Anatomical sublobar resection was widely performed for small-sized nodule located in the deep field of the lung. In this study, we compared surgical outcomes between anatomical sublobar resection in left upper lobe including segmentectomy and subsegmentectomy. We also applied a technique based on bronchovascular patterns for subsegmentectomy.A hundred and fifty-one patients underwent anatomical sublobar resection of left upper lobe in our hospital in the period from January 2018 to December 2019 and they were retrospectively reviewed. Patients' characteristics and surgical outcome of the subsegmentectomy group (n = 71) were analyzed and compared to those of patients of the segmentectomy group (n = 80). The bronchovascular patterns of left upper lobe in these patients were also classified, and a technique of subsegmentectomy was introduced by cases description.Compared to segmentectomy, the subsegmentectomy group had longer operative time [141min, interquartile range (IQR): 125-161 vs. 127.5 min, interquartile range (IQR): 114.75-148.25; P = 0.001] and more estimated blood loss [50 mL, IQR: 30-50 vs. 30 mL, IQR: 20-50; P = 0.02]. Branching pattern of bronchus was classified into 3 types in left upper division, and 2 types in lingular division. Branching pattern of pulmonary artery was classified into 7 types in left upper division and 2 types in lingular division. Branching pattern of pulmonary vein was classified into 3 types in left upper division and VSubsegmentectomy is a safe procedure with surgical outcome comparable to the one of segmentectomy except operative time and estimated blood loss. Subsegmentectomy based on bronchovascular patterns in left upper lobe is feasible.

Gestational trophoblastic disease (GTD) encompasses a range of trophoblastic disorders from hydatidiform mole (HM), to malignant gestational trophoblastic neoplasia (GTN). The exact molecular mechanisms of GTN remain unknown. Dysregulation and dysfunction of programmed cell death 4 (PDCD4)have been observed in many cancers. The roles of PDCD4 in GTD have not been previously reported.A total of 161 cases of formalin-fixed, paraffin-embedded trophoblast blocks, and 36 cases of fresh trophoblast tissues were collected, including normal first trimester placentas, HM, and invasive HM. Choriocarcinoma cells JAR and JEG-3 were employed. The expressions of PDCD4 and small ubiquitin-like modifier 2/3 (SUMO2/3) were examined by immunohistochemistry, quantitative reverse transcription PCR and Western blotting in trophoblastic tissues and cells. The relationship between SUMOylation and PDCD4 was investigated. The effects of PDCD4 on proliferation, invasion, and migration of choriocarcinoma cells were evaluated by Cell Counting Kit-8 and transwell assays post siRNA transfection. Extracellular MatrixAdhesion Profiler PCR Array was used to screen the downstream molecules of PDCD4.PDCD4 was significantly repressed in HM tissues. Loss of PDCD4 expression was demonstrated in 90% invasive HMs. Choriocarcinoma cells also displayed with suppressed PDCD4 expression. The varied expression of PDCD4 was paralleled by SUMO2/3. Inhibition of SUMOylation reduced the expression of PDCD4. Silencing of PDCD4promoted proliferation/migration/invasion, upregulatedMMP3/MMP8/ITGB2, and downregulated TIMP1/TIMP2 in choriocarcinoma cells.Our results suggest that reduced SUMOylation is one reason for suppressed PDCD4 in GTD. Loss of PDCD4 likely determines the malignant phenotype of GTN by dysregulating some members of the MMPs/TIMPs/integrins complex.

