1. Improved Metal Cation Optosensing Membranes through the Incorporation of Sulphated Polysaccharides

Abstract

Optosensing chitosan-based membranes have been applied for the detection of heavy metals, especially in drinking water. The novelty of this study is based on the use of sulphated polysaccharides, in such optosensing membranes, aiming at an improved analytical performance. The sulphated polysaccharides, such as ulvan, fucoidan and chondroitin sulfate, were extracted from by-products and wastes of marine-related activities. The membranes were developed for the analysis of aluminum. The variation in the visible absorbance of the sensor membranes after the contact between the chromophore and the aluminum cation was studied. The membranes containing sulphated polysaccharides showed improved signals when compared to the chitosan-only membrane. As for the detection limits for the membranes containing ulvan, fucoidan and chondroitin sulfate, 0.17 mg L−1, 0.21 mg L−1 and 0.36 mg L−1 were obtained, respectively. The values were much lower than that obtained for the chitosan-only membrane, 0.52 mg L−1, which shows the improvement obtained from the sulphated polysaccharides. The results were obtained with the presence of CTAB in analysis solution, which forms a ternary complex with the aluminum cation and the chromophore. This resulted in an hyperchromic and batochromic shift in the absorption band. When in the presence of this surfactant, the membranes showed lower detection limits and higher selectivity

Abstract

Marine algae are recognised sources of bioactive compounds that have attracted great interest as nutritional supplements for aquaculture fish. Intensive rearing conditions often expose fish to husbandry-related stressors, rendering fish more susceptible to disease and reducing production yields. The present work evaluated the potential of two marine algae extracts (Fucus vesiculosus and Nannochloropsis gaditana) as nutritional supplements to mitigate stress effects in meagre (Argyrosomus regius) exposed to an acute handling stress (AS). A plant-based diet was used as a control, and three other diets were prepared, which were similar to the control diet but supplemented with 1% of each algal extract or a combination of the two extracts (0.5% each). The effects of supplemented diets on stress biomarkers, antioxidant enzyme activities, and immune response were analysed in fish exposed to AS after 4 weeks of feeding. Supplemented diets did not affect growth performance but the inclusion of F. vesiculosus promoted higher feed efficiency, as compared to the control group. Dietary algal extracts supplementation reduced plasma glucose levels, increased white blood cell counts, and reduced the expression of pro-inflammatory genes when compared with the control. N. gaditana supplementation led to a reduction in hepatic antioxidant enzyme activity and glutathione levels, while F. vesiculosus supplementation increased muscle glutathione reductase activity and reduced lipid peroxidation. These findings support the potential of algal extracts as nutraceuticals in aquafeeds to enhance the ability of fish to cope with husbandry-related stressful conditions and ultimately improve fish health and welfare.

3. Brighter is darker: the Hamilton-Zuk hypothesis revisited in lizards

Abstract

Several studies of lizards have made an erroneous interpretation of negative relationships between spectral brightness and parasite load, and thus provided misleading support for the Hamilton–Zuk hypothesis (HZH). The HZH predicts that infected hosts will produce poorer sexual ornamentation than uninfected individuals as a result of energetic trade-offs between immune and signalling functions. To test whether there is a negative relationship between spectral brightness and pigment content in the skin of lizards, we used spectrophotometry to quantify the changes in spectral brightness of colour patches of two species after chemically manipulating the contents of orange, yellow and black pigments in skin samples. Carotenoids were identified using high- performance liquid chromatography. In addition, we compared the spectral brightness in the colour patches of live individuals with differential expression of nuptial coloration. Overall, the analyses demonstrated that the more pigmented the colour patch, the darker the spectrum. We provide a comprehensive interpretation of how variation in pigment content affects the spectral brightness of the colour patches of lizards. Furthermore, we review 18 studies of lizards presenting 24 intraspecific tests of the HZH and show that 14 (58%) of the tests do not support the hypothesis

4. Methodological improvements for detecting and identifying scats of an expanding mesocarnivore in south-western Europe

Abstract

The use of scats is a widespread non-invasive method in ecological studies of mammalian carnivores. However, their low detectability and the incorrect species assignment may be important sources of bias. We aimed to optimize the detection and identification of scats of Egyptian mongoose (Herpestes ichneumon), using the red fox (Vulpes vulpes) as a comparative model. Based on molecular identification of scats we assessed: (1) the accuracy of species morphological identification (Field-ID); (2) whether post-field laboratory analyses (FL-ID) improve Field-ID; (3) species-specific morphological differences of scats, and (4) whether specific field surveys increase the detectability of mongoose scats. Out of 175 collected scats, 81 were genetically identified. Field-ID accuracy was over 75.6% for the mongoose and over 45.0% for the red fox. Misidentified mongoose scats mainly belonged to stone marten (50.0%) while misidentified red fox scats mainly belonged to mongoose (63.6%). After applying FL-ID, accuracy increased to 93.1% for the Egyptian mongoose and 76.2% for the red fox. Morphological scat differences were only significant for the scat diameter, with red fox scats being significantly thicker than those of mongoose. More mongoose scats were found along ecotones (mean ± SE: 4.05 ± 1.45 scats/transect) than along trails and roads (0.05 ± 0.05 scats/transect), while red fox scats were found similarly in both transect types. The application of post-field analysis to scats and focusing the search along ecotones optimized both the identification accuracy and the detection probability of Egyptian mongoose scats, although searching for scats in other structures should not be discarded if the aim is the study of habitat use or selection by the species. Our results can be useful for scat identification in studies on this and other carnivore species.

