2. Neuroprotective effects of Sideritis trojana essential oil in AlCl3-induced amnesic rats.

In this study, the effects of Sideritis trojana essential oil on memory, anxiety, and depression were investigated in rats subjected to AlCl
3 -induced dementia. The major constituents of S. trojana essential oil were analyzed as carvacrol (42.32%), carvone (18.69%), and α-pinene (4.54%). Rats were divided into five groups: Control, positive control (donepezil), AlCl3 -only applicated, AlCl3 with 40% S. trojana essential oil applicated, and AlCl3 with 80% S. trojana essential oil applicated groups. The essential oil was administered for 30 days, after that, the rats were subjected to behavioral tests. In the forced swimming test, S. trojana essential oil application demonstrated antidepressant-like effects. In the elevated plus maze and open field tests, S. trojana essential oil application exhibited anxiolytic-like effects. Furthermore, the application of S. trojana essential oil ameliorated memory impairment induced by AlCl3 by increasing spontaneous alternation in the Y-maze test. Furthermore, S. trojana essential oil application reduced the level of malondialdehyde and increased catalase activity, demonstrating antioxidant activity. Molecular docking analyses were performed between the major compounds of S. trojana essential oil and the AMPA receptor to evaluate possible mechanisms of action leading to neuroprotection. Carvacrol, carvone, and α-pinene were shown to possess strong binding abilities to the AMPA receptor, which may result in allosteric modulation of the receptor. This study demonstrated the neuroprotective effects of S. trojana essential oil in an animal model induced by AlCl3 application, suggesting its potential protective role against central nervous system disorders. Purpose: This study aimed to determine the serum sclerostin levels in kidney transplant recipients, dialysis patients, and patients with non-dialysis chronic kidney disease (CKD), and evaluate their associations with carotid intima-media thickness (CIMT) and carotid artery plaque presence. Methods: The study groups included kidney transplant recipients (n = 61), dialysis patients (n = 43), non-dialysis CKD patients (n = 43), and healthy controls (n = 19). Serum sclerostin levels were measured by the ELISA method. All participants underwent imaging tests for the evaluations of CIMT and carotid plaque presence. Results: Serum sclerostin levels were highest in dialysis patients, followed by the non-dialysis CKD group. Kidney transplant patients and healthy controls had the lowest sclerostin levels. There was a positive correlation between CIMT and sclerostin levels in the CKD group. Sclerostin levels were significantly high in individuals with calcified plaques. Conclusion: The CIMT and serum sclerostin levels of kidney transplant recipients compared to those of CKD patients with and without dialysis may suggest that transplantation may prevent further atherosclerosis, with sclerostin levels promising a predictive role. The most important side effect of gentamicin (GM) is nephrotoxicity. p-Coumaric acid (PCA) is a phenolic compound that scavenges free radicals, reduces fibrosis, and tissue damage. This study investigates the protective effect of PCA on tissue damage and kidney function in gentamicin-induced nephrotoxicity (GIN). Thirty-five rats were separated into five groups and each group contained seven animals: control group, ethanol group, GM group, PCA group, and GM + PCA group. At the end of the seven-day treatment, the rats were sacrificed after blood and kidney tissue samples were taken. While serum urea, creatinine, and neutrophil gelatinase-associated lipocalin (NGAL) levels increased significantly in the GM group compared to the control, they showed a significant decrease in the GM + PCA group compared to the GM. Serum tumor necrosis factor-α (TNF-α) and tissue malondialdehyde (MDA) levels were significantly increased in the GM group compared to the control. While the tissue total oxidant status (TOS) and oxidative stress index (OSI) values of the GM group were significantly higher than the control, they showed a significant decrease in the GM + PCA group compared to the GM. In the histopathological examination, significant tubular necrosis and tubulointerstitial inflammation were detected in the proximal tubules in the GM group compared to the control, while a significant decrease was observed in the severity of these findings in the GM + PCA group compared to the GM. This study shows that PCA has biochemical and histopathological ameliorating effects on GIN in the rat model. [ABSTRACT FROM AUTHOR]