Introduction In this study we examined the hypothesis that a hypoxic intrauterine environment causes mitochondrial dysfunction of trophoblasts in fetal growth restriction (FGR). Methods The mtDNA content, mRNA levels of mitochondrial encoded genes (ND6, COX I), mitochondrial membrane proteins (COX I, COX IV and VDAC), HIF-1α and BINP3 (mitophagy receptor) protein levels were examined in FGR placentas and normal placentas. The mitochondrial function (ATP production and mitochondrial membrane potential-ΔΨm) and above related proteins were further examined in hypoxic HTR-8/SVneo cells induced by cobalt chloride (CoCl2). Mitophagy and its regulating mechanism under hypoxia in FGR was also investigated. Results Compared with normal controls, both FGR placentas and CoCl2-treated trophoblast cells demonstrated statistically lower mtDNA content, reduced mRNAs of mitochondrial encoding genes, and decreased mitochondrial membrane proteins, accompanied by increased HIF-1α. Mitochondrial functions were impaired as demonstrated by decreased ATP production, and, reduced ΔΨm in CoCl2-treated cells. Meanwhile, mitophagy was markedly enhanced as indicated by increased LC3 fluorescent puncta in mitochondria of hypoxic trophoblastic cells. The upregulated BINP3 expression was demonstrated in FGR placentas as well as in hypoxic trophoblastic cells. Discussion We demonstrated that hypoxic conditions lead to impaired mitochondrial function in trophoblasts in FGR. Reduced mtDNA may be associated with enhanced mitophagy via activating HIF-1α/BINP3 signalling pathway, that may, in turn, affect nutrition and energy transfer to the growth-restricted fetus.

Inter-turn short-circuit (ITSC) faults do not necessarily produce high temperatures but have special heat distribution and characteristics. Therefore, a new recognition solution for diagnosing faults is proposed, based on the fault overlay method, and the convolutional neural network (CNN) is trained to achieve the automatic identification of infrared images. In this method, through the coverage of layers, the proposed image augmentation method is carried out and simulates the fault data of increasing training. We produce 43 fault traces through the fault overlay method on the three-phase winding of a transformer and use 90 infrared images of transformers in normal operation combined with them to enhance the amount of image data. The fault recognition ability is realized based on CNN model training, including analysis of experimental results of grayscale and color images, and Gaussian noise. In the test of the practical case, a short-circuit test of the 11.4 kV dry-type transformer is carried out, and the ITSC fault is identified when the load is about 15%. The fault characteristic block on this thermal image is 36.3 degrees, which verifies the identification available by this method, and has a certain reference value for the development of infrared image diagnosis technology for power equipment.

Objective: This study aims to investigate the feasibility of single-port video-assisted thoracoscope (SPVATS) in treating thoracic oesophageal squamous cell carcinoma (TESCC) using McKeown approach. Materials and Methods: Totally 10 McKeown approach-based SPVATS surgeries (8 males and 2 females, aged 42–68 years) were carried out from January 2015 to December 2015 to treat TESCC, including one case in upper thoracic segment, 5 cases in median thoracic segment and 4 cases in inferior thoracic segment. All the cases were pathologically diagnosed as SCC pre-operatively. SPVATS was performed to free thoracic oesophagus and dissect the lymph nodes, and laparoscopy was performed to free stomach and to perform oesophagus-left gastric collum anastomosis. Results: All the patients were successfully completed SPVATS, with average thoracic surgery time as 150 min, intra-operative blood loss as 30–260 ml (average 90 ml), and post-operative hospital stay as 9–16 days (average 12 days). Conclusions: SPVATS was technically feasible and safe in treating TESCC using McKeown approach, with less trauma and rapid post-operative recovery, and hence, it could be used as a new surgical option for McKeown approach-based TESCC treatment.

The aim of this study was to explore the significance of upper mediastinal lymph node dissection performed by video-assisted thoracic surgery in the treatment of middle thoracic esophageal carcinoma.The clinical and pathological data from 128 patients with middle thoracic esophageal carcinoma who underwent surgery from January 2013 to December 2015 using a right chest-abdomen-neck approach combined with thoracoscopy and laparoscopy in the Jieyang People's Hospital of Huangdong province were analyzed retrospectively.The lymph node metastasis rates of the thoracic left para-recurrent laryngeal nerve (1, 2, and 4L zones) and right para-recurrent laryngeal nerve (1R zone) were 30.47% and 28.12% in 128 cases, respectively. The metastasis rates of the 2R, 4R, and 5 zones were 4.69%, 3.91%, and 5.47%, respectively.The upper mediastinal region was the most common location for lymph node metastasis from middle thoracic esophageal carcinoma, and upper mediastinal lymph node dissection performed by video-assisted thoracic surgery was safe and complete. It also reduced the risk of para-recurrent laryngeal nerve injury, residual tumor, and the postoperative recurrence rate.

