Objectives: This systematic review was designed to summarize randomized controlled trials of intra-articular administration of non-steroidal anti-inflammatory drugs (NSAIDs) for temporomandibular disorders. Methods: Randomized controlled trials regarding intra-articular injections of non-steroidal anti-inflammatory drugs for temporomandibular disorders were included in the review. The final search was conducted on 16 June 2024 in the Bielefeld Academic Search Engine, PubMed, and Scopus databases. Results: Of the 173 identified studies, 6 were eligible for review. In trials comparing arthrocentesis alone to arthrocentesis with NSAIDs, slight differences in joint pain were noted. For tenoxicam, differences were under 1 point on a 0–10 scale after 4 weeks, with inconsistent results. Piroxicam showed no significant difference, and pain levels were minimal in both groups. For maximum mouth opening (MMO), tenoxicam showed no significant difference. Piroxicam increased MMO by nearly 5 mm, based on one small trial with bias concerns. Conclusions: Currently, there is no strong scientific evidence supporting the injection of NSAIDs into the temporomandibular joint to relieve pain or increase jaw movement. Preliminary reports on piroxicam with arthrocentesis and tenoxicam or diclofenac without rinsing justify further research. [ABSTRACT FROM AUTHOR]

Due to tenoxicam (TX)'s poor aqueous solubility (0.072 mg/ml), it is poorly absorbable in the GIT, and the long-term oral administration of TX may cause severe GIT disturbances. Topical administration of TX can help in bypassing the GIT adverse effects. Therefore, in the present work, we constructed different pluronic/lecithin organogels (PLOs) for topical delivery of TX. PLO was constructed simply via direct mixing of an aqueous pluronic solution with lecithin solution. The prepared PLO formulations were characterized for their physicochemical properties including pH, drug content, visual inspection, viscosity, and spreadability. Also, the in vitro release and kinetic studies were carried out to investigate the mechanism of drug release. Moreover, the in vivo studies were carried out by investigating the anti-inflammatory and analgesic activities using albino male rats. The results showed that the modified PLOs have good physicochemical properties. The viscosity of the modified gels is a direct proportionality with both lecithin and pluronic concentrations. Also, subsequently, the drug release rate is directly proportional to gel viscosity. Moreover, the in vivo studies showed that the modified PLOs (F19) showed a significant (< 0.05%) paw edema inhibition and pain analgesia compared with other investigated groups. Also, the results indicated that the increase in dose is accompanied by higher activity and a longer duration of action which extended to 12 h. Introduction This study aimed to examine the effect of commonly used non-antibiotic drugs (dexamethasone and tenoxicam), on treatment of Pseudomonas aeruginosa infections, antibiotic resistance and virulence in this pathogen. Methods Four antibiotics (gentamicin, cefepime, ciprofloxacin and meropenem) were investigated. The proteolysis and hemolysis were selected as virulence factors for investigation. In this work, we selected the following final concentrations: dexamethasone (0.0052 µg/mL) and tenoxicam (2.7 µg/mL) to be used in combination with antibiotics or alone for investigation of their effects on antibiotic resistance and virulence in P. aeruginosa isolates. Results The drugs either increased or decreased antibiotic resistance in only 0-3 isolates, which indicates that the investigated drugs did not significantly affect the antibiotic resistance. Interestingly, our study demonstrated that both dexamethasone and tenoxicam increased the hemolytic activity of the investigated isolates. On the other hand, our results indicated that no overall final increasing or decreasing effect could be observed for dexamethasone on the proteolytic activity, while tenoxicam increased the proteolytic activity of the investigated isolates. Interestingly, by real-time PCR dexamethasone has shown significant down-regulation of virulence genes namely algD, plcH and toxA, apparently, in case of combination with ciprofloxacin and with gentamicin in one isolate. However, a negative influence was observed in another isolate. Unfortunately, in the case of tenoxicam the only positive effect was observed in the combination with gentamicin in one isolate. Conclusions Resistance of P. aeruginosa against gentamicin and ciprofloxacin may be affected by combining these antibiotics with dexamethasone or tenoxicam. [ABSTRACT FROM AUTHOR]

