Abstract Medical image registration is vital for disease diagnosis and treatment with its ability to merge diverse information of images, which may be captured under different times, angles, or modalities. Although several surveys have reviewed the development of medical image registration, they have not systematically summarized the existing medical image registration methods. To this end, a comprehensive review of these methods is provided from traditional and deep-learning-based perspectives, aiming to help audiences quickly understand the development of medical image registration. In particular, we review recent advances in retinal image registration, which has not attracted much attention. In addition, current challenges in retinal image registration are discussed and insights and prospects for future research provided.

Three new species of the genus Belisana Thorell, 1898 are described from karst caves in Guangxi, China: Belisana langping Zhang, Li & Yao, sp. nov. (♂♀), B. lingui Zhang, Li & Yao, sp. nov. (♂♀), and B. tianyang Zhang, Li & Yao, sp. nov. (♂♀). In addition, a list of all Belisana species from Guangxi is also provided.

Abstract Purpose Glycolysis and immune metabolism play important roles in acute myocardial infarction (AMI). Therefore, this study aimed to identify and experimentally validate the glycolysis-related hub genes in AMI as diagnostic biomarkers, and further explore the association between hub genes and immune infiltration. Methods Differentially expressed genes (DEGs) from AMI peripheral blood mononuclear cells (PBMCs) were analyzed using R software. Glycolysis-related DEGs (GRDEGs) were identified and analyzed using the Database for Annotation, Visualization, and Integrated Discovery (DAVID) for functional enrichment. A protein–protein interaction network was constructed using the STRING database and visualized using Cytoscape software. Immune infiltration analysis between patients with AMI and stable coronary artery disease (SCAD) controls was performed using CIBERSORT, and correlation analysis between GRDEGs and immune cell infiltration was performed. We also plotted nomograms and receiver operating characteristic (ROC) curves to assess the predictive accuracy of GRDEGs for AMI occurrence. Finally, key genes were experimentally validated using reverse transcription-quantitative polymerase chain reaction (RT-qPCR) and western blotting using PBMCs. Results A total of 132 GRDEGs and 56 GRDEGs were identified on the first day and 4–6 days after AMI, respectively. Enrichment analysis indicated that these GRDEGs were mainly clustered in the glycolysis/gluconeogenesis and metabolic pathways. Five hub genes (HK2, PFKL, PKM, G6PD, and ALDOA) were selected using the cytoHubba plugin. The link between immune cells and hub genes indicated that HK2, PFKL, PKM, and ALDOA were significantly positively correlated with monocytes and neutrophils, whereas G6PD was significantly positively correlated with neutrophils. The calibration curve, decision curve analysis, and ROC curves indicated that the five hub GRDEGs exhibited high predictive value for AMI. Furthermore, the five hub GRDEGs were validated by RT-qPCR and western blotting. Conclusion We concluded that HK2, PFKL, PKM, G6PD, and ALDOA are hub GRDEGs in AMI and play important roles in AMI progression. This study provides a novel potential immunotherapeutic method for the treatment of AMI.

Abstract Background The detection and management of intracranial aneurysms (IAs) are vital to prevent life-threatening complications like subarachnoid hemorrhage (SAH). Artificial Intelligence (AI) can analyze medical images, like CTA or MRA, spotting nuances possibly overlooked by humans. Early detection facilitates timely interventions and improved outcomes. Moreover, AI algorithms offer quantitative data on aneurysm attributes, aiding in long-term monitoring and assessing rupture risks. Methods We screened four databases (PubMed, Web of Science, IEEE and Scopus) for studies using artificial intelligence algorithms to identify IA. Based on algorithmic methodologies, we categorized them into classification, segmentation, detection and combined, and then their merits and shortcomings are compared. Subsequently, we elucidate potential challenges that contemporary algorithms might encounter within real-world clinical diagnostic contexts. Then we outline prospective research trajectories and underscore key concerns in this evolving field. Results Forty-seven studies of IA recognition based on AI were included based on search and screening criteria. The retrospective results represent that current studies can identify IA in different modal images and predict their risk of rupture and blockage. In clinical diagnosis, AI can effectively improve the diagnostic accuracy of IA and reduce missed detection and false positives. Conclusions The AI algorithm can detect unobtrusive IA more accurately in communicating arteries and cavernous sinus arteries to avoid further expansion. In addition, analyzing aneurysm rupture and blockage before and after surgery can help doctors plan treatment and reduce the uncertainties in the treatment process.

This study aimed to explore the influence of feeding regimens on carcase traits and meat quality of Gangba lambs. Thirty healthy Gangba ram lambs at nine months of age and an average body weight of 14.08 ± 0.79 kg were randomly distributed into three treatment groups (10 replicates per group). The groups are as follows: a grazing (G) group without any supplementation and housing; a semi-grazing (SG) group provided with 200–300 g of concentrates and free access to oat hay off-pasture; and a stall-feeding (SF) group with 400–500 g of concentrates and free access to oat hay. Abstract Background The strong invasiveness and rapid expansion of dengue virus (DENV) pose a great challenge to global public health. However, dengue epidemic patterns and mechanisms at a genetic scale, particularly in term of cross-border transmissions, remain poorly understood. Importation is considered as the primary driver of dengue outbreaks in China, and since 1990 a frequent occurrence of large outbreaks has been triggered by the imported cases and subsequently spread to the western and northern parts of China. Therefore, this study aims to systematically reveal the invasion and diffusion patterns of DENV-1 in Guangdong, China from 1990 to 2019. Methods These analyses were performed on 179 newly assembled genomes from indigenous dengue cases in Guangdong, China and 5152 E gene complete sequences recorded in Chinese mainland. The genetic population structure and epidemic patterns of DENV-1 circulating in Chinese mainland were characterized by phylogenetics, phylogeography, phylodynamics based on DENV-1 E-gene-based globally unified genotyping framework. Results Multiple serotypes of DENV were co-circulating in Chinese mainland, particularly in Guangdong and Yunnan provinces. A total of 189 transmission clusters in 38 clades belonging to 22 subgenotypes of genotype I, IV and V of DENV-1 were identified, with 7 Clades of Concern (COCs) responsible for the large outbreaks since 1990. The epidemic periodicity was inferred from the data to be approximately 3 years. Dengue transmission events mainly occurred from Great Mekong Subregion-China (GMS-China), Southeast Asia (SEA), South Asia Subcontinent (SASC), and Oceania (OCE) to coastal and land border cities respectively in southeastern and southwestern China. Specially, Guangzhou was found to be the most dominant receipting hub, where DENV-1 diffused to other cities within the province and even other parts of the country. Genome phylogeny combined with epidemiological investigation demonstrated a clear local consecutive transmission process of a 5C1 transmission cluster (5C1-CN4) of DENV-1 in Guangzhou from 2013 to 2015, while the two provinces of Guangdong and Yunnan played key roles in ongoing transition of dengue epidemic patterns. In contextualizing within Invasion Biology theories, we have proposed a derived three-stage model encompassing the stages of invasion, colonization, and dissemination, which is supposed to enhance our understanding of dengue spreading patterns. Conclusions This study demonstrates the invasion and diffusion process of DENV-1 in Chinese mainland within a global genotyping framework, characterizing the genetic diversities of viral populations, multiple sources of importation, and periodic dynamics of the epidemic. These findings highlight the potential ongoing transition trends from epidemic to endemic status offering a valuable insight into early warning, prevention and control of rapid spreading of dengue both in China and worldwide. Abstract Background The utilization of live yeast (Saccharomyces cerevisiae, YE) in dairy cows is gaining traction in dairy production as a potential strategy to improve feed efficiency and milk yield. However, the effects of YE on dairy cow performance remain inconsistent across studies, leaving the underlying mechanisms unclear. Hence, the primary aim of this study was to investigate the impact of YE supplementation on lactation performance, ruminal microbiota composition and fermentation patterns, as well as serum antioxidant capacity and immune functions in dairy cows. Results Supplementation with YE (20 g/d/head) resulted in enhancements in dairy cow’s dry matter intake (DMI) (P = 0.016), as well as increased yields of milk (P = 0.002) and its components, including solids (P = 0.003), fat (P = 0.014), protein (P = 0.002), and lactose (P = 0.001) yields. China exhibits remarkable diversity of the spider genus Belisana Thorell, 1898, with 62 species recorded to date. However, the largest number of Belisana species was found in Yunnan Province (23 ssp.), while only seven species were found in Guizhou Province.In this paper, Belisana wangchengi sp. nov. as a new species is described from Guizhou Province, China.