ObjectiveThe aims of this study were to make clear whether miR-21 was dysregulated in hydatidiform mole (HM) tissues and choriocarcinoma (CCA) cells, to elucidate whether aberrant miR-21 expression would affect the function of CCA cells, and to find out whether there was a relationship between miR-21 andAKT,PDCD4, andPTENin CCA cells.MethodsFresh and formalin-fixed, paraffin-embedded trophoblastic tissues (normal first trimester placentas and HMs) were retrieved from the biobank in the International Peace Maternity and Child Health Hospital, Shanghai Jiao Tong University. Choriocarcinoma JAR and JEG-3 cells were cultured. Expression of miR-21 in trophoblast cells and tissues was examined by quantitative real-time polymerase chain reaction. Location and distribution of miR-21 in trophoblast tissues were determinated by in situ hybridization and fluorescent in situ hybridization. The effect of miR-21 on JAR and JEG-3 cells was tested by miR-21 mimics and inhibitor transfection, followed by cell viability assay, flow cytometric analysis, and Transwell analysis. Interaction between miR-21 and its target genes in CCA cells was verified by quantitative real-time polymerase chain reaction, Western blot, and luciferase report system.ResultsWe originally found miR-21 was markedly upregulated in HM tissues compared with normal first trimester placentas. The expression of miR-21 was exclusively confined in trophoblastic layers. Furthermore, we discovered miR-21 was significantly increased in JAR and JEG-3 cells compared with normal primary human trophoblastic cells. Moreover, we demonstrated miR-21 could promote proliferation, migration, and invasion of CCA cells. We furthermore proved miR-21 negatively regulated PDCD4 and PTEN in CCA cells and targeted toPDCD43′UTR directly. In addition, we confirmed that miR-21 could activate Akt pathway by phosphorylating Akt at Ser 473.ConclusionsOur results suggested miR-21 was responsible for aggressive phenotype of gestational trophoblastic disease and had the potential diagnostic and therapeutic values for gestational trophoblastic neoplasm.

Detecting the faults related to the operating condition of induction motors is a very important task for avoiding system failure. In this paper, a novel methodology is demonstrated to detect the working condition of a three-phase induction motor and classify it as a faulty or healthy motor. The electrical current signal data is collected for five different types of fault and one normal operating condition of the induction motors. The first part of the methodology illustrates a pattern recognition technique based on the empirical wavelet transform, to transform the raw current signal into two dimensional (2-D) grayscale images comprising the information related to the faults. Second, a deep CNN (Convolutional Neural Network) model is proposed to automatically extract robust features from the grayscale images to diagnose the faults in the induction motors. The experimental results show that the proposed methodology achieves a competitive accuracy in the fault diagnosis of the induction motors and that it outperformed the traditional statistical and other deep learning methods.

The Philippines consists of 7100 islands, many of which still use fossil fuel diesel generators as the main source of electricity. This supply can be complemented by the use of renewable energy sources. This study uses a Philippine offshore island to optimize the capacity configuration of a hybrid energy system (HES). A thorough investigation was performed to understand the operating status of existing diesel generator sets, load power consumption, and collect the statistics of meteorological data and economic data. Using the Hybrid Optimization Models for Energy Resources (HOMER) software we simulate and analyze the techno-economics of different power supply systems containing stand-alone diesel systems, photovoltaic (PV)-diesel HES, wind-diesel HES, PV-wind-diesel HES, PV-diesel-storage HES, wind-diesel-storage HES, PV-wind-diesel-storage HES. In addition to the lowest cost of energy (COE), capital cost, fuel saving and occupied area, the study also uses entropy weight and the Technique for Order Preference by Similarity to an Ideal Solution (TOPSIS) method to evaluate the optimal capacity configuration. The proposed method can also be applied to design hybrid renewable energy systems for other off-grid areas.

