Four previously undescribed highly oxygenated diterpenoids (1–4), zeylleucapenoids A–D, characterized by halimane and labdane skeletons, were isolated from the aerial parts of Leucas zeylanica. Their structures were elucidated primarily by NMR experiments. The absolute configuration of 1 was established by theoretical ECD calculations and X-ray crystallographic analysis, whereas those for 2−4 were assigned by theoretical ORD calculations. Zeylleucapenoid D (4), with an IC50 value of 38.45 μM in RAW264.7 macrophages and nontoxic activity for zebrafish embryo, and obviously inhibited pro-inflammatory cytokines TNF-α and IL-6 in a dose-dependent manner. Compound 4 inhibited of the expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2). Furthermore, molecular docking analysis indicated the possible mechanism of action for 4 may be bind to targets by hydrogen- and hydrophobic- bond interactions.

The phytochemical investigation of the stems of Homalium stenophyllum afforded seven new phenolic glycosides (1−5 and 8−9) and two known compounds (6 and 7). Their structures were elucidated by comprehensive analyses of NMR spectroscopic, mass spectrometric data and chemical hydrolysis. Additionally, their anti-inflammatory activities against the NO production in LPS-induced macrophages were evaluated.

A new polyketide, 9(E)-3-(5,8-dihydroxynon-3-en-1-yl)-5,6-dihydroxy-2,6-dimethylcyclohex-2-enone (1), and one known compound, 5-chloroisorotiorin (2), were isolated from the mangrove-derived fungus Phomopsis sp. TJM1-5. Their structures were determined by extensive NMR and MS analyses, while the absolute configurations of C-5 and C-6 were established based on experimental and calculated electronic circular dichroism spectra. Compound 2 was isolated from the genus of Phomopsis for the first time. Compounds 1 and 2 were evaluated for their antibacterial activity against five pathogenic bacteria.

Four new flavone derivatives (1–4), together with six known flavones (5-10), were isolated from the stems of Dracaena angustifolia. The complete structures of new compounds were elucidated by analyses of spectroscopic data and quantum chemistry calculations. Compounds 2, 9 and 10 displayed weak anti-inflammatory activity with IC50 values of 45, 33 and 61 μM, respectively.

A preliminary phytochemical investigation of the stems of the endangered plant Ulmus elongata led to the isolation of a new coumarin derivative (named ulmuselactone A, 1) and eight known compounds (2-9). The new structure was elucidated by detailed analysis of comprehensive spectroscopic methods, and its absolute configuration was established by comparing experimental and calculated electronic circular dichroism (ECD) spectra. The isolated compounds were evaluated for their antibacterial activities.

Five new tetralones, daldiniones A-E (1-5), three new chromones, 7-hydroxy-5-methoxy-2,3-dimethylchromone (9), 5-methoxy-2-propylchromone (10), and 7-ethyl-8-hydroxy-6-methoxy-2,3-dimethylchromone (11), and two new lactones, helicascolides D and E (16 and 17), together with nine known metabolites (6-8, 12-15, and 18-19) were isolated from the mangrove-derived fungus Daldinia eschscholtzii HJ004. The structures and absolute configurations of the new compounds were determined by analyzing MS and NMR data and utilizing GIAO based 13C NMR chemical shift calculations and quantum chemical electronic circular dichroism (ECD) calculations. Compounds 9, 13, and 18 showed inhibitory activities against α-glucosidase with IC50 values of 13, 15, and 16 μM, respectively.

A new isopentylated dibenzodioxocinone, canescenin A (1), and a new isopentylated pyran-3,5-dione derivative, canescenin B (2), were isolated from an extract of the deep-sea-derived fungus Penicillium canescens SCSIO z053. Their structures were elucidated by spectroscopic analysis. It was rare to obtain pyran-3,5-dione derivatives from nature. Antibacterial, cytotoxic, and antiviral activities of 1 and 2 were also evaluated.

