The antibacterial and anti-inflammatory effect of thioglycosides has already been established. This study investigates the effects of thioglycosides extracted from white mustard, specifically the "Bamberka" variety, in the context of oral hygiene. The aim of the study is to clarify an evidence-based link between the documented antibacterial and anti-inflammatory effects attributed to thioglycosides and their practical application in oral care. A randomized, single-blinded (patient-blinded) clinical study was performed on 66 patients using mustard-based toothpaste for oral hygiene. The patients were examined at baseline and after 6 and 12 months. The values of the Approximal Plaque Index (API), the Plaque Index (PI), and Bleeding on probing (BOP) were taken into consideration. The results show a significant reduction in plaque accumulation, especially after 6 months of using mustard-based toothpaste in all examined parameters. This suggests that thioglycosides from mustard contribute to a considerable decrease in dental plaque accumulation, confirming their potential in natural oral care solutions, which is indicated in the main conclusions or interpretations. AbstractNovel class of triazine sulfonamide thioglycosides was designed and synthesized. Those novel structures comprising three essential and pharmacological significant moieties such as the triazine, sulfonamide, and thioglycosidic scaffolds. The triazine sulfonamides were furnished via a direct approach starting from potassium cyanocarbonimidodithioate, then the corresponding triazine sulfonamide thioglycosides were generated using the peracylated α-d-gluco- and galacto-pyranosyl bromides. Anti-viral evaluation of compounds in vitro against HCoV-229E virus revealed that some compounds possess promising activity. Compounds 4a, 4b, 4d, 6d and 6e indicate from moderate to low antiviral activity against low pathogenic coronavirus 229E in comparison with remdesivir at a concentration of 100 µg/mL. Additionally their in vitro anti-proliferative effects against NCI 60 cancer cell lines cell lines were also investigated. The lyotropic properties of alkyl thioglycosides with varying sugar headgroup (lactose, cellobiose, maltose, galactose, or glucose) and alkyl chain length (octyl, decyl, or dodecyl chains) are investigated by surface tensiometry, visual observation, and fluorescence spectroscopy. The results substantiate that the glycosidic S‐linkage confers considerably different solution aggregation behavior on these surfactants relative to their O‐linked counterparts, where the properties of the latter are known. The materials properties of the aggregated structures from the alkyl thioglycosides vary considerably. Micelles are formed by octyl thiocellobioside and all alkyl thiomaltosides. Turbid aggregate solutions are formed by the alkyl thioglucosides and octyl thiogalactoside, whereas the longer chain alkyl thiogalactosides are minimally soluble. Fluorescence spectroscopy of these systems confirms their aggregation in lamellar‐like structures. The alkyl thiocellobiosides and alkyl thiolactosides form hydrogels from these low‐molecular weight materials at concentrations almost an order of magnitude lower than gels using other low‐molecular weight materials. Here, hydrogels form at concentrations <0.3 wt% with some forming hydrogels at concentrations as low as 0.03 wt% from alkyl thiocellobiosides and thiolactosides, with hydrogel properties differing significantly with this slight change in the sugar headgroup. Alkyl thiocellobiosides form a nanofiber network and alkyl thiolactosides form globular hydrogels. The synthesis of glycosides generally requires the use of building blocks that need to be readily prepared, avoiding tricky reaction steps and boring purifications. Idose-type glycosides have numerous biological activities and have been widely used as anticoagulant drugs and