This study sought to identify candidate biomarkers associated with gastric cancer (GC) prognosis based on an integrated bioinformatics analysis.First, the GSE54129 and GSE79973 data sets were downloaded from the Gene Expression Omnibus (GEO) database. The differentially expressed genes (DEGs) identified between the 2 data sets were screened using the limma software package in R, and the intersection DEGs were obtained by a Venn analysis. Subsequently, gene clustering and a functional analysis were performed to explore the roles of the DEGs. The protein-protein interaction (PPI) network of the genes in clusters was constructed using the Search Tool for the Retrieval of Interacting Genes/Proteins. A survival analysis evaluated the associations between the candidate genes and the overall survival of GC patients. A drug-gene interaction analysis and an external data set analysis were conducted using The Cancer Genome Atlas-Stomach Adenocarcinoma (TCGA-STAD) data set to validate the prognostic genes.We extracted 421 intersection DEGs from the 2 GEO data sets. The ever-increasing demands of very high precision components have forced manufacturers to improve equipment to meet the demand. The main factor is the identification and compensation of errors in machine tools. In this paper, a new real-time method is presented to identify and compensate the error with space gauge, by use of which the composition of errors is directly compensated by measuring the deformation of the points in machine bed. Then, a simple single-axis machine tool is used to determine the relationship of deformation between gauging point and control point to verify the feasibility of the method. Furthermore, due to the simplicity of the implement and, the method can be easily used in an industrial production with minimal investment. Based on the data of experiment, the coefficients of the model are determined. The model can be used to predict the residual life of specimen under biaxial compressive loading with constant confined stress.

In this study, a transcriptional repressor was cloned and characterized from the Chinese shrimp Fenneropenaeus chinensis (Osbeck, 1765) and named FcTR. The results indicate that the full-length cDNA of 1397 bp has an open reading frame encoding a polypeptide of 354 amino acids. The calculated molecular mass of the mature protein was found to be 39.9 kDa and theoretical pI 5.59. The FcTR has four ZnF-C2H2 domains and two putative transcriptional repressor regulating G2/M transition domains (SFP1). A phylogenetic analysis of transcriptional repressor sequences shows that FcTR has a high homology with the invertebrate transcriptional repressors from Megachile rotundata (Fabricius, 1793), Apis mellifera Linnaeus, 1758, Camponotus floridanus (Buckley, 1866), Acromyrmex octospinosus echinatior (Forel, 1899) and Apis florea Fabricius, 1787 (similarity 72%). A qRT-PCR analysis indicated that FcTR was expressed in intestines, hepatopancreas, muscles and gills, and its profile was increased significantly post WSSV (white spot syndrome virus) challenge followed by a decreased trend in all examined tissues. These results indicate that FcTR might be involved in the immune defence response to WSSV in F. chinensis as a negative regulator.

The low cycle loading tests of 3 pieces of recycled concrete latticed walls under different side column reinforcements and different axial pressure ratios are conducted. The failure mode, failure process, hysteresis curve, ductility, and energy dissipation of the walls are studied. The experiment results indicate that the failure mode of recycled concrete latticed wall under the cyclic load is shearing failure. It can enhance the ductility of the wall, distortion ability, and the earthquake resistance performance, with increasing the axial load ratio. It can enhance the ultimate load and restrain the crack development by increasing the reinforcement of the side columns.

A highly sensitive and specific TaqMan real-time PCR was used to quantify hepatopancreatic parvovirus (HPV) type III infections in wild broodstocks and hatchery-reared postlarvae (PL) of Fenneropenaeus chinensis. Totals of 159 and 162 wild brooders from three locations were captured, and 140 and 180 PL were obtained from seven and six commercial hatcheries in 2007 and 2008, respectively. Among the three wild broodstock groups from 2007, only 1 group showed HPV infection and 3.2% of 159 brooders were positive for HPV infection. In 2008, HPV infections were observed from all three wild broodstock groups with 1.93×10(4) copies/mg tissue of pleopods. Of 162 brooders, 26.6% were positive for HPV infection. No PL from the two hatcheries collected in 2007 showed HPV infection, and PL from the rest of the five hatcheries had up to 1.74×10(6) copies/ng of DNA, and PL from three hatcheries showed HPV infections with over 1,000 copies/ng of DNA. The PL from all seven hatcheries collected in 2008 showed up to 2.10×10(5) HPV copies/ng of DNA. PL from two hatcheries showed less than 100 copies/ng of DNA, but PL from the rest of the hatcheries showed HPV infections with over 1,000 copies/ng of DNA. These results show that HPV type III is widely distributed in Korea in addition to previously reported HPV type I, and they can be effectively detected by type-specific realtime PCR.

The results of prediction show that the suggested method is better than the Miner rule.

