Cells are 3D objects. Therefore, volume EM (vEM) is often crucial for correct interpretation of ultrastructural data. Today, scanning EM (SEM) methods such as focused ion beam (FIB)-SEM are frequently used for vEM analyses. While they allow automated data acquisition, precise targeting of volumes of interest within a large sample remains challenging. Here, we provide a workflow to target FIB-SEM acquisition of fluorescently labeled cells or subcellular structures with micrometer precision. The strategy relies on fluorescence preservation during sample preparation and targeted trimming guided by confocal maps of the fluorescence signal in the resin block. Laser branding is used to create landmarks on the block surface to position the FIB-SEM acquisition. Using this method, we acquired volumes of specific single cells within large tissues such as 3D cultures of mouse mammary gland organoids, tracheal terminal cells in Drosophila melanogaster larvae, and ovarian follicular cells in adult Drosophila, discovering ultrastructural details that could not be appreciated before., (© 2021 Ronchi et al.)

Sepsis is defined as a systemic inflammatory response to infection. This study is aimed to evaluate the effects of experimental sepsis on the proliferation and apoptosis of granulosa and theca cells in the rat ovary. 28-day-old immature Wistar-Albino female rats were treated with pregnant mare serum gonadotrophin to develop the first generation of preovulatory follicles. Sepsis was induced by cecal ligation and puncture (CLP). Following in vivo 5-Bromo-2-deoxyuridine (BrdU) labeling, animals were sacrificed and ovaries were embedded in paraffin and Epon. Besides electron microscopic evaluation, BrdU, cleaved caspase-3, p27 immunostaining, and TUNEL labeling were performed. In CLP-operated animals, cleaved caspase-3 immunoreactivity was significantly increased in Graafian follicles. TUNEL and BrdU labeling in the ovarian follicles were not statistically different between CLP and sham-operated rats. In septic animals, p27 immunoreactivity was increased significantly in the nuclei of oocytes and decreased in the cytoplasm of granulosa and theca cells in multilaminar primary follicles compared to the sham group. In ultrastructural evaluation, increased apoptosis was observed in theca interna and granulosa cells in both the early and late stages of follicles in the CLP group. In conclusion, experimentally-induced sepsis leads to apoptosis in ovarian follicles at advanced stages of development. Polycystic ovary syndrome (PCOS) is a clinical syndrome characterized by hyperandrogenism, oligo/anovulation, and polycystic ovary. Autophagy is an intracellular system that degrades cytosolic proteins and organelles. The relationship between autophagy and PCOS has not been clarified. We found that p62 and ubiquitin were significantly increased in theca cells of women with PCOS using immunohistochemistry. Autophagy inhibition by palmitic acid and chloroquine in bovine theca cells increased p62 and ubiquitin and induced the expression of cytochrome P450 17A1 (CYP17A1) and plasminogen activator inhibitor-1 (PAI-1) mRNA. Furthermore, palmitic acid and chloroquine exposure significantly increased reactive oxygen species (ROS) and activated p38 and c-Jun N-terminal kinase (JNK). Inhibition of p38 and JNK significantly reduced CYP17A1 and PAI-1 mRNA expression. We showed that inhibition of autophagy in theca cells may have contributed to the pathogenesis of PCOS, based on CYP17A1 and PAI-1 mRNA expression via the ROS/p38 and JNK signalling pathways., (Copyright © 2020 Elsevier B.V. In mammalian ovaries, the theca layers of growing follicles are critical for maintaining their structural integrity and supporting androgen synthesis. Through combining the postnatal monitoring of ovaries by abdominal magnetic resonance imaging, endocrine profiling, hormonal analysis of the follicular fluid of growing follicles, and transcriptomic analysis of follicular theca cells, we provide evidence that the exposure of ovine fetuses to testosterone excess activates postnatal follicular growth and strongly affects the functions of follicular theca in adulthood. Prenatal exposure to testosterone impaired androgen synthesis in the small antral follicles of adults and affected the expression in their theca cells of a wide array of genes encoding extracellular matrix components, their membrane receptors, and signaling pathways. Most expression changes were uncorrelated with the concentrations of gonadotropins, steroids, and anti-Müllerian hormone in the recent hormonal environment of theca cells, suggesting that these changes rather result from the long-term developmental effects of testosterone on theca cell precursors in fetal ovaries. Disruptions of the extracellular matrix structure and signaling in the follicular theca and ovarian cortex can explain the acceleration of follicle growth through altering the stiffness of ovarian tissue. We propose that these mechanisms participate in the etiology of the polycystic ovarian syndrome, a major reproductive pathology in woman.

Excessive secretion of androgens including androstenedione and testosterone in theca cells frequently causes female infertility in mammals. Melatonin is a potent inhibitor of androgen production in gonadal cells of several species in a membrane receptor-dependent manner. However, the function of melatonin in steroidogenesis of porcine theca cells remains unclear. Here we report that melatonin inhibits androgen biosynthesis independently of its membrane receptors in pigs. Using flow cytometry, immunofluorescence and RT-PCR we showed that the vast majority of cells isolated from the theca layer of antral follicles are indeed theca cells. Furthermore, we demonstrated that of the two of melatonin membrane receptors encoded in the porcine genome, theca cells exclusively express melatonin receptor 1B. Cell counting analysis indicated that different concentrations of melatonin did not alter the normal viability and proliferation of theca cells. Additionally, hormone radioimmunoassay and qPCR respectively showed that a high concentration of melatonin significantly repressed both androgen production and expression of steroidogenic genes involving StAR, CYP11A1, HSD3β and SET (P < 0.05), but did not impair progesterone production. Interestingly, these effects were not reversed by N-acetyl-2-benzyltryptamin, a melatonin membrane receptor antagonist. Immunoexpression of 3β-hydroxysteroid dehydrogenase (3β-HSD), cytochrome P450c17 (P450c17), androgen receptor (AR), and steroid contents were studied in the ovaries of immature female Wistar rats killed between postnatal days 1 and 30. During days 1-7, ovarian somatic structures lacked AR, 3β-HSD and P450c17, except for the surface epithelium, which featured the presence of these three proteins, suggestive of its androgen responsiveness and steroidogenic function. On day 10, AR appeared in many somatic structures, including the granulosa layers, which coincided with the P450c17 immunoexpression in some theca/interstitial cells, and an increase in ovarian androgen concentration. On the following days a further rise in ovarian androgen and progesterone contents paralleled an increase in 3β-HSD and P450c17 immunoexpression in the theca layer cells and primary interstitial cells. However, the development of the follicles constituting the first follicular wave was aberrant, since they lacked AR expression until the preantral stage and were characterized by a delayed onset and much lower expression of the thecal P450c17. They could not ovulate, since ovarian content of estradiol was too low to evoke the LH surge. The clusters of the secondary interstitial cells found on day 30 exhibited predominant expression of 3β-HSD over P450c17, suggesting more intensive progesterone than androgen synthesis in these structures., (Copyright © 2011 Elsevier GmbH. Background: In vitro maturation of ovarian follicles, in combination with cryopreservation, might be a valuable method for preserving and/or restoring fertility in mammals with impaired reproductive function. Several culture systems capable of sustaining mammalian follicle growth in vitro have been developed and many studies exist on factors influencing the development of in vitro grown oocytes. However, a very few reports concern the ultrastructural morphology of in vitro grown follicles., Methods: The present study was designed to evaluate, by transmission and scanning electron microscopy, the ultrastructural features of isolated mouse preantral follicles cultured in vitro for 6 days in a standard medium containing fetal calf serum (FCS). The culture was supplemented or not with FSH., Results: The follicles cultured in FCS alone, without FSH supplementation (FCS follicles), did not form the antral cavity. They displayed low differentiation (juxta-nuclear aggregates of organelles in the ooplasm, a variable amount of microvilli on the oolemma, numerous granulosa cell-oolemma contacts, signs of degeneration in granulosa cell compartment). Eighty (80)% of FSH-treated follicles formed the antral cavity (FSH antral follicles). These follicles showed various ultrastructural markers of maturity (spreading of organelles in ooplasm, abundant microvilli on the oolemma, scarce granulosa cell-oolemma contacts, granulosa cell proliferation). Areas of detachment of the innermost granulosa cell layer from the oocyte were also found, along with a diffuse granulosa cell loosening compatible with the antral formation. Theca cells showed an immature morphology for the stage reached. Twenty (20)% of FSH-treated follicles did not develop the antral cavity (FSH non-antral follicles) and displayed morphological differentiation features intermediate between those shown by FCS and FSH antral follicles (spreading of organelles in the ooplasm, variable amount of microvilli, scattered granulosa cell-oolemma contacts, signs of degeneration in granulosa cell compartment)., Conclusions: It is concluded that FSH supports the in vitro growth of follicles, but the presence of a diffuse structural granulosa cell-oocyte uncoupling and the absence of theca development unveil the incomplete efficiency of the system. The present study contributes to explain, from a morphological point of view, the effects of culture conditions on the development of mouse in vitro grown follicles and to highlight the necessity of maintaining efficient intercellular communications to obtain large numbers of fully-grown mature germ cells.

