Evidence that exendin-4, a glucagon-like peptide-1 analog, might be used to treat poorly healing wounds under diabetic and nondiabetic conditions has gained increasing interest. Little is known, however, about the effects of the drug on the production by dermal fibroblasts of key extracellular matrix and regulatory compounds. Therefore, we used human skin fibroblasts cultured in normo- (1 g/l = 5.6 mmol/l glucose) or hyperglycemic (4.5 g/l = 25 mmol/l glucose) culture medium to test the effects of exendin-4 (0 – 100 nmol/l) on fibroblast functions crucial for the wound healing process. Exendin-4 increased the proliferative and metabolic activities, as measured by the BrdU (bromodeoxyuridine) and MTT (3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide) assays, respectively, of fibroblasts cultured in normoglycemic medium. Under hyperglycemic conditions, the drug had no effect on proliferation and reduced metabolic fibroblast activity. Exendin-4 decreased metalloproteinase-9 (MMP-9) secretion in the normoglycemic milieu only and increased tissue inhibitor of metalloproteinase-1 (TIMP-1) concentration in fibroblast colonies under both normo- and hyperglycemic experimental conditions. Exendin-4 increased the fibroblast growth factor-1 (FGF-1) concentration in cell colonies maintained in the normoglycemic milieu but decreased FGF-1 release when fibroblasts were grown in hyperglycemic medium. High glucose caused lactic dehydrogenase (LDH) leakage when compared with normoglycemic conditions, and exendin-4 was not able to prevent this effect, although it reduced LDH release from fibroblasts cultured in normoglycemic medium. Finally, exendin-4 increased glycosaminoglycan (GAG) content under both experimental conditions. Our results indicate that exendin-4 effects on the production of the extracellular matrix and regulatory proteins differ in human skin fibroblasts exposed to either normal or high glucose. In general, the beneficial effects of the drug, which may be important for the improvement of wound healing, are more pronounced under normoglycemic conditions, thus indicating that hyperglycemia attenuates the positive effects of exendin-4 on fibroblasts. The real part of the relative permittivity, ϵ', and dielectric loss, tan δ, have been determined at audio frequencies at temperatures from about 5K to 350K for nano-composites composed of BaTiO3 nanoparticles and polycarbonate. The room temperature breakdown strength was also measured and thermal analysis, nuclear magnetic resonance and scanning electron microscopy studies were carried out. For some films the nanoparticles were surface-treated (STNP) while for others they were not (UNP). For concentrations of UNP greater than about 3.4 vol. %, ϵ' is much larger than expected on the basis of laws of mixing. On the other hand, ϵ' for materials made using STNP is well-behaved. Correspondingly, increased loss (ϵ" or tan δ) in the vicinity of room temperature is observed for the materials made from UNP. The anomalously large values of relative permittivity and increased loss are attributed to the presence of large aggregates in the materials made using the UNP. For both UNP-and STNP-based materials, the breakdown strength is found to decrease as nanoparticle concentration increases. The breakdown strength for the materials made using STNP is found to be larger for all concentrations than for those containing UNP despite the presence of large aggregates in some of the STNP-based materials. This shows that breakdown is strongly affected by the nanoparticle surfaces and/or the interface layer. It is also found that the breakdown strength for materials made using UNP increases as particle size increases. Finally, variable temperature and pressure proton nuclear magnetic resonance relaxation measurements were made to assess the effect of nanoparticle inclusion on polymer motion, and the effects were found to be very minor. The work function of nano-crystalline zinc oxide (ZnO) thin films was examined using photoemission spectroscopy (PES). Colloidally dispersed ZnO nano-particles were electrospray-deposited in vacuum to form nano-crystalline thin films. The samples showed an immediate work function reduction by 0.35 eV during ultraviolet photoemission spectroscopy (UPS) measurements. This artifact was detected and quantified through low intensity x-ray photoemission spectroscopy (LIXPS) measurements, which use a very low photon flux. This prevented significant photochemical changes on the measured surface, i.e. the true work function unaffected by the UPS artifact can be measured. Annealing