Abstract

International audience; Background: The objective of this study was to demonstrate that synchrotron K-edge subtraction tomography (SKES-CT) can simultaneously track therapeutic cells and their encapsulating carrier, in vivo, in a rat model of focal brain injury using a dual-contrast agent approach. The second objective was to determine if SKES-CT could be used as a reference method for spectral photon counting tomography (SPCCT). Methods: Phantoms containing different concentrations of gold and iodine nanoparticles (AuNPS/INPs) were imaged with SKES-CT and SPCCT to assess their performances. A pre-clinical study was performed in rats with focal cerebral injury which intracerebrally received AuNPs-labelled therapeutic cells encapsulated in a INPs-labelled scaffold. Animals were imaged in vivo with SKES-CT and back-to-back with SPCCT. Results: SKES-CT revealed to be reliable for quantification of gold and iodine, whether alone or mixed. In the preclinical model, SKES-CT showed that AuNPs remained at the site of cell injection, while INPs expanded within and/or along the lesion border, suggesting dissociation of both components in the first days post-administration. Compared to SKES-CT, SPCCT was able to correctly locate gold, but not completely located iodine. When SKES-CT was used as reference, SPCCT gold quantification appeared very accurate both in vitro and in vivo. Iodine quantification by SPCCT was also quite accurate, albeit less so than for gold. Conclusion: We here provide the proof-of-concept that SKES-CT is a novel method of choice for performing dual-contrast agent imaging in the context of brain regenerative therapy. SKES-CT may also serve as ground truth for emerging technologies such as multicolour clinical SPCCT.

Abstract

The term extracellular vesicles (EVs) has been recommended to describe various membrane-bound vesicles secreted by most living cells and found in various biological fluids. They gained growing interest as mediators of cell-cell communication and for their roles in different patho-physiological processes. In addition, they were recently considered as disease biomarkers and new drug delivery systems. However, it is still difficult to link a biological function to a specific EV population among the heterogenous EV mixture secreted in the extracellular space due to limitations of optimal isolation methods. EV classification according to their size as small (\textless200 nm) and large (\textgreater200 nm) vesicles is also completed by the identification of selected proteins, nucleic acids and lipids. In this review, we summarized briefly knowledge about the composition and role of EV lipids that received less attention compared to their protein and nucleic acid content. Lipids are not only essential structural components of EVs, but can give important information on their biogenesis. Especially, we discussed our recent data showing the utility of bis(monoacylglycero)phosphate (BMP), a specific endolysosomal lipid marker, that could sign the endosomal origin of small EVs, classically named as exosomes.

Abstract

International audience; Oral administration of nanoparticles (NPs) is a promising strategy to overcome solubility and stability issues of many active compounds. However, this route faces major obstacles related to the hostile gastrointestinal (GI) environment, which impairs the efficacy of orally administered nanomedicines. Here, we propose nanocomposites as a promising approach to increase the retention time of NPs in the intestinal tract by using bio-and mucoadhesive matrixes able to protect the cargo until it reaches the targeted area. A microfluidic-based approach has been applied for the production of tailored nanoemulsions (NEs) of about 110 nm, used for the encapsulation of small hydrophobic drugs such as the antiinflammatory JAK-inhibitor tofacitinib. These NEs proved to be efficiently internalized into a mucus-secreting human intestinal monolayer of Caco-2/ HT29-MTX cells and to deliver tofacitinib to subepithelial human THP-1 macrophage-like cells, reducing their inflammatory response. NEs were then successfully encapsulated into alginate hydrogel microbeads of around 300 μm, which were characterized by rheological experiments and dried to create a long-term stable system for pharmaceutical applications. Finally, ex vivo experiments on excised segments of rats′ intestine proved the bioadhesive ability of NEs embedded in alginate hydrogels compared to free NEs, showing the advantage that this hybrid system can offer for the treatment of intestinal pathologies.

Abstract

Biofilm formation on abiotic surfaces has become a major public health concern because of the serious problems they can cause in various fields. Biofilm cells are extremely resistant to stressful conditions, because of their complex structure impedes antimicrobial penetration to deep-seated cells. The increased resistance of biofilm to currently applied control strategies underscores the urgent need for new alternative and/or supplemental eradication approaches. The combination of two or more methods, known as Hurdle technology, offers an excellent option for the highly effective control of biofilms. In this perspective, the use of functional enzymes combined with biosourced antimicrobial such as essential oil (EO) is a promising alternative anti-biofilm approach. However, these natural antibiofilm agents can be damaged by severe environmental conditions and lose their activity. The microencapsulation of enzymes and EOs is a promising new technology for enhancing their stability and improving their biological activity. This review article highlights the problems related to biofilm in various fields, and the use of encapsulated enzymes with essential oils as antibiofilm agents. KEY POINTS: • Problems associated with biofilms in the food and medical sectors and their subsequent risks on health and food quality. • Hurdle technology using enzymes and essential oils is a promising strategy for an efficient biofilms control. • The microencapsulation of enzymes and essential oils ensures their stability and improves their biological activities.

