Abstract: The group velocity of 'space-time' wave packets $-$ propagation-invariant pulsed beams endowed with tight spatio-temporal spectral correlations $-$ can take on arbitrary values in free space. Here we investigate theoretically and experimentally the maximum achievable group delay that realistic finite-energy space-time wave packets can achieve with respect to a reference pulse traveling at the speed of light. We find that this delay is determined solely by the spectral uncertainty in the association between the spatial frequencies and wavelengths underlying the wave packet spatio-temporal spectrum $-$ and not by the beam size, bandwidth, or pulse width. We show experimentally that the propagation of space-time wave packets is delimited by a spectral-uncertainty-induced `pilot envelope' that travels at a group velocity equal to the speed of light in vacuum. Temporal walk-off between the space-time wave packet and the pilot envelope limits the maximum achievable differential group delay to the width of the pilot envelope. Within this pilot envelope, the space-time wave packet can locally travel at an arbitrary group velocity and yet not violate relativistic causality because the leading or trailing edge of superluminal and subluminal space-time wave packets, respectively, are suppressed once they reach the envelope edge. Using pulses of width $\sim$4ps and a spectral uncertainty of $\sim$ 20 pm, we measure maximum differential group delays of approximately $\pm$ 150 ps, which exceed previously reported measurements by at least three orders of magnitude.

Abstract: Transmission spectroscopy of exoplanets has revealed signatures of water vapor, aerosols, and alkali metals in a few dozen exoplanet atmospheres. However, these previous inferences with the Hubble and Spitzer Space Telescopes were hindered by the observations' relatively narrow wavelength range and spectral resolving power, which precluded the unambiguous identification of other chemical species$-$in particular the primary carbon-bearing molecules. Here we report a broad-wavelength 0.5-5.5 $\mu$m atmospheric transmission spectrum of WASP-39 b, a 1200 K, roughly Saturn-mass, Jupiter-radius exoplanet, measured with JWST NIRSpec's PRISM mode as part of the JWST Transiting Exoplanet Community Early Release Science Team program. We robustly detect multiple chemical species at high significance, including Na (19$\sigma$), H$_2$O (33$\sigma$), CO$_2$ (28$\sigma$), and CO (7$\sigma$). The non-detection of CH$_4$, combined with a strong CO$_2$ feature, favours atmospheric models with a super-solar atmospheric metallicity. An unanticipated absorption feature at 4$\mu$m is best explained by SO$_2$ (2.7$\sigma$), which could be a tracer of atmospheric photochemistry. Abstract: International audience; A chiral 3D coordination compound, [Gd2(L)2(ox)2(H2O)2], arranged around a dinuclear Gd unit has been characterized by X-ray photoemission and X-ray absorption measurements in the context of density functional theory studies. Core level photoemission of the Gd 5p multiplet splittings indicates that spin orbit coupling dominates over j–J coupling evident in the 5p core level spectra of Gd metal. Indications of spin–orbit coupling are consistent with the absence of inversion symmetry due to the ligand field. Density functional theory predicts antiferromagnet alignment of the Gd2 dimers and a band gap of the compound consistent with optical absorption.

Abstract: International audience; Since the invention and further development of lasers in the 1960s, photonics has grown into a field that permeates virtually every aspect of modern life. As photonics deals with the generation, transmission, modulation, amplification, conversion, and detection of light, glasses have played crucial and central roles in a multitude of applications given glasses' diverse range of composi-tions, properties, and forms. They are also low-cost materials, recyclable and generally easy to fabricate. In homage to the United Nations International Year of Glass 2022, this paper reviews the past, present, and future of photonic glasses.

Abstract: Silica nanoparticles, prepared by the Stober method, have been doped with Eu 3+ , Dy 3+ , or processed to result in Au nanoparticles on the silica surface. The luminescence of the rare earth (RE)-doped SiO 2 particles has been studied as a function of the nature of the RE, their concentration and also of the presence of Au nanoparticles at the surface of the SiO 2 nanoparticles. We have shown that the Eu 3+ emission is observable over the experimental conditions examined, whereas it was not possible to observe any emission for Dy 3+ doped materials. No enhancement of the Eu 3+ emission was observed following the adsorption of gold nanoparticles at the surface of the SiO 2 nanoparticle, however an excitation at 250 nm leads to both the emission of the matrix and Eu 3+ showing an energy transfer from the SiO 2 matrix to Eu 3+ ions.

Abstract: We introduce and study the barcode entropy for geodesic flows of closed Riemannian manifolds, which measures the exponential growth rate of the number of not-too-short bars in the Morse-theoretic barcode of the energy functional. We prove that the barcode entropy bounds from below the topological entropy of the geodesic flow and, conversely, bounds from above the topological entropy of any hyperbolic compact invariant set. As a consequence, for Riemannian metrics on surfaces, the barcode entropy is equal to the topological entropy. Abstract: International audience; Spectral beam combining (SBC) by volume Bragg gratings (VBGs) recorded in photo-thermo-refractive (PTR) glass is a powerful tool for laser applications that require higher radiance than a single laser unit can achieve. The beam-combining factor (BCF) is introduced as a tool to compare various beam-combining methods and experiments. It describes the change of radiance provided by a beam-combining system but is not affected by the initial beam quality of the combined lasers. A method of optimization of VBGs providing the maximum efficiency of SBC has been described for an arbitrary number of beams. An experiment confirming the proposed modeling for a two-beam SBC system by a single VBG has demonstrated a total combined power of 301 W with a channel separation of 0.25 nm, combining efficiency of 97%, close to diffraction limited divergence with M2=1.18, BCF of 0.77, and spectral radiance of 770 TW/(sr⋅m2⋅nm), the highest to date for SBC.

Abstract: International audience; Tasks such as analysis, design optimization, and uncertainty quantification can be computationally expensive. Surrogate modeling is often the tool of choice for reducing the burden associated with such data-intensive tasks. However, even after years of intensive research, surrogate modeling still involves a struggle to achieve maximum accuracy within limited resources. This work summarizes various advanced, yet often straightforward, statistical tools that help. We focus on four techniques with increasing popularity in the surrogate modeling community: (i) variable screening and dimensionality reduction in both the input and the output spaces, (ii) data sampling techniques or design of experiments, (iii) simultaneous use of multiple surrogates, and (iv) sequential sampling. We close the paper with some suggestions for future research.

