline{\Xi}}^{+} $$ are also presented to test isospin symmetry and the vector meson dominance model.

Chemo-dynamic therapy (CDT) has a great potential in tumor extirpation. It entails producing hypertoxic reactive oxygen species (ROS) that damage the DNA of tumor cells and other biomacromolecules. However, the efficiency of CDT is severely hampered by the massive presence of glutathione (GSH) in tumor cells and the interference of ROS defense systems, such as Mutt homolog 1 (MTH1) protein sanitizes ROS-oxidized nucleotide pools. In this research, DNA-mediated self-assembly nanoparticles (HTCG@TA NPs) were engineered with high-performance amplified oxidative damage and gene therapy effect for synergistic anti-tumor treatment. Cu2+ was converted into Cu + by redox reactions to deplete GSH while H2O2 was catalyzed to generate hydroxyl radicals (·OH). As a result, the ROS level was evidently improved. Moreover, controllable-released TH588 prevented MTH1-mediated DNA repairing, thus aggravated oxidative damage to tumor cells. Meanwhile, the released functional nucleic acid G3139 downregulated the expression of Bcl-2, and accelerated the apoptosis of tumor cells. In conclusion, the HTCG@TA demonstrated significant effect in oxidative damage amplification and tumor inhibition both in vitro and in vivo, which has provided a new outlook for the clinical application of chemo-dynamic tumor treatment and synergistic gene therapy with self-delivery nanoplatforms.

Abstract In a context characterized by the dual issues of educational inequality and problems with fertility, the internal relationship between these two factors requires further in-depth exploration. On the basis of data drawn from the China Family Panel Studies (CFPS) from 2010 to 2018, this paper investigates whether “super high schools” in cities represent a form of educational inequality, uses a combination of a two-way fixed effect model with the instrumental variable method to discuss the influence of this situation on family fertility and the corresponding transmission mechanism, develops a proportional risk regression model, and expands our understanding of the influence of educational inequality on the second-child birth interval. This study reveals that educational inequality significantly suppresses the number of family births and that the paths of action associated with this effect include both the explicit and implicit costs of education. The inhibitory effect of educational inequality on the number of family births varies according to urban grade, urban or rural classification, and the education levels attained by women. In addition to decreasing the number of family births, educational inequality increases the second-child birth interval. This paper not only enriches related research on family multichild behavior but also provides an educational reference for attempts to address the issue of continuously low fertility rates in China and establish a connection between population and educational economics.

Abstract This research aimed to assess the validity of ultrasound scans with new features in detecting fetal anal atresia and verify the effectiveness of these new features. Additionally, we aimed at investigating the perinatal incidence of anal atresia. This multicenter prospective study recruited 94,617 normal gravidas and 84 gravidas with anal atresia fetuses. The gold standard for diagnosing perinatal anal atresia is routine neonatal anus examinations. The incidence calculation was based on the results of the gold standard. The validity of our new approach was evaluated via a diagnostic test involving all 94,701 subjects. The effectiveness of our new features was assessed through an ablation study in a randomly established new dataset, with the ratio of anal atresia to non-anal atresia cases of 1:4. The annual perinatal incidence of anal atresia between 2019 and 2023 ranges from 0.57‰ to 1.29‰. Our new method performed great regarding the Youden index, diagnostic odds ratio (DOR), area under the curve (AUC) of the receiver operating characteristic curve (ROCC), AUC of the precision-recall curve (PRC), F1-score, and Cramer’s V. In the ablation study, our new approach surpassed its competitors concerning Youden index, DOR, AUC of the ROCC, and AUC of the PRC. Ultrasound scans show high validity and clinical value in detecting fetal anal atresia. Our new ultrasound features significantly promote the detection of fetal anal atresia.

Abstract A delicate balance between neural stem cell (NSC) quiescence and proliferation is important for adult neurogenesis and homeostasis. Small ubiquitin-related modifier (SUMO)-dependent post-translational modifications cause rapid and reversible changes in protein functions. However, the role of the SUMO pathway during NSC reactivation and brain development is not established. Here, we show that the key components of the SUMO pathway play an important role in NSC reactivation and brain development in Drosophila. Depletion of SUMO/Smt3 or SUMO conjugating enzyme Ubc9 results in notable defects in NSC reactivation and brain development, while their overexpression leads to premature NSC reactivation. Smt3 protein levels increase with NSC reactivation, which is promoted by the Ser/Thr kinase Akt. Warts/Lats, the core protein kinase of the Hippo pathway, can undergo SUMO- and Ubc9-dependent SUMOylation at Lys766. This modification attenuates Wts phosphorylation by Hippo, leading to the inhibition of the Hippo pathway, and consequently, initiation of NSC reactivation. Moreover, inhibiting Hippo pathway effectively restores the NSC reactivation defects induced by SUMO pathway inhibition. Overall, our study uncovered an important role for the SUMO-Hippo pathway during Drosophila NSC reactivation and brain development.

