Background: HIV-1-associated neurocognitive disorders (HAND) in stable patients undergoing antiretroviral therapy (ART) may result from ongoing immune dysregulation and chronic inflammation. A contributing factor may result from the unstable HLA class I allele, HLA-C*07. Objective: To assess the genetic profile of killer-cell immunoglobulin-like receptors (KIR), human leukocyte antigens (HLA), and immune activation or senescence markers and their association with HAND in stable HIV-1 patients receiving ART. Methods: An observational cross-sectional study was carried out with 96 patients with asymptomatic or symptomatic HAND. HLA and KIR as well as immune activation/senescence biomarkers in peripheral blood cells were assessed by SSO-Luminex typing and flow cytometry, respectively. Results: HLA-C*07 is associated with symptomatic HAND. The frequency of two copies of HLA-C*07 was higher in patients with symptomatic than with asymptomatic HAND (12.0 vs. 2.2%, ρ < 0.001). The percentage of senescent CD8
+ CD28- T-cells was higher in patients with two copies of HLA-C*07 (ρ < 0.05). In patients with symptomatic HAND, the percentages of non-senescent CD8+ CD28+ T cells were inversely proportional to the number of copies of the HLA-C*07 (ρ < 0.05). Conclusion: Patients with symptomatic HAND showed a higher frequency of the homozygotic unstable HLA-C*07 allotype, which could be associated with neurocognitive complications. Two copies of HLA-C*07 were associated with immune senescent T lymphocyte profiles characterized by the loss of CD28 expression. [ABSTRACT FROM AUTHOR]- Published

- Abstract
Els canvis en la forma de viure de les noves generacions requereixen una arquitectura que recolzi les noves formes d'utilitzar l'habitatge. Les persones busquen espais flexibles que puguin adaptar-se a les seves necessitats i preferències canviants, permetent un major grau de personalització a la llar. Transformacions globals com l'evolució del treball cap al treball remot o des de casa accelerat per la pandèmia de Covid-19, els canvis en l'estructura familiar i els rols en aquesta, els canvis demogràfics i l'envelliment de la població, són factors que influeixen en la demanda d'habitatges adaptables. Tanmateix, altres factors com l'augment de la consciència sobre l'eficiència energètica i la comoditat, la creixent adopció de dispositius mòbils i la popularitat dels assistents virtuals, ens fan intuir una creixent demanda de llars intel·ligents. A la recerca de llars que s'adaptin a les necessitats i preferències dels ocupants de manera fàcil i eficient, la domòtica en combinació amb l'arquitectura flexible podria ser una solució per adaptar-se a les necessitats actuals i futures d'habitatge? En el transcurs del treball estudiarem les condicions que envolten la creació dels sistemes de mobiliari domòtic, el paper que poden tenir en l'arquitectura i quines prediccions sobre la seva implementació en podem fer., Los cambios en la forma de vivir de las nuevas generaciones requieren una arquitectura que apoye las nuevas formas de utilizar la vivienda. Las personas buscan espacios flexibles que puedan adaptarse en sus necesidades y preferencias cambiantes, permitiendo un mayor grado de personalización al hogar. Transformaciones globales como la evolución del trabajo hacia el trabajo remoto o desde casa acelerado por la pandemia de Covid-19, los cambios en la estructura familiar y los roles en esta, los cambios demográficos y el envejecimiento de la población, son factores que influyen en la demanda de viviendas adaptables. Aun así, otros factores como el aumento de la conciencia sobre la eficiencia energética y la comodidad, la creciente adopción de dispositivos móviles y la popularidad de los asistente virtuales, nos hacen intuir una creciente demanda de hogares inteligentes. En busca de hogares que se adapten a las necesidades y preferencias de los ocupantes de manera fácil y eficiente, la domótica en combinación con la arquitectura flexible podría ser una solución para adaptarse a las necesidades actuales y futuras de vivienda? En el transcurso del trabajo estudiaremos las condiciones que rodean la creación de los sistemas de mobiliario domótico, el papel que pueden tener en la arquitectura y qué predicciones sobre su implementación podemos hacer., New generations changing lifestyles require an architecture that supports different ways of using homes. People seek flexible spaces that can adapt to their changing needs and preferences, allowing a greater sense of personalization. The evolution of work towards remote accelerated by the Covid-19 pandemic, as well as modifications in family structure and roles, demographic movements and the aging of the population, are all contributing to the demand for adaptable housing. Other factors, such as rising awareness of energy efficiency and comfort, the adoption of mobile devices and the popularity of virtual assistants, suggest a growth in desire for smart homes. When looking for homes that adapt to the needs and preferences of the occupants easily and efficiently, could home automation in combination with flexible architecture be a solution to adapt to current and future housing needs? In the course of the work, we will study the conditions surrounding the creation of domotic furniture systems, the role they can play in architecture, and which predictions we can make.