A single nucleotide variant in mitochondrial DNA (mtDNA) 1555A>G is associated with drug-induced hearing loss. For the 1555A>G mutation site, 1555A wild-type and 1555G mutant-type plasmids were constructed, respectively. In this study, a PCR method based on the TaqMan amplification refractory mutation system was proposed to detect mtDNA 1555A>G. A common upstream primer, a common TaqMan probe, and two downstream allele-specific primers with mismatched bases were designed. One-step amplification and detection of the wild-type and mutant type at the 1555 site were realized for the deafness-related gene through two reactions. Based on this detection method, the minimum detection limit of the wild-type and mutant type detection systems for plasmids was 50 copies/μL. The minimum sensitivity for the detection of nucleic acids in real dried blood spot (DBS) samples was 0.1 ng/μL. In the normal DBS DNA sample, the detection limit of the mutation abundance reached 0.78%. The specificity of the detection method was 100%, and the coefficient of variation was less than 3.36%. This approach was validated using clinical DNA extracted from 113 DBS samples of newborns. Additionally, it showed 100% agreement with bi-directional Sanger sequencing. It can be used as an optional method for the clinical detection of deafness-related genes.

Abstract Eye health has become a global health concern and attracted broad attention. Over the years, researchers have proposed many state‐of‐the‐art convolutional neural networks (CNNs) to assist ophthalmologists in diagnosing ocular diseases efficiently and precisely. However, most existing methods were dedicated to constructing sophisticated CNNs, inevitably ignoring the trade‐off between performance and model complexity. To alleviate this paradox, this paper proposes a lightweight yet efficient network architecture, mixed‐decomposed convolutional network (MDNet), to recognise ocular diseases. In MDNet, we introduce a novel mixed‐decomposed depthwise convolution method, which takes advantage of depthwise convolution and depthwise dilated convolution operations to capture low‐resolution and high‐resolution patterns by using fewer computations and fewer parameters. We conduct extensive experiments on the clinical anterior segment optical coherence tomography (AS‐OCT), LAG, University of California San Diego, and CIFAR‐100 datasets. The results show our MDNet achieves a better trade‐off between the performance and model complexity than efficient CNNs including MobileNets and MixNets. Specifically, our MDNet outperforms MobileNets by 2.5% of accuracy by using 22% fewer parameters and 30% fewer computations on the AS‐OCT dataset.

In order to provide a theoretical basis for the application of organic selenium (Se) in the production and cultivation of Scrophularia ningpoensis Hemsl. We investigated the effects of selenomethionine (SeMet) on the growth and physiological characteristics of S. ningpoensis seedlings. The results showed that SeMet significantly improved the antioxidant capacity by enhancing the activities of antioxidant enzymes such as superoxide dismutase (SOD), peroxidase (POD), catalase (CAT) and ascorbate peroxidase (APX) in the leaves of S. ningpoensis, which significantly reduced the content of malondialdehyde (MDA) and hydrogen peroxide (H2O2), as compared to the control. SeMet also significantly improved the water metabolism by increasing the transpiration rate, stomatal conductance, water use efficiency (WUE), relative water content, and water saturation deficit of S. ningpoensis leaves. Moreover, SeMet significantly enhanced photosynthetic performance by decreasing non-photochemical quenching (NPQ) and increasing the soil and plant analyser development (SPAD) value, net photosynthetic rate, PSII actual photochemical efficiency Y(II), photochemical quenching (qP), PSII photochemical effective quantum yield (Fv'/Fm') and apparent electron transport rate (ETR). Meanwhile, SeMet significantly improved the plant's height, basal diameter, root/shoot ratio and dry weight of shoots and roots in S. ningpoensis. Various SeMet 30 and 60 mg/L SeMet concentrations demonstrated better effects on the growth and physiological characteristics of S. ningpoensis. The above results indicate that appropriate concentrations of SeMet can enhance the growth of S. ningpoensis and can be improved by increasing its antioxidant capacity, water metabolism, and photosynthetic performance. This provides a theoretical foundation for using organic selenium in growing and producing S. ningpoensis.

Due to the deformable surface and unknown biomechanical properties of human skin tissue, it brings a large impact on the contact position and force control of robotic massage. In this paper, an adaptive hybrid impedance control method is proposed. To adapt the massage robot to the environmental changes of muscle tissues in the human massage area, a human muscle tissue model was established, and a recursive least squares algorithm was optimized to identify the model parameters online. To determine the parameters of the impedance controller, a fuzzy system between the parameters of the skin tissue model and the robot contact dynamics model was developed. To keep the robot with proper end-effector orientation and stable contact force during the massage process, an adaptive force-position hybrid control strategy is proposed. On the one hand, a position controller for adaptive attitude adjustment is used to control the move position and the normal direction of the robot. On the other hand, a force controller based on impedance control is used to ensure a stable contact force during dynamic interaction with human. Furthermore, since massage is a human contact task, the adaptive impedance control strategy also improves the interaction safety performance and contact detection. The effectiveness and feasibility of the massage system using adaptive fuzzy impedance control adapted to the variation of human muscle tissue is verified through experiments on the robot massage movement.

In the context of traditional energy shortage and climate warming, the development of solar energy, as a clean and renewable energy, is crucial. As an effective way to utilize solar energy resources, photovoltaic (PV) power generation technology has been widely used around the world. Using remote sensing images to extract PV panel information, including location, area, has a positive effect on understanding the development status, planning and construction of regional PV new energy. In this study, a semantic segmentation network called HCT-Net, combined with the hybrid neural networks and the swarm intelligence optimization algorithms, is designed to segment solar PV panels from remote sensing images automatically and accurately. To address the problem of inconsistent segmentation within PV regions, a hybrid encoder, which combines a convolutional neural network and a Transformer, is designed to extract local features with rich detail information and global features with global context dependencies, resulting in enhanced feature representations. The foreground relation module is designed to solve the problem of mis-segmentation of the background into PVs. This module strengthens the model’s focus on the target object and suppresses the feature representations of non-PVs by explicitly learning the similarity relationship between the global PV feature representation and the feature representations of other objects, and by adaptively assigning weights according to the similarity. The swarm intelligence optimization algorithm is applied to adjust the learning rate and the balance coefficient of the composite loss function of HCT-Net during training. Experimental results show that compared with the current mainstream semantic segmentation network, the method in this study effectively alleviates the problem of inconsistent segmentation within PV regions and mis-segmentation and has advantages in the complete and accurate extraction of PV panels.