Abstract Temperature plays an important impact on rock mechanical properties. In this paper, the mechanical properties, fracture mechanism and constitutive model of marble under 20–120 °C and 15 MPa are studied. The results show marble deformation can be divided into four stages: compaction, linear elasticity, crack propagation and post-peak failure. Stress–strain curve is not obviously affected by temperature. Macroscopic fracture characteristics change from shear failure to tensile mixed failure with temperature increasing. With the increase of temperature, the strength of marble tends to decrease, indicating that temperature increase has a weakening effect on marble, and there are temperature-sensitive areas of 20–60 °C and temperature sub-sensitive areas of 60–120 °C. The elastic modulus of marble decreases and Poisson’s ratio increases with increasing temperature. The energy evolution law of marble under different temperature is basically the same, which shows that before crack initiation, the energy dissipation is less, and after the damage and yielding occurs, the energy dissipation increases quickly. The energy dissipation in the failure process is mainly used for crack initiation-connection-penetration, as well as plastic deformation caused by friction and slip of cracks, and the plastic deformation and energy dissipation have good linear characteristics. The statistical damage constitutive model based on three-parameter Weibull distribution function can effectively reflect the characteristics of post-peak plastic deformation and strain softening. The weakening effect of marble at 20–120 °C is related to its internal moisture excitation. With the increase of temperature, water is stimulated to absorb and attach to the original relatively dry interface, which plays a role in lubrication. The relative motion friction resistance between solid particles or crack surfaces decreases, which leads to crack initiation and friction energy consumption reduction, changes the specific surface energy of rocks and weakens the strength of marble. The results provide a theoretical basis for predicting and evaluating the long-term stability and safety of surrounding rock of underground deep engineering in complex environment with high ground temperature and high geo-stress.

China is one of the countries in the world with the most frequent occurrence of continental earthquakes and the greatest impacts of earthquake disasters. In particular, the Qinghai-Tibet Plateau is a region characterized by a wide spatial distribution of continental earthquakes, shallow epicenters, complex seismic origins, and significant disaster losses, making it highly susceptible to seismic events. The need for seismic geological survey is becoming increasingly urgent in this area. Based on regional geological eological data，high-precision satellite remote sensing images and DEM elevation data，this paper interprets the active faults in the region of Xigaze. The results show that the active faults in the Xigaze area are mainly concentrated in the rift valleys and their periphery, with tectonic activity varying spatially. The tectonic activity of active faults in the eastern rift is stronger than that in the western rifts. Additionally，the tectonic activity of active faults in each rift also shows a clear segmentation feature. At the same time, this paper comprehensively considers the characteristics of active faults, topography and geomorphology and stratigraphic lithology in the Xigaze area. It preliminarily evaluates the potential risk of earthquake disasters in 204 towns under its jurisdiction. The results show that there are 57 towns with “extremely high risk” “high risk” and “high-medium risk” of earthquake disasters, accounting for about 28% of the total number of towns in the region.

Abstract The branching fraction of D + → K S 0 π 0 e + ν e $$ {D}^{+}\to {K}_S^0{\pi}^0{e}^{+}{\nu}_e $$ is measured for the first time using 7.93 fb −1 of e + e − annihilation data collected at the center-of-mass energy s $$ \sqrt{s} $$ = 3.773 GeV with the BESIII detector operating at the BEPCII collider, and is determined to be B D + → K S 0 π 0 e + ν e = 0.881 ± 0.017 stat . ± 0.016 syst . % $$ \mathcal{B}\left({D}^{+}\to {K}_S^0{\pi}^0{e}^{+}{\nu}_e\right)=\left(0.881\pm {0.017}_{\textrm{stat}.}\pm {0.016}_{\textrm{syst}.}\right)\% $$ . Based on an analysis of the D + → K S 0 π 0 e + ν e $$ {D}^{+}\to {K}_S^0{\pi}^0{e}^{+}{\nu}_e $$ decay dynamics, we observe the S-wave and P-wave components with fractions of f S-wave = (6.13 ± 0.27stat. ± 0.30syst. )% and f K ¯ ∗ 892 0 = 93.88 ± 0.27 stat . ± 0.29 syst . % $$ {f}_{{\overline{K}}^{\ast }{(892)}^0}=\left(93.88\pm {0.27}_{\textrm{stat}.}\pm {0.29}_{\textrm{syst}.}\right)\% $$ , respectively. From these results, we obtain the branching fractions B D + → K S 0 π 0 S − wave e + ν e = 5.41 ± 0.35 stat . ± 0.37 syst . × 10 − 4 $$ \mathcal{B}\left({D}^{+}\to {\left({K}_S^0{\pi}^0\right)}_{S-\textrm{wave}}{e}^{+}{\nu}_e\right)=\left(5.41\pm {0.35}_{\textrm{stat}.}\pm {0.37}_{\textrm{syst}.}\right)\times {10}^{-4} $$ and B D + → K ¯ ∗ 892 0 e + ν e = 4.97 ± 0.11 stat . ± 0.12 syst . % $$ \mathcal{B}\left({D}^{+}\to {\overline{K}}^{\ast }{(892)}^0{e}^{+}{\nu}_e\right)=\left(4.97\pm {0.11}_{\textrm{stat}.}\pm {0.12}_{\textrm{syst}.}\right)\% $$ . Objective: To extract Astragalus polysaccharides (APS) from Astragalus membranaceus using compound enzymes (including papain, pectinase and cellulase), and to analyze the effect of process conditions on APS extraction. Methods: Orthogonal experiments were conducted to determine the proportion of compound enzymes. The response surface method was used to optimize the conditions of extracting APS with compound enzymes. The optimal process condition was obtained, and the α-glucosidase-inhibiting effect of APS was evaluated by pNPG method. Results: For 5 g of Astragalus herbs, the optimal ratio of papain, pectinase and cellulase were 88000 U, 65000 U and 6000 U, respectively. The optimal extraction condition was as follows: time of 2.82 h, temperature at 60.34 ℃, pH of 5.11, and solid-liquid ratio of 1:34.46 g/mL. The yield of APS under the optimal condition was up to 22.79%±0.14%. The half-inhibition concentration (IC50) of APS for α-glucosidase was 7.42 μg/mL. Conclusion: The yield of APS by compound enzymes is significantly higher than that by single enzymes, and APS can effectively inhibit the activity of α-glucosidase.

