Resumen del póster presentado a la XXIX International Yeast Conference Genetics and Molecular Biology (ICYGMB), celebrada en Göteborg (Gothenburg-Sweden) del 18 al 22 de agosto de 2019., The yeast Saccharomyces cerevisiae is the main microorganism carrying out the alcoholic fermentation to convert grape juice to wine. The grape juice is characterized by plenty sugar concentration, in the form of glucose and fructose, that causes hyperosmotic stress, but scarce nitrogen sources, so nitrogen limitation usually produces starvation during most fermentations. These peculiar conditions make the crosstalk between nutrients signalling pathways a critical event not only for cell survival, but also for proper fermentation performance in metabolically active cells. As nitrogen is scarce, the TORC1 pathway must be relevant in the process. TORC1 activity was measured by the phosphorylation state of two of its readouts, the proteins Rps6p and Par32p. The analysis of both TORC1 targets indicates that their branches pathway are activated only during the first hours of winemaking pointing out that signalling of nitrogen shortage may be taking place earlier than expected, when nitrogen is still available. Possibly, other nutrient signalling pathways could be active to promote growth. One of these pathways is the glucose repression pathway, controlled by protein kinase Snf1p, mainly regulating the use of carbon sources alternative to glucose. Snf1p-dependent phosphorylation patterns demonstrate that surprisingly this pathway is also active only during the first hours of winemaking, when glucose and fructose are present at high concentration much longer. The activity of the Ras/cAMp/PKA pathway has also been investigated by measuring cAMP levels, which show a 2.5-fold reduction between the first hours of fermentation and stationary phase cultures, when the pathway is not activated. Stress conditions at the beginning of fermentation may give rise to a certain level of kinase activation, despite the high abundance of sugars in the must. Nitrogen availability also influences the activity of yeast nitrogen catabolic repression (NCR), mediated by the transcription factor Gln3p, which allows cells to repress the use of poor nitrogen sources when optimal nitrogen sources are present. The analysis of Gln3p phosphorylation state along fermentation indicates that NCR seems to be inactive at the beginning of the fermentation, although yeast assimilable nitrogen (YAN) is not low. The coordination between nutrient signalling pathways allows to integrate different inputs, such as nitrogen shortage and high sugar concentration, to respond to the specifically environmental conditions during the alcoholic fermentation of the grape must, which are very harsh.

Esta tesis aborda el análisis de las rutas de señalización de nutrientes de cepas vínicas de S. cerevisiae en condiciones de vinificación para comprender su respuesta frente a las condiciones adversas de disponibilidad de fuentes de nitrógeno y carbono en el mosto. El mosto de uva se caracteriza por un elevado contenido en azúcares y una limitación de fuente de nitrógeno, lo que podría explicar la mayor tolerancia de las cepas vínicas, respecto a las de laboratorio, frente a rapamicina (inhibidor del complejo TORC1) y a análogos estructurales no metabolizables de la glucosa, sugiriendo una organización diferencial de flujos metabólicos. Se han realizado dos experimentos de evolución dirigida, frente a rapamicina y frente a un análogo estructural de la glucosa (2-desoxiglucosa), que han permitido obtener cepas más tolerantes a dichos compuestos, pero sin fenotipos con aplicabilidad industrial. También se ha analizado el estado de activación de las rutas de señalización de nutrientes a través del estudio del estado de fosforilación de proteínas diana de las mismas, y se ha encontrado que las rutas TORC1 y Snf1 solo están activas en las primeras horas de vinificación, mientras que la ruta RAS/cAMP/PKA se mantiene activa a lo largo de todo el proceso, sugiriendo que ésta es la ruta reguladora principal en esas condiciones. Dado que el mosto de uva presenta una concentración deficitaria de nitrógeno, se estudiaron, además de TORC1, otras rutas que también responden a disponibilidad de nitrógeno, como el control general de aminoácidos (GAAC) y la represión por catabolito de nitrógeno (NCR), encontrándose que la proteinquinasa Sch9p y el factor transcripcional Gcn4p influyen profundamente en la fisiología celular, en el metabolismo y en la longevidad cronológica. Mutaciones en los genes codificantes de estas proteínas alteran, de manera similar, el metabolismo central del carbono y la producción de glicerol en vinificación y, sin embargo, tienen diferente impacto sobre el resto del metabolismo. Se realizó también un análisis fenómico global de las cepas y mutantes de deleción en genes de proteínas diana de las rutas de señalización de nutrientes. Los resultados mostraron la importancia de las proteínas Ras2p y Bcy1p, recalcando la importancia de la ruta Ras/cAMP/PKA en vinificación, así como la relación entre Snf1p y Gln3p. También mostraron que el gen MKS1, regulador negativo de la ruta retrógrada, es una buena diana para la obtención de cepas mejoradas de aplicabilidad industrial en vinificación. El conjunto de resultados obtenidos demuestra que no existe división entre la señalización de fuentes de carbono y de nitrógeno en vinificación, sino una comunicación cruzada adecuadamente regulada entre todas las rutas de señalización de nutrientes que permite que el metabolismo celular se adecúe a las condiciones ambientales., [EN]: Alcoholic fermentation of grape must is mainly carried out by the yeast Saccharomyces cerevisiae. This thesis deals with the analysis of nutrient signalling pathways of Saccharomyces cerevisiae wine strains in winemaking conditions to understand their response to low conditions of nitrogen and carbon availability of sources in the must. Grape must is characterized by high sugar content and limited nitrogen sources, which could explain the greater tolerance of wine strains, compared to laboratory strains, against rapamycin (inhibitor of the TORC1 complex) and non-metabolizable structural analogues of glucose, suggesting a differential organization of metabolic fluxes. Two experiments of directed evolution have been carried out, one against rapamycin ( an inhibitor of TOR activity) and the other against 2-deoxyglucose (a structural analogue of glucose), which have allowed us to obtain strains more tolerant to those compounds however none of them displayed improved phenotypes for winemaking. The activation state of nutrient signalling pathways has also been analysed through the study of the phosphorylation state of their target proteins, and it has been found that both the TORC1 and SNF1 pathways are only active during the first hours of vinification, while the RAS/cAMP/PKA pathway remains active throughout the entire process, suggesting that this is the main regulatory pathway in these conditions. Since grape must has a nitrogen deficit concentration, other pathways that also respond to nitrogen availability, such as the general amino acid control (GAAC) and nitrogen catabolite repression (NCR), were studied in addition to TORC1. Sch9p protein kinase and the transcriptional factor Gcn4p deeply impact cell physiology, metabolism and chronological longevity. Deletion of SCH9 and overexpression of GCN4 similarly alter central carbon metabolism and glycerol production in winemaking but they have different impact on the rest of metabolic pathways. A global phenomic analysis of deletion mutants in genes coding for target proteins from the nutrient signalling pathways was also performed. The results showed the significance of Ras2p and Bcy1p proteins, emphasizing the importance of the Ras/cAMP/PKA pathway in winemaking, as well as the relationship between Snf1p and Gln3p. They also showed that the MKS1 gene, a negative regulator of the retrograde pathway, is a good target for the improvement of strains for industrial winemaking. The main general conclusion from the obtained results is that no division exists between signalling of carbon and nitrogen sources in vinification conditions, but a properly regulated crosstalk between all nutrient signalling pathways that allows cellular metabolism to adapt to the singular environmental conditions during winemaking.

