In this study, an Artificial Neural Network (ANN) model is used to solve the complex task of producing fresh cheese with the desired quality parameters. The study focuses on kombucha fresh cheese samples fortified with ground wild thyme, supercritical fluid extract of wild thyme, ground sage and supercritical fluid extract of sage and optimizes the parameters of chemical composition, antioxidant potential and microbiological profile. The ANN models demonstrate robust generalization capabilities and accurately predict the observed results based on the input parameters. The optimal neural network model (MLP 6-10-16) with 10 neurons provides high r2 values (0.993 for training, 0.992 for testing, and 0.992 for validation cycles). The ANN model identified the optimal sample, a supercritical fluid extract of sage, on the 20th day of storage, showcasing specific favorable process parameters. These parameters encompass dry matter, fat, ash, proteins, water activity, pH, antioxidant potential (TP, DPPH, ABTS, FRAP), and microbiological profile. These findings offer valuable insights into producing fresh cheese efficiently with the desired quality attributes. The Thymus serpyllum L. is a widespread in the steppe bioсenosеs of southern and eastern Ukraine. It is well cultivated in specialized farms. A promising direction of modern phytochemical research of wild and cultivated Thymus L. species is the determination of the accumulation of biologically active polyphenolic compounds that exhibit pronounced biological activity. The purpose of the work is to determine the accumulation of polyphenolic compounds in Thymus serpyllum L. herb at the flowering using TLC and HPLC methods. The Thymus serpyllum L. herb was harvested in steppe biocenoses of southern and eastern Ukraine at the flowering, during the period of biologically active polyphenolic compounds maximum accumulation. For experimental studies, alcohol extracts from Thymus serpyllum L. herb (1:100) were used. The substances component composition was studied by TLC method Biostep CD 60 densitometer (Germany) and HPLC Agilent 1260 Infinity HPLC System Open LABCDS Software (Japan). TLC and HPLC methods revealed up to 25 polyphenolic compounds. Experimentally were determined 13 flavonoids and 12 hydroxycinnamic acids. All these biologically active compounds are well known in modern phytotherapy for their pronounced antimicrobial, anti-inflammatory and antioxidant activity. During the flowering period up to 25 polyphenolic compounds were identified and quantified in the ethanolic extract (1:100) obtained from Thymus serpyllum L. herbal raw material. Of these, 13 compounds were attributed to flavonoids with a defined total content (0.3 mg/100 mg) and 12 to hydroxycinnamic acids (0.9 mg/100 mg). The highest accumulation of hydroxycinnamic acids in the Thymus serpyllum L. herbal raw material were characterized by rosmarinic acid (12.77 + 1.19%), chlorogenic acid (8.55 + 0.79%) and p-coumaric acid (7.41 + 0.69%). Of flavonoids, the highest content were established for rutin (8.45 + 0.79%), luteolin (3.87 + 0.37%), luteolin-7-O-β-D-glucopyranoside (3.86 ± 0.33%), apigenin-7,4'-diglucoside (3.86 ± 0.32%), quercetin (3.85 ± 0.34%) and apigenin (3.81 ± 0.36%). The results obtained by TLC and HPLC methods indicate the perspective of polyphenolic compounds research for the purpose of determining the biological activity of extracts and carrying out the standardization of Thymus serpyllum L. herb. The study of the polyphenolic compounds accumulation in the Thymus serpyllum L. plant raw materials is important for this standardization and obtaining effective phytopreparations.

