Abstract Lumbar hernia is a rare lateral abdominal wall hernia. Various surgical repair strategies have been recorded, but there is currently no unified standard. A Chinese surgeon recently revealed a novel technique for treating primary lumbar hernia called retroperitoneal totally endoscopic prosthetic repair (R-TEP). We have made a further exploration of this method and successfully used it in the treatment of secondary lumbar hernia. We successfully performed R-TEP on three patients with lumbar hernias. All patients were female with an average age of 64 years (51–71 years). Two patients each had a primary upper lumbar hernia, while one patient had a secondary lumbar hernia. With a mean operative time of 77 min (60–105 min), all operations were performed successfully. The average visual analogue scale (VAS) was 1.3 points (1–2 points) on the second day following surgery. The mean postoperative hospital stay was 2.3 days (2–3 days). No postoperative complications occurred. During a mean follow-up period of 19 months (10–24 months), there was no recurrence of the hernia, chronic pain or mesh infection. Therefore, R-TEP is safe and effective for both primary and secondary lumbar hernia. Anti-adhesive coated meshes are not required, making this a cost-effective procedure that is worthy of recommendation.

Abstract Background The issue of whether or not splenic hilum lymph nodes (SHLN) should be excised in radical gastrectomy with D2 lymph node dissection remains controversial. In this study, we identified the clinicopathological features in patients with gastric cancer that could serve as predictive risk factors of SHLN metastasis. Methods We searched Medline, Embase, PubMed, and Web of Science databases from inception to May 2020 and consulted the related references. Overall, 15 articles evaluating a total of 4377 patients were included for study. The odds ratios (OR) of each risk factor and corresponding 95% confidence intervals (CI) were determined using the Revman 5.3 software. Results Our meta-analysis revealed tumor size greater than 5 cm (p < 0.01), tumor localization in the greater curvature (p < 0.01), diffuse type (Lauren’s classification) (p < 0.01), Borrmann types 3–4 (p < 0.01), poor differentiation and undifferentiation (p < 0.01), depth of invasion T3–T4 (p < 0.01), number of lymph node metastases N2–N3 (p < 0.01), distant metastasis M1 (p < 0.01), TNM stages 3–4 (p < 0.01), vascular invasion (p = 0.01), and lymphatic invasion (p < 0.01) as potential risk factors of SHLN metastasis. Moreover, positivity of Nos. 1, 2, 3, 4sa, 4sb, 4d, 6, 7, 9, 11, and 16 lymph nodes for metastasis was strongly associated with SHLN metastasis. Conclusions Tumor size, tumor location, Lauren’s diffuse type, Borrmann type, degree of differentiation, T stage, N stage, M stage, TNM stage, vascular invasion, lymphatic infiltration, and other positive lymph nodes are risk factors for SHLN metastasis.

Abstract Temporal transcriptional variation is a major contributor to functional evolution and the developmental process. Parthenogenetic water fleas of the genus Daphnia (Cladocera) provide an ideal model to characterize gene expression patterns across distinct developmental stages. Herein, we report RNA-seq data for female Daphnia mitsukuri at three developmental stages: the embryo, juvenile (three timepoints) and adult. Comparisons of gene expression patterns among these three developmental stages and weighted gene co-expression network analysis based on expression data across developmental stages identified sets of genes underpinning each of the developmental stages of D. mitsukuri. Specifically, highly expressed genes (HEGs) at the embryonic developmental stage were associated with cell proliferation, ensuring the necessary foundation for subsequent development; HEGs at the juvenile stages were associated with chemosensory perception, visual perception and neurotransmission, allowing individuals to enhance detection of potential environmental risks; HEGs at the adult stage were associated with antioxidative defensive systems, enabling adults to mount an efficient response to perceived environmental risks. Additionally, we found a significant overlap between expanded gene families of Daphnia species and HEGs at the juvenile stages, and these genes were associated with visual perception and neurotransmission. Our work provides a resource of developmental transcriptomes, and comparative analyses that characterize gene expression dynamics throughout development of Daphnia.

Abstract The characterization of phospholipid membrane permeability for small molecules is crucial for many applications in drug discovery and biomedical research in general. Here, chemosensor‐laden vesicles offer an attractive platform for permeability assays. In this work, the stability of immobilized chemosensor‐filled vesicles is explored on anti‐fouling polymer brush surfaces for potential use in monitoring small molecule membrane permeability. The study focuses on the development of a method for immobilizing sensor‐loaded vesicles into arbitrary patterns on surfaces and characterizing their stability under changing temperatures and compositions. As substrate to enable intact, long‐term stable vesicle immobilization reactive polymer brushes with anti‐fouling properties are used. Utilizing microchannel cantilever spotting, biotin moieties are introduced on the polymer brush surface, enabling stable tethering of vesicles through streptavidin‐biotin interactions. The immobilized vesicles are monitored through fluorescence microscopy for their response to analytes under changing environmental parameters and vesicle composition. A higher stability of immobilized vesicles compared to free‐floating ones is observed, with permeability only at elevated temperatures. By tuning vesicle compositions, permeability at lower temperatures can be raised again. Overall, the study provides insights into a novel approach for vesicle immobilization, showcasing the potential of surface‐bound vesicles for applications in microfluidic systems and as biosensors in various assays.