Objective: To assess emotional fluctuations, physical and mental health status, and indicators closely related to red blood cells, such as RIO kinase 3 (Riok3), MAX interactor 1 (Mxi1), and microRNA 191 (miR191), in participants with different levels of red blood cells. Methods: Participants who underwent physical examinations at Dongfang Hospital between April and October 2019 were divided into healthy, blood deficiency, and anemia groups (30 individuals in the healthy and blood deficiency group respectively, and 13 in the anemia group). The physical and mental conditions of the participants were evaluated through questionnaires, and emotional fluctuations were assessed through an emotion-inducing experiment, in which participants watched video segments designed to induce specific emotions. Relative expression levels of miR191, Riok3, and Mxi1 from venous blood samples were also determined. Results: The main psychological factors identified in the anemia and blood deficiency groups were obsessive-compulsive symptoms, depression, anxiety, and other negative emotions. Relative gene expression levels indicated that miR191 was upregulated and Riok3 and Mxi1 were downregulated in both the blood deficiency and anemia groups. Regarding the emotional score of disgust on video stage, the main effect was significant (F = 335.58, P < .001), which showed that watching the three videos caused participants to have a dominant emotion, and there is a difference on group (F = 5.35, P = .01), with higher disgust scores in the anemia and blood deficiency groups. The symptoms of blood deficiency and anemia, such as weakness in limbs were significantly negatively correlated with Riok3 and Mxi1 expression (r = −0.38 and −0.31 respectively), but was significantly positively correlated with miR191 expression (r = 0.29). Conclusion: We determined that a close relationship exists between red blood cell levels and emotional status. Our findings suggest that individuals with anemia and blood deficiency are more likely to experience psychological problems and negative emotions, particularly disgust. We also demonstrate that emotional regulation is related to mir191-Riok3-Mxi1 pathway activity, identifying these pathway components are potential targets for genetic therapies in combination with psychological therapy.

At present, the manufacturer collaborates on selling and recycling through an online platform. We explores the mode selection of closed-loop supply chain (CLSC) under the reselling or agency modes in forward SC and under the delegated-recycling mode or agency-recycling mode in reverse SC considering data intelligence level with an online platform. The results show that both the retail price of intelligent connected products (ICPs) and the demand of new ICPs are positively correlated with data intelligence level of new ICPs. The recycling price of old ICPs is negatively relevant to data intelligence level of old ICPs. However, the recycling quantity of old ICPs is positively related to data intelligence level of old ICPs. Increasing data intelligence level of new ICPs is benefit to enterprises. Moreover, it is very unconventional that the demand of new ICPs is increasing when the retail price of new ICPs increases. In addition, the manufacturer can choose the optimal selling mode and recycling mode by setting the selling commission rate and the recycling commission rate.

Abstract Background Molecular diagnostic technology is the foundation of precision medicine, which has the advantages of good specificity, high sensitivity, strong targeting, rapiddiagnosis, etc. It has a wide range of applications in the field of pediatrics. However, molecular diagnostic technology is characterized by complicated experimental operation, high difficulty in data analysis and interpretation, in consistent standards among laboratories, and difficult standardization of technical processes. Enhancing the application value of molecular diagnostic technology in pediatrics and promoting the high‐quality development of the discipline requires careful consideration by relevant management and professionals. Methods This study firstly outlines the development history of molecular diagnostic technology. Then, it analyzes the application of molecular diagnostic technology in the field of pediatrics. Finally, it explores the countermeasures for the management of molecular diagnostic laboratories. Results This study highlights the importance of molecular diagnostic technology in providing information and decision‐making basis for disease prevention, prediction,diagnosis, treatment and regression. It has a wide range of applications in the molecular diagnosis of pediatric hereditary diseases, malignant tumors and infectious diseases. In addition, the countermeasures for the management of molecular diagnostic laboratories are proposed from the five aspects of laboratory, personnel team construction, standardized management,multidisciplinary cross‐discipline, research and translation, safety managementand ethical supervision, and management upgrading and modernization. Conclusions Molecular diagnostic technology, as the basis of precision medicine, has become one of the important frontier fields in the development of contemporary pediatric medicine. Enhanced laboratory capacity in molecular diagnostic techniques can improve outcomes in the prevention, prediction, diagnosis, treatment, prognosis and research of pediatricdiseases, and lay the groundwork for child healthcare.

Soil enzymes play important roles in soil C and nutrient cycling. However, the effects of N addition on soil enzyme kinetics and the underlying mechanisms remain unclear. Thus, we aimed to determine the effects of short-term N addition on the soil properties, microbial properties, maximum reaction rate (Vm, which is attained at saturating substrate concentrations), and Michaelis constant (Km, where a high Km indicates low substrate affinity) of microbial C- (β-1,4-glucosidase and cellobiohydrolase), N- (β-1,4-N-acetylglucosaminidase and L-leucine aminopeptidase), and P-degrading (acid phosphatase and alkaline phosphatase) enzymes in subtropical coniferous (Pinus taiwanensis) and broadleaf (Castanopsis faberi) forests. In the broadleaf forest, N addition increased the Vm and substrate-binding affinities (decline in Km) of C- and P-degrading enzymes by triggering a P deficiency response in microorganisms (i.e., increased microbial biomass N:P ratio). These findings indicate that the soil enzyme kinetics followed the optimal foraging strategy in response to N addition. Moreover, N addition reduced the proportion of complex organic molecules in dissolved organic matter (DOM; e.g., reduced abundance of humic-like fluorophores and humification index), suggesting that N addition increased soil DOM quality and thus increased the affinities of C-degrading enzymes. N addition increased the abundance of Acidobacteria and Chloroflexi but reduced the abundance of Proteobacteria and Rhizobiales, indicating a shift in microbial community toward efficient P acquisition. N addition affected bacterial composition and thus indirectly influenced N- and P-degrading enzymes. In the coniferous forest, N addition significantly increased the Vm of C-degrading enzymes but did not change other enzyme kinetics, which could be partly attributed to the unchanged N availability and microbial properties. Collectively, our findings provide insights into the relationship between enzyme kinetics, DOM quality, and microbial properties, which are important for predicting soil nutrient cycling and parameterizing models of C cycling under N deposition.

Along with the increasing low-carbon demand of the power system, the access of a high percentage of renewable energy resources to the distribution network has a large impact on the voltage fluctuation of the system and reduces the operational reliability. In this paper, we consider utilizing the reactive capacity of distributed resources to participate in system voltage regulation to reduce node loss of load probability (LOLP) caused by node overvoltage faults and propose an overvoltage risk regulation strategy for the interaction between distribution network operators (DSOs) and distributed users in the framework of the Stackelberg game. First, the nodes are clustered and analyzed based on the two-dimensional indexes of node voltage regulation ability, and different voltage regulation compensation tariffs are assigned. Second, the cost-benefit model of voltage regulation for the leader and follower sides and the node LOLP model are constructed to measure the reliability of the system. The Stackelberg game is used to co-optimize the two parties’ compensation tariffs and voltage regulation strategies. The optimal solution of voltage regulation under the equilibrium of the game is obtained by solving using the particle swarm optimization (PSO) algorithm. Based on the IEEE-33 node system, a case study is carried out to verify that the proposed overvoltage risk regulation strategy can maximize the benefits of the regulator participants while enhancing the operational reliability of the system.

ObjectiveTo explore the clinical effect of bladder cancer patients with Fear of Cancer Recurrence (FCR) after applying the gratitude extension construction theory nursing program.Methods168 patients with bladder cancer hospitalized in the Department of Urology from December 2021 to June 2023 in a hospital are study subjects. The experimental subjects are uniformly designed as an experimental group and a control group, with 52 participants in each group. The former receives routine nursing care, while the later receives nursing interventions based on gratitude extension construction theory. The baseline data, Quality of life Questionnaire-core 30, Quality of Life Questionnaire-non Invasive Bladder Cancer 24, Fear of Progression Questionnaire-Short Form, gratitude level questionnaire, Self-Rating Depression Scale, Self-rating Anxiety Scale, patient compliance behavior score, Overall Survival, and Progression-free Survival are evaluated.ResultsThe basic data revealed no statistical significance. The quality of life questionnaire-core 30 and quality of life questionnaire-noninvasive bladder cancer 24 was no significant difference before treatment and after treatment for 1 month. After 9 months, There was a significant difference in pre-treatment scores. The experimental group had no significant difference before and after treatment. For the overall survival rates, the two groups were 67.25% and 79.56%. The progression-free survival rates were 56.35% and 72.35%, respectively, with statistical difference. The compliance rates were 86.54% and 98.08%. The compliance rate of the experimental group exceeded the control group. After 3, 6, and 12 months, the gratitude level questionnaire score and the fear of progression questionnaire-short form in the experimental group were improved. After 3, 6, and 12 months, the control group had no statistically significant difference in the gratitude level questionnaire and the fear of progression questionnaire-short form scores. Compared with the control group, the scores on the gratitude level questionnaire and the fear of progression questionnaire-short form were significantly higher after 3, 6, and 12 months of intervention.ConclusionAfter applying the gratitude extension construction theory nursing program, the FCR of bladder cancer patients is significantly reduced. The quality of life and compliance rate are significantly improved, and anxiety and depression are relieved.