Abstract

International audience; Targeted radionuclide therapy is a revolutionary tool for the treatment of highly spread metastatic cancers. Most current approaches rely on the use of vectors to deliver radionuclides to tumor cells, targeting membrane-bound cancer-specific moieties. Here, we report the embryonic navigation cue netrin-1 as an unanticipated target for vectorized radiotherapy. While netrin-1, known to be reexpressed in tumoral cells to promote cancer progression, is usually characterized as a diffusible ligand, we demonstrate here that netrin-1 is actually poorly diffusible and bound to the extracellular matrix. A therapeutic anti-netrin-1 monoclonal antibody (NP137) has been preclinically developed and was tested in various clinical trials showing an excellent safety profile. In order to provide a companion test detecting netrin-1 in solid tumors and allowing the selection of therapy-eligible patients, we used the clinical-grade NP137 agent and developed an indium-111-NODAGA-NP137 single photon emission computed tomography (SPECT) contrast agent. NP137-111 In provided specific detection of netrin-1-positive tumors with an excellent signal-to-noise ratio using SPECT/CT imaging in different mouse models. The high specificity and strong affinity of NP137 paved the way for the generation of lutetium-177-DOTA-NP137, a novel vectorized radiotherapy, which specifically accumulated in netrin-1-positive tumors. We demonstrate here, using tumor cell-engrafted mouse models and a genetically engineered mouse model, that a single systemic injection of NP137-177 Lu provides important antitumor effects and prolonged mouse survival. Together, these data support the view that NP137-111 In and NP137-177 Lu may represent original and unexplored imaging and therapeutic tools against advanced solid cancers.

Abstract

Paracetamol (PCT) and chloramphenicol (CPL) can have unfavorable impacts on human health, as well as on natural ecosystems. These substances contribute to the aquatic environment’s contamination and disturb the performance of municipal wastewater treatment systems, causing ecosystem disruption and microbial resistance. In this study, activated carbon produced from sawdust (ACs) was synthesized utilizing the chemical activation process for the removal of both PCT and CPL compounds from an aqueous solution. ACs has a primarily microporous structure with a significant specific surface area of 303–1298 m2/g, total pore volume of 0.462 cm3/g and bimodal distribution of pores of 0.73–1.7 nm. The removal efficiencies for PCT and CPL with the low-cost activated carbon, determined at the optimum dose (750 mg/L for PCT and 450 mg/L for CPL), were significantly high at 85% and 98%, respectively. The adsorption kinetics for both pharmaceuticals exhibited a quick initial decline. For PCT and CPL adsorption, the equilibrium was attained after just 20 and 90 min, respectively. The Langmuir isotherm model and the pseudo-second-order kinetics model offered the best fits for the adsorption of both compounds. Additionally, the central composite design (CCD) and Box–Behnken design (BBD) were used to optimize the experimental adsorption conditions using a response surface methodology (RSM). On the basis of the findings, it is evident that activated carbon made from sawdust may be used as a new, effective alternative adsorbent for removing PCT and CPL in aqueous environments.

Abstract

Leishmania parasites are the causative agents of visceral or cutaneous leishmaniasis in humans and of canine leishmaniosis. The macrophage is the predilected host cell of Leishmania in which the promastigote stage is transformed into amastigote. We previously showed changes in the fatty acid composition (FA) of lipids in two strains of Leishmania donovani upon differentiation of promastigote to amastigote, including increased proportions of arachidonic acid (AA) and to a less extent of docosahexaenoic acid (DHA). Here, we carried out supplementation with AA or DHA on two Leishmania infantum strains, a visceral (MON-1) and a cutaneous (MON-24), to evaluate the role of these FA in parasite/macrophage interactions. The proportions of AA or DHA in total lipids were significantly increased in promastigotes cultured in AA- or DHA-supplemented media compared to controls. The content of FA-derived oxygenated metabolites was enhanced in supplemented strains, generating especially epoxyeicosatrienoic acids (11,12- and 14,15-EET) and hydroxyeicosatetraenoic acids (5- and 8- HETE) from AA, and hydroxydocosahexaenoic acids (14- and 17-HDoHE) from DHA. For both MON-1 and MON-24, AA-supplemented promastigotes showed higher infectivity towards J774 macrophages as evidenced by higher intracellular amastigote numbers. Higher infectivity was observed after DHA supplementation for MON-24 but not MON-1 strain. ROS production by macrophages increased upon parasite infection, but only minor change was observed between control and supplemented parasites. We propose that under high AA or DHA environment that is associated with AA or DHA enrichment of promastigote lipids, FA derivatives can accumulate in the parasite, thereby modulating parasite infectivity towards host macrophages.

Abstract

International audience; This article focuses on the class of linear systems coupled with a nonlinearity satisfying a sector bound or a monotonic condition. First, we present a set of sufficient conditions based on Linear Matrix Inequalities (LMIs) to establish if such a system defines a contraction. Then, we propose a contractive state-feedback control law designed via the use of LMIs which may include nonlinear terms. Furthermore, the design is extended to the case in which an integral action is included in order to achieve constant output regulation. Following a similar approach, the design of contractive observers for the aforementioned class of systems is also considered. Finally, based on previous conditions, we establish a new nonlinear separation principle in which the feedback and the observer gains may be designed independently. Throughout the paper, we illustrate our results by means of practical examples.

