1. Biotechnological Potential and Safety Evaluation of Dextran- and Riboflavin-Producing Weisella cibaria Strains for Gluten-Free Baking

Abstract: Gluten consumption causes several immunological and non-immunological intolerances in susceptible individuals. In this study, the dextran-producing Weissella cibaria BAL3C-5 and its derivative, the riboflavin-overproducing strain BAL3C-5 C120T, together with a commercial bakery yeast, were used to ferment gluten-free (GF)-doughs obtained from corn and rice flours at two different concentrations and supplemented with either quinoa, buckwheat, or chickpea to obtain laboratory-scale GF bread. The levels of dextran, riboflavin, and total flavins were determined in the fermented and breads. Both strains grew in fermented doughs and contributed dextran, especially to those made with corn plus quinoa (~1 g/100 g). The highest riboflavin (350–150 µg/100 g) and total flavin (2.3–1.75 mg/100 g) levels were observed with BAL3C-5 C120T, though some differences were detected between the various doughs or breads, suggesting an impact of the type of flour used. The safety assessment confirmed the lack of pathogenic factors in the bacterial strains, such as hemolysin and gelatinase activity, as well as the genetic determinants for biogenic amine production. Some intrinsic resistance to antibiotics, including vancomycin and kanamycin, was found. These results indicated the microbiological safety of both W. cibaria strains and indicated their potential application in baking to produce GF bread.

Abstract: The article has the purpose to analyze the role of disability and its inclusion in the history of the tourism in Italy in the twentieth-century. Departing from the Italian law regarding the disabled people and their inclusion in the contemporary Italian society, and continue considering the typologies of tourism, that in last century, allowed the people with disability to have safe and confortable travel condition. The focus of the article is a case study: the summer camps for children and young guests of the Istituto dei Ciechi in Milan, with particular attention to the period of great changes from the mid-fifties to the mid-seventies. Through unpublished documents, we analyze the organization and the leasure time activities of young people at the sea and mountain.

Es recomana als professionals de la seguretat que revisin i actualitzin regularment les seves polítiques i controls de seguretat i que implementin programes de formació i conscienciació per als empleats per ajudar a prevenir violacions de seguretat.

Abstract: Ageing is the main risk factor common to most primary neurodegenerative disorders. Indeed, age-related brain alterations have been long considered to predispose to neurodegeneration. Although protein misfolding and the accumulation of toxic protein aggregates have been considered as causative events in neurodegeneration, several other biological pathways affected by brain ageing also contribute to pathogenesis. Here, we discuss the evidence showing the involvement of the mechanisms controlling neuronal structure, gene expression, autophagy, cell metabolism and neuroinflammation in the onset and progression of neurodegenerative disorders. Furthermore, we review the therapeutic strategies currently under development or as future approaches designed to normalize these pathways, which may then increase brain resilience to cope with toxic protein species. In addition to therapies targeting the insoluble protein aggregates specifically associated with each neurodegenerative disorder, these novel pharmacological approaches may be part of combined therapies designed to rescue brain function.

Abstract: X-ray quasi-periodic eruptions (QPEs) represent a recently discovered example of extreme X-ray variability associated with supermassive black holes. These are high-amplitude bursts recurring every few hours that are detected in the soft X-ray band from the nuclei of nearby galaxies whose optical spectra lack the broad emission lines typically observed in unobscured active galaxies. The physical origin of this new X-ray variability phenomenon is still unknown and several theoretical models have been presented. However, no attempt has been made so far to account for the varying QPE recurrence time and luminosity in individual sources, nor for the diversity of the QPE phenomenology in the different known erupters. We present a semi-analytical model based on an extreme mass-ratio inspiral (EMRI) system where the secondary intersects, along its orbit, a rigidly precessing accretion disc surrounding the primary. We assume that QPEs result from emission from an adiabatically expanding, initially optically thick gas cloud expelled from the disc plane at each impact. We produced synthetic X-ray light curves, which we then compared with X-ray data from four QPE sources: GSN 069, eRO-QPE1, eRO-QPE2, and RX J1301.9+2747. Our model aptly reproduces the diversity of QPE properties between the considered objects and it is also able to naturally account for the varying QPE amplitudes and recurrence times in individual sources. Future implementations will enable us to refine the match with the data and to estimate the system parameters precisely, making additional use of multi-epoch QPE data. We briefly discuss the nature of the secondary object, as well as the possible implications of our findings for the EMRI population at large.

