The cytotoxic profile and antiproliferative and mitochondrial effects of triterpene acid conjugates with mitochondriotropic lipophilic triphenylphosphonium (TPP + ) and F16 cations were evaluated. Maslinic and corosolic acids chosen as the investigation objects were synthesized from commercially available oleanolic and ursolic acids. Study of the cytotoxic activity of TPP + and F16 triterpenoid derivatives against six tumor cell lines demonstrated a comparable synergistic effect in the anticancer activity, which was most pronounced in the case of MCF-7 mammary adenocarcinoma cells and Jurkat and THP-1 leukemia cells. The corosolic and maslinic acid hybrid derivatives caused changes in the progression of tumor cell cycle phases when present in much lower doses than their natural triterpene acid precursors. The treatment of tumor cell lines with the conjugates resulted in the cell cycle arrest in the G1 phase and increase in the cell population in the subG1 phase. The cationic derivatives of the acids were markedly superior to their precursors as inducers of hyperproduction of reactive oxygen species and more effectively decreased the mitochondrial potential in isolated rat liver mitochondria. We concluded that the observed cytotoxic effect of TPP + and F16 triterpenoid conjugates is attributable to the ability of these compounds to initiate mitochondrial dysfunctions. A series of new 1,3-thiazole derivatives of maleopimaric acid 6a-f , 7a-f were synthesized and evaluated for anticancer, antibacterial and antifungal activities. Evaluation of cytotoxic activity against human embryonic kidney 293 cells (HEK293), human neuroblastoma cell line (SH-SY5Y), hepatocellular carcinoma cell line (HepG2) and human T-cell lymphoblast-like line (Jurkat), showed that introduction of the aminothiazole fragment at position 6 of the diterpenoid molecule leads to decrease of cell viability. Substance 3 was found to be the most active against all tested cell lines, inhibiting cell viability with IC 50 values in the range of 2-24 μM. The structure-activity relationship of these compounds was studied and the results show that the compounds 6c and 7e exhibited in vitro antifungal activity against Candida albicans and also possessed antibacterial profile against Enterobacter aerogenes , Klebsiella pneumoniae , Staphylococcus aureus , Streptococcus pyogenes , Escherichia coli and Proteus vulgaris .

The results of recent studies indicate that the hyperactivation of the glutamatergic system plays an important role in the pathophysiology of multiple sclerosis (MS). In addition to the well-known direct toxic effect of the excessive extracellular level of the glutamate neurotransmitter on neurons, additional mechanisms of glutamate-induced cell damage have been described in the literature, including effects on oligodendrocytes, astrocytes, endothelial cells and immune cells. The study of these toxic effects will reveal the possible link between various pathological hallmarks of MS, such as axonal damage, oligodendrocyte death, demyelination, neurodegeneration, autoimmune reactions and dysfunction of blood-brain barrier. Understanding the mechanisms underlying the glutamate toxicity will contribute to the development of new therapeutic approaches for the diagnosis and treatment of patients with MS. This review focuses on the mechanisms that lead to an increase in the concentration of neurotransmitter glutamate and excitotoxicity in the context of the pathogenesis of this disease. Also the authors present the data on existing and currently developed medicines and therapeutic approaches to regulate the activity of the glutamatergic system.

To determine the role of NMDA receptors in the functional regulation of immunocompetent cells, comparative assay was carried out for genes expressed in the mononuclears in peripheral blood of healthy persons under normal conditions and after blockade of these receptors. The genes, whose expression changed in response to blockade of NMDA receptors in mononuclears, encode the products involved in regulation of the major functions of immune cells, such as proliferation (IL4, VCAM1, and CDKN2A), apoptosis (BAX, MYC, CDKN2A, HSPB1, and CADD45A), activation (IL4R, IL4, VCAM1, and CDKN2A), and differentiation (IL4, VCAM1, and BAX).

The study demonstrated that chitooligosaccharides with a molecular weight of 5–10 kDa and a degree of acetylation of 65% exhibited an auxin-like effect in wheat plants and also played an important role in regulating the activity of polysaccharide (chitin)–specific anion isoenzymes of peroxidase oxidizing indole acetic acid. Changes in the kinetic parameters of the interaction of the wheat anionic chitin-specific peroxidase with pI ∼3.5 with chitin oligomers in the presence of indoleacetic acid were pH-depended and indicated that chitooligosaccharides significantly impair the ability of the enzyme for oxidation at pH levels of 4.2 and 6.0. It can be assumed that chitooligosaccharides not only induce protective plant systems but also increase the accumulation of auxin in plant tissues, thus adversely affecting a number of components of the plant protective system against pathogens.

