Background The prevalence of diabetes and cardiovascular diseases in China has been increasing annually, and it has been demonstrated that diabetes can exacerbate the adverse effects of cardiovascular diseases through nutritional and inflammatory pathways. The prognostic nutritional index (PNI) is a marker of immunonutrition that reflects the inflammation, immune status and nutritional status of an individual. Due to its advantages of simplicity, rapidity, accessibility, and reliability, research on PNI has been increasing, yet its role in cardiovascular diseases has been less explored. Objective To investigate the relationship between PNI and major in-hospital adverse cardiovascular events (MACE) in patients with acute ST-elevation myocardial infarction (STEMI) complicated by type 2 diabetes mellitus (T2DM) following percutaneous coronary intervention (PCI) . Methods A retrospective analysis of 1 053 STEMI patients with T2DM who underwent PCI at the First Affiliated Hospital of Xinjiang Medical University from January 2015 to June 2023 was conducted. The patients were divided into the MACE (n=177) and non-MACE (n=876) groups according to the occurrence of MACE during hospitalization, and further categorized into the high PNI (n=686) and low PNI (n=367) groups according based on the cutoff value of PNI to predict the occurrence of in-hospital MACE after PCI in patients with STEMI and T2DM. Univariate and multivariate Logistic regression analyses identified factors influencing in-hospital MACE after PCI in patients with STEMI and T2DM. Receiver operating characteristic (ROC) curves were plotted to evaluate the predictive value of PNI for in-hospital MACE, and the area under the curve (AUC) was calculated. Pearson correlation analysis or Spearman rank correlation analysis was used to explore the correlation between PNI and cardiovascular disease risk factors. Results The differences between the MACE and non-MACE groups in gender, age, history of hypertension, serum glucose, Killip classification, lactate dehydrogenase, heart rate, myoglobin, creatine kinase-MB isoenzyme (CK-MB), white blood cell count, brain natriuretic peptide (BNP), PNI, Global Registry of Acute Coronary Events (GRACE) score, hemoglobin, platelet count, lymphocyte count, albumin, total protein, creatinine, urea nitrogen and triacylglycerol were statistically significant (P

The accurate diagnosis and treatment of gliomas depends largely on the understanding of the P53 gene status. In our study, we presented a robust deep learning model, CTD-RegNet (improved RegNet integrating CNN, vision transformer, and truth discovery), tailored for predicting P53 gene status in gliomas. Our model addressed common challenges of existing deep learning models, such as incomplete feature extraction and uncertainty. First, the model used the RegNet network as a basis for predicting P53 gene mutations by skillfully extracting heterogeneous features. Next, the RegNet network was enhanced by integrating the CNN and ViT modules to optimise feature extraction and computational efficiency. Finally, using the truth discovery algorithm, we iteratively refined model uncertainties, thereby improving prediction accuracy. Our experiments demonstrated the effectiveness of the CTD-RegNet model, achieving an impressive accuracy of 95.57% and an AUC score of 0.9789, outperforming existing P53 gene status prediction models. The non-invasive nature of our model minimised the economic burden and physical and psychological stress on patients, while providing critical insights for accurate clinical diagnosis and treatment of gliomas.

The sustainability and additive manufacturing of dielectric insulators are the development direction of the power system. Introducing dynamic covalent bonds in light-based 3D printing have attracted considerable attention as the reversible crosslinks allow for the reprocessing of printed objects. However, there generally exists a trade-off between mechanical strength, glass transition temperature (Tg), and reconfigurability for dynamic covalent networks. The reconfiguring process of the dynamic covalent network often requires high mobility of molecular chains and large free volumes, which in turn decreases the mechanical strength, Tg, and electrical insulating performance. Herein, we demonstrate a novel strategy for developing a kind of mechanically robust and sustainable vitrimer by building a rigid-flexible coupling inter-penetration network (IPN). Specifically, a two-stage curing approach was used to prepare high-performance 3D-printing vitrimers by using the plant oil-epoxy hybrid resin, which brings a lot of ester bonds and β-hydroxyl ester for the crosslinking network. Computational techniques with molecular dynamics calculation are used for the design and optimization of the crosslinking network, and then the optimized IPN is prepared by digital light processing 3D printing and subsequent heat curing. In the IPN, the epoxy backbone is rigid to enhance the Tg and tensile strength, while the plant-based methacrylate is flexible to guarantee topological rearrangement at elevated temperatures. Compared to reported epoxy vitrimers, the resultant IPN exhibits simultaneous high Tg (111 °C), outstanding tensile strength and toughness (tensile strength of 70 MPa, elongation at break of 17.58 %), good topological rearrangement, and excellent dielectric properties (permittivity less than 4, breakdown strength of 49.3 kV/mm). This work provides a new strategy for balancing the strength, toughness, electrical insulating and sustainability of 3D-printed thermosets.