The immunohistochemical (IHC) localization of oestrogen receptor alpha (ERα) was studied in the developing left ovary of 14.5-day-old chick embryos. The study was focused in particular on distinguishing in cortex and medulla the different cell categories that proved positive to the reaction, in order to gain further understanding of gonadal cell interactions during ovarian development. Immunostained cells were observed in both the cortex and medulla, but the reactivity for ERα was discontinuous, probably due to variable cell requirements. In the cortex, positivity was observed in cells of the ovarian surface epithelium, in germ cells and in prefollicular cells. In the medulla, positivity was found in the following cell categories: interstitial cells, poorly differentiated somatic cord cells, including those delimiting lacunae, germ cells and their accompanying cells of epithelial origin. Furthermore, the IHC results showed that the intracellular localization of the antigen was cytoplasmic, nuclear, or both. The significance of ERα presence and intracellular localization was discussed in relation and as supplementary to previous research by various Authors. In particular, as regards the unusual cytoplasmic immunoreactivity, a gradual shift of ERα localization from cytoplasmic to nuclear during the embryonic period is suggested., (© 2010 Blackwell Verlag GmbH.)

The postnatal mouse ovary is rich in quiescent and early-growing oocytes, each one surrounded by a layer of somatic granulosa cells (GCs) on a basal lamina. As oocytes start to grow the GCs change shape from flattened to cuboidal, increase their proliferation and form multiple layers, providing a unique model for studying the relationship between cell shape, proliferation and multilayering within the context of two different intercommunicating cell types: somatic and germ cells. Proliferation of GCs was quantified using immunohistochemistry for Ki67 and demonstrated that, unusually, cuboidal cells divided more than flat cells. As a second layer of GCs started to appear, cells on the basal lamina reached maximum packing density and the axes of their mitoses became perpendicular to the basal lamina, resulting in cells dividing inwards to form second and subsequent layers. Proliferation of basal GCs was less than that of inner cells. Ultrastructurally, collagen fibrils outside the basal lamina became more numerous as follicles developed. We propose that the basement membrane and/or theca cells that surround the follicle provide an important confinement for rapidly dividing columnar cells so that they attain maximum packing density, which restricts lateral mitosis and promotes inwardly oriented cell divisions and subsequent multilayering.

The morphological and endocrine aspects of the ovarian interstitial tissue of adult female viscachas were investigated to establish the probable function and the biological significance of this compartment in this rodent. Pregnant and nonpregnant adult female viscachas were used. The histological characteristics, histochemical properties, and ultrastructural features of the interstitial tissue were studied. A morphometric study was carried out to measure the relative area of lipid droplets. The progesterone and androstenedione levels in ovarian tissue as well as in serum were determined by radioimmunoassay. In this species, the histological observations showed an abundant interstitial tissue that contained a large amount of lipids. The cholesterol and its esters were present in nonpregnant females and were scarce in pregnant animals. The most ultrastructural differences were observed at mid-pregnancy. At this stage, the interstitial cells showed features that suggested higher steroidogenic activity. Furthermore, during mid-pregnancy, the relative area of lipid droplets was smaller. Both progesterone and androstenedione levels in ovarian tissue and serum were higher during pregnancy. Our results suggest that the interstitial tissue may be storage of precursor substances for the steroidogenesis via. These precursors are probably used when the endocrine requirements are high, that is, during the pregnancy. Thus, this compartment may contribute to the normal gestation of Lagostomus. However, the relation between the interstitial tissue and the pregnancy is complex, and further studies are needed to clearly establish it., ((c) 2007 Wiley-Liss, Inc.)

The ultrastructure of the follicular wall in primordial, previtellogenic and vitellogenic follicles of the sexually immature ostrich is described in the present study. The follicular wall consists of a zona radiata, granulosa cell layer, basal lamina and thecal layer. Cytoplasmic processes from the plasma membranes of the granulosa cell layer and the ovocyte form the zona radiata in previtellogenic and vitellogenic follicles. The granulosa cell layer transforms from simple cuboidal epithelium in primordial follicles to simple columnar or pseudostratified columnar epithelium in previtellogenic and vitellogenic follicles. Transosomes were observed along the apical and lateral plasma membranes of granulosa cells. The thecal layer in previtellogenic and vitellogenic follicles consists of interna and externa components. The fibroblasts in the theca externa contain microfilaments, which are thought to be actin filaments. The study revealed ultrastructural features, which are associated with the transportation of yolk precursors and nutrients into the ovoplasm. In addition, the study indicates that, although the cells in the theca externa contain microfilaments, they do not possess the ultrastructural characteristics of smooth muscle cells.

In the hen ovary, each preovulatory follicle in the hierarchy, irrespective of its size and the level of its maturity is exposed to the preovulatory LH surge in each ovulatory cycle of an egg laying sequence. In the present study, the thecal weight and membrane protein content of theca layers at different stages of hen ovulatory cycle were assessed. Hens were killed 2 h (stage I), 9 h (stage II), 16 h (stage III), and 23 h (stage IV) after oviposition. The first (F1), second (F2), third (F3), fourth (F4) and fifth (F5) largest yellow follicles were utilized. In all follicles except F1, the thecal weight rose considerably between stages I and III (P < 0.05) followed by a slight cessation of the thecal growth at stage IV. The mean content of the theca membrane protein in F1-F5 follicles was lowest at stage III, increasing at stage IV (P < 0.05), although, in the case of individual follicles the difference was significant (P < 0.05) in F3 follicles only. Estradiol-17beta levels in the plasma were lowest (but not significant) at stage III, and a fourfold increase in the plasma progesterone concentration occurred at stage IV. These findings demonstrate for the first time the ovulatory cycle-related alterations in the thecal weight and membrane protein content in the hen preovulatory follicles. Data suggest that the preovulatory rise in ovarian steroid hormones is probably involved in transient termination of the growth and induction of differentiation of the theca in preovulatory follicles as they pass from one category to the next.

We developed a culture system in which two types of ovarian follicular cells were allowed to attach to opposite sides of a collagen membrane. Using this in vitro cell culture system, we studied the effects of granulosa- and theca-cell interaction on the morphology, structure, and function of bovine ovarian follicular cells. In the first part of the study, we explored how the interaction between theca and granulosa cells affects the morphology and structure of the cells. This study was done using follicular cells collected from bovine ovarian follicles at the early developmental stage. Granulosa cells cultured alone were flattened, and formed a monolayer sheet. By contrast, granulosa cells cultured with theca cells were convex, and formed multilayer sheets. Theca cells cultured alone were thin, flat, and spindle-shaped. Theca cells cultured with granulosa cells were also spindle-shaped; however, they appeared convex and more densely packed when compared with theca cells cultured alone. In the second part of the study, the possible role of the cellular interaction in the control of differentiation and growth of granulosa and theca cells was investigated. When follicular cells were isolated from the early stage of follicular development, theca cells reduced progesterone and inhibin production by granulosa cells and augmented the growth of granulosa cells. When the cells were isolated from the late stage of follicular development, by contrast, theca cells augmented hormonal production by granulosa cells, and did not affect the growth of granulosa cells. The growth and androstenedione production by theca cells were increased by the presence of granulosa cells, irrespective of the origin of follicular cells. These results demonstrated that communication between two types of follicular cells results in reciprocal modulation of their morphology, structure, growth, and function. Cellular interactions seem to be one of the major factors controlling the differentiation and growth of the follicular cells during the follicular maturation process. In contrast to granulosa and theca cells cultured alone, cells in the coculture seemed to possess morphological and functional characteristics more similar to those of cells in the growing follicular wall in vivo. Thus, we speculate that the interaction between these two types of follicular cells is essential for the maintenance of original structure and function of the bovine follicular wall.