Na presente dissertação, na modalidade de Projeto para obtenção do grau de Mestre em Arquitetura Paisagista, é apresentado uma proposta desenvolvida a nível de Anteprojeto para reabilitação de um troço em meio urbano da ribeira de Cobres, localizada em Almodôvar. A água é um recurso natural finito e é fundamental para a existência de vida no nosso planeta. Consciencializar a população sobre a importância de conservar, preservar e sobretudo proteger a água é um grande desafio. A reabilitação e a integração de rios e ribeiras nos espaços urbanos são uma preocupação atual, nomeadamente no sentido de atingir uma melhoria nos fluxos ecológicos. A ribeira de Cobres em Almodôvar é um desses casos. Ao longo do seu trajeto, os impactos causados pelo Homem são bastantes visíveis. Em grande parte, a ribeira permanece ao abandono, artificializada e degradada. Nalguns trechos encontra-se canalizada e em ocasiões de elevada precipitação regista-se a ocorrência de cheias. Esta proposta de projeto de reabilitação desta ribeira visa contribuir para alcançar um futuro sustentável, que permite um aumento na melhoria da qualidade de vida da população. Reabilitar e integrar a ribeira no meio urbano é o fator chave para a criação de um elo entre o meio urbano e a Natureza. Espera-se que o presente trabalho contribua para demonstrar a importância de salvaguardar, proteger e requalificar os cursos de água em meio urbano e que a proposta de intervenção projetada para esta ribeira se possa tornar num exemplo, de algum modo, consciencializar a importância dos meios hídricos e a falta que eles fazem no meio urbano. In the present dissertation, i developed a project to obtain the Master's degree in Landscape Architecture, it was a developed Antproject proposal for Rehabilitation of a section in the urban area of the “ribeira de Cobres”. Water is a natural limited resource and is essential for the existence of life on our planet. Making the population aware of the importance of conserving, preserving and specially protecting water is a major challenge. The rehabilitation and integration of rivers and streams in urban spaces is a current concern, for example in order to achieve an improvement in ecological flows. The Cobres stream in Almodôvar is one of these situations. Along its path, the impacts caused by Man were devastating. For the most part, the stream remains abandoned, full of humans wast. In some part is channeled and on occasions of high rainfall there is the occurrence of floods. This proposal for a rehabilitation project for this stream is the way forward to achieve a sustainable future, with an increase in the quality of life of the population. Restoring and integrating the stream into the urban environment is the key factor in creating a link between the urban environment and Nature. It is expected that the present work will contribute to demonstrate the importance of safeguarding, protecting and requalifying water courses in urban areas and that the intervention proposal designed for this stream can become an example, in some way, to raise awareness of the importance of water resources and the lack of them in the urban environment

- Abstract
This study described the clinical, virological, and serological responses of immunologically naïve and vaccinated horses to African horse sickness virus (AHSV) serotype 9. Naïve horses developed a clinical picture resembling the cardiac form of African horse sickness. This was characterized by inappetence, reduced activity, and hyperthermia leading to lethargy and immobility-recumbency by days 9-10 post-infection, an end-point criteria for euthanasia. After challenge, unvaccinated horses were viremic from days 3 or 4 post-infection till euthanasia, as detected by serogroup-specific (GS) real time RT-PCR (rRT-PCR) and virus isolation. Virus isolation, antigen ELISA, and GS-rRT-PCR also demonstrated high sensitivity in the post-mortem detection of the pathogen. After infection, serogroup-specific VP7 antibodies were undetectable by blocking ELISA (b-ELISA) in 2 out of 3 unvaccinated horses during the course of the disease (9-10 dpi). Vaccinated horses did not show significant side effects post-vaccination and were largely asymptomatic after the AHSV-9 challenge. VP7-specific antibodies could not be detected by the b-ELISA until day 21 and day 30 post-inoculation, respectively. Virus neutralizing antibody titres were low or even undetectable for specific serotypes in the vaccinated horses. Virus isolation and GS-rRT-PCR detected the presence of AHSV vaccine strains genomes and infectious vaccine virus after vaccination and challenge. This study established an experimental infection model of AHSV-9 in horses and characterized the main clinical, virological, and immunological parameters in both immunologically naïve and vaccinated horses using standardized bio-assays.

