- Abstract
Introduction: Knowledge on the human gut microbiota in health and disease continues to rapidly expand. In recent years, changes in the gut microbiota composition have been reported as a part of the pathology in numerous neurodegenerative diseases. Bacterial extracellular vesicles (EVs) have been suggested as a novel mechanism for the crosstalk between the brain and gut microbiota, physiologically connecting the observed changes in the brain to gut microbiota dysbiosis. Methods: Publications reporting findings on bacterial EVs passage through the blood–brain barrier were identified in PubMed and Scopus databases. Results: The literature search yielded 138 non-duplicate publications, from which 113 records were excluded in title and abstract screening step. From 25 publications subjected to full-text screening, 8 were excluded. The resulting 17 publications were considered for the review. Discussion: Bacterial EVs have been described with capability to cross the blood–brain barrier, but the mechanisms behind the crossing remain largely unknown. Importantly, very little data exists in this context on EVs secreted by the human gut microbiota. This systematic review summarizes the present evidence of bacterial EVs crossing the blood–brain barrier and highlights the importance of future research on gut microbiota-derived EVs in the context of gut-brain communication across the blood–brain barrier.

- Abstract
Purpose: To report clinical features and potential disease markers of inherited retinal dystrophy (IRD) caused by the biallelic c.148delG variant in the tubby-like protein 1 (TULP1) gene. Methods: A retrospective observational study of 16 IRD patients carrying a homozygous pathogenic TULP1 c.148delG variant. Clinical data including fundus spectral-domain optical coherence tomography (SD-OCT) were assessed. A meta-analysis of visual acuity of previously reported other pathogenic TULP1 variants was performed for reference. Results: The biallelic TULP1 variant c.148delG was associated with infantile and early childhood onset IRD. Retinal ophthalmoscopy was primarily normal converting to peripheral pigmentary retinopathy and maculopathy characterized by progressive extra-foveal loss of the ellipsoid zone (EZ), the outer plexiform layer (OPL), and the outer nuclear layer (ONL) bands in the SD-OCT images. The horizontal width of the foveal EZ showed significant regression with the best-corrected visual acuity (BCVA) of the eye (p < 0.0001, R2 = 0.541, F = 26.0), the age of the patient (p < 0.0001, R2 = 0.433, F = 16.8), and mild correlation with the foveal OPL-ONL thickness (p = 0.014, R2 = 0.245, F = 7.2). Modelling of the BCVA data suggested a mean annual loss of logMAR 0.027. The level of visual loss was similar to that previously reported in patients carrying other truncating TULP1 variants. Conclusions: This study describes the progression of TULP1 IRD suggesting a potential time window for therapeutic interventions. The width of the foveal EZ and the thickness of the foveal OPL-ONL layers could serve as biomarkers of the disease stage.

7. Delivery mode and perinatal antibiotics influence the infant gut bacteriome and mycobiome:a network analysis

- Abstract
Both exposure to antibiotics at birth and delivery via Caesarean section influence the gut bacteriome’s development in infants. Using 16S rRNA and internal transcribed spacer sequencing on the Ion Torrent platform, we employed network analysis to investigate the bacterial and fungal interkingdom relationships in the gut microbiome from birth to age 18 months in a prospective cohort study of 140 infants. The gut microbiome at ages six and 18 months revealed distinctive microbial interactions, including both positive and negative associations between bacterial and fungal genera in the gut ecosystem. Perinatal factors, delivery mode and intrapartum antibiotic exposure affected the associations between bacterial and fungal species. In infants exposed and unexposed to perinatal antibiotics, the gut microbiome formed distinct networks for the bacteriome and mycobiome. The fungi Saccharomyces, Trichosporon, Pezoloma, Cystofilobasidium, Rigidoporus and Fomitopsis were strongly associated with exposure to antibiotics at birth. Hyaloscypha, Trichosporon, Fomitopsis and Vishniacozyma were strongly associated with the control group that was not exposed to antibiotics. Five distinct networks were formed according to delivery mode. The present study confirms that bacteria and fungi clearly interact in the infant gut ecosystem. Furthermore, perinatal factors appear to influence the relationships between bacteria and fungi in the developing gut microbiome.