anti-infection drugs. Thioglycosides have enhanced stability for acid-mediated or enzymatic hydrolysis, and have a wide range of applications in glycobiology and drug development. Herein, we describe an efficient method for site-selective and stereoselective synthesis of potential bioactive 2-amino-2-deoxy-1,3-dithioidoglycosides via organocatalysis sequential C3-Ferrier rearrangement and Michael addition of 3-O-acetyl-2-nitrogalactals. Both stepwise and one-pot protocols were carried out and work well. This unique thio-glycosylation protocol highlighted the various advantages, including (i) mild reaction conditions; (ii) excellent site-selectivity and stereoselectivity, good to excellent yields; (iii) broad substrate scopes; (iv) being atom-economic and environmentally friendly; (v) the reactions can be scaled up. The biologically important l -hexoses, which are less widespread than d -hexoses, cannot be obtained from natural sources or can only be extracted very costly. Due to the complexity of their synthesis, their commercially available derivatives (which are sold mostly in free form) are also very expensive, which is further exacerbated by the current rapid rise in prices. In the present work, starting from the cheapest d -hexose, d -glucose, using inexpensive and readily available chemicals, a reaction pathway was developed in which the three most expensive l -hexoses (l -idose, l -altrose, and l -talose) were successfully prepared in orthogonally protected thioglycoside form, ready for glycosylation. The l - ido and l - talo derivatives were synthesized by C-5 epimerization of the corresponding 5,6-unsaturated thioglycosides. From the l - ido derivatives, the orthogonally protected thioglycosides of l -altrose were then prepared by C-4 epimerization. Different approaches to the preparation of the key intermediates, 5,6-unsaturated thioglycoside derivatives, were systematically investigated in the presence of various protecting groups (ether and ester) and using commercially available reagents. The conventional synthesis of glycosyl chlorides from thioglycosides relies on sequential oxidation and chlorination. A one-pot synthesis of glycosyl chlorides is warranted as an alternative method. Here, we report a one-pot synthesis of glycosyl chlorides from thioglycoside precursors. The transformation was mediated at low temperatures by bromodiethylsulfonium bromopentachloroantimonate (BDSB) as a mild oxidant with Bu4 NCl as an additive. Armed thioglycosides afforded the corresponding α-glycosyl chlorides in moderate to good yields under the optimized conditions. Low conversions and yields were obtained when the less-reactive disarmed thioglycosides were used. Unexpectedly, BDSB-mediated oxidation of thioglycosides without the addition of Bu4 NCl also afforded the α-glycosyl chlorides in moderate yields. From these derivatives, the orthogonally protected thioglycosides of further two l-hexoses (l-allose and l-glucose) were synthesized by C-4 epimerization. By the method developed, we are able to produce highly functionalized l-gulose derivatives in 9 steps (total yields: 21–23%) and l-galactose derivatives in 12 steps (total yields: 6–8%) starting from d-mannose. Multiple fluorination of glycostructures has emerged as an attractive way of modulating their protein affinity, metabolic stability, and lipophilicity. Here we described the synthesis of a series of mono-, di- and trifluorinated N-acetyl-ᴅ-glucosamine and ᴅ-galactosamine analogs. The key intermediates are the corresponding multiply fluorinated glucosazide and galactosazide thioglycosides prepared from deoxyfluorinated 1,6-anhydro-2-azido-β-ᴅ-hexopyranose precursors by ring-opening reaction with phenyl trimethylsilyl sulfide. Nucleophilic deoxyfluorination at C4 and C6 by reaction with DAST, thioglycoside hydrolysis and azide/acetamide transformation completed the synthesis.

