We investigate imbibition of a monodisperse emulsion into a low-aspect ratio microfluidic channel with the height h comparable to the droplet diameter d. For confinement ratio d/h = 1.2, the tightly confined disk-like droplets in the channel move more slowly compared to the average suspension velocity. Behind the meniscus that drives the imbibition, there is a droplet-free region, separated from the suspension region by a sharp concentration front. The suspension exhibits strong droplet density and velocity fluctuations, but on average, the suspension domain remains uniform. For weaker confinement, d/h = 0.65, the spherical droplets move faster than the average suspension flow, resulting in the formation of a dynamically unstable high-concentration region near the meniscus. We describe the macroscopic suspension dynamics using linear transport equations for the particle-phase flux and suspension flux that are driven by the local pressure gradient. A dipolar particle interaction model explains the observed large density and velocity fluctuations in terms of the dynamics of elongated particle clusters with different orientations.

The mechanisms for phase separation in highly concentrated emulsions when subjected to a thermal phase transition remain to be elucidated. Here, we create a hexagonally close-packed monodisperse emulsion in 2D and show that during a cool-heat cycle, the emulsion fully destabilizes akin to phase separation. The mechanism for this catastrophic destabilization is found to be spontaneous coalescence initiation that synchronously occurs between every solidified droplet and its neighbors. This synchronous coalescence initiation establishes system-wide network connectivity in the emulsion causing large-scale destabilization. This system-wide coalescence initiation is found to be insensitive to droplet size and surfactant type, but dependent on network connectivity and crystal content of individual droplets.

We report a new mechanism for nucleation in a monolayer of hexagonally packed monodisperse droplet arrays. Upon cooling, we observe solidified droplets to nucleate their supercooled neighbors giving rise to an autocatalytic-like mechanism for accelerated crystallization. This collective mode of nucleation depends on the strength and nature of droplet contacts. Intriguingly, the statistical distribution of the solidified droplet clusters is found to be independent of emulsion characteristics except surfactant. In contrast to classical nucleation theory, our work highlights the need to consider collective effects of nucleation in supercooled concentrated emulsions where droplet crowding is inevitable.

In this study, we report the use of digital holography microscopy (DHM) for 3D-resolved flow kinematics and shear rheometry of viscoelastic polymeric fluids. We computationally reconstruct the recorded holograms to visualize the tracer imbued flow volume in microchannels, followed by implementation of particle tracking velocimetry (PTV) to quantitate spatially-resolved velocity fields in 3D. In order to select optimal parameters for DHM-PTV characterization of complex fluids, we studied the effect of hologram recording distance, seeding density and particle size. Using the optimal parameters, we show quantitative characterization of the shear rheology from the velocity fields without any a-priori assumptions of wall boundary condition or constitutive equation. The viscosity versus shear rate data for Newtonian and polyethylene oxide solutions could be measured in the range of ~ 0.05 - 20,000 s-1 with just four input flow rates. This data from holographic shear rheometry was found to be in good agreement with computational fluid dynamics simulations and macrorheometry. The holographic shear rheology technique remained unaffected by wall-slip events and instead provided an avenue to quantitate slip severity. Finally, we discuss holographic visualization of particle migration in microfluidic flows which can limit flow field access while at the same time provide a fingerprint of the suspending fluid rheology., Comment: Corresponding author is Siva A. The transport of deformable objects including polymer particles, vesicles and cells, has been a subject of interest for several decades where the majority of experimental and theoretical studies have been focused on circular tubes. Due to advances in microfluidics, there is a need to study the transport of individual deformable particles in rectangular microchannels where corner flows can be important. In this study, we report measurements of hydrodynamic mobility of confined polymeric particles, vesicles and cancer cells in a linear microchannel with square cross-section. Our operating conditions are such that the mobility is measured as a function of geometric confinement over the range 0.3 < l < 1.5 and at specified particle Reynolds numbers that are within 0.1 < Rep < 2.5. The experimental mobility data of each of these systems is compared with the circular-tube theory of Hestroni, Haber and Wacholder (J. of Fluid Mech., 1970) with modifications made for a square cross-section. For polymeric particles, we find that the mobility data agrees well over a large confinement range with the theory but under predicts for vesicles. The mobility of vesicles is higher in a square channel than in a circular tube, and does not depend significantly on membrane mechanical properties. The mobility of cancer cells is in good agreement with the theory up to l approaches 0.8, after which it deviates. Comparison of the mobility data of the three systems reveals that cancer cells have higher mobility than rigid particles but lower than vesicles, suggesting that the cell membrane frictional properties are in between a solid-like interface and a fluid bilayer. We explain further the differences in the mobility of the three systems by considering their shape deformation and surface flow on the interface. The results of this study may find potential applications in drug delivery and biomedical diagnostics.