Abstract Using e + e − annihilation data sets corresponding to an integrated luminosity of 4.5 fb −1, collected with the BESIII detector at center-of-mass energies between 4.600 and 4.699 GeV, we report the first measurements of the absolute branching fractions B Λ c + → p K L 0 $$ \mathcal{B}\left({\Lambda}_c^{+}\to p{K}_L^0\right) $$ = (1.67 ± 0.06 ± 0.04)%, B Λ c + → p K L 0 π + π − $$ \mathcal{B}\left({\Lambda}_c^{+}\to p{K}_L^0{\pi}^{+}{\pi}^{-}\right) $$ = (1.69 ± 0.10 ± 0.05)%, and B Λ c + → p K L 0 π 0 $$ \mathcal{B}\left({\Lambda}_c^{+}\to p{K}_L^0{\pi}^0\right) $$ = (2.02 ± 0.13 ± 0.05)%, where the first uncertainties are statistical and the second systematic. Abstract The rapid growth of offshore wind farms (OWFs) is driven by concerns for energy security and climate change mitigation. However, their impact on marine environments remains poorly understood due to limited research. This study analyzes the effects of an OWF along China's Jiangsu Coast on seawater quality using data from different development phases. Results show the major pollutants were different across phases. Heavy metal pollution reached alert levels during construction compared to the safe levels observed in the pre-construction and operational phases, mainly due to increases in Pb, Cd, and Hg concentrations. Eutrophication was mild throughout all periods but exhibited a continuous decrease, primarily attributed to reductions in PH and COD concentrations. As a result, the comprehensive pollution level during construction was increased, but it was improved to a clean level during the operational phase. Besides, significant variations were observed in the spatial distribution patterns of major pollutant indices across different scenarios. These changes may stem from a combination effect of land-based pollution, aquaculture, OWF-induced disturbances to atmosphere and hydrodynamics, OWF-related drain and leakage contamination, and marine management policies. Understanding these effects informs OWF optimization, rational wind resource utilization, and marine ecology protection.

Abstract Background Lipid droplet (LD) is a metabolically active organelle, which changes dynamically with the metabolic state and energy requirements of cells. Proteins that either insert into the LD phospholipid monolayer or are present in the cytoplasm, playing a crucial role in lipid homeostasis and signaling regulation, are known as LD-associated proteins. Methods The keywords “lipid droplets” and “metabolic diseases” were used to obtain literature on LD metabolism and pathological mechanism. After searching databases including Scopus, OVID, Web of Science, and PubMed from 2013 to 2024 using terms like “lipid droplets”, “lipid droplet-associated proteins”, “fatty liver disease”, “diabetes”, “diabetic kidney disease”, “obesity”, “atherosclerosis”, “hyperlipidemia”, “natural drug monomers” and “natural compounds”, the most common natural compounds were identified in about 954 articles. Eventually, a total of 91 studies of 10 natural compounds reporting in vitro or in vivo studies were refined and summarized. Results The most frequently used natural compounds include Berberine, Mangostin, Capsaicin, Caffeine, Genistein, Epigallocatechin-3-gallate, Chlorogenic acid, Betaine, Ginsenoside, Resveratrol. These natural compounds interact with LD-associated proteins and help ameliorate abnormal LDs in various metabolic diseases. Conclusion Natural compounds involved in the regulation of LDs and LD-associated proteins hold promise for treating metabolic diseases. Abstract Objectives This study aimed to compare plasma concentrations of anesthetic drugs administered during Cesarean section with low Apgar score in neonates deliveried under general anesthesia and analyze associated risk factors. Methods Data from 76 neonates undergoing Cesarean section under general anesthesia with blood concentrations of anesthetic drugs were analyzed. A low Apgar score was defined as ≤ 7. Perioperative maternal and neonatal data were collected and analyzed. Neonates were divided into a control group (Group CON, n = 65) and a low Apgar score group (Group LAS, n = 11) based on Apgar score. Results There were no significant differences in the plasma concentrations of anesthetic drugs in maternal artery, umbilical vein or umbilical artery blood between the two groups. Risk factors for neonatal low Apgar scores during Cesarean section under general anesthesia were premature delivery (aOR 10.2, 95% CI = 1.8–56.9) and preoperative fetal distress (aOR 9.6, 95% CI = 1.3–69.0). The prediction model was: probability = 1/(e‑Y), Y= -4.607 + 2.318× (premature delivery) + 2.261× (fetal distress) (yes = 1, no = 0). The Hosmer–Lemeshow test showed χ²= 9.587, P = 0.213, and the area under the curve (AUC) was 0.850 (0.670 ~ 1.000). With a cutoff value of 0.695, sensitivity and specificity were 81.8% and 87.7%, respectively. Conclusions There was no correlation between blood concentration of general anesthetic drugs and Apgar score or occurrence of neonatal low Apgar scores. Premature delivery and preoperative fetal distress were identified as independent risk factors for neonatal low Apgar scores after Cesarean section under general anesthesia.