DNA analysis of ñîmplex biological objects (wastewater, soil, archaeological and forensic samples, etc.) is currently of great interest. DNA of these objects is characterized by low suitability for research due to the violation of its integrity and chemical structure; thus, the detection of specific nucleic acid fragments can be achieved by PCR with contiguous primers. In this paper, we present the results that clarify the specific characteristics of PCR with abutting primers. The 3'-ends of these primers are annealed at adjacent nucleotides of complementary chains of DNA target. It has been shown that the proximity of primers enables the formation of specific reaction products with a higher sensitivity and less reaction time. Using artificially damaged DNA and DNA from the soil we demonstrated that the abutting primers provide assured detection of specific DNA fragments. The results of this work may be taken into account in PCR with degraded (fragmented) DNA.

Design and synthesis ofnew derivatives of (-)-cytisine with a wide spectrum of pharmacological activity, represents the potential therapeutic interest for development of drug candidates for neurodegenerative disorders, inflammatory diseases, and treatment of nicotine addiction. We used HEK293 cell line transiently transfected with N F-κB and STATI luciferase reporter constructs to screen the (-)-cytisine derivatives for their potency to modulate basal and induced NF-κB and STAT1 activity. Currently, NF-κB, STAT1 and components of their signaling pathways are considered as attractive targets for pharmacological intervention, primarily in chronic inflammation, cancer, autoimmune, neurodegenerative and infectious diseases. The library of compounds included the derivatives of (-)-cytisine with amino-, amide-, thio- and carboxamide groups at 3, 5 and 12 position of the starting molecule, as well as some bimolecular derivatives. Our experimental data revealed compounds with moderate activating as well as inhibitory effects for basal NF-κB and STATI activity (IC50 or EC50 values are mainly in the micromolar range). The structure-activity relationship analysis demonstrated that the character of activity (activation or inhibition of NFκ-B and STAT1) is determined by the topology of the substituents at the (-)-cytisine molecule, whereas the nature of the substituents mainly contributes to severity of the effect (introduction of aromatic and adamantyl substituents, as well as thionyl or keto groups are of the principal importance). When evaluating the effect of (-)-cytisine derivatives on activity of NF-κB and STATI, induced by specific agents (TNFα and IFNγ, respectively) we observed that some compounds inhibited basal and stimulated activity of NF-κB and STAT1, another compounds showed the dual effect (an increase of basal- and a decrease of stimulated NF-κB activity) and several compounds increase both basal and induced activity of NF-κB and STAT1. Thus, obtained results suggest that one of the possible mechanisms of biological action of (-)-cytisine derivatives is their ability to influence the components of NF-κB and STAT1-dependent signaling pathways.

Background: Noopept (N-phenyl-acetyl-L-prolylglycine ethyl ester) was constructed as a dipeptide analog of the standard cognition enhancer, piracetam. Our previous experiments have demonstrated the cognition restoring effect of noopept in several animal models of Alzheimer disease (AD). Noopept was also shown to prevent ionic disbalance, excitotoxicity, free radicals and pro-inflammatory cytokines accumulation, and neurotrophine deficit typical for different kinds of brain damages, including AD. In this study, we investigated the neuroprotective action of noopept on cellular model of AD, Aβ 25-35-induced toxicity in PC12 cells and revealed the underlying mechanisms., Results: The neuroprotective effect of noopept (added to the medium at 10 μM concentration, 72 hours before Аβ 25-35) was studied on Аβ 25-35-induced injury (5 μM for 24 h) in PC12 cells. The ability of drug to protect the impairments of cell viability, calcium homeostasis, ROS level, mitochondrial function, tau phosphorylation and neurite outgrowth caused by Аβ 25-35 were evaluated. Following the exposure of PC12 cells to Аβ 25-35 an increase of the level of ROS, intracellular calcium, and tau phosphorylation at Ser396 were observed; these changes were accompanied by a decrease in cell viability and an increase of apoptosis. Noopept treatment before the amyloid-beta exposure improved PC12 cells viability, reduced the number of early and late apoptotic cells, the levels of intracellular reactive oxygen species and calcium and enhanced the mitochondrial membrane potential. In addition, pretreatment of PC12 cell with noopept significantly attenuated tau hyperphosphorylation at Ser396 and ameliorated the alterations of neurite outgrowth evoked by Аβ25-35., Conclusions: Taken together, these data provide evidence that novel cognitive enhancer noopept protects PC12 cell against deleterious actions of Aβ through inhibiting the oxidative damage and calcium overload as well as suppressing the mitochondrial apoptotic pathway. Moreover, neuroprotective properties of noopept likely include its ability to decrease tau phosphorylation and to restore the altered morphology of PC12 cells. Therefore, this nootropic dipeptide is able to positively affect not only common pathogenic pathways but also disease-specific mechanisms underlying Aβ-related pathology.