Objective: In our previous papers, we demonstrated that TCDD and PCBs (both coplanar PCB 126 and non-coplanar PCB 153) caused the decreased estradiol secretion in cultured pig ovarian follicles. However, the mechanism of action is not clearly understood. Moreover, to our knowledge, the expression of AhR in pig follicular cells was not described yet., Methods: In order to elucidate if TCDD, PCB 126, and PCB 153 can disrupt ovarian steroidogenesis via AhR-dependent mechanism, granulosa and theca cells were isolated from pig ovarian small follicles and subsequently treated with 32 pg/ml of TCDD, 100 ng/ml of PCB 153, 100 pg/ml of PCB 126 in an in vitro culture. After 3-hour exposure to the chemicals, cells were prepared for immunohistochemical analysis or collected for immunobloting, and the effects of TCDD and PCBs on the levels of AhR protein were studied., Results: Under basal conditions, AhR staining showed a diffuse cytoplasmic pattern of distribution as well in granulosa and theca cells. However, immunocytochemical labeling of AhR in theca cells was less evident and only few cells displayed cytoplasmic staining. Immunoblotts prepared from granulosa and theca cell lysates detected a strong band of 120 kDa indicating AhR protein expression. The level of AhR expression detected by Western blot analysis was higher in granulosa cells than in theca cells. Intensive cytoplasmic and nuclear labeling corresponding to AhR protein in TCDD and PCB 126 treated granulosa cells was observed. In PCB 153 treated cells, the level of AhR staining was comparable with control. Western blot analysis showed a strong band in TCDD-treated grnulosa cells and only slightly differences compared with control in those exposed to PCB 126. The effect of both PCBs on immunocytochemical AhR staining in theca cells was less evident. Immunoblott analysis did not reveal any substantial differences between the control and TCDD- and PCBs-treated theca cells., Conclusions: This study has shown different AhR expression in porcine theca and granulosa cells, in respect of both its intracellular localization and cellular distribution. Both, dioxin and dioxin-like PCB activated AhRs in granulosa but not in theca cells. Differences in AhR-responsiveness of granulosa and theca cells to TCDD and dioxin-like PCB 126 are probably connected with the differences of these cells in estradiol secretion and different proliferative potential.

Theca cells are the endocrine cells associated with ovarian follicles that play an essential role in fertility by producing the androgen substrate required for ovarian estrogen biosynthesis. Theca cells differentiate from the interfollicular stroma in response to proteins secreted from growing follicles. The most common endocrine cause of infertility is associated with excessive proliferation of theca cells and ovarian hyperandrogenism. Cell facts: -ovarian androgen-producing cells; -are associated only with developing follicles; -over-activity of theca cells causes infertility due to hyperandrogenism; -under-activity of theca cells causes infertility due to lack of estrogen. The diabetes (db/db) mutation (i.e., leptin membrane receptor defect) promotes a progressive, hypercytolipidemia within ovarian follicular granulosa, thecal and interstitial layers of C57BL/KsJ mice which manifests an infertile, acyclic hypogonadal syndrome. The current studies focus on the structural, cytochemical and gluco-/lipo-metabolic changes which induce cellular lipoapoptosis and the resulting cytostructural disruption of db/db follicular populations, relative to littermate control indices, following the expression of progressive ovarian hypercytolipidemia. Control (normal: +/+ and +/?) and diabetes (db/db) genotype groups were prepared for high resolution light microscopic (HRLM) analysis of cytolipidemia and nuclear apoptosis (TUNEL-labeled 3'-DNA fragmentation) indices and compared to the transmission electron (TEM) microscopic analysis of ovarian follicular samples collected from 8-16-week-old groups. Compared to controls, the db/db mutation induced a dramatic increase in cytolipid vacuole volume and density within all ovarian follicular layers. TEM analysis revealed that the lipid vacuoles initially aggregated along the inner membrane compartments of affected thecal and granulosa cells in response to the interstitial and vaso-lipidemic-hyperglycemic conditions which characterized the ovarian microenvironment of db/db follicles. Progressive cytoplasmic movement of lipid pools into the perinuclear compartment of affected granulosa cells induced nuclear isolation from cytoplasmic organelles that were displaced towards peripheral intracellular compartments. Cytochemical analysis of lipid vacuole accumulations indicated attraction towards, and incorporation within, the nuclear envelope of hyperlipidemic cells. Co-localization of nuclear apoptotic 3'-DNA fragments within identified hyperlipidemic granulosa cells was coincident with the cytochemical and ultrastructural identification of lipid penetration through the nuclear envelope in db/db mutants. These results are the first cytochemical evidence that the lipometabolic disturbances in db/db mutants, which promote hypercytolipidemia-induced premature ovarian involution, are coincident with lipoapoptosis-induced nuclear dissolution within follicular granulosa layers. The lipidemia-induced alterations in cellular and nuclear architecture suggests that the disturbances in glucose and lipid metabolic cascade activities in diabetes (db/db) mutants disrupts follicular cytointegrity, culminating in nuclear disregulation (as indicated by lipoapoptosis) which results in premature reproductive tract organo-involution and manifest reproductive sterility.

Theca-interstitial (T-I) cells play a fundamental role in the control of ovarian function. Steroidogenic activity and growth of the T-I cells are regulated by many paracrine and endocrine factors. However, little is known about the mechanisms controlling T-I death. In an in vitro model of apoptosis, purified rat T-I cells were cultured for 24 h with serum and subsequently for up to an additional 24 h with serum or in serum-free medium with or without insulin, insulin-like growth factors (IGF-I and IGF-II) and LH or 8-bromo-cyclic AMP (8Br-cAMP). Apoptosis was identified by histological assessment of nuclear morphology and by detection of internucleosomal cleavage and quantified by determination of [alpha32P]-dideoxy-ATP 3'-end labeling of low molecular weight DNA. Serum withdrawal resulted in nuclear condensation and fragmentation into apoptotic bodies of T-I cells and led to pronounced DNA cleavage. Insulin (10 nM) protected T-I cells from apoptosis, reducing DNA fragmentation by 39 +/- 8% compared to serum-free controls. IGF-I (10 nM) and IGF-II (10 nM) had comparable antiapoptotic effects, decreasing DNA fragmentation by 55 +/- 9% and 37 +/- 14%, respectively. In contrast, LH (100 ng/ml) and 8Br-cAMP (1 mM) augmented the pro-apoptotic effect of serum withdrawal, increasing DNA fragmentation by 85 +/- 55% and 72 +/- 42%, respectively. The antiapoptotic effects of insulin and IGFs and the pro-apoptotic effect of LH, acting via the cAMP system, may be important in the maintenance of T-I homeostasis. Moreover, excessive levels of insulin and free IGFs may lead to T-I cell hyperplasia characteristic of conditions such as polycystic ovary syndrome.

The distribution of androgen receptor (AR) and cytochrome P450 aromatase was investigated in paraffin sections of pregnant pig ovary. Ovarian follicles and corpora lutea were isolated from ovaries obtained on Days 10, 18, 32, 71 and 90 post coitum (p.c.). Androgen receptor was localized in the nuclei of granulosa cells of follicles of various sizes. In addition, some follicles demonstrated staining in the nuclei of theca interna cells. Stroma cells also exhibited a positive immunostaining. At early and mid pregnancy (up to Day 71) AR was expressed in the nuclei of luteal cells. Corpora lutea of Day 71 showed mainly cytoplasmic staining while on Day 90 almost all luteal cells showed staining exclusively in the cytoplasm. Immuno-staining for the presence of cytochrome P450 aromatase was very faint in all investigated ovarian structures. The results could suggest that the process of androgen aromatization plays a negligible role in the ovary of the pregnant pig.

The diabetes (db/db) and obese (ob/ob) genotype mutations induce a progressive, hypercytolipidemic condition within the ovarian compartments of the female reproductive tract that results in sterility and premature organ involution in C57BL/KsJ mice. The current studies focus on the ultrastructural changes that occur within the ovarian interstitial, thecal, and follicular granulosa cell layers during the progressive expression of these mutations which promote tissue cytolipidemia-induced organoinvolution. Control (normal: +/?), diabetes (db/db), and obese (ob/ob) genotype groups were prepared for high resolution light (HRLM) and transmission electron microscopic (TEM) analysis of ovarian tissue samples collected from 4 (young)- to 20 (aged)-week-old mice, allowing for the progressive influences of the mutational aberrations on tissue structure to be evaluated. Compared to controls, both (ob/ob) and (db/db) mutations induced a dramatic increase in ovarian interstitial, thecal and follicular granulosa cytolipid vacuole accumulations, which increased in density between 4 and 20 weeks of age. Initially, lipid vacuoles aggregated in the interstitial and thecal regions of ovarian follicles in response to the hyperglycemic-hypertriglyceridemic metabolic conditions typical of both (ob/ob) and (db/db) groups. Progressive cytoplasmic movement of the lipid pools established a perinuclear isolation from associated cytoplasmic organelles. Progressive lipid accumulations forced cytoplasmic organelles to peripheral cell compartments and altered the follicular cell profile towards that of adipocyte-like entities relative to controls. The progressive hypercytolipidemia-induced alterations in cell structure disrupted normal tissue continuity, which culminated in premature ovarian organo-involution and female reproductive sterility.