Abstract Background Bi-allelic variants in DNAH11 have been identified as causative factors in Primary Ciliary Dyskinesia, leading to abnormal respiratory cilia. Nonetheless, the specific impact of these variants on human sperm flagellar and their involvement in male infertility remain largely unknown. Methods A collaborative effort involving two Chinese reproductive centers conducted a study with 975 unrelated infertile men. Whole-exome sequencing was employed for variant screening, and Sanger sequencing confirmed the identified variants. Morphological and ultrastructural analyses of sperm were conducted using Scanning Electron Microscopy and Transmission Electron Microscopy. Western Blot Analysis and Immunofluorescence Analysis were utilized to assess protein levels and localization. ICSI was performed to evaluate its efficacy in achieving favorable pregnancy outcomes for individuals with DNAH11 variants. Results In this study, we identified seven novel variants in the DNAH11 gene in four asthenoteratozoospermia subjects. These variants led the absence of DNAH11 proteins and ultrastructure defects in sperm flagella, particularly affecting the outer dynein arms (ODAs) and adjacent structures. The levels of ODA protein DNAI2 and axoneme related proteins were down regulated, instead of inner dynein arms (IDA) proteins DNAH1 and DNAH6. Two out of four individuals with DNAH11 variants achieved clinical pregnancies through ICSI. The findings confirm the association between male infertility and bi-allelic deleterious variants in DNAH11, resulting in the aberrant assembly of sperm flagella and contributing to asthenoteratozoospermia. Importantly, ICSI emerges as an effective intervention for overcoming reproductive challenges caused by DNAH11 gene variants.

Abstract Systemic infection with Candida albicans poses a significant risk for people with weakened immune systems and carries a mortality rate of up to 60%. However, current therapeutic options have several limitations, including increasing drug tolerance, notable off-target effects, and severe adverse reactions. Over the past four decades, the progress in developing drugs to treat Candida albicans infections has been sluggish. This comprehensive review addresses the limitations of existing drugs and summarizes the efforts made toward redesigning and innovating existing or novel drugs through nanotechnology. The discussion explores the potential applications of nanomedicine in Candida albicans infections from four perspectives: nano-preparations for anti-biofilm therapy, innovative formulations of “old drugs” targeting the cell membrane and cell wall, reverse drug resistance therapy targeting subcellular organelles, and virulence deprivation therapy leveraging the unique polymorphism of Candida albicans. These therapeutic approaches are promising to address the above challenges and enhance the efficiency of drug development for Candida albicans infections. By harnessing nano-preparation technology to transform existing and preclinical drugs, novel therapeutic targets will be uncovered, providing effective solutions and broader horizons to improve patient survival rates. Abstract This study explores tourism implicit conflict and its spatial dynamics in rural China. It proposes that residents in different spatial structures within tourism communities perceive implicit conflicts differently. Data were collected through public participatory geographic information system (PPGIS) mapping and semi-structured interviews. The findings reveal that conflict perceptions vary by residential structures. Residents in core and peripheral areas experienced heightened economic and cultural conflicts. In contrast, those in expanded areas were more concerned with issues of tourism governance. The study also highlights two key spatial aspects of perceived conflict: internality, the overlap within their residential areas, and significant externality, which spreads across the entire ancient town. From a combined spatial and psychological perspective, this study proposes the concept of spatial relative deprivation to explain resident’s perceived tourism implicit conflict. This study affirms that qualitative-spatial analysis sheds light on socio-cultural phenomena, contributing to a more nuanced understanding of the underlying conflict mechanisms in a spatially explicit manner. The findings suggest that tourism planning should consider the intrinsic and micro-values of communities.

Choerospondias axillaries (CA) is an important fast-growing afforestation tree species in southern China, and its fruit has medicinal and edible value. High performance liquid chromatography was used to determine the composition and content of sugar and acid in CA fruits from different provenances, and cluster analysis was conducted on different provenances. The results showed that the total sugar content of CA fruit ranged from 49.31 to 139.41 mg/g, with sucrose accounting for the highest proportion of total sugar, followed by glucose, and fructose was the lowest. The total acid content of CA fruit ranged from 47.97 to 82.81 mg/g, with citric acid accounting for 67.09% of the total acid, followed by ascorbic acid, quinic acid, tartaric acid, and malic acid. Cluster analysis was conducted on 20 CA fruits, which were divided into 4 categories. It was recommended to develop N19 fruit had the highest content of sucrose and glucose, and the highest sweetness value, sugar-acid ratio and sweet-acid ratio. It can be suggested to be developed as a high-sugar fresh food source. N02 fruit with high sugar and high acid content can be used as a raw material for fruit cake processing. This result provides an important reference for the quality evaluation and rational development and utilization of CA.

Abstract Background Ribosome profiling (or Ribo-seq) is a technique that provides genome-wide information on the translational landscape (translatome). Across different plant studies, variable methodological setups have been described which raises questions about the general comparability of data that were generated from diverging methodologies. Furthermore, a common problem when performing Ribo-seq are abundant rRNA fragments that are wastefully incorporated into the libraries and dramatically reduce sequencing depth. To remove these rRNA contaminants, it is common to perform preliminary trials to identify these fragments because they are thought to vary depending on nuclease treatment, tissue source, and plant species. Results Here, we compile valuable insights gathered over years of generating Ribo-seq datasets from different species and experimental setups. We highlight which technical steps are important for maintaining cross experiment comparability and describe a highly efficient approach for rRNA removal. Furthermore, we provide evidence that many rRNA fragments are structurally preserved over diverse nuclease regimes, as well as across plant species. Using a recently published cryo-electron microscopy (cryo-EM) structure of the tobacco 80S ribosome, we show that the most abundant rRNA fragments are spatially derived from the solvent-exposed surface of the ribosome. Conclusion The guidelines presented here shall aid newcomers in establishing ribosome profiling in new plant species and provide insights that will help in customizing the methodology for individual research goals.

