Background: Development of multidrug resistance in Uropathogenic Escherichia coli (UPEC) makes treatment of Urinary Tract Infections (UTIs) a major challenge. This study was conducted to investigate the effect of trans-resveratrol (t-RSV) at a subinhibitory concentration (sub-MIC-t-RSV) on phenotypic and genotypic expression of virulence factors of clinical isolates of UPEC and develop a nanoformulation of t-RSV. Fifty-five clinical UPEC strains were investigated for the presence of virulence factors by phenotypic methods and PCR detection of virulence genes. The effect of sub-MIC-t-RSV was studied on the phenotypic and genotypic expression of virulence factors. t-RSV-loaded nanoemulgel formulation was prepared and characterized. Results: Out of the 55 tested isolates, 50.9% were biofilm producers, 23.6% showed both mannose-sensitive and mannose-resistant hemagglutination, 21.8% were serum-resistant, 18.2% were hemolysin producers, while 36.4% showed cytotoxic effect on HEp-2 cells. A total of 25.5% of the isolates harbor one or more of hly-A, cnf-1 and papC genes, while 54.5% were positive for one or more of fimH, iss and BssS genes. A concentration of 100 µg/mL of t-RSV effectively downregulates the phenotypic and genotypic expression of the virulence factors in positive isolates. A stable t-RSV-nanaoemulgel with droplet size of 180.3 nm and Zetapotential of -46.9 mV was obtained. Conclusion: The study proves the effective role of t-RSV as an antivirulence agent against clinical UPEC isolates in vitro and develops a stable t-RSV-nanoemulgel formulation to be assessed in vivo. Electrochemical biosensor based on horseradish peroxidase (HRP) modified gold nanoparticle/sulfur-doped graphene (AuNP/SG) has been developed for the quantification of trans-resveratrol (Res). Gold nanoparticle (AuNP) was selected as the carrier for HRP due to its excellent electrochemical catalytic activity, biocompatibility, and conductivity, while sulfur-doped graphene (SG) was selected as the support for AuNP due to the defects introduced in the carbon network by sulfur doping can facilitate the high dispersion state of AuNP and HRP. The results show that the excellent sensing performance of the HRP/AuNP/SG is attributed to the high charge polarization of material after sulfur dropping and the co-catalysis of HRP and AuNP. For the detection of Res, linear response can be observed with high sensitivity and low detection limit (4 × 10−8 M). Further, this electrochemical biosensor presents strong reproducibility, stability, and anti-interference capability in detecting Res. Mastalgia is the most common manifestation of mastopathy. Up to 70 % of women experiencing breast pain report a significant decrease in their quality of life; however, hormone therapy is not indicated and suitable for everyone. The main aim of a clinician in this case is to exclude cancer and choose appropriate treatment to manage pain. First-line therapy for such patients may not necessarily include medications.The aim of this work was to identify the most effective non-pharmacological treatments for patients with mastalgia. We conducted a search of publications assessing various methods of mastalgia management in both Russian and foreign databases (PubMed, CyberLeninka, Elibrary, Google Scholar). We used the following key words: “mastalgia”, “cyclic mastalgia“, and “non-cyclic mastalgia“.Visual demonstration of the fact the woman has no risk of breast cancer (after examination) and proper explanation can reduce complaints of mastalgia without any additional treatment. Recommendation to change a bra also brings a significant relief to the majority of women. Reduced consumption of methylxanthines and fats along with sufficient intake of fiber and liquid can be beneficial for patients. Dietary modifications with certain nutrients (such as indole-3-carbinol and trans-resveratrol) might significantly improve the quality of life of patients with mastalgia. Relaxation practices can also mitigate mastalgia.Most patients with mastalgia can be managed without any medications if they are reassured about the absence of cancer risk, choose a well-fitting and supportive bra, have psychoemotional support, and modify their diet.

Neuropsychiatric disorders are anticipated to be a leading health concern in the near future, emphasizing an outstanding need for the development of new effective therapeutics to treat them. Stilbenes, with resveratrol attracting the most attention, are an example of multi‐target compounds with promising therapeutic potential for a broad array of neuropsychiatric and neurological conditions. This review is a comprehensive summary of the current state of research on stilbenes in several neuropsychiatric and neurological disorders such as depression, anxiety, schizophrenia, autism spectrum disorders, epilepsy, traumatic brain injury, and neurodegenerative disorders. We describe and discuss the results of both in vitro and in vivo studies. The majority of studies concentrate on resveratrol, with limited findings exploring other stilbenes such as pterostilbene, piceatannol, polydatin, tetrahydroxystilbene glucoside, or synthetic resveratrol derivatives. Overall, although extensive preclinical studies show the potential benefits of stilbenes in various central nervous system disorders, clinical evidence on their therapeutic efficacy is largely missing. This study aims to determine whether drying is a suitable preservation method for gilaburu fruit and the changes in the bioactive components of gilaburu fruit (Viburnum opulus L.) at the end of the drying process. In this study, gilaburu fruits were dried in a cabinet dryer at different temperatures (50°C, 60°C, and 70°C). The analyses of trans-resveratrol, water-soluble vitamins, organic acids, and phenolic compounds were made using the HPLC method, while total phenolic contents and antioxidant activity were spectrophotometric. As a result of drying of gilaburu fruit at 50°C, 6 °C, and 70°C, the highest component loss was observed at 70°C. Losses of 73.64% and 84.08%, respectively, were detected in the total phenolic substance and antioxidant capacity content of gilaburu fruit after drying at 70°C. While the trans-resveratrol content was 1.26±0.05 (g/100 g dry weight (DW)) in fresh fruit, it reduced to 0.31±0.03, 0.30±0.01 and 0.21±0.01 after drying at 50°C, 60°C and 7 °C, respectively. In terms of vitamins, the highest loss was seen in niacin. The contents of ascorbic acid, pyridoxine, niacin and thiamine contents of fresh gilaburu fruit decreased after drying at 50°C, 60°C and 70°C. In addition, drying kinetics of water-soluble vitamins, total phenolic contents, antioxidant activity, and trans-resveratrol were modeled. The Page model best described the drying behavior of fruits at 70°C, and the parabolic model at both 50°C and 60°C. Thermal degradation of water-soluble vitamins, total phenolic contents, antioxidant activity, and trans-resveratrol were fitted in the first-order kinetic model.