The aim of this study was to evaluate the rat ovarian morphological and function changes after pinealectomy (px). Two months after px, young female Wistar rats were sacrificed and the right ovaries were analysed morphologically and the left ovaries were used for steroid receptor binding experiments. Blood was collected and steroid hormone and melatonin levels were measured using radioimmunoassay kits. Results revealed that in the px group the rat ovaries had an increase in the number of atretic follicles and interstitial cells. These cells showed hyperactivity features on transmission electron microscopy and morphometric analysis (p < 0.05 compared with control and sham groups). Px-group serum showed an increase in estradiol (p < 0.05) and a decrease in progesterone levels (p < 0.05) compared with other groups. Moreover, progesterone receptor expression was lower than control and sham groups (p < 0.05). We postulate that pinealectomy leads to many morphological alterations of rat ovaries that are associated with functional changes in steroidogenesis and a decrease in progesterone receptor expression.

The goal of this study was to investigate histological changes of the rat ovary treated with either insulin or insulin plus human chorionic gonadotropin (hCG). The study was conducted in Celal Bayar University, School of Medicine, Animal Research Laboratory. Eighteen adult female Wistar rats were divided into three groups to receive saline, or insulin, or insulin plus hCG for 4 weeks. At the end of treatment the rats were sacrificed and the ovaries were evaluated with hematoxylin and eosin. There was no abnormal change in rats treated with saline. A thickened capsule, stromal hypertrophy and stromal cell hyperplasia, and no developing follicles, were observed in the insulin-only group. A thin capsule, developing follicles and corpora lutea, and normal theca cells and stroma were observed in the insulin-plus-hCG group. We conclude that insulin may lead to histological changes similar to stromal hyperthecosis and polycystic ovary syndrome, and may be one of the factors causing follicular arrest.

The amounts of lysophospholipase D (LPLD) and the ovarian protein kinase C delta (PKCdelta) increase during the course of pregnancy. Because LPLD is involved in the production of the bioactive phospholipid lysophosphatidic acid (LPA), we examined whether stimulation with LPA would influence PKCdelta in the ovary. We used immunoblotting and immunohistochemical methods to show that stimulation of bovine theca cells with LPA leads to an unexpected redistribution of PKCdelta from the cytosol to the perinuclear area and that in the presence of LH, LPA induces a complete nuclear translocation of PKCdelta. These effects of LPA are dose dependent, can be mimicked by phorbol ester, and are inhibited by a PKCdelta inhibitor, rottlerin. Concomitantly, under the same experimental conditions both LPA and the phorbol ester PMA (4beta-phorbol-12-myristate-13-acetate) augment LH-stimulated progesterone accumulation in this cell system. This functional effect of LPA and PMA is abolished in cells pretreated with rottlerin. It is unclear whether the nuclear localization of PKCdelta indicates a specific function of the enzyme in the bovine ovary. Because PKCdelta supports a luteotropic function in rodent models, a similar role in the bovine ovary is also likely.

Unlabelled: Several authors have shown the effect of LH or hCG gonadotrophins on steroid secretion in the chick embryo ovary. However, the effects of luteinising hormone or human chorionic gonadotropin on chick embryo ovary innervation have not been studied. The purpose of the present work was to study the subcellular aspects of interstitial cells innervation in chick embryo ovaries cultured with LH or hCG. Coob's White Rock female chick embryos at 7, 11, 15 and 19 days of incubation (in ovo) were employed. Explants of the left and right ovaries were cultured separately for 4 days in: 1--Basic medium (control): minimum essential medium (MEM-GIBCO) with 10% bovine fetal serum, 2--Basic medium with added LH or hCG (problems). The cultures were processed to be studied ultrastructurally (TEM). Left and right ovary explants were fixed two hours at room temperature in Karnovsky's fixative. The tissues were then post-fixed in osmium tetroxide in 0.1 M cacodylate buffer for one hour and after a rinse in buffer were dehydrated in acetone: afterwards they were embedded in Araldite resin. Thick sections stained with 1% Toluidine blue were examined and photographed using a Zeiss Photo II microscope. Thin sections were stained with uranyl acetate and lead citrate and examined in a Siemens ElmisKop 101 electron microscope., Controls: In both ovaries the isolated interstitial cells and the appearance of nerve fibers and nerve endings were observed in the explants of 11 day cultured during four days and in a similar way at the age of 15 days of in ovo development., Problems: With LH or hCG nerve fibers and nerve endings were observed both in the right gonad and in the medulla of the left ovary in close contact with the steroid-producing interstitial cells. Ultrastructurally, the interstitial cells are recognized by their typical organoids: SER, Golgi apparatus, mitochondria with tubular crests and abundant lipidic droplets. The innervation of these cells were seen in ovaries at 7 days development, cultured during 4 days. The present state corresponds to that of 11 days development in ovo. The nerve endings had mitochondria, microtubules and small and medium-sized vesicles, some with an electrodense content. These results in vitro suggest that innervation of the ovaries is controlled by an indirect mechanism via the hypothalamic-pituitary system and another local with production neurotrophic factors modulated by LH.

Our observations of bovine follicles indicated that the original histological classifications of atresia were inaccurate. A detailed histological, ultrastructural and immunohistochemical study of antral follicles from bovine ovaries collected from an abattoir and from animals whose large follicles had been monitored by ultrasonography was conducted to investigate this further. Nidogen and CD68 were immunolocalized to observe the follicular basal lamina and macrophages, respectively. In randomly collected ovaries, approximately one quarter of all antral follicles were undergoing antral atresia, as designated in this study. Antral atresia was characterized by early destruction of the layers of the membrana granulosa closest to the antrum, whereas the most basal cells remained intact. Numerous pyknotic nuclei were observed in the most antral layers and in the antrum close to the membrana granulosa. This is the classic description of atretic follicles and was observed at all sizes of follicle development and almost universally in large follicles (> 5 mm in diameter), including dominant follicles. Basal atretic follicles, as designated in this study, were almost as prevalent as the antral atretic follicles, and were characterized by initial destruction of the most basal layer of granulosa cells, whereas the cells in the most antral layers remained associated with each other and were predominantly healthy. Pyknotic nuclei and the nuclei of dying basal cells budded into apoptotic bodies were observed rarely. The basal lamina of basal atretic follicles was often breached by macrophages, which were phagocytosing dying basal granulosa cells. The theca was characterized by an increased deposition of collagen, and the cells were orientated randomly, rather than lying parallel to the membrana granulosa as in healthy follicles. Basal atresia occurred in small (< 5 mm in diameter) follicles only. Importantly, these basal atretic follicles were originally identified incorrectly in the literature. Thus, on the basis of the results of this study and another on the expression of steroidogenic enzymes in atretic follicles, it is suggested that the standard biochemical methods for measuring steroid hormone concentrations in follicular fluids to assess atresia should be re-evaluated.

In a previous work we demonstrated the relationship between nerve fibers and nerve endings and interstitial cells (estrogen-producing cells) from the atrophic right ovary and the medulla in the left functioning ovary during embryogenesis in the chick, in ovo. Besides, the local production of neurotrophins by steroidogenic cells is probably involved in the control of ovarian innervation. The objective of the present study was to analyze ultrastructurally the innervation during the differentiation of chick ovary cultured with 17-beta-estradiol. Explants of right and left ovaries from seven to nineteen days in ovo development were cultured separately for 4 days in MEM (controls) or in the presence of 17-beta-estradiol (problems). In controls the electron microscopic examination of the innervation explants from chick embryo ovaries revealed that the interstitial cells are well innervated. Nerve fibres and nerve endings were observed in close contact with steroid-producing cells, a similar pattern of innervation that those of the fifteen days ovaries in ovo development. Problems cultured from seven days showed nerve fibres and nerve endings at difference to controls. These results in vitro suggest that innervation of the ovaries is controlled by indirect mechanism via the hypothalamic-pituitary system and local production factors. More experiments are necessary to confirm this results.