Abstract Background Poly (ADP-ribose) polymerase inhibitors (PARPi) are approved for the treatment of BRCA-mutated breast cancer (BC), including triple-negative BC (TNBC) and ovarian cancer (OvCa). A key challenge is to identify the factors associated with PARPi resistance; although, previous studies suggest that platinum-based agents and PARPi share similar resistance mechanisms. Methods Olaparib-resistant (OlaR) cell lines were analyzed using HTG EdgeSeq miRNA Whole Transcriptomic Analysis (WTA). Functional assays were performed in three BRCA-mutated TNBC cell lines. In-silico analysis were performed using multiple databases including The Cancer Genome Atlas, the Genotype-Tissue Expression, The Cancer Cell Line Encyclopedia, Genomics of Drug Sensitivity in Cancer, and Gene Omnibus Expression. Results High miR-181a levels were identified in OlaR TNBC cell lines (p = 0.001) as well as in tumor tissues from TNBC patients (p = 0.001). We hypothesized that miR-181a downregulates the stimulator of interferon genes (STING) and the downstream proinflammatory cytokines to mediate PARPi resistance. BRCA1 mutated TNBC cell lines with miR-181a-overexpression were more resistant to olaparib and showed downregulation in STING and the downstream genes controlled by STING. Extracellular vesicles derived from PARPi-resistant TNBC cell lines horizontally transferred miR-181a to parental cells which conferred PARPi-resistance and targeted STING. In clinical settings, STING levels were positively correlated with interferon gamma (IFNG) response scores (p = 0.01). In addition, low IFNG response scores were associated with worse response to neoadjuvant treatment including PARPi for high-risk HER2 negative BC patients (p = 0.001). OlaR TNBC cell lines showed resistance to platinum-based drugs. OvCa cell lines resistant to platinum showed resistance to olaparib. Knockout of miR-181a significantly improved olaparib sensitivity in OvCa cell lines (p = 0.001). Conclusion miR-181a is a key factor controlling the STING pathway and driving PARPi and platinum-based drug resistance in TNBC and OvCa. The miR-181a-STING axis can be used as a potential marker for predicting PARPi responses in TNBC and OvCa tumors.

Abstract Background GeneXpert MTB/RIF (Xpert) assay was applied widely to detect Mycobacterium tuberculosis (MTB) and rifampicin resistance. Methods Retrospectively investigated the association among treatment histories, phenotypic drug susceptibility testing (pDST) results, and clinical outcomes of patients infected with probe A absent mutation isolate confirmed by Xpert. Results 63 patients with only probe A absent mutation and 40 with additional pDST results were analyzed. 24 (60.0%) patients had molecular-phenotypic discordant rifampicin (RIF) susceptibility testing results, including 12 (12/13, 92.3%) new tuberculosis (TB) patients and 12 (12/27, 44.4%) retreated ones. 28 (28/39, 71.8%) retreated patients received first-line treatment regime within two years with failed outcomes. New patients had better treatment outcomes than retreated ones (successful: 83.3% VS. 53.8%; P value = 0.02). The clinical results of RIF-susceptible TB confirmed by pDST were not better than RIF-resistant TB (successful: 62.5% VS. 50.0%; P value = 0.43). INH-resistant TB and INH-susceptible TB had similar treatment outcomes too (successful: 61.5% VS. 50.0%; P value = 0.48). 11 (11/12, 91.7%) new patients treated with the short treatment regimen (STR) had successful outcomes. Conclusions More than half of mono probe A absent isolates had RIF molecular-phenotypic discordance results, especially in new patients. Probe A mutations were significantly associated with unsuccessful clinical outcomes, whether the pDST results were RIF susceptible or not. STR was the best choice for new patients. Trial registration retrospectively registered in Wuhan Jinyintan Hospital (No. 2021-KY-16).

To supply more insights into the roles of putrescine (Put) in alleviating salt stress in wheat crops, we explored the effects of Put on ascorbate (ASC) and glutathione metabolism, photosynthetic performance, and ion homeostasis in leaves of salt-stressed wheat seedlings. Our results displayed that salt stress increased the activities of enzymes in ASC and glutathione metabolism, including ascorbate peroxidase, dehydroascorbate reductase, monodehydroascorbate reductase, glutathione reductase, gamma-glutamylcysteine synthetase, and l-galactono-1,4-lactone dehydrogenase, which increased reduced ascorbate (AsA), dehydroascorbate (DHA), reduced glutathione (GSH), oxidised glutathione (GSSG), total ASC and total glutathione contents. Whereas, salt stress induced higher increases in the contents of DHA and GSSG than those of AsA and GSH, which led to reduced AsA/DHA and GSH/GSSG. Meanwhile, salt stress reduced photosynthetic rate (Pn), maximum photochemical efficiency of PSII (Fv/Fm), and the contents of chlorophyll and carotenoids, and destroyed Na+/K+ homeostasis, which further inhibited plant growth. In comparison with salt stress alone, Put strengthened the activities of the above enzymes, which further increased the above metabolites contents, as well as AsA/DHA and GSH/GSSG in leaves of salt-treated seedlings. In this way, Put reduced malondialdehyde content and electrolyte leakage. Besides, Put also increased Pn, Fv/Fm, and above pigments contents, and maintained Na+/K+ homeostasis. Meanwhile, Put increased plant height and biomass of salt-treated seedlings. The present findings clearly implied that Put enhanced salt tolerance of wheat crops by strengthening ASC and glutathione metabolism, photosynthetic performance, and maintaining ion homeostasis in leaves. Therefore, Put can be applied to strengthen the salt tolerance of wheat crops in production and cultivation.

Hydroxyapatite (HAP) is a material renowned for its exceptional capabilities in adsorbing and exchanging heavy metal ions, making it a widely employed substance within the environmental domain. This study aims to present a novel material, namely copper–HAP (Cu–HAP), which was synthesized via an ion exchange method. The resulting material underwent comprehensive characterization using scanning electron microscopy, X-ray diffraction, Fourier-transform infrared spectroscopy, and Brunauer–Emmett–Teller (BET) analysis. Subsequently, based on the principle of the Fenton-like oxidation reaction, the material was used for the degradation of phenol. The outcomes of the investigation revealed that the optimal preparation conditions for the catalyst were achieved at a temperature of 40 °C, a pH value of 5, and a relative dosage of Cu–HAP at 100 mg/g. Under the reaction conditions of a catalyst dosage of 2 g/L, a 30% hydrogen peroxide concentration of 30 mM, a phenol concentration of 20 mg/L, a pH value of 6, a temperature of 40 °C, and the degradation rate of phenol impressively reached 98.12%. Furthermore, the degradation rate remained at 42.31% even after five consecutive cycles, indicating the promising potential of Cu–HAP in the treatment of recalcitrant organic compounds within this field. HIGHLIGHTS A novel copper-based catalyst (Cu–HAP) has been synthesized and utilized for the catalytic degradation of phenol.; Cu–HAP improved the efficiency of H2O2 in the traditional Fenton reaction.; Cu–HAP surface has a rich pore structure.; The phenol removal rate can still reach 42.31% after five times of reuse.;