It is shown how global exponential stability can still be obtained by adding an anti-windup loop to the integral action. As first contribution towards the solution of this regulation problem, we give some new results related to the characterization of global exponential stability of the equilibrium of a subclass of semi-linear dynamical systems.

Abstract

In food industries, microbial contaminations are difficult to control due to the recurrent formation of biofilms that hinders antimicrobials penetration and efficiency. An understanding of Salmonella Enteritidis biofilms behavior under flow conditions is a key to develop efficient preventive and control strategies. S. Enteritidis biofilms displayed 5.96, 6.28 and 6.80 log CFU cm−2 under 0.006 cm s−1, 0.045 cm s−1, and 0.087 cm s−1 flow velocities, respectively. Biofilms exposed to higher nutrient conditions under greater flow rates, induced significantly more biofilm biomass. To control biofilms, the disinfection efficiency of thymol (THY) was assessed under dynamic conditions by encapsulation it into two types of nanocapsules: monolayer (ML) nanocapsules prepared with a single carrier material (maltodextrin), and layer-by-layer (LBL) nanocapsules prepared by combining two carrier materials (maltodextrin and pectin). A combined mixture of ML and LBL nanocapsules at ½ their minimal inhibitory concentrations induced 99.99% eradication of biofilms developed under the highest flow conditions, after 5 h. ML nanocapsules decreased significantly bacterial counts during the first 0.5 h, while LBL nanocapsules eliminated the remaining bacterial cells and ensured a protection from bacterial contamination for up to 5 h by releasing THY in a sustained manner over time due to the thicker shell wall structure. International audience; In this paper we focus on nonlinear systems composed by a linear term plus a nonlinearity satisfying a monotonic or a sector bound condition. We present sufficient conditions based on LMIs for the design of a feedback control law possessing an infinite gain margin property that makes the closed-loop system to define a contraction. Then, we analyze the connection between infinite gain margin contractive feedbacks and optimization problems by showing how such an infinite gain control design minimizes a given cost function. A practical case of study is given to illustrate our results.

No. of bitstreams: 0 Previous issue date: 2021-04-01 In this study, we evaluated the effect of complexation and microencapsulation with pea protein on the antioxidant activity of protein hydrolysates from bycatch in Brazil. The zeta potential values of complexes changed from negative to positive with the increase of pea protein as a result of positively charged complexes formation. The increase in the ratio of pea protein/hydrolysates also resulted in increased turbidity in all samples. Particle size measurements indicated that the complexes tended to form larger aggregates (ranged from 61.5 ± 1.7 μm to 183 ± 2.8 μm). The 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity of the pea protein/fish protein hydrolysate complexes was higher than that of the protein hydrolysates alone. Moreover, increasing levels of pea protein did not affect the antioxidant activity of fish protein hydrolysates. The complexes of the Paralonchurus brasiliensis were chosen for the microencapsulation process by spray-drying. The results revealed that spray-drying did not have a significant effect (P > 0.05) on the protein hydrolysate antioxidant activity when they were complexed with pea protein. Thus, this work suggests that the complexation with pea protein and subsequent microencapsulation by spray-drying is an efficient way to protect the biological activity of protein hydrolysates obtained from bycatch. This study provides evidence for the potential use of bycatch from shrimp fisheries as functional ingredients or nutraceuticals. International audience; This study aimed to improve the physical, mechanical, and biological properties of a monolayer pectin (P) film containing nanoemulsified trans-Cinnamaldehyde (TC) by incorporating it between inner and outer layers of ethylcellulose (EC). The nanoemulsion had an average size of 103.93 nm and a zeta potential of −46 mV. The addition of the nanoemulsion increased the opacity of the film, reduced its moisture absorption capacity, and improved its antimicrobial activity. However, the tensile strength and elongation at break of the pectin films decreased after the incorporation of nanoemulsions. Multilayer films (EC/P/EC) showed a higher resistance to breaking and better extensibility compared to monolayer films. The antimicrobial activity of both mono and multilayer films was effective in inhibiting the growth of foodborne bacteria during storage of ground beef patties at 8 °C for 10 days. This study suggests that biodegradable antimicrobial multilayer packaging films can be effectively designed and applied in the food packaging industry.; Cette étude visait à améliorer les propriétés physiques, mécaniques et biologiques d'un film monocouche de pectine (P) contenant du trans-cinnamaldéhyde (TC) nanoémulsifié en l'incorporant entre les couches internes et externes d'éthylcellulose (EC). La nanoémulsion avait une taille moyenne de 103,93 nm et un potentiel zêta de -46 mV. L'ajout de la nanoémulsion a augmenté l'opacité du film, réduit sa capacité d'absorption de l'humidité et amélioré son activité antimicrobienne. Cependant, la résistance à la traction et l'élongation à la rupture des films de pectine ont diminué après l'incorporation des nanoémulsions. Les films multicouches (EC/P/EC) ont montré une plus grande résistance à la rupture et une meilleure extensibilité que les films monocouches. L'activité antimicrobienne des films monocouches et multicouches a permis d'inhiber la croissance des bactéries d'origine alimentaire pendant le stockage de galettes de bœuf haché à 8 °C pendant 10 jours. Cette étude suggère que les films d'emballage multicouches biodégradables et antimicrobiens peuvent être conçus et appliqués efficacement dans l'industrie de l'emballage alimentaire.