Abstract Biomolecular condensates are broadly implicated in both normal cellular regulation and disease. Consequently, several chemical biology and optogenetic approaches have been developed to induce phase separation of a protein of interest. However, few tools are available to perform the converse function – dissolving a condensate of interest on demand. Such a tool would aid in testing whether the condensate plays specific functional roles. Here we show that light-gated recruitment of a solubilizing domain, maltose-binding protein (MBP), results in rapid and controlled dissolution of condensates formed from proteins of interest. Our optogenetic MBP-based dissolution strategy (OptoMBP) is rapid, reversible, and can be spatially controlled with subcellular precision. We also provide a proof-of-principle application of OptoMBP by disrupting condensation of the oncogenic fusion protein FUS-CHOP and reverting FUS-CHOP driven transcriptional changes. We envision that the OptoMBP system could be broadly useful for disrupting constitutive protein condensates to probe their biological functions.

Abstract Background H2S imbalances in the intestinal tract trigger Crohn's disease (CD), a chronic inflammatory gastrointestinal disorder characterized by microbiota dysbiosis and barrier dysfunction. However, a comprehensive understanding of H2S generation in the gut, and the contributions of both microbiota and host to systemic H2S levels in CD, remain to be elucidated. This investigation aimed to enhance comprehension regarding the sulfidogenic potential of both the human host and the gut microbiota. Results Our analysis of a treatment-naive CD cohorts' fecal metagenomic and biopsy metatranscriptomic data revealed reduced expression of host endogenous H2S generation genes alongside increased abundance of microbial exogenous H2S production genes in correlation with CD. While prior studies focused on microbial H2S production via dissimilatory sulfite reductases, our metagenomic analysis suggests the assimilatory sulfate reduction (ASR) pathway is a more significant contributor in the human gut, given its high prevalence and abundance. Subsequently, we validated our hypothesis experimentally by generating ASR-deficient E. coli mutants ∆cysJ and ∆cysM through the deletion of sulfite reductase and L-cysteine synthase genes. This alteration significantly affected bacterial sulfidogenic capacity, colon epithelial cell viability, and colonic mucin sulfation, ultimately leading to colitis in murine model. Further study revealed that gut microbiota degrade sulfopolysaccharides and assimilate sulfate to produce H2S via the ASR pathway, highlighting the role of sulfopolysaccharides in colitis and cautioning against their use as food additives. Conclusions Our study significantly advances understanding of microbial sulfur metabolism in the human gut, elucidating the complex interplay between diet, gut microbiota, and host sulfur metabolism. We highlight the microbial ASR pathway as an overlooked endogenous H2S producer and a potential therapeutic target for managing CD. Video Abstract

Abstract Background Sirolimus is increasingly utilized in treating diseases associated with mTOR pathway overactivation. Despite its potential, the lack of evidence regarding its long-term safety across all age groups, particularly in pediatric patients, has limited its further application. This study aims to assess the long-term safety of sirolimus, with a specific focus on its impact on growth patterns in pediatric patients. Methods This pooled analysis inlcudes two prospective cohort studies spanning 10 years, including 1,738 participants (aged 5 days to 69 years) diagnosed with tuberous sclerosis and/or lymphangioleiomyomatosis. All participants were mTOR inhibitor-naive and received 1 mg/m²/day of sirolimus, with dose adjustments during a two-week titration period to maintain trough blood concentrations between 5 and 10 ng/ml (maximum dose 2 mg). Indicators of physical growth, hematopoietic, liver, renal function, and blood lipid levels were all primary outcomes and were analyzed. The adverse events and related management were also recorded. Results Sirolimus administration did not lead to deviations from normal growth ranges, but higher doses exhibited a positive association with Z-scores exceeding 2 SD in height, weight, and BMI. Transient elevations in red blood cell and white blood cell counts, along with hyperlipidemia, were primarily observed within the first year of treatment. Other measured parameters remained largely unchanged, displaying only weak correlations with drug use. Stomatitis is the most common adverse event (920/1738, 52.9%). In adult females, menstrual disorders were observed in 48.5% (112/217). Conclusions Sirolimus’s long-term administration is not associated with adverse effects on children’s physical growth pattern, nor significant alterations in hematopoietic, liver, renal function, or lipid levels. Abstract Background Prior antibiotic exposure has been identified as a risk factor for VAP occurrence, making it a growing concern among clinical practitioners. But there is a lack of systematic research on the types of antibiotics and the duration of exposure that influence VAP occurrence in children at current. Methods We retrospectively reviewed 278 children admitted to the Pediatric Intensive Care Unit (PICU) and underwent invasive mechanical ventilation (MV) between January 2020 and December 2022. Of these, 171 patients with MV duration ≥ 48 h were included in the study, with 61 of them developing VAP (VAP group) and the remaining 110 as the non-VAP group. We analyzed the relationship between early antibiotic exposure and VAP occurrence. Results The incidence of VAP was 21.94% (61/278). Abstract The processes h c → γP (P = η′, η, π 0) are studied with a sample of (27.12 ± 0.14) × 108 ψ(3686) events collected by the BESIII detector at the BEPCII collider. The decay h c → γη is observed for the first time with the significance of 9.0 σ, and the branching fraction is determined to be (3.77 ± 0.55 ± 0.13 ± 0.26) × 10 −4, while B $$ \mathcal{B} $$ (h c → γη′) is measured to be (1.40 ± 0.11 ± 0.04 ± 0.10) × 10 −3, where the first uncertainties are statistical, the second systematic, and the third from the branching fraction of ψ(3686) → π 0 h c . The combination of these results allows for a precise determination of R h c = B h c → γη B h c → γ η ′ , $$ {R}_{h_c}=\frac{\mathcal{B}\left({h}_c\to {\pi}^0\gamma \eta \right)}{\mathcal{B}\left({h}_c\to {\pi}^0\gamma {\eta}^{\prime}\right)}, $$ which is calculated to be (27.0 ± 4.4 ± 1.0)%. The results are valuable for gaining a deeper understanding of η − η′ mixing, and its manifestation within quantum chromodynamics. No significant signal is found for the decay h c → γπ 0, and an upper limit is placed on its branching fraction of B $$ \mathcal{B} $$ (h c → γπ 0) < 5.0 × 10 −5, at the 90% confidence level.