- Abstract
While trade finance has been recognized as a key element in international trade, there is a persistent gap between supply and demand. On the other hand, distributed ledger technology (DLT) has given birth to the decentralised finance (DeFi) phenomenon, promising to revolutionize banking and the whole financial sector. This paper enquires whether a DeFi-based business model could address the trade finance gap problematic. To this end, it presents a sketch of a DeFi trade finance business model, showing how it could address the reasons behind the trade finance gap, while at the same time developing a broader meaning of the DeFi concept itself

Abstract Background Type I Interferons (IFNs) are well known cytokines which exert antiviral activity, antitumor activity and immunomodulatory effects. Single-nucleotide polymorphisms (SNP) and deletions in the gene coding for IFNA2 have been shown to influence the level of expression in vitro. The indel polymorphism -305_-300delAACTTT showed the strongest effect in vitro. To analyse the worldwide distribution of this polymorphism we analyzed five different populations (586 Vietnamese, 199 Central Africans, 265 Brazilians, 108 Kaingang and 98 Guarani). To investigate a possible association with susceptibility to infectious diseases we determined the polymorphism in malaria patients suffering either mild or severe malaria and in a cohort of hepatitis C virus infected individuals. Results We could detect the indel polymorphism in all populations analysed. There was no association with this polymorphism and the outcome of malaria but we found an increase of this indel polymorphism in hepatitis C virus positive individuals compared to healthy controls (p = 0.014). Conclusion Polymorphisms in genes involved in the interferon pathway have been implicated in the resistance or susceptibility against cerebral malaria and HBV. Here we show that an indel polymorphism, which mediates a disadvantageous effect in HBV patients, may also play a disadvantageous role in HCV infections stressing the importance of a fully functional interferon pathway.

[ES]: Para favorecer el uso de dosis de inseminación de semen de porcino congelado-descongelado a nivel comercial, sería interesante conocer de antemano si el semen de verraco congelará bien o mal. El objetivo de este estudio fue evaluar el 'ChemSensor' (cromatógrafo de gases-masas unido a un software de análisis quimiométrico) como herramienta precoz de discriminación entre eyaculados según su congelabilidad. Para ello se utilizaron 33 eyaculados de verracos de raza Ibérica de los cuales se cogió una alícuota de 1 ml de semen y se analizó con el 'ChemSensor', el resto del eyaculado fue congelado-descongelado usando un protocolo estandarizado, para determinar su congelabilidad (buenos y malos congeladores) en función del porcentaje de espermatozoides con la membrana plasmática intacta y el porcentaje de espermatozoides móviles totales post-descongelación. Para el análisis con el 'ChemSensor', las muestras se volatilizaron e ionizaron descomponiéndolas en diferentes iones con un tamaño determinado, que se usaron como variables de discriminación, para separar los eyaculados en función de su congelabilidad. El 'ChemSensor' fue capaz de discriminar todos los eyaculados de los buenos congeladores; y dentro del grupo de los malos congeladores solamente una muestra fue clasificada erróneamente. Debido al reducido número de muestras, la distancia obtenida entre grupos (2,62), aunque no muy elevada, podría considerarse como significativa, aunque sería necesario un mayor número de muestras para poder crear un modelo matemático más robusto. En conclusión, el 'ChemSensor' es una posible herramienta adecuada para la discriminación precoz de eyaculados de cerdo Ibérico en función de su congelabilidad. [EN]: For the use of insemination dose of frozen-thawed boar sperm at the commercial level, it would be very interesting to know, in advance, if sperm from a boar will freeze well or poorly. So the aim of this study was to evaluate the ability of the “ChemSensor” (gas chromatograph-mass coupled with chemometric analysis software) as an early discrimination tool of Iberian pig ejaculates according to their freezability. For this, 33 ejaculates from Iberian breed boars were used and an aliquot of 1 ml of semen from each one was analyzed with the “ChemSensor”, the rest of the ejaculate was frozen-thawed using a standardized protocol to determine their freezability (good and bad freezers), depending on the percentage of intact plasma membrane and total motile post-thaw sperm. For analysis of “ChemSensor”, sperm samples were volatilized and ionized, decomposing into different ions with a determined size, which were used as discrimination variables to determine if it is possible the separation of ejaculates into two freezability groups. The “ChemSensor” was able to discriminate all the ejaculates of the good freezers; and inside of bad freezers group only one sample was misclassified. Due to the small number of samples, the distance obtained between groups (2.62), although not very high, it could be considered significant, although a larger sample in order to create a more robust mathematical model would be necessary. In conclusion, the “ChemSensor” is a possible suitable tool for the early discrimination of Iberian pig ejaculates according to their freezability.