Test systems for monitoring activities and the search for substances activating or inhibiting transcription factors as biotargets have been designed on the basis of luciferase constructs containing binding sites for transcription factors CREB, NFAT, NF-kB, p53, STAT1, GAS, VDR, HSF1, and HIF1alpha. An assessment of the functional activity of reporter constructs has been carried out using their transient transfection into HEK293 cells followed by treatment with specific inducers. The functional activity of all reporter constructs was observed based on the increased luciferase expression. In order to evaluate the efficiency of the suggested test systems, aspirin was used. Incubation of cells transfected with the above-mentioned constructs treated with aspirin was accompanied by the suppression of NF-kB, HIF1alpha, GAS, VDR, and HSF binding activity. The findings revealed for NF-kB, NFAT, and STAT1 confirm the published data concerning the mechanisms of aspirin action. The detected effects of this drug on the HIF1alpha, GAS, VDR, and CREB activity have been demonstrated for the first time.

Transgenic tobacco plants expressing the AINTEGUMENTA gene of rape under control of the 35S promoter and the promoter of dahlia mosaic virus were obtained. The transgenic plants were characterized by increase in the length of the leaves, flower sizes, stem height, and weight of seeds; at the same time, the degree of increase was greater in the case of use of the dahlia mosaic virus promoter as a regulator of transcription. Ectopic expression of the AINTEGUMENTA gene promoted prolongation of leaf growth, while sizes of epidermal cells of the leaves remained unchanged.

Under action of growth-stimulating concentrations of bioregulator stifun on wheat plants, an increase of functional activity of nucleoli of meristematic cells; contents of lectin (wheat germ agglutinin); and activity of proteinases, tripsin inhibitors, and ATPase activity was established. The pool of free amino acids was increased under bioregulator use. Levels of methionine, phenylalanine, cysteine, lysine, and tyrosine were increased. It is likely that stifun could activate protein biosynthesis in wheat plants.

Elevation of intracellular Ca²⁺ in T-lymphocytes as a consequence of T cell antigen receptor activation triggers transcriptional programs resulting in effector cytokine secretion and immune response coordination. Increase of Ca²⁺ concentration in T-lymphocytes follows both the Ins(1,4,5)P(3)-dependent release from an intracellular store and subsequent influx from extracellular milieu. Flow cytometry and the fluorescent dye Fluo-4AM have been used to demonstrate that noncompetitive NMDA receptor antagonist (+)-MK801 inhibits Ca²⁺ influx in T cells induced by thapsigargin. Combination of thapsigargin and (+)-MK801 with following incubation does not affect Ca²⁺ mobilization from intracellular stores, while decreased Ca²⁺ entry was observed. Overall data indicate that the ion channel blocker (+)-MK801 is able to inhibit the Ca²⁺ influx and confirm our suggestion about involvement of NMDA receptor in the store-operated Ca²⁺ entry mechanisms in human T-lymphocytes. To identify the signal transduction pathways associated with NMDA receptors in mitogen-stimulated T-lymphocytes, the cells were incubated with (+)-MK801, then activity of key phosphorylated protein kinases of MAP-activated (pERK1/2, pSAPK/JNK, p-p38), Ca²⁺-dependent (pCaMKII), PI3/Akt-dependent (pGSK-3β), and PKC-activated (pPKCθ) pathways were detected. The data we obtained demonstrate that (+)-MK801 treatment leads to more prominent decrease in Ras-activated protein kinases pERK1/2 and Rac-activated proteins p-p38 and pSAPK/JNK, as compared to DAG-dependent pPKCθ and Ca²⁺-dependent pCaMKII. These results show that NMDA receptors are mainly involved in regulation of Ras/Rac-dependent signaling in T-lymphocytes.