Bu çalışmada Ölüm Eşiği Deneyimleri(ÖED) din psikolojisi çerçevesindeele alınmaktadır. Bu çerçevede, yaşayanların hayatlarında dinive psikolojik açıdan köklü değişikliklere neden olan bu deneyimdegörülen temel karakteristiklere, deneyim öncesi, esnası ve sonrasındakipsiko-sosyal durumlara ışık tutulmaktadır. Konuya ilişkin kavramanalizlerini ve ÖED literatürünü içermenin yanı sıra çalışmanınen özgün katkısı, nitel araştırma modellerinden mülakat tekniğiyleelde edilen veriler vasıtasıyla olmuştur. Amaçlı örnekleme yöntemiylebelirlenen ve kartopu tekniğiyle ulaşılan, ÖED yaşamış 13 kişiylemülakat yapılmıştır. Within this framework, the aim is to shed light on NDEs’ basic features as well as the psychosocial attitudes observed during pre-, mid- and post-experience which have profound effects that change the experiencers’ religious beliefs and psychological states. The study does not only include relevant concept analysis and NDE literature but also bears an original approach thanks to the data collected by qualitative re- search models adopted through interviews. Bu araştırmanın konusu, klinik olarak ölü kabul edilen insanların ruhlarının fiziksel bedenlerini terk etmesiyle farklı bir mekana gitmelerini; bedenlerinin dışında kendilerini veya etrafında olanları izlemelerini; ışık, ses, tünel vb. fenomenleri tecrübe etmelerini, geri döndükten sonra tüm yaşadıklarını detaylarıyla anlatmalarını içeren Ölüm Eşiği Deneyimleridir (ÖED). Son yıllarda bilimsel çalışmalarla yeni bir aşamaya evrilen bu deneyimler, aslında insanlık tarihi kadar eski bir geçmişe sahiptir. ÖED'lerin anlaşılması ve hakkıyla değerlendirilmesi, ancak onunla bağlantılı olan ruh, ruh-beden ilişkisi ve ölüm'ün bilinmesiyle mümkündür. Semavî dinlerden diğer dinî geleneklere, felsefeden mit ve efsanelere kadar her alanda kendine önemli bir yer bulan ÖED'ler, bu deneyimi yaşayan insanların hayatlarında derin dinî ve psikolojik değişikliklere yol açmaktadır. Din Psikolojisi açısından ÖED'nin ele alındığı bu tezde, ÖED'nin temel karakteristikleri, deneyim öncesi, esnası ve sonrası olmak üzere kapsamlı bir şekilde araştırılmıştır. Böylece daha önceki birikimin sağladığı zemin üzerinde, nitel araştırma modeli kullanılmıştır. Amaçlı örnekleme (purposive sampling) yöntemiyle belirlenen ve kartopu tekniğiyle ulaşılan ÖED yaşamış 13 kişiyle mülakat yapılmıştır. ÖED öncesine dair inançlar, kişilik yapıları, ruhsal haller gibi durumlarla ilgili bulgular elde edilirken; ÖED esnasına ilişkin literatürdeki bilişsel, afektif, paranormal ve transandantal unsurlara uygun olarak `hız, sesler, yaşamın göz önünden geçmesi, tünel, ışık, huzur, mutluluk, sevgi, aşk, birleşme, BDD, duyu ötesi algı, başka bir alem, başka bir varlıkla karşılaşma` gibi unsurlar; ÖED sonrasına ilişkin ise inançta, ibadette, kişilikte, ölüm ve sonrasına bakışta, empati ve iyilikte değişiklikler şeklindeki bulgulara ulaşılmıştır. This paper is about Near Death Experiences (NDEs) occurring to people who are considered clinically dead, and defined by characteristic features such as a feeling of separating from the physical body, entering into a foreign dimension, observing self from outside the body while witnessing events going on around them, providing a detailed recollection of these events when they return to their body, and the general perception of a profound feeling of love, peace and light. These experiences have been reported since ancient times, however recent scientific studies have taken them into a new step. To understand and rightfully assess NDEs, one must know about the related concepts, namely soul, soul-body relation and death. Having an important place in many subjects from Abrahamic religions to traditional beliefs, philosophy, myths and legends; NDEs have profound effects that change the experiencer's religious beliefs and psychological state. The present study uses psychology of religion approach to the phenomenon and provides an extensive research on their basic features, along with pre-, mid- and post-experience factors. Thus, a qualitative research model was applied while taking pre-existing conditions into account. A total of 13 near death experiencers selected using purposive sampling methods and contacted through the snowball technique were interviewed. While the findings related to the beliefs, personality types, psychological states before NDE obtained about pre-experience; the findings as `speed, voices, life-review, tunnel, light, peace, happiness, love, unity, OBE, extrasensory