The aim was to investigate eutectic transition during tableting and storage. Mixtures of lidocaine and series of NSAIDs with increasing melting point were used as model systems to guide formulators to scaleup eutectic forming materials gaining enhanced dissolution while avoiding deleterious physical changes. Physical mixtures of NSAIDs with lidocaine were prepared at eutectic forming ratio. These were directly compressed, dry co-ground before compression, or compressed after wet granulation. Dissolution of tablets was compared to corresponding dry co-ground mixture. Thermograms of direct compressed tablet were compared to co-ground mixture and pure compound. Stability of direct compressed tablets was assessed. Tableting initiated eutexia which enhanced dissolution of NSAIDs. Eutexia was associated with tablet softening in case of low melting point ketoprofen and aceclofenac. Wet granulation hastened eutexia developing unacceptable tablet in case ketoprofen and aceclofenac. Tablets prepared by direct compression of physical mixtures underwent gradual eutectic transition upon storage with the magnitude of eutectic transition reducing with increased melting point of NSAIDs. Ketoprofen was physically unstable but aceclofenac degraded chemically as well. Tenoxicam and meloxicam tablets were physically and chemically stable. Direct compression after physical mixing is the best tableting technique, but low melting point drugs should consider different strategy before compression. [ABSTRACT FROM AUTHOR]

Background and aim: Pain after soft tissue injuries in and around the ankle is a troublesome process in terms of patient comfort and mobilization. The aim of this study was to compare the analgesic efficacy of intravenous ibuprofen and intravenous tenoxicam in patients with acute musculoskeletal pain due to ankle injury. Methods: We conducted a prospective, double-blind, randomized controlled study in a tertiary hospital. The patients were divided into two groups as those administered IV 400 mg ibuprofen and IV 20 mg tenoxicam. After the treatment of the patients, visual analog scale (VAS) scores were recorded at 15, 30, 60, and 120 min. VAS scores were compared with the effectiveness of drugs, their side effects, and the need for rescue drugs. Results: One hundred and twenty-four patients were included in the study. There were 62 patients in the tenoxicam group and 62 patients in the ibuprofen group. When VAS scores were compared, it was found that the VAS scores of the ibuprofen group were statistically significantly lower (p < 0.001). When the ΔVAS scores were compared, it was observed that the ΔVAS scores of the ibuprofen group were statistically significantly higher from 30 min (p < 0.001). There was a statistically significant difference in favor of ibuprofen between the two drug groups in terms of the need for rescue analgesics (p < 0.001). Conclusıon. The analgesic efficacy of intravenous ibuprofen and tenoxicam is equal after an ankle injury. However, after 30 min of drug administration, ibuprofen provides more effective analgesia than tenoxicam. Tenoxicam, a selective cyclooxygenase (COX)-2 inhibitor, has potent analgesic and anti-inflammatory effects and is frequently used for out-of-hospital pain control. Even though other non-steroidal anti-inflammatory drugs were incriminated in Stevens–Johnson syndrome (SJS) and toxic epidermal necrolysis (TEN) appearance, the literature is scarce regarding this agent. We report a case of tenoxicam-induced toxic epidermal necrolysis, detailing the multidisciplinary approach in a patient presenting skin detachment of 90% of the total body surface area, with concomitant ocular, oral, nasal, and vaginal mucosae involvement. A skin biopsy confirmed the diagnosis. The immediate cessation of the incriminated drug and rapid initiation of systemic steroids, along with topical therapies, and isolation into a specific environmental condition to limit skin infection were the cornerstones of therapeutic management. The patient was discharged with skin hyperpigmentation area and mild anxiety as long-term sequels. This report emphasized that severe or complicated cases should be transferred to a specialized burn center to reduce mortality risk and long-term morbidity. [ABSTRACT FROM AUTHOR]

The search for effective systems to facilitate the release of poorly bioavailable drugs is a forefront topic for the pharmaceutical market. Materials constituted by inorganic matrices and drugs represent one of the latest research strategies in the development of new drug alternatives. Our aim was to obtain hybrid nanocomposites of Tenoxicam, an insoluble nonsteroidal anti-inflammatory drug, with both layered double hydroxides (LDHs) and hydroxyapatite (HAP). The physicochemical characterization on the base of X-ray powder diffraction, SEM/EDS, DSC and FT-IR measurements was useful to verify the possible hybrids formation. In both cases, the hybrids formed, but it seemed that the drug intercalation in LDH was low and, in fact, the hybrid was not effective in improving the pharmacokinetic properties of the drug alone. On the contrary, the HAP–Tenoxicam hybrid, compared to the drug alone and to a simple physical mixture, showed an excellent improvement in wettability and solubility and a very significant increase in the release rate in all the tested biorelevant fluids. This study presents a new dual drug delivery system (DDDS) of poly(lactic acid) (PLA) with different morphologies (spheres and fibers). One approach for delivering Norfloxacin-Tenoxicam (NOR-TENO) drugs based on PLA microspheres was prepared by encapsulating TENO into PLA via emulsion-evaporation method to form TENO/microspheres then NOR was then adsorbed onto the surface of these microspheres by loading process. Another approach was designed by integrating a mixture of NOR-TENO drugs into the PLA matrix, then through the electrospinning process; NOR-TENO/electrospun fibers were fabricated. The microspheres and electrospun fibers had been characterized through FTIR, XRD, and SEM. Release behaviors of NOR-TENO were determined at pH (5.4, 6.7 and 7.4), where the release of the two drugs was faster in the two systems at pH 7.4. Antimicrobial activity of NOR-TENO/microspheres and NOR-TENO/electrospun fibers against S. aureus, B. cereus, and E. coli bacteria was examined in vitro, and NOR-TENO/electrospun fibers have better potential in killing bacteria. In vitro cytotoxicity assay of NOR-TENO/microspheres against HepG2, MCF-7, and HCT116 tumor cells were tested, and NOR-TENO/microspheres was highly cytotoxic to MCF-7 cells (IC50 = 21.87 μg・ml–1) with the lowest cytotoxic effect on WI-38 normal cells (the half maximal inhibitory concentration IC50 = 58.12 μg・ml–1). In vivo study of DMBA-induced breast cancer in rats had been accomplished for NOR-TENO/microspheres, which offer promising results in breast cancer treatment.