La procura nace como resultado de la necesidad de representar a los litigantes en el proceso. Debido a la velocidad a la que se desarrolla la sociedad, esta labor se ve afectada en cuanto a competencias se refiere; aparecen nuevas vías de representar a las partes en su modalidad telemática y ello genera un nuevo debate sobre la necesidad de intervención de este sujeto y el futuro de la profesión. En el presente trabajo se realiza un estudio que versa sobre la figura del procurador a lo largo de los años, desde sus inicios en el derecho romano hasta el planteamiento de su existencia en el futuro, y los cambios a realizar para garantizar su perduración, bajo la opinión de varias instituciones y autores.

“La duración prolongada del tratamiento ortodóncico pone en riesgo la integridad bucal del paciente, debido a que la aparatología fija permite la retención de placa dentobacteriana e impide realizar una higiene bucal adecuada, favoreciendo la desmineralización, caries dental e hiperplasia gingival. La aplicación de fuerzas prolongadas favorece la resorción radicular, lo cual es un factor importante e irreversible que se debe considerar durante el tratamiento ortodóncico. La duración del tratamiento ortodóncico se prolonga aún más si se realizan extracciones. El resultado obtenido en este trabajo permite arribar a las siguientes conclusiones; el uso de vibraciones mecánicas podría tener un efecto favorable durante el tratamiento ortodóncico si se emplean durante mayor tiempo, al ser vibraciones de alta frecuencia podrían disminuir en gran medida la duración del tratamiento ortodóncico, sin tener efectos colaterales como la reabsorción radicular; además, el cierre de espacios es una fase importante a considerar dentro de la ortodoncia acelerada para la disminución del mismo, por lo tanto, en base a los resultados obtenidos en este estudio, se sugiere realizar estudios con la aplicación de vibraciones mecánicas de alta frecuencia hasta culminar el cierre de espacios en masa para confirmar las expectativas del mismo, con una muestra mayor y homogénea.”

El presente estudio de maestría guarda relación con la implementación de una unidad didáctica dirigida a mejorar los niveles de argumentación en los estudiantes del grado 3°B de la institución educativa Centro Auxiliar de Servicios Docentes (CASD), sede Santa Eufrasia. La metodología que enmarca el trabajo es cuantitativa, teniendo en cuenta que el procedimiento inició con una evaluación diagnóstica, la cual fue el punto de partida para el diseño de la intervención a través de la unidad didáctica, y finalmente, se efectuó una evaluación de salida con el fin de analizar los resultados del proceso. Durante la aplicación de la unidad didáctica surgieron interrogantes por parte de los estudiantes, lo que obligó a modificar o cambiar algunas secuencias de dicha unidad, obteniendo resultados satisfactorios en lo que respecta a la argumentación y al manejo de la alimentación saludable, y fue observable en los estudiantes una mayor sensibilidad y conciencia frente al área en cuestión.

