We investigated whether the associations that self-concept clarity had with indicators of psychological distress were mediated by the forms of escapism referred to as self-suppression (engaging in activities in an effort to avoid unpleasant mental processes) and self-expansion (engaging in activities in order to experience growth). The indirect associations that self-concept clarity had with psychological distress through self-suppression and self-expansion were examined in college students (Study 1; N = 270), Cosplayers (Study 2; N = 45), and Furries (Study 3; N = 191). Across these three studies, self-concept clarity had very large associations with psychological distress and negative associations with self-suppression that ranged from small to large in magnitude. Further, engaging in activities for the purpose of self-suppression mediated the associations that self-concept clarity had with various indicators of psychological distress with the most consistent associations emerging for depression. In contrast, self-concept clarity did not have consistent associations with the motivation to engage in activities for the purpose of self-expansion nor did self-expansion consistently mediate the associations that self-concept clarity had with the indicators of psychological distress. These findings indicate that engaging in activities for the purpose of self-suppression may play an important role in the link between low self-concept clarity and psychological distress.

Aim of the Study: The study aimed to develop and evaluate two new hot chocolate recipes (P1 – Red Recipe and P2 – Green Recipe) and compare them with a commercial hot chocolate product (P3 – “La Festa”). The focus was on creating recipes that are nutritionally comparable to natural or dietetic products, with reduced sugar and increased dietary fibre content. Materials and Methods: The study employed sensory evaluation methods, specifically the Mann-Whitney and Wilcoxon Sign Rank tests, to assess appearance, consistency, flavour, taste, and smell of the hot chocolate recipes. Additionally, a comprehensive nutritional analysis was conducted to compare the protein, lipid, carbohydrate, fibre, and caloric content of the recipes against the commercial product. Results: The sensory evaluation revealed that Recipe P1 significantly outperformed both Recipe P2 and the commercial product P3 in all tested attributes. Nutritionally, Recipes P1 and P2 were similar, both showing lower sugar and higher fibre content than typical market products. However, no significant sensory differences were noted between Recipe P2 and the commercial hot chocolate P3. Conclusions: The study concludes that Recipe P1 is superior in sensory qualities with a favourable nutritional profile, making it an appealing option for health-conscious consumers. Recipe P2, while nutritionally akin to P1, shares more sensory similarities with traditional hot chocolate, offering a healthier alternative without sacrificing familiar flavours. This research underscores the feasibility of creating healthier versions of popular beverages without compromising taste and sensory appeal.

Ragweed pollen allergy is the most common seasonal allergy in western Romania. Prolonged exposure to ragweed pollen may induce sensitization to pan-allergens such as calcium-binding proteins (polcalcins) and progression to more severe symptoms. We aimed to detect IgE sensitization to recombinant Amb a 9 and Amb a 10 in a Romanian population, to assess their potential clinical relevance and cross-reactivity, as well as to investigate the relation with clinical symptoms. rAmb a 9 and rAmb a 10 produced in Escherichia coli were used to detect specific IgE in sera from 87 clinically characterized ragweed-allergic patients in ELISA, for basophil activation experiments and rabbit immunization. Rabbit rAmb a 9- and rAmb a 10-specific sera were used to detect possible cross-reactivity with rArt v 5 and reactivity towards ragweed and mugwort pollen extracts. The results showed an IgE reactivity of 25% to rAmb a 9 and 35% to rAmb a 10. rAmb a 10 induced basophil degranulation in three out of four patients tested. Moreover, polcalcin-negative patients reported significantly more skin symptoms, whereas polcalcin-positive patients tended to report more respiratory symptoms. Furthermore, both rabbit antisera showed low reactivity towards extracts and showed high reactivity to rArt v 5, suggesting strong cross-reactivity. Our study indicated that recombinant ragweed polcalcins might be considered for molecular diagnosis.

Abstract Waning antibody responses after COVID-19 vaccination combined with the emergence of the SARS-CoV-2 Omicron lineage led to reduced vaccine effectiveness. As a countermeasure, bivalent mRNA-based booster vaccines encoding the ancestral spike protein in combination with that of Omicron BA.1 or BA.5 were introduced. Since then, different BA.2-descendent lineages have become dominant, such as XBB.1.5, JN.1, or EG.5.1. Here, we report post-hoc analyses of data from the SWITCH-ON study, assessing how different COVID-19 priming regimens affect the immunogenicity of bivalent booster vaccinations and breakthrough infections (NCT05471440). BA.1 and BA.5 bivalent vaccines boosted neutralizing antibodies and T-cells up to 3 months after boost; however, cross-neutralization of XBB.1.5 was poor. Interestingly, different combinations of prime-boost regimens induced divergent responses: participants primed with Ad26.COV2.S developed lower binding antibody levels after bivalent boost while neutralization and T-cell responses were similar to mRNA-based primed participants. In contrast, the breadth of neutralization was higher in mRNA-primed and bivalent BA.5 boosted participants. Combined, our data further support the current use of monovalent vaccines based on circulating strains when vaccinating risk groups, as recently recommended by the WHO. We emphasize the importance of the continuous assessment of immune responses targeting circulating variants to guide future COVID-19 vaccination policies.

(1) Background: The aim of this study was to analyze the impact of pharmacogenetic-guided antidepressant therapy on the 12-month evolution of the intensity of depressive symptoms in patients with recurrent depressive disorder (RDD) in comparison to a control group of depressive subjects who were treated conventionally. (2) Methods: This prospective longitudinal study was conducted between 2019 and 2022, and the patients were evaluated by employing the Hamilton Depression Rating Scale (HAM-D), Hamilton Anxiety Rating Scale (HAM-A) and the Clinical Global Impressions Scale: Severity and Improvement. We followed them up at 1, 3, 6, and 12 months. (3) Results: Of the 76 patients with RDD, 37 were tested genetically (Group A) and 39 were not (Group B). Although the patients from Group A had statistically significantly more severe MDD at baseline than those from Group B (p < 0.001), by adjusting their therapy according to the genetic testing, they had a progressive and more substantial reduction in the severity of RDD symptoms [F = 74.334; η2 = 0.674; p < 0.001], indicating a substantial association with the results provided by the genetic testing (67.4%). (4) Conclusions: In patients with RDD and a poor response to antidepressant therapy, pharmacogenetic testing allows for treatment adjustment, resulting in a constant and superior reduction in the intensity of depression and anxiety symptoms.

Abstract Metallosis with subsequent cardiac involvement is a possible long‐term complication of hip arthroplasty. We report the case of a young female referred to our centre for the suspicion of cardiac amyloidosis presenting with low electrocardiogram voltage, left ventricular hypertrophy, pericardial effusion, and global and longitudinal systolic impairment with apical sparing pattern. Her medical history was remarkable for arthroplasty in the context of congenital hip dysplasia. Two years prior to presentation, she underwent revision surgery for prosthesis malfunction, and tissue metallosis was initially documented. At the current presentation, cobalt metallosis was confirmed, as the circulating cobalt and chromium levels were severely elevated. The accurate diagnosis prompted the removal of the cobalt source with extensive tissue debridement and the use of chelating agents. Reversal of the cardiac abnormalities occurred as the circulating cobalt levels returned to normal.

Mangalia area harbors in the western Black Sea a distinctive marine environment thriving under specific hydrochemical conditions, largely influenced by a significant number of sulphurous springs occurring in shallow marine waters. These springs led to the designation of the area as part of the Natura 2000 Marine Protected Area (MPA) network at European level (Underwater Sulphurous Springs from Mangalia - ROSAC0094), as unique hydro-geomorphological features in the region. In 2021 and 2023, two research cruises led by GeoEcoMar investigated underwater sulphurous springs primarily located offshore of Mangalia (Constanţa County, Romania). The study area, located between 17-29 meters water depth and 1.8-3 km offshore, encompasses two marine protected areas: the Underwater Sulphurous Springs from Mangalia (ROSAC0094) and Cape Aurora (ROSCI0281). The research combined geophysical and geochemical techniques and sediment sampling. Considering the susceptibility of these natural systems to human activities such as fishing and dredging, as well as the impact of ecological and climate changes, this paper offers significant insights contributing to the development of effective conservation and management strategies for these environments. The surveys were conducted for benthic habitats mapping, with the objective of improving our understanding of these ecosystems’ distribution, composition and dynamics. As these sulphurous waters are rich in methane, a powerful greenhouse gas, our results also contribute to the inventory of greenhouse gas sources. The results presented in this paper provide valuable new insights into this specific environment, contributing to the understanding of its complex functioning and evolution.