A novel hydroanthraquinone possessing an unprecedented hexacyclic spiro-fused ring system, anthrininone A (1), and two new anthraquinones with a 4,5-disubstituted butylaminolate unit, anthrininones B and C (2 and 3), together with six known analogues including 6-O-methylalaternin (4) and so on, were obtained from the deep-sea derived fungus Alternaria tenuissima DFFSCS013. Their structures were determined by spectroscopic methods, and the absolute configurations were further determined by single crystal X-ray diffraction analysis for 1 and quantum chemical calculations for 2 and 3, respectively. Compounds 1–4 had significant inhibition activity against indoleamine 2,3-dioxygenase 1 (IDO1), and 2–4 also exhibited selective inhibition activity against different protein tyrosine phosphatases. In addition, 1 could obviously stimulate intracellular calcium levels at a concentration of 10 μM. Their structure–bioactivity relationship was also discussed.

Eight undescribed 3,4-seco-norlabdane diterpenoids, callnudoids A-H, as well as two known analogues were isolated from the leaves of Callicarpa nudiflora. The structures were elucidated using spectroscopic methods and were compared with published NMR spectroscopic data. The absolute configurations of callnudoids D and E were defined based on ECD data or single-crystal X-ray diffraction. Callnudoids A-C are the highly modified labdane diterpenoids featuring rearranged 3,4-seco-ring and the formation of an undescribed cyclohexene moiety via C2-C18 cyclization. They only contain 15 carbon atoms on the carbon skeleton. Callnudoid D represents the unusual 3,4-seco-15,16-norlabdane diterpenoid with C13-C17 cyclization, and a putative biosynthesis pathway for callnudoids A, B, D, and E was proposed. All compounds were evaluated for their anti-inflammatory activities by inhibiting the lipopolysaccharide (LPS)-induced nitric oxide (NO) released in RAW264.7 cells; callnudoids A-E and H, and methylcallicarpate obviously inhibited pro-inflammatory cytokines TNF-α and IL-1β in a dose-dependent manner.

Two new 2,5-diketopiperazines derivatives (

Four unsaturated fatty acid derivatives including three new pantheric acids (1–3), together with three known polyketides (5–7), were isolated from a culture broth of the marine-derived fungus Aspergillus sp. SCAU150. Their complete structures were determined by NMR and HRESIMS data analyses. The antifungal activity of the isolated compounds above was evaluated and 2 was found to show moderated activity toward the phytopathogenic fungus Fusarium solani bio-80814 with an inhibition zone diameter of 6 mm under 5 µg/disc.

Two new 2,5-diketopiperazines derivatives (1-2), together with eight known analogs (3-10), were isolated from a culture broth of an endophytic fungus Nigrospora camelliae-sinensis S30, derived from mangrove Lumnitzera littorea. Their complete structures were determined by a detailed analysis of spectroscopic data and ECD calculations. The antimicrobial activity and neuroprotective activity of these isolated compounds were also evaluated.

A new phenol derivative, 3-chloro-5-hydroxy-4-methoxyphenylacetic acid methyl ester (1), along with five known compounds methyl 4-hydroxyphenylacetate (2), cytosporone B (3), (R)-striatisporolide A (4), (R)-butanedioic acid (5) and ergosterol (6) were isolated from the mangrove-derived fungus Eupenicillium sp. HJ002. Their structures were established by spectroscopic methods, GIAO based 13C NMR chemical shift calculations and comparison with the data of literature. Compounds 1–5 were isolated from Xylocarpus granatum Koening-derived fungus for the first time.

Chemical investigations of the flowers of Praxelis clematidea resulted in the isolation of 12 known compounds (1���12). Their structures were elucidated by extensive spectroscopic studies and chemical evidence. The structure and absolute configuration of 1 was precisely determined by a combination of 2 D NMR and quantum chemical calculations of ECD spectra analysis. Compounds 1, 4, 11 were isolated from the genus Praxelis for the first time, while 1���6, 8���12 were isolated from P. clematidea for the first time. The antibacterial activity of compounds 1, 2, 7, 9 were evaluated.

Eight new tetramic acid derivatives, cladosporiumins A–H (1–8) were isolated from a culture broth of Cladosporium sp. SCSIO z0025 derived from the deep-sea sediment collected from Okinawa Trough. Their structures were elucidated by extensive spectroscopic data analysis. Compounds 1–3 were unique 3-acyltetramic acids with a hexyl enic alcohol side chain and a six-membered lactone ring substituted at C-3 of the 2, 4-pyrrolidinedione skeleton. The absolute configurations of chiral carbons of the 2, 4-pyrrolidinedione unit in 1–4 and 7 were established by quantum chemical calculations of the electronic circular dichroism (ECD) spectra. And the absolute configurations of chiral carbons of the six-membered lactone ring in 1–3 were established by density functional theory calculation of their 13C NMR chemical shifts. In addition, all the compounds were tested for cytotoxic, antibacterial, anti-biofilm, and AchE inhibitory activities.