The objective of present study was to find out an accurate, rapid and nondestructive method to detect total acid content (TA) of pitaya with visible/near-infrared spectrometry, wavelet transform (WT) and successive projections algorithm (SPA), which will provide scientific basis for non- destructive measurement of pitaya. Maya2000 fiber-optic spectrumeter was used to collect spectral data of pitaya on the wavelength in the range of 380~1 099 nm; and then with the methods of WT denosing pretreatment, SPA and partial least squares regression (PLSR) quantitative forecasting model of TA of pitaya was established. The result showed that the precision of WT-SPA-PLSR model, which combine the WT with SPA, was better than that of PLSR model based on the whole wave variables. The relation coefficient of the PLSR model (Rp) that predicted TA based on the original spectrum of all samples as the input variables was 0.851 394 and RMSEP was 0.086 848. The original spectrum variable of the all samples were processed by using wavelet function dbN(N=2, 3, …, 10) for wavelet decomposition and de-noising. The optimal results of noise reduction were decomposed in level 2 using wavelet function db4 (db4-2). The Rp of WT-PLSR model was 0.915 635 and RMSEP was 0.066 752. The prediction of model using wavelet transform de-noising was improved significantly. After the original spectrum processed by db10-3 and SPA, 12 preferred variables were selected from 570 spectrum variables, such as 530, 545, 604, 626, 648, 676, 685, 695, 730, 897, 972, 1 016 nm spectrum variables. The WT-SPA-PLSR model based on these 12 variables as input variables was established. Rp of the WT-SPA-PLSR prediction model was 0.882 83 and RMSEP was 0.077 39. SPA algorithm was suitable for the selection of spectrum variables which could effectively obtain the spectrum variables which were strong correlation with TA and increase the accuracy and stability of the prediction model. The results indicated that the nondestructive detection for TA of pitaya based on the diffuse reflectance visible/near-infrared spectrometry, WT and SPA was feasible.

Gestational trophoblastic disease (GTD) encompasses a range of trophoblast-derived disorders. The most common type of GTD is hydatidiform mole (HM). Some of HMs can further develop into malignant gestational trophoblastic neoplasia (GTN). Aberrant expression of microRNA (miRNA) is widely reported to be involved in the initiation and progression of cancers. MiRNA expression profile also has been proved to be the useful signature for diagnosis, staging, prognosis, and response to chemotherapy. Till now, the profile of miRNA in the progression of GTD has not been determined. In this study, a total of 34 GTN and 60 complete HMs (CHM) trophoblastic tissues were collected. By miRNA array screening and qRT-PCR validating, six miRNAs, including miR-370-3p, -371a-5p, -518a-3p, -519d-3p, -520a-3p, and -934, were identified to be differentially expressed in GTN vs. CHM. Functional analyses further proved that miR-371a-5p and miR-518a-3p promoted proliferation, migration, and invasion of choriocarcinoma cells. Moreover, we demonstrated that miR-371a-5p was negatively related to protein levels of its predictive target genes BCCIP, SOX2, and BNIP3L, while miR-518a-3p was negatively related to MST1 and EFNA4. For the first time, we proved that miR-371a-5p and miR-518a-3p directly targeted to 3′-UTR regions of BCCIP and MST1, respectively. Additionally, we found that miR-371a-5p and miR-518a-3p regulated diverse pathways related to tumorigenesis and metastasis in choriocarcinoma cells. The results presented here may offer new clues to the progression of GTD and may provide diagnostic biomarkers for GTN.

Heme oxygenase-1 (HMOX-1) is a microsomal enzyme that exerts anti-apoptotic and cytoprotective effects. In the present study, HMOX-1 was demonstrated to be overexpressed and able to be induced by doxorubicin in breast cancer cell lines. Knockdown of HMOX-1 using short interfering (si)RNA enhanced the cytotoxicity of doxorubicin in MDA-MB-231 and BT549 cells. Knockdown of HMOX-1 downregulated B cell lymphoma (Bcl)-2 and Bcl-extra large expression, and significantly enhanced doxorubicin-induced apoptosis in MDA-MB-231 and BT549 cells. Additionally, knockdown of HMOX-1 upregulated light chain 3B expression and markedly increased the accumulation of autophagic vacuoles in MDA-MB-231 and BT549 cells treated with doxorubicin. These results indicated that HMOX-1 may be involved in conferring the chemoresistance of breast cancer cells, by preventing apoptosis and autophagy. Therefore, HMOX-1 may represent a potential therapeutic target for enhancing the cytotoxicity and efficacy of doxorubicin during the treatment of breast cancer.