Purpose of the study – to study the efficacy and safety of tenoxicam (Texared®) in patients with active axial spondyloarthritis.Material and methods. The study included 35 patients with active axial spondyloarthritis with BASDAI≥4.0. Patients were given continuous oral tenoxicam (Texared®, Dr. Reddy’s Laboratories) at a dosage of 20 mg/day. Subsequently, 5 patients were excluded from the study due to discontinuation of the drug after 5–10 days of administration. 30 patients were included in the final analysis. Initially and after 30 days, to assess the severity of pain and stiffness, activity, patients filled out questionnaires in electronic form using Google forms, a general assessment of pain in the lower back and the intensity of night pain by the patient, subjective sleep characteristics were carried out. The doctor calculated the BASDAI, ASDAS-CRP, BASMI indices, and evaluated the activity according to the doctor’s opinion. The baseline blood pressure level was determined, and a patient diary was issued for ambulatory blood pressure measurement in the morning/evening for 30 days. After 30 days, the patient’s ambulatory blood pressure control was assessed. At baseline and after 30 days, biochemical blood parameters were studied, including a complete blood and urine test.Results and conclusion. In patients with axial spondyloarthritis with high and very high activity, a positive effect of tenoxicam (Texared®) therapy on disease activity was noted. The effect of Texared® develops with regular use already during the first 2 weeks, and after 4 weeks there is a clear decrease in the severity of pain in the lower back, a decrease in the duration of morning stiffness. The drug is well tolerated, has a favorable safety profile, no serious adverse events and few side effects that do not require discontinuation of therapy.

Tenoxicam (TNX) is a new non-steroidal anti-inflammatory drug that shows a superior anti-inflammatory effect and has the advantages of a long half-life period, a fast onset of action, a small dose, complete metabolism, and good tolerance. Some compounds often have tautomerism, and different tautomers exist in different crystalline forms. TNX is such a compound and has three tautomers. TNX always exists as the zwitterionic form in cocrystals. When the salt is formed, TNX exists in the enol form, which exhibits two conformations depending on whether a proton is gained or lost. Currently, the crystal structure of the keto form is not in the Cambridge Structural Database (CSD). Based on the analysis of existing crystal structures, we derived a simple rule for what form of TNX exists according to the pKa value of the cocrystal coformer (CCF) and carried out validation tests using three CCFs with different pKa values, including p-aminosalicylic acid (PAS), 3,5-dinitrobenzoic acid (DNB), and 2,6-dihydroxybenzoic acid (DHB). The molecular surface electrostatic potential (MEPS) was combined with the pKa rule to predict the interaction sites. Finally, two new cocrystals (TNX-PAS and TNX-DNB) and one salt (TNX-DHB) of TNX were obtained as expected. The differences between the cocrystals and salt were distinguished by X-ray diffraction, vibration spectra, thermal analysis, and dissolution measurements. To further understand the intermolecular interactions in these cocrystals and salt, the lattice energy and energy decomposition analysis (EDA) were used to explain them from the perspective of energy. The results suggest that the melting point of the CCF determines that of the cocrystal or salt, the solubility of the CCF itself plays an important role, and the improvement of the solubility after salt formation is not necessarily better than that of API or its cocrystals. Purpose: In this study, the preparation of self-assembled polymer-free in situ forming nanovesicles (ISNs) based on non-ionic surfactants (NISs) is presented. Methods: A 22·41 full factorial experimental design was adopted for the development of novel polymer-free ISNs loaded with tenoxicam utilizing the emulsion method. The type of NIS (Brij® 52 or Span® 60), the cholesterol percentage (30, 50, or 60 w/w%), and the internal phase percentage (20 or 30 v/v%) were chosen as the independent variables. Percentage drug released after 1 h (Q1), vesicle particle size (PS), and mean dissolution time (MDT) were the dependent variables. Selected formulation was investigated morphologically using transmission electron microscopy. Results: Results revealed that the formation had spherical dense shape. All independent factors significantly affected the percentage drug release after the first hour (Q1), and the MDT, while only the type of NIS had a significant effect on PS. The highest control of drug release was observed in formulation containing Span® 60 with lower internal phase percentage (MDT = 20.06 ± 0.40 h) as well as the smallest PS (123.75 ± 16.68 nm). Conclusion: The obtained results indicated the potentiality of the invented ISNs in controlling the release of tenoxicam in a desirable economical biphasic pattern compared to other in situ formulations. The solution mixture of the non-steroidal anti-inflammatory drugs ketoprofen, meloxicam and tenoxicam was degraded through systems, composed of different photocatalysts based on TiO2 (Fe and Cu) and the hydrogen peroxide oxidant. The monitoring was performed by UV-Vis spectroscopy. Under sunlight radiation, a reduction in peaks was observed with the use of impregnated photocatalysts. After 60 min, the sun/H2O2/Fe-TiO2 system reached degradations of 46.5% and 93.2% at 260 and 367 nm, respectively, and was selected for further studies. The degradation kinetic reached 92 and 96% of degradation after 180 min, for the λ of 260 and 367 nm, respectively. The kinetic curve could be represented by the empirical model proposed by Nichela and co-authors, indicating that besides the heterogeneous photocatalysis that occurs at the surface of the TiO2 there is also the joint effect of the photo-Fenton process. After the treatment, there was no toxicity to cress and lettuce seeds. However, a sensitivity of the thyme seeds to the compounds formed during the treatment was verified. After the fifth treatment cycle, the supported photocatalyst showed degradation higher than 82%. These results indicate that this system is suitable for the treatment of effluents containing pharmaceutical compounds.