Four new alkaloids, nonialkaloids A–D (1–4) and six known analogues (5–10) were isolated from the noni juice. Among the new compounds, 1 and 2 are indole alkaloids with a seven-membered fused N-heterocyclic ring, 3 and 4 are quaternary ammonium derivatives. The structures were elucidated by extensive NMR and MS analysis, while the absolute configurations of the stereogenic carbons were established based on quantum-chemical electronic circular dichroism calculations or the modified Mosher's method. All the isolates were tested for α-glucosidase inhibitory activities. Compounds 1 and 3 displayed potent inhibitory activity against α-glucosidase with the IC50 values of 413.7 and 364.4 μM, respectively.

Biofilm formation of Staphylococcus aureus is one of its mechanisms of drug resistance. Antibiofilm screening of 106 compounds from marine-derived fungi displayed that 12 compounds inhibited S. aureus biofilm formation by >50% at the concentration of 100 μg/ml, and only secalonic acid D (SAD) and B inhibited by >90% at 6.25 μg/ml without inhibiting cell growth after 24-h incubation. Meanwhile, it was found that the double bond between C-1 and C-10 of citrinin derivatives and the C-C connection position of two chromone monomers may be important for their anti-biofilm activities. Moreover, SAD slightly facilitated biofilm eradication and influenced its architecture. Furthermore, SAD slowed the cell growth rate in the preceding 18-h incubation and differentially regulated transcriptional expression of several genes, such as agr, isaA, icaA, and icaD, associated with biofilm formation in planktonic and biofilm cells, which may be the reason for the anti-biofilm activity of SAD. Finally, SAD acted synergistically against S. aureus growth and biofilm formation with other antibiotics. These findings indicated that various natural products from marine-derived fungi, such as SAD, could be used as a potential biofilm inhibitor against S. aureus.

A new linear peptide simplicilliumtide I (1) and four new cyclic peptides simplicilliumtides J–M (2–5) together with known analogues verlamelins A and B (6 and 7) were isolated from the deep-sea-derived fungal strain Simplicillium obclavatum EIODSF 020. Their structures were elucidated by spectroscopic analysis, and their absolute configurations were further confirmed by chemical structural modification, Marfey’s and Mosher’s methods. Compounds 2, 6, and 7 showed significant antifungal activity toward Aspergillus versicolor and Curvularia australiensis and also had obvious antiviral activity toward HSV-1 with IC50 values of 14.0, 16.7, and 15.6 μM, respectively. A new cyclic pentapeptide and three new linear peptides, namely, aspergillipeptides D–G (1–4), were isolated from a culture broth of marine gorgonian-derived fungus Aspergillus sp. SCSIO 41501. Their structures were elucidated by spectroscopic analysis, and their absolute configurations were confirmed by Marfey’s method. Compounds 1 and 2 showed evident antiviral activity against herpes simplex virus type 1 (HSV-1) with IC50 values of 9.5 and 19.8 µM under their non-cytotoxic concentrations against a Vero cell line, respectively, and 1 also had antiviral activity against acyclovir-resistant clinical isolates of HSV-1.

Three new lactones penicilactones A&minus, C (1&minus, 3) were obtained from the mangrove-derived fungus Penicillium sp. TGM112. Their structures and absolute configurations were determined by detailed NMR, MS spectroscopic data, Mo2(OAc)4-induced electronic circular dichroism (ECD), and circular dichroism (CD) spectroscopy. Compound 1 showed antibacterial activity against Staphylococcus aureus with an MIC value of 6.25 &mu, g/mL. Compound 2 showed insecticidal activity against newly hatched larvae of Culex quinquefasciatus with the LC50 value of 78.5 (&plusmn, 0.58) &mu, g/mL.

