The field trial evaluating the effect of biochar on physical and hydro-physical properties of a Dystric Cambisol was carried out at the Research Grassland Station Vatín. The observed variants of the biochar experiment were as follows: 1) the control without biochar, 2) the biochar rate of 15 t/ha applied to the topsoil, 3) the biochar rate of 30 t/ha applied to the topsoil and 4) the biochar rate of 45 t/ha applied to the topsoil. It was found out that even the highest rate of biochar 45 t/ha did not affect saturated hydraulic conductivity of the topsoil at the depth of 0.05-0.20 m. When monitoring bulk density, it was confirmed that the biochar rate of 45 t/ha significantly reduced bulk density from 1218 kg/m3 to 1169 kg/m3. In the crop rotation without manure and clover growing the application of biochar at the rate of 45 t/ha resulted in the relevant decline in the average value of bulk density from 1197 kg/m3 to 1138 kg/m3. In the Norfolk crop rotation there were the inconclusive differences in bulk density. The rate of 45 t/ha had a highly noticeable impact on porosity. It increased from 53.75% (0 t/ha) to 55.60%. The biochar doses did not affect the value of field capacity. A potential effect of adjuvants/wetting agents added to the spray mixture on the water stability of soil aggregates (WSA) in agricultural soil was studied. Nine sites were chosen in the Czech Republic. Each site was mapped using representative soil pits (depth min. 1.3 m). A total of 54 mixed samples were collected from topsoil horizons on the selected sites. The samples were exposed to the action of four different types of wetting agents (organosilicone wetting agent; methyl ester of rapeseed oil; mixture of methyl ester palmitic and oleic acids; isodecyl alcohol ethoxylate), which are the most common wetting agents used in agriculture in the Czech Republic. WSA was determined before and after the addition of wetting agents (WA). Initial WSA values were at the same level in a majority of sampling points. Two sites were an exception, on which Haplic Luvisols and Relictistagnic Fluvisols occurred. These soil types featured the lowest WSA values. After the addition of WA across the sampling points, average WSA values exhibited a demonstrable trend: WSA of control sample (without the WA application) was at all times higher than in samples with the addition of WA. If the measured WSA values are compared in terms of overall means, it is evident that the control variant always exhibited the highest WSA value (on average 44.04 %) and the variants with the application of WA showed always WSA values lower by min 16 %. The worst effect on WSA was that of wetting agents whose basic component was methyl ester of rapeseed. These wetting agents caused a decrease in WSA by more than 50 %. All soil samples were also analysed for basic soil parameters (glomalin, oxidizable carbon - C-ox, pH, Na, P, Ca, K, Mg) in order to determine their potential influence on aggregate stability and to possibly eliminate the negative impact of WA. Objective. Dehydrogenase activity after the biological activation of biochar by the native soil microorganisms was studied. The main aim was to improve biochar properties by activation and make it more friendly for the soil microflora. Materials and methods. The activation was reached by aerating with the soil solution for two weeks. No special inoculum of microorganisms was applied. The following treatments in four replicates were prepared: conventional raw biochar (BR), activated biochar (BA), mineral fertilizer DAM 390 (NF), and control (C). A statistical test for comparing treatments means (Fisher p≤0.05; program STATISTICA 12.0; StatSoft software Inc., Tulsa, Oklahoma, USA) was used. Results. Statistically significant differences in the dehydrogenase activity between the treatments BR, BA, and C were found. Application of mineral fertilizers had a negative effect and increasing of nitrogen leaching was observed. Conclusions. El objetivo principal era mejorar las propiedades del biocarbón mediante su activación y hacerlo más amigable para la microflora del suelo. Materiales y métodos. La activación se logró mediante la aireación del suelo con la solución durante dos semanas. No se aplicó ningún inóculo especial de microorganismos. Se prepararon los siguientes tratamientos en cuatro réplicas: biocarbón crudo convencional (BR), biocarbón activado (BA), fertilizante mineral DAM 390 (NF) y control (C). Se utilizó una prueba estadística para comparar las medias de los tratamientos (Fisher p≤0.05; programa STATISTICA 12.0; StatSoft software Inc., Tulsa, Oklahoma, USA). Resultados. Se encontraron diferencias estadísticamente significativas en la actividad deshidrogenasa entre los tratamientos BR, BA y C. La aplicación de fertilizantes minerales tuvo un efecto negativo y se observó un aumento de la lixiviación de nitrógeno. Conclusiones. La activación del biocarbón es un método adecuado para mejorar la condición de la biota del suelo en comparación con el biocarbón convencional.