Geospatial objects pose significant challenges, including dense distribution, substantial interclass variations, and minimal intraclass variations. These complexities make achieving precise foreground object segmentation in high-resolution remote sensing images highly challenging. Current segmentation approaches often rely on the standard encoder–decoder architecture to extract object-related information, but overlook the inherent uncertainty issues that arise during the process. In this article, we aim to enhance segmentation by introducing an uncertainty-guided decoding mechanism and propose the uncertainty-guided segmentation network (UGSNet). Specifically, building upon the conventional encoder–decoder architecture, we initially employ the pyramid vision transformer to extract multilevel features containing extensive long-range information. We then introduce an uncertainty-guided decoding mechanism, addressing both epistemic and aleatoric uncertainties, to progressively refine segmentation with higher certainty at each level. With this uncertainty-guided decoding mechanism, our UGSNet achieves accurate geospatial object segmentation. To validate the effectiveness of UGSNet, we conduct extensive experiments on the large-scale ISAID dataset, and the results unequivocally demonstrate the superiority of our method over other state-of-the-art segmentation methods.

Abstract Background DFR is a crucial structural gene in plant flavonoid and polyphenol metabolism, and DFR knockout (DFR-KO) plants may have increased biomass accumulation. It is uncertain whether DFR-KO has comparable effects in tobacco and what the molecular mechanism is. We employed the CRISPR/Cas9 method to generate a knockout homozygous construct and collected samples from various developmental phases for transcriptome and metabolome detection and analysis. Results DFR-KO turned tobacco blossoms white on homozygous tobacco (Nicotiana tabacum) plants with both NtDFR1 and NtDFR2 knockout. RNA-seq investigation of anthesis leaf (LF), anthesis flower (FF), mature leaf (LM), and mature root (RM) variations in wild-type (CK) and DFR-KO lines revealed 2898, 276, 311, and 101 differentially expressed genes (DEGs), respectively. DFR-KO primarily affected leaves during anthesis. According to KEGG and GSEA studies, DFR-KO lines upregulated photosynthetic pathway carbon fixation and downregulated photosystem I and II genes. DFR-KO may diminish tobacco anthesis leaf photosynthetic light reaction but boost dark reaction carbon fixation. DFR-KO lowered the expression of pathway-related genes in LF, such as oxidative phosphorylation and proteasome, while boosting those in the plant–pathogen interaction and MAPK signaling pathways, indicating that it may increase biological stress resistance. DFR-KO greatly boosted the expression of other structural genes involved in phenylpropanoid production in FF, which may account for metabolite accumulation. The metabolome showed that LF overexpressed 8 flavonoid metabolites and FF downregulated 24 flavone metabolites. In DFR-KO LF, proteasome-related genes downregulated 16 amino acid metabolites and reduced free amino acids. Furthermore, the DEG analysis on LM revealed that the impact of DFR-KO on tobacco growth may progressively diminish with time. Conclusion The broad impact of DFR-KO on different phases and organs of tobacco development was thoroughly and methodically investigated in this research. DFR-KO decreased catabolism and photosynthetic light reactions in leaves during the flowering stage while increasing carbon fixation and disease resistance pathways. However, the impact of DFR-KO on tobacco growth steadily declined as it grew and matured, and transcriptional and metabolic modifications were consistent. This work offers a fresh insight and theoretical foundation for tobacco breeding and the development of gene-edited strains.

BackgroundBabesia is a unique apicomplexan parasite that specifically invades and proliferates in red blood cells and can be transmitted via blood transfusion, resulting in transfusion-transmitted babesiosis. However, detecting Babesia in blood before transfusion has not received enough attention, and the risk of transfusing blood containing a low density of Babesia microti (B. microti) is unclear, possibly threatening public health and wellness.PurposeThis study aimed to determine the lower detection limit of B. microti in blood and to evaluate the transmission risk of blood transfusion containing low-density B. microti.MethodsInfected BALB/c mouse models were established by transfusing infected whole blood with different infection rates and densities of B. microti. Microscopic examination, nested Polymerase Chain Reaction (nested PCR), and an enzyme-linked immunosorbent assay (ELISA) were used to evaluate the infection status of the mouse models. Meanwhile, the nested PCR detection limit of B. microti was obtained using pure B. microti DNA samples with serial concentrations and whole blood samples with different densities of B. microti-infected red blood cells. Thereafter, whole mouse blood with a B. microti density lower than that of the nested PCR detection limit and human blood samples infected with B. microti were transfused into healthy mice to assess the transmission risk in mouse models. The infection status of these mice was evaluated through microscopic examination, nested PCR tests, and ELISA.ResultsThe mice inoculated with different densities of B. microti reached the peak infection rate on different days. Overall, the higher the blood B. microti density was, the earlier the peak infection rate was reached. The levels of specific antibodies against B. microti in the blood of the infected mice increased sharply during the first 30 days of infection, reaching a peak level at 60 days post-infection, and maintaining a high level thereafter. The nested PCR detection limits of B. microti DNA and parasite density were 3 fg and 5.48 parasites/μL, respectively. The whole blood containing an extremely low density of B. microti and human blood samples infected with B. microti could infect mice, confirming the transmission risk of transfusing blood with low-density B. microti.ConclusionWhole blood containing extremely low density of B. microti poses a high transmission risk when transfused between mice and mice or human and mice, suggesting that Babesia detection should be considered by governments, hospitals, and disease prevention and control centers as a mandatory test before blood donation or transfusion.