Objective: In this study, we aimed to investigate the effect of 2 different dosages of tenoxicam in the prevention of propofol injection pain. Methods: A total of 120 patients between the ages of 20-50 years who were scheduled for elective surgery were included in this prospective. Patients were randomly divided into 3 groups. Group 1 received 5 mL saline, group 2 received 10 mg tenoxicam in 5 mL saline, and group 3 received 20 mg tenoxicam in 5 mL saline intravenously as a pretreatment. Venous occlusion was applied for 60 seconds with a rubber tourniquet after the injection was completed. After injecting propofol, the pain at the injection site of the patient was questioned according to the Verbal Rating Scale. Results: The overall pain incidence during propofol injection was 85% in group 1, 75% in group 2, and 60% in group 3 (P = .039). While there was no significant difference between groups 1 and 2 (P = .264), there was a significant difference between groups 1 and 3 (P = .012). Moreover, there was a significant decrease in the level of severe pain in group 3 compared to group 1 (P = .008). There was no significant difference between the groups in terms of mild and moderate pain levels (P > .05). Conclusions: We found that 20 mg of tenoxicam pretreatment was effective in reducing the incidence and severity of propofol injection pain compared to the control saline group, but the 10 mg dose did not significantly reduce the injection pain. Transdermal delivery of non-steroidal anti-inflammatory drugs (NSAIDs) is an effective route of drug administration, as it directs the drug to the inflamed site with reduced incidence of systemic adverse effects such as gastric hemorrhage and ulcers. Tenoxicam (TNX) is a member of NSAIDs that are marketed only as oral tablets due to very poor absorption through the skin. The current study intended to formulate and characterize a hydrogel loaded with nanostructured lipid carriers (NLCs) to enhance the transdermal delivery of TNX. Six formulations of TNX were formulated by slight modifications of high shear homogenization and ultrasonication method. The selected formula was characterized for their particle size, polydispersity index (PDI), zeta potential, entrapment efficiency (EE), in-vitro drug release and ex-vivo skin permeation studies. Moreover, the effectiveness of the developed formula was studied in-vivo using carrageenan-induced paw edema and hyperalgesia model in irradiated rats. Formula F4 was chosen from six formulations, as the average diameter was 679.4 ± 51.3 nm, PDI value of about 0.02, zeta potential of −4.24 mV, EE of 92.36%, globules nanoparticles without aggregations and absence of interactions in the developed formula. Additionally, the in-vivo study showed the efficacy of formula F4 (TNX-NLCs hydrogel) equivalent to oral TNX in reducing the exaggerated inflammatory response induced by carrageenan after irradiation. In conclusion, the present findings suggest that TNX-NLCs hydrogel could be a potential transdermal drug delivery system alternative to the oral formulation for the treatment of various inflammatory conditions.