Abstract

International audience; We study the minimum time problem for a simplified model of a ship towing a long spread of cables. Constraints are on the curvature of the trajectory as well as on the shape of what represent the spread of cables here. This model turns out to be the same as a cart towing two trailers and rolling without sleeping on a plane in uniform translation. We analyse the Hamiltonian system describing the extremal flow given by Pontrjagin maximum principle. We detail the equilibria of the system and prove that, contrary to the case of one trailer studied previously by part of the authors, it is not solvable by quadratures. Preliminary numerical results are given.

International audience; We recently reported (R. Castro Contreras, B. Guicheret, B. F. Machado, C. Rivera-Cárcamo, M. A. Curiel Alvarez, B. Valdez Salas, M. Ruttert, T. Placke, A. Favre Réguillon, L. Vanoye, C. de Bellefon, R. Philippe and P. Serp, J. Catal., 2019, 372, 226–244) that a structure/activity correlation exists in Pd/C catalysts for myrcene hydrogenation, which integrates the Pd dispersion, and the surface concentration of oxygen groups and defects of the support. Here, through a combined experimental–theoretical study, we provide an explanation of the influence of these three structural characteristics of Pd/C catalysts for alkene hydrogenation. Highly dispersed Pd nanoparticles (PdNP) are necessary to activate dihydrogen. A high concentration of surface defects on the support is necessary to stabilize Pd single atoms (PdSA), which coexist with PdNP on Pd/C catalysts. A high concentration of oxygenated surface groups is also necessary on the support to allow hydrogen spillover. We demonstrate that such a combination allows cooperative catalysis to operate between PdNP and PdSA that involves the formation of PdSA–H species, which are much more active than PdNP–H for alkene hydrogenation but also isomerization. Importantly, we also report an efficient method to control the ratio between PdSA and PdNP in Pd/C catalysts of similar loadings and show that the control of this ratio allows the development of a new generation of stable and highly active catalysts integrating the ultra-rational use of precious metals in short supply. Indeed, for myrcene hydrogenation, activity variations of several orders of magnitude were measured as a function of the value of this ratio.

Abstract

International audience; Nisin is a small peptide produced byLactococcus lactisssplactisthat is currently industrially produced. This preservative is often used for growth prevention of pathogenic bacteria contaminating the food products. However, the use of nisin as a food preservative is limited by its low production during fermentation. This low production is mainly attributed to the multitude of parameters influencing the fermentation progress such as bacterial cells activity, growth medium composition (namely carbon and nitrogen sources), pH, ionic strength, temperature, and aeration. This review article focuses on the main parameters that affect nisin production byLactococcus lactisbacteria. Moreover, nisin applications as a food preservative and the main strategies generally used are also discussed.

Abstract

Superparamagnetic nanoparticles are attracting significant attention. Therefore, being explored in microsystems for a wide range of applications. Typical examples include lab-on-a-chip and microfluidics for synthesis, detection, separation, and transportation of different bioanalytes, such as biomolecules, cells, and viruses to develop portable, sensitive, and cost-effective biosensing systems. Particularly, microfluidic systems incorporated with magnetic nanoparticles and, in combination with magnetoresistive sensors, shift diagnostic and analytical methods to a microscale level. In this context, nanotechnology enables the miniaturization and integration of a variety of analytical functions in a single chip for manipulation, detection, and recognition of bioanalytes reliably and flexibly. In consideration of the above, recent development and benefits are elaborated herein to discuss the role of magnetic nanoparticles inside the microchannels to design highly efficient disposable point-of-care applications from transportation to the detection of bioanalytes.

Abstract

International audience; The present work focuses on the development of a performing numerical methodology to solve the steady state Population Balance Equation (PBE) including nucleation, independent size growth and loose agglomeration as crystallization mechanisms. The methodology is based on the solution of two PBEs: one for the isolated crystallites and one describing the loose agglomerates formation. Both are solved by a discretization method and only the last one is reformulated as a fixed point problem. The algorithm solving PBE for agglomeration includes the crossed-secant algorithm as a fixed point acceleration method. The numerical PBE solution method is first validated by comparison to analytical solutions and then applied to the neodymium oxalate precipitation in order to compare to experimental results in a wide range of operating conditions. The methodology is tested under highly restrictive numerical conditions: narrow tolerances, a large amount of points in the discretization scheme and a zero vector as initial condition. The crossed-secant method demonstrates to improve the robustness of the standard fixed point iterations by ensuring the convergence of the agglomerates PBE when penalizing conditions are applied and by reducing the number of iterations otherwise. In all cases, the developed methodology predicts accurately the crystal size distribution under the experimental uncertainty in a reasonable computation time and number of iterations.

Such results are obtained by using sector conditions that are involved in the Lyapunov analysis in order to ensure input-to-state stability. A simulation case study shows the effectiveness of the proposed redesign in denoising and outlier attenuation with increased accuracy and precision.