A new isopentylated dibenzodioxocinone, canescenin A (

The present study investigated the fungal diversity in four different deep-sea sediments from Okinawa Trough using high-throughput Illumina sequencing of the nuclear ribosomal internal transcribed spacer-1 (ITS1). A total of 40,297 fungal ITS1 sequences clustered into 420 operational taxonomic units (OTUs) with 97% sequence similarity and 170 taxa were recovered from these sediments. Most ITS1 sequences (78%) belonged to the phylum Ascomycota, followed by Basidiomycota (17.3%), Zygomycota (1.5%) and Chytridiomycota (0.8%), and a small proportion (2.4%) belonged to unassigned fungal phyla. Compared with previous studies on fungal diversity of sediments from deep-sea environments by culture-dependent approach and clone library analysis, the present result suggested that Illumina sequencing had been dramatically accelerating the discovery of fungal community of deep-sea sediments. Furthermore, our results revealed that Sordariomycetes was the most diverse and abundant fungal class in this study, challenging the traditional view that the diversity of Sordariomycetes phylotypes was low in the deep-sea environments. In addition, more than 12 taxa accounted for 21.5% sequences were found to be rarely reported as deep-sea fungi, suggesting the deep-sea sediments from Okinawa Trough harbored a plethora of different fungal communities compared with other deep-sea environments. To our knowledge, this study is the first exploration of the fungal diversity in deep-sea sediments from Okinawa Trough using high-throughput Illumina sequencing.

Oxalicumone A (POA1), a novel dihydrothiophene-condensed sulfur chromone isolated from the marine fungus Penicillium oxalicum SCSGAF 0023, showed cytotoxicity against several cancer cells previously. In this study, its anti-cancer activity and underlying mechanism of this action were investigated in leukemia cells like KG-1a, HL60, U937, and K562. The results showed that POA1 inhibited dose-/time-dependently cell growth and induced apoptosis in leukemia cells. Also, POA1 caused cleavages of caspase-3, 8, 9 and PARP1, loss of mitochondrial membrane potential, up-regulations of phosphorylated p38 and JNK, and activation of endoplasmic reticulum stress (ER stress). Furthermore, 4-PBA (an ER stress inhibitor) but not SP600125 and SB203580 (JNK and p38 inhibitor, respectively) could largely inhibit POA1-induced growth suppression. Additionally, 4-PBA obstructed mitochondrial depolarization and cleavage of PARP1. These data suggested that ER stress pathway might be an important mediator in POA1-induced apoptosis. In conclusion, POA1 may have antitumor effects in leukemia cells through the induction of ER stress pathway.

A new symmetric 16-membered macrodiolide, trichobotryside A (1), and two new asymmetric 18-membered macrodiolides trichobotrysides B–C (2–3) were obtained from the deep-sea-derived fungus Trichobotrys effuse DFFSCS021. Their structures were elucidated by extensive spectroscopic analysis. The absolute configurations of 1 and 2 were established by the methanolysis and modified Mosher method. And the methanolysis of both 1 and 2 produced the new compound of 4R,7S,9S-trihydroxyl-2-en-1-methyl decanoate (4). It was rare to obtain an 18-membered macrodiolide from nature. Compound 1 showed strong antifouling activity against Bugula neritina and Balanus amphitrite larvae settlement.

Fourteen ansamycin derivatives including seven new herbimycins G–L (1–6) and divergolide O (7), and seven known analogues were isolated from a culture broth of the marine-derived Streptomyces sp. SCSGAA 0027. Their complete structures were determined by detailed analysis of spectroscopic data and quantum chemical calculations. Compounds 1–5 and 7 featured an additional eight-membered O-heterocycle that has rarely been reported for ansamycins, and the Z,Z- and E,E-configurations for Δ2,Δ4 were reported for the first time in geldanamycin analogues. Compound 1 exhibited weak inhibition activity towards Hsp90α with an IC50 value of 96 µM, 2–5 showed mild cytotoxicity against four human cancer cell lines with IC50 values ranging from 13 μM to 86 μM, and 7 had moderate anti-HSV-1 activity with an IC50 value of 19 µM and very weak cytotoxicity towards Vero cell. The possible biosynthetic pathways for 1–5 were proposed. And their structure-bioactivity relationship was also discussed.