Objective: This investigation aimed to explore the potential of non-ionic surfactant based niosomal vesicles encapsulating tenoxicam (TN; anti-rheumatic drug) for the treatment of rheumatic diseases., Material and Methods: Mechanical dispersion technique with controlled pressure was employed to prepare different niosomal formulations. The effects of different ratios of surfactant (span-60), lipid, and sodium deoxycholate on noisome's physicochemical properties have been examined. Moreover, inhibition of TNF-α in lipopolysaccharide-activated cultured Human leukemia monocytic (THP-1) cells were demonstrated to assess the in vitro inflammation profile. Finally, the optimized niosomal formulation (TN3) was prepared in gel matrix consist of carbopol 934 (termed as TN34) and stability was also tested at 4±2 ̊C, 25±2 ̊C, 37±2 ̊C and 45±2 ̊C for 6 months., Results: The optimized niosomal formulation exhibited a small vesicle size (165±14nm) and high drug encapsulation (79.64±1.5%). Niosomal gel formulation TN34 showed pH (6.7), viscosity (6810±3.34 cps), spreadability (19.11±1.87gm.cm/sec) and also displayed sustained release pattern of drug release (98.16±0.07% TN released from gel matrix in 24h) in vitro release study. TN34 exhibited substantial anti-inflammatory response, with ∼75% inhibition of TNF-α in 48h. For the first time ever, useful fluorescent (FL) carbonaceous materials (CMTS) were isolated from incense ash using facile procedure on two steps; dispersion of the CMTS in water followed by filtration. The CMTS were characterized using the following techniques; dynamic light scattering (DLS), transmission electron microscopy (TEM) and Fourier transform infrared (FT-IR) spectroscopy. The CMTS exhibit excitation wavelength dependent fluorescence emission, so it can be used as a FL probe. The FL probe was employed for sensing and quantitative determination of two members of oxicam family (tenoxicam (TEN) and meloxicam (MEL)) that belongs to non-steroidal anti-inflammatory drugs (NSAIDs). The method is based on the quenching of the FL intensity of the isolated CMTS by inner filter effect mechanism (IFE). The FL intensity decreases in linear relationship with increasing the concentrations of the two cited drugs within the range of 4.0 – 30.0 µg/mL with mean percentage recoveries of 100.04 ± 0.95 and 100.07 ± 1.06 with detection limits of 1.31 µg/mL and 1.06 µg/mL for TEN and MEL, respectively. Finally, the developed sensing system was validated as per ICH guidelines and it was proved to be accurate and precise and applied successfully for quantitative determination of the two cited drugs in their capsule dosage forms with excellent percentage recoveries reaching to 97.66 ± 0.39and 98.19 ± 1.12 for TEN and MEL, respectively. Tenoxicam (TNX) is a new non-steroidal anti-inflammatory drug that shows a superior anti-inflammatory effect and has the advantages of a long half-life period, a fast onset of action, a small dose, complete metabolism, and good tolerance. Some compounds often have tautomerism, and different tautomers exist in different crystalline forms. TNX is such a compound and has three tautomers. TNX always exists as the zwitterionic form in cocrystals. When the salt is formed, TNX exists in the enol form, which exhibits two conformations depending on whether a proton is gained or lost. Currently, the crystal structure of the keto form is not in the Cambridge Structural Database (CSD). Based on the analysis of existing crystal structures, we derived a simple rule for what form of TNX exists according to the pKa value of the cocrystal coformer (CCF) and carried out validation tests using three CCFs with different pKa values, including p-aminosalicylic acid (PAS), 3,5-dinitrobenzoic acid (DNB), and 2,6-dihydroxybenzoic acid (DHB). The molecular surface electrostatic potential (MEPS) was combined with the pKa rule to predict the interaction sites. Finally, two new cocrystals (TNX-PAS and TNX-DNB) and one salt (TNX-DHB) of TNX were obtained as expected. The differences between the cocrystals and salt were distinguished by X-ray diffraction, vibration spectra, thermal analysis, and dissolution measurements. To further understand the intermolecular interactions in these cocrystals and salt, the lattice energy and energy decomposition analysis (EDA) were used to explain them from the perspective of energy. The results suggest that the melting point of the CCF determines that of the cocrystal or salt, the solubility of the CCF itself plays an important role, and the improvement of the solubility after salt formation is not necessarily better than that of API or its cocrystals.