Flowering time, plays a crucial role in tobacco ecological adaptation besides its substantial influence on tobacco production and leaf quality. Meanwhile, it is sensitive to biotic or abiotic challenges. The plant hormones Gibberellins (GAs), controlling a number of metabolic processes, govern plants growth and development. In this study, we created a late flowering mutant HG14 through knocking out NtGA3ox1 by CRISPR/Cas9. It took around 13.0 and 12.1 days longer to budding and flowering compared to wild type Honghuadajinyuan. Nearly all of the evaluated agronomic characters deteriorated in HG14, showing slower growth and noticeably shorter and narrower leaves. We found that NtGA3ox was more prevalent in flowers through quantitative reverse transcription PCR analysis. Transcriptome profiling detected 4449, 2147, and 4567 differently expressed genes at the budding, flowering, and mature stages, respectively. The KEGG pathway enrichment analysis identified the plant-pathogen interaction, plant hormone signal transduction pathway, and MAPK signaling pathway are the major clusters controlled by NtGA3ox1 throughout the budding and flowering stages. Together with the abovementioned signaling pathway, biosynthesis of monobactam, metabolism of carbon, pentose, starch, and sucrose were enriched at the mature stage. Interestingly, 108 up- and 73 down- regulated DEGs, impairing sugar metabolism, diterpenoid biosynthesis, linoleic and alpha-linolenic acid metabolism pathway, were continuously detected accompanied with the development of HG14. This was further evidenced by the decreasing content of GA metabolites such as GA4 and GA7, routine chemicals, alkaloids, amino acids, and organic acids Therefore, we discovered a novel tobacco flowering time gene NtGA3ox1 and resolved its regulatory network, which will be beneficial to the improvement of tobacco varieties.

The double-hole rice vacuum seed meter is critical equipment for the planting precision of rice direct seeding. The effects of shaped holes and seed disturbance on the precision of rice bunch planting were investigated to improve the precision of bunch planting with the double-hole rice vacuum seed meter. A test bench with the rice vacuum seed meter was set up to analyze the trends in the quality of feed index, miss index, and multiple index of seed meters with different shaped holes at different speeds and vacuum pressures. Based on the optimal hole structure, different seed disturbance structures were designed to investigate the influence of the seed disturbance structure on the precision of bunch planting. A multiple linear regression model was established for the relationship between the disturbance structure, vacuum pressure, rotational speed, and the precision of bunch planting. Discrete element numerical simulation experiments were carried out to analyze the effect of disturbance structures on seeds. The planting precision of the seed meter with the shaped hole was significantly higher than that of the seed meter without the shaped hole while the shaped hole B was the optimum structure. Disturbance structure affects the quality of feed index, multiple index rate, and miss index. The planting precision of the seed disturbance structure II was better than the other structures. At a speed of 60 rpm and vacuum pressures of 2.0 kPa, 2.4 kPa, and 2.8 kPa, the qualities of feed index of seed disturbance structure II were 90%, 91.11%, and 89.17%, respectively, and the miss indexes were 2.96%, 1.94%, and 1.57%, respectively. At high rotational speeds, the precision of rice bunch planting with the seed disturbance structure is better than that without the seed disturbance structure. In the simulation test, the seed velocity and total force magnitude of the meter without disturbance structures were less than those with the disturbed structure. Simulation experiments showed that the seed disturbance structure breaks up the stacked state of seeds. Research has shown that the shaped hole holds the seed in a stable suction posture, which helps to increase the seed-filling rate. Seed disturbance improves seed mobility, thereby enhancing the precision of bunch planting.

This investigation considered the effects of both internal and external excitation vibrations on the efficacy of the seed dispenser in a rice precision hole seeder. Through comprehensive field tests, we analyzed vibrational characteristics during direct seeder operations and established a vibration seeding test bed for systematic examination of these effects. Time-domain analysis of the vibration data revealed a predominantly vertical vibration direction, with notably higher levels in sandy loam soil compared to clay loam. A correlation was observed between increased engine size and rotary ploughing speeds, as well as forward speed and elevated vibration amplitudes. Frequency domain analysis pinpointed the primary vibration frequency of the machinery within the 0–170 Hz range, remaining consistent across different operating conditions. Crucially, bench test results indicated that seeding accuracy and dispersion were significantly influenced by vibration frequencies, particularly within the 70–130 Hz range, where a decrease in accuracy and increase in dispersion were noted. A regression model suggested a complex, non-linear relationship between seeding performance and vibration frequency. These insights highlight the necessity for a robust mechanism to effectively address these vibrational impacts. This study paves the way for enhancing the operational efficiency of the rice precision hole seeder, aiming to achieve the design goals of minimized vibrations in the paddy power chassis.