Phenolic compounds are crucial for grape berry quality, wine, and human health. The factors that dramatically affect the composition and levels of metabolites are biotic and abiotic plant stressors. Wounding is one of these stressors. However, little is known about its effect on the accumulation and biosynthesis of the whole phenolic profile. This study examined the effects of artificial leaf wounding on metabolite accumulation in 'Merlot' grapes. Twenty-year-old vines underwent ten wounding treatments at different times. Results showed diverse responses with practical implications for managing berry maturity and quality. Early-season treatments accelerated ripening, while late-season treatments halted sugar accumulation. Late treatments increased total phenolics, but responses varied in the phenolic profile. T5 had the highest total phenolics, while T1 exhibited the highest tannin content. Wounding treatments increased total anthocyanins compared to control. Flavonoids and hydroxybenzoic acids responded differently, with T9 showing the highest (+)-Catechin and Syringic acid contents. Vanillic acid T6 had the highest (–)-Epicatechin control content. Except for T7, all treatments had higher trans-resveratrol levels than control. Injury direction and timing had numerical and content effects, although not statistically significant. 'Merlot' grapes can perceive artificial wounds and mount a robust defense response. Wounding enhances the production of beneficial compounds, improving ripeness, chemical composition, wine quality, and human health. More research is needed for optimal wounding strategies, environmental friendliness, berry ripening, and leaf photosynthetic responses to injury. BackgroundDietary supplements, like small-quantity lipid-based nutrient supplements (SQ-LNS), are used in intervention programs to prevent undernutrition among women and young children in low-income countries. An objective marker is needed to track consumption of supplements to evaluate the effectiveness of these programs.ObjectiveThe aim of this study was to evaluate saccharin and resveratrol as potential adherence markers for tracking recent consumption of a single serving of SQ-LNS in women.MethodsForty-seven healthy nonpregnant women 18-45 y of age were assigned to consume a single dose of SQ-LNS (20 g) containing either 10 mg sodium saccharin or 5 mg trans-resveratrol, under supervision. On the day before and for 2 d following SQ-LNS consumption, urine samples were collected each day for 24 h as 3 consecutive 4-h collections and one 12-h overnight collection. Urinary concentrations of saccharin and trans-resveratrol-3-O-sulfate, a resveratrol metabolite, were measured by ultra-high-performance liquid chromatography interfaced to a mass spectrometer with electrospray ionization [UHPLC-(ESI-)MS/MS]. Urinary concentrations (μmol/L urine) of saccharin and trans-resveratrol-3-O-sulfate were plotted against time, and receiver operating characteristic (ROC) curves were used to determine the discriminative capacity of each compound, at each post-consumption time point compared with baseline, to detect recent consumption of SQ-LNS. Cutoff values to differentiate supplement consumption from nonconsumption of each marker were developed using the closest-to-(0,1)-corner cut-point approach.ResultsForty-five participants were included in the analysis. Urinary concentrations of saccharin and trans-resveratrol-3-O-sulfate increased within 4 h of SQ-LNS consumption. Urinary concentration cutoff values for saccharin (13.4 µmol/L) and trans-resveratrol-3-O-sulfate (0.7 µmol/L) allowed for 78% and 89% sensitivity, respectively, and 100% specificity in detecting consumption of SQ-LNS within the first 12 h after consumption.ConclusionsUrinary concentrations of saccharin and trans-resveratrol-3-O-sulfate reflect consumption of SQ-LNS containing those compounds during the first 12 h post-consumption with high sensitivity and specificity in healthy women and may be useful objective adherence markers for tracking consumption of SQ-LNS.

Saccharin and trans-resveratrol were incorporated into small quantity lipid-based nutritional supplements (SQ-LNS) to be evaluated as the markers of consumption for nutritional intervention studies. Forty-seven healthy women consumed a single supplement with either 8.6 mg of saccharin or 5 mg of trans-resveratrol, and urine was collected for 4 h. A rapid 11 min method employing multiple reaction monitoring and ultrahigh performance liquid chromatography coupled to a triple quadrupole mass spectrometer was developed to measure saccharin and resveratrol metabolites in urine simultaneously. The linear dynamic range of the method was from 3 to 1000 ng mL-1, with the correlation coefficient of 0.999 and limits of quantification from 15.28 to 53.03 ng mL-1. Sample preparation was simple dilution with an average recovery of 97.8%. Ion suppression was observed with urine concentrations >10%. Mean levels of saccharin and resveratrol-3-O-sulfate in urine were 5.481 ± 4.359 and 3.440 ± 4.160 nmol L-1, respectively. We developed and validated a method to measure saccharin and trans-resveratrol metabolites in urine to objectively corroborate the consumption of SQ-LNS for the first time in nutrition intervention studies.

The present study proposes a method for designing small bioactive nanoparticles using silk fibroin as a carrier to deliver hydrophobic polyphenols. Quercetin and trans-resveratrol, widely distributed in vegetables and plants, are used here as model compounds with hydrophobic properties. Silk fibroin nanoparticles were prepared by desolvation method and using various concentrations of ethanol solutions. The optimization of the nanoparticle formation was achieved by applying Central Composite Design (CCD) and the response surface methodology (RSM). The effects of silk fibroin and ethanol solution concentrations together with the pH on the selective encapsulation of phenolic compounds from a mixture were reported. The obtained results showed that nanoparticles with an average particle size of 40 to 105 nm can be prepared. The optimized system for the selective encapsulation of the polyphenols on the silk fibroin substrate was determined to be 60% ethanol solution and 1 mg/mL silk fibroin concentration at neutral pH. The selective encapsulation of the polyphenols was achieved, with the best results being obtained in the case of resveratrol and quercetin and encapsulation of gallic and vanillic acids being rather poor. Background: Lung adenocarcinoma (LUAD) is a dominant tumor with high morbidity and mortality. In spite of innovations in surgery, chemotherapy, and targeted therapies, the prognosis for patients with LUAD remains unsatisfactory. Therefore, it is particularly important to study the biological markers of the occurrence, development, and prognosis of this disease. Methods: In this study, 3 datasets were selected from the Gene Expression Omnibus (GEO) database to screen differentially expressed genes (DEGs). Then, Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses were performed using Database Annotation, Visualization and Integrated Discovery (DAVID). In addition, a protein-protein interaction network was performed and 10 key genes were analyzed. Gene and protein expression were analyzed using The Cancer Genome Atlas (TCGA) and The Human Protein Atlas (HPA) databases, respectively. Then, overall survival (OS) rate was analyzed using Kaplan-Meier plotter database. The chemical components were downloaded from the HERB database. Finally, the chemical constituents were docked with 10 targets by molecular docking. By using in vitro studies, we evaluated the effects of curcumin and trans-resveratrol on the proliferation of LUAD cells. Results: A total of 122 upregulated and 434 downregulated genes were identified. Then, 10 key genes (glyceraldehyde-3-phosphate dehydrogenase (GAPDH), interleukin-1 beta (IL1B), von Willebrand factor (VWF), matrix metalloproteinase-9 (MMP9), CD44 antigen (CD44), transcription factor Jun (JUN), platelet endothelial cell adhesion molecule (PECAM1), hematopoietic progenitor cell antigen CD34 (CD34), peroxisome proliferator-activated receptor gamma (PPARG), and collagen alpha-1(I) chain (COL1A1)) were obtained. TCGA results showed that GAPDH, MMP9, and COL1A1 were upregulated, while IL-1B, VWF, CD44, JUN, PECAM1, CD34, and PPARG were downregulated in cancer tissues. Kaplan-Meier plotter analysis also showed that high expression of GAPDH, PPARG, and COL1A1, and low expression of VWF, PECAM1, CD34, PPARG, and COL1A1 were associated with poor prognosis in LUAD. Combined with gene expression, staging, immunohistochemistry, and OS prognostic analysis, the results showed that COL1A1 was a potential target for the occurrence and prognosis of LUAD. In addition, 2 key components (curcumin and trans-resveratrol) had good docking effect with COL1A1. We found that curcumin and trans-resveratrol significantly inhibited the proliferation of LUAD cells and significantly decreased the expression of COL1A1 and MMP9. Conclusion: In conclusion, our study identified the potential target of LUAD and the main traditional Chinese medicine components (curcumin and trans-resveratrol) for LUAD treatment. Pelvic pain is one of the most common and significant clinical symptoms of endometriosis. The complex pathogenesis and paradoxical nature of endometriosis-associated pain, its chronicity and centralization, a marked decrease in the quality of life of patients, the lack of an "ideal" and effective remedy with no limitation for all patients, as well as the progressive and recurrent nature of the disease are factors that determine the need for novel, additional therapeutic options. The pain mechanisms in endometriosis and the potential pathogenetic effects of trans-resveratrol and indole-3-carbinol on various aspects of endometriosis-associated pain are addressed based on the literature sources presented in electronic databases PubMed, CyberLeninka, and Google Scholar. The described pharmacological properties of the compounds suggest that the combined use of trans-resveratrol and indole-3-carbinol is a pathogenetically justified and promising treatment for complex endometriosis-associated pain.