Abstract

International audience; An original one dimensional population balance model (PBM)-based model of liquid-liquid extraction columns is reported. Compared to existing simulators, ColHySE implements a more realistic description of the flow patterns in the contactor, and predicts its effect on the local droplet-droplet interactions (i.e. breakage and coalescence rates). Proper turbulent properties, extracted from single-phase flow CFD simulations, are used in the source terms of the PBM to evaluate locally the inhomogeneous breakage and coalescence rates, using the averaged Coulaloglou and Tavlarides kernels (Castellano et al., 2018). The sensitivity of the predicted droplets mean diameter, d32, and the holdup, , to the parameters of the used empirical and phenomenological models, on the one hand, and to the operating conditions of the column, on the other hand, was studied. Although some model parts must be refined, and an experimental validation remains necessary, the results confirm that the 1D-PBM methodology used in ColHySE is relevant for predicting the interfacial area in the pulsed column as a function of the operating conditions and geometry, hence highlighting its relevance to study the hydrodynamic stability and the tendency to flooding. The sensitivity analysis has moreover highlighted the needs for an improved slip velocity model.

Abstract

Essential oils active components emerged as captivating antimicrobials to control biofilms developed on food contact surfaces. Free and nanoencapsulated carvacrol (CAR) and thymol (THY) were assessed as antimicrobials against Salmonella Enteritidis biofilms adhered to stainless steel. The developed spherical nanocapsules using the spray-drying technique showed an average size ranged between 159.25 and 234.76 nm and zeta potential values ranged between 23.60 and 24.66 mV. The minimal inhibitory concentrations (MIC) of free THY and CAR were both 1.25 mg L-1. Nanoencapsulation reduced MIC values to 0.62 mg L-1 (THY) and 0.31 mg L-1 (CAR). Furthermore, the exposure to free and nanoencapsulated CAR and THY induced a destabilization of bacterial membranes with obvisous morphological deformations and a pronounced leakage of potassium ions and green fluorescent proteins. Eradication of S. Enteritidis biofilms developed on stainless steel was achieved following a 15 min treatment with nanoencapsulated CAR and THY at 2 MIC. Free antimicrobial exposures induced up to 4.27 log CFU cm(-2) reductions. Additionally, the ecotoxicity tests against Daphnia magna crustaceans reported a non-toxicity of both free and nanoencapsulated CAR and THY after 48 h exposure. Thereby, both CAR and THY antimicrobials proved to be promising natural surface disinfectants that require further exploration and incorporation in food industries.

International audience; Optimal control of nonlinear systems is a difficult problem which has been addressed by both the Control Theory (CT) and Reinforcement Learning (RL) communities. Frequently, the former relies on the linearization of the system thus obtaining only local guarantees. The latter relies on data to build model-free controllers, focused solely on performances. In this paper we propose a methodology to combine the advantages of both approaches, casting the formulation of an optimal local Linear Quadratic Regulator (LQR) into a Deep RL problem. Our solution builds on the linear framework to derive a learnt nonlinear controller showing local stability properties and global performances.

Abstract

International audience; In this paper, we are interested in the estimation of Particle Size Distributions (PSDs) during a batch crystallization process in which particles of two different shapes coexist and evolve simultaneously. The PSDs are estimated thanks to a measurement of an apparent Chord Length Distribution (CLD), a measure that we model for crystals of spheroidal shape. Our main result is to prove the approximate observability of the infinite-dimensional system in any positive time. Under this observability condition, we are able to apply a Back and Forth Nudging (BFN) algorithm to reconstruct the PSD.

Abstract

We address the problem of dynamic output feedback stabilization at an unobservable target point. The challenge lies in according the antagonistic nature of the objective and the properties of the system: the system tends to be less observable as it approaches the target. We illustrate two main ideas: well chosen perturbations of a state feedback law can yield new observability properties of the closed-loop system, and embedding systems into bilinear systems admitting observers with dissipative error systems allows to mitigate the observability issues. We apply them on a case of systems with linear dynamics and nonlinear observation map and make use of an ad hoc finite-dimensional embedding. More generally, we introduce a new strategy based on infinite-dimensional unitary embeddings. To do so, we extend the usual definition of dynamic output feedback stabilization in order to allow infinite-dimensional observers fed by the output. We show how this technique, based on representation theory, may be applied to achieve output feedback stabilization at an unobservable target.

Abstract

Bioactive aminooxime ligands based on optically pure (R)-limonene have been synthesized in two steps. Their ruthenium (II) cationic water-soluble complex was prepared by a reaction between dichloro (para-cymene) ruthenium (II) dimers and aminooxime ligands in a 1:2 molar ratio. Antibacterial and antibiofilm activities of the synthetized complex were assessed against Escherichia coli, Staphylococcus aureus, Listeria monocytogenes, and Enterococcus faecalis. The results revealed that the ruthenium (II) complex has higher antibacterial and antibiofilm activities in comparison with free ligands or the enantiopure (R)-limonene. Moreover, microencapsulation of this complex reduced its cytotoxicity and improved their minimum inhibitory concentration and antibiofilm activity toward the considered bacteria. The ruthenium (II) complex targets the bacterial cell membrane, which leads to rapid leakage of intracellular potassium. Our study suggests that the developed ruthenium (II) complexes could be useful as an alternative to conventional disinfectants.