V úvodní teoretické části jsou popsány význam a mechanismy účinku acetylcholinu a acetylcholinesterasy v organismu, dále základní vlastnosti, metabolismus a interakce kofeinu s receptory. V experimentální části se práce zabývá možnostmi konstrukce biosenzoru pro stanovení inhibice acetylcholinesterasy kofeinem. Aktivita enzymu a stanovení kinetických parametrů Km, Vmax, Ki nebo IC50 byla zkoumána spektrofotometrickou Ellmanovou metodou a elektrochemickými metodami. Z těchto byly použity square-wave voltametrie a potenciometrická titrace v pH-statu. Butyrylcholinesterasa je enzym tvořený v játrech patřící do skupiny cholinesteras. Butyrylcholinesterasa nemá v organismu fyziologický substrát a její doposud neznámý význam v organismu je předmětem výzkumu. Stanovení aktivity tohoto enzymu se nejčastěji provádí pomocí Ellmanovy metody. Butyrylcholinesterasa je považována za ideální rozpoznávací prvek přírodního původu a v kombinaci s fyzikální senzorickou částí je využívána pro přípravu biosenzorů. Acetylcholinesterasa (AChE) je enzym, který se podílí na urychlení rozpadu acetylcholinu na synapsi. Bez přítomnosti AChE by tento rozpad trval neúměrně dlouho a vedení vzruchů by nebylo možné. Právě toto způsobují organofosfátové a karbamátové pesticidy a bojové látky. V diplomové práci jsme tyto látky prokazovali Ellmanovou metodou, což je metoda relativně spolehlivá, rychlá, reprodukovatelná a levná. Pro zavedení do praxe jsme používali 96 jamkové mikrotitrační destičky s imobilizovanou AChE. Toto uspořádání umožňuje rychlé stanovení velkého množství vzorků v jeden okamžik a tím značnou úsporu času při stanovování inhibitorů AChE. Jako alternativní chromogenní substrát jsme používali indoxylacetát., Acetylcholinesterase is enzyme which accelerates decomposition of acetylcholine on synapse. Následující práce je věnována stanovení tkáňových markerů oxidačního stresu v modelu laboratorního zvířete. Teoretická část práce se věnuje oxidačnímu stresu. Obsahuje vyjmenované reaktivní formy kyslíku a dusíku a význam přechodných kovů. Důsledky oxidačního stresu jako je lipidová peroxidace, poškození DNA, poškození proteinů a změny v metabolismu iontů. Dále je věnována pozornost antioxidačním bariérám chránícím organizmus před působením reaktivních forem. Nízko a vysokomolekulárním antioxidantům, enzymovým antioxidantům a dalším. Na konci teoretické části jsou uvedeny příklady metod používaných ke stanovení oxidačního stresu. Experimentální část se věnuje vyšetření markerů oxidačního stresu v modelu laboratorního zvířete, kterým byl podán čistý destilovaný sirný yperit a sodná sůl 2-merkaptoethanolsulfonové kyseliny. K tomuto účelu bylo využito několika vybraných metod. Stanovení nízkomolekulárních antioxidantů metodou, stanovení malondialdehydu a dalších látek reagujících s 2-thiobarbiturovou kyselinou, stanovení glutation reduktázy a stanovení karboxylovaných proteinů., The following thesis was focused on assay of oxidative stress markers in a laboratory animal model. Theoretical part is focused on role of oxidative stress in the body. It contains a list of reactive oxygen and nitrogen species and an importance of heavy metals. The consequences of oxidative stress, such as lipid peroxidation, DNA damage, protein degradation and alteration in basal metabolism. Attention is also focused on antioxidant barriers protecting the body against the effects. Low and high molecular antioxidants, enzymatic antioxidants and others are exampled in the thesis. The terminal chapters of theoretical part are focused on examples of methods suitable for oxidative stress assay. Experimental part is focused on examination of oxidative stress markers in a laboratory animal model. The animals were exposed to sulfur mustard. A separate group of the animals was treated by 2-mercapto-ethanesulfonate known as MESNA preparation. For purpose of the experiment, we have chosen several biochemical methods. Diplomová práce byla zaměřena na stanovení nízkomolekulárních antioxidantů a markerů oxidačního stresu v biologických vzorcích. V teoretické části byla věnována pozornost oxidačnímu stresu jako takovému. Byly popsány různé cesty vniku reaktivních forem kyslíku a dusíku (RONS), jakožto hlavních původců poškození biomolekul. Proteiny, lipidy a DNA jsou modifikovány RONS, a tím dochází k rozvoji patologických procesů. V dalších kapitolách byla věnována pozornost antioxidačním obranným systémům (AOS), jako jsou antioxidační enzymy a nízkomolekulární antioxidanty. Pro experimentální část byl proveden výběr metod, s ohledem na dostupnost metody z hlediska vybavení a výpovědní hodnoty sledovaného parametru. Rovněž byla zkoumána toxikologie sirného yperitu (bojové označení HD), jeho mechanizmus účinku a vliv na oxidační stres. V experimentální části byly vybrané metody testovány na reálných vzorcích. Pro stanovení celkové antioxidační kapacity byla použita metoda FRAP, pro zkoumání produktů oxidačního stresu, přesněji sekundární produkt neenzymové oxidace lipidů