Purpose: To evaluate the effect of anterior-segment structure on vault and position after implantable collamer lens (ICL) implantation using ultrasound biomicroscopy. Methods: The retrospective case-control study included insufficient vault eyes (750 µm). The preoperative biometric parameters of the anterior-segment structure and basic data between the three groups were analyzed using one-way analysis of variance. Results: There were significant differences (P < 0.05) between the three groups in maximum ciliary body thickness (CBTmax), iris-zonule distance (IZD), and trabecular-ciliary angle (TCA). The vault gradually decreased as CBTmax decreased and TCA increased. In the pairwise comparison, the CBTmax comparison between the insufficient vault (750 µm) group was statistically significant (P = 0.024, 95% CI: −0.17–0.017 µm); the TCA comparison between the insufficient vault (750 µm) group was statistically significant (P = 0.005, 95% CI: 1.78°–12.15°); The IZD comparison between the insufficient vault (750 µm) group was statistically significant (P = 0.037, 95% CI: 0.0027–0.1119 µm). The analysis of 284 ICL haptics locations showed that there were 16.67%, 32.69%, and 70.83% haptics located in the ciliary sulcus in three groups, respectively. Conclusion: The vault and ICL haptics position are related to anterior-segment structure. A thinner and posteriorly positioned ciliary body would increase the risk of low vault and fewer ICL haptics located in the ciliary sulcus after ICL implantation. This provides guidance for the selection of the ICL size and placement position before surgery.

Alfalfa (Medicago sativa L.) plays an important role in the development of animal husbandry in the karst region of southwestern China, and karst environmental stress has a significant impact on the germination of alfalfa seeds. This study subjected alfalfa seeds to calcium salt stress (0–100 mM), drought stress (0–0.53 MPa), and pH stress (pH 3–9). Germination indicators (germination rate, germination potential, germination index, and vigor index), seedling morphological indicators (shoot length and root length), and biomass indicators were measured to assess seed stress resistance. The results showed that mild drought stress (5% PEG solution) and weak alkaline stress (pH 8) promoted seed germination, and a 20 mM CaCl2 solution significantly increased the germination rate. The root system of seedlings was more sensitive to the three types of stress. Under moderate calcium stress (40 mM), only a slight decrease in tissue water content was observed. Under moderate drought stress (10% PEG), fresh weight and tissue water content decreased, but dry weight significantly increased. Under alkaline stress (pH 9), both biomass indicators and tissue water content increased. This study provides a theoretical reference for selecting plants suitable for cultivation in karst environments.

In mammalian species, neural tissues cannot regenerate following severe spinal cord injury (SCI), for which stem cell transplantation is a promising treatment. Neural stem cells (NSCs) have the potential to repair SCI; however, in unfavourable microenvironments, transplanted NSCs mainly differentiate into astrocytes rather than neurons. In contrast, bone mesenchymal stem cells (BMSCs) promote the differentiation of NSCs into neurons and regulate inflammatory responses. Owing to their easily controllable mechanical properties and similarities to neural tissue, gelatin methacrylate (GelMA) hydrogels offer remarkable cell biocompatibility and regulate the differentiation of NSCs. Therefore, in this study, we propose co-culturing NSCs and BMSCs within low-modulus GelMA hydrogel scaffolds to promote regeneration following SCI. In vitro comparisons revealed that the viability, proliferation, migration, and neuron differentiation capacity of cells in these low-modulus scaffolds exhibit substantially superior performance compared to those in high-modulus hydrogel scaffolds. To the best of our knowledge, this study is the first to report that NSCs/BMSCs co-culture implants can remarkably enhance motor function recovery in SCI rats, reduce the area of spinal cord cavities, stimulate neuron regeneration, and suppress scar tissue formation. Thus, this hydrogel system loaded with co-cultured cells represents a promising therapeutic approach for SCI repair.

Study region: Hubei province, Yangtze River Basin, Central China Study focus: Risk and loss analysis constitutes a critical element of flood disaster management. Nevertheless, the relationship between risk and loss has been infrequently examined in the literature. Here we developed a comparative analysis framework for assessing flood risk and loss in Hubei Province. The risk model incorporates 22 indicators in terms of hazard, exposure, vulnerability and emergency response & recovery capability. Indicator weights were determined through a combination of objective and subjective weighting methods based on the game theory. Flood risk and loss levels were classified using fuzzy comprehensive evaluation and grey correlation analysis respectively. New hydrological insights: Annual risk and loss maps exhibit a spatial pattern of higher levels in the eastern regions and lower levels in the western regions. However, the relationship between risk and loss was not as well as expected during designated flood periods. While regions impacted by disasters typically demonstrated higher risk levels, the risk in the regions unaffected by flood disasters appeared to be overestimated. By analyzing flood risk and loss across various temporal scales, this research contributes a novel perspective for the evaluation of flood risk models.

Abstract Aberrant regulation of unfolded protein response (UPR)/endoplasmic reticulum (ER) stress pathway is associated with cancer development, metastasis, and relapse, and the UPR signal transducer ATF6 has been proposed as a diagnostic and prognostic marker for many cancers. However, a causal molecular link between ATF6 activation and carcinogenesis is not established. Here, it is found that tumor protein D52 (TPD52) integrates ER stress and UPR signaling with the chaperone machinery by promoting S2P‐mediated cleavage of ATF6. Although TPD52 has been generally considered as an oncogene, TPD52 is identified as a novel tumor suppressor in bladder cancer. Significantly, attenuation of the ER stress via depletion of TPD52 facilitated tumorigenesis in a subset of human carcinomas. Furthermore, the APCCdc20 E3 ligase is validated as the upstream regulator marking TPD52 for polyubiquitination‐mediated proteolysis. In addition, inactivation of Cdc20 sensitized cancer cells to treatment with the ER stress inducer in a TPD52‐dependent manner. Thus, the study suggests that TPD52 is a novel Cdc20 substrate that may modulate ER stress to prevent tumorigenesis.