Abstract: Pathology registers can be a useful tool to overcome obstacles in the identification and management of familial hypercholesterolemia since childhood. In 2018, the LIPIGEN pediatric group was constituted within the Italian LIPIGEN study to focus on FH subjects under 18 years. This work aimed at discussing its recent progress and early outcomes. Demographic, biochemical, and genetic baseline characteristics were collected, with an in-depth analysis of the genetic defects. The analysis was carried out on 1,602 children and adolescents (mean age at baseline 9.9 ± 4.0 years), and almost the whole cohort underwent the genetic test (93.3%). Overall, the untreated mean value of LDL-C was 220.0 ± 97.2 mg/dl, with an increasing gradient from subjects with a negative (N = 317; mean untreated LDL-C = 159.9 ± 47.7 mg/dl), inconclusive (N = 125; mean untreated LDL-C = 166.4 ± 56.5 mg/dl), or positive (N = 1,053; mean untreated LDL-C = 246.5 ± 102.1 mg/dl) genetic diagnosis of FH. In the latter group, the LDL-C values presented a great variability based on the number and the biological impact of involved causative variants. The LIPIGEN pediatric group represents one of the largest cohorts of children with FH, allowing the deepening of the characterization of their baseline and genetic features, providing the basis for further longitudinal investigations for complete details.

In order to ensure a successful event, the establishment has striven to expose the modern and cosmopolitan face of Milan, inside and outside the “white city”.

Abstract: The Latest Danian Event (LDE, ~ 62.2 Ma) is characterized by global changes in the carbon cycle as indicated by two negative δ13C excursions of up to ~ 1‰. These δ13C shifts are accompanied by a 2–3 °C warming of both surface and deep waters based on benthic and planktic foraminiferal δ18O measurements, and the LDE has, thus, been considered as a so-called hyperthermal event. The event lasted for 200 kyr and has been identified in various ocean basins and shallow marine settings. Here, we present a compilation of data from three deep-sea cores covering the Pacific, North and South Atlantic oceans as well as a southern Tethyan shelf section to document the response of planktic foraminifera assemblages to ocean warming. In all studied successions, we observe the disappearance of the planktic foraminifer genus Praemurica on a global scale that took place close to the onset of the LDE. Moreover, on the long run, praemuricids were contemporaneously replaced by morozovellids. Both the decline of Praemurica and a temperature increase started between 200 and 260 kyr before the LDE onset and were punctuated by the LDE itself. In this paper, we propose two mechanisms that have controlled the environmental changes associated with this event, (1) increased activity of the North Atlantic Igneous Province acting on long time-scales, and (2) changes of orbital parameters resulting in insolation changes on shorter time-scales. In contrast to a proposed muted benthic foraminiferal response, the planktic community has been substantially impacted by the LDE as indicated by changes in planktic foraminifera faunas and calcareous nannofossils. 20. Record summers in Europe: Variations in drought and heavy precipitation during 1901–2018

Abstract: During the last 20 years some very hot and dry summers affected Europe, resulting in regionally record‐breaking high temperature or low precipitation values. Long‐term changes of such extremely hot and dry summers are of great relevance for our society, as they are connected with manifold negative impacts on human society, natural ecosystems, and diverse economic sectors. Long‐term variations in drought and five record drought summer half years are studied based on 63 stations across Europe with high‐quality precipitation and temperature time series spanning the period 1901–2018. Eight drought indices are deployed to analyse drought intensity, frequency, and duration; four of them purely precipitation‐based and four integrating potential evapotranspiration in the computation. Additionally, three heavy precipitation indices and simultaneous increases in drought and heavy precipitation are studied. The five driest summer half years over Europe are identified (1947, 2018, 2003, 1921, and 1911). They are analysed by aggregating eight drought indices into the aggregated drought evaluation index (ADE) for five subregions. The ADE shows increasing summer drought conditions over most of Europe, except for some stations in northern Europe. The increase in drought conditions during the warm part of the year is particularly pronounced for indices integrating evapotranspiration in their definition. At the same time, the intensity of heavy precipitation events shows a positive trend, as well as an increased contribution to total precipitation. Several stations in central Europe show simultaneously increasing drought conditions and increasing heavy precipitation events. This increases the risks connected with precipitation extremes., Drought and heavy precipitation trends are studied for selected stations with long time series. Both extreme events are related to specific impacts on different economic sectors and thus society. Stations that simultaneously show increasing trends in drought and heavy precipitation mainly occur in central Europe. This indicates a probably higher exposure to these risks in central Europe as well as a demand for broader adaptation options in this region.