Nucleophilic substitution of the chlorine atom in 4,5-dichloro-1,2-dithiol-3-one using 1-thio-β-d-glucopyranose and its 2,3,4,6-tetra-O-acetyl derivative in acetonitrile resulted in two new 5-thioglucoside conjugates of 4-chloro-1,2-dithiol-3-one. The obtained thioglucosides showed cancer-preventive properties in a model of mouse epidermal JB6Cl41 P+ cells in soft agar, while one of them manifested antitumor activity in in vivo experiments in CD1 mice on a model of solid Ehrlich carcinoma. A streamlined iterative assembly of thio‐oligosaccharides was developed by aqueous glycosylation. Facile syntheses of various deoxythio sugars with the sulfur on different positions from commercially available starting materials were described. These syntheses featured efficient chemical methods including our recently reported BTM‐catalyzed site‐selective acylation. The resulting deoxythio sugars could then be used for the Ca(OH)2‐promoted protecting group‐free S‐glycosylation in water at room temperature. The aqueous glycosylation reaction proceeded smoothly to afford the corresponding 1,2‐trans S‐glycosides in good yields with high chemo‐ and stereoselectivity. An appropriate choice of protecting groups for the thiol in the glycosyl donor was necessary for the development of iterative synthesis of thio‐oligosaccharides. A new class of thioglycosides comprising a diazenyl pyridine moiety was assembled from the regioselective reaction of the acetylated α-glycopyranosyl bromide with hitherto unreported 2-mercaptonicotinonitriles. Moreover, the deacetylation of S-glycoside derivatives utilizing of methanolic ammonia solution afforded the free hydroxy thioglycosides in quantitative yields. The identity of the newly obtained derivatives has been achieved via spectroscopic and elemental analyses. All of the newly synthesized derivatives exhibited significant antimicrobial activity towards certain selected microorganisms (bacteria and fungi). Generally, S-glycoside derivatives demonstrated outstanding antibacterial and antifungal activities. Amongst them, the acetylated derivatives showed the most distinguished antifungal and antibacterial activities with a promising minimum inhibitory concentration (MIC) values in comparison with the approved drugs; Amphotericin B and Cefotaxime.

A series of new substituted triazolo[4,5-d]pyrimidine derivatives linked to thienopyrimidine ring system were prepared as a hybrid heterocyclic systems, as possible nucleobases analogs, starting from the key carboxamide derivative 2 and its azide precursor via heterocyclization reactions and their structures were characterized. Glycosylation of the prepared triazolopyrimidine derivatives was performed and afforded, regioselctively, the corresponding thienopyrimidine-triazolopyrimidine hybrid N1-glycosides and their thioglycoside analogues in good yields. The synthesized glycosyl heterocycles were studied for their cytotoxic activity against HepG-2 and MCF-7 human cancer cells and significant results were obtained. Compounds 7a, 8 b, 9 b, 9a and 7 b demonstrated promising activities comparable to the activity of the doxorubicin for (HepG-2) cell line. Furthermore, a number of the afforded triazolopyrimidine glycosides were found potent against cancer cells (MCF-7). Furthermore, docking simulation the promising thienopyrimidine analogues 7-13 was done against EGFR kinase to provide a binding model that could serve in discovery of further anticancer agents. Pseudomonas aeruginosa is a highly prevalent gram-negative bacterium that is becoming more difficult to treat because of increasing antibiotic resistance. As chemotherapeutic treatment options diminish, there is an increased need for vaccines. However, the creation of an effective P. aeruginosa vaccine has been elusive despite intensive efforts. Thus, new paradigms for vaccine antigens should be explored to develop effective vaccines. In these studies, we have focused on the synthesis of two L-rhamnose–bearing epitopes common to glycoforms I and II of the outer core domain of Pseudomonas aeruginosa lipopolysaccharide, α-L-Rha-(1→6)-α-D-Glc-(1→4)-α-D-GalN-(Ala)-α-aminooxy (3) and α-L-Rha-(1→3)-β-D-Glc-(1→3)-α-D-GalN-(Ala)-α-aminooxy (4), respectively. The target trisaccharides were both prepared starting from a suitably protected galactosamine glycoside, followed by successive deprotection and glycosylation with suitably protected D-glucose and L-rhamnose thioglycosides. Global deprotection resulted in the formation of targets 3 and 4 in 22 and 35% yield each. Care was required to modify basic reaction conditions to avoid early deprotection of the N-oxysuccinamido group. In summary, trisaccharides related to the L-rhamnose–bearing epitopes common to glycoforms I and II of the outer core domain of Pseudomonas aeruginosa lipopolysaccharide have been prepared as their aminooxy glycosides. The latter are expected to be useful in chemoselective oxime-based bioconjugation reactions to form Pseudomonas aeruginosa vaccines.