5. Prunus lusitánica L.: An endangered plant species relict in the central region of mainland Portugal

Abstract

Prunus lusitanica L. is a paleotropical relic species with an Ibero-Maghrebian distribution, which is presently considered as an endangered species, recognized by the Natura 2000 Network (92/43/EEC) as a priority habitat for conservation in Europe. The mountains in the Portuguese mainland central region offer the best location for this species to occur. The main objective of this study is to measure the current conservation status of the communities of P. lusitanica, through the collection of field data, such as the number of existing individuals of each population and their location, which will then be comparatively analyzed based on the previous literature, published at least 15 years ago. Soil characterization analysis was carried out and the main threats to conservation were identified. As a result, a decline of approximately 40% was observed in the number of individuals and in the quality of their habitat. The main threats to their conservation were found to be the seasonal occurrence of rural fires and the expansion of invasive species, such as Acacia dealbata Link and Ailanthus altissima (Mill.) Swingle. Finally, we present the main management methodologies that should be considered

6. Discordant patterns of introgression across a narrow hybrid zone between two cryptic lineages of an Iberian endemic newt

Abstract

The study of natural hybrid zones can illuminate aspects of lineage divergence and speciation in morphologically cryptic taxa. We studied a hybrid zone between two highly divergent but morphologically similar lineages (south-western and south-eastern) of the Iberian endemic Bosca's newt (Lissotriton boscai) in SW Iberia with a multilocus dataset (microsatellites, nuclear and mitochondrial genes). STRUCTURE and NEWHYBRIDS analyses retrieved few admixed individuals, which classified as backcrosses involving parental individuals of the south-western lineage. Our results show asymmetric introgression of mtDNA beyond the contact from this lineage into the south-eastern lineage. Analysis of nongeographic introgression patterns revealed asymmetries in the direction of introgression, but except for mtDNA, we did not find evidence for nonconcordant introgression patterns across nuclear loci. Analysis of a 150-km transect across the hybrid zone showed broadly coincident cline widths (ca. 3.2–27.9 km), and concordant cline centres across all markers, except for mtDNA that is displaced ca. 60 km northward. Results from ecological niche modelling show that the hybrid zone is in a climatically homogenous area with suitable habitat for the species, suggesting that contact between the two lineages is unlikely to occur further south as their distributions are currently separated by an extensive area of unfavourable habitat. Taken together, our findings suggest the genetic structure of this hybrid zone results from the interplay of historical (biogeographic) and population-level processes. The narrowness and coincidence of genetic clines can be explained by weak selection against hybrids and reflect a degree of reproductive isolation that is consistent with cryptic speciation.

7. Geographic patterns of stress in insular lizards reveal anthropogenic and climatic signatures

Abstract

Anthropization of insular ecosystems may have negative impacts on native populations of lizards, which provide core ecosystem services on islands. We aimed to identify environmental factors to explain the interlocal variation in faecal glucocorticoids, parasite intensity, and body condition in populations of insular lizards. A cross-sectional design during the summer of 2017 and 2018 was used to sample 611 adult lizards, Gallotia galloti. Interlocal variation of three stress indicators was analysed in response to environmental variables across a wide environmental gradient in Tenerife (Canary Islands): (i) concentration of faecal glucocorticoids, (ii) intensities of infection by hematic parasites, and (iii) body condition. The data, with low spatial autocorrelation, were analysed using multimodel inference and model cross-validation. Bioclimatic variables associated with the extreme hot and dry climate of summer were the most informative predictors. Interlocal variation in faecal corticosterone in males was best fitted to a model that included the maximum temperature of the warmest month, although the best predictor was habitat anthropization. The thermal annual range, associated with extreme thermal events, was positively related to faecal corticosterone in females. Extreme hot temperatures were positively related to the median parasite intensities in both sexes, while the highest mean intensities of infection were found in females from the most xeric coastal localities. None of the predictors tested, including faecal glucocorticoids, explained individual or interlocal variation in body condition. Effects of human pressure and climate change on insular populations of lizards can be additive. However, the uncoupled relationship found between body condition and the faecal glucocorticoid content suggests that current negative effects may be aggravated during drought periods in summer. Abstract

Accurate analyses of the diets of predators are key to understand trophic interactions and defining conservation strategies. Diets are commonly assessed through analysis of non‐invasively collected scats, and the use of faecal DNA (fDNA ) analysis can reduce the species misidentifications that could lead to biased ecological inference. We review the scientific literature since publication of the first paper on amplifying fDNA , in order to assess trends in the use of genetic non‐invasive sampling (gNIS ) for predator species identification in scat‐based diet studies of North American and European terrestrial mammalian carnivores (Carnivora). We quantify error rates in morphology‐based predator species identification. We then provide an overview of how applying gNIS would improve research on trophic interactions and other areas of carnivore ecology. We found that carnivore species identity was verified by using gNIS in only 8% of 400 studies of carnivore diets based on scats. The median percentage of false positives (i.e. samples wrongly identified as belonging to the target species) in morphology‐based studies was 18%, and was consistent regardless of species’ body size. We did not find an increasing trend in the use of gNIS over time, despite the existing technical capability to identify almost all carnivore species. New directions for fDNA studies include employing high‐throughput sequencing (HTS ) and DNA metabarcoding to identify the predator species, the individual predator, the entire assemblage of consumed items, and the microbiome of the predator and pathogens. We conclude that HTS protocols and metagenomic approaches hold great promise for elevating gNIS as a fundamental cornerstone for future research in ecology and conservation biology of mammals.