Abstract: Mobility data describe the movement of objects in a time interval, such as vehicles, pedestrians, animals. Commonly, mobility data take the form of spatial trajectories, i.e. sequences of timestamped positions in a coordinated space. For example, GPS trajectories are a popular class of spatial trajectories collected outdoors. In this project, we consider a variety of mobility data, including indoor trajectories collected using a positioning infrastructure based on UltraWide Band (UWB). Goal of the project is to investigate on recent Moving Object database solutions for the management of UWB trajectories with focus on recent commercial databasesfor IOT data. In particular, the goal is to analyze a recent Moving Object database called MobilityDB.

Abstract: In cereals with hollow internodes, lodging resistance is influenced by morphological characteristics such as internode diameter and culm wall thickness. Despite their relevance, knowledge of the genetic control of these traits and their relationship with lodging is lacking in temperate cereals such as barley. To fill this gap, we developed an image analysis–based protocol to accurately phenotype culm diameters and culm wall thickness across 261 barley accessions. Analysis of culm trait data collected from field trials in seven different environments revealed high heritability values (>50%) for most traits except thickness and stiffness, as well as genotype-by-environment interactions. The collection was structured mainly according to row-type, which had a confounding effect on culm traits as evidenced by phenotypic correlations. Within both row-type subsets, outer diameter and section modulus showed significant negative correlations with lodging (<−0.52 and <−0.45, respectively), but no correlation with plant height, indicating the possibility of improving lodging resistance independent of plant height. Using 50k iSelect SNP genotyping data, we conducted multi-environment genome-wide association studies using mixed model approach across the whole panel and row-type subsets: we identified a total of 192 quantitative trait loci (QTLs) for the studied traits, including subpopulation-specific QTLs and 21 main effect loci for culm diameter and/or section modulus showing effects on lodging without impacting plant height. Providing insights into the genetic architecture of culm morphology in barley and the possible role of candidate genes involved in hormone and cell wall–related pathways, this work supports the potential of loci underpinning culm features to improve lodging resistance and increase barley yield stability under changing environments.

Abstract: Biogeographical studies have traditionally focused on readily visible organisms, but recent technological advances are enabling analyses of the large-scale distribution of microscopic organisms, whose biogeographical patterns have long been debated. Here we assessed the global structure of plankton geography and its relation to the biological, chemical, and physical context of the ocean (the ‘seascape’) by analyzing metagenomes of plankton communities sampled across oceans during the Tara Oceans expedition, in light of environmental data and ocean current transport. Using a consistent approach across organismal sizes that provides unprecedented resolution to measure changes in genomic composition between communities, we report a pan-ocean, size-dependent plankton biogeography overlying regional heterogeneity. We found robust evidence for a basin-scale impact of transport by ocean currents on plankton biogeography, and on a characteristic timescale of community dynamics going beyond simple seasonality or life history transitions of plankton.

Abstract: The physical processes that operate within, and beneath, a volcano control the frequency, duration, location and size of volcanic eruptions. Volcanotectonics focuses on such processes, combining techniques, data, and ideas from structural geology, tectonics, volcano deformation, physical volcanology, seismology, petrology, rock and fracture mechanics and classical physics. A central aim of volcanotectonics is to provide sufficient understanding of the internal processes in volcanoes so that, when combined with monitoring data, reliable forecasting of eruptions, vertical (caldera) and lateral (landslide) collapses and related events becomes possible. To gain such an understanding requires knowledge of the material properties of the magma and the crustal rocks, as well as the associated stress fields, and their evolution. The local stress field depends on the properties of the layers that constitute the volcano and, in particular, the geometric development of its shallow magma chamber. During this decade an increasing use of data from InSAR, pixel offset and structure-from-motion, as well as dense, portable seismic networks will provide further details on the mechanisms of volcanic unrest, magma-chamber rupture, the propagation of magma-filled fractures (dikes, inclined sheets and sills) and lateral and vertical collapse. Additionally, more use will be made of accurate quantitative data from fossil and active volcanoes, combined with realistic numerical, analytical and machine-learning studies, so as to provide reliable models on volcano behaviour and eruption forecasting.