In the framework of a protein–ligand-fishing strategy to identify proteins that bind to trans-resveratrol, a natural phenolic compound with pharmacological benefits, we have developed magnetic nanoparticles covalently linked to trans-resveratrol through three different derivatives and examined their aggregation behavior in aqueous solution. The monodispersed magnetic core (18 nm diameter) with its mesoporous silica shell (93 nm diameter) exhibited a notable superparamagnetic behavior useful for magnetic bioseparation. The hydrodynamic diameter, deduced from dynamic light scattering analysis, of the nanoparticle increased from 100 to 800 nm when the aqueous buffer changed from pH 10.0–3.0. A size polydispersion occurred from pH 7.0–3.0. In parallel, the value of the extinction cross section increased according to a negative power law of the UV wavelength. This was mainly due to light scattering by mesoporous silica, whereas the absorbance cross section remained very low in the 230–400 nm domain. The three types of resveratrol-grafted magnetic nanoparticles exhibited similar scattering properties, but their absorbance spectrum was consistent with the presence of trans-resveratrol. Their functionalization increased their negative zeta potential when pH increased from 3.0 to 10.0. The mesoporous nanoparticles were monodispersed in alkaline conditions, where their anionic surface strongly repulsed each other but aggregated progressively under van der Waals forces and hydrogen bonding when negative zeta potential decreased. The characterized results of nanoparticle behavior in aqueous solution provide critical insight for further study of nanoparticles with proteins in biological environment. [ABSTRACT FROM AUTHOR]

Agro-industrial by-products are a sustainable source of natural additives that can replace the synthetic ones in the food industry. Grape pomace is an abundant by-product that contains about 70% of the grape's polyphenols. Polyphenols are natural antioxidants with multiple health-promoting properties. They are secondary plant metabolites with a wide range of solubilities. Here, a novel extraction process of these compounds was developed using enzymes that specifically liberates target polyphenols in the appropriate hydroalcoholic mixture. Tannase, cellulase, and pectinase retained 22, 60, and 52% of their activity, respectively, in ethanol 30% v/v. Therefore, extractions were tested in ethanol concentrations between 0 and 30% v/v. Some of these enzymes presented synergistic effects in the extraction of specific polyphenols. Maximum yield of gallic acid was obtained using tannase and pectinase enzymes in ethanol 10% v/v (49.56 ± 0.01 mg L−1 h−1); in the case of p-coumaric acid, by cellulase and pectinase treatment in ethanol 30% v/v (7.72 ± 0.26 mg L−1 h−1), and in the case of trans-resveratrol, by pectinase treatment in ethanol 30% v/v (0.98 ± 0.04 mg L−1 h−1). Also, the effect of enzymes and solvent polarity was analysed for the extraction of malvidin-3-O-glucoside, syringic acid, and quercetin. Previous studies were mainly focused on the maximization of total polyphenols extraction yields, being the polyphenolic profile the consequence but not the driving force of the optimization. In the present study, the basis of a platform for a precise extraction of the desire polyphenols is provided. Antibiotic resistance due to bacterial biofilm formation is a major global health concern that makes the search for new therapeutic approaches an urgent need. In this context,, trans-resveratrol (RSV), a polyphenolic natural substance, seems to be a good candidate for preventing and eradicating biofilm-associated infections but its mechanism of action is poorly understood. In addition, RSV suffers from low bioavailability and chemical instability in the biological media that make its encapsulation in delivery systems necessary. In this work, the anti-biofilm activity of free RSV was investigated on Staphylococcus aureus and, to highlight the possible mechanism of action, we studied the anti-adherence activity and also the cell wall damage on a MRSA strain. Free RSV activity was compared to that of RSV loaded in liposomes, specifically neutral liposomes (L = DOPC/Cholesterol) and cationic liposomes (LG = DOPC/Chol/GLT1) characterized by a galactosylated amphiphile (GLT1) that promotes the interaction with bacteria. The results indicate that RSV loaded in LG has anti-adherence and anti-biofilm activity higher than free RSV. On the other side, free RSV has a higher bacterial-growth-inhibiting effect than encapsulated RSV and it can damage cell walls by creating pores; however, this effect can not prevent bacteria from growing again. This RSV ability may underlie its bacteriostatic activity.

A grapevine shoot extract (GSE) was obtained using ultrasound-assisted extraction and characterized. The main phenolic constituents were identified as stilbenoids. Among them, trans-resveratrol and trans-ε-viniferin stood out. The GSE was administered to an isoproterenol-induced myocardial injury animal model. The extract alleviated the associated symptoms of the administration of the drug, i.e., the plasma lipid profile was improved, while the disturbed plasma ion concentration, the cardiac dysfunction markers, the DNA laddering, and the necrosis of myocardial tissue were diminished. This effect could be related to the anti-oxidative potential of GSE associated with its antioxidant properties, the increased levels of endogenous antioxidants (glutathione and enzymatic antioxidants), and the diminished lipid peroxidative markers in the heart. The results also revealed angiotensin-converting enzyme (ACE)-inhibitory activity, which indicated the potential of GSE to deal with cardiovascular disease events. This work suggests that not only trans-resveratrol has a protective role in heart function but also GSE containing this biomolecule and derivatives. Therefore, GSE has the potential to be utilized in the creation of innovative functional ingredients.

In this study, we compared the polyphenolic composition of the roasted grapevine wood chips of four Vitis vinifera cultivars—namely, Sorbara, Grasparossa, Malbo Gentile, and Spergola. These waste byproducts have the potential as infusion chips for the aging of alcoholic beverages and vinegars, contributing to an enriched sensory profile. Roasting amplifies aromatic nuances and triggers the depletion of crucial bioactive compounds, including polyphenols. We investigated the extent of polyphenolic loss in the ethanolic extract of roasted grapevine chips to repurpose this waste byproduct and assess its potential. We assessed the levels of trans-resveratrol, trans-ε-viniferin, trans-piceatannol, and the main resveratrol trimer. Our findings indicated a significant decrease in polyphenol content as the roasting temperature increased, from 16.85–21.12 mg GAE/g for grapevine chips roasted at 120 °C to 3.10–7.77 mg GAE/g for those roasted at 240 °C. This study also highlights notable genotypic differences in polyphenolic content. Among the red grape cultivars analyzed, Sorbara exhibited the highest levels (7.77–21.12 mg/GAEg), whereas the white grape cultivar Spergola showed the lowest polyphenolic content (3.10–16.85 mg/GAEg). These findings not only contribute to the scientific understanding of polyphenol stability but also hold practical implications for the enhancement of aged beverages, as well as advancing sustainable practices in the viticulture industries.