The objective of this study was to spectrophotometric determinate the total phenolic, flavonoid, hydroxycinnamic acid, and flavonol content of orange, pineapple, and apple juices fortified with wild thyme (Thymus serpyllum L.), Dalmatian sage (Salvia officinalis L.), and wild thyme-Dalmatian sage (3: 1, v / v) extracts, and to evaluate their DPPH radical scavenging activity as a contribution to the development of a new functional beverage. The plant extracts addition increased the amount of phenolic compounds in fruit juices and improved their antioxidant properties. The highest concentrations of bioactive compounds and the greatest DPPH radical activity were obtained by adding Dalmatian sage extract to orange juice. Our study provides the novelty of fortifying fruit juices with wild thyme and Dalmatian sage extracts and offers significant potential for the creation of functional beverages. Especially their antineoplastic properties have been thoroughly studied with respect to their putative application in chemoprevention and adjuvant therapy of cancer. However, the most known biological effects of Lamiaceae have been ascribed to their essential oil fractions, whereas their (poly)phenolic metabolites being also abundant in these plants, are much less recognized, nevertheless contributing to their beneficial properties, such as anti-cancer actions. The aim of this study was to evaluate the impact of dried aqueous extracts from common thyme (Thymus vulgaris L.) (ExTv), wild thyme (Thymus serpyllum L.) (ExTs), sweet marjoram (Origanum majorana L.) (ExOm), and peppermint (Mentha × piperita L.) (ExMp), as well as (poly)phenolic compounds: caffeic acid (CA), rosmarinic acid (RA), lithospermic acid (LA), luteolin-7-O-β-glucuronide (Lgr), luteolin-7-O-rutinoside (Lr), eriodictyol-7-O-rutinoside (Er), and arbutin (Ab), on unstimulated Jurkat cells, in comparison with their effect on staurosporine-stimulated Jurkat cells. Jurkat T cells were incubated with different concentrations of ExTv, ExTs, ExOm, ExMp, Lgr, LA, Er, Lr, RA, CA, or Ab. Subsequently, staurosporine was added to half of the samples and flow cytometry combined with fluorescence-activated cell sorting analysis was conducted, which allowed for the selection of early and late apoptotic cells. Both ExTs and ExOm stimulated apoptosis of Jurkat cells and enhanced the proapoptotic effect of staurosporine. Conversely, ExTv and ExMp demonstrated no clear effect on apoptosis. CA and RA raised the staurosporine-induced apoptotic effect. The impact of Er and Lgr on Jurkat cells showed fluctuations depending on the compound concentration. Neither Er nor Ab altered staurosporine-induced apoptosis in Jurkat cells, whereas Lgr seemed to weaken the proapoptotic action of staurosporine. The most evident observation in this study was the pro-apoptotic action of ExTs and ExOm observed both in staurosporine-unstimulated and stimulated Jurkat cells. Additionally, an enhancement of staurosporine-induced apoptosis by caffeic and rosmarinic acids was reported. Functional beverages based on herbal extracts are highly demanded products due to the presence of bioactives with promising health benefits and interesting and characteristic sensory properties. Mediterranean medicinal and aromatic herbs contain a wide range of bioactives (non-volatile polyphenols, volatile terpenes) that are important constituents of herbal extracts and essential oils. The antioxidant capacity and potential health benefits of these bioactives could be associated with their synergistic effects. Therefore, this study aimed to characterize the non-volatile and volatile bioactives of sage (Salvia officinalis L.), wild thyme (Thymus serpyllum L.) and laurel (Laurus nobilis L.) aqueous extracts and their two- and three-component mixtures as well as their antioxidant capacity. The content of total phenols, flavonoids, hydroxycinnamic acids and flavonols was determined spectrophotometrically. Individual polyphenols were analyzed by LC-MS/MS, the volatiles were analyzed by HS-SPME/GC-MS, and the antioxidant capacity was analyzed by ORAC and DPPH assays. The results showed that aqueous extracts of all examined herbs and their mixtures contained a high content of phenolic compounds ranging from 0.97 to 2.79 g L−1 of the sample, among which the most common were flavonols. At the same time, mono- and sesquiterpenes were the main volatiles. All extracts showed high antioxidant capacity, especially L. nobilis (781.62 ± 5.19 μmol TE mL−1 of the sample in the DPPH assay; 1896.10 ± 8.77 μmol TE mL−1 of the sample in the ORAC assay) and the two-component mixture of L. nobilis and T. serpyllum (679.12 ± 5.19 μmol TE mL−1 in the DPPH assay; 1913.38 ± 8.77 μmol TE mL−1 in the ORAC assay). Mixtures of herbal extracts have been shown to possess additive or synergistic effects, consequently contributing to higher