Abstract

Recent studies from several groups have indicated that abnormal or ectopic expression and function of adrenal receptors for various hormones may regulate cortisol production in ACTH-independent hypercortisolism. Gastric inhibitory polypeptide (GIP)-dependent Cushing's syndrome has been described in patients with either unilateral adenoma or bilateral macronodular adrenal hyperplasia; this syndrome results from the large adrenal overexpression of the GIP receptor without any activating mutation. We have conducted a systematic in vivo evaluation of patients with adrenal Cushing's syndrome in order to identify the presence of abnormal hormone receptors. In macronodular adrenal hyperplasia, we have identified, in addition to GIP-dependent Cushing's syndrome, other patients in whom cortisol production was regulated abnormally by vasopressin, ß-adrenergic receptor agonists, hCG/LH, or serotonin 5HT-4 receptor agonists. In patients with unilateral adrenal adenoma, the abnormal expression or function of GIP or vasopressin receptor has been found, but the presence of ectopic or abnormal hormone receptors appears to be less prevalent than in macronodular adrenal hyperplasia. The identification of the presence of an abnormal adrenal receptor offers the possibility of a new pharmacological approach to control hypercortisolism by suppressing the endogenous ligands or by using specific antagonists for the abnormal receptors.

Abstract

Time-resolved vibrational spectroscopy constitutes an invaluable experimental tool for monitoring hot-carrier induced surface reactions. However, the absence of a full understanding on the precise microscopic mechanisms causing the transient spectral changes has been limiting its applicability. Here we introduce a robust first-principles theoretical framework that successfully explains both the nonthermal frequency and linewidth changes of the CO internal stretch mode on Cu(100) induced by femtosecond laser pulses. Two distinct processes engender the changes: electron-hole pair excitations underlie the nonthermal frequency shifts, while electron-mediated vibrational mode coupling gives rise to linewidth changes. Abstract

Climate change is expected to alter the input of nitrogen (N) sources in the Eastern Canadian Arctic Archipelago and Baffin Bay due to increased discharge from glacial meltwater and permafrost thaw. Since dissolved inorganic N is generally depleted in surface waters, dissolved organic N (DON) could represent a significant N source fueling phytoplankton activity in Arctic ecosystems. Yet, few DON data for this region exist. We measured concentrations and stable isotope ratios of DON (δ15N) and nitrate (NO3−; δ15N and δ18O) to investigate the sources and cycling of dissolved nitrogen in regional rivers and marine samples collected in the Eastern Canadian Arctic Archipelago and Baffin Bay during the summer of 2019. The isotopic signatures of NO3− in rivers could be reproduced in a steady state isotopic model by invoking mixing between atmospheric NO3− and nitrified ammonium as well as NO3− assimilation by phytoplankton. DON concentrations were low in most rivers (≤4.9 μmol N L−1), whereas the concentrations (0.54–12 μmol N L−1) and δ15N of DON (−0.71–9.6‰) at the sea surface were variable among stations, suggesting dynamic cycling and/or distinctive sources. In two regions with high chlorophyll‐a, DON concentrations were inversely correlated with chlorophyll‐a and the δ15N of DON, suggesting net DON consumption in localized phytoplankton blooms. We derived an isotope effect of 6.9‰ for DON consumption. Our data helps establish a baseline to assess future changes in the nutrient regime for this climate‐sensitive region. Plain Language Summary: Primary productivity in the Arctic Ocean surface waters is limited by nitrogen supply. We investigated dissolved inorganic and organic nitrogen dynamics in the Eastern Canadian Arctic Archipelago and Baffin Bay surface ocean waters as well as adjacent rivers. We used the isotopic composition (N and O) of both dissolved organic and inorganic nitrogen (DON and DIN, respectively) to explore the sources and transformations of nitrogen. Nitrate in rivers was from both the atmosphere and nitrified ammonium. Nitrate was also consumed by phytoplankton. DON concentrations were low in rivers compared with inorganic nitrogen (i.e., nitrate). This observation contrasts with previous data collected in the Eurasian and U.S. western coastal Arctic Ocean, where rivers represent a significant source of DON to the coastal ocean. We observed variable DON concentrations and isotopic composition in the Eastern Canadian Arctic Archipelago and Baffin Bay surface ocean waters, suggesting different sources and/or a dynamic DON cycling. We additionally found evidence for DON consumption in regions of highest chlorophyll‐a. These data are important to better understand how Greenland's melting ice sheet will impact nutrient delivery and primary productivity in the region. Abstract

Inferences on soil biogeochemical processes based on metagenomic profiles is a challenging task due to enormous diversity of soil microbes and the fragile linkage between gene abundance and functioning. Here we used the biological sink of H2 as a case study to test the hypothesis that [NiFe]-hydrogenase gene distribution and expression profiles explain variations in H2 oxidation rate measured in soil collected in poplar monoculture, larch plantation and farmland. Shotgun metagenomic and metatranscriptomic analyses of soil samples exposed to elevated or low H2 concentration led to the identification of 45 genes encoding the large subunit of [NiFe]-hydrogenases belonging to 8 distinct phyla. Our results indicate that despite significant sequencing effort, retrieved hydrogenase sequences are not in themselves adequate surrogates of H2 oxidation activity in these soils. In fact, land-use exerted a greater influence than H2 exposure on both hydrogenase gene distribution and expression though expression of certain genes responded to H2. We argue that approaches relying on PCR/RT-PCR amplicon sequencing or quantification combined with physicochemical parameters are currently the best option to infer the activity of H2-oxidizing bacteria and probably other specialist functional guilds with similar population size in soil.