New approaches to the detection of impaired nucleotides based on the allele specific ligation of a "C probe" followed by rolling circle amplification have been developed. The detection of amplification products was realized by using enzymatic and deoxyribozyme digestion of fluorescently-labeled DNA-RNA-DNA chimeric oligonucleotide structures in cycling probe technology (CPT) in real-time mode.

The outbreaks of hantavirus infections in some regions of the Russian Federation in some years involve considerable material and social losses. In this connection, the designing of the most effective types of vaccines is an urgent task. The authors have created plasmid constructions containing the gene of hemorrhagic fever with renal syndrome virus, with whose Intramuscular injection there is a specific immune response and plasmid DNA is detectable in the adjacent tissues within a month after injection.

Competitiveness and genetic variation of the Rhizobium galegae strains from the collection of the All-Russia Institute of Agricultural Microbiology, Russian Academy of Agricultural Sciences, causing nodulation of oriental goat's rue under conditions of Bashkortostan soils (lacking this rhizobial species) were studied. It was demonstrated that of all the tested strains, the strains CIAM 0702 and CIAM 0704, each carrying two megaplasmids of 1500 and 2000 MDa, were the most competitive. RAPD (random amplified polymorphic DNA) analysis showed that R. galegae strains were able to intensively exchange the genetic material in the host plant rhizosphere. We did not succeed in detecting the local root nodule bacteria that were either initially able to infect oriental goat's rue or had adapted to infecting this species due to various genetic rearrangements.

Correlation was investigated between blood-plasma levels of C3(H2)O (conformation pattern of C3 component of the complement) and tumor-associated marker CA-125 in patients with ovarian cancer before and after chemotherapy. Since a drop in CA-125 level after chemotherapy was associated with similar changes in C3(H2)O fraction, it seems reasonable to suggest that change in conformation of C3 is a response of the immune system to cancer.

We have used the Rat Atlas cDNA Array ("BD Bioscience") to assess changes in mRNA expression of 588 genes in rat brain after acute treatment of 2-aminoadamantane compound--Ladasten. Drug exhibits the psychostimulating and anxyolitic actions. The analysis of results of hybridization on macrochips and their corroboration by quantitative real-time RT-PCR has allowed to reveal 12 genes, expression of which changes in response to ladasten in rat brain cells. The GAT3 and CARBH genes should be considered as primary pharmacologically significant targets and the changes of their functional conditions allows to explain the distinct mechanisms of anxyolitic properties of the drug. It was shown that Ladasten induced genes are involved in the different signalling pathways (APC, Rb, PKCIP, PMCA), genes encoding the cytosceletal proteins (Tubal, actin), synaptic proteins (Syn IA&IB, PLP) and metabolism enzymes (Gapdh, NSE). It is possible to assume, that proteins, encoded by the given genes participate in the compensatory and/or neuroplastic adaptation to biochemical effects of Ladasten.

A significant heterogeneity between bacteria Rhizobium galegae bv. officinalis and R. galegae bv. orientalis forming the nitrogen-fixing symbiosis with Galega officinalis and G. orientalis, respectively, and not forming any single cross-inoculation group, was found by means of RAPD and RFEL methods. The high level of sequence similiraty between lectins of these plants indicates at their close relationship. However the sequences of lectin sugar binding peptides (SBP) of G. orientalis (TYCNPGWDPRDR) and G. officinalis (TFYNEEWDLVIKDEH) were highly diverged. Amino acids of SBP which are involved in linkage of Ca2+ and Mn2+ ions responsible for stabilization of spatial structure of carbohydrate-binding "pocket", keep their position in peptide. It suggests that lectins participate in Rhizobium-legume symbiosis and that carbohydrate-binding site plays a key role in this process.