The purpose of the study was identified by the effort to complement the standard sports training programme with a portfolio of innovative means and methods to optimise the process of achieving performance in gymnastics competitions. The group of subjects investigated was female and consisted of 19 gymnasts, aged 10 - 12 years. They were tested initially and finally to determine the effects that going through the training program had on the analysed indicators. The Matorin, Flamingo, Balance Plate Position Maintenance and the "Y" tests were used in the research. We used descriptive statistics (Arithmetic Mean, Standard Deviation, Median, Confidence Interval and Coefficient of Variation) and significance tests which were used to assess the statistical significance of the difference between the means for two sets of scores (STUDENT paired sample t-test and Wilcoxon test). Based on the results recorded, we identified a significant difference between the mean values for the two tests. Following the results obtained, we can confirm that they help to form an overall picture from the perspective of motor skills on gymnasts and that there is a significant effectiveness of the motor intervention program in the sports training process.

Orbital angular momentum of light and the associated Laguerre–Gauss modes have garnered increasing interest due to the many applications they promise in the fields of telecommunications and quantum information, as well as charged particle acceleration. Here, we use the numerical simulation of the dynamics of a large number of identical, charged, relativistic, but classical particles, forming a statistical ensemble and seen as test-particles in a time-dependent external electromagnetic field, to get insight into the process of a head-on collision with a pulsed Laguerre–Gauss mode. We have identified two steady-states regimes (low a0 and large a0), where the distribution of the final angular momentum gained by the particles as a function of their initial coordinates is well-behaved and does not change its structure with variations of the reduced amplitude. For small reduced amplitudes a0 of the incoming electromagnetic field, we have also found an analytic approximation of the expression for the angular momentum transferred to the particles after the pulse has left their region. Using both numerical integration and the analytic approximation, we have investigated the effects of the field’s gradient, initial phase of the field, polarization type, and azimuthal order m.

Objective: To describe the adaptations made and to examine interrater reliability and feasibility of administering a telehealth version of the Multiple Sclerosis Functional Composite (tele-MSFC). Design: The Multiple Sclerosis Functional Composite (MSFC) is a commonly used, in-person clinical outcome assessment. It is composed of the timed 25-Foot Walk Test (T25FWT), Nine-Hole Peg Test (NHPT), and Paced Auditory Serial Addition Test (PASAT). The MSFC was adapted for videoconference administration as part of a larger clinical trial. One of the adaptations included administering a timed 12.5-Foot Walk Test (T12.5FWT) for participants who did not have adequate space in their homes for the T25FWT. Participants, examiners, and raters completed surveys online about their satisfaction and experience with tele-MSFC. Setting: Participants underwent the tele-MSFC in their homes using a laptop or smartphone while examiners scored the tele-MSFC in real-time at a remote location. Participants: Community-dwelling adults (n=61) with mild-to-moderate multiple sclerosis (MS) symptoms. Interventions: Not applicable. Main Outcome Measure: Tele-MSFC. Results: Intraclass correlation coefficients (ICC) assessed interrater reliability between the examiner and 2 independent raters who later scored a recording of the tele-MSFC. Interrater reliability was excellent (ICC>0.90) for all tests, including the T12.5FWT. Participants were highly satisfied with tele-MSFC. However, challenges included adequate space for T25FWT, technical difficulties, and safety and privacy considerations of individuals with moderate impairments who were requested to have their caregivers present during testing. Conclusion: The tele-MSFC is reliable and feasible to administer with adaptations for community-dwelling adults with mild to moderate MS symptoms. Further validation of T12.5FWT is needed.

IntroductionSarcoidosis is a multi-system inflammatory disease of unknown origin with heterogeneous clinical manifestations varying from a single organ non-caseating granuloma site to chronic systemic inflammation and fibrosis. Gene expression studies have suggested several genes and pathways implicated in the pathogenesis of sarcoidosis, however, due to differences in study design and variable statistical approaches, results were frequently not reproducible or concordant. Therefore, meta-analysis of sarcoidosis gene-expression datasets is of great importance to robustly establish differentially expressed genes and signalling pathways.MethodsWe performed meta-analysis on 22 published gene-expression studies on sarcoidosis. Datasets were analysed systematically using same statistical cut-offs. Differentially expressed genes were identified by pooling of p-values using Edgington’s method and analysed for pathways using Ingenuity Pathway Analysis software.ResultsA consistent and significant signature of novel and well-known genes was identified, those collectively implicated both type I and type II interferon mediated signalling pathways in sarcoidosis. In silico functional analysis showed consistent downregulation of eukaryotic initiation factor 2 signalling, whereas cytokines like interferons and transcription factor STAT1 were upregulated. Furthermore, we analysed affected tissues to detect differentially expressed genes likely to be involved in granuloma biology. This revealed that matrix metallopeptidase 12 was exclusively upregulated in affected tissues, suggesting a crucial role in disease pathogenesis.DiscussionOur analysis provides a concise gene signature in sarcoidosis and expands our knowledge about the pathogenesis. Our results are of importance to improve current diagnostic approaches and monitoring strategies as well as in the development of targeted therapeutics.

The vortex photon beam induced nuclear reaction is studied. The interaction formalism of nuclei with vortex photons is developed and incorporated into the statistical reaction model to calculate reaction cross-sections. For 138 nuclei of high nuclear astrophysics and structure interest, the cross-sections of γ-ray emission and neutron production from the decay of the giant resonances (GR) populated by vortex photons, e.g., (γvo,γemit) and (γvo,nprod.), are computed. It is shown that for the (γvo,γemit) and (γvo,nprod.) cross-sections, the GR contribution of an individual L is either enhanced or suppressed depending on the parameters of vortex γ-rays, and the contribution from the GR of a specific L can be identified and deduced. To this end, a novel method to exclusively determine the γ-strength function (γSF) for the GR of a specific L is proposed considering the (γvo,γemit) and (γvo,nprod.) measurements, and the feasibility studies demonstrate that the γSF for the giant quadrupole resonance can be extracted. Furthermore, the astrophysical reaction rates of the vortex photon induced reactions in p-process are investigated. It is indicated that photo-nuclear reactions induced by vortex photons will bring new insights in nuclear physics and astrophysics research.

PurposePrevious studies have demonstrated that the majority of patients with an inborn error of immunity (IEI) develop a spike (S)-specific IgG antibody and T-cell response after two doses of the mRNA-1273 COVID-19 vaccine, but little is known about the response to a booster vaccination. We studied the immune responses 8 weeks after booster vaccination with mRNA-based COVID-19 vaccines in 171 IEI patients. Moreover, we evaluated the clinical outcomes in these patients one year after the start of the Dutch COVID-19 vaccination campaign.MethodsThis study was embedded in a large prospective multicenter study investigating the immunogenicity of COVID-19 mRNA-based vaccines in IEI (VACOPID study). Blood samples were taken from 244 participants 8 weeks after booster vaccination. These participants included 171 IEI patients (X-linked agammaglobulinemia (XLA;N=11), combined immunodeficiency (CID;N=4), common variable immunodeficiency (CVID;N=45), isolated or undefined antibody deficiencies (N=108) and phagocyte defects (N=3)) and 73 controls. SARS-CoV-2-specific IgG titers, neutralizing antibodies, and T-cell responses were evaluated. One year after the start of the COVID-19 vaccination program, 334 study participants (239 IEI patients and 95 controls) completed a questionnaire to supplement their clinical data focusing on SARS-CoV-2 infections.ResultsAfter booster vaccination, S-specific IgG titers increased in all COVID-19 naive IEI cohorts and controls, when compared to titers at 6 months after the priming regimen. The fold-increases did not differ between controls and IEI cohorts. SARS-CoV-2-specific T-cell responses also increased equally in all cohorts after booster vaccination compared to 6 months after the priming regimen. Most SARS-CoV-2 infections during the study period occurred in the period when the Omicron variant had become dominant. The clinical course of these infections was mild, although IEI patients experienced more frequent fever and dyspnea compared to controls and their symptoms persisted longer.ConclusionOur study demonstrates that mRNA-based booster vaccination induces robust recall of memory B-cell and T-cell responses in most IEI patients. One-year clinical follow-up demonstrated that SARS-CoV-2 infections in IEI patients were mild. Given our results, we support booster campaigns with newer variant-specific COVID-19 booster vaccines to IEI patients with milder phenotypes.