In the present study, we used TaqMan real-time polymerase chain reaction to quantify and compare infection of white spot syndrome virus (WSSV) with shrimp production of Fenneropenaeus chinensis cultured in outdoor ponds along the west coast of the South Korea. In 2007, a total of 60 specimens in summer and 116 specimens in autumn were collected from 12 growing-out ponds and 12 harvest ponds respectively. Pond harvest data were obtained from farmers. Of the summer samples, all specimens were WSSV positive, with a wide range of 12.4–7.0 × 107 (mean 7.5 × 106) copies ng−1 DNA; shrimp production was 1.7 metric tonnes per hectare (mt ha−1). Of the 116 autumn-sample specimens, 81 (69.8%) were WSSV positive; WSSV infection had been decreased dramatically, to 0–7.2 (mean 3.5) copies ng−1 DNA. Glutamine synthetase (GS) is considered to be one of the oldest existing functioning genes in evolution and plays a key role in two major biochemical pathways: in liver GS catalyzes ammonia detoxification, whereas in neural tissues it also functions in recycling of the neurotransmitter glutamate. In this study, a cytosolic glutamine synthetase was cloned and characterized from the Chinese Shrimp Fenneropenaeus chinensis, and named as FcGS. The results indicated that the full-length DNA of 1,756 bp had an open reading frame without intron, which encoded a polypeptide of 366 amino acids with several conserved active site residues. The calculated molecular mass of mature protein was 40.7 kDa and theoretical pI was 6.47. A phylogenetic analysis of GS sequences showed that FcGS clustered with the invertebrate group as expected. qRT-PCR analysis indicated that FcGS was expressed in intestines, hepatopancreas, muscles and gills, and its profile was up-regulated post-WSSV challenge in hepatopancreas and gills. Our results suggested that FcGS might participate in the regulation of shrimp immune response toward WSSV invasion. These data would be helpful to better understand the WSSV resistance mechanism of farming shrimps.

A TaqMan real-time PCR was conducted to quantify the white spot syndrome virus (WSSV) infections of three Litopenaeus vannamei hatcheries and 30 commercial grow-out ponds from different farms, during summer and autumn in South Korea. A total of 98.89% postlarvae showed WSSV-positive with very low WSSV infection load, 0.0–34.3 (mean of 3.2) copies/ng DNA, while all farm samples were WSSV-positive with a mean load of 9.8 × 10 6 and 2.1 × 10 5 copies/ng DNA in summer and autumn, respectively. In summer, nine (52.9%) farms showed WSSV infections with fewer than 10 3 copies/ng DNA and a mean shrimp production of 1.05 mt/ha; meanwhile, eight (47.1%) farms showed high infections (i.e., >10 3 copies/ng DNA) with a mean shrimp production of 0.85 mt/ha. In autumn, a total of eight (61.5%) farms showed infections with fewer than 10 3 copies/ng DNA, with mean shrimp production of 4.53 mt/ha; five (38.5%) farms showed high infection (i.e., >10 3 copies/ng DNA), with a mean shrimp production of 0.42 mt/ha. The blue crab, Portunus trituberculatus, is one of the most important fishery resources in the Yellow Sea of Korea, but the wild stock of this species has been reduced due to over-fishing and the destruction of the natural habitat in this area. Hatchery-produced seeds of blue crab have been released into the sea to enhance stock, but information on viral infection in the larvae as well as wild crabs of this species is very limited. In the present study, TaqMan real-time PCR was applied to quantify white spot syndrome virus (WSSV) in hatchery-produced larvae and wild populations of P. trituberculatus in South Korea. Out of 140 P. trituberculatus zoea from seven commercial hatcheries, 96.4% were WSSV-positive. The mean WSSV copies were 6.0 per ng DNA, or 3216.0 per larva. In 222 adult crabs from four wild populations captured in different seasons, the WSSV-positive rate was 79.3%, and the WSSV load was 5.2 copies per ng DNA, or 2116.5 copies per mg tissue. Both the WSSV-positive rate and the load of the winter population were significantly lower than those of the other three populations. Statistical analysis showed no significant correlations between WSSV infection loads and growth (CL, CW, and BW). The results suggest that low viral load of WSSV may not affect the growth of P. trituberculatus in a wild environment.