18. Intestinal microRNAs and bacterial taxa in juvenile mice are associated, modifiable by allochthonous lactobacilli, and affect postnatal maturation

Abstract

The gut microbiota affects intestinal microRNA (miRNA) signatures and is modified by host-derived luminal miRNA. This suggests the existence of close miRNA-microbiota relationships that are critical to intestinal homeostasis. However, an integrative analysis of these relationships and their evolution during intestinal postnatal maturation is lacking. We provide a system-level longitudinal analysis of miRNA-microbiota networks in the intestine of mice at the weaning transition, including tissue and luminal miRNA and luminal microbiota. To address causality and move toward translational applications, we used allochthonous probiotic lactobacilli to modify these longitudinal relationships and showed that they are critical for intestinal maturation in early life. These findings contribute to understand mechanisms that underlie the maturation of the intestinal ecosystem and suggest that interventions aiming at maintaining, or restoring, homeostasis cannot prescind from considering relationships among its components.The interplay between the intestinal microbiota and host is critical to intestinal ontogeny and homeostasis. MicroRNAs (miRNAs) may be an underlying link. Intestinal miRNAs are microbiota-dependent and, when shed in the lumen, affect resident microorganisms. Yet, longitudinal relationships between intestinal tissue miRNAs, luminal miRNAs, and luminal microorganisms have not been elucidated, especially in early life. Here, we investigated the postnatal cecal miRNA and microbiota populations, their relationship, and their impact on intestinal maturation in specific pathogen-free mice; we also assessed if they can be modified by intervention with allochthonous probiotic lactobacilli. Abstract

The presence of neighbouring plants and predation risk can affect trophic interactions between plants and herbivores. We hypothesized that the relative preference for neighbouring species would determine winter herbivory and that predation risk would modulate browsing pressure. We tested these hypotheses using feeding trials in two regions with high white-tailed deer (Odocoileus virginianus (Zimmermann, 1780)) densities: Outaouais and Anticosti Island (Quebec, Canada). In each, we selected a species of interest and compared browsing rates and time spent foraging with neighbours relatively preferred or avoided. In a subexperiment, we included coyote (Canis latrans Say, 1823) urine to test for threat-sensitive foraging and interactions with neighbour effects. In Outaouais, time spent foraging on the focal species was reduced by the presence of potentially avoided neighbours and deer reduced browsing with increased perceived predation risk. On Anticosti, browsing rates on the focal species increased with avoided neighbours, with no effect of the predator urine. Anticosti deer have been in a predator-free environment for more than 120 years, likely reducing antipredator behaviours. This study demonstrates both neighbour effects and threat-sensitive foraging, phenomena that could interact and thus would benefit from being studied together to better represent trophic interactions in natural environments. Nous avons teste ces hypotheses par des tests d'alimentation dans deux regions caracterisees par de fortes densites de cerfs de Virginie (Odocoileus virginianus (Zimmermann, 1780)) : l'Outaouais et l'ile d'Anticosti (Quebec, Canada). Dans chacune de ces regions, nous avons selectionne une espece d'interet et compare les frequences de broutement et le temps passe a l'approvisionnement a ceux de plantes compagnes preferees ou evitees. Dans le cadre d'une sous-experience, nous avons inclus de l'urine de coyote (Canis latrans Say, 1823) pour verifier si l'approvisionnement et les interactions avec les especes compagnes etaient sensibles au risque de predation percu. En Outaouais, la presence d'especes compagnes potentiellement evitees reduisait le temps passe a brouter l'espece d'interet, et un plus grand risque predation percu entrainait une reduction du broutement par les cerfs. Dans l'ile d'Anticosti, le broutement de l'espece d'interet augmentait en presence d'especes compagnes evitees, sans effet de l'urine de predateur. Les cerfs de l'ile d'Anticosti evoluent dans un milieu exempt de predateurs depuis plus de 120 ans, ce qui a vraisemblablement diminue leurs comportements antipredateur. Abstract