Abstract

Geographical Information Systems (GIS) are often used to assess and monitor the environmental impacts caused by mining activities. The aim of this work was to develop a new application to produce dynamic maps for monitoring the temperature variations in a self-burning coal waste pile, under a GIS open source environment—GIS-ECOAL (freely available). The performance of the application was evaluated with distributed temperature measurements gathered in the S. Pedro da Cova (Portugal) coal waste pile. In order to obtain the temperature data, an optical fiber cable was disposed over the affected area of the pile, with 42 location stakes acting as precisely-located control points for the temperature measurement. A monthly data set from July (15 min of interval) was fed into the application and a video composed by several layouts with temperature measurements was created allowing for recognizing two main areas with higher temperatures. The field observations also allow the identification of these zones; however, the identification of an area with higher temperatures in the top of the studied area was only possible through the visualization of the images created by this application. The generated videos make possible the dynamic and continuous visualization of the combustion process in the monitored area.

Abstract

Well-dispersed loads of powdered metals, metal oxides, several carbon allotropes, or nanoclaysare incorporated into polyamide 6 microcapsules (PAMC) via a one-step in situ fabricationtechnique. It is based on activated anionic polymerization (AAP) of e-caprolactam in ahydrocarbon solvent performed in the presence of therespective micro- or nanosized loads. The forming PAMCare with controllable shape, size and load content. Theresults obtained show that the melt processing of PAMCis a facile method to fabricate thermoplastic hybridswith high mechanical resistance and tailored electro-magnetic characteristics. Abstract

The role of natural selection in promoting reproductive isolation has received substantial renewed interest within the last two decades. As a consequence, the study of ecological speciation has become an extremely productive research area in modern evolutionary biology. Recent innovations in sequencing technologies offer an unprecedented opportunity to study the mechanisms involved in ecological speciation. Genome scans provide significant insights but have some important limitations; efforts are needed to integrate them with other approaches to make full use of the sequencing data deluge. An international conference 'Advances in Ecological Speciation' organized by the University of Porto (Portugal) aimed to review current progress in ecological speciation. Using some of the examples presented at the conference, we highlight the benefits of integrating ecological and genomic data and discuss different mechanisms of parallel evolution. Finally, future avenues of research are suggested to advance our knowledge concerning the role of natural selection in the establishment of reproductive isolation during ecological speciation. © 2013 John Wiley & Sons Ltd.

Abstract

3,4-dihydro-12-hydroxy-2,2-dimethyl-2H,6H-pyrano[3,2-b]xanthen-6-one (compound 1) is a cytotoxic dihydropyranoxanthone exhibiting antiproliferative effects, inducing S-phase cell cycle arrest, and increasing the percentage of apoptotic cells in leukemia cell lines. Nevertheless, the poor aqueous solubility of compound 1 is a major drawback not only for its potential use in therapy but even for the in vitro assessment of its biological activity. Polymeric nanoparticles formulations were developed as potential carriers to overcome problems related with low water solubility of compound 1. The objective of this work was to develop and validate a specific, sensitive and simple HPLC method for the quantitative analysis of the prenylated xanthone (compound 1), which was entrapped in PLGA nanoparticles for the first time.Chromatographic separation was performed with a reversed-phase C18 column, using methanol: water (85:15, v/v) containing 1 % (v/v) acetic acid as a mobile phase at a flow rate of 1 ml/min and quantification was made by UV detection at 254 nm. The isocratic system required 10 minutes of chromatographic run. The method was shown to be linear (r> 0.999) over the concentration range of 0.50-3.00 µg/ml and precise at the intra-day and inter-day levels as reflected by the relative standard deviation values (lower than 1.5% and 1.6%, respectively). The mean recovery ranged from 97.53 to 104.28 % (RSD: 0.027%) and from 98.49 to 101.81% (RSD: 0.019 %) for nanospheres and nanocapsules, respectively.A simple, linear, sensitive, accurate, and precise HPLC method suitable for the quantification of compound 1 incorporated in polymeric nanoparticles was developed and validated.

Abstract

The standard (p° = 0.1 MPa) molar enthalpies of formation, in the crystalline phase, of the cyclic anhydrides of glycine and DL-alanine, at T = 298.15 K, were derived from the standard molar energies of combustion in oxygen, measured by static bomb combustion calorimetry. For these compounds, the standard molar enthalpies of sublimation, at T = 298.15 K, were determined from the temperature-vapour pressure dependence, obtained by the Knudsen mass-loss effusion method. Through the values for the heat capacity differences between the gas and the crystalline phases of the studied compounds, the standard (po = 0.1 MPa) molar enthalpies, entropies and Gibbs energies of sublimation, at T = 298.15 K, were derived. From the experimental values, the standard molar enthalpies of formation, in the gaseous phase, at T = 298.15 K, of glycine anhydride and DL-alanine anhydride were calculated as -(321.0 ± 1.9) kJ·mol-1 and -(382.7 ± 2.0) kJ·mol-1, respectively. A theoretical study at the G3 and G4 levels has been carried out, and the calculated enthalpies of formation have been compared with the experimental values. © 2012 Elsevier Ltd. Abstract

The main objective of this report is to show the usefulness and versatility of the Mathematica program to simulate enzyme linear pathways and to depict the effect of changing the Vmax and/or Km values of one or more enzymes on the course of the reaction. In addition, analysis of the different types of inhibition of the first enzyme of the pathway by its end product is viewed with the reservoir model for enzyme kinetics. All the data shown here are quantitatively related to the kinetic constants of the implicated enzymes. Particular attention has been paid to calculate the time needed to achieve half of the possible total synthesis of the final product of a metabolic pathway. © 2013 Elsevier Ltd.