Microsatellite DNA markers were used to investigate the genetic diversity of Penaeus (Fenneropenaeus) chinensis, a topic of controversy in previous studies. A total of 210 individual shrimps were collected in seven geographic locations in the Yellow and Bohai Seas: Liaodong Bay (LD), Bohai Bay (BH), Haizhou Bay (HZ), Rushan Bay (RS), Haiyangdao fishing ground (HYD), the west coast (KW), and the south coast of the Korean Peninsula (KS). A new population of P. chinensis was identified in KS. A total of 109 alleles were identified from seven loci. Among the 49 population loci, 34 showed significant deviation from the Hardy–Weinberg equilibrium. Cluster analysis using UPGMA revealed a close genetic relationship of P. chinensis between BH and LD and between RS and HZ, while the shrimp from KW and KS diverged. Genetic distance and AMOVA analysis identified three areas with distinct populations: the Bohai and Yellow Seas along the Chinese coast; the west coast of Korea, and the south coast of Korea. These results are consistent with tagging–recapture data and some molecular DNA marker studies, but different from results by mitochondrial gene methods.

The WSSV infections in 3 broodstocks showed a wide range, from 0 to 2.28 × 10 6 (with a mean of 1.50 × 10 4 ) copies ng − 1 of DNA. Out of 159 brooders assayed, 39 (24.5%) were negative and 120 (75.5%) were positive; 153 (96.2%) showed less than 100 copies (mean 10.2 copies), 111 (69.8%) showed less than 10 copies and only 6 individuals (3.8%) showed high infections with a range of 2.36 × 10 2 to 2.28 × 10 6 copies ng − 1 of DNA. In 210 postlarvae, a range of 2.6 to 713.6 (with a mean of 220) copies g − 1 of DNA was observed. The mean WSSV copy number in the postlarvae was 7.9 × 10 5 , which was equivalent to 8.5 × 10 5 copies mg − 1 of postlarvae weight. A total of 87.1% of postlarvae were WSSV positive and except for two hatcheries (H 4 and H 7 ), the postlarvae of all the other five hatcheries were positive. Even postlarvae from the same hatchery, especially of hatcheries H 4 and H 5 , showed a wide range of WSSV infection resulting in higher infections than other hatcheries. There were 34.3% of the postlarvae assayed in the present study that showed very low infection, with less than 10 copies ng − 1 of DNA. Based on our results, it is recommended to pre-screen broodstock or larvae for selective breeding, stocking in production systems or stock enhancement.

Viral diseases are major emerging problems of shrimp that have affected the production, and even complete losses for shrimp farms. In this study, we developed a sensitive TaqMan real-time PCR method to quantify white spot syndrome virus (WSSV) and hepatopancreatic parvovirus (HPV) in the shrimp and pond water in which fleshy shrimp, Fenneropenaeus chinensis, and Pacific white shrimp, Litopenaeus vannamei, are reared. WSSV and HPV in pond seawaters ranged from to and from 0 to copies/L of seawater, respectively. Of 20 ponds analyzed, all pond water and shrimp were positive for WSSv. L. vannamei showed higher susceptibility to WSSV than F chinensis. HPV was detected only in the pond water for F chinensis. No infected snails and infested water were found.The measures of schistosomiasis control after the disaster are effective in Lushan County, and the goal to prevent major plague after the earth quake is achieved.

Primers were designed for eight microsatellite loci from Chinese shrimp Fenneropenaeus chinensis. Microsatellites were used to characterize three wild populations from the China coast of the Yellow and Bohai Seas (HB), and the west coast (KX) and south coast of the Korean Peninsula (KN). A total of sixty-one alleles were obtained, and the average observed heterozygosity ranged from 0.660 to 0.756. Six of the 24 population-locus cases showed a significant departure from the Hardy–Weinberg equilibrium, three of them from population KN, two from KX and one from HB. The Fst values indicated that genetic variation was greater within populations than between populations. Analysis using unweighted pair group method with arithmetic mean showed that the relationship between populations HB and KX was closer than between KN and the other two populations. Polymorphic information contents of the eight microsatellites ranged from 0.598 to 0.918. These results indicated that all eight microsatellite loci would be useful for the analysis of genetic variation in Chinese shrimp (F. chinensis) populations.

Fatigue tests were conducted on tapered plain concrete prism specimens under triaxial constant-amplitude tension-compression cyclic loading. The low stress of the cyclic loading was taken as 0.2fc and the upper stress ranged from 0.20ft, to 0.65ft. Three constant lateral pressures were 0. 1fc, 0.2fc and 0.3fc respectively. Based on the results, the three-stage evolution rule of the fatigue stiffness, maximum(minimum) longitudinal strain and damage were analyzed, and a unified S-N curve to calculate fatigue strength factors was worked out. The results show that the fatigue strength and fatigue life under triaxial constant-amplitude tension-compression cyclic loading are smaller than those under uniaxial fatigue condition. Moreover, the secondary strain creep rate is related to the fatigue life, a formula for describing their relation was derived. The investigation of this paper can provide information for the fatigue design of concrete structures.