The past few decades have seen a marked expansion in market demand for food supplements with therapeutic value. Due to this demand, the recovery of vine waste for obtaining certain phytochemicals or plant synthesized compounds with health-promoting activities can be an important economic component, principally with the agreement of the European Union for resveratrol as a new food ingredient. For the sake of economic capitalization, it is necessary to determine optimum extraction parameters for maximum recovery. In this paper, we have determined the optimum parameters for the solvent extraction of trans-resveratrol from vine prunings. We tested different extraction conditions: 35 different types of solvents, 10 types of solid-to-liquid ratios, 10 extraction times, 10 types of granulosity of the ground material and 7 consecutive extractions on the same material. The optimal parameters determined were: solvent ethanol:diethyl ether 4:1 ratio, 1:35 solid liquid ratio g/mL, 4 days for extraction time, 500 µm–350 µm granulosity of powdered material and one extraction on the material. The objective of this study was to determine the influence of chitosan treatments, applied in vineyard for plan protection, on the phenolic quality of grapes. The study was conducted for two consecutive years (2020-2021) in the wine center of Murfatlar, Romania, on 'Fetească neagră', an ancient but well-known indigenous Romanian grape variety for red wines. Chitosan is meant to offer an alternative for the classic treatment with Bordeaux mixture (BM), which represented the control variant. A combination treatment with half dose of chitosan and half dose of BM was also applied and evaluated. Chemical composition at harvest time was evaluated by measuring the content of sugar, total acidity, pH, total anthocyanin potential (ApH1), anthocyanins extractable at wine pH (ApH3.2), total polyphenol index, skin tannins, seed tannins and seeds maturity. In wines the colour parameters were determined by spectrophotometry and individual phenolic compounds by UHPLC-HRMS. In grapes, accumulation of anthocyanins increased with the total dose of chitosan applied in the vineyard, with higher values in 2020 when temperatures were higher and the rainfall values were typical for the region. Accumulation of tannins in grape skins followed a similar trend. In wine, chitosan determined a significant 72% increase in colour intensity in 2020. Among the individual phenols gallic acid was predominant, with higher values in the rainier year (2021) and significant increases determined by chitosan treatment (especially in 2021 when it increased by 97% as compared to BM treatment). Catechin and epicatechin recorded important increases in the less favourable year (2021), with confirmed increases in both years elicited by the chitosan. Quercetin and myricetin were not influenced by the chitosan treatment, but their increase was corelated with higher temperatures and inversely corelated to the amount of rainfall. Transresveratrol ranged between 4.3-8.0 mg L-1 in 2020 and 5.0-6.5 mg L-1 in 2021, with an important increase determined by the chitosan treatment in 2020 (89% increase compared to BM treatment). This paper presents the results of the testing during the winemaking process using commercial active dry yeasts (CADY) in parallel with yeast biomass produced starting from yeasts isolated from Pietroasa area (Romania) (PAY). PAYs were obtained based on autochthonous yeasts isolated from Busuioacă de Bohotin (BB) and Tămâioasa Românească (TR) grape varieties. Two variants of must for each grape variety were taken in study, i.e. obtained from early harvested grapes (TRD and BBD) and from late harvested grapes (TRS and BBS). Winemaking experiments were carried out with 7 types of commercial yeasts and two types of commercial nutrients. The alcoholic concentration of the experimental wines varied from 11.8 % for WTRD to 18.4% for WBBD. The WBBD & S samples showed a higher average trans-resveratrol content (1.716 and 1.555 mg/L, respectively) compared to the WTRD & S wine samples (0.590 and 1.365 mg/L, respectively). Resveratrol is a compound increasingly studied for its many beneficial properties for health. However, it is a highly unstable photosensitive compound, and therefore it is necessary to encapsulate it to protect it if you want to use it in a commercial product. Emulsions are systems that allow the encapsulation of active ingredients, protecting them and allowing their release in a controlled manner. They are highly used systems in the pharmaceutical, cosmetic and food industries. The main objectives of this work are to study the feasibility of encapsulating resveratrol in concentrated water-in-oil-in-water double emulsions and the effect produced by adding the double emulsion with optimal formulation to a commercial cream for cosmetic applications. The effect of the selected optimal double emulsion on a commercial cream was studied, analyzing droplet size distribution, morphology, stability and rheology. The main conclusion of this work is that incorporating 1/3 of concentrated double emulsion W1/O/W2 into a commercial moisturizing cream had a positive physical effect and produced cream with a resveratrol concentration of up to 0.0042 mg/g. • UV-C applied to grapevine cuttings for the first time induces abiotic stress, causing physiological and biochemical changes and affecting the growth of the cuttings. • The marked increase of trans -resveratrol and antioxidant activity in leaves highlights the potential of UV-C and increases the efficiency of plant defense mechanisms. • UV-C application increases photosynthetic processes through an increase in photosynthetic pigments, promoting plant metabolism and enhancing development. In this study, UV-C was applied to the cuttings of Cabernet Sauvignon and Cabernet Franc cv.'s at three different durations to simulate abiotic stress, with subsequent monitoring of resultant changes upon the formation of at least six leaves. In both grape varieties, total phenolic compounds and antioxidant capacity in leaves reached the highest levels after 60 min of UV-C treatment. Trans -resveratrol content reached its highest level with an increase of 157 %. The activity of superoxide dismutase enzyme reached its highest level with an increase of 630 %, especially in Cabernet Franc. Total chlorophyll, chlorophyll a, chlorophyll b contents reached their highest levels after 60 min of UV-C treatment. UV-C treatments did not affect shoot number. However, parameters such as fresh and dry shoot weight, shoot length, fresh and dry root weight reached their highest levels after 45 min of UV-C stress. In correlation analysis, the highest correlation was found between trans -resveratrol and chlorophyll b. In principal component analysis, 6 min of UV-C stress had the highest loading on component 1 and 45 min of UV-C stress had the highest loading on component 2. These results suggest that UV-C treatment may be a potential method to improve grape quality and plant health by inducing important physiological responses in grapevines. The methods for producing natural resveratrol are of big interest due to the numerous health benefits of this substance and its increasing use in functional foods, food supplements and para-pharmaceutical preparations, especially with the recent permission of resveratrol as a novel food ingredient in the European Union (Commission Implementing Decision (EU) 2016/1190). Since annually a large amount of waste is produced in vine pruning, the aim of this study was to evaluate the potential of vine shoots from ten grape varieties (Fetească Albă, Fetească Neagră, Muscat Ottonel, Muscat de Hamburg, Tămâioasă, Coarnă Neagră, Merlot, Cabernet Sauvignon, Fetească Regală and Burgund Mare) from two different locations to be used as a natural source of trans-resveratrol. To reach this goal, two different solvents were explored, two different storage periods, three different time of collecting of samples in two different years. Moreover, a correlation between the type of vine (red or white) and trans-resveratrol content was analyzed. Maceration solvent diethyl-ether:ethanol, 1:4 ratio, showed a higher yield of extraction of trans-resveratrol than methanol 80%. Increasing the storage time by another 6 weeks positively affected the concentration of resveratrol for 5 samples, and for another 7 negatively. The time of collecting the samples had a high influence on the resveratrol content on shoots samples, highlighting the fact that autumn is the optimal time for harvesting. No correlation could be established among the grape varieties (red or white) and the trans-resveratrol content. In this research, the amounts of trans-resveratrol, (+)-catechin, (-)- epicatechin, and malvidin-3-glucoside which are among phenolic compounds as well as the total phenolic compound, total anthocyanin, and antioxidant capacity changes were examined in the Boğazkere (Vitis vinifera L.) grape, pomace before press, pomace after press and wine. In all parameters examined, a decrease was seen when proceeding from grape to wine, only (+)- catechin and (-)- epicatechin levels were detected to be higher in the prepress pomace. Total phenolic compound, total anthocyanin, and antioxidant capacity levels were found highest in grape samples, respectively, 8018 mg GAE/kg; 1606 mg/kg; 16.05 µmol troloxl/g with ABTS method; 7.75 µmol trolox/g with DPPH method; 4.78 µmol trolox/g with FRAP method. The highest amount of trans-resveratrol content was obtained from grape berry with 3.57 mg/kg, the highest (+)- catechin content was obtained from the pomace before press sample with 54.66 mg/kg, the highest amount of (-)- epicatechin was obtained from pomace before press sample 20.38 mg/kg and the highest amount of malvidin-3-glucoside amount was determined as 510 mg/kg in grape. According to the results of the research, it was understood that the pomace of the Boğazkere variety, which is one of the grape varieties mostly processed as wine in our country, has a high level of antioxidant capacity and phenolic content. Vine shoots resulting from vine pruning can become valuable products of interest for many industries. Research conducted to date showed that resveratrol is found in many parts of the vine. The aim of the present study was to determine the resveratrol content in Romanian vine sprouts samples by direct injection into the HPLC instrument coupled with diode array detector after being filtered through 0.45 μm PTFE membrane filters. The resveratrol content varied from 5.44 mg/100 g to 10.27 mg/100 g in ethanolic extracts. For aqueous extract, the resveratrol was found in only two samples in very low concentration. Thise paper aimed to contribute to the research on the capitalization of residual products from viticulture industry.