Abstract

The homeodomain transcription factor Prrxl1/DRG11 has emerged as a crucial molecule in the establishment of the pain circuitry, in particular spinal cord targeting of dorsal root ganglia (DRG) axons and differentiation of nociceptive glutamatergic spinal cord neurons. Despite Prrxl1 importance in the establishment of the DRG-spinal nociceptive circuit, the molecular mechanisms that regulate its expression along development remain largely unknown. Here, we show that Prrxl1 transcription is regulated by three alternative promoters (named P1, P2, and P3), which control the expression of three distinct Prrxl1 5′-UTR variants, named 5′-UTR-A, 5′-UTR-B, and 5′-UTR-C. These 5′-UTR sequences confer distinct mRNA stability and translation efficiency to the Prrxl1 transcript. The most conserved promoter (P3) contains a TATA-box and displays in vivo enhancer activity in a pattern that overlaps with the zebrafish Prrxl1 homologue, drgx. Regulatory modules present in this sequence were identified and characterized, including a binding site for Phox2b. Concomitantly, we demonstrate that zebrafish Phox2b is required for the expression of drgx in the facial, glossopharyngeal, and vagal cranial ganglia.

Abstract

Vertebrate odd-skipped related genes (Osr) have an essential function during the formation of the intermediate mesoderm (IM) and the kidney structures derived from it. Here, we show that these genes are also crucial for limb bud formation in the adjacent lateral plate mesoderm (LPM). Reduction of zebrafish Osr function impairs fin development by the failure of tbx5a maintenance in the developing pectoral fin bud. Osr morphant embryos show reduced wnt2b expression, and increasing Wnt signaling in Osr morphant embryos partially rescues tbx5a expression. Thus, Osr genes control limb bud development in a non-cell-autonomous manner, probably through the activation of Wnt2b. Finally, we demonstrate that Osr genes are downstream targets of retinoic acid (RA) signaling. Therefore, Osr genes act as a relay within the genetic cascade of fin bud formation: by controlling the expression of the signaling molecule Wnt2ba in the IM they play an essential function transmitting the RA signaling originated in the somites to the LPM.

Abstract

The tshz genes comprise a family of evolutionarily conserved transcription factors. However, despite the major role played by Drosophila tsh during the development of the fruit fly, the expression and function of other tshz genes have been analyzed in a very limited set of organisms and, therefore, our current knowledge of these genes is still fragmentary. In this study, we perform detailed phylogenetic analyses of the tshz genes, identify the members of this gene family in zebrafish and describe the developmental expressions of two of them, tshz2 and tshz3b, and compare them with meis1, meis2.1, meis2.2, pax6a, and pax6b expression patterns. The expression patterns of these genes define a complex set of coexpression domains in the developing zebrafish brain where their gene products have the potential to interact.

Abstract

The relative stabilities of 2,2′- and 3,3′-bithiophenes were evaluated by experimental thermochemistry and the results compared with data obtained from state of the art calculations, which were also extended to 2,3′bithiophene. The standard (p° = 0.1 MPa) molar enthalpies of formation of crystalline 2,2′-bithiophene and 3,3′bithiophene were calculated from the standard molar energies of combustion, in oxygen, to yield CO2 (g) and H2SO4 ·115H2O, measured by rotating-bomb combustion calorimetry at T = 298.15 K. The vapor pressures of these two compounds were measured as a function of temperature by Knudsen effusion massloss technique. The standard molar enthalpies of sublimation, at T = 298.15 K, were derived from the Clausius-Clapeyron equation. The experimental values were used to calculate the standard (p° = 0.1 MPa) enthalpies of formation of the title compounds in the gaseous phase; the results were analyzed and interpreted in terms of enthalpic increments and molecular structure. Standard ab initio molecular orbital calculations at the G3(MP2)//B3LYP level were performed. Enthalpies of formation, using homodesmotic reactions, were calculated and compared with experimental data. The computational study was also extended to the isomeric compound 2,3'-bithiophene. Detailed inspections of the molecular and electronic structures of the compounds studied were carried out. Finally, bond dissociation enthalpies (BDE) and enthalpies of formation of thienyl radicals were also computed. © 2009 American Chemical Society.

Abstract

Thermophysical and thermochemical studies have been carried out for crystalline parabanic acid. The thermophysical study was made by differential scanning calorimetry, DSC, over the temperature interval between T = (263 and 473) K. Two phase transitions were found: at T = (392.3 ± 1.6) K with the enthalpy of transition of (2.1 ± 0.4) kJ · mol−1 and at T = (509.8 ± 1.5) K, when the compound was scanned to its fusion temperature. The standard (p∘ = 0.1 MPa) molar enthalpy of formation, at T = 298.15 K, for crystalline parabanic acid was determined using static-bomb combustion calorimetry as −(590.2 ± 1.0) kJ · mol−1. The standard molar enthalpy of sublimation, at T = 298.15 K, was derived from the variation of their vapour pressures, measured by the Knudsen-effusion method, with the temperature. These two thermochemical parameters yielded the standard molar enthalpy of formation in the gaseous phase, at T = 298.15 K, as −(470.8 ± 1.2) kJ · mol−1.

