International audience; Phelan-McDermid syndrome (PMS) is a 22q13.3 deletion syndrome that presents with a disturbed development, neurological and psychiatric characteristics, and sometimes other comorbidities like seizures. The epilepsy manifests itself in a variety of seizure semiologies. Further diagnostics using electroencephalogram (EEG) and brain magnetic resonance imaging (MRI) are important in conjunction with the clinical picture of the seizures to decide whether anticonvulsant therapy is necessary. As part of the development of European consensus guidelines we focussed on the prevalence and semiology of epileptic seizures in PMS associated with a pathogenic variant in the SHANK3 gene or the 22q13 deletion involving SHANK3, in order to then be able to make recommendations regarding diagnosis and therapy.

THOC6 is the genetic basis of autosomal recessive THOC6 Intellectual Disability Syndrome (TIDS). THOC6 facilitates the formation of the Transcription Export complex (TREX) tetramer, composed of four THO monomers. The TREX tetramer supports mammalian mRNA processing that is distinct from yeast TREX dimer functions. Human and mouse TIDS model systems allow novel THOC6-dependent TREX tetramer functions to be investigated. Biallelic loss-of-function (LOF) THOC6 variants do not influence the expression and localization of TREX members in human cells, but our data suggests reduced binding affinity of ALYREF. Impairment of TREX nuclear export functions were not detected in cells with biallelic THOC6 LOF. Instead, mRNA mis-splicing was observed in human and mouse neural tissue, revealing novel insights into THOC6-mediated TREX coordination of mRNA processing. We demonstrate that THOC6 is required for regulation of key signaling pathways in human corticogenesis that dictate the transition from proliferative to neurogenic divisions that may inform TIDS neuropathology.

Copolymer hydrogel (PVA-g-AA) having varied PVA (Polyvinyl alcohol) and AA (Acrylic acid) content is prepared by gamma induced radiation polymerization. The parameters affecting the gel fraction yield have been studied. The gel fraction and the swelling property are found to be 92.39 and 905% respectively at an absorbed dose of 20 kGy. Structural and property characteristics were determined by Fourier Transform Infrared (FTIR) spectrometer and Differential Scanning Calorimetry (DSC). The surface morphology of PVA and copolymer has been studied with Scanning Electron Microscope (SEM). The factors affecting the metal uptake such as pH, time, and initial feed metal concentration were investigated. It is found that at pH 5 and after 240 minutes the maximum adsorption amount are 178, 161, 117, and 110 mg/g for Pb2+, Zn2+, Co2+, and Ni2+ respectively.

Internal migration plays a crucial role in influencing real changes in local area population size and in facilitating the efficient functions of regional and national labor market and housing. This study is conducted to investigate the elements at the household level and others at the provincial level, which have impacts on the decision of personal migration. We used the database of household living standards surveys in 2010, 2012, and 2014 in Vietnam. Logit model was applied with array data to analyze determinants influencing the decision of personal migration. The results show that households with low living standards have a higher probability of migration. The provincial competitiveness index has a positive impact on the probability of households with migrants. The results provide the local authority with the foundation for regulation and distribution of the labor market on an effective basis. Based on the findings, some implications are proposed in the context of emerging countries and Vietnam as the case study

The evolutionarily conserved hedgehog (Hh) pathway is essential for organogenesis and plays critical roles in postnatal tissue maintenance and renewal. A unique feature of the vertebrate Hh pathway is that signal transduction requires the primary cilium (PC) where major pathway components are dynamically enriched. These factors include smoothened (SMO) and patched, which constitute the core reception system for sonic hedgehog (SHH) as well as GLI transcription factors, the key mediators of the pathway. Here, we report biallelic loss-of-function variations in SMO in seven individuals from five independent families; these variations cause a wide phenotypic spectrum of developmental anomalies affecting the brain (hypothalamic hamartoma and microcephaly), heart (atrioventricular septal defect), skeleton (postaxial polydactyly, narrow chest, and shortening of long bones), and enteric nervous system (aganglionosis). Cells derived from affected individuals showed normal ciliogenesis but severely altered Hh-signal transduction as a result of either altered PC trafficking or abnormal activation of the pathway downstream of SMO. In addition, Hh-independent GLI2 accumulation at the PC tip in cells from the affected individuals suggests a potential function of SMO in regulating basal ciliary trafficking of GLI2 when the pathway is off. Thus, loss of SMO function results in abnormal PC dynamics of key components of the Hh signaling pathway and leads to a large continuum of malformations in humans.