Structural luteolysis shows striking interspecies differences. Morphological changes in the corpus luteum (CL) of the cyclic hamster have been studied alongside the potential involvement of known luteolytic hormones. Ovaries from intact Syrian golden hamsters killed at 1100 h on days 1 and 2 and at 1100 and 1700 h on days 3 and 4 of the estrous cycle were dissected for histological study. The day of ovulation, the day of estrus, was arbitrarily designated day 1 of the estrous cycle. Steroidogenic cells in the CL were scarcely luteinized on day 1 and reached full luteinization on day 2. On the morning of day 3, initial regressive changes (accumulation of lipid droplets, invasion by neutrophils, and accumulation of phagocytic cells) were observed. These regressive changes increased progressively and apoptotic cells as well as phagocytic cells containing phagocytized apoptotic cells were abundant on the evening of day 3. On the morning of day 4, apoptotic cells/bodies and phagocytic cells containing phagocytized material were extremely abundant throughout the CL. However, steroidogenic cells with intact nuclei and well-preserved blood vessels were also found. Surviving cells in the CL showed progressive morphological changes. These cells showed morphological features intermediate between luteal and interstitial cells in the evening of day 4 and were virtually indistinguishable from interstitial cells on day 1 of the following cycle. Additional animals were injected at 1100 h on day 2 with: (a) the dopaminergic agonist CB154 (0.4 mg) to block prolactin secretion, (b) the anti-estrogen LY117018 (1.6 mg) or the anti-androgen Flutamide (3 mg) to block estrogen or androgen receptors, respectively, and (c) progesterone (2 mg) to prevent the fall in serum progesterone concentrations. Ovaries from these animals were collected at 1700 h on day 3 and at 1000 h on day 4. The luteolytic process was not affected by any treatment. These data indicate that, in contrast to its close relatives (e.g., the rat), structural luteolysis in the hamster is independent of the apoptotic inducing luteolytic hormones. In addition, differences in the cellular mechanisms responsible for CL elimination were also present. In order to understand the fine structure and distribution of the interstitial glandular cells (IGCs) and associated elements in the human fetal ovary, we studied human fetal ovaries at 16 weeks post fertilization (p. f.) by transmission electron microscopy. Semithin sections revealed voluminous typical IGCs usually grouped in clusters, located in the interstitium among the ovigerous cords. Isolated primordial follicles were seen in the cords located close to the interstitium in which IGCs were present. Besides the main ultrastructural characteristics of steroid secreting cells, the IGCs showed lipofuscin granules and stacks of annulate lamellae in their cytoplasm. Fibrocytes, macrophages and mast cells were detected close to the IGCs. In particular, the fibrocytes were located around the IGCs, with which they occasionally formed focal cell contacts. Fibrocytes issued numerous long projections, which, together with collagen fibers, surrounded the clusters of IGCs and small vessels (mainly capillaries), often extending into the intercellular spaces among IGCs. These data indicated that, already at the initiation of folliculogenesis, the IGCs are present numerously in a close association with the ovigerous cords. The morphological aspects of IGCs were comparable to that of fetal testis interstitial (Leydig) cells and hilar cells in adult ovary, and suggest that fetal IGCs may be source of adult ovary hilar cells. In addition, we have here demonstrated for the first time that IGCs are associated with stromal cells whose distribution seems to support IGCs microtopography. Fetal ovarian fibrocytes revealed a structural arrangement similar to that of the "compartmentalizing cells" previously described in the adult testis. Macrophages and mast cells presumably have a role as local modulators of steroid synthesis. Mast cells may also affect fibrocyte organization and vascular permeability. We thus suggest that IGCs and associated cells may form a glandular unit in the human fetal ovary similar to that in the adult testis, and this structure is likely involved in early steroid secretion during gonadal differentiation.

The present study was designed to determine whether rat pre-antral follicles can grow under in-vitro conditions. Emphasis is on whether follicular interaction is involved in in-vitro follicle culture, and furthermore its role in follicular development has been assessed. Pre-antral follicles were isolated mechanically from 10-day old rat ovaries. They were divided into small (50 microm < diameter < 100 microm) and large (120 microm < diameter < 200 microm) pre-antral follicles and cultured individually or in groups for 6 days in medium with or without fetal calf serum (FCS). Based on morphological criteria, large pre-antral follicles cultured in groups in serum-free medium had significantly higher survival rates than those cultured individually. In the presence of FCS, no significant difference was detected with respect to the survival. However, the large pre-antral follicles cultured in groups had a significantly greater increase in diameter than those cultured individually. Furthermore, follicles cultured in groups in FCS-containing medium exhibited significantly more follicular cell proliferation than those in serum-free medium, based on DNA measurement. The present culture system (with or without FCS) proved to be insufficient for small pre-antral follicles to stimulate growth comparable to that of large pre-antral follicles. The transmission electron microscopical (TEM) study revealed the ultrastructural differences between follicles cultured in FCS-containing and serum-free media. Taken together, the results suggest that interfollicular factors are involved in follicle development in vitro, which especially at the early folliculogenesis stage plays a positive role in terms of follicular growth as well as survival. Objective: To localize (by the light and electron microscopy) and partially characterize the antigen recognized by the Mab4E6 in rat ovaries., Methods: Monoclonal antibody (4E6) against a rat ovarian granulosa cell antigen was prepared and identified the 40kDa protein specific for rat steroid producing cells. The localization of this antigen was studied by light and electron microscopic immunocytochemistry., Results: The immunocytochemical observation suggested that the recognized antigen was localized in granulosa and thecal cells in all stages of follicular development. The intensity of immunostaining was found to depend on the developmental stage. In granulosa and thecal stage (health follicle) Mab 4E6 binding molecule was localized on the membranes of rough endoplasmic reticulum (RER) and on the surface of lipid droplets in close association with the RER. In atretic follicles we established that the final destination of the visualized antigen is in structures which we refer as the autophagic vacuoles in close contact with the steroidogenic organelles. In addition, we observed Mab 4E6 binding molecule in the cytoplasm of luteal cells. Leydig cells and adrenocortical cells., Conclusions: The results indicate that the 40kDa antigen may be common to all of rat steroidogenic organs. Our results suggest that the 40kDa protein may be associated with the processes governing steroidogenesis and/or follicular development.

We have shown recently that the bovine corpus luteum (CL) possesses specific luteal cell surface membrane binding sites for progesterone. We have now confirmed and extended these observations to compare the subcellular distribution of these binding sites in developing, mature and regressing CL. The median buoyant densities of luteal progesterone binding sites from early-, mid- and late-luteal phase CL were similar (though three of five density profiles for late-luteal phase CL showed association of steroid binding with a fraction with a lower density), and clearly resolved from nuclear, mitochondrial, lysosomal, peroxisomal, Golgi-endoplasmic reticulum-lysosomal and smooth endoplasmic reticulum markers. Specific binding of [3H]progesterone overlapped with the distributions of 5'-nucleotidase and luteinizing hormone receptor (luteal cell surface membrane markers) in both control and digitonin-treated gradients at all stages of the luteal phase. Since steroidogenic 'large luteal' and 'small luteal' cells of the CL are derived from the granulosa cells (GC) and theca of the preovulatory follicle, we also investigated whether similar receptors were present in the follicle, and describe for the first time specific membrane binding sites for progesterone in purified GC and thecal membranes from healthy bovine follicles of different sizes. Specific binding increased linearly with GC and thecal membrane protein concentration; however, it was detectable only when digitonin was included in the binding incubation. Binding sites were specific for progesterone; unlabelled progesterone competed for [3H]progesterone binding at low concentrations (IC50, 35 and 31 nmol/l) compared with testosterone (IC50, 905 and 870 nmol/l) and delta4-androstenedione (IC50, 1050 and 660 nmol/l) for GC and thecal receptors respectively. In contrast, oestradiol, oestrone, pregnenolone, cortisol, cholesterol, and a genomic progesterone receptor antagonist, RU486, competed poorly. Steroid binding was present in GC and thecal membranes of follicles of all sizes, but [3H]progesterone binding to GC membranes decreased significantly with increasing follicle size (P<0.02), perhaps indicating developmental regulation of GC membrane non-genomic progesterone receptors in the preovulatory bovine follicle. We suggest that these membrane steroid receptors may be involved in the autocrine/paracrine regulation of follicular function by progesterone.