Several solid–solid transitions have been detected in imidazolidin-2-one. The standard (p∘ = 0.1 MPa) molar enthalpies of formation, at T = 298.15 K, for crystalline imidazolidin-2-one and N,N′-trimethyleneurea [tetrahydropyrimidin-2(1H)-one], were determined using static-bomb combustion calorimetry. The standard molar enthalpies of sublimation, at T = 298.15 K, for the two compounds were derived from the variation of their vapour pressures, measured by the Knudsen effusion method, with the temperature. These two thermochemical parameters yielded the standard molar enthalpies of formation of the two cyclic urea compounds studied in the gaseous phase at T = 298.15 K. These values are discussed in terms of molecular structural contributions and interpreted on the bases of the “benzo-condensed effect” and of the ring strain of imidazolidin-2-one.

The enthalpies of combustion, vaporization, and sublimation were measured by rotating-bomb combustion calorimetry, Calvet microcalorimetry, correlation gas chromatography, and Knudsen effusion techniques and the gas-phase enthalpies of formation, at T = 298.15 K, were determined. Standard ab initio molecular orbital calculations at the G2 and G3 levels were performed, and a theoretical study on the molecular and electronic structures of the compounds studied has been conducted. Calculated enthalpies of formation using atomization and isodesmic reactions are compared with the experimental data. Experimental and theoretical results show that 2-acetylthiophene is thermodynamically more stable than the 3-isomer. A comparison of the substituent effect of the acetyl group in benzene and thiophene rings has been carried out.

Abstract

The standard (p = 0.1 MPa) molar energies of combustion in oxygen, at T = 298.15 K, of 1-naphthalenemethanol and 2-naphthalenemethanol were measured by static bomb calorimetry. The values of the standard molar enthalpies of sublimation, at T = 298.15 K, were obtained by Calvet microcalorimetry. Combining these results the standard molar enthalpies of formation of the compounds, in the gas phase, at T = 298.15 K, have been calculated. Additionally, ab initio second order Mller-Plesset calculations were also used at the MP2(FULL)/6-31G(d), MP2/6-31 + G(d), and MP2(FULL)/6-31 + G(d) levels. The calculation of the energies of appropriate isodesmic reactions allowed the estimation of the standard molar enthalpies of formation in the gas phase for the compounds. The experimental measurements and both sets of quantum chemical calculations were in good agreement for both isomers.

Abstract

Thiophene-based compounds have widespread use in modern drug design, biodiagnostics, electronic and optoelectronic devices, and conductive polymers. The present study reports an experimental and computational thermochemical study on the relative stabilities of 2- and 3-thiopheneacetic acid methyl esters. The enthalpies of combustion and vaporization were measured by a rotating-bomb combustion calorimeter, Calvet microcalorimetry, and correlation gas chromatography, and the gas-phase enthalpies of formation at T = 298.15 K were determined. Standard ab initio molecular orbital calculations at the G3 level were performed, and a theoretical study of the molecular and electronic structure of the compounds studied was carried out. Abstract

The ability to reengineer object-oriented legacy systems has become a vital matter in today’s software industry. Early adopters of the object-oriented programming paradigm are now facing the problems of transforming their object-oriented “legacy” systems into full-fledged frameworks. To address this problem, a series of workshops have been organised to set up a forum for exchanging experiences, discussing solutions, and exploring new ideas. Typically,these workshops are organised as satellite events of major software engineering conferences,such as ECOOP’97 [1], ESEC/FSE’97 [3], ECOOP’98 [7], ECOOP’99 [6],ESEC/FSE’99 [4], ECOOP’03 [2], ECOOP’04 [16]. The last of this series so far has been organised in conjunction with ECOOP’05 and this report summarises the key discussions and outcome of that workshop.8 As preparation to the workshop, participants were asked to submit a position paper which would help in steering the workshop discussions. Moreover, researchers working on dynamic analysis were invited to compare the results of their approaches by applying their tools on a common case (ArgoUML). As a result, we received 10 position papers, of which 9 authors were present during the workshop. Together with 3 organisers and 5 participants without position paper, the workshop numbered 17 participants. 34. Rumex induratus Leaves: Interesting Dietary Source of Potential Bioactive Compounds

Abstract

The phenolic compounds of Rumex induratus leaves were determined by HPLC-DAD-MS/MS-ESI. The results revealed a profile composed of 19 compounds: caffeoyl-hexoside, two p-coumaroyl-hexoside isomers, feruloyl-hexoside, sinapoyl-hexoside, 6-C-hexosyl-quercetin, 8-C-hexosyl-luteolin, 6-C-hexosyl-luteolin, 6-C-hexosyl-apigenin, 3-O-hexosyl-quercetin, 3-O-rutinosyl-quercetin, 7-O-hexosyl-diosmetin, 3-O-rutinosyl-isorhamnetin, 7-O-(acetyl)-pento-hexosyl-diosmetin, 6-C-hexosyl-genkwanin, and four unidentified O-glycosyl-C-glycosylflavones. The quantification of the identified phenolics by HPLC-DAD showed 6-C-hexosyl-luteolin as the main compound. Organic acid composition was determined by HPLC-UV, revealing a high content of oxalic acid. R. induratus was also investigated for its capacity to act as a scavenger of DPPH and superoxide radicals. Good antioxidative results were obtained against both radicals. Unlike other species of the genus Rumex, R. induratus did not present any anthraquinone derivative.