To refine the grain size and improve the mechanical properties of ultrahigh-strength aluminum alloy (Al-10Zn-1.9Mg-1.6Cu-0.12Zr), the Al-Ti-B-Er grain refiner was prepared by the melt reaction method using the aluminum melt and Al + Ti + B precursor. The results exhibit that the Al-Ti-B-Er grain refiner is mainly composed of a block TiAl3 phase, and loose agglomerated nano-sized TiB2 and Al3Er phases. The microstructure of ultrahigh-strength aluminum is significantly affected by the Al-Ti-B-Er refiner, which changes from dendrite to equiaxial grain with increasing Al-Ti-B-Er content, and the size of the eutectic phase is significantly refined. The high-efficiency refinement of Al-Ti-B-Er is due to Er promoting the uniform distribution of TiAl3 particles and the formation of loose agglomerated nano-sized TiB2 particles. The optimal addition content of Al-Ti-B-Er into ultrahigh-strength aluminum alloys is 1 wt%, whose grain size is approximately 40 µm. Additionally, the strength and ductility of ultrahigh-strength aluminum alloys are simultaneously improved by adding 1wt% Al-Ti-B-Er after the T6 treatment, reaching 756 MPa and 20%, respectively. This enhancement in strength and ductility is mainly attributed to grain refinement and the eutectic phase refinement.

Low-grade glioma (LGG), a common primary tumor, mainly originates from astrocytes and oligodendrocytes. Increasing evidence has shown that peroxisomes function in the regulation of tumorigenesis and development of cancer. However, the prognostic value of peroxisome-related genes (PRGs) in LGG has not been reported. Therefore, it is necessary to construct a prognostic risk model for LGG patients based on the expression profiles of peroxisome-related genes. Our study mainly concentrated on developing a peroxisome-related gene signature for overall survival (OS) prediction in LGG patients. First, according to these peroxisome-related genes, all LGG patients from The Cancer Genome Atlas (TCGA) database could be divided into two subtypes. Univariate Cox regression analysis was used to find prognostic peroxisome-related genes in TCGA_LGG dataset, and least absolute shrinkage and selection operator Cox regression analysis was employed to establish a 14-gene signature. The risk score based on the signature was positively associated with unfavorable prognosis. Then, multivariate Cox regression incorporating additional clinical characteristics showed that the 14-gene signature was an independent predictor of LGG. Time-dependent ROC curves revealed good performance of this prognostic signature in LGG patients. The performance about predicting OS of LGG was validated using the GSE107850 dataset derived from the Gene Expression Omnibus (GEO) database. Furethermore, we constructed a nomogram model based on the gene signature and age, which showed a better prognostic power. Gene ontology (GO) and Kyoto Encylopedia of Genes and Genomes (KEGG) analyses showed that neuroactive ligand-receptor interaction and phagosome were enriched and that the immune status was decreased in the high-risk group. Finally, cell counting kit-8 (CCK8) were used to detect cell proliferation of U251 and A172 cells. Inhibition of ATAD1 (ATPase family AAA domain-containing 1) and ACBD5 (Acyl-CoA binding-domain-containing-5) expression led to significant inhibition of U251 and A172 cell proliferation. Flow cytometry detection showed that ATAD1 and ACBD5 could induce apoptosis of U251 and A172 cells. Therefore, through bioinformatics methods and cell experiments, our study developed a new peroxisome-related gene signature that migh t help improve personalized OS prediction in LGG patients.

Background In CheckMate 9LA, nivolumab plus ipilimumab with chemotherapy prolonged overall survival (OS) versus chemotherapy regardless of tumor PD-L1 expression or histology. We report updated efficacy and safety in all randomized patients with a minimum 4-year follow-up and an exploratory treatment-switching adjustment analysis in all treated patients who received chemotherapy and subsequent immunotherapy.Methods Adults with stage IV/recurrent non-small cell lung cancer (NSCLC), no sensitizing EGFR/ALK alterations, and ECOG performance status ≤1 were randomized 1:1 to nivolumab 360 mg every 3 weeks plus ipilimumab 1 mg/kg every 6 weeks with chemotherapy (two cycles) or chemotherapy (four cycles, with optional maintenance pemetrexed for the nonsquamous population). Assessments included OS, progression-free survival, and objective response rate. Exploratory analyses included efficacy by tumor PD-L1 expression and histology and in patients who discontinued nivolumab plus ipilimumab with chemotherapy due to treatment-related adverse events (TRAEs), and a treatment-switching adjustment analysis using inverse probability of censoring weighting.Results With a 47.9-month minimum follow-up for OS, nivolumab plus ipilimumab with chemotherapy continued to prolong OS over chemotherapy in all randomized patients (HR 0.74, 95% CI 0.63 to 0.87; 4-year OS rate: 21% versus 16%), regardless of tumor PD-L1 expression (HR (95% CI): PD-L1

Abstract The rapid reversal of deep neuromuscular blockade (NMB) is important but remains challenging. This study aimed to evaluate the efficacy and safety of adamgammadex versus sugammadex in reversing deep rocuronium‐induced NMB. This multicenter, randomized, phase IIb study included 80 patients aged 18–64 years, American Society of Anesthesiologists (ASA) grade 1–2, undergoing elective surgery under general anesthesia with rocuronium. Patients were randomized to the adamgammadex 7, 8, and 9 mg/kg group or the sugammadex 4 mg/kg group. The primary efficacy variable was the time to recovery of train‐of‐four ratio (TOFr) to 0.9. The secondary efficacy variables were the time to recovery of TOFr to 0.7, antagonistic success rate of the recovery of TOFr to 0.9 within 5 min, and incidence rate of recurarization within 30 min after drug administration. The explorative efficacy variable was the time to recovery of the corrected TOFr to 0.9 (actual/baseline TOF ratio). Adamgammadex 7, 8, and 9 mg/kg and sugammadex 4 mg/kg groups did not significantly differ in all efficacy variables. Importantly, adamgammadex 9 mg/kg permitted reversal within a geometric mean of 2.9 min. According to the safety profile, adamgammadex achieved good tolerance and low incidence of drug‐related adverse events compared with the 4 mg/kg sugammadex. Adamgammadex 7, 8, and 9 mg/kg facilitated rapid reversal of deep rocuronium‐induced NMB and had good tolerance and low incidence of drug‐related adverse events. Therefore, adamgammadex is a potential and promising alternative to sugammadex.

The addition of antibiotics as growth promoters to ruminant feed can result in bacterial resistance and antibiotic residues in ruminant products. Correspondingly, there is serious public concern regarding the presence of antibiotic residue in ruminant products and the consequent threat to human health. As a result, the addition of plants and their products to ruminant feeds, as an alternative to antibiotics, has received much attention recently. Garlic and its products are rich in organosulphur compounds, which have a variety of biological activities and have been widely used as natural additives in animal production. This review presents recent knowledge on the addition of garlic products (powder, skin, oil, leaf and extracts) to the diets of ruminants. In this paper, garlic products are evaluated with respect to their chemical composition, bioactive compounds, and their impacts on the rumen ecosystem, antioxidant status, immune response, parasitic infection, growth performance and product quality of ruminants. This review provides valuable guidance and a theoretical basis for the development of garlic products as green, highly efficient and safe additives, with the aims of promoting ruminant growth and health, reducing methane emissions and improving ruminant product quality. Garlic extracts have the potential to control parasite infections by decreasing the faecal egg count. Garlic powder, oil and allicin are able to reduce the methane emissions of ruminants. Organosulphur compounds such as allicin, which is present in garlic products, have the potential to inhibit membrane lipid synthesis of the archaeal community, thus influencing the population of methanogenic archaea and resulting in a reduction in methane emissions. Some garlic products are also able to increase the average daily gain (garlic skin, water extract, and leaf) and the feed conversion ratio (garlic skin and leaf) of ruminants. Garlic stalk silage fed to sheep has the potential to improve the nutritional value of mutton by increasing the concentrations of linoleic and linolenic acids and essential amino acids. Sheep fed a diet containing garlic powder or oil are able to produce milk with higher concentrations of the conjugated linoleic acids and n-3 fatty acids, which has health benefits for consumers, due to the widely recognized positive impact of n-3 polyunsaturated fatty acids and conjugated linoleic acids on human heart health, improving platelet aggregation, vasodilation and thrombotic tendency. Overall, garlic products have the potential to enhance growth performance and product quality and reduce parasite infections, as well as methane emissions of ruminants.