Aims: To explore the expression of miR-24-3p in human arteries with arteriosclerosis obliterans (ASO) as well as the role of miR-24-3p in the pathogenesis of ASO. Methods: We used quantitative real-time PCR (qRT-PCR) and in situ hybridization to monitor miR-24-3p expression in human arteries. To investigate the effect of miR-24-3p on human arterial smooth muscle cells (HASMCs), we applied cell counting and EdU assays to monitor proliferation and transwell and wound healing assays to investigate migration and flow cytometry to investigate apoptosis. Furthermore, we applied 3'-untranslated region (3'-UTR) luciferase assays to investigate the role of miR-24-3p in targeting platelet-derived growth factor receptor B (PDGFRB) and c-Myc. Results: MiR-24-3p was mainly located in the media of arteries and was downregulated in ASO arteries compared with normal arteries. Platelet-derived growth factor BB (PDGF-BB) treatment reduced the expression of miR-24-3p in primary cultured HASMCs. MiR-24-3p mimic oligos inhibited the proliferation and migration, and promotes apoptosis of HASMCs. Our 3'-UTR luciferase assays confirmed that PDGFRB and c-Myc were targets of miR-24-3p. Conclusion: The results suggest that miR-24-3p regulates the proliferation and migration of HASMCs by targeting PDGFRB and c-Myc. The PDGF/miR-24-3p/PDGFRB and PDGF/miR-24-3p/c-Myc pathways may play critical roles in the pathogenesis of ASO. These findings highlight the potential for new therapeutic targets for ASO.

Fetal growth restriction (FGR) threatens perinatal health and is correlated with increased incidence of fetal original adult diseases. Most cases of FGR were idiopathic, which were supposed to be associated with placental abnormality. Decreased circulating placental growth factor (PGF) was recognized as an indication of placental deficiency in FGR. In this study, the epigenetic regulation of PGF in FGR placentas and the involvement of PGF in modulation of trophoblast activity were investigated. The expression level of PGF in placental tissues was determined by RT-qPCR, immunohistochemistry and ELISA. DNA methylation profile of PGF gene was analyzed by bisulfite sequencing. Trophoblastic cell lines were treated with ZM-306416, an inhibitor of PGF receptor FLT1, to observe the effect of PGF/FLT1 signaling on cell proliferation and migration. We demonstrated that PGF was downregulated in placentas from FGR pregnancies compared with normal controls. The villous expression of PGF was positively correlated with placental and fetal weight. The CpG island inside PGF promoter was hypomethylated without obvious difference in both normal and FGR placentas. However, the higher DNA methylation at another CpG island downstream exon 7 of PGF was demonstrated in FGR placentas. Additionally, we found FLT1 was expressed in trophoblast cells. Inhibition of PGF/FLT1 signaling by a selective inhibitor impaired trophoblast proliferation and migration. In conclusion, our data suggested that the PGF expression was dysregulated, and disrupted PGF/FLT1 signaling in trophoblast might contribute to placenta dysfunction in FGR. Thus, our results support the significant role of PGF in the pathogenesis of FGR.

Lung adenocarcinoma (AC) is one of the most deadly malignancies. The disease has a low five-year survival rate; therefore, the identification of novel therapeutic agents is required. This study aimed to investigate the effect of small interfering RNA (siRNA) targeting hypoxia‑inducible factor 1α (HIF‑1α) on the growth of AC A549 cells. A549 cells were transfected with various concentrations of HIF‑1α or control siRNA, and the effect on HIF‑1α expression was analyzed using quantitative polymerase chain reaction and western blot analysis. The effects of HIF-1α siRNA on growth inhibition and apoptosis were then assessed using standard methods. HIF‑1α siRNA treatment significantly reduced HIF‑1α mRNA and protein expression in A549 cells. Furthermore, the downregulation of HIF-1α expression inhibited the growth of A549 cells and induced apoptosis of A549 cells by upregulating caspase-3 expression. The present in vitro study demonstrates that the downregulation of HIF‑1α is capable of suppressing AC A549 cell growth, through the induction of apoptosis. This suggests that HIF‑1α inhibition may represent a promising strategy for the treatment of AC.