Some stages of low-temperature signal transduction causing appropriate cold stress response in plants are considered. The effects of Ca2+ chelators, Ca2+ channel blockers, and protein kinase inhibitors on protoplasts and plants of cabbage suggest that the initial stages of cold signal transduction are the change in membrane fluidity followed by the activation of calcium channels and elevation of Ca2+ influx into the cytoplasm. Increased concentration of Ca2+ in cytoplasm activates calcium-dependent protein kinase most likely participating in induction of transcription factors necessary for the expression of cold-regulated genes, in particular csp5. The protein kinase inhibitors staurosporine and wortmannin insignificantly repress the expression of csp5.

This review deals with recent data on the structure and biochemical properties of dehydrins, proteins that are normally synthesized in maturating seeds during their desiccation, and also in vegetative tissues of plants treated with abscisic acid or exposed to environmental stress factors that result in cellular dehydration. The dehydrins are considered as stress proteins involved in formation of plant protective reactions against dehydration. The generally accepted classification of dehydrins is based on their structural features, such as the presence of conserved sequences, designated as Y-, S-, and K-segments. The K-segment representing a highly conserved 15 amino acid motif (EKKGIMDKIKEKLPG) forming amphiphilic alpha-helix has been found in all dehydrins. The pathways of regulation of dehydrin gene expression, putative functions of dehydrins, and molecular mechanisms of their actions are discussed.

The process of accumulation of knowledge on wheat and related wild species during the 20th century is briefly reviewed with special reference to the evidence of the recent years on evolution of polyploid wheats and the role of diploid species. The latter serve as potential donors of the genomes, detection of which is particularly important because of the continuing speciation in the tribe Triticeae and artificial development of synthetic forms. The arguments in favor of the donor role for various diploid wheat species and aegilopses from the section Sitopsis are compared. It is stated that in the formation of the both lines of polyploid wheats turgidum-aestivum and timopheevi, diploid Aegilops speltoides acted as a maternal form. In addition to plasmatic genomes, this aegilops species introduced into them also the B and G nuclear subgenomes. A comparison of nucleotide sequences in the variable part of the promoter of evolutionary conserved rRNA genes in polyploid wheats with their counterparts in diploid wheats and aegilopses confirmed the accepted wheat phylogenies.

The effects of the phytohormones 6-benzylaminopurine (BAP) and 24-epibrassinolide (EB) on the nucleolar sizes in the interphase nuclei of root meristem were studied using the silver-staining procedure in wheat species with different ploidy levels (a polyploid series). In addition, the effects of the phytohormones on the cell mitotic activity in the roots of 5-day-old seedlings were studied. The higher the wheat species ploidy level, the higher its sensitivities to BAP and EB were. In diploid wheat, the maximum increase in the nucleolar organizing region (NOR) activity was observed after treatment with considerably higher phytohormone concentrations compared to tetra- and hexaploid wheat species. The phytohormone treatment increased both the sizes and the number of nucleoli in meristematic cells of seedling roots in all wheat species studied. It was assumed that the differences between the responses of wheat species with three different ploidy levels to different concentrations of phytohormones were related to their effects on the methylation/demethylation of cytosine residues in the rDNA promoter region.

The review considers the mechanisms of gene transcription regulation by phytohormones, which play an important role in controlling major genetic programs in plants. The focus is cis-regulatory elements, which occur in the phytohormone-responsive gene promoters and, along with trans-acting factors and other proteins, ensure the effective reception of the hormonal signal.

The rRNA gene promoter regions of three diploid wheats Triticum boeoticum Boiss, Triticum urartu Tum. ex Gandil, and Triticum monococcum L. have been sequenced and analyzed. It has been found that the rRNA promoter initiation regions of diploid wheats contain the sequences, which differ from the evolutionary conserved TATAGTAGG (+1 is underlined) motif of monocots. The transient expression assay in wheat protoplasts confirmed that cloned sequences are active promoters. Deletion analysis showed that the promoter sequence localized between -113 and +15 (relative to +1) is enough to direct RNA polymerase I-dependent transcription.