By providing a range of values rather than a point estimate, accurate interval forecasting is critical to the success of investment decisions in exchange rate markets. This work proposes a sliding-window metaheuristic optimization for interval-valued time series forecasting using multi-output least squares support vector regression (MLSSVR). The hyperparameters in MLSSVR are finetuned using an accelerated particle swarm optimization algorithm to yield the best predictions and fastest convergence. The proposed system has a graphical user interface that is developed in a computing environment and functions as a stand-alone application. The system is validated using stock prices as well as exchange rates and outputs are compared with published results. Finally, the proposed interval time series prediction method is tested in two case studies; one involves the daily Australian dollar and Japanese yen rates (AUD/JPY) and the other involves US dollar and Canadian dollar rates (USD/CAD). The proposed model is promising for interval time series forecasting.

The photosystem II PsbS protein of thylakoid membranes is responsible for regulating the energy-dependent, non-photochemical quenching of excess chlorophyll excited states as a short-term mechanism for protection against high light (HL) stress. However, the role of PsbS protein in long-term HL acclimation processes remains poorly understood. Here we investigate the role of PsbS protein during long-term HL acclimation processes in wild-type (WT) and npq4-1 mutants of Arabidopsis which lack the PsbS protein. During long-term HL illumination, photosystem II photochemical efficiency initially dropped, followed by a recovery of electron transport and photochemical quenching (qL) in WT, but not in npq4-1 mutants. In addition, we observed a reduction in light-harvesting antenna size during HL treatment that ceased after HL treatment in WT, but not in npq4-1 mutants. When plants were adapted to HL, more reactive oxygen species (ROS) were accumulated in npq4-1 mutants compared to WT. Gene expression studies indicated that npq4-1 mutants failed to express genes involved in plastoquinone biosynthesis. These results suggest that the PsbS protein regulates recovery processes such as electron transport and qL during long-term HL acclimation by maintaining plastoquinone biosynthetic gene expression and enhancing ROS homeostasis.

Hirschsprung disease (HSCR) is a complex genetic disease characterized by absence of ganglia in the intestine. HSCR etiology can be explained by a unique combination of genetic alterations: rare coding variants, predisposing haplotypes and Copy Number Variation (CNV). Approximately 18% of patients have additional anatomical malformations or neurological symptoms (HSCR-AAM). Pinpointing the responsible culprits within a CNV is challenging as often many genes are affected. Therefore, we selected candidate genes based on gene enrichment strategies using mouse enteric nervous system transcriptomes and constraint metrics. Next, we used a zebrafish model to investigate whether loss of these genes affects enteric neuron development in vivo. This study included three groups of patients, two groups without coding variants in disease associated genes: HSCR-AAM and HSCR patients without associated anomalies (HSCR-isolated). The third group consisted of all HSCR patients in which a confirmed pathogenic rare coding variant was identified. We compared these patient groups to unaffected controls. Predisposing haplotypes were determined, confirming that every HSCR subgroup had increased contributions of predisposing haplotypes, but their contribution was highest in isolated HSCR patients without RET coding variants. CNV profiling proved that specifically HSCR-AAM patients had larger Copy Number (CN) losses. Gene enrichment strategies using mouse enteric nervous system transcriptomes and constraint metrics were used to determine plausible candidate genes located within CN losses. Validation in zebrafish using CRISPR/Cas9 targeting confirmed the contribution of UFD1L, TBX2, SLC8A1, and MAPK8 to ENS development. In addition, we revealed epistasis between reduced Ret and Gnl1 expression and between reduced Ret and Tubb5 expression in vivo. Rare large CN losses—often de novo—contribute to HSCR in HSCR-AAM patients. We proved the involvement of six genes in enteric nervous system development and Hirschsprung disease., Author summary Hirschsprung disease is a congenital disorder characterized by the absence of intestinal neurons in the distal part of the intestine. It is a complex genetic disorder in which multiple variations in our genome combined, result in disease. One of these variations are Copy Number Variations (CNVs): large segments of our genome that are duplicated or deleted. Patients often have Hirschsprung disease without other symptoms. However, a proportion of patients has additional associated anatomical malformations and neurological symptoms. We found that CNVs, present in patients with associated anomalies, are more often larger compared to unaffected controls or Hirschsprung patients without other symptoms. Furthermore, Copy Number (CN) losses are enriched for constrained coding regions (CCR; genes usually not impacted by genomic alterations in unaffected controls) of which the expression is higher in the developing intestinal neurons compared to the intestine. We modelled loss of these candidate genes in zebrafish by disrupting the zebrafish orthologues by genome editing. For several genes this resulted in changes in intestinal neuron development, reminiscent of HSCR observed in patients. The results presented here highlight the importance of Copy Number profiling, zebrafish validation and evaluating all CCR expressed in developing intestinal neurons during diagnostic evaluation.