Due to factors such as a small amount of fertilizer application during rice topdressing and slow machine speed, the ordinary fertilizer discharge device fails to distribute the fertilizer uniformly and accurately as required, making it difficult to meet the needs of precise rice topdressing. This research focuses on the development of an offset spiral tooth fertilizer discharge device suitable for rice topdressing. The study analyzes the amount of fertilizer discharged in one cycle, the fertilizer force, and the motion of the fertilizer particles. In order to enhance the uniformity of the fertilizer discharge device at a low speed and small volume, the discrete element method is employed to conduct experimental research on the key structural parameters that affect the one-cycle amount of discharged fertilizer and the dynamics of the fertilizer discharge device. Through single-factor tests, it was found that the angle, height, number of spiral teeth, and diameter of the fertilizer discharge wheel are closely associated with the fertilizer discharge performance. To further investigate the impact of these four parameters on the fertilizer discharge performance, a regression combination test of the four factors is conducted based on the range optimized by the single-factor tests. A multi-objective mathematical model, considering the key parameters of fertilizer uniformity coefficient, one-cycle amount of fertilizer, and fertilizer discharge torque, is established at three speeds: 20, 55, and 90 rpm. The response surface method is utilized to analyze the influence of the interaction factors on the fertilizer discharge performance. The optimal combination of the key structural parameters was determined as follows: spiral tooth angle of 35.42°, tooth height of 9.02 mm, discharge wheel diameter of 57.43 mm, and tooth amount of 9.37. The bench test results of the device, using the optimal parameter combination and a fertilizer discharge speed of 0–90 rpm, were obtained for four commonly used rice fertilizers. The maximum variation coefficient of fertilizer discharge was found to be 10.42%. The one-cycle amount of fertilizer discharge was measured to be 19.82 ± 0.72 (A Kang), 17.20 ± 0.69 (Ba Tian), 20.34 ± 0.54 (Yaran), and 14.51 ± 0.44 (granular urea). The fertilizer discharge torque remained stable. These results indicate that the achieved optimization meets the precise fertilizer application requirements and can provide technical support for precise topdressing operations.

2-acetyl-1-pyrroline (2AP) is a highly effective volatile compound that gives fragrance to numerous plant species and food. Mutation(s) in the betaine aldehyde dehydrogenase 2 (BADH2) gene results in the accumulation of 2AP. However, the function of BADH genes in tobacco (Nicotiana tabacum L.) remains poorly understood. In this study, we successfully obtained four betaine aldehyde dehydrogenase (BADH) genes from tobacco. Phylogenetic analysis of the protein sequences showed that two of the four BADH genes were closely related to the wolfberry (Lycium barbarum) BADH gene (LbBADH1), so we named them NtBADH1a and NtBADH1b, respectively. The other two BADH genes were orthologues of the tomato (Solanum lycopersicum) aminoaldehyde dehydrogenase 2 (SlAMADH2) gene, and were named NtBADH2a and NtBADH2b, respectively. Expression analysis revealed that the biological functions of NtBADH1a and NtBADH1b were different from those of genes NtBADH2a and NtBADH2b. We introduced mutations into NtBADH1a, NtBADH1b, NtBADH2a and NtBADH2b in tobacco using the CRISPR/Cas9 system and identified transgenic Ntbadh mutant tobacco lines. Single mutants (Ntbadh1a, Ntbadh1b, Ntbadh2a and Ntbadh2b) and double mutants (Ntbadh1a-Ntbadh1b and Ntbadh2a-Ntbadh2b) harbored deletion or insertion of nucleotides, both of which led to the production of a frameshift, preventing protein accumulation. A popcorn-like scent was noticeable in tobacco leaves from the Ntbadh2a-Ntbadh2b double mutant, but not from any single mutant or the Ntbadh1a-Ntbadh1b double mutant or the wild type. Consistent with this observation, we only detected 2AP in fresh leaves from the Ntbadh2a-Ntbadh2b double mutant. These findings indicate that only the combined inactivation of NtBADH2a and NtBADH2b results in 2AP accumulation in tobacco, which was not related to NtBADH1.

The parameters of the discrete element simulation model for rice field soils serve as valuable data references for investigating the dynamic characteristics of the walking wheel of high-speed precision seeding machinery in paddy fields. The research specifically targets clay loam soil from a paddy field in South China. Calibration of essential soil parameters was achieved using EDEM_2022 software (and subsequent versions) discrete element simulation software, employing the Edinburgh Elasto-Plastic Adhesion (EEPA) nonlinear elastic-plastic contact model. The tillage layer and plough sub-base layer underwent calibration through slump and uniaxial compression tests, respectively. Influential contact parameters affecting slump and axial pressure were identified through a Plackett–Burman test. The optimal contact parameter combinations for the discrete element model of the tillage layer and plough sub-base layer were determined via a quadratic rotational orthogonal test. The accuracy of the discrete element simulation model’s parameters for paddy field soils was further validated through a comparative analysis of the simulation test’s cone penetration and the field soil trench test. Results indicate that the Coefficient of Restitution, surface energy, Contact Plasticity Ratio, and Tensile Exp significantly influence slump (p < 0.05). Additionally, the Coefficient of Restitution, Contact Plasticity Ratio, coefficient of rolling friction, and Tangential Stiff Multiplier significantly impact axial pressure (p < 0.05). Optimal contact parameters for the plough layer were achieved with a particle recovery coefficient of 0.49, a surface energy of 18.52 J/m2, a plastic deformation ratio of 0.45, and a tensile strength of 3.74. For the plough subsoil layer, optimal contact parameters were a particle recovery coefficient of 0.47, a coefficient of interparticle kinetic friction of 0.32, a plastic deformation ratio of 0.49, and a tangential stiffness factor of 0.31. Results from the cone penetration test reveal no significant disparity in compactness between the actual experiment and the simulation test. The calibrated discrete element model’s contact parameters have been verified as accurate and reliable. The findings of this study offer valuable data references for understanding the dynamic characteristics of the walking wheel of the entire machinery in high-speed precision seeding in paddy fields.