perception, another realm, encountering with others` obtained about mid-experience as cognitive, affective, paranormal and transcendental components are in line with the existent literature on NDEs and also differences in belief, personality, view for death and after-life, emphatizing and benevolence are obtained about post-experience. Amaç: Hemoglobinopatiler, globin fonksiyonlarını bozan bir grup hastalıktır. HPLC, hemoglobinopati taramalarında tüm dünyada en sık kullanılan tekniktir. Çalışmada, Türkiye'de bir üçüncü basamak hastane laboratuvarında 3 yıllık retrospektif hemoglobinopati sıklığı değerlendirilmesi amaçlanmıştır. Gereç ve Yöntem: Üniversite hastanemize başvuran 2461 hastanın 3 yıllık laboratuvar ve hastane kayıtları incelenmiştir. Hemoglobinopati taramasında Tosoh G8 HPLC cihazı kullanılmıştır Bulgular: HbA2%3,5 olan 668, %3,5HbA2%4 olan 19 sonuç saptanmıştır.19 sonuçtan 10'u betatalasemi minör olarak kabul edilmiştir. HbA2 %4 olan 649 sonuç vardır ve sonculara göre 602 hasta beta-talassemi minör olarak kabul edilmiştir. HbA2 düzeylerine göre, hastaların %25'i beta-talasemi minör olarak değerlendirilmiştir. HbF%2 olan 391 hasta saptanmış ve HbA2%3,5 olan 138 kayıt ayrıntılı incelemeye alınmıştır. Varyant analizi veri kümesi, 53 hasta içermektedir. HbA2%3,5 ve HbF%2 olan, varyant pikleri mevcut kayıtlar incelenmiş ve 28 vaka saptanmıştır. En sık görülen Hb varyantları sırasıyla HbH, HbS, HbD, HbC, HbE ve HbO-Arab'dır. Sonuç: Çalışmada en sık varyantlar sırasıyla HbH, HbAS, HbAD, HbAC, HbAE ve HbO-Arab olarak bulunmuştur. Beta talasemi minör sıklığı ülkemizin gelen popülasyonundan yüksek olarak %25 bulunmuştur. Özellikle hematolojik malignensiler, herediter sferositoz ve demir eksikliği anemisi gibi eşlik eden durumlar hemoglobinopati değerlendirmesinde akılda tutulmalıdır., Objective: Hemoglobinopathies are an array of disorders that affect hemoglobin (Hb) function. The aim of this study is to evaluate 3-year retrospective data of a tertiary hospital in Turkey according to prevalence of hemoglobinopathies. Material and Methods: A three-year data of 2461 patients admitted to our university hospital laboratory were assessed by scanning laboratory and hospital records. Tosoh G8 HPLC instrument was used for screening. Results: 668 results with HbA23.5% and 19 results with 3.5% HbA2 4% were detected. 10 of 19 patients were considered as beta-thalassemia. Number of patients with HbA2 4% were 649 and 602 of them were assessed as beta-thalassemia-minor. 25% of patients were assessed as beta-thalassemiaminor, based on HbA2 levels. 391 patient results were found to be HbF2% and 138 of them with HbA23.5% were further investigated. Variant analysis dataset contains 53 patients. Samples with HbA23.5% and HbF2% showing variant peaks were scanned and 28 cases were detected. The most common variants were found to be HbH, HbS, HbD, HbC, HbE and HbO-Arab, respectively. Conclusion: In this study, the most common variants were found to be HbH, HbAS, HbAD, HbAC, HbAE and HbO-Arab, respectively. Moreover, the frequency of beta-thalassemia minor was found as 25% which is higher than general population prevalence of our country. Accompanying diseases especially hematological malignancies, hereditary spherocytosis and IDA should be kept in mind during evaluating hemoglobinopathy analysis.

Amaç: Kronik Böbrek Hastalığında, kronik inflamasyon ve aterosklerozla ilişkili olarak kardiyovasküler hastalık riski artmaktadır. Hemodiyaliz ve periton diyaliz alan hastalara göre belirgin bir düzelme görülse de renal transplantasyon sonrası inflamatuvar süreç devam etmektedir. TNF reseptör ailesinin bir üyesi olan Decoy reseptör 3 (DcR3), inflamasyon ve ateroskleroz ile ilişkili bulunmuştur. Bu çalışmadaki amacımız, kronik böbrek ve renal transplant hastalarında; DcR3 seviyeleri ile inflamasyon ve ateroskleroz arasındaki ilişkiyi göstermektir. Bu amaçla, serum DcR3, hücreler arası adezyon molekülü-1 (intercellular adhesion molecule-1 (ICAM-1)), vasküler hücre adezyon molekülü-1 (vascular cell adhesion molecule-1 [VCAM-1]), interlökin-8 (interleukin-8 [IL-8]) seviyelerinin ölçülmesi, endotelyal disfonksiyonun karotid intima-media kalınlığı (KİMK) ve karotid arter plak varlığı ile değerlendirilmesi hedeflenmiştir.Gereç ve Yöntem: Çalışmaya 50 renal transplantasyon yapılan (Grup 1), 40 hemodiyaliz (HD) alan (Grup 2), 30 pre-diyaliz KBY (Grup 3) ve 30 sağlıklı kontrol (Grup 4) olmak üzere toplam 150 birey dahil edildi. Serum DcR3, VCAM-1, ICAM-1 ve IL-8 seviyeleri ELISA yöntemiyle çalışıldı. KİMK ve karotid arter plak