The sense of taste is an important sentinel governing what should or should not be ingested by an animal, with high pH sensation playing a critical role in food selection. Here we explore the molecular identities of taste receptors detecting the basic pH of food using Drosophila melanogaster as a model. We identify a chloride channel named alkaliphile (Alka), which is both necessary and sufficient for aversive taste responses to basic food. Alka forms a high-pH-gated chloride channel and is specifically expressed in a subset of gustatory receptor neurons (GRNs). Optogenetic activation of alka-expressing GRNs is sufficient to suppress attractive feeding responses to sucrose. Conversely, inactivation of these GRNs causes severe impairments in the aversion to high pH. Altogether, our discovery of Alka as an alkaline taste receptor lays the groundwork for future research on alkaline taste sensation in other animals.

Abstract Background Identification of micropapillary and solid subtypes components in small-sized (≤ 2 cm) lung adenocarcinoma plays a crucial role in determining optimal surgical procedures. This study aims to propose a straightforward prediction method utilizing preoperative available indicators. Methods From January 2019 to July 2022, 341 consecutive patients with small-sized lung adenocarcinoma who underwent curative resection in thoracic surgery department of Xuanwu Hospital, Capital Medical University were retrospectively analyzed. The patients were divided into two groups based on whether solid or micropapillary components ≥ 5% or not (S/MP5+ and S/MP5-). Univariate analysis and multivariate logistic regression analysis were utilized to identify independent predictors of S/MP5+. Then a nomogram was constructed to intuitively show the results. Finally, the calibration curve with a 1000 bootstrap resampling and the receiver operating characteristic (ROC) curve were depicted to evaluate its performance. Results According to postoperative pathological results, 79 (23.2%) patients were confirmed as S/MP5+ while 262 (76.8%) patients were S/MP5-. [Introduction] To ensure the safety, reliability and economy of power supply, the paper evaluates the power consumption meteorological risk of Yinchuan City from 2014 to 2022. [Method] A power consumption meteorological risk assessment model was established by risk index, sensitivity index and vulnerability index, and the correlation analysis, percentile method, normalization and other methods based on meteorological data from automatic stations and electricity load data. [Result] The results show that the peak of electricity load appeared in heating period and transition period (January, March, October to December) and Summer (June to July), and the lower values in April and September. The results of correlation analysis show that the higher temperature, the greater the electrical load. High temperature intensity, high temperature frequency, daily electricity consumption, power supply load, population density and community power supply facilities are selected as indicators of power consumption by the correlation analysis and the expert questionnaire survey, and the weights are 0.5, 0.5, 0.98,

Insights into the interdisciplinary response to scientific collaboration remain scarce in interdisciplinary fields. The present work focuses on the effect of scientific collaboration on disciplinary diversity in the field of climate change using multiple statistical methods. The results show that research collaboration at the author and country/region levels has significantly positive effects on variety and DIV (an integrated diversity indicator), although these effects are limited. Additionally, the associations between the values of variety, disparity, and DIV and the number of departments are significantly positive, and similar results are found regarding relationships between these indicators and the number of countries/regions at the country/region and integrated levels. However, scientific collaboration has a negative effect on the balance of references at all levels. This study can improve our understanding of how scientific collaboration affects the multidimensional aspects of interdisciplinary research, and facilitate cross-disciplinary collaboration in the future.