This morphological study on the ovary of the seal focuses on two structures of special interest: the subsurface crypts and the interstitial cells. Ovaries from 110 seals (from 1,5 months to 12 years) were used. Most of the seals of our study originated from German North Coast, a few from Island. The ovaries were examined by light microscopic as well as by transmission and scanning electron microscopic techniques. The seal's ovary conforms to the basic structure of the ovary of other mammalian species. However, the surface epithelium cell layer lowers itself into an epithelium lined - broadly branching system of subsurface crypts. The size and extent of these crypts and the proliferation of the epithelium appear to be mainly related to the reproductive activity. The ultrastructure of crypt epithelium cells indicates a remarkable metabolic activity. In juvenile ovaries cell buds proliferate from the crypt epithelium and grow as plug-like strands into the cortical stroma. These cells show ultrastructural changes during their proliferation and appear to transform into steroid producing interstitial cells. In the adult seal the interstitial cells are completely differentiated established and, according to their origin, can be divided into epithelial and fibroblastic interstitial cells. It is suggested that a major function of the crypts is the formation of the ovarian blastema with particular reference to ovarian interstitial cells. The intensive proliferation of interstitial cells is in agreement with the extraordinary high level of 350 pmol estrogen/l blood serum.

The germinal disc of the female gamete and its overlying follicular wall (the germinal disc region) is hypothesized to play an important role in the regulation of follicular growth. The role of the germinal disc region in follicular growth in chickens was investigated by destroying the germinal disc region or an area opposite the germinal disc (control) of the second largest follicle 22-23 h before ovulation of the F1 follicle, by localized freezing with solid CO2. Structural changes of the follicular wall (non-frozen region) were observed by electron microscopy 10-20 h after the destruction of the germinal disc region. Development of the inner structure of mitochondria in granulosa cells and accumulation of lipid droplets in thecal cells were observed in follicles 15 h after destruction of the germinal disc region. Twenty hours after destruction of the germinal disc region, follicles showed early signs of atresia (bursting atresia). Degenerative changes in follicles, including hydrolysis by lysosomal enzymes, were present in thecal fibroblast-like cells. Control follicles, in which an area opposite the germinal disc region was frozen 20 h before examination, had no degenerative features. These results provide further evidence that the germinal disc region is required for follicular growth.

To study ovarian follicular development, we established a procedure to reconstruct a follicular wall in vitro by using three-dimensional collagen gel. Granulosa cells and thecal cells were dissociated from median follicles of porcine ovaries. Isolated thecal cells were embedded in collagen gel and a medium containing isolated granulosa cells was overlaid. A monolayer culture of recombined granulosa and thecal cells in collagen gel was used for the control. To examine the synergism of granulosa cells and thecal cells in this culture system, isolated granulosa and thecal cells were cultured either singly or in combination. The culture media were assayed for steroids by RIA. Cultured granulosa cells in this culture system were epitheloid and grown in multilayer aggregates. In contrast, cells cultured by the conventional method were as flat as a monolayer sheet. Ultrastructural findings for these cells showed abundant organella. Gap junctions were often observed between these cells and the basal lamina-like structure. Estradiol production by both culture methods was significantly higher than the sum of that produced by granulosa and thecal cells cultured separately. Androstenedione accumulation was increased by this culture system, but was suppressed by the conventional culture system. These findings suggest that the constitution of the ovarian follicular wall plays an important role in morphological and functional interaction between granulosa and thecal cells.

Diazepam-binding inhibitor (DBI) is the precursor of a family of peptides, including an octadecaneuropeptide (ODN), that share with DBI the ability to specifically displace benzodiazepines (BZD) from their receptors. An association of ODN with the peripheral type BZD receptors (PBR) has been reported in the brain and a few peripheral tissues. In order to investigate whether DBI and PBR are present in ovarian tissue, we have localized DBI by means of immunocytochemistry, in situ hybridization and autoradiography of PBR in the rat ovary. Immunocytochemical localization was achieved by means of rabbit antibodies developed against rat ODN. Immunostaining was located in the cytoplasm of the theca interna, corpus luteum and interstitial gland cells, but not in the granulosa cells. Hybridization signal obtained following in situ hybridization with a [35S]-labelled single-stranded RNA probe complementary to DBI mRNA was observed in all the steroid-secreting cells, including granulosa cells of developing and mature follicles. Autoradiographic localization of PBR obtained by incubating ovary sections with [3H] PK11195, a ligand selective for PBR, revealed the presence of specific labelling in all the steroid-secreting cells. These results, which demonstrate for the first time that the ovarian steroid-secreting cells contain both PBR and its endogenous ligand, suggest that the BZD receptor might be involved in the regulation of ovarian function.

Paracrine/autocrine factors like epidermal growth factor (EGF) and transforming growth factor (TGF alpha) produced in the mammalian ovary modulate ovarian functions such as cell proliferation, differentiation, and steroidogenesis. To study similar activities in the avian ovary, we have developed a culture system for chicken granulosa cells using either serum containing or serum-free media. In the defined medium, granulosa cells for the F1, F2, and F3 follicles of the chicken ovary proliferated as indicated by enhanced levels of DNA, synthesised progesterone in similar amounts to those observed in acute experiments, and responded to luteinizing hormone (LH) during and after the culture period. The morphology of the cells and profile of growth in both media (although better in serum than in serum-free medium) are similar to those reported for mammalian cells. EGF stimulated granulosa cell proliferation over 72 hr in culture while inhibiting basal and LH-stimulated progesterone production; these effects were greatest in the less mature cells taken from the F3 follicles. Theca-conditioned medium (TCM) from chicken ovarian theca cell cultures mimicked the effects of EGF when cultured with granulosa cells. Taken together, these results suggest that EGF/TGF alpha-like peptides have a role in avian ovarian function. Moreover, the effects observed for TCM suggest that these peptides may be present in the theca layer with a paracrine function in the adjacent granulosa.

As a step towards understanding the role of prostaglandin F2 alpha (PGF2 alpha) in ovarian function, a rabbit antiserum against purified PGF2 alpha receptor (PGF2 alpha-R) was produced. This report details the use of this antiserum in immunohistochemical staining of ovaries of non-pregnant and pregnant rats to ascertain which cell types, in vivo, possess PGF2 alpha-R. In non-pregnant rats, three ovarian cell subpopulations contain immunoreactive PGF2 alpha-R. These include: a subpopulation of the cells found in corpora lutea, a subpopulation of the thecal cells surrounding secondary and mature (Graafian) follicles, and a subpopulation of primary and secondary interstitial cells. The ovarian tissues and cell types in which immunoreactive PGF2 alpha-R cannot be demonstrated include: the serosa overlying the ovary and its vessels, the coelomic epithelium and its underlying cortical stroma, medullary stroma and vessels, granulosa cells of primary, secondary and mature follicles, the oocyte, and the blood vessels and stroma within corpora lutea. PGF2 alpha-R immunohistochemical staining of corpora lutea from non-pregnant animals was examined both prior to the start of luteolysis and during luteolysis. During luteolysis, cells undergoing apoptosis stained for the presence of PGF2 alpha-R. PGF2 alpha-R immunohistochemical staining was also examined in corpora lutea during pregnancy and until 4 days postpartum. The major findings here were the apparent large increase in staining intensity of granulosa-lutein cells during pregnancy, and the loss of PGF2 alpha-R immunopositivity of the granulosa-lutein cells during the postpartum period. In summary, three ovarian cell subpopulations, all of which can secrete steroids, possess immunoreactive PGF2 alpha-R.