Tobacco belongs to the family Solanaceae, which easily forms continuous cropping obstacles. Continuous cropping exacerbates the accumulation of autotoxins in tobacco rhizospheric soil, affects the normal metabolism and growth of plants, changes soil microecology, and severely reduces the yield and quality of tobacco. In this study, the types and composition of tobacco autotoxins under continuous cropping systems are summarized, and a model is proposed, suggesting that autotoxins can cause toxicity to tobacco plants at the cell level, plant-growth level, and physiological process level, negatively affecting soil microbial life activities, population number, and community structure and disrupting soil microecology. A combined strategy for managing tobacco autotoxicity is proposed based on the breeding of superior varieties, and this approach can be combined with adjustments to cropping systems, the induction of plant immunity, and the optimization of cultivation and biological control measures. Additionally, future research directions are suggested and challenges associated with autotoxicity are provided. This study aims to serve as a reference and provide inspirations needed to develop green and sustainable strategies and alleviate the continuous cropping obstacles of tobacco. It also acts as a reference for resolving continuous cropping challenges in other crops.

Hybridization of surface‐bound DNA with complementary strands is the basis of many biotechnological applications. Herein, the structure of interfacial coatings between substrate and bound DNA is a crucial element for hybridization behavior. Herein, three reactive surfaces for constructing DNA‐sensing platforms, namely, plain gold films on silicon, poly(bisphenolA‐co‐epichlorohydrin) (PBAG) surfaces with a brush‐like bilayer structure, and dibenzocyclooctyne monolayers (both on glass), are compared. Fluorescence imaging is employed to survey the effect of coating structure and conformation on hybridization performance. To better understand the interfacial structural properties and chemistry of the coated films, atomic force microscopy, water contact angle measurements, and X‐ray photoelectron spectroscopy are employed to characterize the surface morphology. DNA probe microarrays are created on the different platforms via microchannel cantilever spotting, and their performance for hybridizing with the DNA counterparts is assessed. While all three platforms work reliable for DNA detection, a protein‐binding assay reveals that PBAG surfaces offer the highest hybridization efficiency among these approaches. The results of the present work have significant implications for comprehension of the interactions between the DNA hybridization efficiency and the physico‐chemical properties of surface coatings and can inform the fabrication of DNA sensors.

To elucidate the vibrational characteristics of power chassis in paddy fields, we examined the Yanmar VPG6G rice transplanter across diverse terrains, including paddy fields, dry land, and concrete roads. Vibrational acceleration measurements, taken in longitudinal, transverse, and vertical orientations at key chassis locations, revealed noteworthy findings. The Mizuta power chassis exhibited its lowest root-mean-square (RMS) vibrational acceleration on concrete, while the highest was observed on paddy fields. The acceleration power spectra predominantly peaked between 1~14 Hz, with peak values amplifying as speed increased. Additionally, pendant orientation frequencies exceeded those of longitudinal and lateral directions. Both front and rear wheels mirrored the vibrational accelerations of the rear axle, but dynamic load coefficients for the front wheels consistently surpassed the rear, particularly at elevated speeds. This research not only enhances our understanding of terrain-induced vibrations and the intricate dynamics between terrain and tires but also lays the groundwork for designing optimized vibration-damping solutions tailored to prevalent road conditions.

The widespread use of chemical herbicides has jeopardized concerns about food safety and ecological consequences. To address these issues and reduce reliance on chemical herbicides, a physical weed control device was developed for the tillering stage in paddy fields. This device features a biomimetic duckbill-like vibration chain that effectively controls weed outbreaks. The chain penetrates the soft surface soil of the paddy field under gravity and rapidly stirs the soil through vibration, leading to the detachment of the weed roots anchored in the surface layer. Simultaneously, the device avoids mechanical damage to rice seedlings rooted in deeper soil. This study aimed to investigate the effects of chain structural parameters (the number of chain rows, vibration amplitude, and length of chains) and operational parameters (vibration frequency and working velocity) on weed control efficiency and rice seedling damage. Through a central composite regression field test, the optimal device structure and operational parameters were determined. The optimization results demonstrated that a vibration amplitude of 78.8 mm, a chain length of 93.47 cm, and 3.4 rows of chains, along with a vibration frequency and working velocity ranging from 0.5 to 1.25 m/s, achieved an optimal weeding effect. Under the optimal parameter combination, field test results demonstrated that approximately 80% of the weeds in the field were effectively cleared. This indicates that the design of the biomimetic duckbill-like vibration chain weeding device exhibits a relatively superior weeding performance, offering a practical solution for the management of weeds in rice fields.