Anthomyiidae is a cosmopolitan and diverse family of Calyptratae, and is routinely considered to play key roles in both ecology and agriculture. The higher-level phylogenetic classification of Anthomyiidae has been highly controversial, necessitating further molecular data for precise reconstruction of phylogenetic relationships. In this study, we successfully acquired and annotated 18 new mitogenomes of anthomyiids. Moreover, the mitogenomes of the following genera Eustalomyia, Hyporites, Leucophora, Emmesomyia and Eutrichota are reported for the first time. The 18 mitogenomes are compared with confamilial species to assess genetic variation and to better understand evolutionary relationships within the family Anthomyiidae. In comparisons among 13 mitochondrial protein coding genes (PCG), the calculation of evolutionary rate exhibited nad1 as the fastest evolving gene in Anthomyiidae. Among the anthomyiids investigated, cox2 and nad4 had the lowest genetic distance across the 13 PCGs, suggesting a high degree of conservation for these two genes. Herein, we conducted phylogenetic analyses of the newly sequenced mitogenomes along with 11 known anthomyiids to investigate the interrelationships of Anthomyiidae. Our results indicate that Anthomyiidae is a monophyletic lineage and sister group to Scathophagidae, confirming prior findings based on morphological and molecular analyses. We recovered two subfamilies as monophyletic (Myopininae, Pegomyinae) while Anthomyiinae was polyphyletic. The great species diversity of anthomyiid flies limits the availability of mitogenomes for accurately resolving the phylogeny of Anthomyiidae. Nonetheless, our study provides novel insight into the molecular taxonomy, evolution, and phylogeny of the family Anthomyiidae.

Abstract Patients with parathyroid carcinoma (PC) are often diagnosed postoperatively, due to incomplete resection during the initial surgery, resulting in poor outcomes. The aim of our study was to investigate the pre-surgery indicators of PC and try to develop a predictive model for PC utilizing machine learning. Evaluation of pre-surgery neuropsychological function and confirmation of pathology were carried out in 133 patients with primary hyperparathyroidism in Beijing Chaoyang Hospital from December 2019 to January 2023. Patients were randomly divided into a training cohort (n = 93) and a validating cohort (n = 40). Analysis of the clinical dataset, two machine learning including the extreme gradient boosting (XGBoost) and the least absolute shrinkage and selection operator (LASSO) regression were utilized to develop the prediction model for PC. Logistic regression analysis was also conducted for comparison. Significant differences in elevated parathyroid hormone and decreased serum phosphorus in PC compared to (BP). Abstract Background There is a substantially increasing need for general practitioners (GPs) for future unpredictable pandemic crises, especially at the community-based health services (CBHS) level to protect the vast and varied grassroot-level population in China. Thus, it is crucial to understand the factors that affect Chinese medical students’ GP career choices and commitments to CBHS. Methods Leveraging the self-administered data collected across the country, this study conducted logistic regressions with 3,438 medical students. First, descriptive statistics of outcome variables and independent variables were provided. Then, stepwise logistic regression models were built, starting from adding individual characteristics, and then familial and institutional characteristics. Last, post-estimation was conducted to further assess whether there were significant marginal effects. Results Results showed that women students were 24% less likely to choose GP careers but were 1.25 times more likely to commit to CBHS than their men peers, holding other individual, familial, and institutional characteristics constant. In addition, students who major in GP-orientated were more likely to choose GP careers and commit to CBHS, respectively, than those who major in clinical medicine. Furthermore, familial characteristics like annual income and mother’s educational level only significantly predicted commitments to CBHS. Notably, sex-related differences in GP career choices and commitments to CBHS – by different regions – were observed. Conclusions Understanding the factors that affect medical students’ GP career choices sheds light on how medical education stakeholders can make informed decisions on attracting more medical students to GP-orientated majors, which in turn cultivates more GP professionals to meet the nation’s demand for GPs. In addition, by understanding the factors that influence medical students’ commitment to CBHS, policymakers could make beneficial policies to increase medical students’ motivations to the grassroot-level health institutions, and devote to CBHS as gatekeepers for a large population of residents’ health.