Abstract

Topical drug treatment aims at providing high concentrations of drugs at the site of application so as to avoid adverse systemic effects associated with oral administration. Smart polymers, or stimuli-responsive polymers, are able to respond to a stimulus by showing physical or chemical changes in their behaviour as, for example, the delivery of the drug carried by them. The thermo-responsive nature of Pluronic® F-127 (Basf, Ludwigshafen, Germany) makes it an excellent candidate for the delivery of drugs at various application sites. In recent years, PF-127, and later, Pluronic lecithin organogels (PLO), have attracted particular interest in the design of dermal and transdermal delivery systems with a view to promoting, improving or retarding drug permeation through the skin, bearing in mind that for topical delivery systems, accumulation in the skin with minimal permeation is desired, while for systemic delivery, the opposite behaviour is preferred. In this review, we discuss the properties and characteristics of PF-127 and Pluronic lecithin organogels (PLO), and present many examples and advantages of the application of these polymeric systems in topical and transdermal administration of drugs. Therefore, this study aimed to determine the primary structure and investigate peptides obtained from the cutaneous secretions of the amphibian, Leptodactylus vastus, as a source of bioactive molecules. The peptides obtained possessed the amino acid sequences, GVVDILKGAAKDLAGH and GVVDILKGAAKDLAGHLASKV, with monoisotopic masses of [M + H]± = 1563.8 Da and [M + H]± = 2062.4 Da, respectively. The molecules were characterized as peptides of the class of ocellatins and were named as Ocellatin-K1(1-16) and Ocellatin-K1(1-21). Functional analysis revealed that Ocellatin-K1(1-16) and Ocellatin-K1(1-21) showed weak antibacterial activity. However, treatment of mice with these ocellatins reduced the nitrite and malondialdehyde content. Moreover, superoxide dismutase enzymatic activity and glutathione concentration were increased in the hippocampus of mice. In addition, Ocellatin-K1(1-16) and Ocellatin-K1(1-21) were effective in impairing lipopolysaccharide (LPS)-induced reactive oxygen species (ROS) formation and NF-kB activation in living microglia. We incubated hippocampal neurons with microglial conditioned media treated with LPS and LPS in the presence of Ocellatin-K1(1-16) and Ocellatin-K1(1-21) and observed that both peptides reduced the oxidative stress in hippocampal neurons. Furthermore, these ocellatins demonstrated low cytotoxicity towards erythrocytes. Abstract

23 pags., 11 figs., 14 tabs., A small-scale ITC benchmarking study was performed involving 9 biophysics laboratories/facilities, to evaluate inter-laboratory and intra-laboratory basal levels of uncertainty. Our prime goal was to assess a number of important factors that can influence both the data gathered by this technique and the thermodynamic parameter values derived therefrom. In its first part, the study involved 5 laboratories and 13 different instruments, working with centrally prepared samples and the same experimental protocol. The second part involved 4 additional laboratories and 6 more instruments, where the users prepared their own samples according to provided instructions and did the experiments following the same protocol as in the first part. The study design comprised: (1) selecting a minimal set of laboratories; (2) providing very stable samples; (3) providing samples not requiring preparation or manipulation; and (4) providing a well-defined and detailed experimental protocol. Thus, we were able to assess: (i) the variability due to instrument and data analysis performed by each user on centrally prepared samples; (ii) the comparability of data retrieved when using 4 different software packages to analyze the same data, besides the data analysis carried out by the different users on their own experimental results; and (iii) the variability due to local sample preparation (second part of the study). Centro de Investigación en Sanidad Animal (CISA), Marine algae are recognised sources of bioactive compounds that have attracted great interest as nutritional supplements for aquaculture fish. Intensive rearing conditions often expose fish to husbandry-related stressors, rendering fish more susceptible to disease and reducing production yields. The present work evaluated the potential of two marine algae extracts (Fucus vesiculosus and Nannochloropsis gaditana) as nutritional supplements to mitigate stress effects in meagre (Argyrosomus regius) exposed to an acute handling stress (AS). A plant-based diet was used as a control, and three other diets were prepared, which were similar to the control diet but supplemented with 1% of each algal extract or a combination of the two extracts (0.5% each). The effects of supplemented diets on stress biomarkers, antioxidant enzyme activities, and immune response were analysed in fish exposed to AS after 4 weeks of feeding. Supplemented diets did not affect growth performance but the inclusion of F. vesiculosus promoted higher feed efficiency, as compared to the control group. Dietary algal extracts supplementation reduced plasma glucose levels, increased white blood cell counts, and reduced the expression of pro-inflammatory genes when compared with the control. N. gaditana supplementation led to a reduction in hepatic antioxidant enzyme activity and glutathione levels, while F. vesiculosus supplementation increased muscle glutathione reductase activity and reduced lipid peroxidation. These findings support the potential of algal extracts as nutraceuticals in aquafeeds to enhance the ability of fish to cope with husbandry-related stressful conditions and ultimately improve fish health and welfare., This research was funded by U. Abstract

