lncRNAs, a group of eukaryotic cell genome-encoded transcripts, have been demonstrated to exert a notable impact on tumorigenesis. LINC01535, belonging to the lncRNA family, was reported to have an aberrant expression in certain types of cancers and thus affect cancer progression. Nevertheless, the expression pattern and potential roles of LINC01535 in clear cell renal cell carcinoma (ccRCC) remain to be elucidated. Here, LINC01535 expression was detected in ccRCC by RT-qPCR, cell proliferation by CCK-8 assays, and invasion by transwell assays. Besides, effects of LINC01535 on in vivo tumor growth were investigated by xenograft tumor models. The miR-146b-5p/LINC01535/TRIM2 interaction was evaluated via luciferase reporter assays. This study showed downregulation of LINC01535 in ccRCC. Moreover, LINC01535 upregulation attenuated in vitro ccRCC development and hindered in vivo tumor growth. Furthermore, LINC01535 sponged miR-146b-5p which had a negative correlation with LINC01535, and TRIM2 was a direct target of miR-146b-5p and mediated by LINC01535. Mechanically, LINC01535/miR-146b-5p/TRIM2 axis affected ccRCC progression by mediating the PI3K/Akt signaling. All in all, our observations suggest the LINC01535/miR-146b-5p/TRIM2 axis as a crucial role in ccRCC.

The members of the Nedd4-like E3 family participate in various biological processes. However, their role in clear cell renal cell carcinoma (ccRCC) is not clear. This study systematically analyzed the Nedd4-like E3 family members in ccRCC data sets from multiple publicly available databases. NEDD4L was identified as the only NEDD4 family member differentially expressed in ccRCC compared with normal samples. Bioinformatics tools were used to characterize the function of NEDD4L in ccRCC. It indicated that NEDD4L might regulate cellular energy metabolism by co-expression analysis, and subsequent gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis. A prognostic model developed by the LASSO Cox regression method showed a relatively good predictive value in training and testing data sets. The result revealed that NEDD4L was associated with biosynthesis and metabolism of ccRCC. Since NEDD4L is downregulated and dysregulation of metabolism is involved in tumor progression, NEDD4L might be a potential therapeutic target in ccRCC.

To assess the feasibility, safety, and efficiency of bilateral inguinal lymphadenectomy using simultaneous double laparoscopies for penile cancer.We reviewed retrospectively the records of 65 patients who underwent inguinal lymph nodes dissection (ILND) for penile cancer from January 2012 to May 2019. Treatments included open ILND (OILND, 19 patients), video-endoscopic inguinal lymphadenectomy (VEIL) using single laparoscopy (S-VEIL, 24 patients), and VEIL using double laparoscopies (D-VEIL, 22 patients). We evaluated the peri-operative and short-term oncological outcomes of the three groups.The mean operative time of D-VEIL (105.91 ± 10.87 minutes) was significantly shorter than the other two groups, OILND shorter than S-VEIL (160.47 ± 13.74 minutes, 191.67 ± 17.80 minutes, respectively) (P0.001). Intraoperative blood loss in the S-VEIL and D-VEIL groups were 53.54 ± 8.78 and 48.41 ± 13.22 ml, respectively; they were significantly lower than that of the OILND group (99.74 ± 9.64 ml; P0.001). The numbers of unilateral and total lymph nodes harvested were similar in all groups. The complication rates in the S-VEIL group (4.2%) and the D-VEIL group (4.5%) were significantly lower than that in the OILND group (63.2%; P0.001). Compared with open surgery (13.53 ± 1.74 days for hospitalization; 11.37 ± 1.92 days for the left side of drain, 11.95 ± 1.84 days for the right side), the two VEIL groups had significantly shorter drainage tube residence time (7.42 ± 2.02 and 7.32 ± 1.52 days, respectively for the left side; 7.63 ± 1.81 and 7.27 ± 1.58 days, respectively for the right side), shorter postoperative hospitalization (9.46 ± 1.64 and 9.00 ± 1.83 days, respectively) (P0.001). There were no statistically significant differences in rates of regional recurrence and short-term survival among the three groups.Bilateral inguinal lymphadenectomy using double laparoscopies simultaneously can provide adequate oncological outcomes safely and efficiently, and carry significantly lower morbidity than OILND, at a median follow-up of 33.5 months. It is a more time-saving surgical approach for penile cancer patients who need bilateral ILND.