Abstract

International audience; Dysfunctional elastin turnover plays a major role in the progression of atherosclerotic plaques. Failure of tropoelastin cross-linking into mature elastin leads to the accumulation of tropoelastin within the growing plaque, increasing its instability. Here we present Gd4-TESMA, an MRI contrast agent specifically designed for molecular imaging of tropoelastin within plaques. Gd4-TESMA is a tetrameric probe composed of a tropoelastin-binding peptide (the VVGS-peptide) conjugated with four Gd(III)-DOTA-monoamide chelates. It shows a relaxivity per molecule of 34.0 ± 0.8 mM-1 s-1 (20 MHz, 298 K, pH 7.2), a good binding affinity to tropoelastin (KD = 41 ± 12 μM), and a serum half-life longer than 2 h. Gd4-TESMA accumulates specifically in atherosclerotic plaques in the ApoE-/- murine model of plaque progression, with 2 h persistence of contrast enhancement. As compared to the monomeric counterpart (Gd-TESMA), the tetrameric Gd4-TESMA probe shows a clear advantage regarding both sensitivity and imaging time window, allowing for a better characterization of atherosclerotic plaques.

Abstract

International audience; Molecular structure modifications of camel milk (CaM) and cow milk (CM) mixtures during coagulation were investigated combining mid-infrared (MIR) spectroscopic monitoring with Parallel Factor Analysis (PARAFAC) and particle-size measurements. To evaluate the structure evolution at the molecular level, five different milk formulations were prepared using the following volume fractions of CaM in the mixtures: 100, 75, 50, 25, and 0. Regarding MIR spectroscopy, wavelength ranges located between 3,000 and 2,800 cm(-1) corresponding to fatty acids; from 1,700 to 1,500 cm(-1), related to amide I and II bands; and in the 1,500-900 cm(-1) range called the fingerprint region were considered for the characterization of milk coagulation kinetics. MIR spectroscopy with PARAFAC was allowed for the identification of modifications at the molecular level depending on the coagulation time and milk composition. This was also confirmed by canonical correlation analysis, which demonstrated a high degree of correlation between the casein particle-size distributions and MIR spectra measured during coagulation. Novelty impact statement PARAFAC coupled with MIR spectroscopy were successfully used to monitor coagulation of CM, CaM, and their mixtures (CaM, CM, 1CaM:1CM, 1CaM:3CM, 3CaM:CM1; v/v). PARAFAC coupled with MIR spectroscopy were successfully used to differentiate between physicochemical phenomenon occurring during coagulation of different mixtures of CM, CaM and their mixtures. Canonical correlation analysis (CCA) of MIR spectra and particle-size distribution during milk coagulation demonstrated a strong relationship between the formation of casein aggregates and the variation of the MIR spectral data.

Abstract

The reconstruction mechanisms built by the human auditory system during sound reconstruction are still a matter of debate. The purpose of this study is to refine the auditory cortex model introduced in [9], and inspired by the geometrical modelling of vision. The algorithm transforms the degraded sound in an 'image' in the time-frequency domain via a short-time Fourier transform. Such an image is then lifted in the Heisenberg group and it is reconstructed via a Wilson-Cowan differo-integral equation. Numerical experiments on a library of speech recordings are provided, showing the good reconstruction properties of the algorithm., arXiv admin note: substantial text overlap with arXiv:2004.02450

Abstract

The article deals with the output regulation of a Korteweg-de-Vries (KdV) system subject to a distributed disturbance. The control input and the output are located at the boundary. To achieve this objective, we follow a Lyapunov approach. For that, inspired by the strictication methodology proposed in [42] in the finite-dimensional context, we construct an ISS-Lyapunov functional for the KdV equation thanks to the use of an observer designed via the backstepping approach. Then, thanks to this Lyapunov functional, we apply the forwarding method in order to solve the desired output regulation problem.

Abstract

In recent years, many biocatalytic processes have been developed for the production of chemicals and pharmaceuticals. In this context, enzyme immobilization methods have attracted attention for their advantages, such as continuous production and increased stability. Here, enzyme immobilization methods and a collection of nitrilases from biodiversity for the conversion of 3-cyanopyridine to nicotinic acid were screened. Substrate conversion over 10 conversion cycles was monitored to optimize the process. The best immobilization conditions were found with cross-linking using glutaraldehyde to modify the PMMA beads. This method showed good activity over 10 cycles in a batch reactor at 30 and 40°C. Finally, production with a new thermostable nitrilase was examined in a continuous packed bed reactor, showing very high stability of the biocatalytic process at a flow rate of 0.12 ml min-1 and a temperature of 50°C. The complete conversion of 3-cyanopyridine was obtained over 30 days of operation. Future steps will concern reactor scale-up to increase the production rate with reasonable pressure drops.

Abstract

International audience; We consider the problem of dynamic output feedback stabilization at an unobservable target point. The challenge lies in according the antagonistic nature of the objective and the properties of the system: the system tends to be less observable as it approaches the target. In the literature, switching techniques rapidly appeared as a suitable approach to deal with this issue. On a case of systems with linear conservative dynamics and nonlinear output, this approach is used in conjunction with an embedding into bilinear systems that admit observers with dissipative error. Combining these two elements, global stabilization by means of a dynamic periodic time-varying output feedback is proved, and numerical simulations are provided.