Abstract Objective Although several mucosal flap techniques have been reported to improve the outcomes in Draf IIb and Draf III procedures, there is scant knowledge on frontal ostium neo‐osteogenesis after reconstruction with mucosa flap. This study evaluates the potential benefits of mucosa flaps on frontal ostium neo‐osteogenesis after frontal sinus drill‐out procedures. Methods Forty‐three patients who underwent extended Draf IIb and Draf III were enrolled. Among them, 20 patients had frontal neo‐ostium (FNO) reconstructed by mucosal flap (group A), and 23 patients did not have neo‐ostium reconstruction (group B). The cross‐sectional area of FNO, frontonasal bone, and the amount of frontal neo‐osteogenesis (FNOG) were measured with OsiriX®. In addition, the Global Osteitis Scoring Scale (GOSS), Lund–Mackay score (LMS), and Lund–Kennedy score (LKS) were also evaluated. Abstract Background Hyperlipidemia, a heterogeneous group of disorders characterized by elevated plasma lipids in the blood, causes severe health problems, leading to fatty liver disease and nonalcoholic fatty liver disease. Thymoquinone, the major active chemical component of Nigella sativa, reportedly exerts a vast array of biological effects. Various studies have reported that Thymoquinone protects against liver injury. Aims The aim of this study was to investigate the possible protective effects of Thymoquinone against liver injury in hyperlipidemia-induced LDL-R−/− mice. Methods Eight-week-old male LDL-R−/− mice were randomly divided into three groups: a control group fed a normal diet and two groups fed a high-cholesterol diet or high-cholesterol diet mixed with Thymoquinone. All groups were fed different diets for 8 weeks. Blood samples were obtained from the inferior vena cava and collected in serum tubes. The samples were then stored at − 80 °C until used. Longitudinal sections of liver tissues were fixed in 10% formalin and then embedded in paraffin for histological evaluation. The remainder of the liver tissues were snap-frozen in liquid nitrogen for reverse transcription-polymerase chain reaction or western blotting. Results Our results demonstrated that Thymoquinone administration significantly reduced liver histological alterations by hyperlipidemia. Thymoquinone mitigated hyperlipidemia-induced liver injury as indicated by the suppression of metabolic characteristics, liver biochemical parameters, pyroptosis indicators, a macrophage marker, and the phosphatidylinositide 3-kinase signaling pathway. Conclusions Thymoquinone is a potential therapeutic agent for hyperlipidemia-induced liver injury.

Abstract Brain-computer interfaces (BCIs) have attracted considerable attention in motor and language rehabilitation. Most devices use cap-based non-invasive, headband-based commercial products or microneedle-based invasive approaches, which are constrained for inconvenience, limited applications, inflammation risks and even irreversible damage to soft tissues. Here, we propose in-ear visual and auditory BCIs based on in-ear bioelectronics, named as SpiralE, which can adaptively expand and spiral along the auditory meatus under electrothermal actuation to ensure conformal contact. Participants achieve offline accuracies of 95% in 9-target steady state visual evoked potential (SSVEP) BCI classification and type target phrases successfully in a calibration-free 40-target online SSVEP speller experiment. Interestingly, in-ear SSVEPs exhibit significant 2nd harmonic tendencies, indicating that in-ear sensing may be complementary for studying harmonic spatial distributions in SSVEP studies. Moreover, natural speech auditory classification accuracy can reach 84% in cocktail party experiments. The SpiralE provides innovative concepts for designing 3D flexible bioelectronics and assists the development of biomedical engineering and neural monitoring.

Soil desertification is an important challenge in global soil management, and effectively and stably restoring soil function is an urgent problem. Using synthetic microbial communities (SynComs) is a burgeoning microbial strategy aimed at enhancing soil nutrients through functional synergies among diverse microorganisms; nevertheless, their effectiveness in restoring desertified soils remains unknown. In this study, we conducted a two-year field experiment using a SynCom constructed by in situ probiotic bacteria and set up control, chemical fertilizer, and combined SynCom–chemical fertilizer (combined fertilizer) treatments to investigate the linkage between microbial communities and soil multifunctionality in the soil surface layer (0–10 cm). Both the bacterial and fungal communities differed the most under the combined fertilizer treatment compared to the control. The bacterial communities differed more under treatments of the SynCom than the chemical fertilizer, while the fungal communities differed more under the chemical fertilizer treatment than the SynCom treatment. Regarding soil function, the SynCom strengthened the correlation between enzyme activities and both bacterial communities and functional properties. pH and available potassium were the main influencing factors under the chemical fertilizer and combined fertilizer treatments. The beta-diversity of the bacterial communities was significantly correlated with soil multifunctionality. Random forest analyses showed that the SynCom significantly enhanced the bacterial communities, driving soil multifunctionality, and that some potential microbial taxa drove multiple nutrient cycles simultaneously. In summary, the SynCom effectively increased the abundance of most carbon, nitrogen, and phosphorus functional genes as well as soil enzyme activities. The bacterial community composition contributed significantly to soil multifunctionality. Hence, the development of novel microbial agents holds significant potential for improving soil functionality and managing desertification.

Abstract Vibrational energy harvesters, which can convert mechanical energy distributed widely in the surrounding environment to electrical energy in a convenient, eco‐friendly and sustainable way, have attracted great attention in both academia and industry. In this study, a resilient electret film‐based vibrational energy harvester with a V‐shaped counter electrode is introduced, simulated and constructed. A negatively charged fluorinated polyethylene propylene (FEP) electret film with a wavy shape was adopted in the devices, achieving simultaneously a stable embedded biased voltage and a large tensile deformation during vibration. The influences of the factors on the performance of the device, including the initial stretching state of the resilient electret film, seismic mass and depth of the V‐shape counter electrode, were analyzed comprehensively with finite element simulation and compared to experiments. Further, the structure of the device was optimised for generating a high output power, and a good agreement between the simulation and experimental data was achieved. Additionally, the resonant frequency of the device can be easily tuned between 28 and 68 Hz by merely adjusting the initial stretching state of the wavy FEP electret film, guaranteeing great superiority for broad bandwidth energy harvesting applications. For an optimised energy harvester with a volume of only 15 × 5 × 1.7 mm3 and a tiny seismic mass of 25 mg, and a normalized output power referring to 1 × g (g is the gravity of the Earth) up to 547 μW was obtained at its resonant frequency of 28 Hz. These results demonstrate that such a miniaturised vibrational energy harvester is a promising electrical energy supplier for low‐power‐consumption electronic devices, in particular in wireless sensor networks.

Abstract Neutralizing antibodies exert a potent inhibitory effect on viral entry; however, they are less effective in therapeutic models than in prophylactic models, presumably because of their limited efficacy in eliminating virus-producing cells via Fc-mediated cytotoxicity. Herein, we present a SARS-CoV-2 spike-targeting bispecific T-cell engager (S-BiTE) strategy for controlling SARS-CoV-2 infection. This approach blocks the entry of free virus into permissive cells by competing with membrane receptors and eliminates virus-infected cells via powerful T cell-mediated cytotoxicity. S-BiTE is effective against both the original and Delta variant of SARS-CoV2 with similar efficacy, suggesting its potential application against immune-escaping variants. In addition, in humanized mouse model with live SARS-COV-2 infection, S-BiTE treated mice showed significantly less viral load than neutralization only treated group. The S-BiTE strategy may have broad applications in combating other coronavirus infections.