The multi-hole steam ejection system is widely applied in practical engineering applications, whereas the heat transfer mechanisms in unstable condensation conditions are not very clear yet. In this study, the separate-effect mechanism tests of single/double-hole sparger, and 88-hole scaled ADS sparger tests are carried out to investigate the unstable steam spraying condensation performances. The experimental results indicate that for the multi-hole steam unstable condensation process, the plumes expand and contract in high frequency, and the instantaneous heat transfer coefficients are estimated quantitatively. The multi-hole steam condensation regimes are significantly determined by the steam mass flow flux and water sub-cooling degree, then the double-hole condensation regimes map is proposed based on the key inputting parameters. Moreover, the simplified lumped condensation model for the multi-hole condensation process is developed, in which the upper and lower heat transfer coefficient values limitations are evaluated to be 4.3–5.9 MW/(m2 °C) and 0.89–0.94 MW/(m2 °C), respectively. The ADS 1–3 scaled 88-hole steam condensation results further validate the appropriation of the lumped condensation model, with the heat transfer coefficients estimated as 0.17–2.9 MW/(m2 °C).

The CYCIAE-230 cyclotron is a proton beam accelerator dedicated to cancer therapy. The low-level radiofrequency (LLRF) system of CYCIAE-230 cyclotron is designed based on the previous LLRF systems which have been successfully commissioned for CYCIAE-100 and CYCIAE-14 cyclotrons. This design includes four feedback control loops, namely, tuning loop, amplitude loop, phase loop, and balance loop. The LLRF system for CYCIAE-230 cyclotron have the same amplitude, phase, and tuning loops as the previous designs. Meanwhile, the balance loop makes the LLRF system stand out. It is enabled after the amplitude loop is closed and controls the homogeneity of the accelerating fields of the four cavities. From the bird view of the control system of the cyclotron, the LLRF system is an embedded control unit which communicates with the program logical controller with Profibus-DP (Distributed Peripherals). In this paper, the desktop experiment has been performed with a small-scale copper cavity with low power. This small-scale cavity has an unloaded quality factor ( Q 0 ) of around 7800 and has push–pull and push–push resonate modes, which are the characteristic parameters of the cavities of CYCIAE-230 cyclotron. The design and performance of the LLRF system were discussed based on the desktop experiment results in this paper.

Accumulating evidence reports that Capn4 plays an important role in the development and progression of various malignant cancers. However, whether Capn4 is involved in prostate cancer remains unclear. Therefore, the aim of this study was to investigate the expression, biological function and regulatory mechanism of Capn4 in prostate cancer. Herein, we found that Capn4 was highly expressed in prostate cancer cell lines compared with normal prostate cells. Capn4 gene silencing markedly suppressed the growth, invasion and Wnt/β-catenin signaling of prostate cancer cells, whereas Capn4 overexpression showed an oncogenic effect. Moreover, silencing of β-catenin significantly blocked the oncogenic effect of Capn4 overexpression. Bioinformatics analysis predicted that Capn4 was a potential target gene of microRNA-520b (miR-520b), which has been reported as a tumor suppressive miRNA in various cancers. The dual-luciferase reporter assay confirmed that miR-520b directly bound to the 3'-untranslated region of Capn4. Real-time quantitative PCR and Western blot analysis showed that miR-520b negatively regulated Capn4 expression in prostate cancer cells in vitro. Furthermore, we found that miR-520b was significantly downregulated in prostate cancer cell lines and tissues. In addition, miR-520b expression was inversely correlated with Capn4 expression in prostate cancer clinical specimens. Overexpression of miR-520b mimicked the tumor suppressive effect of Capn4 siRNA, whereas inhibition of miR-520b had an oncogenic effect. Restoration of Capn4 significantly blocked the antitumor effect of miR-520b in prostate cancer cells. Overall, our findings demonstrate an oncogenic role of Capn4 in prostate cancer and show that its expression is epigenetically regulated by miR-520b. Our results reveal that suppression of Capn4 by miR-520b inhibits the growth and invasion of prostate cancer cells associated with downregulated Wnt/β-catenin signaling, indicating an important role of the miR-520b/Capn4/Wnt/β-catenin regulation axis in the molecular pathogenesis of prostate cancer. Our study suggests that miR-520b and Capn4 may represent potential and novel therapeutic targets for prostate cancer.

Prostate cancer (PCa) is a common malignancy in male urinary system. Cell division cycle-associated protein 1 (CDCA1) is expressed highly in many cancer cells. Yet, whether CDCA1 play an important role in PCa progression is uncertain. pH low insertion peptide (pHLIP), a PH-induced transmembrane structure, can pass through the cell membrane into intracellular in an acidic environment. In this study, we try to confirm the expression status of CDCA1 in the PCa patients' tissues and PCa cell line. In addition, to make the CDCA1-siRNA efficiently targeting the PCa cells, pHLIP and CDCA1-siRNA were combined with disulfide bond to become effector molecules. By the characteristics of the pHLIP allosteric occurring in cancer tissue acidic microenvironment, CDCA1-siRNA may be transported specificity into prostatic cancer cells and released in the cytoplasm. The interference effect of the effector molecules on the CDCA1 was detected in vitro and in vivo. The results showed that CDCA1 was highly expressed in PCa cell line and human PCa clinical samples. Knock down CDCA1 significantly inhibit the growth and promote the apoptosis of prostatic cancer cells. In the intracellular translocation experiment, CDCA1-siRNA could be delivered into cytoplasma at pH 6.2, but not at pH 7.4. In the in vivo test, the tumor size was reduced obviously in the NOD/SCID mice treated with pHLIP-CDCA1-siRNA compared to the CDCA1-siRNA and the bioluminescent signal of Cy5-pHLIP-CDCA1-siRNA was focused detected in the tumor site. Our findings indicated that CDCA1 might be a very key molecule regulating survival and proliferation of PCa. pHLIP-CDCA1-siRNA might be a promising targeting therapy for PCa.