malondyaldehyd (MDA), metoda TBARS a byla stanovena hladina hlavního intracelulárního antioxidantu glutationu (GSH). Stanovení antioxidačních enzymů, jako je glutathionreduktasa (GR) a glutation-S-transferasa (GST) bylo rovněž provedeno. Jako poslední marker byl zkoumán apoptotický enzym kaspasa 3. Byl proveden experiment s potkany kmene Wistar, kterým byl profylakticky podán antioxidant epigallokatechin gallát (EGCG) v různých dávkách a po 1 h byly vystaveni sirnému yperitu (HD). Popsanými metodikami byly zkoumány změny parametrů oxidačního stresu a antioxidační obrany organizmu. Získané výsledky byly diskutovány v závěru práce., This thesis was focused on determination of low molecular antioxidants and markers of oxidative stress in biological samples. In theoretical part of the thesis, was aimed our attention to oxidative stress itself. In the beginning of thesis, we described a few ways of reactive oxygen and nitrogen species (RONS) formation, which are potent to attack important biological molecules. Proteins, lipids and DNA are modified and pathology can appear consequently. The second view was focused on antioxidant defense systems, like antioxidant enzymes and low molecular antioxidants. We also have chosen the methods suitable for experimental part of the thesis, with the view on availability in light of accessories and scientific value of the tracked markers. Next intention was concentrated on sulphur mustards (HD), as an chemical warfare and his effect on oxidative stress. In an experimental part of this thesis, we have chosen methods tested on biological samples. The method FRAP was used for determination of total antioxidant capacity. TBARS method was chosen for assessment of lipids peroxidation represented by malondialdehyde (MDA). Another method determinates total glutathione (GSH) level. Next two methods served for determination of two major antioxidant enzymes, glutathione reductase (GR) and glutathione-S-transpherase (GST). Consequently, we estimated apoptosis by assay of enzyme, caspase 3. Wistar rats were used in the experiment. They were treated with an antioxidant epigallocatechin gallate (ECGC) prior to HD poisoning. Alteration in oxidative stress and antioxidant properties in the animal body were investigated using the chosen methods. Oxidační procesy doprovázející oxidační stres hrají v organismu důležitou roli ve fyziologických i patologických procesech. Vzájemně kooperující antioxidační systémy zajišťují v těchto procesech rovnováhu. Jedním z mnoha faktorů, které ruší rovnováhu ve prospěch oxidačního poškození organismu, je ionizující záření. Hlavním cílem experimentu bylo zaměřit se na stanovení markerů oxidačního stresu a jeho dopad na tkáně laboratorních potkanů exponovaných ionizujícímu gama záření a v další části pak účinku aplikované látky HI-6, na modulaci oxidačního stresu po vystavení ionizujícímu záření. Tyto účinky byly zkoumány na samcích laboratorních potkanů Wistar. Látka HI-6 jim byla aplikována intramuskulárně. Ozáření bylo provedeno celotělově přístrojem 60Co, kterým exponovaná zvířata obdržela dávku 7,5 Gy. Pro stanovení markerů oxidačního stresu jsme použili prověřené analytické metody se spektrofotometrickou detekcí. Podařilo se potvrdit předpokládaný negativní účinek ionizujícího záření, projevený nárůstem markerů lipidové peroxidace v tělních tekutinách i tkáních indukující oxidační stres. Tyto projevy byly současně doprovázeny poklesem hladiny antioxidantů v tělních tekutinách, naopak mírným vzrůstem v játrech. Látka HI-6 byla prokázána jako potenciální radioprotektivum. Další část experimentu zaměřená na sledování vlivu dávky záření na jednotlivé markery však ukázala na jeho ambivalentní dopad. They do very important part in organism in physiological and pathological processes. Mutual cooperative antioxidant systems provide balance in these processes. One of many factors which disturb balance in benefit oxidizing damage of organism is ionizing radiation. Main goal of experiment was to focuse on determination of markers and its incidence of tissues on white rats which are exposed by ionizing gama radiation. In next part as a result of applied dose HI-6 for modulation of oxidizing stress after exposure ionizing radiation. These results were examinated on males Wistar rats. Preparation HI-6 was applied them intramuscularly. Radiation was realized on whole body by 60Co source. Exposed animals got dose 7.5 Gy. We used verified analytical method with spectrophotometric detection for determination of markers. We succeeded in fact of negative effect ionizing radiation which displays rising of lipid peroxidation markers in body fluid and tissues in induction oxidative stress. These exposures were at the same time attended by decline of antioxidative level in body fluids but slightly rise in liver. Preparation HI-6 was positive as potential radioprotector. Next part of experiment was focused on monitoring dose of radiation on each marker. It shows his ambivalent impact.