Tourism is called upon to play the role of facilitator of sustainable development, to lead collaboration with interest groups and local communities to decide jointly on the development of destinations. After the last difficult and challenging years, tourism has shown its resilience and ability to once again represent the percentage of GDP of many of the countries it owned before the pandemic. The WTO launched in 2021 the "Best Tourism Villages" initiative with the aim of transforming tourism into an engine for rural development and well-being and bringing about real change in local communities by creating jobs, supporting local businesses and preserving customs and traditions. The initiative recognizes villages that are an example of a rural tourist destination with cultural and natural assets, that preserve and promote the values, products and lifestyles of communities, and that have a clear commitment to innovation and sustainability. Romania is very well positioned at international level, from the point of view of rural tourism. Six localities have entered the competition "Best Tourism Villages" (2022), dedicated to the recognition of villages that are remarkable examples of rural tourist destinations. These are Biertan, Răşinari and Râu Sadului (Sibiu County), Ciocăneşti (Suceava), Costeşti (Vâlcea), Eibenthal (Mehedinţi). In the work we analysed the criteria that formed the basis for the selection of the three villages in Sibiu County, namely: the cultural and natural resources in the destination, the promotion and conservation of cultural resources, the economic, social and environmental sustainability, the governance and prioritization of tourism, infrastructure and connectivity, health, safety and security, as well as the photos and presentation videos. Răşinari village is the big winner of Romania's second edition of the world competition Best Tourism Villages, being selected by the specialists of the WTO, from over 130 applications, coming from 57 countries.

The present study concerned the connection between the desire for power and romantic commitment in LGBTQ relationships as well as the roles that indicators of relationship functioning (e.g., relationship satisfaction) and perceptions of power played in this association. These associations were examined in community members (N = 113) involved in an LGBTQ romantic relationship. The desire for power had a negative association with romantic commitment that was mediated by perceptions of the relationship and moderated by perceived power. Similar to the results of previous studies concerning heterosexual relationships, these results suggest that issues surrounding the desire for power may be intimately connected with romantic commitment in LGBTQ relationships.

The aim of the study is to perform a comparison between conventional impression versus intraoral and extraoral scanning, plaster models compared to virtual and printed models, in terms of working time and fidelity. Material and method: The study evaluates two groups of practitioners: 30 dentists and 30 dental technicians, regarding registration and reproduction of the prosthetic field. The time required to perform the operations was evaluated in relation to variables that can influence the final results. Conventional impressions were monophasic and biphasic. In the digital flow, an intraoral scanner (Medit, i700) and a laboratory scanner (Medit T310) were used. Also, a printing machine (Elegoo Saturn) was used to print the models. Results and discussions: The average time allocated for conventional impressions is between 8min and 20sec and 25min and 40sec. The scanning time varied between 5 min 30 sec and 12 min 40 sec. Conclusions: The digital impression requires less working time compared to the conventional one, the latter cannot be completely abandoned, at least in terms of mobile prostheses. The fidelity of the prosthodontic field reproduction influences the adaptation of the restorations.

Aim of the study is an ex-vivo time analysis comparing conventional and digital impression methods for prosthodontic restorations. Insights gleaned from ex-vivo experiments hold promise for enhancing existing practices. This primary experimental investigation will prioritize the consideration of both clinician and patient requirements when determining treatment modalities. Materials and Method: The study involved five stages: preparing an artificial arch, measuring time for gingival retraction with cords and paste, assessing time for conventional and digital impressions, and data analysis. A Frasaco maxillary model with 16 teeth was utilized, 14 prepared using cylindrical-conical burs. Gingival retraction cords and paste were applied, and impression times were recorded for both conventional and digital methods. Results and Discussions: Time analyses showed varying durations for gingival retraction techniques, with the paste method significantly faster than retraction cords. Digital impressions exhibited shorter scanning times compared to conventional methods, especially for fewer number of teeth. However, conventional two-step impressions took longer but provided better cervical area fidelity. This study highlighted the efficiency of digital scanning, offering shorter impression times and reduced patient discomfort. Research also emphasized the differences between retraction methods, with varied effects on periodontal tissues and impression quality. Comparison studies indicated benefits and challenges associated with different impression techniques. Conclusion: The findings underscored the need for practitioners to choose impression methods based on time efficiency and patient needs. Optical scanning proved quickest for fewer teeth, while retraction cord and paste methods require careful consideration. While the study didn’t explore gingival tissue effects, it highlighted crucial considerations for impression materials and chemical solutions. These insights offer practitioners guidance in selecting optimal techniques for effective and efficient dental procedures.

The present study explored potential factors that may mediate the relationship between narcissism and conspicuous consumption, as the underlying reasons for this connection remain uncertain. Specifically, we investigated the following variables as potential mediators of the links between narcissistic personality traits and conspicuous consumption: consumer need for uniqueness, materialism, approval seeking, material meaning, consumer sexual signaling, and need for social status. In a sample of 2,944 Israeli community members, our study revealed that extraverted, antagonistic, neurotic, and communal narcissism frequently exhibited positive indirect connections with conspicuous consumption through these mediating factors, with the most robust associations often being observed for antagonistic narcissism. However, a clear departure from this pattern became apparent when the anticipated mediation of the connections between narcissistic personality traits and conspicuous consumption via the need for social status did not unfold as predicted. In contrast, extraverted, antagonistic, and communal narcissism exhibited negative indirect associations with conspicuous consumption through the need for social status. Collectively, these findings indicate both commonalities and distinctions in how narcissistic personality traits are associated with conspicuous consumption via specific mediators. The discussion delves into the significance of these outcomes in shedding light on why conspicuous consumption holds allure for individuals with narcissistic tendencies.

ABSTRACT: Objectives: Elderly become more susceptible to lower respiratory tract infections, resulting in antibiotic prescriptions. Immunoglobulins (Ig) play an important role in host defense and protection against infections. Therefore, we aimed to investigate whether lower Ig levels are a risk factor for antibiotic use in the general elderly population. Methods: After exclusion of current antibiotic users, Cox proportional-hazards regression models were performed to investigate the effect of stable serum IgM, IgG and IgA levels on time to first antibiotic prescription within the Rotterdam Study. Regression models were adjusted for age, sex, body mass index, smoking status and diabetes. We introduced quadratic terms and additionally categorized Igs to explore and quantify potential non-linearity of the association. The restricted cubic splines technique was used to plot the natural log of the hazard across Ig level. Results: In total, 8,639 participants were included (mean age 64 years, 57% female, medium follow-up 3.2 years). No significant association between IgM and time to antibiotic prescription was observed. IgG and IgA levels (in g/L) showed a U-shaped relationship with time to antibiotic prescription (linear IgG HR 0.959, 95% CI 0.930–0.989; quadratic IgG² HR 1.002, 95% CI 1.000–1.003; linear IgA HR 0.949, 95% CI 0.910–0.990; quadratic IgA² HR 1.009, 95% CI 1.004–1.013). Conclusion: Both low and high IgG and IgA levels were associated with a higher incidence of antibiotic prescriptions in stable middle-aged and older individuals. Increased awareness for the potential increased infection risk when persons have low or high Ig levels, even within the reference ranges, is needed.

Abstract Background Inflammation plays a key role in the development of dementia, but its link to early biomarkers, particularly those in plasma or neuroimaging, remains elusive. This study aimed to investigate the association between serum immunoglobulins and biomarkers of dementia. Methods Between 1997 and 2009, serum immunoglobulins (IgA, IgG and IgM) were measured in dementia-free participants of the population-based Rotterdam Study. A random subset of participants had assessment of biomarkers in plasma (total tau (t-tau), neurofilament light chain (NfL), amyloid-β40 (Aβ-40), amyloid-β42 (Aβ-42), while another subset of participants underwent neuroimaging to quantify brain volume, white matter structural integrity and markers of cerebral small vessel disease. Linear regression models were constructed to determine cross-sectional associations between IgA, IgG, IgM and biomarkers of dementia, with adjustment for potential confounders. Multiple testing correction was applied using the false discovery rate. As a sensitivity analysis, we re-ran the models for participants within the reference range of immunoglobulins, excluding those using immunomodulating drugs, and conducted a stratified analysis by APOE-ε4 carriership and sex. Results Of 8,768 participants with serum immunoglobulins, 3,455 participants (65.8 years [interquartile range (IQR): 61.5–72.0], 57.2% female) had plasma biomarkers available and 3,139 participants (57.4 years [IQR: 52.7–60.7], 54.4% female) had neuroimaging data. Overall, no associations between serum immunoglobulins and biomarkers of dementia remained significant after correction for multiple testing. However, several suggestive associations were noted: higher serum IgA levels concurred with lower plasma levels of Aβ-42 (standardized adjusted mean difference: -0.015 [95% confidence interval (CI): -0.029−-0.002], p = 2.8 × 10–2), and a lower total brain volume, mainly driven by less gray matter (-0.027 [-0.046−-0.008], p = 6.0 × 10–3) and more white matter hyperintensities (0.047 [0.016 – 0.077], p = 3.0 × 10–3). In sensitivity analyses, higher IgM was linked to lower t-tau, Aβ-40, and Aβ-42, but also a loss of white matter microstructural integrity. Stratified analyses indicate that these associations potentially differ between carriers and non-carriers of the APOE-ε4 allele and men and women. Conclusions While associations between serum immunoglobulins and early markers of dementia could not be established in this population-based sample, it may be valuable to consider factors such as APOE-ε4 allele carriership and sex in future investigations.