By tradition, herbal infusions have been mainly consumed for their pleasant taste, but, nowadays, the consumer, along with the pleasantness of drinking a savory beverage, also looks for their health benefits. Grapes and grape/wine by-products are a rich source of health-promoting compounds, presenting great potential for the development of new beverages. Moreover, grape-infusion preparation is no more than a sustainable or green way of extracting polyphenols and other nutraceutical compounds from grapes and grape leaves. In this review, we summarize the benefits of drinking grape infusions and discuss the sustainable processes of extracting potential nutraceutical compounds from grapes and grape by-products, which are often considered fermentation waste and are discarded to the environment without proper treatment.

Cissus quadrangularis is a nutrient-rich plant with a history of use in traditional medicine. It boasts a diverse range of polyphenols, including quercetin, resveratrol, β-sitosterol, myricetin, and other compounds. We developed and validated a sensitive LC-MS/MS method to quantify quercetin and t-res biomarkers in rat serum and applied this method to pharmacokinetic and stability studies. The mass spectrometer was set to negative ionization mode for the quantification of quercetin and t-res. Phenomenex Luna (C18(2), 100 A, 75 × 4.6 mm, 3 µ) column was utilized to separate the analytes using an isocratic mobile phase consisting of methanol and 0.1% formic acid in water (82:18). Validation of the method was performed using various parameters, including linearity, specificity, accuracy, stability, intra-day, inter-day precision, and the matrix effect. There was no observed significant endogenous interference from the blank serum. The analysis was completed within 5.0 min for each run, and the lower limit of quantification was 5 ng/mL. The calibration curves showed a linear range with a high correlation coefficient (r2 > 0.99). The precision for intra- and inter-day assays showed relative standard deviations from 3.32% to 8.86% and 4.35% to 9.61%, respectively. The analytes in rat serum were stable during bench-top, freeze-thaw, and autosampler (−4 °C) stability studies. After oral administration, the analytes showed rapid absorption but underwent metabolism in rat liver microsomes despite being stable in simulated gastric and intestinal fluids. Intragastric administration resulted in higher absorption of quercetin and t-res, with greater Cmax, shorter half-life, and improved elimination. No prior research has been conducted on the oral pharmacokinetics and stability of anti-diabetic compounds in the Ethanolic extract of Cissus quadrangularis EECQ, making this the first report. Our findings can provide the knowledge of EECQ’s bioanalysis and pharmacokinetic properties which is useful for future clinical trials.

The present study proposes a method for designing small bioactive nanoparticles using silk fibroin as a carrier to deliver hydrophobic polyphenols. Quercetin and trans-resveratrol, widely distributed in vegetables and plants, are used here as model compounds with hydrophobic properties. Silk fibroin nanoparticles were prepared by desolvation method and using various concentrations of ethanol solutions. The optimization of the nanoparticle formation was achieved by applying Central Composite Design (CCD) and the response surface methodology (RSM). The effects of silk fibroin and ethanol solution concentrations together with the pH on the selective encapsulation of phenolic compounds from a mixture were reported. The obtained results showed that nanoparticles with an average particle size of 40 to 105 nm can be prepared. The optimized system for the selective encapsulation of the polyphenols on the silk fibroin substrate was determined to be 60% ethanol solution and 1 mg/mL silk fibroin concentration at neutral pH. The selective encapsulation of the polyphenols was achieved, with the best results being obtained in the case of resveratrol and quercetin and encapsulation of gallic and vanillic acids being rather poor. Thin-layer chromatography confirmed the selective encapsulation and the loaded silk fibroin nanoparticles exhibited antioxidant activity.

Glucocorticoids are metabolized by the CYP3A isoform of cytochrome P450 and by 11-β-hydroxysteroid dehydrogenase type 1 (11β-HSD-1). Experimental data suggest that post-traumatic stress disorder (PTSD) is associated with an increase in hepatic 11β-HSD-1 activity and a concomitant decrease in hepatic CYP3A activity. Trans-resveratrol, a natural polyphenol, has been extensively studied for its antipsychiatric properties. Recently, protective effects of trans-resveratrol were found in relation to PTSD. Treatment of PTSD rats with trans-resveratrol allowed the rats to be divided into two phenotypes. The first phenotype is treatment-sensitive rats (TSR), and the second phenotype is treatment-resistant rats (TRRs). In TSR rats, trans-resveratrol ameliorated anxiety-like behavior and reversed plasma corticosterone concentration abnormalities. In contrast, in TRR rats, trans-resveratrol aggravated anxiety-like behavior and decreased plasma corticosterone concentration. In TSR rats, hepatic 11β-HSD-1 activity was suppressed, with a concomitant increase in CYP3A activity. In TRR rats, the activities of both enzymes were suppressed. Thus, the resistance of PTSD rats to trans-resveratrol treatment is associated with abnormalities in hepatic metabolism of glucocorticoids. The free energy of binding of resveratrol, cortisol, and corticosterone to the human CYP3A protein was determined using the molecular mechanics Poisson–Boltzmann surface area approach, indicating that resveratrol could affect CYP3A activity.