8. The bacteriome, mycobiome and extracellular vesicles of the neonatal and infant gut microbiota

- Abstract
The human gut microbiota is comprised of a variety of microbes, such as bacteria, fungi, viruses, and archaea. Furthermore, the gut contains bacterial extracellular vesicles (bEVs), small particles secreted by bacteria. The health effects of the gut microbiota have been explored but it is yet unclear when the gut microbiota starts developing. The womb was earlier assumed to be sterile, but recent studies imply that the fetus may already be exposed to microbes in the womb. Colonization of the gut is usually assessed from the first-pass meconium, the first stool passed after birth, which is assumed to serve as a proxy for the fetal gut microbiota. This cohort study of 513 mother-infant pairs was undertaken to elucidate the timing of gut microbiota development and factors affecting it. The first-pass meconium and later stool samples were collected from the infants and placenta and amniotic fluid from the mothers. 16S rRNA gene sequencing was performed to characterize the bacteriome and bEVs in the samples and internal transcribed spacer (ITS) gene sequencing to characterize the mycobiome. Questionnaires were used to collect information from the families on environmental and lifestyle conditions during pregnancy in order to assess the effect of prenatal factors on the microbial composition of the initial gut microbiota. A detectable meconium bacteriome and mycobiome were characterized, but no detectable placenta or amniotic fluid microbiota could be identified. The meconium microbiota was mainly affected by birth factors, including the mode of delivery and use of intrapartum antibiotics. Prenatal factors had little effect on the composition of the meconium bacteriome. The bEVs characterized in the meconium were found to be unaffected by the birth factors. Birth factors affected gut mycobiome development in the infants at least until the age of 18 months. It is concluded that the neonatal gut is probably colonized during birth, and that the birth factors influen, Tiivistelmä Ihmisen suoliston mikrobiomi koostuu monenlaisista mikrobeista, kuten bakteereista, sienistä, viruksista ja arkeista. Lisäksi suolisto sisältää bakteeriperäisiä ekstrasellulaarisia vesikkeleitä (bEV), jotka ovat pieniä partikkeleita, joita bakteerit erittävät. Suoliston mikrobiomin terveysvaikutuksia on tutkittu jo laajasti, mutta suoliston mikrobiomin alkuperää ei vielä tunneta tarkkaan. Aikaisemmin kohtua on pidetty sikiölle steriilinä kasvuympäristönä, mutta viimeaikainen tutkimus osoittaa, että sikiö saattaa altistua mikrobeille kohdussa. 25 páginas, 6 figuras, 2 tablas, Characterization of the genetic landscape of Alzheimer's disease (AD) and related dementias (ADD) provides a unique opportunity for a better understanding of the associated pathophysiological processes. We performed a two-stage genome-wide association study totaling 111,326 clinically diagnosed/'proxy' AD cases and 677,663 controls. We found 75 risk loci, of which 42 were new at the time of analysis. Pathway enrichment analyses confirmed the involvement of amyloid/tau pathways and highlighted microglia implication. Gene prioritization in the new loci identified 31 genes that were suggestive of new genetically associated processes, including the tumor necrosis factor alpha pathway through the linear ubiquitin chain assembly complex. We also built a new genetic risk score associated with the