The monumental trees growing in the cities, especially those from the parks, green spaces and alignments, which potentially cause a great risk to people, are required from time to time to be checked about their health conditions. The aim of this article was to assess the health state of the seven monumental trees from Brasov using ultrasonic tomography with an ARBOTOM 2D software (Rinn Tech, Germany). The material under research was the seven monumental trees that grows in Brasov area, respectively: silver fir tree, sweet chestnut tree, beech tree, ginkgo, horsechestnut tree, sycamore tree and plane tree, whose health status was checked using a sound tomograph which allows to assess the inside of the trunk in a non-invasive way. It could be observed that of the seven monumental trees investigated, two of them are in good status of health (Ginkgo tree and sycamore tree), three specimens show visible effect of decay processes of wood (65% horsechestnut tree, 75% sweet chestnut tree and 31% plane tree) and two specimens respectively silver fir tree and beech tree have the cross-section of their trunks with a beginning of rot in a percentage of 65%.

We investigated the roles that suspicious jealousy and reactive jealousy might play in the associations between borderline personality features (BPF) and mate retention behaviors. Study 1 (N = 406) found that BPF had positive indirect associations with benefit-provisioning behaviors and cost-inflicting behaviors through suspicious jealousy but not through reactive jealousy. Study 2 (N = 334 (a dyadic sample of 167 romantic couples)) revealed actor effects such that BPF had positive indirect associations with benefit-provisioning behaviors and cost-inflicting behaviors through suspicious jealousy for both men and women. In addition, the positive association between BPF and benefit-provisioning behaviors was mediated by reactive jealousy in women but not in men. The only partner effect that emerged from these analyses showed that BPF in women were negatively associated with the benefit-provisioning behaviors reported by their male partners. Discussion focuses on the implications of these results for the function that jealousy might serve in the strategies used by individuals with BPF to maintain their romantic relationships.

Aristolochic acids, compounds derived from Aristolochiaceae plant species, are associated with significant renal nephrotoxicity and carcinogenicity. Aristolochic acid I (AAI), the most predominant and potent of these compounds, is a primary etiological agent in acute and chronic kidney diseases such as Aristolochic Acid Nephropathy (AAN) and Balkan Endemic Nephropathy (BEN). Due to the kidneys’ critical role in xenobiotic excretion, they are the primary organs affected by AAI toxicity. Recent in vitro and in vivo studies have highlighted mitochondrial dysfunction as a crucial factor in the pathogenesis of these kidney diseases. This review provides an update on the recent advances in understanding the causes of acquired mitochondrial dysfunction within the context of AAN and BEN. Key findings include the identification of mitochondrial DNA depletion, loss of mitochondrial membrane potential, and decreased ATP production as significant contributors to kidney damage. Additionally, oxidative stress markers and inflammatory mediators have been implicated in disease progression. Potential therapeutic approaches, such as the use of antioxidants like vitamin C and catalpol, have shown promise in mitigating AAI-induced cytotoxicity. Furthermore, future predictive approaches like pharmacogenomics could pave the way for novel mitochondria-targeted treatments. A comprehensive characterization of mitochondrial function, its underlying molecular mechanisms, and specific biomarkers could offer valuable insights and potential therapeutic options, significantly impacting the current management of AAN and BEN.

The topic of ragweed pollen (RW) versus house dust mites (HDMs) has often been deliberated, but the increasing incidence of co-sensitization between them has been scarcely addressed. Utilizing Sprague Dawley rats, we explored the effects of co-sensitization with the combination of HDMs and RW pollen extracts in correlation with high-fructose diet (HFrD) by in vitro tracheal reactivity analysis in isolated organ bath and biological explorations. Our findings unveiled interrelated connections between allergic asthma, dyslipidemia, and HFrD-induced obesity, shedding light on their compounding role through inflammation. The increased CRP values and airway hyperresponsiveness to the methacholine challenge suggest a synergistic effect of obesity on amplifying the existing inflammation induced by asthma. One of the major outcomes is that the co-sensitization to HDMs and RW pollen led to the development of a severe allergic asthma phenotype in rats, especially in those with HFrD. Therefore, the co-sensitization to these allergens as well as the HFrD may play a crucial role in the modulation of systemic inflammation, obesity, and airway reactivity.

This research investigates novel polymeric composite materials for automotive interior trim applications. The composites utilize recycled polypropylene (PPr) matrix and carboxymethylcellulose (CMC) as filler (PPr/CMC: 100/0, 95/5, and 90/10 wt.%). The materials were processed by extrusion and injection molding. Considering their intended application, the composites were evaluated for resistance to key climatic factors, i.e., temperature, humidity, and UV radiation. In addition, structural analyses and FTIR analyses were performed to assess potential heterogeneity and thermal stability. Following FTIR tests, the incorporation of carboxymethyl cellulose in polypropylene is confirmed by the detection of characteristic CMC bands for -OH, C=O, and C-O-C groups. The results indicate slight structural heterogeneity in the 5% and 10% CMC composites. However, no thermal distortions were observed in either the composites or the PPr matrix itself. The behavior of PPr/CMC composites under the action of the mentioned climatic factors has been assessed from the variation of dielectric characteristics with frequency. The strong polarization of CMC leads to a sharp increase in composites electrical conductivity after submersion in water for 480 h, suggesting weakening of the composite structure. After exposure to UV radiation, a sharp increase in conductivity is observed even after the first cycle (72 h) of UV radiation. Following the experimental results obtained in our study, it is recommended to use the PPr +10% CMC composite for obtaining different interior ornaments (carpets, supports, etc.). At the same time, the use of these materials also has the advantage of lightening the mass of the vehicle due to their lower density than polymers.

Background and Objectives: Dentigerous cysts are one of the most frequent pathologies associated with unerupted or impacted teeth. Such cysts show a male predilection and a preference for the mandibular region. Also, they commonly occur in the second and third decades of life, with only 9% occurring in the first decade. The aim of this work is to apply and study the therapeutic algorithms developed for dentigerous cysts and their outcomes, from the early diagnostic stage to the complete healing phase of pediatric patients diagnosed with this medical condition. Materials and Methods: The study included 19 pediatric patients diagnosed with dentigerous cysts who underwent the enucleation and extraction or conservative attitude of the associated tooth. The bony healing was also followed-up 9 months after the surgery. Results: A higher incidence in the posterior area of the mandible and maxilla was observed, as well as a higher incidence in boys. The 9 months postoperative radiographic assessment showed that the bony defects were completely healed. Conclusions: A thorough understanding of the nature of the lesion backed by a good clinical history and by state-of-the-art radiographic and radiologic examinations can go a long way in helping the surgeon to choose the correct therapeutic approach and to ameliorate the medical condition in the best long-term interest of the young patient. The considered dentigerous cyst cases demonstrated that an early diagnosis and treatment of this pathology is followed by a responsive treatment.