The reaction of glycosyl donor phenyl 2,3,4,6‐tetra‐O‐benzyl‐1‐thio‐β‐D‐glucopyranoside with NIS/TfOH(cat.) was systematically studied under various reaction conditions. Neither the molecular sieve pore size nor amount of NIS activator was found to have an effect on the α/β‐ratio in the reaction with L‐menthol as glycosyl acceptor. Increasing concentration and the amount of triflic acid catalyst, however was found to increase the β‐selectivity in certain cases. Our group has previously reported that 3,3‐difluoroxindole (HOFox) is able to mediate glycosylations via intermediacy of OFox imidates. Thioglycoside precursors were first converted into the corresponding glycosyl bromides that were then converted into the OFox imidates in the presence of Ag2O followed by the activation with catalytic Lewis acid in a regenerative fashion. Reported herein is a direct conversion of thioglycosides via the regenerative approach that bypasses the intermediacy of bromides and eliminates the need for heavy‐metal‐based promoters. A novel method for the glycosylation of selenoglycosides activated by iodosylbenzene was developed. The glycosylation reaction conditions were mild, fast, and efficient, with a high tolerance to diverse protecting groups and a wide substrate scope, which is advantageous for synthesizing complex glycosides. In addition, selenoglycosides were shown to be orthogonal to thioglycosides under the promotion of iodosylbenzene. Notably, a high yield of the poorly reactive glucuronidation reaction product was obtained by acetyl-protected selenoglycoside. Finally, the orthogonal one-pot synthesis of β-(1→6) oligoglucans demonstrated the usefulness of this method in oligosaccharide synthesis.

GH20 human β-N-acetylhexosaminidases (hsHex) and GH84 human O-GlcNAcase (hOGA) are involved in numerous pathological processes and emerged as promising targets for drug discovery. Based on the catalytic mechanism and structure of the catalytic domains of these β-N-acetylhexosaminidases, a series of novel naphthalimide moiety-bearing thioglycosides with different flexible linkers were designed, and their inhibitory potency against hsHexB and hOGA was evaluated. The strongest potency was found for compound 15j (Ki = 0.91 µM against hsHexB; Ki > 100 µM against hOGA) and compound 15b (Ki = 3.76 µM against hOGA; Ki = 30.42 µM against hsHexB), which also exhibited significant selectivity between these two enzymes. Besides, inhibitors 15j and 15b exhibited an inverse binding patterns in docking studies. The determined structure–activity relationship as well as the established binding models provide the direction for further structure optimizations and the development of specific β-N-acetylhexosaminidase inhibitors.

New sugar hydrazones and their derived oxadiazolyl acyclic nucleoside analogs in addition to the derived thioglycosides incorporating pyridine, furan or thiophene have been synthesized via a multi-component reaction (MCR). The acetylated derivatives of hydrazones and the derived deprotected thioglycosides of the prepared acetylated analogs have been also synthesized. Anticancer activity of the products has been tested against adenocarcinomic human alveolar basal epithelial (A549), human prostatic adenocarcinoma (PC3) and human colorectal carcinoma (HCT116) cell lines in addition to their effect on human normal retinal pigmented epithelial cell line (RPE1) using the MTT assay. New 1,4-bithiazolyl-5-oxopyrrole derivatives have been synthesized via a multicomponent reaction (MCR) and their structures characterized. N1-Phenyl- and N1-amino-substituted 1,2,4-triazole derivatives linked with bithiazolyl-5-oxopyrrole system also have been synthesized. Glycosylation of the latter s-triazole compounds by acetylated glycosyl halides has led to the corresponding thioglycoside derivatives. Cytotoxic activity data of the synthesized compounds against colon (HCT-116) and breast (MCF-7) human cancer sells determined by the MTT assay indicate two 1,2,4-triazole thioglycosides as potentially active in comparison with doxorubicin against MCF-7 cancer. The synthesis of two glucuronoxylans is described, which both consist of a pentaxylan backbone and a glucuronic acid linked to the 2 position in the fourth xylose residue from the reducing end. The two target molecules differ in the 4 position of the glucuronic acid where one is unsubstituted while the other contains a methyl ether. The pentaxylan backbone is assembled in four glycosylation reactions with phenyl thioglycoside donors. The couplings are performed by preactivation of the donor with in‐situ‐generated p‐nitrobenzenesulfenyl triflate prior to addition of the acceptor. The glucuronic acids are then attached by Koenigs‐Knorr glycosylations followed by deprotections. The syntheses employ a total of 8 steps from monosaccharide building blocks and afford the two glucuronoxylans in 12 and 15 % overall yield. 1,3-Dibromo-5,5-dimethylhydantoin (DBDMH), an inexpensive, non-toxic and stable reagent, is a competent activator of thioglycosides for glycosidic bond formation. Excellent yields were obtained when triflic acid (TfOH) or trimethylsilyl trifluoromethanesulfonate (TMSOTf) were employed as co-promoters in solution or automated glycan assembly on solid phase.