Trichobotrysins A–F (1–6), a class of new tetramic acid derivatives with a decalin ring, were characterized from the culture of Trichobotrys effuse DFFSCS021 derived from the deep sea sediment collected from the South China Sea. Their structures were elucidated by extensive spectroscopic analysis. Compounds 1, 2, and 4 exhibited significant selective cytotoxicity against human carcinoma KG-1a cell line with IC50 values of 5.44, 8.97, and 6.16 μM, and obvious antiviral activity towards HSV-1 with IC50 values of 3.08, 9.37, and 3.12 μM, respectively. This is the first time to report the anti-HSV-1 activity of this type of compounds.

Cyclopiamides B–J (1–9), nine new oxindole alkaloids were isolated from the deep-sea-derived fungus Penicllium commune DFFSCS026. Their structures were elucidated by extensive spectroscopic data analysis. Compound 4 contains a rare 2H-pyrrole[2,1-b]pyridine unit, and compounds 8 and 9 are complex polycyclic and polyhydroxy alkaloids. The absolute configurations of 6 and 8 were established by molecular mechanics calculation, quantum chemical computations, and single crystal X-ray diffraction analysis, respectively.

Several ansamycins have been reported to inhibit bacterial biofilm formation and accelerate the eradication of developed biofilms, but little is known about the effect of hygrocin C, an ansamycin, on bacterial biofilm formation. Here, hygrocin C was isolated from the marine-derived Streptomyces sp. SCSGAA 0027 and reported for the first time to be capable of inhibiting the biofilm formation of Staphylococcus aureus and Bacillus amyloliquefaciens SCSGAB0082 with the production of anti-microbial lipopeptides from South China Sea gorgonian Subergorgia suberosa at concentrations of less than minimum inhibitory concentrations. Moreover, hygrocin C also promoted the eradication of developed biofilms, affected the biofilm architecture, and lowered the extracellular polymeric matrix formation, cell motility, and surface hydrophobicity in B. amyloliquefaciens, which was in accordance with the inhibition of biofilm formation. Furthermore, transcriptome analysis revealed that hygrocin C altered the transcripts of several genes associated with bacterial chemotaxis and flagellar, two-component system and the synthesis of arginine and histidine, which are important for bacterial biofilm formation. In conclusion, hygrocin C could be used as a potential biofilm inhibitor against S. aureus and B. amyloliquefaciens. But further genetic investigations are needed to provide more details for elucidation of the molecular mechanisms responsible for the effects of hygrocin C on B. amyloliquefaciens biofilm formation.

Five new pteridic acids C-G (1-5) were isolated from a culture broth of the marine-derived actinomycete Streptomyces sp. SCSGAA 0027. Their complete structures were elucidated on the basis of NMR data, modified Mosher's method and quantum chemical calculations. Furthermore, their cytotoxicity, antiviral and antimicrobial activities were also evaluated.

Biofilm formation of

Four new compounds (1–4), including two brevianamides and two mycochromenic acid derivatives along with six known compounds were isolated from the deep-sea-derived fungus Penicillium brevicompactum DFFSCS025. Their structures were elucidated by spectroscopic analysis. Moreover, the absolute configurations of 1 and 2 were determined by quantum chemical calculations of the electronic circular dichroism (ECD) spectra. Compound 9 showed moderate cytotoxicity against human colon cancer HCT116 cell line with IC50 value of 15.6 μM. In addition, 3 and 5 had significant antifouling activity against Bugula neritina larval settlement with EC50 values of 13.7 and 22.6 μM, respectively. The NMR data of 6, 8, and 9 were assigned for the first time.

Seventeen lactones including eight territrem derivatives (1–8) and nine butyrolactone derivatives (9–17) were isolated from a marine-derived fungus Aspergillus terreus SCSGAF0162 under solid-state fermentation of rice. Compounds 1–3 and 9–10 were new, and their structures were elucidated by spectroscopic analysis. The acetylcholinesterase inhibitory activity and antiviral activity of compounds 1–17 were evaluated. Among them, compounds 1 and 2 showed strong inhibitory activity against acetylcholinesterase with IC50 values of 4.2 ± 0.6, 4.5 ± 0.6 nM, respectively. This is the first time it has been reported that 3, 6, 10, 12 had evident antiviral activity towards HSV-1 with IC50 values of 16.4 ± 0.6, 6.34 ± 0.4, 21.8 ± 0.8 and 28.9 ± 0.8 μg·mL−1, respectively. Antifouling bioassay tests showed that compounds 1, 11, 12, 15 had potent antifouling activity with EC50 values of 12.9 ± 0.5, 22.1 ± 0.8, 7.4 ± 0.6, 16.1 ± 0.6 μg·mL−1 toward barnacle Balanus amphitrite larvae, respectively.