Most of the old oilfields in China have entered the stage of ultra-high water cut development. In order to guide drilling wells and further exploitation, it is necessary to describe the remaining oil distribution at present accurately and quantitatively. At the same time, the focus of work should be shifted to the study of remaining oil distribution between wells. For the oilfields with wide area, large thickness, long history of production and many injection production wells, the calculation of numerical simulation is too large and the grid precision is coarse. Although the traditional modeling method of mixing new and old wells can ensure the grid precision, it cannot represent the current situation of the reservoir and play a guiding role in production, so it needs a new way, ensures the plane and vertical precision of the model (accuracy 10×10×0.5m), and through certain constraints, can accurately describe the current plane and vertical oil saturation distribution law of the oilfield, which provides a new direction for further exploitation of the oilfield.

The CYCIAE230 is a low-current, compact superconducting cyclotron designed for proton therapy. The Radio Frequency system consists of four RF cavities and applies second harmonic to accelerate beams. The driving power for the cavity system is estimated to be approximately 150 kW. The LLRF controller is a self-made device developed and tested at low power using a small-scale cavity model. In this paper, the resonator systems of an S.C. cyclotron in history are reviewed. Contrary to those RF systems, the cavities of the CYCIAE230 cyclotron connect two opposite dees. Two high-power RF windows are included in the system. Each window carries approximately 75 kW RF power from the driver to the cavities. Thus, the RF system for the CY-CIAE230 cyclotron is operated in driven push–pull mode. The two-way amplifier-coupler-cavity systems are operated with approximately the same amount of RF power but 180° out of phase compared with each other. The design, as well as the technical advantage and limitations of this operating mode, of the CYCIAE230 cyclotron RF system is analyzed.

In advanced third generation reactor AP1000, the C-shape Passive Residual Heat Removal Heat Exchanger (PRHR HX) immerged in the In-containment Refueling Water Storage Tank (IRWST) is the key equipment in the passive safety system. The mutual influences of the C-shape bundle and the irregular IRWST lead to the complicated heat transfer characteristics. It is still unclear which heat transfer correlations are appropriate for the vertical and horizontal bundle sections, respectively. In the present work, an overall scaled-down IRWST and PRHR HX experimental bench has been built up to simulate the pool boiling performance of the C-shape PRHR HX bundle. The key thermal parameters are measured to evaluate the heat transfer mechanisms and heat transfer coefficients (HTCs) for different bundle sections. The traditional and the revised correlations are further compared with the experimental data of the C-shape PRHR HX bundle. The validated correlations provide applicable and effective references for the reliable operation and system analysis of the passive safety system.

Nonbinary single-nucleotide polymorphisms (SNPs) are potential forensic genetic markers because their discrimination power is greater than that of normal binary SNPs, and that they can detect highly degraded samples. We previously developed a nonbinary SNP multiplex typing assay. In this study, we selected additional 20 nonbinary SNPs from the NCBI SNP database and verified them through pyrosequencing. These 20 nonbinary SNPs were analyzed using the fluorescent-labeled SNaPshot multiplex SNP typing method. The allele frequencies and genetic parameters of these 20 nonbinary SNPs were determined among 314 unrelated individuals from Han populations from China. The total power of discrimination was 0.9999999999994, and the cumulative probability of exclusion was 0.9986. Moreover, the result of the combination of this 20 nonbinary SNP assay with the 20 nonbinary SNP assay we previously developed demonstrated that the cumulative probability of exclusion of the 40 nonbinary SNPs was 0.999991 and that no significant linkage disequilibrium was observed in all 40 nonbinary SNPs. Thus, we concluded that this new system consisting of new 20 nonbinary SNPs could provide highly informative polymorphic data which would be further used in forensic application and would serve as a potentially valuable supplement to forensic DNA analysis.