The laboratory diagnosis of African horse sickness (AHS) is important for: (a) demonstrating freedom from infection in a population, animals or products for trade (b) assessing the efficiency of eradication policies; (c) laboratory confirmation of clinical diagnosis; (d) estimating the prevalence of AHS infection; and (e) assessing postvaccination immune status of individual animals or populations. Although serological techniques play a secondary role in the confirmation of clinical cases, their use is very important for all the other purposes due to their high throughput, ease of use and good cost‐benefit ratio. The main objective of this study was to support the validation of AHS VP7 Blocking ELISA up to the Stage 3 of the World Animal Health Organization (OIE) assay validation pathway. To achieve this, a collaborative ring trial, which included all OIE Reference Laboratories and other AHS‐specialist diagnostic centres, was conducted in order to assess the diagnostic performance characteristics of the VP7 Blocking ELISA. In this trial, a panel of sera of different epidemiological origin and infection status was used. Through this comprehensive evaluation we can conclude that the VP7 Blocking ELISA satisfies the OIE requirements of reproducibility. The VP7 Blocking ELISA, in its commercial version is ready to enter Stage 4 of the validation pathway (Programme Implementation). Specifically, this will require testing the diagnostic performance of the assay using contemporary serum samples collected during control campaigns in endemic countries. [ABSTRACT FROM AUTHOR]

The aim of this study was to evaluate the combined use of fluorescenceand phase contrast microscopy as a new technique for the assessmentof sperm viability. For this, five pools of semen from three fertile boarswere frozen and thawed according to a standard protocol. Afterthawing, sperm samples were stained with 5 ll of propidium iodide(PI, 0.5 mg/ml) and incubated at 37°C in darkness 10 min. Subse-quently, samples were analyzed using a phase contrast microscopy(Nikon Eclipse E400, Tokyo, Japan) coupled with fluorescenceequipment (Nikon C-SHG1) with a mercury lamp (100 W) and aNikon G-2A filter with excitation/barrier filter of 510/590 which allowsa simultaneous excitation of blue and green. After displaying thesample under phase contrast microscopy the fluorescence wasconnected, non-viable sperm showed red color, and viable sperm werenot stained and visible. To determine the validity of this technique thesamples were also analyzed with a dual fluorescent staining (SYBR-14/PI) under fluorescence microscopy (Garner and Johnson, 1995; BiolReprod. 53, 276–284). The results did not show (57.6 vs. 57.2 1.5%live sperm; Garner and Johnson’s and described technique respec-tively) difference between techniques (p > 0.05). In conclusion, thecombined use of fluorescence and phase contrast microcopy using a PIstaining may be used for the determination of sperm viability, saving inthe use of fluorochromes and viable sperm are visible which wouldsimultaneously allow the acrosome state assessment.

The aim of the present study was to determine whether the levels of reactive oxygen species (ROS) substances production and the levels of lipid peroxidation of the sperm membrane were related to the quality that the ejaculates exhibited after cryopreservation in boars. Ejaculates from 42 healthy boars were used in this study and they were cryopreserved with the lactose-egg yolk extender (LEY). Several sperm quality parameters were assessed by flow cytometry in samples incubated for 30 and 150 min at 37°C after thawing: the percentage of sperm with intact plasma membrane (SIPM), intracellular reactive oxygen substances production through mean of DCF fluorescence intensity of total sperm (mean-DCF) and the percentage of viable and non-viable sperm containing oxidized BODIPY (VSOB and NVSOB). In addition, the percentages of total motile (TMS) and progressively motile sperm (PMS) were assessed at the same incubation times with a computer-assisted sperm analysis system. The classification of the ejaculates into good or bad freezers was performed through hierarchical cluster analysis from SIPM and TMS at 150 min post-thawing. The ejaculates of those males classified as good freezers exhibited higher (p 0.05) VSOB, NVSOB and mean-DCF. Therefore, these results show that lipid peroxidation and the amount of reactive oxygen substances in the sperm after cryopreservation are similar between boars classified as good or bad freezers.