According to available literary data, an increased nuclear volume of endocrine gland cells, various structures of the diencephalon and endocrine ovarial structures could be observed when the respective organs and structures were more active; a decreased nuclear volume was revealed when the activity of the respective organ or its structure was suppressed (Mess, 1962; Maracek and Arendarcik, 1976, 1978). Attention was paid to the karyometric analysis of follicular cells of the stratum granulosum (SG) and secretory cells of the theca folliculi interna (TFI) during selection of the dominant ovulatory follicle (DOF). The aim of this study was to determine differences in hte nuclear volumes of the granulosa follicular cells and the secretory cells of the inner theca of the DO1F in comparison with the largest atretic follicles (AF) as well as to verify the use of karyometric variance analysis for the evaluation of the selection process of the dominant ovulatory tertiary follicle. Histological sections were prepared from large DOF and AF from the ovaries of 18 sheep aged 3-5 years and held under standard conditions of a controlled herd. Control group I (n = 3) consisted of Wallachian sheep on day 15 of the sexual cycle (day 0). Excisions of ovaries from the animals of the experimental group (n = 3) were made on day 16 of the cycle after i.m. treatment with 120 micrograms GnRH 10 and 7 hours prior to sampling. Control group II (n = 3) consisted of Tsigai sheep at the 9th or 10th day of the oestrous cycle (hour 0) and three experimental groups (n = 3 each) consisted of Tsigai sheep 24, 48 and 72 hours after i.m. treatment with 125 micrograms cloprostenol. After excision the ovaries were fixed in neutral formaline, cut into 4 mm thick transverse segments and subjected to standard histological processing. After staining with Harris' HE the 5-7 microns thick sections were karyometrically evaluated at a magnification of 2200 x according to the method of Palkovits (1961) using the PC programme Karyotest 03 (Maracek et al., 1991). 200 cells were evaluated from each sample and both cell types; altogether 7200 cells were examined. GnRH treatment (Dirigestran inj. Spofa) increased the nuclear volume of follicular cells (SG) in the dominant ovulatory follicle in the process of selection (Tab. I, Fig. 1). The former, however, reduced the nuclear volume not only of the SG follicular cells of LAF (Tab. II, Fig. 2) but also of the secretory cells of both the DOF and AF theca interna (Tab. II, Figs. 3, 4). Cloprostenol treatment (Oestrophan inj. Spofa) affected the follicular cells of the granulosa in the dominant ovulatory follicle.(ABSTRACT TRUNCATED AT 400 WORDS)

Follicular atresia was studied in the ovary of the electric ray, Torpedo marmorata, by light and electron microscopy. The course of atresia may be divided into four stages. The first two comprise the dissolution of the oocyte and its phagocytosis by the small cells of the granulosa epithelium. The third stage consists of the transformation of the granulosa epithelium into an active glandular structure and is accompanied by the development of a smooth endoplasmic reticulum. The fourth stage is marked by sclerosis and pigmentary degeneration of the atretic follicle. Together these observations suggest an endocrine steroidogenic role for the corpora atretica (preovulatory corpora lutea) in T. marmorata.

Angiotensin II (AII) is present in the mammalian ovary and has been correlated with atresia in follicles. Since the theca interna may be one site at which atresia is initiated, we wished to determine whether AII exerts an effect on theca interna from explanted ovarian follicles of hamsters. Hamsters were sacrified on the morning of proestrus, and ovaries were removed. Preovulatory follicles were excised from the ovaries, and cultured with one of the following components: medium alone (control); medium plus AII (1 x 10(-6) M); the AII-receptor antagonist [Sar1, Ile8] AII (1 x 10(-4) M); or AII plus antagonist. After 72 h, the follicles were processed for transmission electron microscopy (to determine quantities of theca interna organelles involved in the steroid synthetic pathway) or for protein determination (to normalize steroid production rates). The incubation medium was drawn off and analyzed by radioimmunoassay for progesterone, androstenedione, or estradiol-17 beta. There was a significant positive correlation (r = 0.92, P less than 0.01) between follicular androstenedione secretion and area comprising theca interna smooth endoplasmic reticulum. In the theca interna, AII induced a two-fold and 1.6-fold increase in lipid droplet number and area comprising smooth endoplasmic reticulum, respectively (P less than 0.05). Excess antagonist negated the increase in cell organelles and also reduced androstenedione secretion compared with AII alone (P less than 0.05). Most importantly, AII significantly augmented the ratio of androstenedione:estradiol-17 beta secretion by 44% over that of control. The ultrastructural changes observed in this study and the increase in the androstenedione:estradiol-17 beta production ratio are consistent with atresia-like changes in ovarian follicles.(ABSTRACT TRUNCATED AT 250 WORDS)

In order to study the three-dimensional topographic arrangement of the oocyte and zona pellucida, follicular cells and follicle microvasculature, this study applied alkali maceration methods for tissue exposure, the ruthenium red-detergent method for the extracellular matrix visualization, and the vascular corrosion cast technique to rabbits, guinea-pigs and mice ovaries at different stages of follicular development. Macerated samples showed a gradual differentiation of the oocyte surface. This, in primordial follicles, appeared rather smooth, but, with the follicular development, displayed a gradual increase of blebs and microvilli. The latter widely covered the surface of oocytes contained in large or mature follicles. The outer surface of the zona pellucida showed numerous fenestrations, whereas the inner one was smooth. The ruthenium red-detergent method permitted a well detailed view of the filamentous texture of the zona pellucida. The three-dimensional distribution of the contacts between oocyte and neighbouring follicular cells was clearly evaluated in macerated samples. Follicular cells of primary follicles were characterized by their short cytoplasmic processes reaching the oocyte surface. In secondary follicles, these processes issued secondary processes. In larger follicles, the secondary processes of the corona cells were much longer and thinner, and took a tortuous course to reach the oocyte surface, which ran among the numerous oocyte microvilli. This microvillous arrangement greatly increases contact between the oocyte and corona cells, and suggests a coordinated reciprocal control of the activities of both cell types. These data also showed that the spongy and filamentous nature of the zona pellucida is closely dependent upon the temporal differentiation and enormous increase in number of follicular cell projections and their ramifications. Maceration revealed the theca cells surface. In smaller follicles these appeared as fusiform cells which resembled fibroblasts. In larger or mature follicles, many theca cells differentiated to possess morphological features of steroidogenic cells. In addition, these cells delimited a series of intercellular communicating lacunae, continuous with wide pericapillary spaces. The gradual differentiation of the follicle towards a structure having an endocrinal role was further emphasized in vascular corrosion casts. A simple microvascular net made of thin capillaries supplying primary follicles was seen to transform into an elaborate sinusoidal network made of thick permeable capillaries, supplying mature follicles.

Porcine follicular maturation begins by recruitment from a continually proliferating pool of small antral follicles; those receiving the appropriate stimulus differentiate rapidly through a series of structural and functional changes. Such ovarian activity can be induced in prepubertal gilts with a single injection of equine chorionic gonadotropin (eCG). Average follicular diameter in eCG treated females increased from approximately 2 mm before stimulation to 3.5 mm by 24 hr after injection, with subsequent growth to ovulatory size (8 or 9 mm) by 96 hr. Both theca and granulosa layers increased in thickness and complexity, and a prominent capillary bed evolved immediately outside the basement membrane separating the two layers. Cytoplasmic organelles associated with increased metabolic activity and steroidogenesis proliferated within the first 24 hr. Progressive changes included increasing amounts of lipid and rough and smooth endoplasmic reticulum, with the latter occurring in vesicular or lamellar forms and as lipid-associated whorls. Bizarre mitochondrial forms also appeared, often associated with lipids. The amount and proportion of rough and smooth endoplasmic reticulum shifted dramatically as follicles matured. By 24 hr, rough endoplasmic reticulum in thecal cells increased from 4.2 to 7% of cell volume, while the amount in granulosa cells increased from less than 3.5% to more than 10%; the quantity remained relatively constant in the theca but declined to prestimulation values in the granulosa layer. Rough endoplasmic reticulum predominated over smooth in the first 24 hr following stimulation but the proportions were then reversed, so that more than 10% of both layers was composed of smooth endoplasmic reticulum by the time ovulation was imminent. Some follicles had or were in the process of ovulating by 96 hr. Their walls were collapsed into prominent folds with the two cell types beginning to mix. Slight undulations and some regions of discontinuity were observed in basement membranes of large unovulated follicles at this time. In specimens collected at 96 hr poststimulation and processed for retention of lipid, lipid-like material was noticeable in the extracellular matrix surrounding cells that contained organelle configurations suggestive of steroidogenesis.

The purpose of the present study is to provide further evidence in support of the theory which suggests that two cells are involved in the secretion of estrogens and androgens in the theca of the preovulatory ovarian follicles in the domestic fowl. The third (F3), the fourth (F4), and the fifth (F5), in order of decreasing size follicles, were obtained from white Leghorn hens. Theca cells were dispersed by trypsin treatment, and isolated cells were centrifuged in a continuous Percoll gradient (0-100%). Testosterone and 17 beta-estradiol secretion from cell samples in each density gradient fraction were radioimmunologically assayed. The highest secretion of 17 beta-estradiol was registered in cells isolated in density fractions ranging from 1.059 to 1.070 g/ml, while the maximal testosterone production was found in fractions from 1.037 to 1.048 g/ml. The different results were obtained from the three types of follicles studied (F3-F5). Morphological study of the F4 follicle showed typical steroidogenic cells with abundant lipid droplets in the cytoplasm in the theca interna, whereas clusters of poorly differentiated epithelial cells were visualized between fibroblast layers of the theca externa. The secretion of aromatizable androgens by the typical steroidogenic cells of the theca interna and estrogen production in the epithelial cells of the theca externa are postulated.