Abstract: International audience; Applications of quantum information science (QIS) generally rely on the generation and manipulation of qubits. Still, there are ways to envision a device with a continuous readout, but without the entangled states. This concise perspective includes a discussion on an alternative to the qubit, namely the solid-state version of the Mach–Zehnder interferometer, in which the local moments and spin polarization replace light polarization. In this context, we provide some insights into the mathematics that dictates the fundamental working principles of quantum information processes that involve molecular systems with large magnetic anisotropy. Transistors based on such systems lead to the possibility of fabricating logic gates that do not require entangled states. Furthermore, some novel approaches, worthy of some consideration, exist to address the issues pertaining to the scalability of quantum devices, but face the challenge of finding the suitable materials for desired functionality that resemble what is sought from QIS devices.

Abstract: International audience; Photo-thermo-refractive (PTR) glass is a photosensitive multi-component silicate glass that exhibits refractive index change after successive UV-exposure and thermal treatment. The refractive index change was demonstrated to be associated with the precipitation of NaF nano-crystals in the glass matrix. This paper presents a systematic study of the dependence of the refractive index change on dosage of UV-exposure and thermal treatment duration and temperature in already UV-exposed and nucleated samples. It is shown using Avrami plots that the refractive index change in PTR glass is determined by a diffusion-controlled growth of particles from pre-existing nuclei while all coefficients have exponential dependence on temperature (Boltzmann law). The developed model, which includes both photo-chemistry and crystallization processes, allow describing the refractive index change dependence on dosage for a wide range of thermal treatment duration and temperature and an accuracy better than 10%.

Abstract: In the present paper we consider the varying coefficient model which represents a useful tool for exploring dynamic patterns in many applications. Existing methods typically provide asymptotic evaluation of precision of estimation procedures under the assumption that the number of observations tends to infinity. In practical applications, however, only a finite number of measurements are available. In the present paper we focus on a non-asymptotic approach to the problem. We propose a novel estimation procedure which is based on recent developments in matrix estimation. In particular, for our estimator, we obtain upper bounds for the mean squared and the pointwise estimation errors. The obtained oracle inequalities are non-asymptotic and hold for finite sample size.

Abstract: International audience; The classical Kuiper Belt Object (KBO) Arrokoth was surveyed by the New Horizons spacecraft on 1st January 2019, revealing a small bilobed object with a red surface, whose spectral slope lies in the average of the whole KBOs population. This red color has been assigned to reddish organic materials, either inherited from the protosolar disk during accretion, or formed through radiolytic processes in the surface due to exposure to solar or interstellar photons, Solar Wind, Solar Energetic Particles or Galactic Cosmic Rays. We report here a study investigating the radiolytic scenario, based on numerical calculations and experimental simulations run with swift heavy ions (74.8 MeV136Xe19+ and 33.06 MeV58Ni9+), and low-energy 105 keV18O6+ions on CH3OH ice, the only molecule identified at Arrokoth’s surface.

Abstract: International audience; Peregrine soliton (PS) is widely regarded as a prototype nonlinear structure capturing properties of rogue waves that emerge in the nonlinear propagation of unidirectional wave trains. As an exact breather solution of the one-dimensional focusing nonlinear Schrödinger equation with nonzero boundary conditions, the PS can be viewed as a soliton on finite background, i.e., nonlinear superposition of a soliton and a monochromatic wave. A recent mathematical work showed that both nonzero boundary conditions and solitonic content are not pre-requisites for the PS occurrence. Instead, it has been demonstrated that PS can emerge locally, as an asymptotic structure arising from the propagation of an arbitrary large decaying pulse, independently of its solitonic content. This mathematical discovery has changed the widely accepted paradigm of the solitonic nature of rogue waves by enabling the PS to emerge from a partially radiative or even completely solitonless initial data. In this work, we realize the mathematically predicted universal mechanism of the local PS emergence in a water tank experiment with a particular aim to control the point of the PS occurrence in space-time by imposing an appropriately chosen initial chirp. By employing the inverse scattering transform for the synthesis of the initial data, we are able to engineer a localized wave packet with a prescribed solitonic and radiative content. This enabled us to control the position of the emergence of the rogue wave by adjusting the inverse scattering spectrum. The proposed method of the nonlinear spectral engineering is found to be robust to higher-order nonlinear effects inevitable in realistic wave propagation conditions.

Abstract: Blue phosphorene (blue-P) has attracted considerable attention due to its potential applications in optical and electronic devices. However, its synthesis has remained a challenge. Here, we report an experimental investigation of the first steps of blue-P growth on Au(1 1 1) surface by molecular-beam epitaxy. The structure was characterized by in situ low temperature scanning tunneling microscopy, low-energy electron diffraction, combined with density functional theory calculations. We reveal two-dimensional (2D) phosphorus clusters (P-clusters) formed on surface at 150 °C, where the most prevalent structure of P-clusters is composed of triangles with six protrusions. We also demonstrate the transformation of these P-clusters into a single layer of blue-P after post-annealing at 260 °C. Our observation of the growth process is a necessary step for exploring the growth mechanisms further.

Abstract: International audience; The fast relaxation processes in the excited electronic states of functionalized aza-boron-dipyrromethene (aza-BODIPY) derivatives (1-4) were investigated in liquid media at room temperature, including the linear photophysical, photochemical, and nonlinear optical (NLO) properties. Optical gain was revealed for nonfluorescent derivatives 3 and 4 in the near infrared (NIR) spectral range under femtosecond excitation. The values of two-photon absorption (2PA) and excited-state absorption (ESA) cross-sections were obtained for 1-4 in dichloromethane using femtosecond Z-scans, and the role of bromine substituents in the molecular structures of 2 and 4 is discussed. The nature of the excited states involved in electronic transitions of these dyes was investigated using quantum-chemical TD-DFT calculations, and the obtained spectral parameters are in reasonable agreement with the experimental data. Significant 2PA (maxima cross-sections ∼2000 GM), and large ESA cross-sections ∼10(-20) m(2) of these new aza-BODIPY derivatives 1-4 along with their measured high photostability reveal their potential for photonic applications in general and optical limiting in particular.