Reported mole fraction solubilities of trans-resveratrol in aqueous mixtures of acetone in the temperature interval from (273.2 to 313.2) K were correlated by means of some cosolvency models, like multi-linear models of Jouyban-Acree and Jouyban-Acree-van't Hoff and non-linear models of the modified Wilson and CNIBS/R-K model. Apparent thermodynamic quantities of the dissolution processes were computed using the van't Hoff and Gibbs equations. Gibbs energies and enthalpies were positive in all cases, while negative and positive entropies were observed. The plot of enthalpy vs. Gibbs energy of dissolution was non-linear with negative slopes in the region of 0.00 ≤ x1 ≤ 0.70 but positive slopes in the region of 0.70 ≤ x1 ≤ 1.00. Further, by means of the inverse Kirkwood-Buff integrals is observed that trans-resveratrol is preferentially solvated by water in water-rich and acetone-rich mixtures but preferentially solvated by acetone in the composition interval of 0.21 < x1 < 0.77. Retinal and optic nerve damage in glaucoma involves excitotoxicity via N-methyl-D-aspartate (NMDA) receptors. Since, trans-resveratrol (TR) is known to provide neuroprotection, we investigated its protective effects against NMDA-induced retinal and optic nerve injury. Sprague Dawley rats were divided into four groups which received vehicle (PBS), NMDA, and TR 0.4 or TR 4 nmol 24 h prior to NMDA, unilaterally and intravitreally. Seven days post-injection, rats were euthanized; eyeballs were enucleated and subjected to hematoxylin and eosin and terminal transferase dUTP nick end labeling staining while optic nerves were isolated for toluidine blue staining. Retinal morphometry showed that ganglion cell layer (GCL) layer thickness within inner retina (IR), retinal cell count (RCC) per 100-µm length of GCL, RCC per 100-µm2 area of GCL, and RCC per 100 µm2 of IR were significantly higher in both TR-treated groups compared to the NMDA group. No differences were observed between the two dose groups. Optic nerve morphology was in accordance with the retinal morphology whereby TR-treated groups showed significantly lesser degenerative changes compared to NMDA-treated group. Summary: Dietary polyphenols, viz. naringenin and trans‐resveratrol, are widely consumed by the humans. Nevertheless, being oxidation prone and highly unstable, their utilisation in food systems has been considerably limited. Here, we developed a co‐delivery liposome system to improve the storage and antioxidative stability of naringenin and trans‐resveratrol. Mean particle diameter, polydispersity index and transmission electron microscopy, of single and co‐encapsulated liposomes containing polyphenols were evaluated. Higher encapsulation efficiency was noticed in co‐loaded liposomes (RNLIP; 86.89 ± 1.75%) than liposomes separately loaded with naringenin (NLIP; 77.72 ± 2.42%) and trans‐resveratrol (RLIP; 61.98 ± 1.68%), and the effect was in an additive fashion. Co‐loaded liposomes showed the stronger environmental stability, including pH, temperature and sodium chloride than single encapsulation. In addition, co‐encapsulated liposomes exhibited higher antioxidant activity, which was evaluated by DPPH radical‐scavenging activity, ferric reducing power and lipid peroxidation inhibition ability. Therefore, these results confirmed that the strategy of co‐encapsulated trans‐resveratrol (stilbene) and naringenin (flavanone) liposome could maximise polyphenols' application in food systems. Resveratrol (3, 4', 5-trihydroxystilbene) is a natural phenolic compound synthesized by plants in response to various stressful circumstances (mechanical injury, siege of bacteria or fungi and UV exposure). Commercial resveratrol supplements on the market are mostly extracted from Japanese knotweed. As it grows even in heavily polluted environments and the presence of emodin, which is a compound that may have laxative effects and is found in partially purified extracts, additional concerns about the safety of these commercial preparations were raised. An alternative source of resveratrol intensely studied in recent years is vine waste, with recent studies showing high concentrations of resveratrol in extracts. In the last years, the researcher’s attention is focused on obtaining an efficient method of extraction and purification of this compound from the vine waste to show that this material can be used as a raw material for production of commercial supplements of resveratrol.

Allergic rhinitis (AR) is a multifactorial disease triggered by interactions between genes and the environment. Clinical evidence has shown that trans-resveratrol, a widely used drug, significantly ameliorates AR pathology. However, the precise mechanisms underlying this effect remain unclear. This study aimed to elucidate the pharmacological mechanisms of action of trans-resveratrol in patients with AR who exhibit hypoxic symptoms. This will be achieved through microRNA sequencing and signaling pathway screening combined with basic experiments to determine the effects of Trans-resveratrol intervention in this patient population. Network pharmacology was used to determine the therapeutic value of trans-resveratrol in AR. The micro-RNA miR-204-3p was pinpointed by sequencing. Quantitative reverse transcription polymerase chain reaction was used to quantify the expression levels. Haematoxylin and eosin, alcian blue-periodic acid–Schiff, and Masson's trichrome staining were used to assess the effects of hypoxia on nasal mucosa immunohistochemistry and immunofluorescence-localised target proteins. Egl nine homolog 3 (EGLN3) was screened using bioinformatics software. Protein expression was detected by western blotting. Cell growth and death were gauged via Cell Counting Kit-8 and terminal deoxynucleotidyl transferase dUTP nick end labelling staining, respectively. Cell migration was observed using a transwell assay. Enzyme-linked immunosorbent assay was used to measure interleukin (IL)33 levels in the cell supernatants. Flow cytometry was used to verify cell cycle and antigen levels. Electron microscopy was used to visualise the status of the nasal mucosa prior to in vivo expression analysis. Patients with hypoxic AR demonstrated more pronounced nasal mucosal remodelling than that in patients with common AR. Sequencing results indicated that these patients had a reduced expression of miR-204-3p. Through a combination utilizing of bioinformatics analysis and experimental validation, EGLN3 has been identified as a direct target of HIF-1α. The low expression level of miR-204-3p represses EGLN3, resulting in the accumulation of HIF-1α and the activation of the IL33/ST2 signaling pathway. These stimulate the proliferation, survival, and migration of HNEpCs, ultimately contributing to mucosa remodeling and AR progression. Trans-resveratrol notably downregulated the levels of HIF-1α and IL33/ST2, while simultaneously increasing the expression of EGLN3. Downregulation of miR-204-3p initiated a vicious cycle of hypoxic AR via EGLN3/HIF-1α/IL33/ST2. Trans-resveratrol reversed the pathological process of nasal mucosa remodeling of hypoxic AR by exhibiting anti-inflammatory and anti-angiogenic functions via the above signaling pathway. Our study uncovers the underlying mechanism by which hypoxia drives the progression of AR. It presents innovative strategies for addressing inflammatory and hypoxia-related diseases, bridging traditional and modern medicine, and highlighting the potential of natural compounds in clinical practice. [Display omitted] Helicobacter pylori (H. pylori) is a microorganism directly linked to severe clinical conditions affecting the stomach. The virulence factors and its ability to form biofilms increase resistance to conventional antibiotics, growing the need for new substances and strategies for the treatment of H. pylori infection. The trans -resveratrol (RESV), a bioactive polyphenol from natural sources, has a potential activity against this gastric pathogen. Here, Chitosan nanoparticles (NP) containing RESV (RESV-NP) were developed for H. pylori management. The RESV-NP were prepared using the ionic gelation method and characterized by Dynamic Light Scattering (DLS), Nanoparticle Tracking Analysis (NTA) and, Cryogenic Transmission Electron Microscopy (Cryo – TEM). The encapsulation efficiency (EE) and in vitro release rate of RESV were quantified using high-performance liquid chromatography (HPLC). RESV-NP performance against H. pylori was evaluated by the quantification of the minimum inhibitory/bactericidal concentrations (MIC/MBC), time to kill, alterations in H. pylori morphology in its planktonic form, effects against H. pylori biofilm and in an in vitro infection model. RESV-NP cytotoxicity was evaluated against AGS and MKN-74 cell lines and by hemolysis assay. Acute toxicity was tested using Galleria mellonella model assays. RESV-NP showed a spherical shape, size of 145.3 ± 24.7 nm, polydispersity index (PDI) of 0.28 ± 0.008, and zeta potential (ZP) of + 16.9 ± 1.81 mV in DLS, while particle concentration was 3.12 x 1011 NP/mL (NTA). RESV-NP EE was 72 %, with full release within the first 5 min. In microbiological assays, RESV-NP presented a MIC/MBC of 3.9 µg/mL, a time to kill of 24 h for complete eradication of H. pylori. At a concentration of 2xMIC (7.8 µg/mL), RESV-NP completely eradicated the H. pylori biofilm, and in an in vitro infection model, RESV-NP (4xMIC – 15.6 µg/mL) showed a significant decrease in bacterial load (1 Log10CFU/mL) when compared to the H. pylori J99 control. In addition, they did not demonstrate a toxic character at MIC concentration for both cell lines. The use of the RESV-NP with mucoadhesion profile is an interesting strategy for oral administration of substances targeting gastric disorders, linked to H. pylori infections. [ABSTRACT FROM AUTHOR]