42. Efecto de la immunocastración y de la castración quirúrgica sobre los rendimientos productivos y la calidad de la canal en cerdas Ibéricas de cebo

- Abstract
Se utilizaron 192 cerdas Ibéricas (Duroc x Ibérica) con 110 días de vida y 44,94 ± 3,41 kg de peso, para probar el efecto de la castración inmunológica con Improvac® (Pfizer Animal Health). El ensayo se llevó a cabo en el Centro de Pruebas de Porcino del Instituto Tecnológico Agrario de Castilla y León (Itacyl, Consejería de Agricultura y Ganadería). El experimento se diseñó en bloques al azar, con 3 tratamientos (cerdas castradas quirúrgicamente, enteras e inmunocastradas), 48 réplicas totales de 4 cerdas cada una y 16 réplicas por tratamiento. Los animales se pesaron individualmente cada 15 días y se calculó el consumo de pienso, la ganancia de peso y el índice de conversión. Una vez aplicadas las dos dosis de Improvac® (a las 18 y 22 semanas de edad -60 kg y 84,2 kg de peso, respectivamente-) las diferencias de peso vivo entre enteras e inmunocastradas se volvieron estadísticamente significativas (P < 0,05) por encima de los 100 kg, permaneciendo la significación hasta los 170 kg, y presentando éstas últimas mayor consumo, ganancia media diaria e índice de conversión (21,25, 19,24 y 1,44%, respectivamente; P < 0,05). En relación con las castradas y tras la aplicación de la 2ª dosis, las inmunocastradas aumentaron el consumo (12% de media, en función del peso vivo considerado) y la ganancia diaria (22,24%), disminuyendo la conversión un 7,6% (P < 0,05). Por otra parte, las castradas consumieron más (8,35%), con menor ganancia diaria (2,6%) y una mayor conversión (11%) que las enteras (P < 0,05). Un primer lote de 96 cerdas fue enviado a matadero a los 8,2 meses de edad, con pesos de 162,88 kg (castradas), 164,52 kg (enteras) y 170,76 kg (inmunocastradas). Para las cerdas de este lote, el rendimiento de la canal fue de 78,02 vs 79,14 y 78,79%, para enteras, castradas e inmunocastradas respectivamente (P < 0,05), mientras que el peso de piezas nobles fue mayor en las enteras e inmunocastradas (P < 0,05). En las condiciones de nuestro trabajo, podemos concluir que Ia inmunocastración es una alternativa productiva ventajosa a la castración física en cerdas Ibéricas, recomendándose una separación de 10 ± 2 semanas entre la aplicación de la 2ª dosis de Improvac® y el sacrificio para la obtención del máximo beneficio, e igualmente, con las mismas pautas de aplicación, presenta una conversión más favorable a los distintos pesos de sacrificio comerciales en relación con cerdas enteras. La castración quirúrgica es la opción menos interesante.

Contents Sperm cryosurvival rates are not optimal for most species. Therefore, new cryopreservation strategies are needed with the objective of increasing the number of surviving sperm and the quality of those sperm after thawing. Cholesterol plays important roles in many sperm functions, including effects on membrane properties. One of these effects is to stabilize membranes at low temperatures. Thus, species that produce sperm which possess high membrane cholesterol : phospholipid ratios are more resistant to cold shock than sperm with low cholesterol : phospholipid ratios. Therefore, increasing the cholesterol content of sperm membranes may be a strategy that can improve sperm quality after freeze-thawing. In this review, information is presented related to using cyclodextrins pre-loaded with cholesterol for cryopreserving sperm from different species. The topics discussed include both in vitro and in vivo assessments of sperm quality after cryopreservation, as well as how increasing sperm cholesterol content affects other sperm functions.