Abstract: Japan Aerospace Exploration Agency (JAXA) plans a Phobos sample return mission (MMX: Martian Moons eXploration). In this study, we review the related works on the past climate of Mars, its evolution, and the present climate and weather to describe the scientific goals and strategies of the MMX mission regarding the evolution of the Martian surface environment. The MMX spacecraft will retrieve and return a sample of Phobos regolith back to Earth in 2029. Mars ejecta are expected to be accumulated on the surface of Phobos without being much shocked. Samples from Phobos probably contain all types of Martian rock from sedimentary to igneous covering all geological eras if ejecta from Mars could be accumulated on the Phobos surface. Therefore, the history of the surface environment of Mars can be restored by analyzing the returned samples. Remote sensing of the Martian atmosphere and monitoring ions escaping to space while the spacecraft is orbiting Mars in the equatorial orbit are also planned. The camera with multi-wavelength filters and the infrared spectrometer onboard the spacecraft can monitor rapid transport processes of water vapor, dust, ice clouds, and other species, which could not be traced by the previous satellites on the sun-synchronous polar orbit. Such time-resolved pictures of the atmospheric phenomena should be an important clue to understand both the processes of water exchange between the surface/underground reservoirs and the atmosphere and the drivers of efficient material transport to the upper atmosphere. The mass spectrometer with unprecedented mass resolution can observe ions escaping to space and monitor the atmospheric escape which has made the past Mars to evolve towards the cold and dry surface environment we know today. Together with the above two instruments, it can potentially reveal what kinds of atmospheric events can transport tracers (e.g., H2O) upward and enhance the atmospheric escape. Graphical Abstract

A stellar occultation by Neptune's main satellite, Triton, was observed on 5 October 2017 from Europe, North Africa, and the USA. We derived 90 light curves from this event, 42 of which yielded a central flash detection. Aims. We aimed at constraining Triton's atmospheric structure and the seasonal variations of its atmospheric pressure since the Voyager 2 epoch (1989). We also derived the shape of the lower atmosphere from central flash analysis. Methods. We used Abel inversions and direct ray-tracing code to provide the density, pressure, and temperature profiles in the altitude range similar to 8 km to similar to 190 km, corresponding to pressure levels from 9 mu bar down to a few nanobars. Results. (i) A pressure of 1.18 +/- 0.03 mu bar is found at a reference radius of 1400 km (47 km altitude). (ii) A new analysis of the Voyager 2 radio science occultation shows that this is consistent with an extrapolation of pressure down to the surface pressure obtained in 1989. (iii) A survey of occultations obtained between 1989 and 2017 suggests that an enhancement in surface pressure as reported during the 1990s might be real, but debatable, due to very few high S/N light curves and data accessible for reanalysis. The volatile transport model analysed supports a moderate increase in surface pressure, with a maximum value around 2005-2015 no higher than 23 mu bar. The pressures observed in 1995-1997 and 2017 appear mutually inconsistent with the volatile transport model presented here. (iv) The central flash structure does not show evidence of an atmospheric distortion. Abstract: Contains fulltext : 218267.pdf (Publisher’s version ) (Closed access) PURPOSE: Intellectual disability (ID) and autism spectrum disorder (ASD) are genetically heterogeneous neurodevelopmental disorders. We sought to delineate the clinical, molecular, and neuroimaging spectrum of a novel neurodevelopmental disorder caused by variants in the zinc finger protein 292 gene (ZNF292). METHODS: We ascertained a cohort of 28 families with ID due to putatively pathogenic ZNF292 variants that were identified via targeted and exome sequencing. Available data were analyzed to characterize the canonical phenotype and examine genotype-phenotype relationships. RESULTS: Probands presented with ID as well as a spectrum of neurodevelopmental features including ASD, among others. All ZNF292 variants were de novo, except in one family with dominant inheritance. ZNF292 encodes a highly conserved zinc finger protein that acts as a transcription factor and is highly expressed in the developing human brain supporting its critical role in neurodevelopment. CONCLUSION: De novo and dominantly inherited variants in ZNF292 are associated with a range of neurodevelopmental features including ID and ASD. The clinical spectrum is broad, and most individuals present with mild to moderate ID with or without other syndromic features. Our results suggest that variants in ZNF292 are likely a recurrent cause of a neurodevelopmental disorder manifesting as ID with or without ASD.

Abstract: International audience; Virtual steering techniques enable users to navigate in larger Virtual Environments (VEs) than the physical workspace available. Even though these techniques do not require physical movement of the users (e.g. using a joystick and the head orientation to steer towards a virtual direction), recent work observed that users might unintentionally move in the physical workspace while navigating, resulting in Unintended Positional Drift (UPD). This phenomenon can be a safety issue since users may unintentionally reach the physical boundaries of the workspace while using a steering technique. In this context, as a necessary first step to improve the design of navigation techniques minimizing the UPD, this paper aims at analyzing and modeling the UPD during a virtual navigation task. In particular, we characterize and analyze the UPD for a dataset containing the positions and orientations of eighteen users performing a virtual slalom task using virtual steering techniques. Participants wore a head-mounted display and had to follow three different sinusoidal-like trajectories (with low, medium and high curvature) using a torso-steering navigation technique. We analyzed the performed motions and proposed two UPD models: the first based on a linear regression analysis and the second based on a Gaussian Mixture Model (GMM) analysis. Then, we assessed both models through a simulation-based evaluation where we reproduced the same navigation task using virtual agents. Our results indicate the feasibility of using simulation-based evaluations to study UPD. The paper concludes with a discussion of potential applications of the results in order to gain a better understanding of UPD during steering and therefore improve the design of navigation techniques by compensating for UPD.

Abstract: This work reports on the electron-induced modification of NaCl thin film grown on Ag(110). We show using low energy electron diffraction that electron beam bombardment leads to desorption and formation of Cl vacancy defects on NaCl surface. The topographic structure of these defects is studied using scanning tunneling microscopy (STM) showing the Cl defects as depressions on the NaCl surface. Most of the observed defects are mono-atomic vacancies and are located on flat NaCl terraces. Auger electron spectroscopy confirms the effect of electron exposure on NaCl thin films showing Cl atoms desorption from the surface. Using density functional theory taken into account the van der Waals dispersion interactions, we confirm the observed experimental STM measurements with STM simulation. Furthermore, comparing the adsorption of defect free NaCl and defective NaCl monolayer on Ag(110) surfaces, we found an increase of the adhesion energy and the charge transfer between the NaCl film and the substrate due to the Cl vacancy. In details, the adhesion energy increases between the NaCl film and the metallic Ag substrate from 30.4 meV Å−2 for the NaCl film without Cl vacancy and from 39.5 meV Å−2 for NaCl film with a single Cl vacancy. The charge transfer from the NaCl film to the Ag substrate is enhanced when the vacancy is created, from 0.63e− to 1.25e−.