The heat transfer performance of Passive Residual Heat Removal Heat Exchanger (PRHR HX) immerged in the In-containment Refueling Water Storage Tank (IRWST) is of great importance for the efficient and safe removal of the residual heat in the AP1000 plant. The C-shape heating rods bundle is being used in the PRHR HX, yet the heat transfer characteristics of this special C-shape heat exchanger are not completely clear. The Westinghouse Electric Corporation has performed the PRHR HX experiment using three vertical tubes to validate the design of PRHR HX. However, the horizontal sections of the C-shaped bundle were neglected, and attentions were mainly focused on the local heat transfer effect of the vertical tubes. In the present work, an overall scaled-down IRWST and PRHR HX models are built to simulate the thermal hydraulic process in the residual heat removal accident. The natural convection heat transfer characteristics of both the overall IRWST model and the local C-shaped PRHR HX bundle model are researched in the first stage. For the overall IRWST model, the natural convection flow vectors are qualitatively measured by PIV technique. The velocity and temperature distributions are influenced mutually, then thermal stratification phenomenon develops in the overall IRWST. The existence of the “thermal interface” has great influences on the thermal stratification. For the local PRHR HX heat transfer, traditional natural convection correlations for both horizontal and vertical bundles are compared to the experimental data. For the vertical bundle of the PRHR HX model, a revised Rayleigh number is proposed as a criterion for the slender vertical rod bundles, when the vertical rod bundle outside flow cannot be treated as flat plate. Then revised correlations are proposed to predict the PRHR HX average heat transfer coefficient. Moreover, the analysis shows that the forced convection also plays an important role in enhancing the heat transfer effect of the PRHR HX model upper horizontal bundle, so the modified Numix is recommended for the PRHR HX upper horizontal bundle secondary heat transfer calculation.

We in this study investigated the role of lncRNA NR2F2-AS1 in prostate carcinoma (PC). We showed that NR2F2-AS1 was upregulated in PC and positively correlated with CDK4. In PC cells, NR2F2-AS1 overexpression led to upregulated, while NR2F2-AS1 siRNA silencing led to downregulated CDK4. Follow-up study revealed that high levels of NR2F2-AS1 and CDK4 in PC tissues were closely correlated with the poor survival of PC patients. Cell proliferation data showed that NR2F2-AS1 overexpression led to increased, while NR2F2-AS1 siRNA silencing led to proliferation rate of PC cells. Moreover, NR2F2-AS1 also showed positive effects on cell cycle progression. In addition, CDK4 overexpression reduced the effects of NR2F2-AS1 siRNA silencing. Therefore, NR2F2-AS1 positively regulates CDK4 to promote cancer cell proliferation in PC.

The necrobiome is the postmortem community that includes bacteria, fungi, arthropods, and other cadaver-associated organisms. It has been suggested as biological evidence for forensic investigation. Fungi form distinctive mildew spots in colonizing decomposing bodies, converting them into moldy cadavers. However, the postmortem fungal community consists of more than these visible species. Characterizing the succession pattern of the fungal community during decomposition is valuable not only for understanding the ecosystem composition of the cadaver decomposition islands but also for contributing to forensic investigations. In the present study, the fungal composition of pig cadavers and succession patterns during decomposition were investigated with high-throughput sequencing. The succession patterns were easier to discern in outdoor cadavers, compared with those that were placed indoors. The metabarcoding approach revealed trends linking particular fungal taxa with specific postmortem intervals (PMIs). Dominant species increased notably in cadavers and soil. Furthermore, the succession of the soil community was driven by the cadaver decomposition. Significant mycoflora differences were observed between environmental and cadaveric soil. The results obtained suggested that postputrefaction mycoflora have considerable potential for PMI estimation, particularly in cases that involve heavily decomposed bodies. In addition, the diversity of fungal communities revealed by the metabarcoding approach allowed us to discriminate the sites of cadaver decomposition, implying that postputrefaction mycoflora may be helpful in identifying the environment in which a cadaver has been placed, or the original location from which a cadaver has been moved. Our results provide an important step towards developing fungal evidence for use in forensic science and add to the growing body of work on postmortem microbial communities.

The Kongdian Oilfield is located in eastern China. More than 30 years have passed since the oil field was discovered in 1979. The majority of the oil is produced from reservoirs of the Guantao Formation (Miocene). A detailed description and interpretation of core, well-logging and 3-D seismic stratal slices data was carried out to analyze sedimentological characteristics of the Guantao Formation and two fluvial styles have been identified encompassing braided channel and meandering channel. Seven main facies associations were defined: (i) upward-fining channel, (ii) braided channel with mid-channel bar, (iii) meandering channel with point bar, (iv) levee, (v) floodplain, (vi) minor channel, (vii) swamp. Vertical changes in fluvial styles of the Guantao Formation were recognized by variations in preserved thickness, channels stacking pattern, regional paleoflow directions, and channel/floodplain ratios. The fluvial succession is 50–70 m thick in the lower Guantao Formation, composed of braided channels and high channel/floodplain ratio. The alluvial succession is 60 m in thickness in the upper Guantao Formation, the dominant fluvial styles change to meandering channels and channel/floodplain ratio reduces. The main factor that may have affected the fluvial style is the base-level changes. With the rise of the base level the fluvial style is believed to have changed from braided channels to meandering channels.