Background: The relation of between Helicobacter pylori and the symptoms in children is still controversial. Determination of specific immunoglobulin (Ig) G antibodies to H. pylori may represent a useful test to screen the patients with acid peptic disease in childhood. The aim of this study was to investigate the spectrum of clinical symptoms, endoscopic and histologic lesions, and clinical value of serum IgG response to H. pylori in school-aged children residing in the Ural area of Russia for the identification of Helicobacter -related acid-peptic disease., Methods: During 1998, 129 pediatric outpatients (mean age, 12.1 +/- 2.3 years; age range, 10-15 years; 41 boys, 88 girls) were undergoing gastroduodenal endoscopy for evaluation of chronic abdominal pain. H. pylori colonization was determined by histology, urease test, and polymerase chain reaction. H. pylori IgG antibodies were found by using an enzyme-linked immunosorbent assay., Results: There was a high prevalence of H. pylori infection (80%) and peptic ulcers (24%) among the study group. Duodenal ulcers were detected in 31 of the children; all of them were H. pylori positive. Family history of peptic ulcers, nighttime pain associated with nocturnal awakening, fasting pain relieved by food, pain associated with meals, postprandial pain, bitter taste, and heartburn were the clinical signs that helped to distinguish the ulcer-positive children from the ulcer-negative H. pylori group. Duodenal ulcer patients had higher anti- H. pylori IgG titers compared with the levels of IgG antibodies in the infected children without ulcers ( P < 0.001). Peptic ulcer disease was a more common finding in the Ural ethnic group of Asians (Bashkirs) compared with the pediatric population of Russian origin., Conclusions: These results provide further evidence for a causal relation between H. pylori -associated peptic ulcer disease in childhood and relevant clinical symptoms. High titers of anti- H. pylori IgG might serve as a useful noninvasive indicator of ulcer disease.

Polymerase chain reaction (PCR) was used to study the frequencies of the gene of a low-molecular-weight alanine-rich cold shock protein in plants of temperate climate. This gene was demonstrated to be specific for the family Cruciferae. Its variability in this family was studied.

The nucleotide sequences of 280-360-bp domains of lectin genes from 20 legume species belonging to 17 genera have been determined. A computer analysis of the sequences has been performed with the LASERGENE package. Based on this analysis, we constructed the phylogenetic tree of the lectins, which reflects their phylogenetic and evolutionary relationships, and predicted the amino-acid sequences of the corresponding protein domains. Features of the structure of the hydrocarbon-binding lectin domains were elucidated in some species of legume genera from the temperate climatic zone. The domains were highly variable and contained the consensus sequence AspTrePheXxxAsxXxxXxxTrpAspProXxxXxxIns/DelArgHis bearing the bulk of amino acid replacements, insertions, and deletions. An association between legume groups (including species from different genera and tribes) symbiotic with the same rhizobium species and the similarity between the hydrocarbon-binding domains of lectins from these plants was found.

Fragments of the minor S and middle M segments of Hantavirus genome RNA were isolated by reverse transcription-polymerase chain reaction (RT-PCR) from the organs of patients who died from hemorrhagic fever with the renal syndrome (HFRS) during an epidemic outbreak of this infection in the Bashkortostan Republic in 1997-1998. The sizes of the resultant PCR fragments were 317 p. n. for S segment and 416 p. n. for M segment. Their primary structures (GENBANK registration numbers AJ133581 and AJ133582) were determined and they were compared with the nucleotide sequences of the respective genome sites of Hantavirus strain CG1820/Ufa-83 isolated from Clethrionomys in Bashkortostan in 1983. The homologies of nucleotide sequences of the corresponding sites of S and M segments of RNA isolate of 1997 and CG1820/Ufa-83 were 87.7 and 96.6%, respectively. The authors discuss the probability of circulation of hantaviruses differing by individual features of primary structure of genome RNA in Bashkortostan.

A phylogenetic tree was constructed on the basis of the amino acid sequences of the known cyclodextrin glucanotransferases (CDGTs), including those deduced from the nucleotide sequences of Bacillus sp. strain 6.6.3 and Paenibacillus macerans IB-7 genes encoding alpha- and beta-CDGTs. The tree clearly demonstrates the existence of distinct phylogenetic groups of CDGT-producing microorganisms and the divergence of the alpha-, beta-, and gamma-CDGT produced by microorganisms from the genera Bacillus, Paenibacillus, Brevibacillus, and Thermoanaerobacter from a common ancestor, whereas the CDGT of Klebsiella pneumoniae is independent and results from the convergence of different ancestors. The degree of homology of the leader peptide sequences of CDGTs may serve as a criterion of intraspecies relatedness between CDGT-producing microorganisms.