Abstract: During the DARPA SCENICC program, J. Ford, et al., demonstrated that CFBs provide a compelling route to compact, wide angle imagers. Monocentric lenses readily provide diffraction-limited images over wide field but onto a hemispherically curved image surface. They demonstrated visible CFBs can be tapered, cut and polished to relay curved images to flat sensors. We have shown that this provides a volumetric imaging efficiency a hundredfold larger than bulk optics can produce; a hundred times the resolution in the same volume or a hundred times less volume for the same resolution. Ford's work leveraged commercial fiber bundles available for the visible spectrum based on silica. We have developed hybrid fiber bundles using step-index confinement between chalcogenide glass cores and polymer cladding with high index-contrast. The high contrast is necessary to provide tight confinement to the high-index As-Se core with minimal crosstalk between closely spaced cores. Tight confinement also minimizes absorption losses in the PEI polymer cladding. The high contrast of this system also provides a large NA to optimize coupling into the CFB from fast lenses. We introduce disorder into the core radius as a mechanism to further decouple adjacent cores, reduce crosstalk and increase fill-factor. We present coupled-mode theory, modal crosstalk superposition, and finite-element modelling to quantify coupling losses and crosstalk as a function of geometry and disorder. We fabricated preforms, drew small fiber bundles and characterized optical properties of the bundles to aid scale-up to megapixel MWIR CFBs.; Au cours du programme DARPA SCENICC, J. Ford et al. ont démontré que les faisceaux de fibres cohérents (FFC) offrent une voie convaincante aux imageurs compacts à grand angle. Les lentilles monocentriques fournissent facilement des images à diffraction limitée sur un grand champ mais sur une surface d'image incurvée de manière hémisphérique. Ils ont démontré que les FFCs visibles peuvent être effilés, coupés et polis pour transmettre des images incurvées à des capteurs plats. Nous avons montré que cela fournit une efficacité d'imagerie volumétrique cent fois supérieure à ce que l'optique en vrac peut produire ; cent fois la résolution dans le même volume ou cent fois moins de volume pour la même résolution. Les travaux de Ford ont tiré parti des faisceaux de fibres commerciaux disponibles pour le spectre visible à base de silice. Nous avons développé des faisceaux de fibres hybrides utilisant un confinement à saut d'indice entre des noyaux de verre chalcogénure et une gaine polymère à fort contraste d'indice. Le contraste élevé est nécessaire pour fournir un confinement serré au cœur As-Se à indice élevé avec une diaphonie minimale entre des cœurs rapprochés. Un confinement étanche minimise également les pertes par absorption dans le revêtement en polymère PEI. Le contraste élevé de ce système fournit également un grand NA pour optimiser le couplage dans le FFC à partir des objectifs rapides. Nous introduisons du désordre dans le rayon du noyau comme mécanisme pour découpler davantage les noyaux adjacents, réduire la diaphonie et augmenter le facteur de remplissage. Nous présentons la théorie des modes couplés, la superposition de diaphonie modale et la modélisation par éléments finis pour quantifier les pertes de couplage et la diaphonie en fonction de la géométrie et du désordre. Nous avons fabriqué des préformes, dessiné de petits faisceaux de fibres et caractérisé les propriétés optiques des faisceaux pour faciliter la mise à l'échelle des FFCs mégapixels MWIR.

Abstract: We review the study of dissociative recombination and ro-vibrational excitation of diatomic and small polyatomic molecular ions initiating complex organic molecules formation. In particular, we show how Multichannel Quantum Defect Theory (MQDT) and R-matrix methods are used to compute cross sections and rate coefficients for cations in well defined ro-vibrational levels of the ground electronic state, from sub-meV up to few eV collision energies. Abstract: Owing to the existence of periodic channels in phosphorene, this 2D material can be a good candidate for room temperature reversible hydrogen storage. The density functional theory calculations (DFT), including van der Waals interactions (vdW-DF2) coupled with the cooper exchange functional (C09), has been applied to study the potential of phosphorene as a new 2D material for hydrogen storage. Our results show that the adsorption energy (−292 to −277 meV) of H2 on phosphorene is appropriate for storage. The analysis of diffusion pathways between different physisorbed states on phosphorene shows that a single hydrogen molecule diffuses very easily along the open channel (less than 1 meV along the zigzag direction), as compared to 14 meV for diffusion across the channels (along the armchair direction). The potential energy surfaces for the dissociative chemisorption of H2 was computed on highly symmetric sites of phosphorene and the highest activation barrier was found to be 2.77 eV. The very large dissociation energy coupled with a strong physisorption of H2 on phosphorene and facile diffusion, makes this 2D material a promising candidate for H2 storage at room temperature.

Abstract: International audience; Aza-boron dipyrromethenes (aza-BODIPYs) presenting a benzothiadiazole substitution on upper positions are described. The strong electron-withdrawing effect of the benzothiadiazole moiety permits enhancement of the accepting strength and improves the delocalization of the aza-BODIPY core to attain a significant degree of electronic communication between the lower donating groups and the upper accepting groups. The nature of the intramolecular charge transfer is studied both experimentally and theoretically. Linear spectroscopy highlighted the strongly redshifted absorption and emission of the synthesized molecules with recorded fluorescence spectra over 1000 nm. Nonlinear optical properties were also investigated. Strong enhancement of the two-photon absorption of the substituted dyes compared with the unsubstituted one (up to 4520 GM at 1300 nm) results in an approximately 15-20 % improvement of the optical power limiting performances. These dyes are therefore a good starting point for further improvement of optical power limiting in the short-wave IR range.

Abstract: Optical wave packets that are localized in space and time, but nevertheless overcome diffraction and travel rigidly in free space, are a long sought-after field structure with applications ranging from microscopy and remote sensing, to nonlinear and quantum optics. However, synthesizing such wave packets requires introducing non-differentiable angular dispersion with high spectral precision in two transverse dimensions, a capability that has eluded optics to date. Here, we describe an experimental strategy capable of sculpting the spatio-temporal spectrum of a generic pulsed beam by introducing arbitrary radial chirp via two-dimensional conformal coordinate transformations of the spectrally resolved field. This procedure yields propagation-invariant `space-time' wave packets localized in all dimensions, with tunable group velocity in the range from $0.7c$ to $1.8c$ in free space, and endowed with prescribed orbital angular momentum. Abstract: We investigate the effects of private electric vehicles (EVs) on the resilience of distribution systems after disruptions. We propose a framework of network-based multi-agent optimization problems with equilibrium constraints (N-MOPEC) to consider the decentralized decision making of stakeholders in transportation and energy systems. To solve the high-dimensional non-convex problem, we develop an efficient computational algorithm based on exact convex reformulation. Numerical studies are conducted to illustrate the effectiveness of our modeling and computational approach and to draw policy insights. Abstract: International audience; Permanent change of the refractive index of photo-thermo-refractive glass can be achieved by exposure to ultraviolet light followed by thermal development. The dependence of the change on the dosage is nonlinear because the photochemical process of photosensitivity involves saturation. With a holographic recording of Bragg gratings inside glass, a sinusoidally modulated dosage is imprinted into distorted modulation of the refractive index. We investigate this distorted modulation and derive analytical expressions for the amplitudes of its spatial harmonics.