antioxidant capacity. Featured Application: The data obtained can be useful for obtaining high-quality food and pharmacopoeia raw materials from wild thyme with a given chemical composition and controlled biological activity. Plants of the genus Thymus L. are traditionally used in medicine and cooking due to the presence of biologically active compounds in them that have fungicidal, antibacterial and other medicinal properties and original taste qualities. Genetic features and growing conditions cause the elemental composition, responsibly of the synthesised medicinal compounds. However, information on the contents and distributions of elements in the organs of Thymus L. is very limited. This study was to set and compare the elements in organs of wild thyme for different soil and climatic conditions. Two species of wild Thymus L. from Mongolian steppe and on the coast of Lake Baikal were collected during flowering. Twenty-four elements, including Si, in soils, roots, stems, leaves and flowers were simultaneously determined by atomic emission spectrometry. Elemental profiles of two species of wild Thymus L. are described. It is assumed that Si is a necessary element of the plant. Summary: This work compared the effects of pulsed electric fields (PEF) and ultrasound (US) technologies on the extent of cell disintegration of two Mediterranean herb tissues (Origanumvulgare L., Thymusserpyllum L.), as well as on the extractability of phenolic compounds during the subsequent hydroalcoholic extraction (0%–50% ethanol in water, v/v) for up to 4 h. The rate of phenolic compounds extraction decreased with time and was predicted rather satisfactorily (R2 = 0.898–0.989) by the Peleg's model. The application of either PEF or US treatment prior to solid–liquid extraction (SLE) has the potential to reduce duration and concentration of ethanol to achieve the same recovery yield of phenolic compounds. Under optimised PEF (3 kV cm−1, 10 kJ kg−1) and US (400 W, 20 min) treatment conditions, the extracts obtained from either PEF or US pretreated herb samples showed higher total phenolic yield (36% on average) and antioxidant activity (FRAP) (36% on average) as compared to the control extraction, especially when 25% ethanol was used as a solvent. Wild thyme (Thymus serpyllum L.) herbal dust has been recognized as a potential underutilized resource for the recovery of antioxidants. The aim of this paper was to optimize natural deep eutectic solvent (NADES) extraction of polyphenols to obtain improved antioxidant activity of extracts determined by selected in vitro assays (DPPH, FRAP, and ABTS). Twenty different NADES systems were investigated in the first step of the screening of the extraction solvent and l-proline (Pro)–glycerine (Gly) based solvents provided the best results. Preliminary experiments organized by 25−1 fractional factorial design narrowed down the number of extraction factors from five (temperature, extraction time, NADES type, water content and L/S ratio) to three and determined their experimental domain for the final step. A face-centered central composite design with temperature (40–55–70 °C), extraction time (60–120–180 min) and L/S ratio (10–20–30 g NADES/g sample) was applied for influence analysis and process optimization. Multi-response optimization suggested a temperature of 65 °C, time of extraction of 180 min and L/S ratio of 28 g NADES/g DW as optimal extraction parameters. Experimental validation confirmed good agreement between experimental and predicted results in the extract obtained at optimal conditions and the interactions in the most suitable NADES (N16; Pro–Gly–H2O; 1:2:1) were confirmed by the 1H-NMR.

The quality of medicinal raw materials is determined by the content of pharmacologically significant compounds. Chemical composition of some medicinal plant species is relatively constant. However, some species, in particular, creeping thyme are characterized by great chemical polymorphism. Nowadays creeping thyme is collected from nature in Russia. There are many varieties that can be used not only in cookery, but also as a medicine. The article presents the comparative study of the essential oil content and component composition, the total content of polyphenols, tannins and flavonoids in the raw materials of three varieties and one wild population of Thymus serpyllum L. The results of the essential oil composition study suggest that the wild population from VILAR and varieties 'Purpurnovioletovij’ and ‘Variegata’ belong to the species Thymus serpyllum L., while the variety ‘Picantny’ probably is a hybrid.

Вивчено біологічні особливості таких цінних лісових видів, як Carex brevicollis DC, Thymus serpyllum L. та Malva excisa Rchb., що забезпечить успішне збереження їх генофонду й послужить основою для подальшої розробки технологій вирощування для отримання лікарської сировини.