Abstract

International audience; We propose a redesign paradigm for stable estima-tors by introducing a saturation or a dead-zone nonlinearity with adaptive thresholds on the output injection term. Such nonlin-earities allow improving the sensitivity to measurement noise in different scenarios (impulsive disturbances or persistent noise such as sensor bias), while preserving the asymptotic convergence properties of the original observer. These redesigns apply to a broad class of state estimators, including linear observers, observers for input-affine systems, observers for Lipschitz systems , observers based on the circle criterion, high-gain observers, standard and extended Kalman filters. Simulation results confirm the effectiveness of both the stubborn and dead-zone redesigns.

Abstract

International audience; To address the well-known noise sensitivity problems associated with high-gain observers, we insert a low-pass filter on the measurement channel. Considering nonlinear plants in observability canonical form, we first motivate an architecture where the output error is filtered by a linear system parametrized by its arbitrary order and a scalar positive gain. Our main result establishes an exponential finite gain bound for the estimation error, from the measurement noise, this gain being dependent on the high-gain and filter parameters. We also prove bounds depending on the filter parameters characterizing improved high-frequency gains from the measurement noise to the estimation error. The proposed construction is shown to behave desirably in numerical simulations.

Abstract

International audience; Dodecyltrimethylammonium chloride (DTAC) was trapped into maltodextrins/pectin spray dried microcapsules to improve its activity against Salmonella enteritidis and Staphylococcus aureus biofilms. Two different microcapsules were prepared: uncomplexed DTAC-microcapsules (UDM), containing DTAC and maltodextrins; and complexed DTAC-microcapsules (CDM) containing DTAC complexed with pectin and maltodextrins. The minimum inhibitory concentrations (MIC) of both free and microencapsulated DTAC were investigated against S. Enteritidis and S. aureus. The MICs of DTAC were significantly lower when encapsulated. CDM treatment resulted in a 2 and 3.2 log reduction in S. aureus and S. Enteritidis biofilm culturable biomass, respectively. Microencapsulation reduced the cytotoxicity of DTAC by up to 32-fold. Free DTAC and CDM targeted the cell membrane resulting in the leakage of the intracellular molecules and subsequent cell death. The development of DTAC microcapsules reduced the amount of DTAC required to maintain the high standards of cleanliness and hygiene required in the food processing industries.

International audience; Aims: In this study, benzalkonium chloride (BAC) microcapsules were developed for surface disinfection purpose and were evaluated against Listeria monocytogenes and Escherichia coli biofilms.Methods and Results: Microcapsules were prepared with two different strategies: uncomplexed BAC‐microcapsules (UBM), containing BAC and maltodextrins and complexed BAC‐microcapsules (CBM) containing BAC complexed by pectin and maltodextrins. The minimum inhibitory concentrations (MICs) of free and microencapsulated BAC was investigated against two food pathogens: L. monocytogenes and E. coli. The antibiofilm activities of UBM and CBM against L. monocytogenes and E. coli biofilms formed on stainless steel at 37°C were evaluated and compared to BAC used under its free form. MICs of encapsulated BAC were up to 4‐fold lower than those of free BAC. The UBM and CBM showed higher antibiofilm effect when compared to the free BAC.Conclusions: Overall, results demonstrated that microencapsulation enhanced the antibacterial activity of BAC against L. monocytogenes and E. coli biofilms.Significance and Impact of the Study: The application of such BAC microcapsule‐based delivery systems can improve surface disinfection procedures and reduce the required BAC concentrations and the related cytotoxicity of this antimicrobial compound.

Abstract

International audience; The reconstruction mechanisms built by the human auditory system during sound reconstruction are still a matter of debate. The purpose of this study is to propose a mathematical model of sound reconstruction based on the functional architecture of the auditory cortex (A1). The model is inspired by the geometrical modelling of vision, which has undergone a great development in the last ten years. There are however fundamental dissimilarities, due to the different role played by the time and the different group of symmetries. The algorithm transforms the degraded sound in an 'image' in the time-frequency domain via a short-time Fourier transform. Such an image is then lifted in the Heisenberg group and it is reconstructed via a Wilson-Cowan differo-integral equation. Preliminary numerical experiments are provided, showing the good reconstruction properties of the algorithm on synthetic sounds concentrated around two frequencies.