Abstract: International audience; Sperm vitality testing is a basic semen examination that has been described in the World Health Organization (WHO) Laboratory Manual for the Examination and Processing of Human Semen from its primary edition, 40 years ago. Several methods can be used to test sperm vitality, such as the eosin-nigrosin (E-N) stain or the hypoosmotic swelling (HOS) test. In the 6th (2021) edition of the WHO Laboratory Manual, sperm vitality assessment is mainly recommended if the total motility is less than 40%. Hence, a motile spermatozoon is considered alive, however, in certain conditions an immotile spermatozoon can also be alive. Therefore, the differentiation between asthenozoospermia (pathological decrease in sperm motility) and necrozoospermia (pathological decrease in sperm vitality) is important in directing further investigation and management of infertile patients. The causes leading to necrozoospermia are diverse and can either be local or general, testicular or extra-testicular. The andrological management of necrozoospermia depends on its etiology. However, there is no standardized treatment available presently and practice varies among clinicians. In this study, we report the results of a global survey to understand current practices regarding the physician order of sperm vitality tests as well as the management practices for necrozoospermia. Laboratory and clinical scenarios are presented to guide the reader in the management of necrozoospermia with the overall objective of establishing a benchmark ranging from the diagnosis of necrozoospermia by sperm vitality testing to its clinical management.

Abstract: 11 pages, 7 figures, Marine Ecosystem Models (MEMs) provide a deeper understanding of marine ecosystem dynamics. The United Nations Decade of Ocean Science for Sustainable Development has highlighted the need to deploy these complex mechanistic spatial-temporal models to engage policy makers and society into dialogues towards sustainably managed oceans. From our shared perspective, MEMs remain underutilized because they still lack formal validation, calibration, and uncertainty quantifications that undermines their credibility and uptake in policy arenas. We explore why these shortcomings exist and how to enable the global modelling community to increase MEMs’ usefulness. We identify a clear gap between proposed solutions to assess model skills, uncertainty, and confidence and their actual systematic deployment. We attribute this gap to an underlying factor that the ecosystem modelling literature largely ignores: technical issues. We conclude by proposing a conceptual solution that is cost-effective, scalable and simple, because complex spatial-temporal marine ecosystem modelling is already complicated enough, MC and JS acknowledge funding from the EuroMarine 2018 call for Foresight Workshops and Working Groups proposals. Abstract: International audience; Optical composite fibers based on active transition metal (TM)-doped semiconductor crystals are being investigated for use in mid-infrared (mid-IR) fiber laser systems. This study evaluates a candidate glass matrix system assessing its key physical properties and suitability for optical fiber drawing and examines the stability of TM-doped ZnSe crystals during processing. Results indicate that despite excellent refractive index matching between crystal and glass matrix and good fiber draw attributes, the stability of the crystalline dopant within the glass melt is severely impacted by melt conditions. The time and temperature dependence of this stability is mapped by X-ray diffraction (XRD) and second-harmonic generation (SHG) microscopy, illustrating how these tools can be used to track the phase stability and robustness throughout the melting process. Results show that in a range of sulfo-selenide based matrices, dissolution and re-precipitation of Zn-containing crystalline phases is a dominant mechanism.

Abstract: International audience; As pressing needs for exploring molecular interactions in plants soar, conventional sample preparation methods come into question. Though resins used to embed plant tissues have long been assumed to bear no palpable effect on their properties, discrepancies in recent studies exploiting nanoscale microscopy suggest that their impact could be significant at small scales. By juxtaposing the traits of poplar sections prepared with and without embedding, we evaluate the diffusion (penetration depth) of acrylic and epoxy resins commonly used for embedding. Our results unveil critical quantitative differences when probing mechanical properties with a microscale nanoindentation indenter or a nanoscale tip. The latter resolves significant stiffness variations between the compound middle lamellae, the secondary cell wall layers S1 and S2, and the cell corner, not accessible with nanoindentation. Similar observations are drawn from comparing confocal Raman and nanoscale infrared spectroscopy. Our findings shed light on the effect of resin diffusion suggesting acrylic LR White to be the least diffusive for plant cell wall studies.

Abstract: International audience; The Food-Energy-Water (FEW) nexus for urban sustainability needs to be analyzed via an integrative rather than a sectoral or silo approach, reflecting the ongoing transition from separate infrastructure systems to an integrated social-ecological-infrastructure system. As technology hubs can provide food, energy, water resources via decentralized and/or centralized facilities, there is an acute need to optimize FEW infrastructures by considering cost-benefit-risk tradeoffs with respect to multiple sustainability indicators. This paper identifies, categorizes, and analyzes global trends with respect to contemporary FEW technology metrics that highlights the possible optimal integration of a broad spectrum of technology hubs for possible cost-benefit-risk tradeoffs. The challenges related to multiscale and multiagent modeling processes for the simulation of urban FEW systems were discussed with respect to the aspects of scaling-up, optimization process, and risk assessment. Our review reveals that this field is growing at a rapid pace and the previous selection of analytical methodologies, nexus criteria, and sustainability indicators largely depended on individual FEW nexus conditions disparately, and full-scale cost-benefit-risk tradeoffs were very rare. Therefore, the potential full-scale technology integration in three ongoing cases of urban FEW systems in Miami (the United States), Marseille (France), and Amsterdam (the Netherlands) were demonstrated in due purpose finally.

Abstract: International audience; The current WHO 2010 manual for human semen analysis defines leukocytospermia as the presence of peroxidase-positive leukocytes at a concentration >1×106/mL of semen. Granular leukocytes when activated are capable of generating high levels of reactive oxygen species in semen resulting in oxidative stress. Oxidative stress has been correlated with poor sperm quality, increased level of sperm DNA fragmentation and low fertility potential. The presence of leukocytes and pathogens in the semen may be a sign of infection and/or localized inflammatory response in the male genital tract and the accessory glands. Common uro-pathogens including Chlamydia trachomatis, Ureaplasma urealyticum, Neisseria gonorrhoeae, Mycoplasma hominis, and Escherichia coli can cause epididymitis, epididymo-orchitis, or prostatitis. The relationship between leukocytospermia and infection is unclear. Therefore, we describe the pathogens responsible for male genital tract infections and their association with leukocytospermia. The review also examines the diagnostic tests available to identify seminal leukocytes. The role of leukocytospermia in male infertility and its management is also discussed.