Baiyuerius gen. nov., a new genus of the subfamily Coelotinae F. O. Pickard-Cambridge, 1893 is described, including five new species: B. daxi sp. nov. (♀), B. pindong sp. nov. (♂), B. tamdao sp. nov. (♀), B. zhuping sp. nov. (♂) and B. zuojiang sp. nov. (♂♀), from southern China and northern Vietnam. Our molecular phylogenetic analyses support Baiyuerius gen. nov. as monophyletic and as a sister group of the newly established genus Yunguirius Li, Zhao & Li, 2023.

Abstract Biogenesis of organelles requires targeting of a subset of proteins to specific subcellular domains by signal peptides or mechanisms controlling mRNA localization and local translation. How local distribution and translation of specific mRNAs for organelle biogenesis is achieved remains elusive and likely to be dependent on the cellular context. Here we identify Trinucleotide repeat containing-6a (Tnrc6a), a component of the miRNA pathway, distinctively localized to apical granules of differentiating airway multiciliated cells (MCCs) adjacent to centrioles. In spite of being enriched in TNRC6A and the miRNA-binding protein AGO2, they lack enzymes for mRNA degradation. Instead, we found these apical granules enriched in components of the mRNA translation machinery and newly synthesized proteins suggesting that they are specific hubs for target mRNA localization and local translation in MCCs. Consistent with this, Tnrc6a loss of function prevented formation of these granules and led to a broad reduction, rather than stabilization of miRNA targets. These included downregulation of key genes involved in ciliogenesis and was associated with defective multicilia formation both in vivo and in primary airway epithelial cultures. Similar analysis of Tnrc6a disruption in yolk sac showed stabilization of miRNA targets, highlighting the potential diversity of these mechanisms across organs.

Liver fibrosis and hepatocellular carcinoma (HCC) have been worldwide threats nowadays. Liver fibrosis is reversible in early stages but will develop precancerosis of HCC in cirrhotic stage. In pathological liver, excessive H2O2 is generated and accumulated, which impacts the functionality of hepatocytes, Kupffer cells (KCs) and hepatic stellate cells (HSCs), leading to genesis of fibrosis and HCC. H2O2 accumulation is associated with overproduction of superoxide anion (O2•−) and abolished antioxidant enzyme systems. Plenty of therapeutics focused on H2O2 have shown satisfactory effects against liver fibrosis or HCC in different ways. This review summarized the reasons of liver H2O2 accumulation, and the role of H2O2 in genesis of liver fibrosis and HCC. Additionally, nanotherapeutics targeting H2O2 were summarized for further consideration of antifibrotic or antitumor therapy.

The spider genus Asceua Thorell, 1887 contains 34 species, almost entirely limited to Indochina, India, Sri Lanka and China, with a regional distribution. Eleven species of Asceua are currently only known from China, five of them are described only from one sex.Two new spider species of the genus Asceua are reported from China, A. haocongi sp. n. The glycosylation levels of proteins in cancer cells are closely related to cancer invasion and migration. CD44 is a transmembrane glycoprotein that is significantly overexpressed in a variety of tumor cells and has been proven to promote the migration and motility of cancer cells, but the effect of its N-glycosylation modification on CD44 protein function in tumors is less studied. Here, we investigated the effect of six N-glycan chains (N25/57/100/110/120/255) on CD44s localization, function and stability in hepatocarcinoma cells. When the six sites were mutated, we found that CD44s lost its membrane localization in Huh7 and MHCC-97H cells. On this basis, we identified three glycosylation sites on CD44s (N57, N100 and N110) that played key roles in intracellular localization. When N57, N100 and N110 were mutated together, CD44 localized to the cytoplasm, while another three-site mutant (N25/N120/N255) was still anchored to the membrane. In addition, the ability of CD44-N57Q/N100Q/N110Q to promote the metastasis and invasion of Huh7 and 97H cells was weakened compared with that of CD44-N25Q/N120Q/N255Q. Furthermore, CD44-N57Q/N100Q/N110Q accumulated abnormally in the ER, and a high level of the ER stress (ERS) marker BiP was detected at the same time compared with wild-type CD44. When the lysosome inhibitor CQ was added, the content of mutant protein that triggered ERS significantly increased, which indicated that the degradation mode of CD44-N57Q/N100Q/N110Q after ERS was mainly through the lysosomal pathway (ERLAD). The results revealed that the N-glycosylation sites N57, N100 and N110 mutated on CD44s affected its function and degraded it by lysosomes after triggering ERS. These findings provide data for new studies on ER-related degradation, further promote the study of the glycan chain function of CD44 and furnish new ideas for the treatment of liver cancer metastasis.

Myocardial infarction (MI) is one of the complex phenotypes of coronary artery disease, which results from the interaction of multiple genetic and environmental factors. Nicotinamide Adenine Dinucleotide (NAD+) is an important cofactor regulating metabolic homeostasis and a rate-limiting substrate for sirtuin (SIRT) deacetylase. Numerous NAD+ studies have shown that it can be used as an anti-MI treatment. However, there have been few systematic reviews of the overall role of NAD+ in treating MI. MI, which has long been a global health problem, still lacks effective treatment till now, and the discovery of NAD+ provides a new perspective on its adjuvant treatment. This review summarizes the role of NAD+ signaling in SIRTs in alleviating MI.

As a typical chlorophenol, 2,4,6-trichlorophenol (2,4,6-TCP) is highly toxic and difficult to degrade. Here, we prepared a novel material (Cu/HAP@sBC) by combining copper-based hydroxyapatite with biochar, which removes 2,4,6-TCP via activated persulfate (PS). Cu/HAP@sBC was tested using various characterization methods. In addition, the removal of 2,4,6-TCP was up to 100% after 60 min, and the Cu/HAP@sBC/PS system had a broad pH application range (3−10). Based on the results, SO4−•, •OH, •O2−, and 1O2 were the main reactive species. Finally, the possible degradation pathways of 2,4,6-TCP were explored based on the intermediates identified by high-performance liquid chromatography-mass spectrometry (LC-MS).

IntroductionHypomorphic mutations of DCLRE1C cause an atypical severe combined immunodeficiency (SCID), and Epstein-Barr virus (EBV)-related colon lymphoma is a rare complication.Case presentationA teenage boy presented with colon EBV-related colon lymphoma, plantar warts, and a history of recurrent pneumonia. His peripheral blood lymphocyte count and serum level of immunoglobulin (Ig) G were normal, but he exhibited a T+B-NK+ immunophenotype. Genetic analysis by whole exome sequencing revealed compound heterozygous mutations of DCLRE1C (NM_001033855.3), including a novel paternal splicing donor mutation (c.109 + 2T>C) in intron 1, and a maternal c.1147C>T (p.R383X) nonsense mutation in exon 13. Based on his clinical features and genetic results, the diagnosis of atypical SCID with colon lymphoma was established. Our review shows that seven patients, including our patient, have been reported to develop lymphoma, all with hypomorphic DCLRE1C mutations. Among these cases, six had EBV-related B-cell lineage lymphoma, and one had Hodgkin lymphoma with EBV reactivation. Unfortunately, all of the patients died.ConclusionRecognizing the radiosensitivity of the disease is critical for the prognosis. Hematopoietic stem cell transplantation before being infected with EBV is an optimal treatment.