Abstract: Coronavirus disease 2019 (COVID-19) is the greatest threat to global health at the present time, and considerable public and private effort is being devoted to fighting this recently emerged disease. Despite the undoubted advances in the development of vaccines against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the causative agent of COVID-19, uncertainty remains about their future efficacy and the duration of the immunity induced. It is therefore prudent to continue designing and testing vaccines against this pathogen. In this article we computationally designed two candidate vaccines, one monopeptide and one multipeptide, using a technique involving optimizing lambda-superstrings, which was introduced and developed by our research group. We tested the monopeptide vaccine, thus establishing a proof of concept for the validity of the technique. We synthesized a peptide of 22 amino acids in length, corresponding to one of the candidate vaccines, and prepared a dendritic cell (DC) vaccine vector loaded with the 22 amino acids SARS-CoV-2 peptide (positions 50-71) contained in the NTD domain (DC-CoVPSA) of the Spike protein. Next, we tested the immunogenicity, the type of immune response elicited, and the cytokine profile induced by the vaccine, using a non-related bacterial peptide as negative control. Our results indicated that the CoVPSA peptide of the Spike protein elicits noticeable immunogenicity in vivo using a DC vaccine vector and remarkable cellular and humoral immune responses. This DC vaccine vector loaded with the NTD peptide of the Spike protein elicited a predominant Th1-Th17 cytokine profile, indicative of an effective anti-viral response. Finally, we performed a proof of concept experiment in humans that included the following groups: asymptomatic non-active COVID-19 patients, vaccinated volunteers, and control donors that tested negative for SARS-CoV-2. 26. Microalgae colonization of different microplastic polymers in experimental mesocosms across an environmental gradient

Abstract: A variety of organisms can colonize microplastic surfaces through biofouling processes. Heterotrophic bacteria tend to be the focus of plastisphere research; however, the presence of epiplastic microalgae within the biofilm has been repeatedly documented. Despite the relevance of biofouling in determining the fate and effects of microplastics in aquatic systems, data about this process are still scarce, especially for freshwater ecosystems. Here, our goal was to evaluate the biomass develop-ment and species composition of biofilms on different plastic polymers and to in-vestigate whether plastic substrates exert a strong enough selection to drive species sorting, overcoming other niche-defining factors. We added microplastic pellets of high- density polyethylene (HDPE), polyethylene terephthalate (PET), and a mix of the two polymers in 15 lentic mesocosms in five different locations of the Iberian Peninsula, and after one month, we evaluated species composition and biomass of microalgae developed on plastic surfaces. Our results, based on 45 samples, showed that colonization of plastic surfaces occurred in a range of lentic ecosystems covering a wide geographical gradient and different environmental conditions (e.g., nutrient concentration, conductivity, macrophyte coverage). We highlighted that total bio-mass differed based on the polymer considered, with higher biomass developed on PET substrate compared to HDPE. Microplastics supported the growth of a rich and diversified community of microalgae (242 species), with some cosmopolite species. However, we did not observe species-specificity in the colonization of the different plastic polymers. Local species pool and nutrient concentration rather than polymeric composition seemed to be the determinant factor defying the community diversity. Abstract: In semi-arid environments, the effects of irregularly distributed rainfall, flow regulation and water inter-basin transfer enhance the spread of non-native fish to the detriment of native communities. In the River Segura, since the 1980s the number of non-native fish species has progressively increased, also because of the building of water transfer facility connecting the rivers Segura and Tajo. With the aim of highlighting how man-driven changes in the diversity of fish communities affect the diet of top-predators, we compared Eurasian otter Lutra lutra diet in the span of 20 years, i.e. 1997–98 vs. 2016–19. As habitat quality affects the condition of Andalusian barbel Luciobarbus sclateri, the most widespread native fish, we also compared the size of preyed barbels to point out whether human activities may have lowered their profitability to otters. Fish and introduced red swamp crayfish Procambarus clarkii formed the bulk of otter diet in both study periods. In 2016–19 the contribution of non-native species to otter diet increased significantly, both for crayfish and fish, which included ten non-native species. Otter feeding habits faithfully mirrored the variation in the composition of the fish community and confirmed the importance of crayfish as alternative-to-fish prey in the Iberian Peninsula. The average length of preyed barbels was significantly lower in the second study period, consistently with a decline in barbel profitability for otters.

Abstract: Disamistade is the sardinian word for revenge in Barbagia, central part of Sardinia. From 1905 to 1917 in Orgosolo revenge raised among two families with so much cruelty that the autorities decided to intervene and end violence up. The trial against bandits in Sassari became the arena of the fight between the establishment and the community of Orgosolo, in which the revenge code confronted with the Law. In the end, thanks to the ability to convince the court, the defendants were completely acquitted and the code of revenge survived.

Abstract: Maytansinol is a valuable precursor for the preparation of maytansine derivatives (known as maytansinoids). Inspired by its intriguing structure and their success in targeted cancer therapy, we explored the maytansinol acylation reaction. As a result, we were able to obtain a series of derivatives, bearing novel modifications of the maytansine scaffold. We characterized these molecules by docking studies, by a comprehensive biochemical evaluation and by determination of their crystal structures in complex with tubulin. The obtained results shed further light on the intriguing chemical behavior of maytansinoids and confirm the relevance of this peculiar scaffold in the scenario of tubulin binders.