A nanohybrid formulation of silver‑titanium dioxide nanoparticles/poly(lactic acid) (Ag-TiO 2 /PLA) was designed for Norfloxacin/Tenoxicam (NOR/TENO) targeted delivery to maximize the bioavailability and minimize the side effects of the drugs. Ag-TiO 2 nanoparticles were prepared via Stober method. NOR, TENO and a mixture of NOR/TENO (NT) were loaded onto Ag-TiO 2 nanoparticles and coated by PLA via solution casting. The physical interaction between the drugs and carrier was confirmed by Fourier-transform infrared (FTIR) analysis. X-ray diffraction (XRD) demonstrated that Ag-TiO 2 consists of a cubic phase of Ag with two phases of TiO 2 (anatase and brookite). Ag nanoparticle fine spots coated with TiO 2 were collected to form spheres averaging at 100 nm in size. In-vitro release behavior of drugs was studied at different pH (5.4, 7.4) and the release of drug from NT/Ag-TiO 2 /PLA was faster at pH 7.4. Gram-positive and Gram-negative bacteria were used to investigate antibacterial properties of the nanohybrid. Cytotoxicity of the nanohybrid using an MTT assay was studied against different tumor and normal cell lines. It was found that NT/Ag-TiO 2 /PLA has an excellent cytotoxic effect against various bacterial cells and tumor cell lines. Polymorphism is a relatively common phenomenon among pharmaceutical compounds, and one of the main aspects to be considered in the production and development of medications. The investigation of polymorphism associated with oxicams, a group belonging to the class of non-steroidal anti-inflammatory drugs (NSAIDs) has increased in recent years and, in the case of tenoxicam, the existence of four polymorphic forms is reported in the literature. The objective of this study was to characterize the presence of different polymorphic forms of tenoxicam in active pharmaceutical ingredient and oral pharmaceutical formulations, as well as to evaluate the influence on in vitro dissolution. The characterization of the three samples of pharmaceutical ingredient of tenoxicam from different suppliers by X-Ray Diffraction (XRD), Infrared (IR) and dissolution profile indicated the presence of a form III crystalline structure, without presenting significant differences between the in vitro dissolution profiles analyzed, and a Dissolution Efficiency (DE%) of 60.30%, 60.70% and 72.34%, respectively. When the four pharmaceutical specialties of tenoxicam were submitted to XRD analysis, they also presented form III crystalline structures. Despite this, the formulations presented different dissolution profiles and a DE% of 75.23%, 83.69%, 78.19% and 90.63%, respectively, without compromising their quality. However, often polymorphism affects physico-chemical properties of drugs, showing the importance of studying this phenomenon, by correlating the presence of crystalline structures to alterations in the quality of active ingredients and pharmaceutical products.

Aim: The objective of this study is comparing the effects of intra-articular hyaluronic acid (HA) and tenoxicam aplications on joint range of motion (ROM) and pain in patients with gonarthrosis.Patients and Method: This study was designed as prospective, non-randomized controlled, and single-blinded trial. 60 patients diagnosed with Grade II and Grade III osteoarthritis (OA) according to the American College of Rheumatology (ACR) criteria were consecutively enrolled. Patients were divided into two groups whereby the first group received intra-articular HA three times a week and the other group received a single dose intra-articular tenoxicam. Pre-treatment and after-treatment VAS and ROM values of the groups were compared.Results: In the HA Group, 20 patients (66.6%) were female, 10 patients (33.3%) were male and the mean age of them was 59.6 years (range 45-75 years). In the Tenoxicam Group, 24 patients (80%) were female, 6 (20%) patients were male, and the average age of this group was 61.5 years (range 45-75 years). VAS scores knee flexion and extension ROM values on the 1st day, the 15th day, the 1st month,the2nd month, and the 3rd month after the treatment, significantly improved in both groups (p 0.05). Conclusion: It was seen that intra-articular HA and tenoxicam administration seem to be effective and reliable methods in Grade 2-3 knee OA patients; but tenoxicam was more effective in reducing pain and relieving joint movements compared to HA injection in the early periods.

Nonsteroidal anti-inflammatory drugs (NSAIDs) are one of the most important therapeutic agents used for the treatment of a variety of inflammation. Tenoxicam (TNX), which is used to relieve inflammation, swelling, and pain associated with rheumatoid arthritis, is a member of NSAIDs. In this study, copper(II), zinc(II), platinum(II) and palladium(II) complexes of TNX were synthesized and characterized by analytical and instrumental techniques (Ultraviolet and visible absorption spectra (UV–Vis), Liquid chromatography–mass spectrometry (LC-MS), Inductively coupled plasma - optical emission spectrometry (ICP-OES), Differential thermal analysis-Thermogravimetric analysis (DTA-TG)). Fourier-transform infrared (FT-IR) spectra of the complexes were measured and the outcome were supported by density functional theory computations. The proposed structure of these metal complexes are [Cu(TNX)2], [Zn(TNX)2], [Pt(TNX)2] and [Pd(TNX)Cl2]. The anti-inflammatory activities of the synthesized complexes, compared with the drug, were demonstrated for the first time. All metal complexes of the TNX acted more efficiently on the reduction of pro-inflammatory tumor necrosis factor (TNF)-alpha production than that of TNX. The highest anti-inflammatory potential was detected with [Cu(TNX)2]. The receptor-ligand interactions between TNX complexes and TNF-α were revealed by molecular docking calculations.