A novel 2,4,5-triaryl-substituted imidazole (IZCM-1) has been found to display distinct and specific fluorescence enhancement upon binding to parallel G-quadruplexes. Such a sensitive and topology-specific probe is able to light up without affecting the topology or thermal stability of the G-quadruplex sample. Thus, these advantages distinguish IZCM-1 from other G-quadruplex probes.

Intrahepatic cholestasis of pregnancy (ICP) is a pregnancy-specific disorder characterised by raised bile acids in foetal-maternal circulation, which threatens perinatal health. During the progression of ICP, the effect of oxidative stress is underscored. Peroxiredoxin-3 (PRDX3) is a mitochondrial antioxidant enzyme that is crucial to balance intracellular oxidative stress. However, the role of PRDX3 in placental trophoblast cells under ICP is not fully understood. We demonstrated that the level of PRDX3 was downregulated in ICP placentas as well as bile acids–treated trophoblast cells and villous explant in vitro. Toxic levels of bile acids and PRDX3 knockdown induced oxidative stress and mitochondrial dysfunction in trophoblast cells. Moreover, silencing of PRDX3 in trophoblast cell line HTR8/SVneo induced growth arrest and cellular senescence via activation of p38-mitogen-activated protein kinase (MAPK) and induction of p21WAF1/CIP and p16INK4A. Additionally, enhanced cellular senescence, determined by senescence-associated beta-galactosidase staining, was obviously attenuated by p38-MAPK inhibitor SB203580. Our data determined that exposure to bile acid decreased PRDX3 level in human trophoblasts. PRDX3 protected trophoblast cells against mitochondrial dysfunction and cellular senescence induced by oxidative stress. Our results suggest that decreased PRDX3 by excessive bile acids in trophoblasts plays a critical role in the pathogenesis and progression of ICP.

Induction motors face various stresses under operating conditions leading to some failure modes. Hence, health monitoring for motors becomes essential. In this paper, we introduce an effective framework for fault diagnosis of 3-phase induction motors. The proposed framework mainly consists of two parts. The first part explains the preprocessing method, in which the time-series data signals are converted into two-dimensional (2D) images. The preprocessing method generates recurrence plots (RP), which represent the transformation of time-series data such as 3-phase current signals into 2D texture images. The second part of the paper explains how the proposed convolutional neural network (CNN) extracts the robust features to diagnose the induction motor&rsquo, s fault conditions by classifying the images. The generated RP images are considered as input for the proposed CNN in the texture image recognition task. The proposed framework is tested on the dataset collected from different 3-phase induction motors working with different failure modes. The experimental results of the proposed framework show its competitive performance over traditional methodologies and other machine learning methods.

In order to combine the advantages of Surpac in three-demensional modeling and ArcGIS Engine in three-demensional space analysis, the relation of data elements between Surpac and ArcGIS Engine is studied. In this study, the method of data conversion between two models is proposed. Take the model conversion of a large metal mine for example,showing the ability of the method,by adopting the software of Visual C++ to write a interface program of Surpac-ArcGIS Engine. The results show that the method of data conversion which is proposed in this study can switch Surpac and ArcGIS Engine.This switch function can extend the application space of ArcGIS Engine in Civil Engineering field.

Therefore, the position of rockburst, hazard level, as well as how much energy released need to have quantitative analysis, which can provide a safe and effective guideline for mining operation. In this paper, take the Linglong Gold Mine as background, the relationship of surrounding rockmass and rock impact energy index could be derived from simulating the ground stress environment in lab. Based on above equation, FLAC 3D is used for simulating the mining operation of Linglong Gold Mine in regard of the size of rock impact energy in current excavation stress condition, which breakdown the limitation of previous method of getting the indicator of rock burst tendency evaluation from uniaxial compressive full stress-strain curve, and provide the reference for safe mining operation.

This project focus on one wearable device of speechcontroller for bicycle light direction designed with wireless sensor network. In the brand-new design system included two-parts -- wireless speech-recognition and wireless light-direction controller by speech commander. That means user can control the bicycle lights with speech command instead of traditional manual ways which made user in danger condition especially when user riding on and comes across dark environment or encounters trucks in tunnels. This creative product can be used on bicycle handles or on helmet, in addition, it can be put on hand as one fashion watch. The wearable device includes ZigBee module with 2.4GHz wireless communication which sends light control codes to the receiver on the lights. The wearable device can be set functions both single user or group users. When in single user mode, one wearable device controls one direction-light while group mode with one controller can control many bicycles lights.