International audience; The genetics of neurodevelopmental disorders (NDD) has made tremendous progress during the last few decades with the identification of more than 1,500 genes associated with conditions such as intellectual disability and autism. The functional roles of these genes are currently studied to uncover the biological mechanisms influencing the clinical outcome of the mutation carriers. To integrate the data, several databases and curated gene lists have been generated. Here, we provide an overview of the main databases focusing on the genetics of NDD, that are widely used by the medical and scientific communities, and extract a list of high confidence NDD genes (HC-NDD). This gene set can be used as a first filter for interpreting large scale omics dataset or for diagnostic purposes. Overall HC-NDD genes (N = 1,586) are expressed at very early stages of fetal brain development and enriched in several biological pathways such as chromosome organization, cell cycle, metabolism and synaptic function. Among those HC-NDD genes, 204 (12,9%) are listed in the synaptic gene ontology SynGO and are enriched in genes expressed after birth in the cerebellum and the cortex of the human brain. Finally, we point at several limitations regarding the relatively poor standardized information available, especially on the carriers of the mutations. Progress on the phenotypic characterization and genetic profiling of the carriers will be crucial to improve our knowledge on the biological mechanisms and on risk and protective factors for NDD.

Education is the foundation of science and technology development, human resource development to meet the needs of socio-economic development. The teacher is the one who organizes, guides and controls the teaching process and the teaching capacity of teachers determines the quality of education. Teacher training is a key task of schools and the education and training sector. There have been many domestic and foreign studies on teacher training, fostering teaching competence for teachers and management in order to improve the quality of fostering teaching competence for teachers. The paper uses survey method and analyses the results of other research in order to clarify the related issues. The sample has 240 teachers from 6 mountainous provinces in the North of Vietnam, the interview as well as some questions have been designed in the form of both online and paper forms. The results showed that we need to pay more attention to the assessment of teachers' teaching capacity, espeacially teachers of Northern montainous areas. The question mentioned in the article is that although the orientation of teaching towards student capacity development has been applied by many schools and is quite effective, however, the problem of designing and organizing teaching according to the direction of capacity development is a big problem, which means that the teaching capacity of teachers needs to be paid more attention, especially teachers in remote areas.

BackgroundHirschsprung disease (HSCR) is characterized by absence of ganglia in the intestine. Approximately 18% of patients have additional anatomical malformations or neurological symptoms (HSCR-AAM). HSCR is a complex genetic disease in which the loss of enteric ganglia stems from a combination of genetic alterations: rare coding variants, predisposing haplotypes and Copy Number Variation (CNV). Pinpointing the responsible culprits within a large CNV is challenging as often many genes are affected. We investigated if we could find deleterious CNVs and if we could identify the genes responsible for the aganglionosis.ResultsDeleterious CNVs were detected in three groups of patients: HSCR-AAM, HSCR patients with a confirmed causal genetic variant and HSCR-isolated patients without a known causal variant and controls. This study is conducted to investigate the impact of migration on living standards of households with migrants in the context of Vietnam. Data were collected from the results of Vietnam Household Living Standards in the time series. Blinder–Oaxaca decomposition was employed to decompose the source of differences in income between households with migrants and households without migrants. The results show that households with migrants in the multiyear dataset had a higher income than nonmigrant households, and migration had different impacts on expenditure at different quantiles. By conducting quantile regression, migration had positive impacts on expenditures at the 10% and 50% quantile, but no impact at the 90% quantile. Based on the findings, some implications in policies for managers, such as appropriate policies for poor workers in order to improve their living standards, especially poor households in rural or mountainous areas, are proposed.