The presented study revealed that there were changes in the phenolic compounds profile of extract of wild thyme (Thymus serpyllum L.) after γ-irradiation at the dose of 5 kGy. Ethanolic extracts of irradiated and non-irradiated herb were prepared and their compounds were analyzed by RP-HPLC-DAD technique. Between thirty two detected constituents, twelve phenolic compounds classified as hydroxybenzoic and hydroxycinnamic acids derivatives, flavones and flavanones were identified. Among them, caffeic acid derivatives and flavones predominated with the highest content of rosmarinic acid and luteolin-7-O-glucoside, respectively. The aim of the research has been to assay the contents of Zn, Cu, Mn, Fe, and Pb in soil and in the organs of wild Thymus serpyllum L. acquired from three natural habitats from the Kuyavia-Pomerania Province (Poland) not exposed to contamination with metals. As compared with the contents of the geochemical background of the soils in the region and the value of enrichment factor (EF), there was noted a considerable content of Pb in the soil from two locations. The Pb content in plant tissues in one of the three locations was higher than the admissible content specified by the World Health Organization WHO (above 10 mg kg−1 d.w.). As for the Zn content, only the plants from the locations with the relatively youngest phytocenosis met the conditions for herbs to be used for phytotherapy. The Zn content in the dry weight of the plants collected from the other two locations exceeded 50 mg kg−1. The risk of an elevated content of Zn and Pb in the plants makes it necessary to monitor the soil environment and to limit the collection of the plants from natural habitats, as well as to encourage the collection from controlled habitats.

The aim of this study was to valorize Thymus serpyllum herbal dust, a particular fraction distinguished as an industrial waste from filter-tea production. Pressurized liquid extraction (PLE) was used with the aim of overcoming certain obstacles of conventional extraction techniques in terms of shortening extraction time, reducing solvent consumption and energy costs, using “green” solvents and obtaining high yield and quality products. In order to optimize PLE of T. serpyllum herbal dust, the preliminary screening of the independent variables in order to define the most influential parameters and their domain was done first. After the screening, the optimization study using the face-centered central composite experimental design (CCD) with response surface methodology (RSM) was implemented. Additionally, taking into account the high awareness of the positive influence of antioxidants on the human health and associating it with high content of polyphenolic compounds in various members of Lamiaceae family, PLE has proven to be a great approach for antioxidants recovery from T. serpyllum herbal dust.

Herbal remedies like the Thymus serpyllum L. is useful in traditional medicine for the treatment of many diseases especially congestion, and bronchitis. The purpose of this study was to formulate a micro-emulsion, a gel and an ointment containing the plant hydro distilled thymus oil extracted from Thymus serpyllum L. collected from Ziarat, Balochistan. The prepared formulations were subjected to in-vitro and ex vivo study release, High performance Liquid Chromatography (HPLC), Thin Layer Chromatography (TLC), to justify their suitability for topical use. The in-vitro and ex-Vivo release was studied using Franz Cells and using two different kinds of membrane synthetic dialysis cellulose membrane and natural rabbit skin, and the amount of drug released was determined by HPLC at λ 274nm. The three formulations result obtained through dialysis cellulose membrane showed the faster release than the natural rabbit skin. However, the micro-emulsion, gel formulation showed the same release except ointment. The release from the above mentioned formulation can be arranged in the following descending order. micro-emulsion > Gel > Ointment. The best fit of release kinetics was achieved by Krosmeyer- Peppas, the TLC and HPLC identifies the Thymol, isolation and quantification of the marker. This study demonstrates that it is necessary to assess the impact of release and permeability pattern of different