Perinatal depression (PPD) presents a significant public health concern, often influenced by psychological and personality factors. This study investigated the impact of personality traits, particularly neuroticism, and obsessive-compulsive disorder (OCD) symptoms on the severity of PPD. The primary aim was to quantify the contributions of these factors to the risk and severity of PPD to enhance early intervention strategies. A total of 47 pregnant women with depressive symptoms per DSM-5 criteria at “Pius Brinzeu” County Emergency Hospital in Timisoara, Romania, were enrolled in this cross-sectional study, as well as 49 women without depressive symptoms as controls. Personality traits were assessed using the NEO Five-Factor Inventory (NEO-FFI), and OCD symptoms were measured using the Obsessive-Compulsive Inventory (OCI). Depression severity was evaluated using the Edinburgh Postnatal Depression Scale (EPDS). This set of questionnaires were administered antepartum and postpartum. The logistic regression analysis highlighted neuroticism as a significant predictor of PPD severity, with an increase in neuroticism associated with a higher risk of PPD (coefficient = 0.24, p < 0.001). Conversely, openness showed a protective effect (coefficient = −0.13, p = 0.009). Higher OCD symptomatology, particularly ordering and hoarding, were linked with increased depression scores. Specifically, the total OCI score significantly predicted the EPDS score (coefficient = 0.03, p = 0.003). Furthermore, significant increases in EPDS anxiety and depression scores were observed in the perinatal period, indicating worsening of symptoms (anxiety coefficient = 0.51; p < 0.001). The findings suggest that personality traits like neuroticism and OCD symptoms significantly contribute to the severity of PPD. Interventions targeting these specific traits could potentially mitigate the risk and severity of perinatal depression, underscoring the need for personalized treatment plans that consider these psychological dimensions.

Nigeria sits at the crossroads of West and Central Africa; two increasingly critical regions for global health security. To strengthen cross-border collaboration for health security between its neighbors from West and Central Africa, the Federal Ministry of Health, Nigeria through the public health authority at the Points of Entry—Port Health Services, led the design of a multi-lateral framework for cross-border collaboration between Nigeria, the Republic of Benin, the Republic of Chad, the Republic of Cameroon and the Republic of Niger. This framework provides a platform for agreeing on bilateral national and district-level infectious disease surveillance information sharing as well as laying the groundwork for expanded collaboration in incident reporting, joint investigation and response across borders.

Recognizing the crucial role that transport and lifting installations play in various sectors, including industry, construction, and extraction, as well as in mechanizing loading and unloading operations, and the movement of equipment or materials, special attention must be oriented to the diversification and the production of these installations by using the most advanced technologies. The paper presents some operating principles and characteristics of a lifting and transport Spider Crane installation equipment. This installation is considered a more economical and efficient alternative to traditional cranes for works where lifting requirements are below certain limits and significant height. Thus, an analysis of the hydraulic lifting system was carried out, based on the calculation of hydraulic fluid flows for the drive pumps. Also, was evaluated the specific speeds and forces of the hydraulic cylinders, a mechanical analysis of the clamping system with hook, as well as a finite element analysis of some mechanical and hydraulic components.

A nanocrystalline Fe(Co) phase dispersed within an amorphous matrix was achieved after milling of Fe31Co31B30Ta8 powder mixtures up to 200 h by using a high-energy ball mill. The transformations occurring in the powders during milling were studied with the use of X-ray diffraction and scanning electron microscopy techniques. The transformation of the phase depends upon the milling time. With the increase of milling time, various nanostructured phases such as Fe(Co), Co2Ta, Co(Ta,B), fcc-Co, Fe-type borides, and amorphous phase were identified. Magnetic properties were also investigated and correlated with microstructural changes. By increasing milling time, irregular variation of saturation magnetization is identified in relation to the change in the composition of the system. Also, the gradual decrease in grain size to a few nanometers resulted in an increase in coercivity while increasing the milling time up to 200 h. The existence of non-magnetic Ta and B atoms in the neighboring Fe and Co atoms and the increase in the volume fraction of the amorphous phase are retained as the major factors of this hard magnetic behavior of the present system.

The oral cavity’s soft and hard tissues create a conducive environment for microbial proliferation and biofilm development, facilitating the colonization of prosthodontic and implant materials such as titanium (Ti) and zirconia (Zr). This study aimed to compare the efficacy of conventional decontamination methodologies (i.e., chemical and mechanical, using 0.12% digluconate chlorhexidine (CHX) solution-treatment and airflow) to adjunctive laser-based interventions on Ti and Zr substrates inoculated with Staphylococcus (S.) aureus ATCC 25923. Additionally, this investigation sought to elucidate the impact of these treatments on temperature variations and surface integrity, analyzing the laser irradiation effects on these prevalent dental materials. Experimental configurations were delineated for both Ti and Zr samples across four groups: (1) a conventional treatment group (CV); (2) a photodynamic therapy group (PDT); (3) an Er:YAG laser treatment group (Er); (4) a combined PDT and Er:YAG treatment group (PDTEr). Also, a negative control group (C) that received no treatment was considered. The decontamination of the inoculated disc samples was evaluated by quantifying the microbial colonies in colony-forming units per milliliter (CFU/mL). Temperature variations on the surface of the samples were determined during laser treatments. Surface modifications were investigated using scanning electron microscopy (SEM) and optical coherence tomography (OCT). For statistical analysis, Fisher 95% confidence intervals, Hsu’s MCB method, and the Kruskal–Wallis test were applied. With regard to the 105 CFU/mL of the negative control group, results indicated average values equal for each study group to (1) 2.66 CFU/mL for Ti and 2 CFU/mL for Zr for the CV group; (2) 0.33 CFU/mL for Ti and 1 CFU/mL for Zr for the PDT group; (3) 1.25 CFU/mL for Ti and 0 CFU/mL for Zr for the Er group; (4), and 0 CFU/mL for both Ti and Zr for the PDTEr group. Therefore, the combined PDT and Er:YAG treatment (PDTEr) and the singular PDT modality outperformed conventional decontamination methods in eradicating S. aureus biofilms from both Ti and Zr surfaces. Notably, the PDTEr regime achieved a comprehensive elimination of microbial colonies on treated substrates. Surface examination employing OCT demonstrated discernible alterations in the surface morphology of samples subjected to Er:YAG and combined PDT and Er:YAG treatments. Temperature checks during treatments showed no major changes, suggesting the applied laser methods are safe. In conclusion, PDTEr and PDT eliminated bacteria more effectively, but Zr surfaces were more resilient, making them better for microbe-controlling applications. Also, the study demonstrated that the (less costly but lower resolution) OCT method can replace SEM for such investigations.

Posture results from intricate interactions between the central nervous system's control mechanisms, sensory integration, and musculoskeletal responses, encompassing two levels: sustaining tonic muscle activity for posture and making adjustments to maintain balance in response to disturbances. Due to its complexity, assessing posture with simple methods is challenging, necessitating the use of advanced techniques like stabilometric platforms to measure postural stability through the center of pressure oscillations, akin to an inverted pendulum concept, offering a comprehensive understanding of postural control mechanisms. Materials and Methods: The study aims to analyze the trajectory of the center of pressure (COP) in relation to the gender of the subjects, through directional indices (DI) and oscillation ratio (OR), in the process of posture maintenance. The study included a sample of 50 young subjects (21-24 years), sports practitioners, divided into two equal groups. Results: The study findings indicate that gender does not influence COP DI. However, there's a significant disparity between anteroposterior (AP) DI values (0.70) and mediolateral (ML) DI values (0.54). The results suggest a predominant reliance on AP control mechanisms for maintaining balance, regardless of gender. When visual input is absent, there's a slight increase in AP DI (0.72) and a decrease in ML DI (0.52), indicating a shift in balance strategies. In terms of OR, significant differences are observed based on gender, oscillation direction, and visual input. Female subjects exhibit higher AP OR (6.5) compared to males (5.1). Under closed eyes conditions, both genders show decreased AP OR (6.0 for females, 4.7 for males), indicating a reliance on alternative balance strategies in the absence of visual cues. Conclusion: The study highlights the complexity of postural control and empha-sizes the influence of factors like gender and visual input on postural stability metrics, providing valuable insights for further research and clinical practice. The analysis reveals that around 60% of COP oscillations occur in the AP direction, regardless of visual input changes. This underscores the critical role of AP control in maintaining balance, suggesting targeted interventions to enhance stability should prioritize this aspect.

Misophonia is a neurophysiological disorder with behavioral implications, is complex and multifactorial in origin, and is characterized by an atypical and disproportionate emotional response to specific sounds or associated visual stimuli. Triggers include human-generated sounds, mainly sounds related to feeding and breathing processes, and repetitive mechanical sounds. In response to the triggering stimulus, the patient experiences immediate, high-intensity, disproportionate physical and emotional reactions that affect their quality of life and social functioning. The symptoms of misophonia can occur at any age, but onset in childhood or adolescence is most common. Affected children live in a constant state of anxiety, suffer continuous physical and emotional discomfort, and are thus exposed to significant chronic stress. Chronic stress, especially during childhood, has consequences on the main biological systems through the dysregulation of the hypothalamic–pituitary–adrenal axis, including the gastrointestinal tract. Here, we provide arguments for a positive correlation between misophonic pathology and gastrointestinal symptoms, and this hypothesis may be the starting point for further longitudinal studies that could investigate the correlations between these childhood vulnerabilities caused by misophonia and their effect on the gastrointestinal system. Further research to study this hypothesis is essential to ensure correct and timely diagnosis and optimal psychological and pharmacological support.