risk of future AD/dementia or progression from mild cognitive impairment to AD/dementia. The improvement in prediction led to a 1.6- to 1.9-fold increase in AD risk from the lowest to the highest decile, in addition to effects of age and the APOE ε4 allele., This work was funded by a grant (EADB) from the EU Joint Programme – Neurodegenerative Disease Research. This manuscript reports results of a study using Net Impact Method to quantify CSP and approximates corporate financial performance (CFP). We report findings on the CFP-CSP relationship and consider temporal lags between CFP measures and CSP impacts. Our findings are mostly supporting existing research but contrary to earlier research we find non-linear dynamics between CFP and CSP measures on some accounts. Tiivistelmä. Tässä pro gradu -tutkielmassa tarkastellaan maisterivaiheen luokanopettajaopiskelijoiden määritelmiä lahjakkuudelle. Lisäksi tutkielmassa tarkastellaan vastaajien antamien lahjakkuuden määritelmien yhteyttä tutkielmassa esiteltyihin lahjakkuusteorioihin ja -malleihin sekä tarkastellaan sitä, millainen rooli vastaajien käymällä luokanopettajakoulutuksella on ollut heidän antamiensa määritelmien muodostumisessa. Tutkimus on luonteeltaan kvalitatiivinen tutkimus ja lähestymistavaltaan fenomenografinen. Tutkimuksen aineistonkeruu toteutettiin sähköisellä kyselylomakkeella, jota jaettiin Oulun yliopiston kasvatustieteiden tiedekunnan luokanopettajakoulutusta ja siihen liittyviä asioita koskevalla sähköpostilistalla. Vastaajiksi tutkimukseen kertyi yhteensä 17 maisterivaiheen luokanopettajaopiskelijaa, joista jokainen opiskelee Oulun yliopiston kasvatustieteiden tiedekunnassa. Sähköisellä kyselylomakkeella kartoitettiin vastaajien määritelmiä lahjakkuudelle, määritelmien taustalla mahdollisesti vaikuttavia lahjakkuusteorioita ja -malleja sekä sitä, millä tavalla lahjakkuuden aihepiiriä on käsitelty luokanopettajakoulutuksessa, ja kuinka koulutuksen sisällöt ovat vaikuttaneet lahjakkuudelle annettujen määritelmien muodostumiseen. Saatuja vastauksia analysoitiin teoriaohjaavan sisällönanalyysin keinoin. Tutkimuksessa havaittiin, että vastaajien määritelmät lahjakkuudelle ovat hyvin monipuolisia ja sisältävät runsaasti yhtäläisyyksiä toisiinsa nähden. Lahjakkuutta määritellään hyvin käytännönläheisesti esimerkiksi erilaisten taitojen ja kykyjen sekä tekemisen ja oppimisen helppouden kautta. Lisäksi vastaajat näkivät ympäristötekijöiden, perintötekijöiden, sekä sisäisten tekijöiden vaikuttavan lahjakkuuteen. Tarkasteltaessa määritelmien yhteyksiä erilaisiin tutkimuksessa esiteltyihin lahjakkuusteorioihin ja -malleihin, korostui esitellyistä malleista etenkin Gagnén DMGT-malli (2004), vaikka vastaajat eivät kyenneetkään itse nimeämään määritelmiensä taustalla vaikuttavia teoreettisia malleja tai näkemyksiä. Luokanopettajakoulutuksen ei myöskään nähty käsittelevän lahjakkuuden aihepiiriä juurikaan, ja noin puolet vastaajista koki, että heidän määritelmänsä lahjakkuudelle eivät olleet muuttuneet koulutuksen aikana. Tässä tutkielmassa ilmenneet lahjakkuuden määritelmät sisältävät samanlaisia piirteitä kuin tutkimuksissa, joissa on kartoitettu suomalaisten opettajien määritelmiä lahjakkuudelle. Lisäksi tutkimuksessa todettiin, että lahjakkuuden aihepiiriä ei käsitellä juurikaan vastaajien käymässä luokanopettajakoulutuksessa. Samankaltaisia viitteitä aihepiirin vähäisestä käsittelystä on esitetty myös muissa lahjakkuuden aihepiiriä Suomessa tarkastelevissa tutkimuksissa.