To assess whether Beichuan County reaches the national standard of schistosomiasis transmission interruption.Three villages in Beichuan County were sampled as assessment spots, and the schistosomiasis infections of residents and domestic animals, the status of Oncomelania snails and the past records were assessed according to the Criteria for Control and Elimination of Schistosomiasis in China.No local schistosome infections were found in residents and cattle for successive 5 years and no snails were found for 2 successive years in the 3 villages. All the indices reached the national standard of schistosomiasis transmission being interrupted.Beichuan County has reached the national standard of schistosomiasis transmission interruption. However, the endemic surveillance of schistosomiasis still needs to continue.

To understand the epidemic trend of schistosomiasis in Sichuan Province so as to provide the evidence for formulating schistosomiasis control strategy.According to the National Schistosomiasis Surveillance Protocol, the national surveillance sites in Sichuan Province were selected. The schistosomiasis surveillance was carried out continuously from 2005 to 2010.Nine national schistosomiasis surveillance sites were established in Pujiang, Guanghan, Zhongjiang, Fucheng, Dongpo, Danling, Renshou, Xichang and Dechang counties. The Oncomelania hupensis snail area decreased from 351 853 m2 in 2005 to 128 285 m2 in 2010, the snail density from 0.70 to 0.21 per 0.1 m2, the snail infection rate from 0.06% to 0, the positive rate of human serum schistosome antibody from 19.41% to 7.62%, the schistosome infection rate of human population from 1.93% to 0.10%, and the infection rate of livestock from 4.50% to 1.02%. The snails were found mainly in ditch, rice field and other moist field.Though the endemic of schistosomiasis has reached a low level in Sichuan Province, the endemic situation fluctuates at a narrow range in some surveillance sites. Therefore, the surveillance work should be carried out continuously.

To observe the compliance of film-coated praziquantel tablets in the schistosomiasis transmission controlled areas, so as to provide the evidence for the establishment of chemotherapy intervention measures in these areas.In the areas of schistosomiasis transmission controlled in Sichuan Province, 234 people were selected as film-coated praziquantel group (FCPG), and 203 people were selected as praziquantel group (PG). A questionnaire survey was implemented and the compliance of chemotherapy of the 2 groups was compared.In PG, all the people indicated that the praziquantel tablets has bad smell, 98.0% of the people had side-effects, such as nausea, headache, 69.5% thought the medicine was harmful to health subjectively, and 99.5% thought that it was too much dose of tablets for taking one time. In FCPG, all the people indicated that the film-coated praziquantel tablets had no bad smell, 18.8% of the people had side-effects, and 74.4% would like to choose film-coated praziquantel tablets. The rates of knowledge of praziquantel of PG and FCPG were 50.7% and 29.1%, respectively ( chi2 = 21.449, P = 0.01).The film-coated praziquantel tablets have no bad smell and lighter side-effects. The compliance of the film-coated praziquantel tablets is high in the areas of schistosomiasis transmission controlled, so the film-coated praziquantel tablets are worth to popularize and apply.

BACKGROUND/AIMS We analyzed the clinicopathological factors of patients with node-positive gastric cancer, evaluated the prognostic factors associated with long-term survival and clarified the effect of tumor size on long-term survival. METHODOLOGY The study included 591 patients who underwent curative resection for node-positive gastric cancer. Clinicopathological prognostic variables were evaluated as predictors of long-term survival by univariate and multivariate analyses. RESULTS The 5-year survival rate was influenced by tumor size, tumor location, depth on invasion, level of lymph node metastasis, Borrmann classification, histological type, liver metastasis, peritoneal dissemination and disease stage. Of these, independent prognostic factors were depth on invasion and lymph node metastasis. Tumor size is an influence but not independent factor for the prediction of long-term survival in patients with node-positive gastric cancer. CONCLUSIONS In patients with node-positive gastric cancer, two independent prognostic factors were depth on invasion and the status of lymph node metastasis.

Field experiments were conducted in southwestern China to estimate the distances over which cercariae of Schistosoma japonicum are transported and remain infective in village irrigation ditches and to determine the feasibility of interrupting this transport using niclosamide, the molluscicide commonly used for snail control in this environment. The number of worms recovered from bioassay mice downstream from experimentally introduced cercariae, but without niclosamide, decreased about 90% over the first 100 m of ditch and then remained essentially constant for the remaining 300 m of the ditch that was monitored. The niclosamide experiments resulted first in the finding that it was possible to achieve ditch average concentrations close to the target values of 0.25 ppm, but that concentrations at any particular location varied by a factor of up to 3. There was no correlation between water turbidity and niclosamide concentration. An average water concentration of 0.29 ppm resulted in no viable cercariae after a contact time of about 1 min and a travel distance of only 10 m. We regard these results as proof of the feasibility of chemical interruption of transport of S. japonicum cercariae although the toxicity of niclosamide to fish at the levels used here make it a questionable candidate for routine use.