Transdermal delivery of non-steroidal anti-inflammatory drugs (NSAIDs) is an effective route of drug administration, as it directs the drug to the inflamed site with reduced incidence of systemic adverse effects such as gastric hemorrhage and ulcers. Tenoxicam (TNX) is a member of NSAIDs that are marketed only as oral tablets due to very poor absorption through the skin. The current study intended to formulate and characterize a hydrogel loaded with nanostructured lipid carriers (NLCs) to enhance the transdermal delivery of TNX. Six formulations of TNX were formulated by slight modifications of high shear homogenization and ultrasonication method. The selected formula was characterized for their particle size, polydispersity index (PDI), zeta potential, entrapment efficiency (EE), in-vitro drug release and ex-vivo skin permeation studies. Moreover, the effectiveness of the developed formula was studied in-vivo using carrageenan-induced paw edema and hyperalgesia model in irradiated rats. Formula F4 was chosen from six formulations, as the average diameter was 679.4 ± 51.3 nm, PDI value of about 0.02, zeta potential of −4.24 mV, EE of 92.36%, globules nanoparticles without aggregations and absence of interactions in the developed formula. Additionally, the in-vivo study showed the efficacy of formula F4 (TNX-NLCs hydrogel) equivalent to oral TNX in reducing the exaggerated inflammatory response induced by carrageenan after irradiation. In conclusion, the present findings suggest that TNX-NLCs hydrogel could be a potential transdermal drug delivery system alternative to the oral formulation for the treatment of various inflammatory conditions. Pseudomonas aeruginosa is an opportunistic pathogen exhibiting higher resistance to commonly used antibiotics. Microbial resistance to antibiotics is a major problem that hinders attempts to control microbial infections. Quorum sensing inhibitors could help us solve such problem by repressing quorum sensing that controls the production of virulence factors in many pathogens including P. aeruginosa. In this study, the influence of tenoxicam, a non-steroidal anti-inflammatory drug, on quorum sensing in P. aeruginosa was characterized. Treatment of P. aeruginosa with tenoxicam decreased production of many virulence factors such as pyoverdin, rhamnolipids, pyocyanin, elastase, proteases, and hemolysins. Moreover, qRT-PCR revealed a significant reduction in expression of quorum sensing genes in tenoxicam-treated P. aeruginosa in comparison with untreated bacteria. Tenoxicam markedly reduced the capacity of P. aeruginosa to kill mice infection model. Mice injected with tenoxicam-treated P. aeruginosa exhibited higher survival rate as compared with those inoculated with untreated bacteria. Current data clearly demonstrate that tenoxicam has quorum sensing inhibitory effect on P. aeruginosa. Tenoxicam could play a role in reduction of Pseudomonas quorum sensing-dependant virulence factors production, and therefore affect its pathogenesis in the host. In summary, the current study suggests that tenoxicam could be used as adjuvant to antibiotics in the management of diseases caused by P. aeruginosa. [ABSTRACT FROM AUTHOR]

Synthesis of photoluminescence carbon dots from red beetroot and utilizing its extract for spectrofluorimetric determination of tenoxicam. Tenoxicam (TNX) belongs to non-steroidal drugs that effectively treat many inflammatory disorders. In this study, luminescent carbon dots were synthesized from red beetroot as a green fluorescence probe. The synthesized carbon dots' morphology, size, and spectroscopic features were investigated and characterized by spectrophotometric, FTIR, SEM, spectrofluorimetric, and TEM techniques. The synthesized carbon dots possessed an emission at 430 nm and an excitation at 366.5 nm. Meanwhile, TNX is a yellow-colored compound with a spectrophotometric absorbance peak at 370 nm. Based on the inner filter effect mechanism, the emission fluorescence intensity of the synthesized carbon dots has been markedly quenched upon the addition of TNX. In addition, the quenching in the fluorescence intensity was quantitively related to the concentration of TNX at the linear range of 0.25 - 2.5 µg/mL (r2 = 0.9989) with a LOD of 0.08 µg/mL and LOQ of 0.24 µg/mL. The second side of this study is based on enhancing the fluorescence intensity of the acetonitrile extract of red beetroot upon the addition of TNX solution. This enhancement was recorded at a fluorescence emission of 357 nm with a fluorescence excitation of 305.5 nm. In addition, the enhancement in the fluorescence intensity was quantitively related to the concentration of TNX at the linear range of 0.5 - 8.0 µg/mL (r2 = 0.9995) with a LOD of 0.16 µg/mL and LOQ of 0.49 µg/mL. The analytical conditions were well-optimized, and the developed method was validated according to ICH guidelines. Finally, the proposed spectrofluorimetric approaches were applied to determine TNX in vials, tablets, and capsules with good recovery and accepted statistical results. [ABSTRACT FROM AUTHOR]