Background Lung function is an important predictor of health and a marker of physical functioning at older ages. This study aimed to quantify the years of lung function lost according to disadvantaged socioeconomic conditions across the life-course. Methods This multicohort study used harmonised individual-level data from six European cohorts with information on life-course socioeconomic disadvantage and lung function assessed by forced expiratory volume in 1 s (FEV1) and forced vital capacity (FVC). 70 496 participants (51% female) aged 18–93 years were included. Socioeconomic disadvantage was measured in early life (low paternal occupational position), early adulthood (low educational level) and adulthood (low occupational position). Risk factors for poor lung function (e.g. smoking, obesity, sedentary behaviour, cardiovascular and respiratory diseases) were included as potential mediators. The years of lung function lost due to socioeconomic disadvantage were computed at each life stage. Results Socioeconomic disadvantage during the life-course was associated with a lower FEV1. By the age of 45 years, individuals experiencing disadvantaged socioeconomic conditions had lost 4–5 years of healthy lung function versus their more advantaged counterparts (low educational level −4.36 (95% CI −7.33–−2.37) for males and −5.14 (−10.32–−2.71) for females; low occupational position −5.62 (−7.98–−4.90) for males and −4.32 (−13.31–−2.27) for females), after accounting for the risk factors for lung function. By the ages of 65 years and 85 years, the years of lung function lost due to socioeconomic disadvantage decreased by 2–4 years, depending on the socioeconomic indicator. Sensitivity analysis using FVC yielded similar results to those using FEV1. Conclusion Life-course socioeconomic disadvantage is associated with lower lung function and predicts a significant number of years of lung function loss in adulthood and at older ages. Abstract

12 páginas, 9 figuras, 3 tablas., Prunus lusitanica L. is a paleotropical relic species with an Ibero-Maghrebian distribution, which is presently considered as an endangered species, recognized by the Natura 2000 Network (92/43/EEC) as a priority habitat for conservation in Europe. The mountains in the Portuguese mainland central region offer the best location for this species to occur. The main objective of this study is to measure the current conservation status of the communities of P. lusitanica, through the collection of field data, such as the number of existing individuals of each population and their location, which will then be comparatively analyzed based on the previous literature, published at least 15 years ago. Soil characterization analysis was carried out and the main threats to conservation were identified. As a result, a decline of approximately 40% was observed in the number of individuals and in the quality of their habitat. The main threats to their conservation were found to be the seasonal occurrence of rural fires and the expansion of invasive species, such as Acacia dealbata Link and Ailanthus altissima (Mill.) Swingle. Abstract

Several studies of lizards have made an erroneous interpretation of negative relationships between spectral brightness and parasite load, and thus provided misleading support for the Hamilton–Zuk hypothesis (HZH). The HZH predicts that infected hosts will produce poorer sexual ornamentation than uninfected individuals as a result of energetic trade-offs between immune and signalling functions. To test whether there is a negative relationship between spectral brightness and pigment content in the skin of lizards, we used spectrophotometry to quantify the changes in spectral brightness of colour patches of two species after chemically manipulating the contents of orange, yellow and black pigments in skin samples. Carotenoids were identified using high- performance liquid chromatography. In addition, we compared the spectral brightness in the colour patches of live individuals with differential expression of nuptial coloration. Overall, the analyses demonstrated that the more pigmented the colour patch, the darker the spectrum. We provide a comprehensive interpretation of how variation in pigment content affects the spectral brightness of the colour patches of lizards. Abstract

Anthropization of insular ecosystems may have negative impacts on native populations of lizards, which provide core ecosystem services on islands. We aimed to identify environmental factors to explain the interlocal variation in faecal glucocorticoids, parasite intensity, and body condition in populations of insular lizards. A cross-sectional design during the summer of 2017 and 2018 was used to sample 611 adult lizards, Gallotia galloti. Interlocal variation of three stress indicators was analysed in response to environmental variables across a wide environmental gradient in Tenerife (Canary Islands): (i) concentration of faecal glucocorticoids, (ii) intensities of infection by hematic parasites, and (iii) body condition. The data, with low spatial autocorrelation, were analysed using multimodel inference and model cross-validation. Bioclimatic variables associated with the extreme hot and dry climate of summer were the most informative predictors. Interlocal variation in faecal corticosterone in males was best fitted to a model that included the maximum temperature of the warmest month, although the best predictor was habitat anthropization. The thermal annual range, associated with extreme thermal events, was positively related to faecal corticosterone in females. Extreme hot temperatures were positively related to the median parasite intensities in both sexes, while the highest mean intensities of infection were found in females from the most xeric coastal localities. None of the predictors tested, including faecal glucocorticoids, explained individual or interlocal variation in body condition. Effects of human pressure and climate change on insular populations of lizards can be additive. However, the uncoupled relationship found between body condition and the faecal glucocorticoid content suggests that current negative effects may be aggravated during drought periods in summer. Abstract