Abstract: Following the indications reported in Task 6.3.5 of the MADFORWATER project, MADFORWATER partners dedicated a relevant effort to the development of guidelines for the adaptation to other basins in Morocco, Egypt and Tunisia and to other MENA countries of the MADFORWATER tools, technologies and approaches. The general goal of these guidelines is to allow a generic stakeholder (such as a manager of a wastewater treatment plant, a wastewater company, an irrigation company, a farmer, the director of a basin authority, an officer of the Ministry of Irrigation) to perform the necessary activities in order to adapt the MADFORWATER technologies and water management tools to the specific context in which the considered technology or tool needs to be implemented. The adaptation guidelines developed in the framework of MADFORWATER and presented in this deliverable are relative to (i) the MADFORWATER approach for the analysis of water security, (ii) the project’s technologies for wastewater treatment and irrigation, and (iii) the two MADFORWATER decision support tools for wastewater management and for water & land management in agriculture. With regard to these two decision support tools, the adaptation guidelines are integrated by the description of the structure of two training courses that the MADFORWATER partners FHNW, IAMB and UPM can offer to potentially interested stakeholders. The adaptation guidelines presented in this deliverable are integrated by a consistent amount of material – such as the user manuals and computer codes for the decision support tools, the detailed description of the MADFORWATER approach for the assessment of water security and the fact-sheets of the MADFORWATER technologies – freely accessible to any interested stakeholder through the MADFORWATER website or the repositories in which the MADFORWATER dataset have been deposited (mainly, AMS Acta and Zenodo). Abstract: Objectiu: Aquest treball és un petita recerca bibliogràfica que pretén realitzar un sèrie de comparacions amb diferents tipus de tonòmetres. L’objectiu principal és analitzar si els nous dispositius que han aparegut recentment al món de l’òptica poden substituir al tonòmetre de referencia que tothom ha utilitzat algun cop per mesurar la pressió intraocular (PIO). Com molt bé es coneix la PIO és una de les principals eines diagnòstiques per detectar el glaucoma. Mètodes: Per realitzar el treball ens hem basat en diferents estudis apareguts en la darrera dècada, amb la intenció de recol·lectar la informació per poder comparar els tonòmetres. Però per poder fer-ho s’ha de conèixer els seus avantatges i desavantatges. A més a més, per fer-ho una mica més fàcil es classificarà alguns tonòmetres en un grup anomenat Antecedents i els altres en Nous. D’aquesta forma es podrà diferencia la tecnologia antiga de la nova, per veure la evolució amb el pas del temps. Conclusions: Com a conclusió principal es destaca que cap instrument es capaç de substituir el tonòmetre de Goldmann. Tot i que es corregeixin les limitacions d’aquest. Això succeeix perquè no s’aconsegueix saber quina es la PIO real del pacient. Perquè cada tonòmetre aporta un valor lleugerament diferent als altres. Per tant, no se sap quin d’ells és més fiable. Potser en un futur s’hauria d’estudiar alguna forma de obtindré una PIO el més real possible, igual llavors es sabria quin tipus de tonòmetre és més fiable

Abstract: Introducció: Els criteris per a la prescripció refractiva en infants preescolars hipermetrops no són universals i de vegades resulten controvertits. La hipocorrecció és el criteri de prescripció tradicional en els casos no associats a endodesviacions, atès la gran amplitud acomodativa dels infants i l’interès en evitar la interferència en el procés de l’emmetropització. En aquest treball descobrirem si les recomanacions tradicionals estan basades en l’evidència científica i si difereixen dels tractaments refractius més actualitzats. Objectiu: Identificar les evidències de la literatura que avalen els criteris de tractament refractiu de la hipermetropia moderada en infants menors de 7 anys, realitzar una anàlisi dels criteris i recomanacions tradicionals i dur a terme una identificació dels criteris basats en l’evidència científica. Mètode: S’ha realitzat un estudi d’investigació bibliogràfica sobre diversos articles que inclouen l’actualització del tractament refractiu de la hipermetropia moderada, basat en l’evidència, per a nens menors de 7 anys i l’impacte de la prescripció de la hipermetropia moderada o baixa en l’activitat acadèmica i/o en les dificultats de l’aprenentatge o lectura. Conclusions: Pràcticament tots els criteris de prescripció refractiva que disposen els professionals de la salut visual estan basats en la pràctica clínica i encara no existeixen suficients evidències científiques per a la recomanació específica sobre a partir de quin grau d’hipermetropia s’ha de corregir en funció de l’edat i quina és la quantitat hipocorregida aconsellada en la prescripció refractiva de la hipermetropia moderada en infants preescolars