of an identical sample removed all ambient contamination from the ZnO surface with the effect to prevent the work function lowering artifact. This allowed the conclusion that ambient contamination is essential for the artifact to occur, similar to what was observed earlier on indium tin oxide and TiO2 surfaces. In an additional experiment, exposure of the annealed sample to the ambient resulted in a sample that again showed the artifact further demonstrating the necessity of water to be present. This experiment also demonstrated that the solubility enhancing surfactant shell of the nano-crystals does not play a significant role in the artifact, since it was removed during the annealing process. The work functions of nanocrystalline anatase (TiO2) thin films and a rutile single crystal were measured using photoemission spectroscopy (PES). The nanocrystalline titanium dioxide films were deposited in-vacuum using electrospray thin film deposition. A comparison between ultraviolet photoemission spectroscopy (UPS) and low intensity x-ray photoemission spectroscopy (LIXPS) work function measurements on these samples revealed a strong, immediate, and permanent work function reduction (>0.5 eV) caused by the UPS measurements. Furthermore, it was found that regular XPS measurements also reduce the work function after exposure times ranging from seconds to minutes. These effects are similar in magnitude to artifacts seen previously on indium tin oxide (ITO) substrates characterized with XPS and UPS, and are likely related to the formation of a surface dipole through the photochemical hydroxylation of oxygen vacancies present on the TiO2 surface. Hot-Electron Bolometer (HEB) mixers have proven to be the best tool for high-resolution spectroscopy at the Terahertz frequencies. However, the current state of the art NbN mixers suffer from a small intermediate frequency (IF) bandwidth as well as a low operating temperature. \MgB2 is a promising material for HEB mixer technology in view of its high critical temperature and fast thermal relaxation allowing for a large IF bandwidth. In this work, we have fabricated and characterized thin-film ( $\sim$15 nm) <$ 4000 K at 9 K using a 600 GHz source. Using temperature dependencies of the radiation power dissipated in the device we have identified the optical loss in the integrated microantenna responsible as a cause of the limited sensitivity of the current mixer devices. From the analysis of the current-voltage (IV) characteristics, we have derived the effective thermal conductance of the mixer device and estimated the required local oscillator power in an optimized device to be \sim\!\! 1\ \mu\W. For potential applications in superconducting RF cavities, we have investigated the properties of polycrystalline MgB2 films, including the thickness dependence of the lower critical field Hc1. MgB2 thin films were fabricated by hybrid physical-chemical vapor deposition on (0001) SiC substrate either directly (for epitaxial films) or with a MgO buffer layer (for polycrystalline films). When the film thickness decreased from 300 nm to 100 nm, Hc1 at 5 K increased from around 600 Oe to 1880 Oe in epitaxial films and to 1520 Oe in polycrystalline films. Lamotrigine (LTG) [3,5-diamino-6-(2,3-dichlorophenyl)-1,2,4-triazine], an anticonvulsant and antidepressant drug Lamictal®, produces a (photo)toxic response in some patients. LTG absorbs UV light, generating singlet oxygen (1O2) with a quantum yield of 0.22 in CH2Cl2, 0.11 in MeCN and 0.01 in D2O. A small production of superoxide radical anion was also detected in acetonitrile. Thus, LTG is a moderate photosensitizer producing phototoxicity and oxidizing linoleic acid. LTG is a weak 1O2 quencher ( kq = 3.2 × 105 m−1 s−1 in MeCN), but its photodecomposition products in dimethyl sulfoxide (DMSO) quenched 1O2 very efficiently. Upon intense UV irradiation from a xenon lamp, LTG was photobleached rapidly in DMSO and slowly in acetonitrile, alcohol and water. The rate increased significantly when laser pulses at 266 nm were employed. The photobleaching products generated 1O2 twice as strongly as LTG. Photobleaching was usually accompanied by the release of chloride anions, which increased in the presence of ascorbic acid. This suggests the formation of aryl radicals via dechlorination, a process which may be responsible for the photoallergic response observed in some patients. Our results demonstrate that LTG is a moderate generator of 1O2 prone to photodechlorination, especially in a reducing environment, which can contribute to the reported phototoxicity of LTG.