Allergic diseases are showing increasing prevalence in Western societies. They are characterized by a heightened reactivity towards otherwise harmless environmental stimuli. Allergic diseases showing a wide range of severity of symptoms have a significant impact on the quality of life of affected individuals. This study aims to highlight the mechanisms that induce these reactions, how they progress, and which prenatal factors influence their development. Most frequently, the reaction is mediated by immunoglobulin E (IgE) produced by B cells, which binds to the surface of mast cells and basophils and triggers an inflammatory response. The antibody response is triggered by a shift in T-cell immune response. The symptoms often start in early childhood with eczema or atopic dermatitis and progress to allergic asthma in adolescence. An important determinant of allergic diseases seems to be parental, especially maternal history of allergy. Around 30% of children of allergic mothers develop allergic sensitization in childhood. Genes involved in the regulation of the epithelial barrier function and the T-cell response were found to affect the predisposition to developing allergic disorders. Cord blood IgE was found to be a promising predictor of allergic disease development. Fetal B cells produce IgE starting at the 20th gestation week. These fetal B cells could be sensitized together with mast cells by maternal IgE and IgE–allergen complexes crossing the placental barrier via the low-affinity IgE receptor. Various factors were found to facilitate these sensitizations, including pesticides, drugs, exposure to cigarette smoke and maternal uncontrolled asthma. Prenatal exposure to microbial infections and maternal IgG appeared to play a role in the regulation of T-cell response, indicating a protective effect against allergy development. Additional preventive factors were dietary intake of vitamin D and omega 3 fatty acids as well as decreased maternal IgE levels. The effect of exposure to food allergens during pregnancy was inconclusive, with studies having found both sensitizing and protective effects. In conclusion, prenatal factors including genetics, epigenetics and fetal environmental factors have an important role in the development of allergic disorders in later life. Children with a genetic predisposition are at risk when exposed to cigarette smoke as well as increased maternal IgE in the prenatal period. Maternal diet during pregnancy and immunization against certain allergens could help in the prevention of allergy in predisposed children.

Pollen from common ragweed is an important allergen source worldwide and especially in western and southern Romania. More than 100 million patients suffer from symptoms of respiratory allergy (e.g., rhinitis, asthma) to ragweed pollen. Among the eleven characterized allergens, Amb a 6 is a non-specific lipid transfer protein (nsLTP). nsLTPs are structurally stable proteins in pollen and food from different unrelated plants capable of inducing severe reactions. The goal of this study was to produce Amb a 6 as a recombinant and structurally folded protein (rAmb a 6) and to characterize its physicochemical and immunological features. rAmb a 6 was expressed in Spodoptera frugiperda Sf9 cells as a secreted protein and characterized by mass spectrometry and circular dichroism (CD) spectroscopy regarding molecular mass and fold, respectively. The IgE-binding frequency towards the purified protein was evaluated using sera from 150 clinically well-characterized ragweed-allergic patients. The allergenic activities of rAmb a 6 and the nsLTP from the weed Parietaria judaica (Par j 2) were evaluated in basophil activation assays. rAmb a 6-specific IgE reactivity was associated with clinical features. Pure rAmb a 6 was obtained by insect cell expression. Its deduced molecular weight corresponded to that determined by mass spectrometry (i.e., 10,963 Da). rAmb a 6 formed oligomers as determined by SDS-PAGE under non-reducing conditions. According to multiple sequence comparisons, Amb a 6 was a distinct nsLTP with less than 40% sequence identity to currently known plant nsLTP allergens, except for nsLTP from Helianthus (i.e., 52%). rAmb a 6 is an important ragweed allergen recognized by 30% of ragweed pollen allergic patients. For certain patients, rAmb a 6-specific IgE levels were higher than those specific for the major ragweed allergen Amb a 1 and analysis also showed a higher allergenic activity in the basophil activation test. rAmb a 6-positive patients suffered mainly from respiratory symptoms. The assumption that Amb a 6 is a source-specific ragweed allergen is supported by the finding that none of the patients showing rAmb a 6-induced basophil activation reacted with Par j 2 and only one rAmb a 6-sensitized patient had a history of plant food allergy. Immunization of rabbits with rAmb a 6 induced IgG antibodies which strongly inhibited IgE binding to rAmb a 6. Our results demonstrate that Amb a 6 is an important source-specific ragweed pollen allergen that should be considered for diagnosis and allergen-specific immunotherapy of ragweed pollen allergy.

Currently, allergen-specific immunotherapy (AIT) for ragweed allergy is still based on natural allergen extracts. This study aimed to analyse the ability of four commercially available AIT vaccines (CLUSTOID, TYRO-SIT, POLLINEX Quattro Plus and Diater Depot) regarding their ability to induce IgG antibodies against ragweed pollen allergens in rabbits. Accordingly, the IgG reactivity of AIT-induced rabbit sera was tested for ten different ragweed pollen allergens (Amb a 1, 3, 4, 5, 6, 8, 9, 10, 11 and 12) by an ELISA. Furthermore, the ability of rabbit AIT-specific sera to block allergic patients’ IgE binding to relevant ragweed allergens (Amb a 1, 4, 6, 8 and 11) and to inhibit allergen-induced basophil activation was evaluated by an IgE inhibition ELISA and a mediator release assay. Only two AIT vaccines (Diater Depot > CLUSTOID) induced relevant IgG antibody levels to the major ragweed allergen Amb a 1. The IgG responses induced by the AIT vaccines against the other ragweed allergens were low and highly heterogeneous. Interestingly, the kinetics of IgG responses were different among the AIT vaccines and even within one AIT vaccine (Diater Depot) for Amb a 1 (long-lasting) versus Amb a 8 and Amb a 11 (short-lived). This could be due to variations in allergen contents, the immunogenicity of the allergens, and different immunization protocols. The IgE inhibition experiments showed that rabbit AIT-specific sera containing high allergen-specific IgG levels were able to inhibit patients’ IgE binding and prevent the mediator release with Diater Depot. The high levels of allergen-specific IgG levels were associated with their ability to prevent the recognition of allergens by patients’ IgE and allergen-induced basophil activation, indicating that the measurement of allergen-induced IgG could be a useful surrogate marker for the immunological efficacy of vaccines. Accordingly, the results of our study may be helpful for the selection of personalized AIT vaccination strategies for ragweed-allergic patients.

In this article we present the first investigation of the stable isotope composition of groundwater in Romania, East-Central Europe, with a focus on the karst areas. Our aim is twofold: (1) to provide a countrywide map with the distribution of stable oxygen and hydrogen isotope ratios in groundwater, and (2) to assess the recharge patterns of karst water. We collected more than 600 water samples from springs and wells across Romania for stable isotope analyses and monitored in detail the stable isotope composition of the waters as they pass through five cave systems. Our data show a spatial distribution of the stable isotope composition of the groundwater with low values in the mountainous area and high values in the surrounding lowlands and the central Transylvanian Depression. However, waters in karst areas induce departures from this distribution, resulting from the fast (hours to days) transfer of waters from high (ponor) to low (spring) altitudes. Water emerging from the karst springs has generally lower δ values than before sinking through the ponors, thus indicating a substantial contribution of winter recharge through diffuse infiltration and seepage. This contribution results in overall dilution of the water entering through ponors, likely resulting in changes in the chemical composition of the water and diluting potential pollutants. Our data call for careful separation between karst and non-karst spring/well waters, as indiscriminate common treatment might lead to erroneous interpretations.