MicroRNAs (miRNAs) have emerged as important regulators in cancer that are implicated in regulation of various cellular processes. miR-382 has been proposed as a tumor suppressor by several recent studies. However, the function of miR-382 in prostate cancer remains unknown. In this study, we aimed to investigate the potential function of miR-382 in prostate cancer. We found that miR-382 was significantly decreased in prostate cancer specimens and cancer cell lines. The overexpression of miR-382 in prostate cancer cells markedly inhibited cell proliferation, migration, and invasion. In contrast, miR-382 suppression exhibited an opposite effect. Target analysis predicted that chicken ovalbumin upstream promoter transcription factor II (COUP‑TFII) was a direct target of miR-382. This prediction was experimentally confirmed by dual-luciferase reporter assay, real-time quantitative polymerase chain reaction, and western blot analysis. Our results further demonstrated that miR-382 inhibited the downstream genes of COUP‑TFII, including Snail and matrix metalloproteinase 2 (MMP2). Moreover, the restoration of COUP‑TFII expression significantly blocked the inhibitory effect of miR-382 on cell proliferation, migration, and invasion, and Snail expression. Taken together, this study suggests that miR-382 inhibits prostate cancer cell proliferation and metastasis through inhibiting COUP‑TFII, representing an important new mechanism for understanding prostate cancer pathogenesis and providing a novel therapeutic candidate target for prostate cancer therapy.

The effective and reliable operation of the Passive Residual Heat Removal System (PRHRS) is very significant for the safety of the reactor. However, the passive heat sink tank IRWST in PRHRS is vulnerable to get stratified, which has deleterious impacts on the heat transfer effect of the Passive Residual Heat Removal Heat Exchanger (PRHR HX). In the present work, both the numerical simulations and experiments are conducted to investigate the passive baffles designs of the PRHR HX&IRWST. An overall scaled-down separate-effects PRHR HX&IRWST experimental test bench has been built, and the experimental data are used to validate the numerical methods in the prototype PRHR HX model. Different passive baffles Designs A–D are tested employing the commercial CFD software CFX, and then validated by the baffles experiment. The influencing factors of baffles number, thermal conductivity, and baffles shape are investigated. Both the numerical and experimental temperature distributions, natural convection velocity, thermal stratification extent, as well as the Heat Transfer Coefficient are analyzed. The complicated factors combine to determine the overall HTC in baffle design. The numerical and experimental results provide possible improvement ways for the optimization of the PRHR HX baffles design, which have practical engineering application significance in the optimum design of the passive safe system in AP1000 plant.

In AP1000 plant, the Automatic Depressurization System (ADS) 1–3 stages operate to discharge the high-temperature and high-pressure steam from the Reactor Coolant System (RCS) primary side to the large heat sink tank In-containment Refueling Water Storage Tank (IRWST) in accidental conditions. The key equipment’s specific shape and arrangement lead to the complicate flow and heat transfer characteristics in IRWST. In the present work, an overall scaled IRWST&ADS sparger experiment has been built up. The thermocouples matrix, flowmeters, pressure transmitters, heat flux sensors, Particle Image Velocimetry (PIV) technique, and high speed camera are employed for the measurements of the key thermal and flow parameters. The local steam jets condensation phenomena as well as the overall flow and thermal behavior are investigated. The experimental results indicate that the thermal stratification phenomenon is obvious in IRWST. The criteria of Richardson Number and Stratification Number are utilized to predict and evaluate the thermal stratification extent, respectively. An improved ADS arrangement design is further proposed to reduce the thermal stratification. Moreover, the multi-holes lumped “steam condensation column” is modeled with characteristic parameters, then the steam condensation heat transfer coefficient range in chugging condensation process is estimated. The experimental results provide practical engineering application reference for the effective operation of the passive safety system in AP1000 plant.

As a driven accelerator, the CYCIAE-100 cyclotron is designed by China Institute of Atomic Energy for the Beijing Radio Ion-beam Facility project. The cyclotron RF system is designed to use two RF power sources of 100 kW to drive two half-wavelength cavities respectively. Two Dee accelerating electrodes are kept separately from each other inside the cyclotron, while their accelerating voltages are maintained in phase by the efforts of LLRF control. An analog–digital hybrid LLRF system has been developed to achieve cavity tuning control, dee voltage amplitude and phase stabilization etc. The analog subsystems designs are focused on RF signal up/down conversion, tuning control, and dee voltage regulation. The digital system provides an RF signal source, aligns the cavity phases and maintains a Finite State Machine. The digital parts combine with the analog functions to provide the LLRF control. A brief system hardware introduction will be given in this paper, followed by the review of several major characteristics of the digital control in the 100 MeV cyclotron LLRF system. The commissioning is also introduced, and most of the optimization during the process was done by changing the digital parts.