- Abstract
Purpose In order to support people with low back pain (LBP) to stay at work, work arrangements are regarded important. This study aimed to evaluate the effectiveness of a workplace intervention using a participatory approach on work disability of workers with ongoing or recurrent LBP. Methods A total of 107 workers with LBP, with duration of pain for at least two consecutive weeks or recurrent pain of any duration during the last year, were randomized either to the intervention (n = 51) or control group (n = 56). The intervention included arrangements at the workplace, along with individual guidance provided by an occupational physiotherapist (OPT). The randomized intervention study used standard counselling and guidance by an OPT without workplace intervention as a comparison. Surveys were completed at baseline, and 6 and 12 months after baseline. Results There were no statistically significant differences between the intervention and control groups on the primary outcome measure, i.e. self-assessed work ability. We found no between-group differences in perceived health, self-assessed work productivity, number of sickness absence days and severity of back pain. However, there were significant positive within-group changes in the intervention group in the intensity of LBP, perceived health and the number of sickness absence days due to LBP. Conclusion Workplace arrangements are feasible using participatory ergonomics, but more quantitative and qualitative research is needed on its utilization and effectiveness among workers with LBP.

- Abstract
Background: Bacterial extracellular vesicles (EVs) are more likely to cross biological barriers than whole-cell bacteria. We previously observed EV-sized particles by electron microscopy in the first-pass meconium of newborn infants. We hypothesized that EVs may be of bacterial origin and represent a novel entity in the human microbiome during fetal and perinatal periods. Methods: We extracted EVs from first-pass meconium samples of 17 newborn infants and performed bacterial 16S rRNA gene sequencing of the vesicles. We compared the EV content from the meconium samples of infants based on the delivery mode, and in vaginal delivery samples, based on the usage of intrapartum antibiotics. Results: We found bacterial EVs in all first-pass meconium samples. All EV samples had bacterial RNA. Most of the phyla present in the samples were Firmicutes (62%), Actinobacteriota (18%), Proteobacteria (10%), and Bacteroidota (7.3%). The most abundant genera were Streptococcus (21%) and Staphylococcus (17%). The differences between the delivery mode and exposure to antibiotics were not statistically significant. Conclusions: Bacterial EVs were present in the first-pass meconium of newborn infants. Bacterial EVs may represent an important novel feature of the gut microbiome during fetal and perinatal periods.

Introduction: An increasing number of children and adolescents divide their time between their separated parents' homes. Although marital conflict is disadvantageous for children's sleep, little is known about how children of separated parents sleep. The objective was to investigate the association between children's custody arrangements and sleep habits and sleep initiation difficulties. Materials and Methods: Cross-sectional questionnaire data from the 2013 Health Behaviours of School-aged Children was used. The sample included over 7000 adolescents (50% girls), aged 11-15. Nuclear families were used as a reference in all analyses. The sleep issues were defined as follows: Less than 7 h of sleep = insufficient sleep; sleep initiation difficulties >1 per week = insomnia; bedtimes after 11 pm = late bedtimes; more than 2h variability between weekend and weekday bedtimes = jetlag. Short sleep duration, insomnia, late bedtimes and jetlag were respectively used as outcomes from regression analyses where custody forms, gender, and family affluence were used as predictors. Results: The results show differences by custody arrangement, but they are not uniform across the dependent variables. Children and adolescents in sole maternal custody were less likely to sleep as much as recommended (P < .001), more likely to have late bedtimes (P < .001), report sleep initiation difficulties (P < .01) and to report social jetlag between school mornings and weekends (P < .05) compared to those in 2-parent families. Shared physical custody was associated with a higher likelihood of late bedtimes (P < .05) and sleep initiation difficulties (P < .05) compared to those in 2-parent families, but not of sleeping less than recommended or reporting social jetlag. Less-than-equal sharing was generally associated with worse sleep than in 2-parent families. Autosomal genetic analyses of blood lipids have yielded key insights for coronary heart disease (CHD). However, X chromosome genetic variation is understudied for blood lipids in large sample sizes. We now analyze genetic and blood lipid data in a high-coverage whole X chromosome sequencing study of 65,322 multi-ancestry participants and perform replication among 456,893 European participants. Common alleles on chromosome Xq23 are strongly associated with reduced total cholesterol, LDL cholesterol, and triglycerides (min P = 8.5 × 10−72), with similar effects for males and females. Chromosome Xq23 lipid-lowering alleles are associated with reduced odds for CHD among 42,545 cases and 591,247 controls (P = 1.7 × 10−4), and reduced odds for diabetes mellitus type 2 among 54,095 cases and 573,885 controls (P = 1.4 × 10−5). Although we observe an association with increased BMI, waist-to-hip ratio adjusted for BMI is reduced, bioimpedance analyses indicate increased gluteofemoral fat, and abdominal MRI analyses indicate reduced visceral adiposity. Co-localization analyses strongly correlate increased CHRDL1 gene expression, particularly in adipose tissue, with reduced concentrations of blood lipids.