A key problem of honeybee (Apis mellifera mellifera) breeding in the Southern Urals is its cross-breeding with the Caucasian honeybee Apis mellifera caucasica. Mitochondrial DNA (mtDNA) in these subspecies differ in the length of a fragment localized between genes CO-I and CO-II, which can be used as a marker. A pair of 20-mer primers for PCR was chosen by means of computer design in order to determine the fragment size in both of the subspecies. The amplified fragment was shown to have a length of 350 bp in A. m. caucasica and 600 bp in A. m. mellifera. The difference in length results from the different ratio between two main elements P and Q, which comprise a major part of this sequence in these subspecies: a copy of P element and two copies of Q element in A. m. mellifera, and a copy of Q element only in A. m. caucasica. This sharply defined distinction allows us to use PCR for differentiating the subspecies, estimating the heterogeneity in the colonies, and rejecting queens in the selection process because of the maternal inheritance of the studied character. The nucleotide sequence of the amplified mtDNA fragment of A. m. mellifera was determined.

In diploid wheat Triticum monococcum L., a novel family of AT-rich repeated DNA sequences arranged in clusters of tandem arrays of 560-bp fragments is described. The nucleotide sequence of a 556-bp repeat unit was determined. The basic structural unit of this repeated DNA family contains subrepeats with the main motif AAACATTGTTAC. This type of repetitive DNA sequence could evolve via tandem amplification of short subrepeats with some subsequent divergence, and/or via recombination with unique sequences. Structural organization of homologous sequences, revealed by blot hybridization in other representatives of the Triticeae tribe, suggest that this family of repeated DNA is genome-specific.

In the intergenic spacer (IGS) of rDNA of diploid wheats Triticum urartu, T. boeoticum, and T. monococcum, the uncommon motives adjacent to the site of transcription initiation (TIS) are revealed. They are located in the region from -6 to +1 relative to the putative TIS and are not encountered in cereals studied earlier. In T. urartu and T. boeoticum, the motif TACTATG has been revealed, in T. monococcum--TATTATG, while diploid Aegilops speltoides has the motif TATAGTA, typical of the remaining cereal species. The TIS-surrounding rDNA IGS region of diploid wheats was compared to the correspondent known rDNA IGS regions of different plant and animal species.

Relationship was studied between the polymorphism of alpha 1-antitrypsin (Pi), haptoglobin (Hp) and transferrin and the development of chronic unspecific impairment of children lungs. Combination of protein isoforms PiM3M3 and Hp 1-1 was found to increase the risk of the lungs unspecific impairments in persons with lingering pneumonia and caused more severe development of the disease. Relatively low activity of the isoform PiM3M3, as compared with other forms of the PiMM system, and more intensive and nonadequate rate of lipid peroxidation in patients containing HP 1-1 appear to be responsible for aggravation of these impairments.

The organization of 18S and 26S rRNA in 14 plant species, belonging to 8 tribes of the cereal family was studied. In rDNA of all the cereals studied, except maize and reed, the similar character of localization of nucleotide sequences, recognized by restrictases BamHI and EcoRI in 18S and 26S rRNA genes was revealed. The structural organization of rDNA of sainfoin (Papilionaceae) was shown to differ from genes, coding for high molecular rRNA in cereals. The primary structure of subrepeat of non-transcribed rDNA spacer of diploid wheat Tr. urartu, consisting of 132 base pairs was determined. The given subrepeat was hybridized with BamHI-fragments of DNA from cereals and sainfoin. It is shown to hybridize with rDNA of all the cereals studied, and it hardly hybridizes with rDNA of maize and sorghum, but doesn't hybridize with rDNA of sainfoin. The conclusion is made that the size polymorphism of restriction fragments in the coding rDNA region and the level of similarity of subrepeats of rDNA of the non-transcribed spacer may help to reveal the phylogenetic affinity of plants, belonging to different tribes within one family.

Nucleotide sequences of 5.8S rRNA gene and rDNA internal transcribed spacers ITS-1 and ITS-2 were determined in diploid wheat Triticum urartu. It was shown that 5.8S rRNA gene of this wheat species consists of 163 base pairs and GC-content is 59.5%. When comparing 5.8S rRNA sequences in diploid wheat, rice and lupine and also 5.8S rRNA in hexaploid wheat and horse beans a high evolutional conservatism of its structure was revealed. The size of ITS-1 and ITS-2 in Tr. urartu is 219 and 225 base pairs long correspondingly. While comparing structures of similar rDNA regions of Tr. urartu, rice and maize a high level of homology was found only between nucleotides adjoining genes of high molecular rRNAs. In ITS-1 of Tr. urartu an insertion of 5'-GACGACGACATTGTCCGTC-3' was found, which is absent in maize and rice.