BACKGROUND It is known that microRNAs (miRNAs) are a class of small, non-coding RNAs that act as key regulators in various physiological and pathological processes. However, the regulatory mechanisms involving miRNAs in prostate cancer remain largely unknown. Here, we found that miR-103 is down-regulated in prostate cancer and closely associated with tumor proliferation and migration. Our objective was to explore the role of the miR-103 in prostate cancer. METHODS In this study, we measured miR-103 level using real-time polymerase chain reaction in the human prostate cancer cell lines, including PC-3, LNCap, 22Rv1, DU145, and the normal prostate epithelium cell line RWPE-1, a total of 25 pairs of primary prostate cancer tissues and adjacent non-cancerous tissues (NCTs) were measured also. In addition, over-expression of miR-103 in prostate cancer cell lines to determine the role of miR-103 in prostate cancer. RESULTS We found that miR-103 is down-regulated in prostate cancer and closely associated with tumor proliferation and migration. In addition, over-expression of miR-103 apparently inhibits prostate cancer cell proliferation and migration in vitro. Gain-of-function in vitro experiments further show that miR-103 mimics significantly inhibited prostate cancer cell proliferation, invasion and increase the cell cycle in G1 phase, while promoted cell apoptosis. Subsequent dual-luciferase reporter assay identified one of the proto-oncogene PDCD10 as direct target of miR-103. CONCLUSIONS Therefore, our data collectively demonstrate that miR-103 is a proto-oncogene miRNA that can suppress prostate cancer proliferation and migration by down-regulating the oncogene PDCD10, indicating that miR-103 may represent a new potential diagnostic and therapeutic target for prostate cancer treatment. The multi-hole spargers are widely used to discharge the high temperature and high pressure steam for depressurization in most engineering applications due to its high efficiency in heat and mass transfer. However, the steam condensation microscopic mechanisms are very complicated, especially for the multi-hole conditions. In this study, the double-hole spraying steam direct contact condensation (DCC) mechanism and heat transfer characteristics are researched based on the scaled IRWST & ADS experimental facility. The double-hole condensation flow regimes are distinguished on the condensation flow patterns diagram, and the steam condensation analytical models are then developed with characteristic parameters including L, Rex, Rx, δ etc. to quantitatively describe the double-hole condensation features. New empirical correlation for predicting the double-hole steam penetration length L is proposed. Moreover, special temperature distributions along both the steam plumes centerline and middle line reflect the interactions between the adjacent steam plumes, especially for the ellipsoidal plume pattern. In essential, the specific temperature variations reflect the equilibrium and transformation between the thermal dynamic energy and kinetic energy. Finally, the heat transfer coefficients for the double-hole steam condensation are calculated based on the steam plume analytical models. The analysis indicates that the double-hole steam condensation heat transfer coefficients are in same order of magnitude with the single-hole stable condensation case, whereas the lower boundary of heat transfer coefficients deceases to some extent. It provides experimental references for the operation and design of the actual industrial steam condensation applications.

TRIUMF’s ARIEL pre-buncher is powered by a sawtooth waveform which is the combination of 11.79 MHz, 23.57 MHz and 35.36 MHz harmonics. To upgrade the existing analog and digital hybrid LLRF control system, reduce the hardware cost, and achieve more flexible control of the system, an FPGA based digital daughter board is designed. The generation, control, and regulation of these three harmonics are all incorporated digitally inside a single FPGA. To reduce the number of RF interfaces on the motherboard, the new daughter board is designed to control the amplitude and phase of the sawtooth waveform with one channel ADC and DAC. To lock the output phase of the sawtooth waveform to the reference signal, a digital phase-locked loop is designed to lock all three harmonics to the external reference signal. The related work, including the system design of the LLRF control system and the development of the digital phase-locked loop, are presented in this paper.

The Beijing Radioactive Ion-beam Facility project is being constructed by BRIF division of China Institute of Atomic Energy. In this project, a 100 MeV high intensity compact proton cyclotron is built for multiple applications. The first successful beam extraction of CYCIAE-100 cyclotron was done in the middle of 2014. The extracted proton beam energy is 100 MeV and the beam current is more than 20 μA. The RF system of the CYCIAE-100 cyclotron includes two half-wavelength cavities, two 100 kW tetrode amplifiers and power transmission line systems (all above are independent from each other) and two sets of Low Level RF control crates. Each set of LLRF control includes an amplitude control unit, a tuning control unit, a phase control unit, a local Digital Signal Process control unit and an Advanced RISC Machines based EPICS IOC unit. These two identical LLRF control crates share one common reference clock and take advantages of modern digital technologies (e.g. DSP and Direct Digital Synthesizer) to achieve closed loop voltage and phase regulations of the dee-voltage. In the beam commission, the measured dee-voltage stability of RF system is better than 0.1% and phase stability is better than 0.03°. The hardware design of the LLRF system will be reviewed in this paper.