Abstract: International audience; Heterojunctions are used in solar cells and optoelectronics applications owing to their excellent electrical and structural properties. Recently, these energy-efficient systems have also been employed as sensors to distinguish between individual gases within mixtures. Through a simple and versatile functionalization approach using noble metal nanoparticles, the sensing properties of heterojunctions can be controlled at the nanoscopic scale. This work reports the nanoparticle surface functionalization of TiO2/CuO/Cu2O mixed oxide heterostructures, where the gas sensing selectivity of the material is tuned to achieve versatile sensors with ultra-low power consumption. Functionalization with Ag or AgPt-nanoclusters (5–15 nm diameter), changed the selectivity from ethanol to butanol vapour, whereas Pd-nanocluster functionalization shifts the selectivity from the alcohols to hydrogen. The fabricated sensors show excellent low power consumption below 1 nW. To gain insight into the selectivity mechanism, density functional theory (DFT) calculations have been carried out to simulate the adsorption of H2, C2H5OH and n-C4H9OH at the noble metal nanoparticle decorated ternary heterostructure interface. These calculations also show a decrease in the work function by ~2.6 eV with respect to the pristine ternary heterojunctions. This work lays the foundation for the production of a highly versatile array of sensors of ultra-low power consumption with applications for the detection of individual gases in a mixture.

Abstract: Purpose: Myofiber strain, Eff , is a mechanistically relevant metric of cardiac cell shortening and is expected to be spatially uniform in healthy populations, making it a prime candidate for the evaluation of local cardiomyocyte contractility. In this study, a new, efficient pipeline was proposed to combine microstructural cDTI and functional DENSE data in order to estimate Eff in vivo.Methods: Thirty healthy volunteers were scanned with three long-axis (LA) and three short-axis (SA) DENSE slices using 2D displacement encoding and one SA slice of cDTI. The total acquisition time was 11 minutes ± 3 minutes across volunteers. The pipeline first generates 3D SA displacements from all DENSE slices which are then combined with cDTI data to generate a cine of myofiber orientations and compute Eff . The precision of the post-processing pipeline was assessed using a computational phantom study. Transmural myofiber strain was compared to circumferential strain, Ecc , in healthy volunteers using a Wilcoxon sign rank test.Results: In vivo, computed Eff was found uniform transmurally compared to Ecc (-0.14[-0.15, -0.12] vs -0.18 [-0.20, -0.16], P < .001, -0.14 [-0.16, -0.12] vs -0.16 [-0.17, -0.13], P < .001 and -0.14 [-0.16, -0.12] vs Ecc_C = -0.14 [-0.15, -0.11], P = .002, Eff_C vs Ecc_C in the endo, mid, and epi layers, respectively).Conclusion: We demonstrate that it is possible to measure in vivo myofiber strain in a healthy human population in 10 minutes per subject. Myofiber strain was observed to be spatially uniform in healthy volunteers making it a potential biomarker for the evaluation of local cardiomyocyte contractility in assessing cardiovascular dysfunction.

Abstract: International audience; Tipping point dynamics are fundamental drivers for sustainable transition pathways of social-ecological systems (SES). Current research predominantly analyzes how crossing tipping points causes regime shifts, however, the analysis of potential transition pathways from these social and ecological tipping points is often overlooked. In this paper, we analyze transition pathways and the potential outcomes that these may lead to via a stylized model of a system composed of interacting agents exploiting resources and, by extension, the overall ecosystem. Interactions between the social and the ecological system are based on a perception-exploitation framework. We show that the presence of tipping points in SES may yield counter-intuitive social-ecological transition pathways. For example, the high perception of an alarming ecological state among agents can provide short-term ecological benefits, but can be less effective in the long term, compared to a low-perception condition. This work also highlights how understanding non-linear interactions is critical for defining suitable transition pathways of any SES.

Abstract: Collected rainfall records by gauges lead to key forcings in most hydrological studies. Depending on sensor type and recording systems, such data are characterized by different time-resolutions (or temporal aggregations), ta. We present an historical analysis of the time-evolution of ta based on a large database of rain gauge networks operative in many study areas. Globally, ta data were collected for 25,423 rain gauge stations across 32 geographic areas, with larger contributions from Australia, USA, Italy and Spain. For very old networks early recordings were manual with coarse time-resolution, typically daily or sometimes monthly. With a few exceptions, mechanical recordings on paper rolls began in the first half of the 20th century, typically with ta of 1 h or 30 min. Digital registrations started only during the last three decades of the 20th century. This short period limits investigations that require long time-series of sub-daily rainfall data, e.g, analyses of the effects of climate change on short-duration (sub-hourly) heavy rainfall. In addition, in the areas with rainfall data characterized for many years by coarse time-resolutions, annual maximum rainfall depths of short duration can be potentially underestimated and their use would produce errors in the results of successive applications. Currently, only 50% of the stations provide useful data at any time-resolution, that practically means ta = 1 min. However, a significant reduction of these issues can be obtained through the information content of the present database. Finally, we suggest an integration of the database by including additional rain gauge networks to enhance its usefulness particularly in a comparative analysis of the effects of climate change on extreme rainfalls of short duration available in different locations.

Abstract: The emergence of peculiar phenomena in 1D phosphorene chains (P chains) has been proposed in theoretical studies, notably the Stark and Seebeck effects, room temperature magnetism, and topological phase transitions. Attempts so far to fabricate P chains, using the top-down approach starting from a few layers of bulk black phosphorus, have failed to produce reliably precise control of P chains. We show that molecular beam epitaxy gives a controllable bottom-up approach to grow atomically thin, crystalline 1D flat P chains on a Ag(111) substrate. Scanning tunneling microscopy, angle-resolved photoemission spectroscopy, and density functional theory calculations reveal that the armchair-shaped chains are semiconducting with an intrinsic 1.80 ± 0.20 eV band gap. This could make these P chains an ideal material for opto-electronic devices., Similarly to graphene, attempts to fabricate phosphorene by epitaxy or starting from a few layers of bulk black phosphorus have failed so far. Here, the authors present a controllable bottom-up approach to grow atomically thin, crystalline 1D flat phosphorus chains on a Ag(111) substrate.