The study of natural hybrid zones can illuminate aspects of lineage divergence and speciation in morphologically cryptic taxa. We studied a hybrid zone between two highly divergent but morphologically similar lineages (south-western and south-eastern) of the Iberian endemic Bosca's newt (Lissotriton boscai) in SW Iberia with a multilocus dataset (microsatellites, nuclear and mitochondrial genes). STRUCTURE and NEWHYBRIDS analyses retrieved few admixed individuals, which classified as backcrosses involving parental individuals of the south-western lineage. Our results show asymmetric introgression of mtDNA beyond the contact from this lineage into the south-eastern lineage. Analysis of nongeographic introgression patterns revealed asymmetries in the direction of introgression, but except for mtDNA, we did not find evidence for nonconcordant introgression patterns across nuclear loci. Analysis of a 150-km transect across the hybrid zone showed broadly coincident cline widths (ca. 3.2–27.9 km), and concordant cline centres across all markers, except for mtDNA that is displaced ca. 60 km northward. Results from ecological niche modelling show that the hybrid zone is in a climatically homogenous area with suitable habitat for the species, suggesting that contact between the two lineages is unlikely to occur further south as their distributions are currently separated by an extensive area of unfavourable habitat. Taken together, our findings suggest the genetic structure of this hybrid zone results from the interplay of historical (biogeographic) and population-level processes. Diets are commonly assessed through analysis of non‐invasively collected scats, and the use of faecal DNA (fDNA ) analysis can reduce the species misidentifications that could lead to biased ecological inference. We review the scientific literature since publication of the first paper on amplifying fDNA , in order to assess trends in the use of genetic non‐invasive sampling (gNIS ) for predator species identification in scat‐based diet studies of North American and European terrestrial mammalian carnivores (Carnivora). We quantify error rates in morphology‐based predator species identification. We then provide an overview of how applying gNIS would improve research on trophic interactions and other areas of carnivore ecology. We found that carnivore species identity was verified by using gNIS in only 8% of 400 studies of carnivore diets based on scats. The median percentage of false positives (i.e. samples wrongly identified as belonging to the target species) in morphology‐based studies was 18%, and was consistent regardless of species’ body size. We did not find an increasing trend in the use of gNIS over time, despite the existing technical capability to identify almost all carnivore species. New directions for fDNA studies include employing high‐throughput sequencing (HTS ) and DNA metabarcoding to identify the predator species, the individual predator, the entire assemblage of consumed items, and the microbiome of the predator and pathogens. Abstract

Collecting data on the localization of users is a key issue for the MASK (Mobile Airways Sentinel networK: the Allergy Diary) App. Data anonymization is a method of sanitization for privacy. The European Commission's Article 29 Working Party stated that geolocation information is personal data.To assess geolocation using the MASK method and to compare two anonymization methods in the MASK database to find an optimal privacy method., Background Collecting data on the localization of users is a key issue for the MASK (Mobile Airways Sentinel networK: the Allergy Diary) App. Data anonymization is a method of sanitization for privacy. The European Commission’s Article 29 Working Party stated that geolocation information is personal data. To assess geolocation using the MASK method and to compare two anonymization methods in the MASK database to find an optimal privacy method. Methods Geolocation was studied for all people who used the Allergy Diary App from December 2015 to November 2017 and who reported medical outcomes. Two different anonymization methods have been evaluated: Noise addition (randomization) and k-anonymity (generalization). Results Ninety-three thousand one hundred and sixteen days of VAS were collected from 8535 users and 54,500 (58.5%) were geolocalized, corresponding to 5428 users. Noise addition was found to be less accurate than k-anonymity using MASK data to protect the users’ life privacy. Abstract

6 pags., 2 figs., 3 tabs., 2 schs., We report a study on the energetics and structural properties of gallic (1) and ellagic (2) acids. The experimental values of standard enthalpy of formation in solid state at 298.15 K, ΔH (cd) of 1 as (−985.0 ± 2.9 kJ·mol) and 2 as (−1377.9 ± 4.7 kJ·mol) have been determined. The vapour pressure of 1 have been measure by Knudsen effusion methodology and the derived enthalpy of sublimation, Δ H , was combined with the ΔH (cd) in order to derive its gas-phase enthalpy of formation, ΔH (1,g) = −835.7 ± 4.0 kJ·mol. Quantum chemical calculations, at DFT (M05-2X) and composite ab initio Gn (n = 3, 4) levels of theory, provided the consistency of the experimental results and a plausible estimation of ΔH (g) of 2 as (−1128.6 ± 6.4 kJ·mol), which was deduced from the isodesmic-reactions methodology., J.Z.D acknowledges the support funding to IQFR-CSIC (Spain) andperuvian CONCYTEC (INNÓVATE PERÚ, ECIP-1-P-030-14 Grant). Abstract

Background: Change in the prevalence of raised blood pressure could be due to both shifts in the entire distribution of blood pressure (representing the combined effects of public health interventions and secular trends) and changes in its high-blood-pressure tail (representing successful clinical interventions to control blood pressure in the hypertensive population). Our aim was to quantify the contributions of these two phenomena to the worldwide trends in the prevalence of raised blood pressure. Methods: We pooled 1018 population-based studies with blood pressure measurements on 88.6 million participants from 1985 to 2016. We first calculated mean systolic blood pressure (SBP), mean diastolic blood pressure (DBP) and prevalence of raised blood pressure by sex and 10-year age group from 20-29 years to 70-79 years in each study, taking into account complex survey design and survey sample weights, where relevant. We used a linear mixed effect model to quantify the association between (probittransformed) prevalence of raised blood pressure and age-group- and sex-specific mean blood pressure. We calculated the contributions of change in mean SBP and DBP, and of change in the prevalence-mean association, to the change in prevalence of raised blood pressure. Results: In 2005-16, at the same level of population mean SBP and DBP, men and women in South Asia and in Central Asia, the Middle East and North Africa would have the highest prevalence of raised blood pressure, and men and women in the highincome Asia Pacific and high-income Western regions would have the lowest. In most region-sex-age groups where the prevalence of raised blood pressure declined, one half or more of the decline was due to the decline in mean blood pressure. Where prevalence of raised blood pressure has increased, the change was entirely driven by increasing mean blood pressure, offset partly by the change in the prevalence-mean association. Conclusions: Change in mean blood pressure is the main driver of the worldwide change in the prevalence of raised blood pressure, but change in the high-blood-pressure tail of the distribution has also contributed to the change in prevalence, especially in older age groups. © The Author(s) 2018.