ObjectivesThe purpose of this study is to examine the effects of intergenerational support and activities of daily living (ADL) on psychological distress in older adults, and to analyse the effects of the interaction between ADL and intergenerational support on psychological distress in older adults.MethodsA probability sample of 1,065 older adults aged 60 and above was conducted in Xuzhou, China, from 18 June to 26 August 2023. ADL was assessed using the ADL scale combined with the Barthel Index; Intergenerational support was measured using the Intergenerational Support Scale; and psychological distress was measured using the Kessler 10 scale. An ordered multichotomous logistic regression model was constructed to analyse the factors influencing psychological distress in the elderly. The interaction model was constructed by one-way analysis of variance to further analyse the effect of the interaction between financial support, instrumental support and emotional support and ADL on psychological distress in the elderly.ResultsElderly living in rural areas, with an annual household income of less than 30,000 yuan, who engaged in manual labor before retirement, who did not receive financial support and who received instrumental support were more likely to experience psychological distress, and the higher the degree of restriction in ADL, the higher the risk of psychological distress. Perennial ryegrass (Lolium perenne L.) is a key forage species in warm temperate to subtropical regions worldwide. Climate change poses significant challenges to agriculture, particularly through drought and heat stress, which adversely affect ryegrass yield and may be further exacerbated by global warming. Despite numerous research achievements in recent years, a comprehensive bibliometric analysis of the literature on drought and heat stress in perennial ryegrass is lacking. This study provides a quantitative analysis of relevant literature published from 1994 to 2024, utilizing the Web of Science database to evaluate global research trends and priorities. The results indicate a consistent annual growth in publication output, with China and the United States being major contributors, and the journal Crop Science publishing the most papers. Keyword analysis shows that “growth,” “endophytic fungi,” and “yield” are most frequently used in drought stress research, while “growth,” “gene,” and “leaf” are common in heat stress research. Over the past 30 years, research has mainly focused on phenotype, response mechanisms, and drought and heat resistance techniques in perennial ryegrass. Endophyte have become a hot topic in drought stress research in recent years and have also gained attention in heat stress research, suggesting future research directions in this area. Furthermore, there is a need to strengthen research on the molecular mechanisms associated with drought stress in ryegrass, as well as to explore molecular responses to heat stress. The research trend shows increasing attention to the interaction between drought and heat stress, indicating it will become an important direction for future studies. The findings of this study offer valuable insights for guiding future research on perennial ryegrass under drought and heat stress conditions and provide useful information for researchers in related fields.

Water scarcity in arid and semiarid areas highlights the importance of developing water-saving irrigation techniques. Soil extracellular enzyme activities (EEAs) and microbes are important for soil biochemical cycles and plant growth. However, little is understood about how soil EEAs, and microbial communities respond to water-saving irrigation systems and levels. Therefore, a field experiment was carried out over four consecutive wheat-maize seasons to explore the effects of surface irrigation (SI) and drip irrigation (DI) coupled with different irrigation levels (i.e. 65, 50, 35 and 20 mm/event, represents sufficient, mild deficit, moderate deficit and serious deficit irrigation, respectively) on the plant biomass, soil properties, soil EEAs, and bacterial community. The results revealed that mild deficit irrigation showed no significant impact on plant biomass, serious and moderate deficit irrigation significantly decreased crop biomass by 49.0 % and 26.6 %, respectively, compared with sufficient irrigation. However, the crop biomass of serious and moderate deficit irrigation under DI were 15.4 % and 17.4 % higher, respectively, than that under SI. Serious and moderate deficit irrigation significantly increased soil available K accumulation and soil urease activity, decreased bacterial richness, and shifted the bacterial community structure. No significant differences were found between mild deficit and sufficient irrigation. The relative abundances of Firmicutes, Gemmatimonadota, and Myxococcota significantly increased under serious deficit irrigation, while Acidobacteriota significantly decreased. Moderate deficit irrigation under DI did not shift bacterial community structure, while under SI did. Soil pH, electrical conductivity (EC), available K, and plant biomass were the major factors influencing the bacterial community compositions. Serious deficit irrigation reduced the complexity and stability of the soil bacterial co-occurrence network. The network of DI had more significant interactions among bacteria than the SI. Therefore, DI coupled with moderate deficit irrigation is more beneficial to maintain crop yield and the stability of bacterial community structure than SI.