Aims/hypothesis: Type 2 diabetes increases the risk of cardiovascular and renal complications, but early risk prediction could lead to timely intervention and better outcomes. Genetic information can be used to enable early detection of risk. Methods: We developed a multi-polygenic risk score (multiPRS) that combines ten weighted PRSs (10 wPRS) composed of 598 SNPs associated with main risk factors and outcomes of type 2 diabetes, derived from summary statistics data of genome-wide association studies. The 10 wPRS, first principal component of ethnicity, sex, age at onset and diabetes duration were included into one logistic regression model to predict micro- and macrovascular outcomes in 4098 participants in the ADVANCE study and 17,604 individuals with type 2 diabetes in the UK Biobank study. Results: The model showed a similar predictive performance for cardiovascular and renal complications in different cohorts. It identified the top 30% of ADVANCE participants with a mean of 3.1-fold increased risk of major micro- and macrovascular events (p = 6.3 × 10−21 and p = 9.6 × 10−31, respectively) and a 4.4-fold (p = 6.8 × 10−33) higher risk of cardiovascular death. While in ADVANCE overall, combined intensive blood pressure and glucose control decreased cardiovascular death by 24%, the model identified a high-risk group in whom it decreased the mortality rate by 47%, and a low-risk group in whom it had no discernible effect. High-risk individuals had the greatest absolute risk reduction with a number needed to treat of 12 to prevent one cardiovascular death over 5 years. Conclusions/interpretation: This novel multiPRS model stratified individuals with type 2 diabetes according to risk of complications and helped to target earlier those who would receive greater benefit from intensive therapy. Abstract

In this contribution, we present a quantum dynamical approach to study inelastic scattering of diatomic molecules from metal surfaces at normal incidence. The dissipative dynamics obeys a stochastic Schrödinger equation describing the time-evolution of the system as a piecewise deterministic process. Energy exchange between the molecular vibrational degrees of freedom and the metal electrons is represented using operators in tensor product form, which are coupled via anharmonic transition rates calculated from first-order perturbation theory. Full dimensional observables are obtained by averaging over simulations in 4D—including the internal stretch, the distance to the surface, and the orientation angles—at different surface sites. The method is applied to the state-resolved scattering of vibrationally excited NO from Au(111), revealing important channels for quantized energy relaxation. [ABSTRACT FROM AUTHOR]

Abstract

The Hudson Bay complex (HBC) is home to numerous indigenous communities that traditionally have relied heavily on its marine resources. The nutritional quality and stocks of the entire HBC food web depend in large part on the phytoplankton production of bioactive molecules (long chain polyunsaturated fatty acids and carotenoids) and their transfer through trophic levels. The purpose of this study was thus to determine which molecules were produced during spring phytoplankton blooms, as well as the environmental factors driving this production. We investigated 21 stations in 5 sub-regions of the HBC. At the time of sampling, the sub-regions studied had different environmental settings (e.g., ice cover, nutrients, seawater salinity and temperature) conditioning their bloom stages. Pre- and post-bloom stages were associated with relatively low concentrations of bioactive molecules (either fatty acids or carotenoids). In contrast, the highest concentrations of bioactive molecules (dominated by eicosapentaenoic acid and fucoxanthin) were associated with the diatom bloom that typically occurs at the ice edge when silicates remain available. Interestingly, the large riverine inputs in eastern Hudson Bay led to a change in protist composition (larger contribution of Dinophyceae), resulting in lower while more diverse content of bioactive molecules, whether fatty acids (e.g., aa-linolenic acid) or carotenoids (e.g., peridinin). As greater stratification of the HBC is expected in the future, we suggest that a mixotrophic/heterotrophic flagellate-based food web would become more prevalent, resulting in a smaller supply of bioactive molecules for the food web.

Abstract

Evaluating metagenomic software is key for optimizing metagenome interpretation and focus of the Initiative for the Critical Assessment of Metagenome Interpretation (CAMI). The CAMI II challenge engaged the community to assess methods on realistic and complex datasets with long- and short-read sequences, created computationally from around 1,700 new and known genomes, as well as 600 new plasmids and viruses. Here we analyze 5,002 results by 76 program versions. Substantial improvements were seen in assembly, some due to long-read data. Related strains still were challenging for assembly and genome recovery through binning, as was assembly quality for the latter. Profilers markedly matured, with taxon profilers and binners excelling at higher bacterial ranks, but underperforming for viruses and Archaea. Clinical pathogen detection results revealed a need to improve reproducibility. Runtime and memory usage analyses identified efficient programs, including top performers with other metrics. The results identify challenges and guide researchers in selecting methods for analyses.

Abstract

Global warming has pronounced effects on tundra vegetation, and rising mean temperatures increase plant growth potential across the Arctic biome. Herbivores may counteract the warming impacts by reducing plant growth, but the strength of this effect may depend on prevailing regional climatic conditions. To study how ungulates interact with temperature to influence growth of tundra shrubs across the Arctic tundra biome, we assembled dendroecological data from 20 sites, comprising 1153 individual shrubs and 223 63 annual growth rings. Evidence for ungulates suppressing shrub radial growth was only observed at intermediate summer temperatures (6.5 °C–9 °C), and even at these temperatures the effect was not strong. Multiple factors, including forage preferences and landscape use by the ungulates, and favourable climatic conditions enabling effective compensatory growth of shrubs, may weaken the effects of ungulates on shrubs, possibly explaining the weakness of observed ungulate effects. Earlier local studies have shown that ungulates may counteract the impacts of warming on tundra shrub growth, but we demonstrate that ungulates’ potential to suppress shrub radial growth is not always evident, and may be limited to certain climatic conditions.