varlığı non-invaziv olarak ultrason probuyla gerçekleştirildi.Bulgular: Serumda çalışılan tüm parametreler ve KİMK, HD ve pre-diyaliz KBY gruplarında renal transplantasyon ve kontrol gruplarına kıyasla yüksek bulundu. Renal transplantasyon grubunun sonuçları ise kontrol grubundan yüksekti. HD ve pre-diyaliz KBY grupları arasında fark bulunmadı. Hasta gruplarının plak varlıkları arasında fark yoktu. Kontrol grubunda plağa rastlanmadı. Pre-diyaliz KBY grubu kendi içinde değerlendirildiğinde KİMK dışında hiçbir parametrede evreler arasında fark saptanmadı. En yüksek KİMK değerlerinin evre 3'te olduğu görüldü.Sonuç: Renal transplantasyon, inflamatuvar süreçte belirgin bir düzelme sağlasa da inflamasyon sürecini tamamen geri döndürememektedir. Pre-diyaliz KBY ve HD grubunda üremik ortamla ilişkilendirilebilecek inflamatuvar sürecin ateroskleroza zemin hazırladığı söylenebilir. Pre-diyaliz KBY ve HD gruplarında artmış KİMK ve yüzde plak varlıkları bu durumu desteklemektedir.Anahtar kelimeler: DcR3, inflamasyon, ateroskleroz, KİMK, renal transplantasyon Aim: The cardiovascular risk has been increased in Chronic Kidney Disease (CKD) associated with chronic inflammation and atherosclerosis. Inflammatory process continues after renal transplantation. However it has been observed a significant improvement compared to patients with hemodialysis and periton dialysis. Decoy receptor 3 (DcR3), is a member of the TNF receptor superfamily and associated with inflammation and atherosclerosis. The aim of this study is to show the relationship between DcR3 level with inflammation and atherosclerosis in patients with chronic kidney and renal transplant patients. For this purpose, we intend to measure serum levels of DcR3, intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1) and interleukin-8 (IL-8) and evaluation of endothelial dysfunction via measurements of carotid intima-media thickness (CIMT) and presence of plaque. Materials and Methods: A total of 150 participant were obtained from 50 renal transplant patients (Group 1), 40 hemodialysis patients (Group 2), 30 pre-dialysis CKD patients (Group 3) and 30 control (Group 4). Serum DcR3, VCAM-1, ICAM-1 and IL-8 levels measured with ELISA method. CIMT and presence of carotis arter plaque performed by ultrasound probe, non-invasively. Results: All parameters were measured in serum and CIMT were higher in HD and pre-dialysis CKD groups compared to renal transplant and control groups. The levels of renal transplant group were higher from control group. There was no difference between HD and pre-dialysis CKD groups. There was no difference between patient groups in terms of the presence of plaque. Control group didn't show any plaque. Pre-dialysis group was no significant difference between the stages in any parameter except CIMT. CIMT levels was highest in stage 3.Conclusion: Although renal transplantation provides a significant improvement in the inflammatory process, not return completely. Inflammatory process associated with uremic milieu may predispose to atherosclerosis in pre-dialysis CKD and hemodialysis patients. Increased CIMT and presence of plaque in HD and pre-dialysis CKD groups supports this state.Key words: DcR3, inflammation, atherosclerosis, CIMT, renal transplantation. 80

The constriction of vessels due to atherosclerotic lesions causes hypoxia/ischemia and oxidative changes resulting in transformation of free albumin to ischemia-modified albumin (IMA) in the circulation and increased carotid intima–media thickness (cIMT). We investigated the reliability of IMA increase in evaluating atherosclerosis in patients with familial Mediterranean fever (FMF) compared with cIMT. Patients with FMF (n = 58) diagnosed by the Tel-Hashomer criteria in attack-free period and 38 healthy people were included in the study. Patient demographics as well as the clinical and laboratory characteristics of the healthy controls and patients with FMF were noted. The IMA levels and cIMT in patients with FMF were 0.30 ± 0.09 absorbance units (ABSUs) and 1.12 ± 0.27 mm, respectively, and in the control group, IMA levels and cIMT were 0.25 ± 0.07 ABSU and 0.74 ± 0.26 mm, respectively. The IMA levels and cIMT were significantly higher in patients with FMF than in controls (P = .020 and P < .0001, respectively). The IMA values showed positive correlation with cIMT in patients with FMF(r = .302, P = .041). Our results reveal that IMA—an oxidative stress marker—may be an indicator of atherosclerosis in patients with FMF. This finding deserves further investigation. [ABSTRACT FROM AUTHOR]