The rapid and convenient method for identification of all kinds of G-quadruplex is highly desirable. In the present study, a novel colorimetric indicator for a vast variety of G-quadruplex was designed and synthesized on the basis of thiazole orange and isaindigotone skeleton. Its distinct color change enables label-free visual detection of G-quadruplexes, which is due to the disassembly of dye H-aggregates to monomers. This specific detection of G-quadruplex arises from its end-stacking interaction with G-quartet. On the basis of this universal indicator, a facile approach for large-scale identification of G-quadruplex was developed.

12-N-Methylated and non-methylated 5,6-dihydrobenzo[c]acridine derivatives were designed and synthesized as new series of c-myc G-quadruplex binding ligands. Their interactions with c-myc G-quadruplex were evaluated using fluorescence resonance energy transfer (FRET) melting assay, circular dichroism (CD) spectroscopy, surface plasmon resonance (SPR), polymerase chain reaction (PCR) stop assay, and molecular modeling. Compared with the non-methylated derivatives, 12-N-methylated derivatives had stronger binding affinity and stabilizing ability to c-myc G-quadruplex structure, and could more effectively stack on the G-quartet surface. All these derivatives had high selectivity for c-myc G-quadruplex DNA over duplex DNA. The reverse transcription (RT) PCR assay showed that compound 21c could down-regulate transcription of c-myc gene in Ramos cell line containing NHE III(1) element, but had no effect in CA46 cell line with NHE III(1) element removed.

One superior iron and manganese bacteria were separated from the stable operation of porcelain granular BAF filters of removing iron, manganese and ammonia. The bacteria was domesticated at low temperature. By analyzing the sample water containing iron and manganese in the role of iron and manganese bacteria which was not domesticated and domesticated at different temperature, observing the Iron and manganese concentration with time going on, studying the bacteria’s removal of iron and manganese property and the domesticated effect. Studies show that: the selected bacteria with 1% bacterial liquid at proper temperature within 48h ,the removal rate of iron and manganese was 75% and 35% respectively;After domesticated at low temperature, the removal rate of the iron and manganese domesticated bacteria at 10°C was improved 0.4 and 2 times more than the before domesticated; The iron and manganese domesticated bacteria at 10°C did not grow at 4°C,but the bacteria’s removal rate was better than the bacteria cultured at 30°C,and the iron removal rate was improved from 23% to 35%,the manganese removal rate was improved from 5% to 11%.

This paper describes work that has been done on an innovative use of a steel chain as a cable in chain ropeway system developed for use in the mountain orchards of china. This work focuses on the use of Msc Adams in describing the quasi static behavior of the configuration of the chain ropeway. A model was built in solid works and then transferred to Msc Adams which was used to predict the profile of the chain and the tension for various loads. From this study it has been shown that Msc Adams can be a valuable tool in solving problems related to the quasi static and equilibrium state of the chain rope way system and getting an in depth behavior of its dynamics. There is now a foundation to proceed on the dynamic investigation of the chain ropeway system.

A series of mansonone F (MF) derivatives were designed and synthesized. These compounds were found to be strong inhibitors for topoisomerases, with much more significant inhibition for topoisomerase II rather than topoisomerase I. The best inhibitor showed 20 times stronger anti-topoisomerase II activity than a positive control Etoposide. The cytotoxic activity of these MF derivatives was evaluated against human cancer cell lines CNE-2 and Glc-82, which showed that these compounds were potent antitumor agents. The structure–activity relationships (SARs) study revealed that o-quinone group and pyran ring are important for their cytotoxic activity.