Eight new chromones, engyodontiumones A–H (1–8), and three new phenol derivatives (9–11) together with eight known polyketides (12–19) were isolated from the deep-sea-derived fungus Engyodontium album DFFSCS021. Their structures were identified by extensive spectroscopic analysis. Compounds 8 and 16 showed significant selective cytotoxicity against human histiocytic lymphoma U937 cell line with IC50 values of 4.9 and 8.8 μM, respectively. In addition, this is the first time to report that 8, 15 and 16 had mild antibacterial activity against Escherichia coli and Bacillus subtilis, and 15 showed potent antilarval activity against barnacle Balanus amphitrite larval settlement.

Marine-derived microbial secondary metabolites are promising potential sources of nontoxic antifouling agents. The search for environmentally friendly and low-toxic antifouling components guided us to investigate the antifouling potentials of eight novel fungal isolates from deep-sea sediments of the South China Sea. Sixteen crude ethyl acetate extracts of the eight fungal isolates showed distinct antibacterial activity against three marine bacteria (Loktanella hongkongensis UST950701–009, Micrococcus luteus UST950701–006 and Pseudoalteromonas piscida UST010620–005), or significant antilarval activity against larval settlement of bryozoan Bugula neritina. Furthermore, the extract of Aspergillus westerdijkiae DFFSCS013 displayed strong antifouling activity in a field trial lasting 4 months. By further bioassay-guided isolation, five antifouling alkaloids including brevianamide F, circumdatin F and L, notoamide C, and 5-chlorosclerotiamide were isolated from the extract of A. westerdijkiae DFFSCS013. This is the first report about the antifouling potentials of metabolites of the deep-sea-derived fungi from the South China Sea, and the first stage towards the development of non- or low-toxic antifouling agents from deep-sea-derived fungi.

A new pyrrolidinone, nigrospine (1), a new indole alkaloid, nigrospin A (2) and two new citrinins, nigrospins B and C (3-4) were isolated from a culture broth of the marine-derived fungal strain Nigrospora oryzae SCSGAF 0111. Their structures were determined by spectroscopic analysis, and the absolute configurations of 1 and 2 were determined by the Mosher ester technique. Compound 1 contains a rare 2,3-dihydro-benzofuran[2,3-c]2-pyrrolidone skeleton.

Four new xanthene derivatives, penicixanthenes A&ndash, D (1&ndash, 4), and one known compound 5 were isolated from a marine mangrove endophytic fungus Penicillium sp. JY246 that was obtained from the stem of Ceriops tagal. Their structures were determined by detailed NMR, MS spectroscopic data, modified Mosher&rsquo, s method, and calculated electronic circular dichroism data. All of the isolated compounds were examined for insecticidal activity. Compounds 2 and 3 showed growth inhibition activity against newly hatched larvae of Helicoverpa armigera Hubner with the IC50 values 100 and 200 &mu, g/mL, respectively, and compounds 1, 3, and 4 showed insecticidal activity against newly hatched larvae of Culex quinquefasciatus with LC50 values of 38.5 (&plusmn, 1.16), 11.6 (&plusmn, 0.58), and 20.5 (&plusmn, 1) &mu, g/mL, respectively. The four xanthene derivatives have the potential to be developed as new biopesticides.

SCSGAF0086. Five of these compounds (1, 2, 8, 9, and 10) were new, and their structures were determined by spectroscopic methods. Compounds 4, 6, 7, and 17 showed enzyme-inhibitory activities towards several tested enzymes, and 6 and 7 showed strong antifouling activity against Bugula neritina larvae settlement. This is the first time that the antifouling and enzyme-inhibitory activities of these compounds has been reported.

Three new cyclic tetrapeptides, aspergillipeptides A-C (1-3), and one new asteltoxin B (4) were isolated from a culture broth of gorgonian-derived fungal strain Aspergillus sp. SCSGAF 0076. Their structures were elucidated by spectroscopic analysis, and the absolute configurations of aspergillipeptides A-C were determined by Marfey's method.