Abstract Global climate change and socio-economic development have led to a shortage of water and labour resources, which has had a significant impact on rice cultivation. In this study, the application of micro-ridge-furrow planting technology and degradable film mulching in dry direct-seeded rice was investigated to address the factors restricting the development of the rice industry and reduce the impact of rice production on the environment. The effects of a micro-ridge-furrow planting pattern and degradable film mulching on soil temperature, seedling growth, and yield of dry direct-seeded rice in a semiarid region of China were studied through three field experiments: micro-ridge-furrow mulching with traditional plastic film (T1); micro-ridge-furrow mulching with degradable film (T2); and traditional flat-cropping mulching with traditional plastic film (CK). The experimental results demonstrated that the micro-ridge-furrow mulching film planting pattern promoted the germination of rice seeds and improved the soil temperature, plant height, leaf area, dry mass, and grain yield. T2 had the highest average soil temperature (14.68–17.83 ℃ during the day; 14.4–15.74 ℃ at night), leaf area (41.85 cm2 plant−1), root dry mass (45.32 mg plant−1), shoot dry mass (58.46 mg plant−1), root–shoot ratio (0.821), and yield (8.112 t ha−1). In summary, the micro-ridge-furrow mulching with degradable film (T2) is recommended as an efficient planting and mulching pattern for sustainably solving environmental problems and improving grain yield in semiarid regions of China.

Stroke is the main cause of death and disability among Chinese, and neurorestoration is an effective therapeutic strategy for patients with stroke. In recent years, many achievements have been made in stroke neurorestoration, but viewpoints for managing stroke vary per discipline. In order to promote standardization of diagnosis and treatment for stroke neurorestoration, the Chinese Association of Neurorestoratology (CANR; Preparatory) and China Committee of International Association of Neurorestoratology (IANR-China Committee) organized professional experts in the field to integrate fragmented neurorestorative methods and establish clinical diagnostic and therapeutic guidelines for stroke neurorestoration. This guideline includes the diagnosis and staging of stroke and therapeutic recommendations for neurorestoration at different stages of stroke in order to improve survival and quality of life of stroke patients.

To realize the autonomous operation of a terraced rice sowing robot, a set of sowing robot operation path planning algorithms with universal significance for small, irregular terraced plots is proposed. According to the characteristics of terraces and the agronomic requirements of sowing seeding, the operation path mainly includes parallel operation and boundaries surrounding the operation path. The boundary pre-collision detection method is expounded, and the cyclic detection method judges the U-turn area. The Bézier curve fitting algorithm was used to smooth the boundary wrapping path. To verify the feasibility of the algorithm, four typical irregular small fields located in 666.7-hectare terraces of Sama in Hong He Prefecture, Yunnan Province, were randomly selected, and a field map was obtained through Google Earth. An existing seeding robot was used as a model, and the simulation and comparison tests were carried out with the mainstream EHNS algorithm and boundary polyline algorithm under the ROS-kinetic platform in the Cen Village Scientific Research Base of South China Agricultural University. The actual boundaries of the four fields with the same simulation test were used as the map to verify the field experiment. The simulation test results show that the area coverage of the sowing operation is greater than 93.53% and the replay rate is less than 3.46%, and the field test results show that the area coverage of the sowing operation is greater than 94.33% and the replay rate is less than 3.03%. The simulation test is in good agreement with the field test results, indicating that the algorithm has good adaptability, which meets the requirements of a sowing robot for sowing operation path planning and can provide a certain reference for the path planning of irregular field operation robots in hilly and mountainous areas.

This study was designed to investigate the effects of dietary betaine supplementation on growth performance, meat quality and muscle lipid metabolism of growing-finishing pigs. Thirty-six crossbred pigs weighing 24.68 ± 0.97 kg were randomly allotted into two treatments consisting of a basal diet supplemented with 0 or 1200 mg/kg betaine. Each treatment included six replications of three pigs per pen. Following 119 days of feeding trial, dietary betaine supplementation significantly enhanced average daily gain (ADG) (p < 0.05) and tended to improve average daily feed intake (ADFI) (p = 0.08) and decreased the feed intake to gain ratio (F/G) (p = 0.09) in pigs during 100~125 kg. Furthermore, a tendency to increase ADG (p = 0.09) and finial body weight (p = 0.09) of pigs over the whole period was observed in the betaine diet group. Betaine supplementation significantly increased a*45 min and marbling and decreased b*24 h and cooking loss in longissimus lumborum (p < 0.05), tended to increase intramuscular fat (IMF) content (p = 0.08), however had no significant influence on carcass characteristics (p > 0.05). Betaine supplementation influenced the lipid metabolism of pigs, evidenced by a lower serum concentration of low-density lipoprotein cholesterol (p < 0.05), an up-regulation of mRNA abundance of fatty acid synthase and acetyl-CoA carboxylase (p < 0.05), and a down-regulation of mRNA abundance of lipolysis-related genes, including the silent information regulators of transcription 1 (p = 0.08), peroxisome proliferator-activated receptorα (p < 0.05), peroxisome proliferator-activated receptor gamma coactivator-1α (p = 0.07) and carnitine palmitoyl transferase 1 (p < 0.05) in longissimus lumborum. Moreover, betaine markedly improved the expression of microRNA-181a (miR-181a) (p < 0.05) and tended to enhance miR-370 (p = 0.08). Overall, betaine supplementation at 1200 mg/kg could increase the growth performance of growing-finishing pigs. Furthermore, betaine had a trend to improve meat quality and IMF content via increasing lipogenesis and down-regulating the abundance of genes associated with lipolysis, respectively, which was associated with the regulation of miR-181a and miR-370 expression by betaine.