We used volume-of-fluid (VOF) method to perform three-dimensional numerical simulations of droplet formation of Newtonian fluids in microfluidic T-junction devices. To evaluate the performance of the VOF method we examined the regimes of drop formation and determined droplet size as a function of system parameters. Comparison of the simulation results with four sets of experimental data from the literature showed good agreement, validating the VOF method. Motivated by the lack of adequate studies investigating the influence of viscosity ratio ({\lambda}) on the generated droplet size, we mapped the dependence of drop volume on capillary number (0.001 < Ca < 0.5) and viscosity ratio (0.01 < {\lambda} < 15). We find that for all viscosity ratios investigated, droplet size decreases with increase in capillary number. However, the reduction in droplet size with capillary number is stronger for {\lambda} < 1 than for {\lambda} > 1. In addition, we find that at a given capillary number, the size of droplets does not vary appreciably when {\lambda} < 1, while it increases when {\lambda} > 1. We develop an analytical model for predicting droplet size that includes a viscosity-dependent breakup time for the dispersed phase. This improved model successfully predicts the effects of viscosity ratio observed in simulations. Results from this study are useful for the design of lab-on-chip technologies and manufacture of microfluidic emulsions, where there is a need to know how system parameters influence droplet size.

Free-living nematode Caenorhabditis elegans is a powerful genetic model, essential for investigations ranging from behavior to neuroscience to aging, and locomotion is a key observable used in these studies. However, despite the fact that in its natural environment C. elegans moves in three-dimensional (3D) complex media (decomposing organic matter and water), quantitative in vestigations of its locomotion have been limited to two-dimensional (2D) motion. Based on our recent quantitative analysis of 2D turning maneuvers [Phys. Fluids 25, 081902 (2013)] we follow with the first quantitative description of how C. elegans moves in 3D environments. We show that by superposing body torsion and 2D undulations, a burrowing or swimming nematode can rotate the undulation plane. A combination of these roll maneuvers and 2D turns associated with variation of undulation-wave parameters allows the nematode to explore 3D space. We apply our model to analyze 3D chemotaxis of nematodes burrowing in a gel and swimming in water; we conclude that the nematode can achieve efficient chemotaxis in different environments without adjusting its sensory-motor response to chemical signals. The millimeter-long soil-dwelling nematode {\it C. elegans} propels itself by producing undulations that propagate along its body and turns by assuming highly curved shapes. According to our recent study [PLoS ONE \textbf{7}, e40121 (2012)] all these postures can be accurately described by a piecewise-harmonic-curvature (PHC) model. We combine this curvature-based description with highly accurate hydrodynamic bead models to evaluate the normalized velocity and turning angles for a worm swimming in an unconfined fluid and in a parallel-wall cell. We find that the worm moves twice as fast and navigates more effectively under a strong confinement, due to the large transverse-to-longitudinal resistance-coefficient ratio resulting from the wall-mediated far-field hydrodynamic coupling between body segments. We also note that the optimal swimming gait is similar to the gait observed for nematodes swimming in high-viscosity fluids. Our bead models allow us to determine the effects of confinement and finite thickness of the body of the nematode on its locomotion. Droplets moving in a microfluidic loop device exhibit both periodic and chaotic behaviors based on the inlet droplet spacing. We propose that the periodic behavior is an outcome of a dispersed phase conservation principle. This conservation principle translates into a droplet spacing conservation equation. Additionally, we define a simple technique to identify periodicity in experimental systems with input scatter. Aperiodic behavior is observed in the transition regions between different periodic behaviors. We propose that the cause for aperiodicity is the synchronization of timing between the droplets entering and leaving the system. We derive an analytical expression to estimate the occurrence of these transition regions as a function of system parameters. Caenorhabditis elegans, a free-living soil nematode, propels itself by producing undulatory body motion and displays a rich variety of body shapes and trajectories during its locomotion in complex environments. Here we show that the complex shapes and trajectories of C. elegans have a simple analytical description in curvature representation. Our model is based on the assumption that the curvature wave is generated in the head segment of the worm body and propagates backwards. We have found that a simple harmonic function for the curvature can capture multiple worm shapes during the undulatory movement. The worm body trajectories can be well represented in terms of piecewise sinusoidal curvature with abrupt changes in amplitude, wavevector, and phase.

We investigate the dynamics of pairs of drops in microfluidic ladder networks with slanted bypasses, which break the fore-aft structural symmetry. Our analytical results indicate that unlike symmetric ladder networks, structural asymmetry introduced by a single slanted bypass can be used to modulate the relative drop spacing, enabling them to contract, synchronize, expand, or even flip at the ladder exit. Our experiments confirm all these behaviors predicted by theory. Numerical analysis further shows that while ladder networks containing several identical bypasses are limited to nearly linear transformation of input delay between drops, mixed combination of bypasses can cause significant non-linear transformation enabling coding and decoding of input delays., Comment: 4 pages, 5 figures