In an automobile, a stable and durable alternator is extremely important. Based on the basic characteristics of the alternators now on the mainstream market, the hardware simulation method was used to build a simulated “car engine-alternator-auto load-the external environment hardware environment” system. High and low temperature humidity test chamber simulates the vehicle internal environment and the inverter driving power frequency conversion is used to simulate motor car engine power system. It uses fixtures, belts, and pulleys to simulate the connection of the engine to the alternator. In addition it uses electronic load cases to simulate automobile electric power consumption with all kinds of current, voltage, speed signal components to provide feedback signals based on the testing system of alternator to make comparisons. The testing system is designed by analyzing and optimizing the structural mechanics of the whole system according to the requirement of the hardware system layout. For each part of the system factors of design were considered included shape, size, material selection process and heat treatment. Parts and detailed drawings were prepared and later sent for fabrication. The tests indicate that hardware alternator testing system has no major defects that can prevent it from being developed for commercial use.

Deacetoxy/deacetylcephalosporin C synthase (acDAOC/DACS) from Acremonium chrysogenum is a bifunctional enzyme that catalyzes both the ring-expansion of penicillin N to deacetoxycephalosporin C and the hydroxylation of the latter to deacetylcephalosporin C. The R308 residue located in close proximity to the C-terminus of acDAOC/DACS was mutated to the other 19 amino acids. In the resulting mutant pool, R308L, R308I, R308T and R308V showed significant improvement in their ability to convert penicillin analogs, thus confirming the role of R308 in controlling substrate selectivity, the four amino acids all possess short aliphatic sidechains that may improve hydrophobic interactions with the substrates. The mutant R308I showed the highest reactivity for penicillin G, with 3-fold increase in k(cat)/K(m) ratio and 7-fold increase in relative activity.

The method to provide crop biomass status information for precision agriculture by researching the features of leaf area index(LAI) is presented in this paper. The relationship between LAI and spectrum can be identified by spectrum analysis, thereby achieving fast and accurate LAI measurement. The exact total leaf area and the nominal LAI values of citrus leaves can be calculated through a scanning calibration experiment. Analysis system for citrus LAI spectrum data is made up of a data acquisition platform with FieldSpec-FR and computers as its key components to collect spectrum data of leaves and an inversion platform with computers, ViewSpecPro, SPSS and other softwares as its core to calculate the LAI of citrus leaves. The fitting equation of LAI and spectrum information obtained is Y=1.5106X+0.7869 and the calculation error is ±0.0378%. The fitting equation of LAI and dry weight obtained is Y=0.0924X+3.1252 and the calculation error is ±0.3002%. The fitting equation of dry weight and wet weight obtained is Y=0.1807X-0.1133 and the maximum calculation error is 2.2713%.

Air-Fuel Radio (AFR) analyzer technology is basically mastered by monopolies of developed country nowadays. Due to the lack of development in China, it has a strong practical value to study the accurate, rapid response and portable air-fuel ratio analyzer. This article is based on the AFR calculation model microcomputer hardware and software system design, background monitoring software design and debugging and measurement system, and on the choice of universal oxygen sensor calibration laboratory, establishing a wide-range of oxygen sensor output voltage and AFR model. The main features of AFR analyzer are measurement and display of air-fuel ratio, excess air coefficient or oxygen content, via RS232 communication with host computer or via Control Area Network (CAN) bus and vehicle ECU communication function. Test results showed that the error can be controlled at ± 0.03 λ range when comparing the Analyzer measurement values to calculated values. Compared with American Innovate company LM-2 air-fuel ratio analyser, the maximum relative error measured is ±0.08 when exhaust flood or too dilute, the average measurement error is ±0.04 while λ is between 0.8 and 1.3.

Axle is one of the most important parts related to the safety of trailer’s operation on the railroad. In China, the checking of axles is mainly through visual inspection and hammer examinations. The construction of an automation hardware system was simulated by Pro/E, including support, guide rail, sensor, AC servomotor and servo driver conformed to the model. Next, for controlling the operation of the entire system, LabVIEW was used to write a program and to control the rotation of servomotor. The pressure heads controlled by linkage system were utilized to load the axle; meanwhile, through the PCI of NI, the data of pressure and displacement was collected. After calibration, the relative error of pinpointing sensor was 5.207%, and the average relative error was about 1.4%. Analyzed by SPSS, the voltage-load formula's correlation coefficient R is greater than 0.994, while the significance is less than 0.05, showing the regression of great significance. Finally, the capabilities of the simulated axle, such as fatigue, stiffness, intensity and stress, were analyzed carefully.