DNA-based technologies have been increasingly used in species determination of forensically important sarcophagids, as they are often not morphologically distinct, especially for the immature specimens. The mitochondrial genome has been broadly used for species-level identifications. Although Chinese sarcophagid sequences of short fragments (200-600 bp) had been deposited in GenBank, the barcode region and the complete cytochrome oxidase subunit I (COI) and COII sequences are still unavailable. In this study, 78 sarcophagid fly specimens, representing 17 Chinese sarcophagid species, were collected from 29 locations in 18 Chinese provinces. Sequence data of the mitochondrial COI and COII of the most important Chinese flesh fly taxa associated with cadavers were presented for first time, which serve as reference standards for Chinese species determination. Phylogenetic analysis showed that the COI and COII sequences were useful for identifying most sarcophagid species. The results of this research will be conductive for implementation of the Chinese Sarcophagidae in forensic entomology. However, the application of mitochondrial DNA as species identifier requires great circumspection and additional markers and methods should be studied to ensure accuracy of identification in the future.

To study the population data of Y-chromosome STRs (Y-STRs) of Han population resided in Hunan province, we analyzed haplotypes of 26 Y-STRs (DYS19, DYS385a/b, DYS389I, DYS389II, DYS390, DYS391, DYS392, DYS393, DYS437, DYS438, DYS439, DYS448, DYS456, DYS458, DYS481, DYS533, DYS549, DYS570, DYS576, DYS635, DYS643, DYS388, DYS449, DYS460, and YGATAH4) in 310 unrelated male individuals using a commercially available Goldeneye® DNA ID 26Y system. The calculated average gene diversity values ranged from 0.4211 to 0.9590 for DYS438 and DYS385a/b loci, respectively. The discriminatory capacity was 96.77 % with 300 observed haplotypes. Population relationships between Hunan Han and eight other populations available from Y-chromosome haplotype reference database (YHRD) were compared. The results showed that the Han population resided in the Hunan district is significantly different from other populations. Our results also indicated that these 26 Y-STR loci were highly genetically polymorphic in the Hunan Han population and of great value in forensic application.

The species identification of forensically important sarcophagid flies (Diptera: Sarcophagidae) is very difficult and requires strong taxonomic expertise. More and more methods according to the mitochondrial DNA (MtDNA) and nuclear fragments are raised, but the current methods of molecular identification have some defects, such as insufficient discrimination power, high cost, time-consuming and so on. In this study, the CO II sequences of sarcophagid flies were analyzed to establish the SNP-based genotyping system for the first time. Using this system by PSQ, we can identify the species of forensically important Sarcophgid flies with high discrimination power at a low cost in the short time. The SNPs marker system shows a new idea for genetic identification of forensic insects.

Body provides a home for diverse commensal microbiota. Microorganisms such as fungi have major roles in this microbial community stability. Postputrefaction fungi have been recorded in association with decomposed mammalian cadavers in disparate regions. The succession and diversity of these fungi are reviewed with a view to their potential as a forensic tool. The researches of mycology would be an interface to forensic investigation and may provide a means to estimate PMI within serious decomposition. To evaluate the use of succession and diversity of fungi species for PMI estimation, we investigate the Internal Transcribed Spacer (ITS) of fungi community on 3 points of time of rat carcasses conducted by Illumina MiSeq platform. Through high-throughput sequencing, several fungi genus such as Aspergillus , Ophiocordyceps are found in community succession. The community structure of each sample significantly differs in taxon richness and relative abundance patterns within decomposition progress. The succession and diversity of fungi community in corpse decomposition revealed by the indicator of ITS would be a potential forensic tool for PMI estimation.

The RF system of CYCIAE-100 cyclotron has two cavities, which are driven separately by two identical 100-kW RF amplifiers. Due to the power on sequence issue of the three DDSs in the LLRF systems, each time when the system is individually switched on, the phase relationship may not satisfy the requirements of beam acceleration. Instead of adding an extra reset logic to the system, a search and validation algorithm based on the decision tree has been carried out to make sure the phase of the two cavities is correct right after applying power to the cavities, taking advantage of existing hardware resources. In the first year of operation, there are more than 20 times of scheduled shutdown of the cyclotron system. For each time when the cyclotron RF system is completely shutdown and powered on again, the operator confirmed that the phase matching of the two cavities can be done automatically within 30 s. The related work, including the optimization of the phase detector and the development and validation of the algorithm, is reported in this paper.

Through the automatic depressurized system of AP1000 reactor, the steam with high temperature and high pressure from the pressurizer sprays into the water tank, and the pressure in the primary coolant system will diminish to protect the reactor. However, as the steam continues to flow into the tank, the water temperature rises rapidly until boiling occurs, which will affect the local heat transfer process. Therefore, it is necessary to understand the corresponding heat transfer mechanism by means of experiment. To ensure that the experimental results with a scale-down model reflect the actual situations of the prototype we analyzed the process of steam jetting from the pipeline into the water tank, and summarized scaling rules based on equation analysis method, including various flow stages. The results show that the phenomena-based similarity between the model and the prototype should meet: (1) geometrically similar model and prototype; (2) equal thermal parameters and identical initial conditions, which can greatly simplify other similarity parameters; (3) at the blowdown stage, keep the steam mass flux and the nozzle diameter consistent; (4) at the natural convection stage with single phase fluid, the equality in terms of Prandtl number and the Grashof number should be met first, while assessing the relative uncertainty.