For the management of fractured large pores in high-water-bearing reservoirs, the general approach is to use transfer dissection and sealing. Conventional regulators have a limited regulating radius and can only produce blocking in the near-well zone, which is not ideal. Deep dissection technology can expand the radius of action and substantially improve the blocking effect. The existing deep-dissection agent system has problems such as high cost and poor effect, which affect its large-scale application. In this paper, to address these problems, a gel-type dissection modifier cross-linking agent was synthesized and optimized in the laboratory using low-concentration polymer, and the factors affecting the final gel formation effect were experimentally studied. The final polymer concentration was chosen to be 1500 mg/L~3000 mg/L, the poly-crossing ratio was 30:1, the pH was controlled at 7–9, and the temperature was controlled at 30–60 °C; the rubber was formed with good shear resistance and thermal stability, and had good adaptability to the high-mineralization environment. The optimal injection concentration of water-expanded rubber particles for this system was confirmed to be 3000 mg/L. Cryo-electron microscopy was used to observe the morphology of polymer gel formation and the adhesion of nucleated water-expanded particles to the gel, to clarify the mechanism of enhanced retention and sealing of nucleated water-expanded rubber particles by the multiple complex gel system, and finally to verify the sealing performance of the composite sealing system and determine the use effect by indoor simulation experiments with a two-dimensional flat plate model. This study is of great significance for the efficient development of high-water-bearing late reservoirs and further improvement of crude oil recovery.

The soil in some areas of northern China is heavy owing to the presence of clay and stones, which significantly affects the normal operation of a planter as well as the growth of rice. In this regard, this study proposes a seedbed clearing and shaping device for dry direct-seeded rice, which can be used to remove stones in the seeding area, break soil blocks, for soil leveling, and groove forming. The overall structure and roller of the proposed device was developed based on theoretical calculations, discrete element modeling (DEM) simulations, and field tests. The soil-mixing tooth was distributed on the roller based on the double-helix rule, and the two sides of the helix were configured according to the right-hand and left-hand. Subsequently, DEM was used to develop a 33 box-bench design. According to the agronomic requirements and operating speed ratio, the forward speed was set to 0.5 m/s. Furthermore, the optimization parameters combination of the device obtained by simulation experiments was: forward speed 0.5 m/s, soil depth 61 mm, and rotation speed 110 r/min, which obtained a stone removal rate of 85.65%, stone removal efficiency of 35.47 pieces/m, operating resistance of 719.23 N, and torque of 174.89 Nm. The field verification test results indicated that the stone removal rate was 77.23% under the optimization parameters combination, and the mean relative error of the simulated experiments value was 8.42%, which showed that the performance of the proposed device functioned stably and reliably, thereby providing a high-quality seedbed for sowing and rice growth. The developed device represents a useful solution for the seedbed clearing and shaping.

This paper studies the in-line and cross-flow coupling vibration response characteristics of a marine viscoelastic riser subjected to two-phase internal flow and affected by the combined effects of several parameters including the volume fraction of gas phase, sea water flow velocity, viscoelastic coefficient of the marine riser, axial tension amplitude, and the in-line and cross-flow coupling effect taking into account both the geometric and hydrodynamic nonlinearities. On the base of extended Hamilton’s principle for open systems, the dynamic equations of the marine viscoelastic riser subjected to the axial tension and gas-liquid-structure interaction are established. Two distributed and coupled van der Pol wake oscillators are utilized to model the fluctuating lift and drag coefficients, respectively. The finite element method is adopted to directly solve the highly coupled nonlinear fluid-structure interaction equations. Model validations are firstly performed through comparisons with the published experimental data and numerical simulation results, and the characteristic curves of the in-line and cross-flow vibration pattern, the in-line and cross-flow displacement trajectories, the in-line and cross-flow space-time response of displacement, and the in-line and cross-flow space-time response of stress versus different parameters are obtained, respectively. The results show that the volume fraction of gas phase, sea water flow velocity, viscoelastic coefficient of marine riser, axial tension amplitude, the in-line and cross-flow coupling effect, and multiphase internal flow velocity have significant influences on the dynamic response characteristics of the marine viscoelastic riser. Furthermore, the maximum displacements and stresses of the marine viscoelastic riser can be increased or decreased depending on the internal flow velocity, and the critical internal flow velocities result in the increase of mode order for different cross-flow velocities. It is also demonstrated that appropriate viscoelastic coefficients are very important to effectively suppress the maximum displacements and stresses.

In this article, a vortex-induced vibration prediction model of a flexible riser conveying two-phase flow, including geometric and hydrodynamic nonlinearity, is established. A van der Pol wake oscillator is utilized to characterize the fluctuating lift forces. The finite element method is chosen to solve the coupled nonlinear fluid–structure interaction equations. The natural frequencies of the flexible riser are calculated to validate the method through comparisons with results from the literature. The modal analyses show that geometric nonlinearity has a significant effect on the natural frequency, and the critical internal velocity is reduced than those in linear analyses. The impacts of the gas volume fraction as functions of cross-flow velocity on the synchronization region, the displacement amplitudes, and the maximum stresses and frequency spectra have been investigated. The results show that an increase in the gas fraction results in the linear increase in natural frequencies and a wider synchronization region, and an increase in liquid flow rate led to the slight decrease in displacement amplitude and maximum stress within a small flow range.