Common ragweed pollen allergy has become a health burden worldwide. One of the major allergens in ragweed allergy is Amb a 1, which is responsible for over 90% of the IgE response in ragweed-allergic patients. The major allergen isoform Amb a 1.01 is the most allergenic isoform in ragweed pollen. So far, no recombinant Amb a 1.01 with similar allergenic properties to its natural counterpart (nAmb a 1.01) has been produced. Hence, this study aimed to produce a recombinant Amb a 1.01 with similar properties to the natural isoform for improved ragweed allergy management. Amb a 1.01 was expressed in insect cells using a codon-optimized DNA construct with a removable N-terminal His-Tag (rAmb a 1.01). The recombinant protein was purified by affinity chromatography and physicochemically characterized. The rAmb a 1.01 was compared to nAmb a 1.01 in terms of the IgE binding (enzyme-linked immunosorbent assay (ELISA), immunoblot) and allergenic activity (mediator release assay) in well-characterized ragweed-allergic patients. The rAmb a 1.01 exhibited similar IgE reactivity to nAmb a 1.01 in different IgE-binding assays (i.e., IgE immunoblot, ELISA, quantitative ImmunoCAP inhibition measurements). Furthermore, the rAmb a 1.01 showed comparable dose-dependent allergenic activity to nAmb a 1.01 regarding basophil activation. Overall, the results showed the successful expression of an rAmb a 1.01 with comparable characteristics to the corresponding natural isoform. Our findings provide the basis for an improvement in ragweed allergy research, diagnosis, and immunotherapy.

This article develops two metaheuristics optimization techniques, Grey Wolf Optimizer (GWO) and Harris Hawks Optimization (HHO), to handle multi-objective optimal power flow (MOOPF) issues. Multi Objective GWO (MOGWO) and Multi Objective HHO (MOHHO) are the names of the developed techniques. By combining these optimization techniques with Pareto techniques, the non-dominated solution set can be obtained. These developed approaches are characterized by simplicity and have few control parameters. Fuel cost, emissions, real power losses, and voltage deviation were the four objective functions considered. The theories used to determine the best compromise solution and organize the Pareto front options are the fuzzy membership equation and the crowding distance approach, respectively. To validate and evaluate the performance of the presented techniques, two standard IEEE bus systems—30-bus and 57-bus power systems—were proposed. Bi, Tri, and Quad objective functions with 21 case studies are the types of objective functions and the scenarios that were applied in this paper. As compared to the results of the most recent optimization techniques documented in the literature, the comparative analysis results for the proposed methodologies demonstrated the superiority and robustness of MOGWO and MOHHO.

This review aims to explore the intricate relationship among epigenetic mechanisms, stress, and affective disorders, focusing on how early life experiences and coping mechanisms contribute to susceptibility to mood disorders. Epigenetic factors play a crucial role in regulating gene expression without altering the DNA (deoxyribonucleic acid) sequence, and recent research has revealed associations between epigenetic changes and maladaptive responses to stress or psychiatric disorders. A scoping review of 33 studies employing the PRISMA-S (Preferred Reporting Items for Systematic Reviews and Meta-Analyses—Statement) guidelines investigates the role of stress-induced epigenetic mechanisms and coping strategies in affective disorder occurrence, development, and progression. The analysis encompasses various stress factors, including childhood trauma, work-related stress, and dietary deficiencies, alongside epigenetic changes, such as DNA methylation and altered gene expression. Findings indicate that specific stress-related genes frequently exhibit epigenetic changes associated with affective disorders. Moreover, the review examines coping mechanisms in patients with bipolar disorder and major depressive disorder, revealing mixed associations between coping strategies and symptom severity. While active coping is correlated with better outcomes, emotion-focused coping may exacerbate depressive or manic episodes. Overall, this review underscores the complex interplay among genetic predisposition, environmental stressors, coping mechanisms, and affective disorders. Understanding these interactions is essential for developing targeted interventions and personalized treatment strategies for individuals with mood disorders. However, further research is needed to elucidate specific genomic loci involved in affective disorders and the clinical implications of coping strategies in therapeutic settings.

Postpartum depression (PPD) is a significant global health concern with profound implications for mothers, families, and societies. This systematic review aims to synthesize current research findings to understand better how personality traits, as assessed by the NEO Five-Factor Inventory (NEO-FFI), contribute to the development and progression of PPD. Conducted in January 2024, this review searched major databases like PubMed, PsycINFO, and Scopus up to December 2023, focusing on the NEO-FFI’s role in evaluating PPD. Following PRISMA guidelines, studies were selected based on strict criteria, including the exclusive use of NEO-FFI for personality assessment and a focus on postpartum women. A total of seven studies were included after a rigorous two-step screening process, and their data were qualitatively synthesized. The review covered a total of 4172 participants, with a prevalence of clinically significant postpartum depression symptoms ranging from 10.6% to 51.7%. Notably, Neuroticism emerged as a significant predictor of PPD, with odds ratios ranging from 1.07 (95% CI: 0.96–1.20) in some studies to as high as 1.87 (95% CI: 1.53–2.27) in others. In contrast, traits like Extraversion and Conscientiousness generally showed protective effects, with lower scores associated with reduced PPD risk. For instance, Extraversion scores correlated negatively with PPD risk (Beta = −0.171) in one study. However, the impact of other traits such as Openness and Agreeableness on PPD risk was less clear, with some studies indicating negligible effects. The review highlights Neuroticism as a consistent and significant predictor of PPD risk, with varying impacts from other personality traits. The findings suggest potential pathways for targeted interventions in maternal mental health care, emphasizing the need for comprehensive personality evaluations in prenatal and postnatal settings.

This paper presents the obtaining and characterization of recycled polypropylene/strontium ferrite (PP/SrFe12O19) polymer composite materials with applications in the electromagnetic shielding of vehicle interiors (mainly automotive electronics—carcasses) from the electromagnetic radiation emitted mainly by exterior sources—electrical lines and supply sources—in terms of the development of the new electrical vehicles. With this aim, suitable polymer composite materials were developed using SrFe12O19 filler in two forms (powder and concentrate). The recycled PP polymer and composite materials with a PP/SrFe12O19 weight ratio of 75/25 and 70/30 were obtained in two stages, i.e., pellets by extrusion and samples for testing through a melt injection process. The characterization of the obtained materials took into account the requirements imposed by the desired applications. It consisted of determining the mechanical and dielectric properties, and microstructure analyses, along with the determination of the resistance to the action of a temperature of 70 °C, which is higher than the temperatures created during the summer inside vehicles. The performance of these materials as electromagnetic shields was assessed through functional tests consisting of the determination of magnetic permeability and the estimation of the electromagnetic shielding efficiency (SE). The obtained results confirmed the improvement of the mechanical, dielectric, and magnetic properties of the PP/SrFe12O19 composites compared to the selected PP polymers. It is also found that all the composite materials exhibited reflective shielding properties (SER from −71.5 dB to −56.7 dB), with very little absorption shielding. The most performant material was the composite made of PP/SrFe12O19 powder with a weight ratio of 70/30. The promising results recommend this composite material for potential use in automotive shielding applications against electromagnetic pollution.

The yeast Yarrowia lipolytica degrades petroleum compounds, including alkanes, via the monoterminal oxidation pathway, the hydrophobic carbon substrate assimilation is mediated by biosurfactants, and extracellular amphiphilic molecules are produced by the yeast cell. This study focuses on the ability of the strain Y. lipolytica CMGB32 to degrade n-hexadecane by producing biosurfactants with high potential for bioremediation. The hydrocarbon-degrading potential of the yeast strain was observed via a 2,6-dichlorophenolindophenol (DCPIP) test in Bushnell–Hass medium with 1% n-hexadecane, and cell hydrophobicity was expressed as microbial adhesion to hydrocarbons (MATH). Biosurfactant production on yeast peptone (YP) with 1% n-hexadecane was estimated after 72 h using the emulsification index (E24%) against toluene. Crude biosurfactant (cell-free broth) stability tests were performed at different temperatures (4 °C, 70 °C) and NaCl concentrations (2–10%). The effects of a biosurfactant on synthetic wastewater remediation comprised the growth curves (OD measurements) of natural heavy metal degrader Rhodotorula mucilaginosa, determination of nutrients (spectrophotometrically), physico-chemical parameters, and removal capacity of lead and cadmium ions (via inductively coupled plasma mass spectrometry—ICP-MS). The antimicrobial and anti-adherence activities of 20 mg/mL and 40 mg/mL of the biosurfactant against pathogenic Candida krusei strains involved growth observations and the crystal violet microtiter method. The DCPIP decolorization occurred after six days, corresponding to the maximum growth phase of the Y. lipolytica culture. After 72 h, the cells presented high hydrophobicity (82.61% MATH) and stable biosurfactant production (E24% 47%). The crude biosurfactant (5%) increased the growth of R. mucilaginosa strains cultivated on synthetic wastewater cultures contaminated with Pb2+ and Cd2+, increased the conductivity and COD (86%) of the samples, and determined Pb2+ (66%) and Cd2+ (42%) ions reduction. The concentrated biosurfactant inhibited C. krusei growth (70%) and biofilm adherence. In conclusion, Y. lipolytica CMGB32 shows important potential for development of biosurfactant-based technologies for the remediation of heavy-metal- and emerging pathogen-contaminated wastewaters.