The five-beam three-component laser Doppler velocimetry (LDV) is applied to investigate the turbulent flow in a 6 × 6 rod bundle installed with simple grid spacers. LDV measurement has been conducted at four cross sections downstream a grid spacer at five Reynolds numbers ranging from 6600 to 70,300. The flow evolution downstream of the grid spacer is demonstrated through the comparison of the axial mean velocity and Root Mean Square (RMS) velocity at the three cross sections downstream of the grid spacer. All the three components of the flow velocity are measured in the selected subchannels at the outlet cross section of the rod bundle which is dedicated to provide more information on the turbulence statistics in the rod bundle flow. Remarkably high ratio of axial normal stress to the turbulent kinetic energy, v v ¯ / k , is observed even in the subchannel center, which indicates that the turbulence in the rod bundle is anisotropic. Comparing experiment results at the five Reynolds numbers, the low Reynolds number effect is found in the case with Re = 6.6 × 103. The experiment results also imply that the Reynolds number effect in the tight-lattice bundle is weak compare to that in the loose one.

The five-beam three-component laser Doppler system is operated in the three-beam two-component mode to measure the 3 × 3 bundle flow with simple grid spacer. Experiment has been conducted at Re = 15,200 and 29,900. According to the experiment result, the root mean square (RMS) of axial velocity fluctuation shows large value in the gap and the near-wall region of the edge sub-channel which is induced by the axial velocity gradient. Significant intensity of lateral velocity fluctuation is observed which indicates the strong lateral mixing in a 3 × 3 rod bundle. Through the correlation analysis coherent structures have been observed in the gap region. The spectral analysis shows that the LDV measurement complies to the Komogorov spectrum law, f−5/3, well. The low-frequency peak spectral density of the axial velocity fluctuation has been observed in the gap region connecting sub-channels with velocity difference. The performance of the SSG model and the baseline Reynolds stress model are investigated based on the experiment result. The models predict higher axial velocity in the interior sub-channel and lower in the edge and corner ones than the experiment result. Large discrepancy between the calculated and measured axial flow velocity is resulted from failure in calculating the strong negative u ′ w ′ ¯ in the gap region connecting different sub-channels.

Nonbinary single-nucleotide polymorphisms (SNPs) are potential forensic genetic markers because their discrimination power is greater than that of normal binary SNPs, and that they can detect highly degraded samples. We previously developed a nonbinary SNP multiplex typing assay. In this study, we selected additional 20 nonbinary SNPs from the NCBI SNP database and verified them through pyrosequencing. These 20 nonbinary SNPs were analyzed using the fluorescent-labeled SNaPshot multiplex SNP typing method. The allele frequencies and genetic parameters of these 20 nonbinary SNPs were determined among 314 unrelated individuals from Han populations from China. The total power of discrimination was 0.9999999999994, and the cumulative probability of exclusion was 0.9986. Moreover, the result of the combination of this 20 nonbinary SNP assay with the 20 nonbinary SNP assay we previously developed demonstrated that the cumulative probability of exclusion of the 40 nonbinary SNPs was 0.999991 and that no significant linkage disequilibrium was observed in all 40 nonbinary SNPs. Thus, we concluded that this new system consisting of new 20 nonbinary SNPs could provide highly informative polymorphic data which would be further used in forensic application and would serve as a potentially valuable supplement to forensic DNA analysis.

Decomposition is a complex process involving the interaction of both biotic and abiotic factors. Microbes play a critical role in the process of carrion decomposition. In this study, we analysed bacterial communities from live rats and rat remains decomposed under natural conditions, or excluding sarcosaphagous insect interference, in China using Illumina MiSeq sequencing of 16S rRNA gene amplicons. A total of 1,394,842 high-quality sequences and 1,938 singleton operational taxonomic units were obtained. Bacterial communities showed notable variation in relative abundance and became more similar to each other across body sites during the decomposition process. As decomposition progressed, Proteobacteria (mostly Gammaproteobacteria) became the predominant phylum in both the buccal cavity and rectum, while Firmicutes and Bacteroidetes in the mouth and rectum, respectively, gradually decreased. In particular, the arrival and oviposition of sarcosaphagous insects had no obvious influence on bacterial taxa composition, but accelerated the loss of biomass. In contrast to the rectum, the microbial community structure in the buccal cavity of live rats differed considerably from that of rats immediately after death. Although this research indicates that bacterial communities can be used as a “microbial clock” for the estimation of post-mortem interval, further work is required to better understand this concept.