Existing devices for dry direct-seeded rice with film mulching in northern China have limitations such as imprecise sowing, unadjustable sowing depth, and seeding device blocking. In this regard, this study proposes a combined seeding method of ‘mini shovel + telescopic pipe’ for dry direct-seeded rice with film mulching. A precision seeder for dry direct-seeded rice with film mulching was developed through theoretical calculations, discrete element modelling (DEM) simulations, and field experiments. The configuration and diameter of the rollers were obtained. Twelve telescopic pipes were evenly distributed on the circumference of the roller, with a contact ratio exceeding one. This ratio reduced the slip rate of the roller effectively. Subsequently, DEM was used to develop a 33 central composite design. The response surface was established with the sowing depth as the response value. According to agronomic requirements, the sowing depth was set to 20 mm. The optimal combination of working parameters was obtained by optimizing the regression equation. The field experiments showed that the performance of the precision seeder for dry direct-seeded rice with film mulching satisfied the requirements of agricultural production, working stably and reliably. The developed device represents a useful solution for dry direct-seeded rice with film mulching.

The APETALA 2/Ethylene-responsive element binding factor (AP2/ERF) transcription factor gene family is widely involved in the biotic and abiotic stress regulation. Haynaldia villosa (VV, 2n = 14), a wild species of wheat, is a potential gene pool for wheat improvement. H. villosa confers high resistance to several wheat diseases and high tolerance to some abiotic stress. In this study, ERF1-V, an ethylene-responsive element-binding factor gene of the AP2/ERF transcription factor gene family from wild H. villosa, was cloned and characterized. Sequence and phylogenetic analysis showed that ERF1-V is a deduced B2 type ERF gene. ERF1-V was first identified as a Blumeria graminis f. sp. tritici (Bgt) up-regulated gene, and later found to be induced by drought, salt and cold stresses. In responses to hormones, ERF1-V was up-regulated by ethylene and abscisic acid, but down-regulated by salicylic acid and jasmonic acid. Over expression of ERF1-V in wheat could improve resistance to powdery mildew, salt and drought stress. Chlorophyll content, malondialdehyde content, superoxide dismutase and peroxidase activity were significantly differences between the recipient Yangmai158 and the transgenic plants following salt treatment. Furthermore, the expression levels of some stress responsive genes were differences after drought or salt treatments. Although ERF1-V was activated by the constitutive promoter, the agronomic traits, including flowering time, plant height, effective tiller number, spikelet number per spike and grain size, did not changed significantly. ERF1-V is a valuable gene for wheat improvement by genetic engineering.

Growth arrest special 5 (GAS5) is a long non-coding RNA reported to function as an inhibitor in various tumors including cervical cancer. However, the molecular mechanism of GAS5 involved in cervical cancer progression remains far from being elucidated. The expression of GAS5, forkhead box protein O1 and phosphatase and tensin homolog was examined by quantitative reverse transcription polymerase chain reaction qRT-PCR. cell growth, invasion, and apoptosis were assessed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay, colony formation assay, transwell invasion assay, and flow cytometry analysis, respectively. The interaction between GAS5 and miR-196a or miR-205 was confirmed by luciferase reporter assay, RNA immunoprecipitation assay, and qRT-PCR. Xenograft tumor experiments were performed to validate the biological role of GAS5 and its molecular mechanism in cervical cancer in vivo. GAS5 expression was decreased in cervical cancer tissues and cells. GAS5 overexpression suppressed cervical cancer cell proliferation, invasion, and apoptosis. GAS5 was able to directly bind to miR-196a and miR-205 to downregulate their expression. Moreover, GAS5 induced forkhead box protein O1 and phosphatase and tensin homolog expression by repressing miR-196a and miR-205, respectively. Exogenous expression of GAS5 hindered tumor growth in vivo by downregulating miR-196a and miR-205. Upregulation of GAS5 suppressed cell proliferation, invasion, and apoptosis of cervical cancer cells by downregulating miR-196a and miR-205, contributing to our understanding the pathogenesis of cervical cancer and development of long non-coding RNA–mediated clinical therapy against this disease.

Background and purpose An analysis of the ASTER 2 trial revealed similar final recanalisation levels and clinical outcomes in acute large vessel occlusion (LVO) stroke between stent retrieval (SR) alone as a first-line mechanical thrombectomy (MT) technique (SR alone first-line) and concomitant use of contact aspiration (CA) plus SR as a first-line MT technique (SR+CA first-line). The purpose of the present study was to compare the safety and efficacy of SR+CA first-line with those of SR alone first-line for patients with LVO in China.Methods We conducted the present study by using the data from the ANGEL-ACT registry. We divided the selected patients into SR+CA first-line and SR alone first-line groups. We performed logistic regression and generalised linear models with adjustments to compare the angiographic and clinical outcomes, including successful/complete recanalisation after the first technique alone and all procedures, first-pass successful/complete recanalisation, number of passes, 90-day modified Rankin Scale, procedure duration, rescue treatment and intracranial haemorrhage within 24 hours.Results Of the 1233 enrolled patients, 1069 (86.7%) received SR alone first-line, and 164 (13.3%) received SR+CA first-line. SR+CA first-line was associated with more thrombectomy passes (3 (2–4) vs 2 (1–2); β=1.77, 95% CI=1.55 to 1.99, p0.05) between the two groups.Conclusions Patients undergoing SR+CA first-line had more thrombectomy passes and longer procedure duration than patients undergoing SR alone first-line. Additionally, we suggested that SR+CA first-line was not superior to SR alone first-line in final recanalisation level, first-pass recanalisation level and 90-day clinical outcomes in the Chinese population.
