The study was to efficiently degrade lignocellulose, ligninase high-producing strains were screened and bred by mutation and genome rearrangement. Phanerochaete chrysosporium was used as an original strain and treated with atmospheric room temperature plasma (ARTP) and diethyl sulphate (DES). Mutant strains with higher enzyme productivity were screened by the combination of aniline blue decolorization circle method and guaiacol color circle method and used as starting strains for genome shuffling. Through two rounds of genome rearrangement and genetic stability test. The results showed that the high-yield lignin enzyme fusion PR2-24 was obtained, and its lignin peroxidase activity, manganese peroxidase activity and laccase activity were 11.34, 88.35 and 10.82 U/mL respectively, which were 181%, 171% and 203% higher than those of the original strain. Pyoderma gangrenosum (PG) is a rare chronic neutrophilic dermatosis that causes undermining ulcers. Unfortunately, standardization of PG treatment remains a challenge. In this article, we describe a case in which a 69-year-old man presented with a painful ulcer on the right lower leg. The diagnosis of PG was made after excluding other diseases. He had a history of PG on his left lower leg 2 years earlier and was cured by the treatment of systemic corticosteroids and cyclosporin A for 43 days. However, such a treatment was not effective this time. Hence, we applied intravenous immunoglobulin and negative-pressure wound therapy, and the patient was cured. Altogether, this case supports the use of intravenous immunoglobulin as an effective adjuvant for refractory PG, and indicates negative-pressure wound therapy as a treatment option to advance ulcer healing under adequate immunosuppression. A short, enantioselective synthesis of (-)-maximiscin, a structurally intriguing metabolite of mixed biosynthetic origin, is reported. A retrosynthetic analysis predicated on maximizing ideality and efficiency led to several unusual disconnections and tactics. Formation of the central highly oxidized pyridone ring through a convergent coupling at the end of the synthesis simplified the route considerably. The requisite building blocks could be prepared from feedstock materials (derived from shikimate and mesitylene). Strategies rooted in hidden symmetry recognition, C-H functionalization, and radical retrosynthesis played key roles in developing this concise route.