The first-line treatment of osteoarthritis is based on anti-inflammatory drugs, the most currently used being nonsteroidal anti-inflammatory drugs, selective cyclooxygenase 2 (COX-2) inhibitors and corticoids. Most of them present cytotoxicity and low bioavailability in physiological conditions, making necessary the administration of high drug concentrations causing several side effects. The goal of this work was to encapsulate three hydrophobic anti-inflammatory drugs of different natures (celecoxib, tenoxicam and dexamethasone) into core-shell terpolymer nanoparticles with potential applications in osteoarthritis. Nanoparticles presented hydrodynamic diameters between 110 and 130 nm and almost neutral surface charges (between −1 and −5 mV). Encapsulation efficiencies were highly dependent on the loaded drug and its water solubility, having higher values for celecoxib (39–72%) followed by tenoxicam (20–24%) and dexamethasone (14–26%). Nanoencapsulation reduced celecoxib and dexamethasone cytotoxicity in human articular chondrocytes and murine RAW264.7 macrophages. Moreover, the three loaded systems did not show cytotoxic effects in a wide range of concentrations. Celecoxib and dexamethasone-loaded nanoparticles reduced the release of different inflammatory mediators (NO, TNF-α, IL-1β, IL-6, PGE2 and IL-10) by lipopolysaccharide (LPS)-stimulated RAW264.7. Tenoxicam-loaded nanoparticles reduced NO and PGE2 production, although an overexpression of IL-1β, IL-6 and IL-10 was observed. Finally, all nanoparticles proved to be biocompatible in a subcutaneous injection model in rats. These findings suggest that these loaded nanoparticles could be suitable candidates for the treatment of inflammatory processes associated with osteoarthritis due to their demonstrated in vitro activity as regulators of inflammatory mediator production.

Objectives: The electro-oxidation behavior of the non-steroidal anti-inflammatory drug tenoxicam (TX) was studied on multiwalled carbon nanotube (MWCNT)-modified glassy carbon electrode (GCE) by cyclic voltammetry, differential pulse voltammetry (DPV), and square wave voltammetry (SWV). Materials and Methods: The GCE was modified with MWCNT for sensitive determination of TX by voltammetric methods. Results: The current peaks for TX occurred at around 0.520 V for DPV and 0.570 V for SWV when the potential was scanned in the positive direction. The oxidation process of TX showed irreversible and diffusion-controlled behavior. The linear responses were obtained in the range from 2×10-7 to 1×10-5 M with the limit of detection (LOD) 1.43×10-9 for DPV and from 8×10-9 to 8×10-6 with the LOD 9.97×10-10 for SWV in 1 M acetate buffer solution at pH 5.5. Conclusion: Fully validated DPV and SWV were successfully applied for the determination of TX from pharmaceutical dosage form and yielded satisfying results. [ABSTRACT FROM AUTHOR]

ABSTRACTBACKGROUND AND OBJECTIVES:Tenoxicam is widely used in osteoarthritis treatment and we aimedto compare the effectivity of oral and intra-articular administration of tenoxicam in osteoarthri-tis treatment.METHODS: This study was performed between 2011 and 2012 by retrospectively analyzing andcomparing the findings of 60 patients who were clinically and radiologically diagnosed with kneedegenerative osteoarthritis in Bünyan state hospital pain policlinic. 60 patients included in thestudy were divided into two groups. The first group (tenoxicam IA, n = 30) included patientfindings of those subjected to intra-articular injection of 20 mg tenoxicam to the knee oncea week for three weeks and the second group (oral tenoxicam, n = 30) included patients whowere administered 20 mg oral tenoxicam once a day for three weeks. All patients were clini-cally evaluated pre-treatment and in the 1st week, 1st month and 3rd month post-treatmentaccording to specified criteria.RESULTS AND CONCLUSIONS: Twenty two of 60 patients included in the study were male and 38were female. In both groups significant improvements were detected in all of the observedparameters: visual analog scale, Western Ontario McMaster Osteoarthritis Index (pain, physicalactivity, knee stiffness) and Lequesne index scores and in the evaluations performed in 1st week,1st month and 3rd month with respect to pre-treatment values. Besides, a better complianceto treatment and gastrointestinal system tolerability in tenoxicam IA group was also observed.Intra-articular tenoxicam administration could be thought as an alternative treatment methodin patients with knee osteoarthritis who cannot use oral tenoxicam especially due to systemicgastrointestinal system side effects and those who have difficulties in adapting to treatment.