Silica nanoparticles (SiO2-NPs) have been demonstrated to alleviate the adverse impacts of salt or low temperature on crop growth, especially for individual stress. The aim of this study was to elucidate the regulatory effect of SiO2-NPs on plant performance under combined salt and low-temperature stress. Therefore, a phytotron experiment was performed to explore the effects of SiO2-NPs application (0, 50, 100, 200 mg L−1) on the plant growth, ionic content, antioxidant activities, photosynthetic parameters, and osmoregulator concentrations of cotton seedlings subjected to the combined stress of salinity (50, 100, and 150 mmol L−1 NaCl) and low temperature (day and night temperatures of 15 and 10 °C). The results indicated that the combinatorial stress strongly decreased the plant height and leaf area of cotton seedlings, and obviously suppressed the aboveground biomass by 10.26 %, 11.42 %, and 15.70 % with the increase in salinity. While SiO2-NPs application significantly increased the plant growth, photosynthetic rate, transpiration rate, stomatal conductance, superoxide dismutase, catalase and glutathione reductase activities, leaf water potential, K+, and proline contents, and reduced the Na+ content and Na+/K+ ratio of cotton seedlings under the combinatorial stress. However, the effects of SiO2-NPs on reduced glutathione, total soluble sugar and protein content, and peroxidase activity did not exhibit a clear pattern. The aboveground biomass of cotton seedlings subjected to the combinatorial stress was closely correlated with the Na+/K+ ratio, Na+ content, K+ content, proline content, SOD activity, and CAT activity, indicating that SiO2-NPs could alleviate the suppression of combinatorial stress on cotton seedling growth by decreasing the Na+/K+ ratio and increasing the antioxidant capacity.

In this paper, cellulose nanofibers (CNF) were prepared from wheat bran (WB) and the structure of CNF was determined. Fourier transform infrared spectra and X-ray diffractograms showed the groups such as hydroxyl and carboxyl groups and cellulose type 1 structure possessed by CNF, respectively. Scanning electron microscopy exhibited that CNF was filamentous and intertwined. In addition, Pickering emulsions were prepared using CNF and the physicochemical properties of the emulsions were characterized. The results showed that CNF was able to increase the zeta potential and viscosity of the emulsions, thus improving the stability of the emulsions. Moreover, CNF formed a physical barrier by adsorbing at the oil-water interface and near the oil droplets and CNF dispersed in the aqueous phase formed a network structure to restrict the movement of oil droplets, thus improving the stability of the emulsions. These findings may provide some new insights for the potential applications of WB.

Introduction Involved site radiation therapy (ISRT) is a widely used treatment for primary ocular adnexal mucosa-associated lymphoid tissue (MALT) lymphoma with control rates over 85%; however, its complications could reach 30%–50%. Intralesional rituximab injection has shown promising results with remission rates between 65% and 100% and minimal complications. This study aimed to conduct a multicentre randomised controlled clinical trial to compare the safety and efficacy of intralesional rituximab injection versus ISRT on primary ocular adnexal MALT lymphoma.Methods and analysis This is a multicentre randomised controlled trial and will be conducted at eight ophthalmic departments and institutes in China. 108 patients with primary ocular adnexal MALT lymphoma will be enrolled and randomised to undergo intralesional rituximab injection or ISRT. Comprehensive examinations will be performed before and after the treatment. The primary outcome is the cumulative occurrence rate of complications of grade ≥2 within 5 years after treatment commencement. The main secondary outcome is overall response rate, and progression-free survival, time to next treatment and overall survival will also be compared between the groups.Ethics and dissemination Ethical approval has been obtained from the Ethics Committee of the Zhongshan Ophthalmic Center (ID: 2023KYPJ125-3; V.20240818) and the ‘5010 Plan’ evaluation committee at Sun Yat-sen University, Guangzhou, China, and all subcentres. All the participants will be required to provide written informed consent. The results will be disseminated through scientific meetings and published in peer-reviewed journals.Trial registration number NCT06190301.

In the power industry, the proper functioning of insulators is essential for maintaining the stability of the power system. Currently, unmanned aerial vehicle inspection technology is primarily employed to identify defects in insulators. However, this method has limitations, including the potential to overlook internal defects due to camera blind spots. To address this issue, this paper proposes a method for recognizing insulator defects using acoustic signals based on a CNN-GRU network. First, this paper analyzes the causes and acoustic mechanisms of three typical insulator defects and collects their acoustic signal data. Time-frequency domain analysis is conducted to verify the feasibility of using acoustic signals for defect recognition. To more accurately characterize the acoustic signals of insulators and address the limitations of single feature parameters in information representation, this study combines traditional Mel-frequency cepstral coefficients (MFCCs) and inverse Mel-frequency cepstral coefficients (IMFCCs) to create new C-MFCC feature parameters, thereby enhancing the feature extraction algorithm, and principal component analysis is used for dimensionality reduction to prevent redundancy. Additionally, to address the limitations of traditional convolutional neural networks (CNNs), which can only extract spatial features, this paper proposes combining CNN with gated recurrent units (GRUs) to form a CNN-GRU network. This hybrid network capitalizes on the advantages of both models, enabling the extraction of both spatial and temporal features, thereby further enhancing the network’s recognition rate. Finally, the proposed method is compared with other similar methods and evaluated using field-collected data to validate its superiority and feasibility.

Phosphorus is a key indicator for water quality management due to its role in eutrophication. The variety of phosphorus-containing substances necessitates highly sensitive detection of total phosphorus, particularly through automated methods, to ensure water safety. This study involved the independent development of a sensor featuring an automated in situ detection technique. Utilizing potassium persulfate high-temperature oxidation and phosphorus molybdenum blue spectrophotometry, total phosphorus was monitored in situ via sequential injection technology. Additionally, the detection process and reaction conditions of the sensor were optimized, and a temperature compensation algorithm and turbidity correction were applied to mitigate environmental factors. Under optimal conditions, the sensor demonstrated a detection limit of 1.9 µg/L with a range of 6.5–1000 µg/L in seawater, and 1.2 µg/L with a range of 4.1–2000 µg/L in freshwater. The digestion efficiency for five representative phosphorus-containing substances was found to range from 87.3% ± 1.7% to 103.1% ± 0.6%. Notably, the sensor was deployed for in situ operation at a marine experimental station and online at a river monitoring station. With its integration, low power consumption, and high precision, the sensor enabled long-term unattended monitoring, delivering accurate, stable, and reliable results.