Lysimachia nummularia L. is a perennial herbaceous plant rich in bioactive compounds, which can be utilized for medicinal purposes. The present work aims to analyze the phenolic compounds from different parts of the Lysimachia nummularia L. plant using the HPLC technique: Lysimachiae radix (the root part), Lysimachiae herba (the aerial part), Lysimachiae flores (flowers). In order to determine the phenolic compounds, extraction from the three categories of vegetable products was performed with ethanol 70% (v/v) using three extraction methods: (i) Soxhlet extraction, (ii) maceration and (iii) ultrasonic-assisted extraction. The content of polyphenols was determined by the Folin–Ciocalteu method, and the antioxidant activity was evaluated by the DPPH, ABTS, FRAP and CUPRAC methods. The antioxidant activity was correlated with the content of phenolic compounds in the analyzed extracts. The following phenolic compounds were separated, identified and quantified: 3-O-methylgallic, gallic, ferulic, caffeic, chlorogenic, p-coumaric acids and trans-resveratrol. According to the experimental data, the highest content of total polyphenols was observed in the hydroethanolic extract from Lysimachiae flores (22.10 ± 1.48 mg gallic acid/g), which also presented remarkable antioxidant activity.

Resveratrol with a high content in grapes has beneficial effects on inflammation, endothelial function, oxidative stress and cardiovascular and cerebrovascular diseases. Here, we propose a sensitive analytical method basing on the ultrasonic-assisted extraction followed by ultra-high performance liquid mass spectrometry to simultaneously determine trans-resveratrol (RES), trans-resveratrol 3-O-β-D-glucuronide, trans-resveratrol 4ʹ-O-β-D-glucuronide (R4G) in grapes. Single factor experiment and response surface methodology were used to optimize the pretreatment process of grape samples. The optimal conditions were as follows: extraction solvent-ethyl acetate, ultrasonic temperature 62 °C, ultrasonic time 20 min, ultrasonic power 90% and the volume of the extracted solvent 3500 μL. The method showed the good linearity (r ≥ 0.9917), the satisfactory intra-day (%RSD ≤ 11.3%) and inter-day precision (%RSD ≤ 14.5%) and low limit of quantification (0.1 μg/mL). This proposed method was successfully applied to determine RES, R3G, R4G in different grapes, achieving satisfactory recoveries (88.8–116.9%). Thus, this method established in this study is simple and time-saving, which will be helpful to the development and utilization of RES, R3G, R4G in grapes in food and pharmaceutical industries. Reported equilibrium mole fraction solubility values of trans-resveratrol in some aqueous-ethanolic mixtures in the temperature interval from (273.2 to 323.2) K were correlated by means of some cosolvency models, like multi-linear models of Jouyban-Acree and Jouyban-Acree-van't Hoff and non-linear models of the modified Wilson and Buchowski-Ksiazczak. Apparent thermodynamic functions of the dissolution processes were computed using the van't Hoff and Gibbs equations. Gibbs energy and enthalpy were positive in all cases, while negative and positive entropies were observed. The plot of enthalpy vs. Gibbs energy of dissolution was non-linear with negative slopes in the region of 0.00 ≤ x1 ≤ 0.60 but positive slope in the region of 0.60 ≤ x1 ≤ 0.90. Further, by means of the inverse Kirkwood-Buff integrals is observed that trans-resveratrol is preferentially solvated by water in water-rich mixtures but preferentially solvated by ethanol in the composition interval of 0.24 < x1 < 1.00. There is increasing evidence surrounding the health benefits of E-resveratrol; this has triggered interest in stilbenoids in grapes, wine, and by-products. On the one hand, there is an enormous amount of underutilized vine waste, rich in bioactive substances during wine production. On the other hand, there is a growing demand for promising phytochemicals, for dietary and pharmaceutical purposes. Vine shoots are promising sources of stilbenoids; they have economic potential because they are sources of high-value phytochemicals. Recent studies have shown that, due to biosynthesis pathway genes, especially STS (forming trans-resveratrol), which is abundant during storage periods of vine shoots—trans-resveratrol accumulates up to 40-fold. The objective of this research was to determine the most economical part of vine waste to be exploited, and to study the kinetics of resveratrol increase in a 90-day period, to determine the optimal storage period to reach a maximum trans-resveratrol content. Total phenolic content (TPC) and antioxidant activity (AA) were studied to determine possible correlations. In Fetească Neagră vine shoot varieties stored at laboratory temperatures, trans-resveratrol content increased to a maximum (2712.86 mg/kg D.W.) at day 70, and then slightly decreased until day 90. TPC remained constant and there was a slight increase in AA. Vine shoots contained the largest amounts of trans-resveratrol (1658.22 mg/kg D.W.), followed by tendrils (169.92 mg/kg D.W.), and leaves (43.54 mg/kg D.W.). [ABSTRACT FROM AUTHOR]

Resveratrol (trans-3,5,4’-trihydroxystilbene) is a phytoalexin produced naturally by 72 different plant species especially grapevines, Japanese knotweed and pines in response to some exogenous factors such as UV radioation, chemical stress and fungal parasites. With the discovery of the „French paradox” resveratrol had entered in the spotlight of researchers. The French paradox is a term generated in 1992 that refers to the cardioprotective effects of resveratrol that have been discovered based on data from French people that has shown an inverse correlation between wine consumption and the incidence of coronary heart disease (CHD) despite the consumption of a diet in hight saturated fat. The aim of the present study is to determine the trans-resveratrol content in 11 Romanian comercial wine samples by direct injection into the HPLC instrument coupled with diode array detector after being filtered through 0.45 µm PTFE membrane filtres. The total resveratrol content in alayses samples varied from 0.03 mg/L to 6.64 g/L. Pinot Noir wines had the highest average resveratrol content, while the lowest was found for the Sauvignion Blanc variety. All wines analyzed contained resveratrol in variable quantities. This study could contribute to the analysis made on the variety of Romanian wines for resveratrol content.