Abstract: Un estudi realitzat per la Gaceta d’optometria i òptica oftàlmica mostra que el nombre d’usuaris de lents de contacte és molt baix en comparació amb l’ús de les ulleres. El poc coneixement sobre les lents de contacte pot ser la principal raó que justifica aquest fet. A la vegada, també hi ha un gran percentatge d’usuaris de lents de contacte que abandonen el seu ús ja sigui per incomoditat o bé per molèsties. Per tal d’evitar aquesta situació és important l’educació dels nous usuaris, portar un control i un seguiment estricte de les adaptacions. A les pràctiques de OCC (optometria i contactologia clinica) els estudiants realitzen una o cap adaptació de lents de contacte en tot el curs acadèmic. Un dels motius es la manca de demanda de adaptacions. Per aconseguir tenir més pacients ens proposem trobar eines que puguin incrementar la motivació i l’ús de les lents de contacte entre els joves i adolescents. En aquest treball, es presenten diferents tipus de comunicació possibles per fer arribar el nostre missatge (Presentació Power Point, tríptic, campanya a les xarxes socials com ara INSTAGRAM, xerrades dirigides als pares, creació d’una Pàgina web). La creació d’una pàgina web és una via que estaria a l’abast de tothom per tal de poder obtenir la màxima informació amb facilitat i consultar-la en qualsevol moment. Tot i que també s’hauria de donar una gran importància als altres grups d’edats i fer-ne una gran difusió

Abstract: Protein self-assembly is a ubiquitous phenomenon, traditionally studied for its links to amyloid pathologies, which has also gained attention as its physiological roles and possible biotechnological applications emerged over time. It is also known that varying the conditions to which proteins are exposed can lead to aggregate polymorphism. To understand the factors that trigger aggregation and/or direct it toward specific outcomes, we performed a multifaceted structural characterization of the thermally induced self-assembly process of MNEI, a model protein able to form amyloid aggregates under nondenaturing conditions. MNEI is also known for its extreme sweetness which, combined with a considerable thermal stability, makes the protein a promising alternative sweetener. Fourier-transformed infrared spectroscopy and electron microscopy data showed that the presence of NaCl accelerates the kinetics of fibrillar aggregation, while disfavoring the population of off-pathway states that are instead detected by native gel electrophoresis at low ionic strength. NMR studies revealed how NaCl modulates the self-assembling mechanism of MNEI, switching the process from soluble oligomeric forms to fibrils. Comparative analysis demonstrated that the presence of NaCl induces local differences in the protein dynamics and surface accessibility, without altering the native fold. We identified the regions most affected by the presence of NaCl, which control the aggregation process, and represent hot spots on the protein surface for the rational design of new mutants with controlled aggregation propensity.

Abstract: Two new lead(II) coordination polymer complexes (CSCs) (2D), [Pb2(L)2(Br)2]n·H2O (1), [Pb2(HL/)(L/)(H2O)2]n·H2O (2), where L = C6H5NO2 (2-pyridinecarboxylic acid) and L/ = C9H6O6 (1,3,5-tricarboxylic acid), have been synthesized under different experimental conditions. Micrometric crystals (bulk) or microsized materials have been obtained depending on using the branch tube method or sonochemical irradiation. All materials have been characterized by scanning electron microscopy (SEM), powder X-ray diffraction (PXRD) and FT-IR spectroscopy. Single crystal X-ray analyses on complexes 1 and 2 shows that Pb2+ ions are 8-coordinated, 7 and 9-coordinated, respectively. Topological analysis shows that the compound 1 and 2 are 4,6L26 and bnn net, respectively. However, neither the shape nor the morphology is maintained, showing the role of sonochemistry to modulate both morphology and dimensions of the resulting crystalline material, independently of whether we have a 2D coordination polymer (CP). Finally, micro structuration of lead(II) bromide oxide and lead(II) oxide have been prepared by calcination of two different lead (II) CPs at 700 °C that were characterized by SEM and XRD.