The fiber-reinforcement of soil is an effective and reliable ground improvement technique for increasing the strength and stability of soil for various purposes (including retaining structures, embankments, foundations, slopes and pavements). Numerous scholars have developed methods to identify factors that influence the shear strength and to predict the peak friction angle of fiber-reinforced soil (FRS). The accuracy of theoretical and empirical models for predicting the shear strength (peak friction angle) of FRS is questionable because of the difficulty of using these simplified models to describe the complex mechanism of soil-fiber interaction. Solutions to this problem require ever-increasing predictive accuracy, and ML-based methods have been confirmed to provide potential solutions to real-world engineering problems. Therefore, this study develops weighted-feature least squares support vector regression (WFLSSVR) that is optimized by a novel metaheuristic algorithm, jellyfish search (JS) algorithm, to predict the peak friction angle of FRS. Analytical results demonstrate that JS-WFLSSVR outperforms baseline, ensemble, and hybrid machine learning models as well as empirical methods in literature. Notably, analysis of the weight values that were obtained by JS-WFLSSVR enables the identification of new feature combinations that provide much higher accuracy than current models. Therefore, the JS-WFLSSVR model not only significantly provides better predictive accuracy than methods in the literature; it is also a good feature selection method, and can help geotechnical engineers in estimating the shear strength of FRS. Geotechnical engineers can use the proposed model to predict the shear strength and control the quality of FRS structures.

Background Hirschsprung disease (HSCR), which is congenital obstruction of the bowel, results from a failure of enteric nervous system (ENS) progenitors to migrate, proliferate, differentiate, or survive within the distal intestine. Previous studies that have searched for genes underlying HSCR have focused on ENS-related pathways and genes not fitting the current knowledge have thus often been ignored. We identify and validate novel HSCR genes using whole exome sequencing (WES), burden tests, in silico prediction, unbiased in vivo analyses of the mutated genes in zebrafish, and expression analyses in zebrafish, mouse, and human. Results We performed de novo mutation (DNM) screening on 24 HSCR trios. We identify 28 DNMs in 21 different genes. Eight of the DNMs we identified occur in RET, the main HSCR gene, and the remaining 20 DNMs reside in genes not reported in the ENS. Knockdown of all 12 genes with missense or loss-of-function DNMs showed that the orthologs of four genes (DENND3, NCLN, NUP98, and TBATA) are indispensable for ENS development in zebrafish, and these results were confirmed by CRISPR knockout. These genes are also expressed in human and mouse gut and/or ENS progenitors. Importantly, the encoded proteins are linked to neuronal processes shared by the central nervous system and the ENS. Conclusions Our data open new fields of investigation into HSCR pathology and provide novel insights into the development of the ENS. Moreover, the study demonstrates that functional analyses of genes carrying DNMs are warranted to delineate the full genetic architecture of rare complex diseases. Hirschsprung disease (HSCR) is the most common cause of neonatal intestinal obstruction. It is characterized by the absence of ganglia in the nerve plexuses of the lower gastrointestinal tract. So far, three common disease-susceptibility variants at the RET, SEMA3 and NRG1 loci have been detected through genome-wide association studies (GWAS) in Europeans and Asians to understand its genetic etiologies. Here we present a trans-ethnic meta-analysis of 507 HSCR cases and 1191 controls, combining all published GWAS results on HSCR to fine-map these loci and narrow down the putatively causal variants to 99% credible sets. We also demonstrate that the effects of RET and NRG1 are universal across European and Asian ancestries. In contrast, we detected a European-specific association of a low-frequency variant, rs80227144, in SEMA3 [odds ratio (OR) = 5.2, P = 4.7 × 10-10]. Conditional analyses on the lead SNPs revealed a secondary association signal, corresponding to an Asian-specific, low-frequency missense variant encoding RET p.Asp489Asn (rs9282834, conditional OR = 20.3, conditional P = 4.1 × 10-14). When in trans with the RET intron 1 enhancer risk allele, rs9282834 increases the risk of HSCR from 1.1 to 26.7. Overall, our study provides further insights into the genetic architecture of HSCR and has profound implications for future study designs.

The Lao PDR is a landlocked country with 5,920,000 inhabitants for which very few epidemiological studies on cancer have been performed. The aim of the present study was to examine cancer mortality in 2007-2008.A descriptive cancer epidemiology protocol was designed with a data collection form and guideline for both demographics and list of all deaths from all 757 local Health Centers of 17 provinces/ cities. Five indicators, name, age, sex, date of death and the cause of death (ICD-10), were collected for each case. The age-specific cancer mortality rate and ASRs per 100,000 were estimated.There were 448 cancer cases reported from Health Centers within 7 of 17 provinces/cities. Number of person-years was 654,459 for the two-year period. Cancer mortality rates of all sites (ASR) were 116.7 and 97.2 per 100,000 in males and females, respectively. The five most common cancers causing mortality per 100,000 were liver (52.2), followed by colorectal (19.0), lung (17.3), stomach (6.9), and leukemia-lymphoma (7.2) in males and liver (28.4); followed by colorectal (19.0), lung (14.0), cervical uteri (9.2) and stomach (7.1) in females.Liver and colorectal cancers were the first and second most common, respectively, in both males and female.