Conventional techniques that measure the concentration of light elements in metallic materials lack high-resolution performance due to their intrinsic limitation of sensitivity. In that context, scanning microwave microscopy has the potential to significantly enhance the quantification of element distribution due to its ability to perform a tomographic investigation of the sample. Scanning microwave microscopy associates the local electromagnetic measurement and the nanoscale resolution of an atomic force microscope. This technique allows the simultaneous characterization of oxygen concentration as well as local mechanical properties by microwave phase shift and amplitude signal, respectively. The technique was calibrated by comparison with nuclear reaction analysis and nanoindentation measurement. We demonstrated the reliability of the scanning microwave technique by studying thin oxygen-enriched layers on a Ti-6Al-4V alloy. This innovative approach opens novel possibilities for the indirect quantification of light chemical element diffusion in metallic materials. This technique is applicable to the control and optimization of industrial processes.

The impression materials utilized today in dental medicine offer a good reproducibility and are easily accepted by patients. However, because they are polymer-based, they have issues regarding their dimensional stability. In this respect, the present work proposes a new type of dental impression, which is reinforced with rigid mouthguards. The aim of the study is to test the performances of such new impressions by comparing them to conventional ones—from this critical point of view, of the dimensional stability. Three types of polymeric materials were considered for both types of impressions: alginate, condensation silicone, and addition silicone. In order to obtain the new type of impressions, a manufacturing technique was developed, comprising the following phases: (i) conventional impressions were made; (ii) a plaster model was duplicated, and 15 rigid mouthguards were obtained; (iii) they were inserted in the impression technique, with each mouthguard positioned on the cast before the high-consistency material was inserted in the tray and the practitioner took the impression; (iv) the mouthguard remained in the tray and the low-viscosity material was inserted over the mouthguard; (v) the impression was positioned on the model, and after the material hardened, the mouthguard-reinforced impression was analyzed. In the evaluation of the dimensional stability, rigorous statistical analysis was essential to discern the performance differences between conventional and mouthguard-reinforced dental impressions. Statistical analyses employed non-parametric Mann–Whitney U tests because of the non-normal distribution of the data. They indicated a statistically significant improvement in the dimensional stability of addition silicone impressions when reinforced with mouthguards (p < 0.05), showcasing superior performance over conventional methods. Conversely, alginate and condensation silicone reinforced impressions did not exhibit the same level of stability improvement, suggesting the need for further optimization of these materials. In conclusion, from the three considered elastomers, addition silicone was found to be the prime candidate for high-precision dental impressions, with the potential to improve their quality from conventional impressions by utilizing the proposed reinforcing technique.

After a brief introduction highlighting the challenges of fluorine chemistry and the latest developments in the field, this Perspective will discuss how a combination of fluorine and fluorous chemistry together with fluorinated reagents helped to bridge between organic, molecular, macromolecular, supramolecular and biological sciences to create functions in the laboratory of the corresponding author. The reactivity of fluoride as a leaving group is best illustrated by SNAr reactions when it helped to demonstrate single electron transfer-mediated side reactions and through molecular design replaced activated aryl fluorides with aryl chlorides in the synthesis of poly(etherketone)s. Subsequently it was demonstrated how Ni(II) sigma complexes provided an orthogonal approach to the Suzuki-type cross-coupling of arylfluorides, other halides and all aryl C–O based electrophiles. Fluorinated reagents facilitated cylotrimetrization vs cyclotetramerization of bis(methoxy)benzyl chloride and alcohol and the synthesis of the simplest molecular liquid crystals. Triflic acid, methyl triflate facilitated the most tolerant living polymerizations including of cyclic siloxanes, functional vinyl ethers and oxazolines to generate self-organizable dendronized polymers while fluorine, trifluoromethyl and trifluoromethoxy groups facilitated disassembly and reassembly of liquid crystal polyethers and poly(p-phenylenes). Fluorinated stereocenters accessed the first heterochiral recognition in side-chain liquid crystal poly(vinyl ether)s and their model compounds. Alkali metal triflates mediated self-organization of supramolecular nonfluorinated and fluorinated self-assembling minidendrons, dendrons, dendrimers and self-organizable dendronized polymers. The role of fluorinated alkyl groups and of alkali metal triflates in the self-assembly, disassembly and isomorphic replacement analysis, of supramolecular helical columns, of the assembly of helical cogwheel coat and of spherical supramolecular dendrimers forming Frank-Kasper periodic and quasiperiodic arrays was highlighted. A brief discussion of fluorinated amino acids, peptides and peptoids and their potential role in the self-assembly and functions resulted from dendritic dipeptides followed by a discussion of semifluorinated amphiphilic Janus dendrimers as models of biological membranes, including for cell fusion and fission, concludes this Perspective.

BackgroundWheelchair users with a spinal cord injury (SCI) are at a high risk for developing pressure injuries (PIs). Performing weight shifts is a primary method of pressure management for PI prevention; however, individuals with SCI may lack confidence in their abilities to perform adequate pressure relief due to their lack of sensation. Real-time seat interface pressure mapping feedback may provide partial substitution for sensory feedback such that an individual’s confidence is improved. ObjectiveWe aim to examine how confidence for pressure management by wheelchair users with SCI was impacted by providing access to real-time, on-demand seat interface pressure mapping feedback. MethodsAdults with SCI (N=23) completed self-efficacy questions addressing confidence around 4 factors related to performing weight shifts in this longitudinal, repeated-measures study. We evaluated the impact of providing standard PI prevention education and access to live pressure map feedback on confidence levels for performing weight shifts. ResultsAccess to live pressure map feedback while learning how to perform weight shifts resulted in significantly higher confidence about moving far enough to relieve pressure at high-risk areas. Confidence for adhering to the recommended weight shift frequency and duration was not significantly impacted by in-clinic education or use of pressure map feedback. Confidence that performing weight shifts reduces PI risk increased most following education, with slight additional increase when pressure map feedback was added. ConclusionsAccess to live pressure mapping feedback improves confidence about performing weight shifts that relieve pressure when provided in the clinical setting and demonstrates potential for the same in the home. This preliminary exploration of a smartphone-based pressure mapping intervention highlights the value of access to continuous pressure mapping feedback to improve awareness and confidence for managing pressure. Background: The health development of a baby during pregnancy is greatly influenced by prenatal bonding. The purpose of this study is to assess maternal-fetal attachment in the second- and third-trimesters and explore how the mother’s mental health and socioeconomic circumstances may affect it. Additionally, this study takes into account other relevant aspects, and addresses the existing lack of relevant information in Romania. Methods: We conducted a cross-sectional study. Data were collected from 200 pregnant women in the second- and third-trimesters of pregnancy. Personal Information Form (PIF), Depression, Anxiety, and Stress Scale-21 (DASS-21), and Maternal-Foetal Attachment Scale (MFAS) questionnaires were collected. Results: There was no statistically significant correlation between the MFAS and the mental health of the mother. Moreover, subsequent comparative analyses revealed a significant difference only in the case of socio-economic conditions, with women with a moderate socio-economic status showed higher levels of maternal-foetal attachment. Conclusions: The degree of maternal-foetal attachment is not significantly influenced by social characteristics such as age, education level, socio-economic context, or place of residence. The MFAS measured the attachment relationship. However, the study found no statistically significant correlation between the mother’s mental health and this attachment.

The development of an automatic seedling planting system for micro-farms requires testing under laboratory conditions to verify the theoretical relationships between essential functional parameters, working speed and planting time. The constructive dimensional values of the prototype, the results measured in stationary mode directly on the transplanter and the auxiliary equipment and the direct determinations of the working parameters on the soil bin are used. Depending on the characteristics of the soil bin trolley, a range of speeds is chosen at which the machine is tested. The data obtained validate the correct operation of the prototype at speeds close to those determined theoretically for the following indicators: distance between plants per row, planter wheel slippage, misplanted seedlings rate and seedling frequency, with results comparable to existing agronomic standards. Once the appropriate operating speeds of the machine have been obtained, between 0.304 and 0.412 m/s, with planting frequencies between 0.899 and 1.157 s−1 (respectively, 53.94 and 69.42 seedlings per minute), optimizations and adjustments of some machine components can be made, for subsequent testing in real field conditions.