Abstract: [Background]: Since goat was domesticated 10,000 years ago, many factors have contributed to the differentiation of goat breeds and these are classified mainly into two types: (i) adaptation to different breeding systems and/or purposes and (ii) adaptation to different environments. As a result, approximately 600 goat breeds have developed worldwide; they differ considerably from one another in terms of phenotypic characteristics and are adapted to a wide range of climatic conditions. In this work, we analyzed the AdaptMap goat dataset, which is composed of data from more than 3000 animals collected worldwide and genotyped with the CaprineSNP50 BeadChip. These animals were partitioned into groups based on geographical area, production uses, available records on solid coat color and environmental variables including the sampling geographical coordinates, to investigate the role of natural and/or artificial selection in shaping the genome of goat breeds., [Results]: Several signatures of selection on different chromosomal regions were detected across the different breeds, sub-geographical clusters, phenotypic and climatic groups. These regions contain genes that are involved in important biological processes, such as milk-, meat- or fiber-related production, coat color, glucose pathway, oxidative stress response, size, and circadian clock differences. Our results confirm previous findings in other species on adaptation to extreme environments and human purposes and provide new genes that could explain some of the differences between goat breeds according to their geographical distribution and adaptation to different environments., [Conclusions]: These analyses of signatures of selection provide a comprehensive first picture of the global domestication process and adaptation of goat breeds and highlight possible genes that may have contributed to the differentiation of this species worldwide.

Abstract: The plastid-localized phosphoglucose isomerase isoform PGI1 is an important determinant of growth in Arabidopsis thaliana, likely due to its involvement in the biosynthesis of plastidial isoprenoid-derived hormones. Here, we investigated whether PGI1 also influences seed yields. PGI1 is strongly expressed in maturing seed embryos and vascular tissues. PGI1-null pgi1-2 plants had ∼60% lower seed yields than wild-type plants, with reduced numbers of inflorescences and thus fewer siliques and seeds per plant. These traits were associated with low bioactive gibberellin (GA) contents. Accordingly, wild-type phenotypes were restored by exogenous GA application. pgi1-2 seeds were lighter and accumulated ∼50% less fatty acids (FAs) and ∼35% less protein than wild-type seeds. Seeds of cytokinin-deficient plants overexpressing CYTOKININ OXIDASE/DEHYDROGENASE1 (35S:AtCKX1) and GA-deficient ga20ox1 ga20ox2 mutants did not accumulate low levels of FAs, and exogenous application of the cytokinin 6-benzylaminopurine and GAs did not rescue the reduced weight and FA content of pgi1-2 seeds. Seeds from reciprocal crosses between pgi1-2 and wild-type plants accumulated wild-type levels of FAs and proteins. Therefore, PGI1 is an important determinant of Arabidopsis seed yield due to its involvement in two processes: GA-mediated reproductive development and the metabolic conversion of plastidial glucose-6-phosphate to storage reserves in the embryo.

Abstract: The article focuses on two populist experiences which developed in a context of modernization (Italy and Brazil), and were characterized by a strong relationship between a charismatic leader and the poorest. Italy and Brazil, in fact, were two interesting populist cases in global postwar scenario. Achille Lauro from the right, and Jânio Quadros from the left, acted as two successful outsiders. Both of them succeeded in gaining popular consensus at a grass-root level through bizarre and extremely effective electoral campaigns. Their speeches were mainly based on simple messages against bureaucracy and traditional political parties. Moreover, they perfectly knew how to deal with their people.

Abstract: A search for neutrinoless ββ decay processes accompanied with Majoron emission has been performed using data collected during Phase I of the GERmanium Detector Array (GERDA) experiment at the Laboratori Nazionali del Gran Sasso of INFN (Italy). Processes with spectral indices n=1,2,3,7 were searched for. No signals were found and lower limits of the order of 10 23 yr on their half-lives were derived, yielding substantially improved results compared to previous experiments with 76 Ge. A new result for the half-life of the neutrino-accompanied ββ decay of 76 Ge with significantly reduced uncertainties is also given, resulting in T1/22ν=(1.926±0.094)×1021 yr.

Abstract: We present a procedure that allows a reliable determination of the elastic (Young's) modulus of soft samples, including living cells, by atomic force microscopy (AFM). The standardized nanomechanical AFM procedure (SNAP) ensures the precise adjustment of the AFM optical lever system, a prerequisite for all kinds of force spectroscopy methods, to obtain reliable values independent of the instrument, laboratory and operator. Measurements of soft hydrogel samples with a well-defined elastic modulus using different AFMs revealed that the uncertainties in the determination of the deflection sensitivity and subsequently cantilever's spring constant were the main sources of error. SNAP eliminates those errors by calculating the correct deflection sensitivity based on spring constants determined with a vibrometer. The procedure was validated within a large network of European laboratories by measuring the elastic properties of gels and living cells, showing that its application reduces the variability in elastic moduli of hydrogels down to 1%, and increased the consistency of living cells elasticity measurements by a factor of two. The high reproducibility of elasticity measurements provided by SNAP could improve significantly the applicability of cell mechanics as a quantitative marker to discriminate between cell types and conditions.