formulations. Wild thyme (Thymus serpyllum L.) is a popular remedy regarding both traditional and conventional medicine. It is used as the antiseptic, aromatic, expectorant, stomachic, antispasmodic, carminative and preservative substance. For the purpose of this paper, wild thyme essential oil was isolated from the dried herb T. serpyllum by hydrodistillation. Original semi-industrial distillation device SP-130 performing distillation by water and steam was used for the hydrodistillation of T. serpyllum essential oils. The temperature during the hydrodistillation in the device SP-130 ranged from 100°C - 102°C at atmospheric pressure, and the whole process lasted 5 hours. The isolated essential oil is a liquid of light yellow colour and the odour characteristic of the genus Thymus. Obtained yield of essential oil was 0.08 %, with 65 components identified in the tested essential oil. The most represented chemical groups are sesquiturpene hydrocarbons with 35.1%, and oxygenated sesquiturpenes with 34.8%. The main components of essential oil of T. serpyllum were: trans-nerolidol (24.2%), germacrene D (16.0%), thymol (7.3%), δ-cadinene (3.7%) and β-bisabolene (3.3%). The essential oil showed significantly better ability to neutralize DPPH free radicals (IC50 = 0.503 μL / mL) compared with synthetic antioxidants BHA and BHT. Synthetic chemical compounds such as BHA and BHT are used in food industry as antioxidants due to their ability to prolong the shelf-life of foodstuffs by protecting them against deterioration caused by oxidation, such as fat rancidity, colour changes, degradation of the flavor and loss of nutrient value. In recent years, there is a considerable interest in finding natural compounds that could replace sinthetic antioxidants because of adverse toxicological reports on many synthetic compounds. Lamiaceae herbs and their essential oils or extracts application has proven to be the effective preservation agents for the extension shelf-life of foodstuffs, indicating their potential use in food industry as functional ingredients and food additives. The antioxidant activity of extracts from some Lamiaceae herbs is comparable to that of the most common synthetic antioxidants BHT and BHA. Free radicals are a major cause of many degenerative diseases, such as atherosclerosis, cancer, cardiovascular diseases, inflammatory bowel diseases, skin aging, old age dementia and arthritis. Epidemiological data and randomized clinical trials provide sample indications that antioxidants play a fundamental role in the prevention of cancer and cardiovascular diseases. They act as scavengers of reactive oxygen species and metal chelators that protect human cells and reduce oxidative damages. Natural antioxidants are important in the food industry because they can have a double functionality, that is, they can be useful as a food preservation agents while providing important health benefits for humans by maintaining our health and preventing disease. This investigation showed that the essential oil of T. serpyllum can be an important source for the production and application in the food industry as nutritional supplements, functional food components or natural food antioxidants.

The effect of different concentrations (0.05, 0.1 and 0.2%) of Thymus serpyllum L. aqueous extracts on lipid peroxidation process (LP), as well as reduced glutathione content (GSH) in leaves and roots of soybean seedlings were examined 24 and 72 h after the treatment. Our results showed that only highest concentration of the extract used (0.2%) enhanced process of lipid peroxidation, while concentration of 0.1% stimulated GSH accumulation in soybean seedlings.

Evaluation of productivity of arrays of Thymus serpyllum L. in various phytocenoses in East Transbaikalia, conducted by the method of work in key areas, showed that the highest crop capacity of its raw materials accounted for the steppes and depends primarily on the species’s projective cover of the occupied areas. The total area of the identified the jungle is 34006 hectares, the production area is 2210 ha. The determined biological reserve in the investigated area is 841,58 tons, the possible annual harvesting volume is 306,49 tons. The research allowsto attribute this speciesas perspective for harvesting.