Two types of repeated sequences 320 and 510 b. p. long, that hybridize with 32P-5S rRNA, were determined in the genome of diploid wheat Triticum monococcum L. Using pBR327 plasmid, recombinant plasmids pTm5S7 and pTmS12, containing 5S DNA of both types, were constructed. The primary structure of the insertion into plasmid pTm5S7 was determined. The given 5S DNA repeated unit was shown to consist of 327 base pairs and to lack highly diverged sequence between 133 and 214 bases in 3'-flanking region. Eight nucleotide changes in the encoding region, including four changes in the transcription control region were found. Partial or full elimination of 5S DNA repeats were shown in genomes of tetra- and hexaploid wheat species. It was supposed that 5S DNA gene of Triticum monococcum in 327 b. p. repeats should represent a gene with relatively low transcriptional activity due to base changes within the limits of the intragene transcription control region.

Primary structures of 5S rRNA genes and of non-transcribed spacers between them were determined in families of 5S DNA repeats 420 and 500 b.p. long in 8 wheat and Aegilops species. The high conservatism of sequences coding for 5S rRNA, 3'- and 5'-ends of non-transcribed spacers was shown not to depend on the evolutional position, ploidy level and genomic composition of species. The activity of transcription of 5S rRNA cloned genes was determined in vitro. The functional heterogeneity was revealed in each family of repeats due to the existence of exchanges of separate nucleotides within the internal transcription control region. A greater deficiency of CpG dinucleotide was revealed in 5S rRNA genes than in non-transcribed spacers.

Nucleotide sequences of two 5S rRNA genes located in repeated 327 bp long units were determined in diploid wheat Triticum monococcum. They were compared with sequences of 5S rRNA genes of Tr. monococcum and Tr. aestivum which were earlier determined. The differences were revealed in two localizations of the nucleotide sequence in 5S DNA coding regions of Tr. monococcum and - in nine localizations in nontranscribed spacer. It was established that the nucleotide sequence of 5S rRNA gene cloned in pTm5S9 plasmid and 5S DNA coding region in Tr. aestivum have significant homology. Diploid wheat Tr. monococcum was supposed to have 5S rRNA genes with different functional activity within one multigene family.

In genome of diploid aegilops A. tauschii (k773) two types of sequences 420 and 510 b. p. long which hybridize with 5S[32P]rRNA were discovered. Using the plasmid pBR327 the recombinant plasmids pAt5S79 and pAt5S91 containing 5S DNA repeats of Ae. tauschii were constructed. The primary structure of 5S rRNA gene and that of intergene spacer cloned in pAt5S79 was determined. In gene region of Ae. tauschii coding for 5S rRNA, as compared with T. aestivum, there were found 3 base substitutions in positions 24, 36 and 37. The homology of spacer sequences is 79.1%. In 5S rRNA gene of Ae. tauschii GC-content is 53.3%, but in a spacer--48.1%. The terminator of transcription in Ae. tauschii includes 15 AT-base pairs with predomination of T-bases in uncoding chain.

The primary structure of intergenic non-transcribed and external transcribed spacers of rDNA of diploid wheat Triticum urartu, cloned in pTu3 plasmid 2402 b.p. long was determined. The intergenic non-transcribed rDNA spacer of Tr. urartu was shown to consist of 8 subrepeats with an average of 133 b.p. long, heterogeneous in length and nucleotide sequence. A number of repeated sequences was revealed within each subrepeat. While comparing nucleotide sequences of rDNA subrepeats of Tr. urartu and Tr. aestivum a high homology was found (up to 82%). A high similarity between these plant species was also found in the promoter region and in the external transcribed rDNA spacer. Suppression of the nucleolar organizer of 1A chromosome in the presence of 1B and 6B chromosomes of Tr. aestivum is supposed to be connected with the existence of a great number of subrepeats in the intergenic non-transcribed rDNA spacer of B genome donors in polyploid wheat species of turgidum-aestivum row.