We observed the ultrastructure of the cumulus-corona cells (CC cells) surrounding: 1) human preovulatory oocytes unfertilized after in vitro insemination and 2) in vitro-fertilized polypronuclear ova (PO) at the pronuclear stage (3 pronuclei) and during early cleavage, at the 3-8 cell stage (cleaving PO). All the samples were obtained from women who underwent pharmacological hormonal stimulation during in vitro-fertilization procedures. Both cell groups were composed of irregularly rounded CC cells, showing an oval nucleus with one or more reticular nucleoli. Spermatozoa in close contact with CC cells were also seen. Linear and annular gap junctions between neighbouring cells were present, particularly in Group 1. Lipid droplets were present in both groups, appearing slightly more numerous and electron-dense in Group 2. In Group 1, mitochondria were numerous, polymorphic, and provided with cristae varying from lamellar to tubular. In Group 2, mitochondria also showed polymorphism, with bacilliform organelles with tubular cristae being predominant. In both groups cisternae and associated vacuoles and vesicles belonging to the Golgi complex were scattered in the cytoplasm of CC cells. Similarly, tubular and vesicular profiles of smooth endoplasmic reticulum were abundant and uniformly distributed throughout the cytoplasm of CC cells of both groups. In contrast, the abundant rough endoplasmic reticulum in Group 1 was formed by parallel stacks of flattened cisternae, whereas it was less plentiful and not arranged in stacks in Group 2. The CC-cell surface appeared covered by numerous membrane expansions in both groups. The expansions in Group 1 were mainly composed of blebs of various sizes and a few short microvilli, whereas in Group 2 numerous microvilli covered the cell surface. These observations demonstrate that a gradual establishment and maintainance of a steroidosynthetic capability (luteinization) takes place in CC cells, particularly during and shortly after fertilization, as occurs contemporaneously in the granulosa cells of the postovulatory follicle. Our results may be considered as ultrastructural confirmation of the capability of the CC cells to produce small amounts of steroids (estrogens and mainly progesterone). These hormones, alone or together with other substances (proteins, nutrients, growth factors?), might--around the fertilization time--act positively upon the early embryo itself, as well as on the microenvironment in which the embryo develops, both in vivo and in vitro conditions.

In a previous work we demonstrated that germ and epithelial cells of the atrophic right ovary and of the medulla in the left functioning ovary undergo regression while interstitial cells are preserved. The objective of the present study was to analyze the relationship between nerve fibers and nerve endings and interstitial cells from both ovaries during embryogenesis in the chick. To this purpose, left and right ovaries from embryos of 7 to 19 days of development were processed for ultrastructural study and ultracytochemical determination of acid phosphatase. At 7 days, isolated interstitial cells were found in the central region of both ovaries, containing SER, Golgi apparatus, mitochondria with tubular cristae and abundant lipidic vacuoles. Nerve fibers and nerve endings were scarce. From the 11th day, interstitial cells were found grouped in the right ovary and in the medulla of the left one, and were related to nerve bundles surrounded by Schwann cells. The nerve ending, in close contact with the membrane of interstitial cells, contained mitochondria, microtubules and small and big vesicles, some with an electro-dense content. This relationship between interstitial cells and nerve endings and fibers was more frequent from the 15th day. The activity of acid phosphatase localized in germ and epithelial cells presented a more intense reaction in the right ovary and in the medulla of the left ovary at 15 days.(ABSTRACT TRUNCATED AT 250 WORDS)

We have developed a culture system in which bovine granulosa and theca cells are allowed to attach to opposite sides of a collagen membrane. We studied the interaction between theca and granulosa cells by investigating the morphology, proliferation, and steroidogenesis of the cells. Granulosa cells cultured alone were flattened and polygonal and formed monolayer sheets. Granulosa cells cocultured with theca cells formed multilayer sheets. The apical surface of each cell appeared convex. Numerous filopodia spread over the cellular surface connecting cells. Theca cells cultured alone were thin, flat, and spindle-shaped. Theca cells cocultured with granulosa cells were also spindle-shaped; however, the apical surface appeared convex. Cocultured cells were more densely packed than theca cells cultured alone. The number of both granulosa and theca cells in the cocultured group increased approximately twofold compared to control cells cultured alone. Progesterone content per 1 x 10(5) granulosa cells in 24-h culture medium of the cocultured group was reduced to 40% of that of the control group. In contrast, androstenedione content per 1 x 10(5) theca cells of the cocultured group increased approximately threefold compared to androstenedione content of control group. These results indicate that communication between these two types of follicular cells results in reciprocal modulation of their proliferation, morphology, and function.

The embryonic gonads produce and secrete the steroid hormones that induce secondary sex differentiation in different species. The production of estrogens, testosterone and progesterone by interstitial cells of the chick embryo ovary has been demonstrated in experiments in ovo and in vitro. Estrogens would be responsible at the 7th day of in ovo incubation, of the evolution of the functional left ovary. According to Gasc, only the cells of the germinative epithelium of the left ovary possess receptors for estrogens, thus originating the formation of a cortex in this functional gonad and not in the right one. On the other hand, extragonadal factors, such as gonadotrophins, would participate in gonadal differentiation. Involvement of the hypophysis in the morphological and functional differentiation of the ovary in the chick embryo, has been considered in several studies. Woods and Week established the functional hypophysealgonadal axis at 13 days of in ovo incubation. However, the effects of individual gonadotrophins on the interstitial cells of the chick embryo remain unclear. Avila et al. and Grassi Milano observed that when the female gonads were cultured without steroid or gonadotrophic hormones at the start of differentiation an hermaphrodite left ovary and a male right one were formed. Kyparissi and Vakolopoulu observed that the inoculation in ovo of FSH at the end of embryonic development led to an increment in the number and size of follicles in the left ovary. Pittini and Grassi Milano added FSH and LH to cultures of 15 days-old chick embryos testicles and ovaries and found that only male gonads developed similar to normal embryos.(ABSTRACT TRUNCATED AT 250 WORDS)

Ovulation may be achieved and studied in an isolated perfused rabbit ovary upon inclusion of human chorionic gonadotropin (hCG) in the perfusion fluid. The ultrastructural features of the rabbit ovarian follicle prior to ovulation in vitro were compared with those in vivo. The perifollicular vasculature was also examined in in vitro perfused rabbit ovaries during the preovulatory interval. Granulosa cells of the preovulatory follicle share many ultrastructural features in vivo and in vitro; however, only small amounts of smooth endoplasmic reticulum (sER) were observed in granulosa cells in vitro after hCG. Ovulation after hCG in the in vitro preparation tends to occur earlier (6 hours) than in vivo (12 hours). Thus, there may be insufficient time and/or gonadotropin exposure to permit full functional development of granulosa cells, as reflected by reduced amounts of sER. Degradation of collagen fibrils was less prominent in the theca externa and tunica albuginea in vitro than in in vivo. Perifollicular capillaries became dilated after hCG, but interendothelial gaps were not observed. Disappearance of surface epithelium in the apex of follicles was similar in vitro and in vivo.

In previous investigations we could demonstrate that angiitis or angiolytic changes of the vascular wall with loosening or duplication of the basement membrane may be caused by a low calcium diet. In this paper we present experiments using the same experimental procedures which lead to massive necrotic changes of granulosa cells in the ovary. These phenomena are thought to be induced by loosening and swelling of the basement membrane which delimits the granulosa layers from the outermost tissue.

The observations that perifollicular capillaries of the ovary become engorged with prominent edema in the theca interna and externa around the time of ovulation, suggest a preovulatory increase in permeability of perifollicular capillaries. In order to obtain an ultrastructural evidence for this increased capillary permeability, we examined the perifollicular capillaries in the theca interna by using mature rabbit ovaries at various time intervals after the hCG injection for induction of ovulation. We observed a capillary engorgement at 9, 10, 12 hours after the hCG injection, most prominent at 9 hours, and edema in the pericapillary space at 10, 12 hours. Pinocytotic vesicles were seen through the all stages of follicular development and they did not show any significant changes in their size, number and distribution. Fenestrations were observed at 0, 3, 4, 6 hours after the hCG injection, most frequent at 4 hours. Intercellular gaps (500 approximately 1,200 A) between two endothelial cells were demonstrated at 10, 12 hours, and thrombocytes in these gaps were also noticed. The present observations suggest that increased permeability of perifollicular capillaries just prior to ovulation were mainly due to the formation of gaps between the endothelial cells.