33. Impact of Streptococcus salivarius K12 on nasopharyngeal and saliva microbiome:a randomized controlled trial

Background: Probiotic lactobacilli have been ineffective in preventing acute otitis media. In contrast to lactobacilli, alpha-hemolytic streptococci belong to the core microbiome of nasopharynx. Methods: We investigated the effects of Streptococcus salivarius K12 probiotic on the saliva and nasopharyngeal microbiome in 121 children attending daycare. Children were randomly allocated to receive oral K12 product for 1 month or no treatment. We obtained saliva and nasopharyngeal samples at study entry, at 1 and 2 months. The next-generation sequencing of the bacterial 16S gene was performed. Results: After the intervention, the diversity of saliva or nasopharyngeal microbiome did not differ between groups. The proportion of children with any otopathogen did not differ between the groups. At 1 month, the abundance of otopathogens in nasopharynx was lower in K12 group compared with that in control children (34% vs. 55%, P = 0.037). When we compared each otopathogen separately, Moraxella was the only group lower in the treatment group. We could not verify the reduction of Moraxella when an alternative Human Oral Microbiome Database taxonomy database was used. In children receiving K12 product, the mean abundance of S. salivarius was greater in saliva after the intervention (0.9% vs. 2.0%, P = 0.009). Conclusions: The use of S. salivarius K12 probiotic appeared to be safe because it did not disrupt the normal microbiome in young children. Even though a short-term colonization of S. salivarius was observed in the saliva, the impact of S. salivarius K12 probiotic on the otopathogens in nasopharyngeal microbiome remained uncertain.

We derive explicit analytical relations to describe paraxial light beams that represent a particular case of the hypergeometric (HyG) laser beams [J. Opt. Soc. Am. A 25, 262-270 (2008)]. Among these are modified quadratic Bessel--Gaussian beams, hollow Gaussian optical vortices, modified elegant Laguerre--Gaussian beams, and gamma-HyG beams. Using e-beam microlithography, a binary diffractive optical element capable of producing near-HyG beams is synthesized. Theory and experiment are in sufficient agreement. We experimentally demonstrate the ability to rotate dielectric microparticles using the bright diffraction ring of a HyG beam. OCIS codes: 050.4865, 030.4070, 070.2580.