The aim of present study was to evaluate and compare the hypoglycemic activity of different solvents extracts of Thymus serpyllum in rabbits. Diabetes was induced with single intravenous injection of alloxan monohydrate (150mg/kg). Glibenclamide and acarbose were used as standard drugs. The crude powder of Thymus serpyllum (500 mg/kg b.w) significantly reduced blood glucose level in both normal and diabetic rabbits. Various extracts of Thymus serpyllum were compared for their hypoglycemic activity in diabetic rabbits. Ether and aqueous extracts significantly reduced the blood glucose level with maximum effect (p<0.001) produced by aqueous extract, which was selected for further study. Aqueous extract significantly inhibited the rise in glucose level in oral glucose tolerance test. The extract showed synergistic effect with different doses of insulin; however serum insulin level of the diabetic rabbits was not significantly increased by the extract. HbA1c level was significantly (p<0.05) reduced whereas hemoglobin level was significantly increased in three months study. Phytochemical screening of the aqueous extract showed the presence of alkaloids, flavonoids, tannins, terpinoids, reducing sugar and cardiac glycosides. It is concluded that the aqueous extract might be used alone or in combination with insulin to manage diabetes and its associated complications. [ABSTRACT FROM AUTHOR]

Wild thyme (Thymus serpyllum L.) herbal dust has been recognized as a potential underutilized resource for the recovery of antioxidants. The aim of this paper was to optimize natural deep eutectic solvent (NADES) extraction of polyphenols to obtain improved antioxidant activity of extracts determined by selected in vitro assays (DPPH, FRAP, and ABTS). Twenty different NADES systems were investigated in the first step of the screening of the extraction solvent and l-proline (Pro)–glycerine (Gly) based solvents provided the best results. Preliminary experiments organized by 25−1 fractional factorial design narrowed down the number of extraction factors from five (temperature, extraction time, NADES type, water content and L/S ratio) to three and determined their experimental domain for the final step. A face-centered central composite design with temperature (40–55–70 °C), extraction time (60–120–180 min) and L/S ratio (10–20–30 g NADES/g sample) was applied for influence analysis and process optimization. Multi-response optimization suggested a temperature of 65 °C, time of extraction of 180 min and L/S ratio of 28 g NADES/g DW as optimal extraction parameters. Experimental validation confirmed good agreement between experimental and predicted results in the extract obtained at optimal conditions and the interactions in the most suitable NADES (N16; Pro–Gly–H2O; 1:2:1) were confirmed by the 1H-NMR. Variations in the essential oil composition of Thymus serpyllum L., growing wild in Estonia (33 samples) and in some other countries (Russia, Latvia and Armenia, seven samples) were determined. The oil were obtained from Estonia (46 samples) in yields 0.6-4.4 and 1.9-8.2 mL kg-1 in other countries. The T. serpyllum herb grown in Estonia usually did not confirmed to the EP standard in the aspect of the essential oil contents (3.0 mL kg-1). Variations in the essential oil composition of wild thyme were studied using capillary gas chromatographic methods. A total of 94 components were identified. Thymol and carvacrol, mentioned in literature as principial components, are not the main components of the essential oil of wild thyme growing in Estonia. The aim of this study was to valorize Thymus serpyllum herbal dust, a particular fraction distinguished as an industrial waste from filter-tea production. Pressurized liquid extraction (PLE) was used with the aim of overcoming certain obstacles of conventional extraction techniques in terms of shortening extraction time, reducing solvent consumption and energy costs, using "green" solvents and obtaining high yield and quality products. In order to optimize PLE of T. serpyllum herbal dust, the preliminary screening of the independent variables in order to define the most influential parameters and their domain was done first. After the screening, the optimization study using the face-centered central composite experimental design (CCD) with response surface methodology (RSM) was implemented. Additionally, taking into account the high awareness of the positive influence of antioxidants on the human health and associating it with high content of polyphenolic compounds in various members of Lamiaceae family, PLE has proven to be a great approach for antioxidants recovery from T. serpyllum herbal dust. The aim of the research has been to assay the contents of Zn, Cu, Mn, Fe, and Pb in soil and in the organs of wild Thymus serpyllum L. acquired from three natural habitats from the Kuyavia-Pomerania Province (Poland) not exposed to contamination with metals. As compared with the contents of the geochemical background of the soils in the region and the value of enrichment factor (EF), there was noted a considerable content of Pb in the soil from two locations. The Pb content in plant tissues in one of the three locations was higher than the admissible content specified by the World Health Organization WHO (above 10 mg kg−1 d.w.). As for the Zn content, only the plants from the locations with the relatively youngest phytocenosis met the conditions for herbs to be used for phytotherapy. The Zn content in the dry weight of the plants collected from the other two locations exceeded 50 mg kg−1. The risk of an elevated content of Zn and Pb in the plants makes it necessary to monitor the soil environment and to limit the collection of the plants from natural habitats, as well as to encourage the collection from controlled habitats.