Wir betrachten hier solche Strahlen, die durch hochharmonische Erzeugung (HHG) in einem seltenen Gas erzeugt werden, und analysieren die Charakterisierung ihrer Hochladungs-Wirbelstruktur durch einen EUV (Extreme UltraViolet) Hartmann-Wellenfrontsensor. Wir zeigen, dass solche HHG-Strahlen in der Regel aus einer Reihe von zahlreichen Vortex-Modi bestehen. Die Empfindlichkeit der Intensität und Phase des HHG-Strahls auf die infraroten (IR) Laseraberrationen wird mit einem deformierbaren Spiegel untersucht.; Optical beams carrying Orbital Angular Momentum (OAM) are a very active field of research for their prospective applications, especially at short wavelengths. We consider here such beams produced through high-harmonic generation (HHG) in a rare gas and analyze the characterization of their high charge vortex structure by an EUV (Extreme UltraViolet) Hartmann wavefront sensor. We show that such HHG beams are generally composed of a set of numerous vortex modes. The sensitivity of the intensity and phase of the HHG beam to the infrared (IR) laser aberrations is investigated using a deformable mirror.; Les faisceaux optiques portant un Moment Angulaire Orbital (OAM) font l'objet de recherches très actives,en raison de l'intérêt de leurs applications potentielles, particulièrement à courtes longueurs d'onde. Nous nous intéressons ici à des faisceaux produits par Génération d'Harmoniques d'Ordre Elevé (HOE) dans un gaz rare, et analysons la structure du vortex de haute charge grâce à la caractérisation complète obtenue par un capteur de front d'onde de type Hartmann dans l'Extrême UltraViolet (EUV). Nous montrons que ces faisceaux HOE sont généralement constitués d'un ensemble de nombreux modes de vortex. Un miroir déformable nous permet d'étudier la sensibilité de l'éclairement et de la phase du faisceau HOE vis à vis des aberrations du laser infrarouge (IR).

Abstract: Key Points Question What are the long-term outcomes associated with dysbiosis of gut microbiota in very preterm newborns? Findings In this cohort study of 577 very preterm newborns across 24 neonatal intensive care units from a French nationwide cohort, gut microbiota at week 4 after birth showed 6 bacterial patterns that varied according to gestational age, perinatal characteristics, individual treatments, and neonatal intensive care unit strategies. Three clusters were associated with 2-year outcomes after adjustment for these confounders. Meaning Modifying strategies associated with alterations in microbiota, such as promoting enteral nutrition, reducing sedation use, promoting early extubation, or skin-to-skin practice, may be correlated with outcomes in preterm newborns., Importance In very preterm newborns, gut microbiota is highly variable with major dysbiosis. Its association with short-term health is widely studied, but the association with long-term outcomes remains unknown. Objective To investigate in preterm newborns the associations among practice strategies in neonatal intensive care units (NICUs), gut microbiota, and outcomes at 2 years. Design, Setting, and Participants EPIFLORE is a prospective observational cohort study that includes a stool sample collection during the fourth week after birth. Preterm newborns of less than 32 weeks of gestational age (GA) born in 2011 were included from 24 NICUs as part of the French nationwide population-based cohort, EPIPAGE 2. Data were collected from May 2011 to December 2011 and analyzed from September 2016 to December 2018. Exposures Eight NICU strategies concerning sedation, ventilation, skin-to-skin practice, antibiotherapy, ductus arteriosus, and breastfeeding were assessed. A NICU was considered favorable to a practice if the percentage of that practice in the NICU was more than the expected percentage. Main Outcomes and Measures Gut microbiota was analyzed by 16S ribosomal RNA gene sequencing and characterized by a clustering-based method. The 2-year outcome was defined by death or neurodevelopmental delay using a Global Ages and Stages questionnaire score. Results Of 577 newborns included in the study, the mean (SD) GA was 28.3 (2.0) weeks, and 303 (52.5%) were male. Collected gut microbiota was grouped into 5 discrete clusters. A sixth cluster included nonamplifiable samples owing to low bacterial load. Cluster 4 (driven by Enterococcus [n = 63]), cluster 5 (driven by Staphylococcus [n = 52]), and cluster 6 (n = 93) were significantly associated with lower mean (SD) GA (26.7 [1.8] weeks and 26.8 [1.9] weeks, respectively) and cluster 3 (driven by Escherichia/Shigella [n = 61]) with higher mean (SD) GA (29.4 [1.6] weeks; P = .001). Cluster 3 was considered the reference. After adjustment for confounders, no assisted ventilation at day 1 was associated with a decreased risk of belonging to cluster 5 or cluster 6 (adjusted odds ratio [AOR], 0.21 [95% CI, 0.06-0.78] and 0.19 [95% CI, 0.06-0.62], respectively) when sedation (AOR, 10.55 [95% CI, 2.28-48.87] and 4.62 [1.32-16.18], respectively) and low volume of enteral nutrition (AOR, 10.48 [95% CI, 2.48-44.29] and 7.28 [95% CI, 2.03-26.18], respectively) was associated with an increased risk. Skin-to-skin practice was associated with a decreased risk of being in cluster 5 (AOR, 0.14 [95% CI, 0.04-0.48]). Moreover, clusters 4, 5, 6 were significantly associated with 2-year nonoptimal outcome (AOR, 6.17 [95% CI, 1.46-26.0]; AOR, 4.53 [95% CI, 1.02-20.1]; and AOR, 5.42 [95% CI, 1.36-21.6], respectively). Conclusions and Relevance Gut microbiota of very preterm newborns at week 4 is associated with NICU practices and 2-year outcomes. Microbiota could be a noninvasive biomarker of immaturity., This cohort study of 577 very preterm newborns across 24 neonatal intensive care units from a French nationwide cohort investigates associations among practice strategies, gut microbiota, and outcomes at 2 years.