PurposeThe effect of an oral trans-resveratrol-based supplement (Longevinex®) on choroidal thickness, measured using optical coherence tomography (OCT) enhanced depth imaging, was investigated in a prospective study.Materials and methods34 young, healthy participants were randomly divided into two age- and gender-matched groups. They were then assigned in a randomized fashion to treat with either a trans-resveratrol-based group (Longevinex®, Las Vegas) or placebo. All participants underwent ocular imaging with spectral domain (SD)-OCT (Spectralis; Heidelberg Engineering, Heidelberg) at the baseline and then again 1 h following treatment. The choroidal thickness was measured in a masked fashion at the fovea and at four additional points, located at 500 μm and 1000 μm nasal to the fovea and 500 μm and 1000 μm temporal to the fovea.ResultsIn the resveratrol group, the foveal choroidal thickness at the baseline was 267.73 ± 84.19 μm (mean ± SD); it increased to 284.57 ± 92.39 μm 1 h after drug treatment (p = 0.033). The mean choroidal thickness was also significantly increased at each of the four extrafoveal points (all p < 0.05). In the control group, the mean baseline choroidal thickness at the fovea was 269.73 ± 71.40 μm (mean ± SD) and it was 268.43 ± 70.15 μm (mean ± SD) 1 h after the placebo was administered (p = 0.183); there were also no significant differences in choroidal thickness at the four additional points (all p > 0.05) Conclusion: A significant increase in choroidal thickness following oral administration of a trans-resveratrol-based supplement was observed. There was no change in choroidal thickness in the placebo-treated control group. We speculate that the increased choroidal thickness is the result of choroidal vessel vasodilation.

The present work describes a simple, accurate and validated UV spectrophotometric method for determination of trans-resveratrol loaded in ethosomal formulation. The method was validated for different parameters like linearity, precision, specificity, accuracy, limit of detection (LOD), limit of quantitation (LOQ) and robustness as per ICH guidelines. The wavelength maximum of resveratrol in phosphate buffer pH 7.4 was found to be at 307 nm. The method was found to be linear in the range of 8 to 38 μg mL-1 with a correlation coefficient (r²) of 0.9957. The accuracy of the method was studied by recovery study and % recovery was found to be in the range of 99.24 to 100.14 %. The method is simple, accurate and requires relatively inexpensive instruments. The proposed method can be successfully used for determination of trans-resveratrol loaded into ethosomes. Background: Although ionizing radiation (IR) has been of immense value to human life due to its involvement in several fields, it doesn't eliminate that exposure to IR results in an array of biological consequences, including oxidative stress, inflammation, and death. Thus, this study aimed to explore the curative effect of trans-Resveratrol (t-Res) on hepatic and renal injury in a rat model exposed to single and fractionated doses of γ-rays. Methods: Rats exposed to a single dose of IR (6 Gy, as an acute effect) or a fractionated dose of IR (2 Gy/time/3 days, day after day; to imitate a chronic impact) were treated with t-Res. Then, the radio-protective effect of t-Res was investigated via biochemical and histological estimations in the liver and kidney of rats in the different groups. Results: The data displayed a significant amelioration in biochemical and histological indices in the liver and kidney of rats exposed to IR doses and treated with t-Res. Particularly, t-Res reduced the oxidative stress milieu through decreasing HIF-1α, ROS, and MDA levels associated with increased CAT activity and Nrf-2 gene expression. Also, t-Res improved the inflammatory status via a decrease in TNF-α, NF-κB, SOCS-3, and HSP-70 genes expression linked with elevations in SIRT-1 and P53 genes expression. Conclusion: It could be concluded that t-Res had hepatoprotective and renoprotective effects against the deleterious consequences of γ-rays exposure due to its antioxidant and anti-inflammatory properties. Introduction: Amyloid-beta (Aβ) protein is a major component of the extracellular plaque found in the brains of individuals with Alzheimer's disease (AD). In this study, we investigated the effect of trans-resveratrol as an antagonist treatment for moderate to mild AD, as well as its safety and tolerability. Methods: This was a case–control study that enrolled 30 selected patients who had been clinically diagnosed with moderate to mild AD. These patients were randomly divided into two groups, namely, a placebo group (n = 15) and a trans-resveratrol group (n = 15) who received 500 mg trans-resveratrol orally once daily for 52 weeks. Brain magnetic resonance imaging (MRI) examinations were performed on and cerebrospinal fluid (CSF) samples were obtained from all participants before (baseline) and after the study (52 weeks). Enzyme-linked immunosorbent assays were used to determine the levels of plasma Aβ40 and Aβ42 and CSF Aβ40 and Aβ42. Results: The results showed that the changes over the study period in the levels of Aβ40 in the blood and CSF of the patients treated with trans-resveratrol were not statistically significant (P > 0.05). In contrast, patients who received placebo showed a significant decrease in Aβ40 levels compared with that at the beginning of the study (CSF Aβ40: P = 0.024, plasma Aβ40: P = 0.036). Analysis of the images on the brain MRI scans revealed that the brain volume of the patients treated with trans-resveratrol was significantly reduced at 52 weeks (P = 0.011) compared with that of patients in the placebo treatment group, Further analysis indicated that the level of matrix metallopeptidase 9 in the CSF of the patients treated with trans-resveratrol at 52 weeks decreased by 46% compared with that of patients in the placebo group (P = 0.033). Conclusion: These results indicate that trans-resveratrol has potential neuroprotective roles in the treatment of moderate to mild AD and that its mechanism may involve a reduction in the accumulation and toxicity of Aβ in the brain of patients, thereby reducing neuroinflammation. Hepatocellular carcinoma (HCC) is a prevalent liver cancer representing the fourth most lethal cancer worldwide. Trans -Resveratrol (T-R) possesses a promising anticancer activity against HCC. However, it suffers from poor bioavailability because of the low solubility, chemical instability, and hepatic metabolism. Herein, we developed T-R-loaded nanocochleates using a simple trapping method. Nanocarriers were optimized using a comprehensive in-vitro characterization toolset and evaluated for the anticancer activity against HepG2 cell line. T-R-loaded nanocochleates demonstrated monodispersed cylinders (163.27 ± 2.68 nm and 0.25 ± 0.011 PDI) and −46.6 mV ζ-potential. They exhibited a controlled biphasic pattern with minimal burst followed by sustained release for 72 h. Significant enhancements of Caco-2 transport and ex-vivo intestinal permeation over liposomes, with 1.8 and 2.1-folds respectively, were observed. Nanocochleates showed significant reduction of 24 h IC 50 values compared to liposomes and free T-R. Moreover, an efficient knockdown of anti-apoptotic (Bcl-2) and cancer stemness (NANOG) genes was demonstrated. To the best of our knowledge, we are the first to develop T-R loaded nanocochleates and scrutinize its potential in suppressing NANOG expression, 2-folds lower, compared to free T-R. According to these auspicious outcomes, nanocochleates represent a promising nanoplatform to enhance T-R oral permeability and augment its anticancer efficacy in the treatment of HCC.