A practical method for the selective and controlled oxidation of thioglycosides to corresponding glycosyl sulfoxides and sulfones is reported using urea–hydrogen peroxide (UHP). A wide range of glycosyl sulfoxides are selectively achieved using 1.5 equiv of UHP at 60 °C while corresponding sulfones are achieved using 2.5 equiv of UHP at 80 °C in acetic acid. New glycosyl donors have been developed that contained several para‐substituted O‐aryl protecting groups and their stereoselectivity for the glycosylation reaction was evaluated. A highly β‐selective glycosylation reaction was achieved by using thioglycosides that were protected by 4‐nitrophenyl (NP) groups, which were introduced by using the corresponding diaryliodonium triflate. Analysis of the stereoselectivities of several glycosyl donors indicated that the β‐glycosides were obtained through an SN2‐type displacement from the corresponding α‐glycosyl triflate. Thioglycosides are hydrolase‐resistant mimics of O‐linked glycosides that can serve as valuable probes for studying the role of glycosides in biological processes. The development of an efficient, enzyme‐mediated synthesis of thioglycosides, including S‐GlcNAcylated proteins, is reported, using a thioglycoligase derived from a GH20 hexosaminidase from Streptomyces plicatus in which the catalytic acid/base glutamate has been mutated to an alanine (SpHex E314A). This robust, easily‐prepared, engineered enzyme uses GlcNAc and GalNAc donors and couples them to a remarkably diverse set of thiol acceptors. Thioglycoligation using 3‐, 4‐, and 6‐thiosugar acceptors from a variety of sugar families produces S‐linked disaccharides in nearly quantitative yields. The set of possible thiol acceptors also includes cysteine‐containing peptides and proteins, rendering this mutant enzyme a promising catalyst for the production of thio analogues of biologically important GlcNAcylated peptides and proteins. Thioglycosides are stable carbohydrate mimetics frequently used for structural and functional biology studies. A thioglycoligase was developed from a GH20 hexosaminidase, enabling direct S‐GlcNAcylation of a diverse set of thio‐acceptors. Synthesis of a variety of sugar lactols (hemiacetals) has been accomplished in moderate to excellent yields by using bromine-mediated oxidation of thioglycosides. It was found that acetonitrile is the optimal solvent for this oxidation reaction. This approach involving bromine as oxidant is superior to that using N-bromosuccinimide (NBS) which produces byproduct succinimide often difficult to separate from the lactol products. GH20 human β-N-acetylhexosaminidases (hsHex) and GH84 human O-GlcNAcase (hOGA) are involved in numerous pathological processes and emerged as promising targets for drug discovery. Based on the catalytic mechanism and structure of the catalytic domains of these β-N-acetylhexosaminidases, a series of novel naphthalimide moiety-bearing thioglycosides with different flexible linkers were designed, and their inhibitory potency against hsHexB and hOGA was evaluated. The strongest potency was found for compound 15j (Ki = 0.91 µM against hsHexB; Ki > 100 µM against hOGA) and compound 15b (Ki = 3.76 µM against hOGA; Ki = 30.42 µM against hsHexB), which also exhibited significant selectivity between these two enzymes. Besides, inhibitors 15j and 15b exhibited an inverse binding patterns in docking studies. The determined structure-activity relationship as well as the established binding models provide the direction for further structure optimizations and the development of specific β-N-acetylhexosaminidase inhibitors.