Abstract: The GERmanium Detector Array ( Gerda ) experiment at the Gran Sasso underground laboratory (LNGS) of INFN is searching for neutrinoless double beta ( 0Î½Î²Î² ) decay of 76 Ge. The signature of the signal is a monoenergetic peak at 2039Â keV, the QÎ²Î² value of the decay. To avoid bias in the signal search, the present analysis does not consider all those events, that fall in a 40Â keV wide region centered around QÎ²Î² . The main parameters needed for the 0Î½Î²Î² analysis are described. A background model was developed to describe the observed energy spectrum. The model contains several contributions, that are expected on the basis of material screening or that are established by the observation of characteristic structures in the energy spectrum. The model predicts a flat energy spectrum for the blinding window around QÎ²Î² with a background index ranging from 17.6 to 23.8Â Ã Â 10-3 Â cts/(keVÂ kgÂ yr). A part of the data not considered before has been used to test if the predictions of the background model are consistent. The observed number of events in this energy region is consistent with the background model. The background at QÎ²Î² is dominated by close sources, mainly due to 42 K, 214 Bi, 228 Th, 60 Co and Î± emitting isotopes from the 226 Ra decay chain. The individual fractions depend on the assumed locations of the contaminants. It is shown, that after removal of the known Î³ peaks, the energy spectrum can be fitted in an energy range of 200Â keV around QÎ²Î² with a constant background. This gives a background index consistent with the full model and uncertainties of the same size.

Abstract: SummaryAtrial fibrillation (AF) patients are at high risk for thrombotic and vascular events related to their cardiac arrhythmia and underlying systemic atherosclerosis. Ankle-Brachial Index (ABI) is a non-invasive tool in evaluating systemic atherosclerosis, useful in predicting cardiovascular events in general population; no data are available in AF patients. ARAPACIS is a prospective multicentre observational study performed by the Italian Society of Internal Medicine, analysing association between low ABI (≤0.90) and vascular events in NVAF out- or in-patients, enrolled in 136 Italian centres. A total of 2,027 non-valvular AF (NVAF) patients aged > 18 years from both sexes followed for a median time of 34.7 (interquartile range: 22.0–36.0) months, yielding a total of 4,614 patient-years of observation. Mean age was 73 ± 10 years old with 55% male patients. A total of 176 patients (8.7%) experienced a vascular event, with a cumulative incidence of 3.81%/patient-year. ABI≤ 0.90 was more prevalent in patients with a vascular event compared with patients free of vascular events (32.2 vs 20.2%, p< 0.05). On Cox proportional hazard analysis, ABI≤ 0.90 was an independent predictor of vascular events (hazard ratio (HR): 1.394, 95% confidence interval (CI): 1.042–1.866; p=0.02), vascular death (HR: 2.047, 95% CI: 1.255-3.338; p=0.004) and MI (HR: 2.709, 95%> CI: 1.485-5.083; p=0.001). This latter association was also confirmed after excluding patients with previous MI (HR: 2.901, 95% CI: 1.408-5.990, p=0.004). No association was observed between low ABI and stroke/transient ischaemic attack (p=0.91). In conclusion, low ABI is useful to predict MI and vascular death in NVAF patients and may independently facilitate cardiovascular risk assessment in NVAF patients.Note: The review process for this paper was fully handled by C. 50. Kinetic Analysis of Zinc/Cadmium Reciprocal Competitions Suggests a Possible Zn-Insensitive Pathway for Root-to-Shoot Cadmium Translocation in Rice

Abstract: Cadmium (Cd) and zinc (Zn) translocation in rice was studied by analyzing a complete set of competition experiments in which plants were exposed to increasing Zn external concentrations (0.1, 1, and 10 µM), in the absence or presence of 0.1 µM Cd2+, or to different Cd concentrations (0, 0.01, 0.1, and 1 µM) in the presence of 1 µM Zn2+. Results revealed the lack of a full reciprocity when considering the effect of Cd on Zn accumulation, and vice versa, since the accumulation of Zn in the shoots was progressively inhibited by Cd increases, whereas that of Cd was only partially impaired by Zn. Such behaviors probably involved the mechanisms of Cd-xylem loading, as suggested by: i) the analysis of Zn and Cd content in the xylem sap performed in relation to the concentration of the two metals in the mobile fractions of the roots; ii) the analysis of the systemic movement of 107Cd in short term PETIS experiments. From these results we suggest that at least two pathways may mediate root-to-shoot Cd translocation in rice. The former could involve OsHMA2 as Zn2+/Cd2+ xylem loader, whereas the latter appears to involve a Zn-independent system that still needs to be identified.