Wir betrachten hier solche Strahlen, die durch hochharmonische Erzeugung (HHG) in einem seltenen Gas erzeugt werden, und analysieren die Charakterisierung ihrer Hochladungs-Wirbelstruktur durch einen EUV (Extreme UltraViolet) Hartmann-Wellenfrontsensor. Wir zeigen, dass solche HHG-Strahlen in der Regel aus einer Reihe von zahlreichen Vortex-Modi bestehen. Die Empfindlichkeit der Intensität und Phase des HHG-Strahls auf die infraroten (IR) Laseraberrationen wird mit einem deformierbaren Spiegel untersucht.; Optical beams carrying Orbital Angular Momentum (OAM) are a very active field of research for their prospective applications, especially at short wavelengths. We consider here such beams produced through high-harmonic generation (HHG) in a rare gas and analyze the characterization of their high charge vortex structure by an EUV (Extreme UltraViolet) Hartmann wavefront sensor. We show that such HHG beams are generally composed of a set of numerous vortex modes. The sensitivity of the intensity and phase of the HHG beam to the infrared (IR) laser aberrations is investigated using a deformable mirror.; Les faisceaux optiques portant un Moment Angulaire Orbital (OAM) font l'objet de recherches très actives,en raison de l'intérêt de leurs applications potentielles, particulièrement à courtes longueurs d'onde. Nous nous intéressons ici à des faisceaux produits par Génération d'Harmoniques d'Ordre Elevé (HOE) dans un gaz rare, et analysons la structure du vortex de haute charge grâce à la caractérisation complète obtenue par un capteur de front d'onde de type Hartmann dans l'Extrême UltraViolet (EUV). Nous montrons que ces faisceaux HOE sont généralement constitués d'un ensemble de nombreux modes de vortex. Un miroir déformable nous permet d'étudier la sensibilité de l'éclairement et de la phase du faisceau HOE vis à vis des aberrations du laser infrarouge (IR).

Abstract: Six niobium rich glasses presenting different sodium contents were synthetized. On the basis of complementary infrared and Raman analysis, the influence of the sodium content and its role on the structure of the glasses prior to and following poling was examined. Correlative second harmonic generation (SHG)/Raman microscopy on the poled glasses cross section has shown a co-localization of the SHG signal and the structural variations. It also evidenced similar structural rearrangements whether sodium is removed through poling (230–300 °C) or through the alkali content defined by the starting glass compositions (melted at 1300–1500 °C). The effect of the sodium content on the optical properties, prior to and after thermal poling, is also discussed. It was found that refractive index variations induced by poling (ranging between 10−3 and 3 × 10−2) are mainly the result of a density decrease in the poled region rather than compositional and structural changes. The electro-optic origin of the poling-induced second order nonlinear response is confirmed by the Maker Fringe SHG analysis, and the evolution of χ(2) (2–2.5 pm/V) with the sodium content is discussed based on a selection of parameters influencing either the third order susceptibility [χ(3)] or the internal electric field strength of the poled layer.

Abstract: Knowledge of highly excited rovibrational states of ozone isotopologues is of key importance for modelling the dynamics of exchange reactions, for understanding longstanding problems related to isotopic anomalies of the ozone formation, and for analyses of extra-sensitive laser spectral experiments currently in progress. This work is devoted to new theoretical study of high-energy states for the main isotopologue 48O3 = 16O16O16O and for the family of 18O-enriched isotopomers 50O3 = {16O16O18O, 16O18O16O, 18O16O16O} of the ozone molecule considered using a full-symmetry approach. Energies and wave functions of bound states near the dissociation threshold are computed in hyperspherical coordinates accounting for the permutation symmetry of three identical nuclei in 48O3 and of two identical nuclei in 50O3, using the most accurate potential energy surface available now. The obtained vibrational band centers agree with observed ones with the root-mean-squares deviation of about 1 cm-1, making the results appropriate for assignments and analyses of future experimental spectra. The levels delocalized between the three potential wells of ozone isomers are computed and analyzed. The states situated deep in the three (for 48O3) or two (for 50O3) equivalent potential wells have similar energies with negligible splitting. However, the states situated just below the potential barriers separating the wells, are split due to the tunneling between the wells resulting in the splitting of rovibrational sub-bands. We evaluate the amplitudes of the corresponding effects and consider possible perturbations in vibration-rotation bands due to interactions between three potential wells. Theoretical predictions for the splitting of observable band centers are provided for the first time.

Abstract: Significance Bacteria are known to add amino acids (aa) to membrane lipids to resist antimicrobials and escape immune responses. This surface lipid aminoacylation process requires diverting aminoacyl-tRNAs from protein synthesis. While widespread in bacteria, no analogous lipid remodeling system had thus far been evidenced in eukaryotes. We uncovered that most fungi tRNA-dependently add aspartate onto ergosterol (ergosteryl-3β-O-l-aspartate [Erg-Asp]), the major sterol found in fungal membranes. Asp addition is catalyzed by an ergosteryl-3β-O-l-aspartate synthase (ErdS) and its removal by a dedicated hydrolase (ErdH). This pathway is conserved across “higher” fungi, including pathogens. Given the central roles of sterols and derivatives in fungi, we propose that the Erg-Asp homeostasis system might impact membrane remodeling, trafficking, antimicrobial resistance, or pathogenicity., Diverting aminoacyl-transfer RNAs (tRNAs) from protein synthesis is a well-known process used by a wide range of bacteria to aminoacylate membrane constituents. By tRNA-dependently adding amino acids to glycerolipids, bacteria change their cell surface properties, which intensifies antimicrobial drug resistance, pathogenicity, and virulence. No equivalent aminoacylated lipids have been uncovered in any eukaryotic species thus far, suggesting that tRNA-dependent lipid remodeling is a process restricted to prokaryotes. We report here the discovery of ergosteryl-3β-O-l-aspartate (Erg-Asp), a conjugated sterol that is produced by the tRNA-dependent addition of aspartate to the 3β-OH group of ergosterol, the major sterol found in fungal membranes. In fact, Erg-Asp exists in the majority of “higher” fungi, including species of biotechnological interest, and, more importantly, in human pathogens like Aspergillus fumigatus. We show that a bifunctional enzyme, ergosteryl-3β-O-l-aspartate synthase (ErdS), is responsible for Erg-Asp synthesis. ErdS corresponds to a unique fusion of an aspartyl-tRNA synthetase—that produces aspartyl-tRNAAsp (Asp-tRNAAsp)—and of a Domain of Unknown Function 2156, which actually transfers aspartate from Asp-tRNAAsp onto ergosterol. We also uncovered that removal of the Asp modifier from Erg-Asp is catalyzed by a second enzyme, ErdH, that is a genuine Erg-Asp hydrolase participating in the turnover of the conjugated sterol in vivo. Phylogenomics highlights that the entire Erg-Asp synthesis/degradation pathway is conserved across “higher” fungi. Given the central roles of sterols and conjugated sterols in fungi, we propose that this tRNA-dependent ergosterol modification and homeostasis system might have broader implications in membrane remodeling, trafficking, antimicrobial resistance, or pathogenicity.