- Abstract
Tiivistelmä. Tässä kandidaatin tutkielmassa tarkastellaan lahjakkaiden oppilaiden asemaa inklusiivisessa perusopetuksessa sekä erilaisia lahjakkaiden oppilaiden opettamiseen tarkoitettuja toimintamalleja inkluusioperiaatteen mukaisessa toimintaympäristössä. Aiheen käsittely on rajattu suomalaisen perusopetuksen viitekehykseen, mutta lahjakkuuden ja inkluusion käsitteet pohjautuvat kansainvälisiin ja yleisesti viitattuihin tutkimuksiin aiheesta. Suomalaisessa perusopetuksessa ei ole olemassa selkeitä malleja lahjakkuuden käsitteelle tai lahjakkaiden oppilaiden tunnistamiselle. Lahjakkuutta ja siihen liitettäviä ominaisuuksia on pyritty määrittelemään kasvatustieteen kentällä vuosikymmenien saatossa useilla erilaisilla teoreettisilla malleilla, joiden kautta käytännön opetusta on pyritty järjestämään siten, että se palvelisi myös lahjakkaita oppilaita. Lahjakkuuteen usein liitettäviä piirteitä peruskoulumaailmassa ovat keskiverto-oppilasta parempi taitotaso, aito kiinnostus ja keskittyminen aiheeseen sekä luovuus ja ongelmanratkaisukyky, joka mahdollistaa opitun tiedon soveltamisen. Suomalaisessa perusopetuksessa lahjakkaiden oppilaiden opetuksen toimintamallit ilmenevät selkeimmin suuntauksessa opetuksen ja opetustavoitteiden yksilöllistämiseen ja oppiainekohtaisiin eriyttämiskeinoihin, joilla pyritään varmistamaan jokaiselle oppilaalle hänen oppimistaan tukevat ja motivoivat tehtävät. Opetuksen eriyttäminen vaatii yksittäiseltä opettajalta entistä enemmän resursseja ja aikaa, joille voi olla hankalaa löytää tilaa nykymuotoisessa perusopetuksessa, jossa ryhmäkoot ovat kasvussa ja luokassa työskentelee inkluusioperiaatteen myötä entistä monimuotoisempi joukko oppilaita. Keskustelu opetuksen eriyttämisestä keskittyy useimmiten niiden oppilaiden tukemiseen, joilla on haasteita käsiteltävän asian oppimisessa tai esimerkiksi tiedossa olevia diagnosoituja oppimisvaikeuksia. Inkluusioperiaatteen mukaisesti jokaisen oppilaan tulisi saada juuri hänen yksilöllistä oppimistaan tukevaa opetusta. Tässä tutkielmassa tarkastellaan sitä, millaisessa asemassa lahjakkaat oppilaat ovat inklusiivisessa perusopetuksessa niin oppimisen eriyttämisen kuin muiden inkluusioperiaatteiden mukaisten keinojen suhteen.

- Abstract
We critically evaluated the fetal microbiome concept in 44 neonates with placenta, amniotic fluid, and first-pass meconium samples. Placental histology showed no signs of inflammation. Meconium samples were more often bacterial culture positive after vaginal delivery. In next-generation sequencing of the bacterial 16S gene, before and after removal of extracellular and PCR contaminant DNA, the median number of reads was low in placenta (48) and amniotic fluid (46) and high in meconium samples (14,556 C-section, 24,860 vaginal). In electron microscopy, meconium samples showed extracellular vesicles. Utilizing the analysis of composition of microbiomes (ANCOM) against water, meconium samples had a higher relative abundance of Firmicutes, Lactobacillus, Streptococcus, and Escherichia-Shigella. Our results did not support the existence of the placenta and amniotic fluid microbiota in healthy pregnancies. The first-pass meconium samples, formed in utero, appeared to harbor a microbiome that may be explained by perinatal colonization or intrauterine colonization via bacterial extracellular vesicles.

- Abstract
Degradation of freshwater ecosystems requires efficient tools for assessing the ecological status of freshwater biota and identifying potential cause(s) for their biological degradation. While diatoms and macroinvertebrates are widely used in stream bioassessment, the potential utility of microbial communities has not been fully harnessed. Using data from 113 Finnish streams, we assessed the performance of aquatic leaf-associated fungal decomposers, relative to benthic macroinvertebrates and diatoms, in modelling-based bioassessment. We built multi-taxon niche -type predictive models for fungal assemblages by using genus-based and sequence-based identification levels. We then compared the models’ precision and accuracy in the prediction of reference conditions (number of native taxa) to corresponding models for macroinvertebrates and diatoms. Genus-based fungal model nearly equalled the accuracy and precision of our best model (macroinvertebrates), whereas the sequence-based model was less accurate and tended to overestimate the number of taxa. However, when the models were applied to streams disturbed by anthropogenic stressors (nutrient enrichment, sedimentation and acidification), alone or in combination, the sequence-based fungal assemblages were more sensitive than other taxonomic groups, especially when multiple stressors were present. Microbial leaf decomposition rates were elevated in sediment-stressed streams whereas decomposition attributable to leaf-shredding macroinvertebrates was accelerated by nutrients and decelerated by sedimentation. Comparison of leaf decomposition results to model output suggested that leaf decomposition rates do not detect effectively the presence of multiple simultaneous disturbances. The rapid development of global microbial database may soon enable species-level identification of leaf-associated fungi, facilitating a more precise and accurate modelling of reference conditions in streams using fungal communities.