Trabajo presentado a la 28th International Conference on Yeast Genetics and Molecular Biology (ICYGMB), celebrada en Praga (Republica Checa) del 27 de agosto al 1 de septiembre de 2017., Wine alcoholic fermentation is mainly carried out by enological strains of Saccharomyces cerevisiae, hence the importance of the environmental and genetic factors that determine their growth and aging, not only for their relevance on the fermentative process but also by their impact on wine organoleptic properties. When a nutrient is scarce, dietary restriction happens, which consists in the reduction of nutrients supply without inducing malnutrition. Dietary restriction controls longevity through nutrient signaling pathways, as the TORC1 pathway, which promotes growth and protein synthesis when nitrogen sources are abundant. As sugars are plenty in grape juice, generally is nitrogen the limiting nutrient. Thus, the study of the physiological processes by which a selective nitrogen source is used is of great importance. Hence, our interest in TORC1 targets, such as ribosomal Rps6 protein and the nitrogen catabolic repression (NCR) system, that represses the use of poor nitrogen sources when optimal sources are present. Its main effectors are GATA transcriptional factors (Gln3 and Gat1), controlled mainly by TORC1. The behaviour of NCR transcription factors Gat1 and Gln3 is being studied in wine strains. Our results indicate genetic differences between wine and laboratory yeast strains, mainly in their response to nitrogen shortage. Indeed, using phosphorylation of Rps6 as a tool to study TORC1 activity, our data indicate that wine and laboratory yeast strains behave differently against several TOR inhibitors, such as rapamycin or methionine sulfoximine (MSX). Moreover, TORC1 pathway seems to be only active in the first hours of winemaking, indicating that signalling of nitrogen shortage occurs earlier than expected. Under these conditions, other nutrient signalling pathways (Ras/AMPc/PKA or Snf1) could be active to promote growth, and that aspect is being also addressed. In general, TORC1 pathway can integrate the nitrogen signal and coordinate other pathways to respond to the specific environmental winemaking conditions.

Objetivo: comprobar la fiabilidad de las escalas de Braden y EMINA (escalas de valoración del riesgo de úlceras por presión) en atención domiciliaria. Método: estudio observacional de tipo transversal. Ámbito de estudio: Área de Gestión Sanitaria Este de Málaga-Axarquía y Área de Gestión Sanitaria Serranía, ambas de Málaga. Sujetos de estudio: pacientes incluidos dentro del Programa de Inmovilizados sin úlceras por presión. Instrumentos: escalas de Braden y EMINA. Muestra: 30 pacientes, usando una técnica de muestreo aleatorio simple. Resultados: Escala de Braden: Alfa de Cronbach de 0,78 para ambos observadores. Coeficiente de correlación intraclase (CCI) para la puntuación total de la escala, 0,968. El índice kappa ponderado para cada uno de los 6 ítems de la escala oscila entre 0,685 que es el más bajo y corresponde a fricción y deslizamiento y 1,00 para el más alto, que corresponde a percepción sensorial. Escala EMINA: el Alfa de Cronbach oscila entre 0,69 y 0,73 entre observadores. Coeficiente de correlación intraclase para la puntuación total de la escala, 0,974. El índice kappa ponderado para cada uno de los 5 ítems de la escala oscila entre 0,750, el más bajo que corresponde a la actividad, y 1,00 para el más alto, que corresponde a nutrición. Conclusiones: la escala de Braden tiene mayor consistencia interna, aunque el grado de acuerdo cuando se utilizan las escalas de manera categórica es superior en la EMINA. Ambas escalas son instrumentos fiables para el uso en atención domiciliaria, aunque sería conveniente establecer nuevos puntos de corte, para definir o categorizar el riesgo en esta población. Objective: Assessing reliability of the Braden and EMINA scales (Rating Scales for the Risk of pressure sores) in-home care. Method: Cross-sectional study. Population of study from Málaga: area of Heath Management East Málaga-Axarquia and mountainous region. Subjects of study: Patients entered in the programme of immobilize persons without pressure sore. Questionnaires: Braden and EMINA. Sample: 30 patients selected by simple randomisation. Results: Braden Scale: Cronbach's coefficient alpha 0.78 for both ratters. The Intraclass correlation coefficient (ICC) is 0.968. Kappa values for each of the 6 items vary from 0.685 for the friction and slipping item and 1 for the sensorial perception item. EMINA Scale: Cronbach's coefficent alpha 0.69 and 0.73 for each of the two raters, respectively. The Intraclass correlation coefficient (ICC) is 0.974. Kappa values for each of the 6 items vary from 0.750 for the activity item and 1 for the nutrition item Conclusions: The internal consistency of Braden scales is higher than the consistency of the EMINA scale. However, when the individual item agreement between raters was higher for the EMINA than for the Braden. Both scales, as continuous, are reliable instruments to be used for in-home care, although it would be convenient to establish new cut-off points for the scales in order to categorise the risk of pressure sores for this type of population.