Abstract

International audience; Dyes contamination causes serious damages to aquatic life and human health. With the increase in these persistent organic contaminants accumulation around the world, performant environmentally friendly and low-cost materials and techniques are required to protect the ecosystem and humans. In this study, the efficacy of titania-smectite nanocomposites (Sm-TiO2 NCs) at removing indigo blue (IB) dye was assessed by both batch adsorption and hybrid treatment combining adsorption and ultrafiltration (UF) processes. During the adsorption, the effect of different parameters on IB removal such as dose of Sm-TiO2, contact time, pH and IB concentration were studied. The optimal conditions were applied for the Sm-TiO2 during dye removal using adsorption/UF hybrid treatment. At 293 K and pH 2.5, an optimized dose of Sm-TiO2 (100 mg/L) removed 73% of the IB from a 25 mg/L solution in 240 min with an equilibrium adsorption capacity greater than 180 mg/g. The adsorption of IB dye by Sm-TiO2 NCs followed the Langmuir adsorption isotherm model and pseudo-second-order kinetics. The thermodynamic studies revealed that the adsorption process was spontaneous and exothermic. When a hybrid process was applied, a higher IB removal capacity has been obtained with a lower dose of Sm-TiO2 compared to the case of the batch adsorption process. In addition, the presence of Sm-TiO2 NCs has been found to reduce the UF membrane fouling. In this case, the adsorption followed by UF treatment applied in two stages was modified to a single-stage hybrid process. Permeate flux was increased from 71 L/h m 2 (only UF) to 182 L/h m 2 (Sm-TiO2 /UF).

Abstract

International audience; An experimental study of the hydrodynamics of a gas-liquid co-current flow inside a thin sheet of Open Cell Solid Foam (OCSF) confined in a plate reactor is proposed. Image processing is used to quantify the flow patterns when varying the gas and liquid flow rates and buoyancy (horizontal and vertical upflow). The fraction of {\it joint flow} and {\it interface motion} areas, respectively $\varepsilon_m$ and $\varepsilon_i$, are defined as relevant quantities to characterize the different flow regimes and phases interaction. Two distinct regimes are observed for the vertical flow: the low (LA) and high (HA) activity regimes characterized by a low or high phases interaction. In the horizontal configuration, an additional regime is observed and is called pseudo-trickling (PT) regime due to its similarity with the trickling regime observed in vertical downflow packed beds. The transition between LA and HA regimes is compared to the regime transition in trickle beds.

The resulting soft structured catalytic support (S2CS) has been used as a highly efficient tool for the release of dihydrogen from a commercially available alkaline sodium borohydride solution at room temperature. With a diluted solution containing 0.4 wt% NaBH4 and 0.4 wt% NaOH, the hydrogen generation rate was of 76.4 ± 3.18 L h−1 gcat−1 after stabilization of the catalytic activity. The catalytic tool could be used for 10 runs.

Abstract

International audience; Alternated slug flow in capillaries is an efficient way to separate droplets with different compositions for applications in various fields. This paper describes how to solve the main issue, i.e. droplets coalescence, by using a gas bubble as a separator and demonstrates quantitatively how the mass transfer between resulting liquid-(gas-liquid)-liquid flow is negligibly impacted by the gas bubble size distribution generated. Thus residence time of up to 30 min could be achieved avoiding coalescence. The conditions to generate and maintain the alternated flow such as the superficial velocities, viscosities and surface tensions, are provided. The mechanism for mass transfer between droplets is shown to occur mainly through the film at the wall of the capillaries explaining the negligible impact of the gas. Mastering such alternated slug flow opens the door for many applications requiring compartmentation of several reactions. 2

Abstract

Adipose-derived mesenchymal stem cells (ASCs) are well known for their secretory potential, which confers them useful properties in cell therapy. Nevertheless, this therapeutic potential is reduced after transplantation due to their short survival in the human body and their migration property. This study proposes a method to protect cells during and after injection by encapsulation in microparticles of calcium alginate. Besides, the consequences of encapsulation on ASC proliferation, pluripotential, and secretome were studied. Spherical particles with a mean diameter of 500 &micro, m could be obtained in a reproducible manner with a viability of 70% after 16 days in vitro. Moreover, encapsulation did not alter the proliferative properties of ASCs upon return to culture nor their differentiation potential in adipocytes, chondrocytes, and osteocytes. Concerning their secretome, encapsulated ASCs consistently produced greater amounts of interleukin-6 (IL-6), interleukin-8 (IL-8), and vascular endothelial growth factor (VEGF) compared to monolayer cultures. Encapsulation therefore appears to enrich the secretome with transforming growth factor &beta, 1 (TGF-&beta, 1) and macrophage inflammatory protein-1&beta, (MIP-1&beta, ) not detectable in monolayer cultures. Alginate microparticles seem sufficiently porous to allow diffusion of the cytokines of interest. With all these cytokines playing an important role in wound healing, it appears relevant to investigate the impact of using encapsulated ASCs on the wound healing process.

Abstract

A dynamical model of the pathophysiological behaviors of IL18 and IL10 cytokines with their receptors is tested against data for the case of early sepsis. The proposed approach considers the surroundings (organs and bone marrow) and the different subsystems (cells and cyctokines). The interactions between blood cells, cytokines and the surroundings are described via mass balances. Cytokines are adsorbed onto associated receptors at the cell surface. The adsorption is described by the Langmuir model and gives rise to the production of more cytokines and associated receptors inside the cell. The quantities of pro and anti-inflammatory cytokines present in the body are combined to give global information via an inflammation level function which describes the patient's state. Data for parameter estimation comes from the Sepsis 48 H database. Comparisons between patient data and simulations are presented and are in good agreement. For the IL18/IL10 cytokine pair, 5 key parameters have been found. They are linked to pro-inflammatory IL18 cytokine and show that the early sepsis is driven by components of inflammatory character.