Being prepared by a liquid-phase reduction method, sulfur-modified nanoscale zero-valent iron (S-nZVI) was then coated with sodium alginate (SA) to form gel beads (SAS-nZVI) which are capable of removing Pb(II) from water. SAS-nZVI was characterized by SEM, EDS, FTIR, XRD, and BET, and its removal effect on Pb(II) in water, including the effects of pH, adsorbent dosage, shaking time, and initial concentration of lead, was also studied. The results demonstrated that the maximum removal efficiency of Pb(II) by SAS-nZVI was 97.89%, and the maximum uptake was 246.40 mg/g. In the Pb(II) removal behavior study, the pseudo-second-order kinetic model and the Langmuir isotherm model were found to fit the adsorption process well. SAS-nZVI was easier to recycle from the reaction system, and the removal efficiency of SAS-nZVI to Pb(II) in water was still able to reach 82.75% after five cycles. Therefore, this study suggests that SAS-nZVI has a high removal capacity for Pb(II) and great potential in water pollution treatment. HIGHLIGHTS The prepared SAS-nZVI nanocomposites not only reduce agglomeration, and improve oxidation resistance, but also show good dispersion.; SAS-nZVI is more easily separated from the reaction system, avoiding nZVI draining in practical applications, and causing secondary pollution.; SAS-nZVI has better cycle utilization. After five cycles, the Pb(II) removal rate was still 82.75%.;

Abstract Hypoimmunogenic human pluripotent stem cells (hPSCs) are expected to serve as an unlimited cell source for generating universally compatible “off-the-shelf” cell grafts. However, whether the engineered hypoimmunogenic hPSCs still preserve their advantages of unlimited self-renewal and multilineage differentiation to yield functional tissue cells remains unclear. Here, we systematically studied the self-renewal and differentiation potency of three types of hypoimmunogenic hPSCs, established through the biallelic lesion of B2M gene to remove all surface expression of classical and nonclassical HLA class I molecules (B2Mnull), biallelic homologous recombination of nonclassical HLA-G1 to the B2M loci to knockout B2M while expressing membrane-bound β2m-HLA-G1 fusion proteins (B2MmHLAG), and ectopic expression of soluble and secreted β2m-HLA-G5 fusion proteins in B2MmHLAG hPSCs (B2Mm/sHLAG) in the most widely used WA09 human embryonic stem cells. Our results showed that hypoimmunogenic hPSCs with variable expression patterns of HLA molecules and immune compromising spectrums retained their normal self-renewal capacity and three-germ-layer differentiation potency. More importantly, as exemplified by neurons, cardiomyocytes and hepatocytes, hypoimmunogenic hPSC-derived tissue cells were fully functional as of their morphology, electrophysiological properties, macromolecule transportation and metabolic regulation. Our findings thus indicate that engineered hypoimmunogenic hPSCs hold great promise of serving as an unlimited universal cell source for cell therapeutics.

Salvia miltiorrhiza Bunge is a traditional medicinal plant in China and poly-glutamic acid (PGA) is a valuable biopolymer. However, it is unclear whether PGA promotes growth and the accumulation of main medicinal components in S. miltiorrhiza. To elucidate this scientific question, the influences of PGA on the growth, physiological characteristics, and accumulation of main medicinal components in S. miltiorrhiza were explored through a pot experiment. The results revealed that PGA significantly promoted basal diameter, plant height, shoot and root biomass, as well as root volume, compared with control. PGA also increased SPAD value, net photosynthetic rate, actual and maximum photochemical efficiency of photosynthetic system II, photochemical quenching, and electronic transfer rate. Meanwhile, PGA increased transpiration rate, stomatal conductance, water use efficiency, leaf relative water content, and the contents of soluble protein, soluble sugar, and proline. Furthermore, PGA increased the activities of antioxidant enzymes and the contents of antioxidants. The above findings imply that PGA facilitated S. miltiorrhiza growth by enhancing photosynthetic performance, water metabolism, and antioxidant capacity. Additionally, PGA significantly improved the yield of rosmarinic acid, salvianolic acid B, dihydrotanshinone, cryptotanshinone, tanshinone I, and tanshinone ⅡA in roots by up-regulating the transcript levels of genes responsible for their biosynthesis. Our findings indicated that PGA promoted S. miltiorrhiza growth and the accumulation of main medicinal components in roots.

In practice, trade-ins are offered by new energy vehicle producers and include trade-in-for-new fuel vehicles. And trade-in-for-new energy vehicles. By using the game-analytical method, we mainly analyze the optimal pricing strategy of new energy vehicle producers when they provide both of the above mentioned trade-in services. The results show that automobile producers should consider the production cost of new vehicles and the trade-in rebate when they provide a trade-in strategy. On the one hand, when the production cost of new vehicles is high but the trade-in rebate is very low, automobile producers do not provide the trade-in service. On the other hand, when the production cost of new vehicles is very low but the trade-in rebate is very high, automobile producers should provide trade-in services to heterogeneous consumers. Moreover, when the heterogeneous behavior of consumers is strong and the innovation value of new generation fuel vehicles is low, automobile producers should choose the preannounce pricing strategy. Otherwise, these producers should adopt the dynamic pricing strategy. This study investigated the effects of cerium (Ce) on the growth and fruit quality of strawberries under salt stress. The findings revealed that salt stress markedly enhanced the activities of antioxidant enzymes and increased the contents of malonaldehyde (MDA) and hydrogen peroxide (H2O2) in leaves and the contents of anthocyanins, phenolic compounds, vitamin C (Vc), soluble sugar (SS) and titratable acid (TA) in fruits. Ce markedly improved the activities of ascorbate peroxidase, superoxide dismutase, peroxidase and catalase in leaves and the contents of anthocyanins, phenolic compounds, Vc and SS in fruits, but significantly decreased MDA and H2O2 levels in leaves and TA content in fruits under salt stress. However, salt stress significantly decreased the contents of chlorophyll (Chl) and carotenoids (Car), photosynthetic rate (Pn), transpiration rate (Tr), stomatal conductance (gs), relative water content (RWC), plant height and biomass, and fruit weight and sugar-acid ratio (SAR). Compared with salt stress alone, Ce obviously increased Chl and Car contents, Pn, Tr, gs, RWC, plant height and biomass, as well as fruit weight and SAR. The above results suggested that Ce showed beneficial effects on the growth and fruit quality of strawberries under salt stress.

A carbon derivative with Fe-Cu bimetallic organic framework (Fe-Cu-MOF@C) was prepared by microwave synthesis and pyrolysis. Using potassium persulfate (PS) as oxidant and 2,4-dichlorophenol (2,4-DCP) as target pollutant, the optimal preparation conditions of Fe-Cu-MOF@C were studied. The factors affecting the synthesis of Fe-Cu-MOF include microwave power, microwave time, microwave temperature, the molar ratio of metal ions to organic ligands, the molar ratio of iron and copper, etc. In addition, the influence of pyrolysis temperature on the performance of Fe-Cu-MOF@C cannot be ignored. The results show that Fe-Cu-MOF@C has the best catalytic performance when the microwave time is 30 min, the microwave power is 600 W, the microwave temperature is 150 °C, the molar ratio of (Fe2+ + Cu2+)/H2BDC is 10:3, the molar ratio of Fe2+/Cu2+ is 10:1, and the pyrolysis temperature is 700 °C. After 90 min of reaction, 2,4-DCP was completely removed. Repeatable experiments show that Fe-Cu-MOF@C has good stability and its service life can be restored by heat treatment. In this study, a heterogeneous catalyst with strong catalytic capacity, high stability and easy recovery was prepared by a simple and efficient process, which is conducive to the development of advanced oxidation technology and the progress of water environmental protection. HIGHLIGHTS A new method for preparing carbonized derivatives of Fe-Cu bimetallic organic framework (Fe-Cu-MOF@C) based on microwave and pyrolysis was proposed.; This material is an excellent catalyst for PS, which can efficiently and thoroughly degrade chlorophenol compounds in water.; The optimum preparation conditions were determined.; The material has good stability and long service life.;