Extracting water bodies from synthetic aperture radar (SAR) images plays a crucial role in the management of water resources, flood monitoring, and other applications. Recently, transformer-based models have been extensively utilized in the remote sensing domain. However, due to regular patch-partition and weak inductive bias, transformer-based models face challenges such as edge serration and high data dependency when used for water body extraction from SAR images. To address these challenges, we introduce a new model, the Superpixel-based Transformer (SPT), based on the adaptive characteristic of superpixels and knowledge constraints of the adjacency matrix. (1) To mitigate edge serration, the SPT replaces regular patch partition with superpixel segmentation to fully utilize the internal homogeneity of superpixels. (2) To reduce data dependency, the SPT incorporates a normalized adjacency matrix between superpixels into the Multi-Layer Perceptron (MLP) to impose knowledge constraints. (3) Additionally, to integrate superpixel-level learning from the SPT with pixel-level learning from the CNN, we combine these two deep networks to form SPT-UNet for water body extraction. The results show that our SPT-UNet is competitive compared with other state-of-the-art extraction models, both in terms of quantitative metrics and visual effects.

To explore the effects of the introduction order of calcium sources and the bacteria-to-calcium ratio on the microbially induced calcium carbonate precipitation (MICP) product CaCO3 and to achieve the regulation of CaCO3 crystal morphology, the mineralisation products of MICP were compared after combining bacteria and calcium at ratios of 1/9, 2/9, 3/9, 4/9, 5/9, and 6/9. A bacterial solution was combined with a urea solution in two calcium addition modes: calcium-first and calcium-later modes. Finally, under the calcium-first addition method, the output of high-purity vaterite-type CaCO3 was achieved at bacteria-to-calcium ratios of 2/9 and 3/9; under the calcium-later addition method, the output of calcite-type CaCO3 could be stabilised, and the change in the bacteria-to-calcium ratio did not have much effect on its crystalline shape.

Aedes aegypti (Ae. aegypti) is a major vector of dengue virus (DENV) and Zika virus (ZIKV). Understanding the complex interaction mechanisms between mosquito vectors and arboviruses is essential to interrupt virus transmission. This study constructed CYP4C21 knockout (KO) Aag2 cells (Ae. aegypti cells) and confirmed that CYP4C21 KO reduced DENV2 and ZIKV copies in Aag2 cells, which suggests that CYP4C21 may play an important role in mosquito infection with arboviruses. Furthermore, it is the first report of the CYP4 family related to viral infection, which lays the foundation for exploring the role of the CYP4C21 in the interaction of Ae. aegypti and arbovirus and provides novel insights into the function of cytochrome family proteins.

We investigate the properties of non-Gaussian quantum states generated by a three-photon quantum scissor (3-QS). Three different types of inputs to the 3-QS are considered: coherent state (CS), squeezed vacuum state (SVS), and optical field thermal state (TS). We discuss both the detection probability and fidelity of the output states. Our results demonstrate that when the input states have a smaller mean photon number, the output non-Gaussian state can exhibit high detection probability and high fidelity by selecting an appropriate transmissivity. Additionally, we calculate the Wigner functions for the various output states and analyze the non-classical properties by comparing the negative volume of the Wigner functions. The results indicate that the output non-Gaussian states display significant non-classical behavior, particularly when the transmissivity is high, with SVS as the input state showing the strongest effect. Moreover, we compare the performance of the 3-QS with that of the two-photon quantum scissors (2-QS). The findings demonstrate that the non-Gaussian states prepared by the 3-QS possess stronger non-classical characteristics. These results have potential implications for advancing the practical application of the 3-QS in quantum information technology.

Abstract Background Accurately evaluating the lymph node status preoperatively is critical in determining the appropriate treatment plan for non-small-cell lung cancer (NSCLC) patients. This study aimed to construct a novel nomogram to predict the probability of lymph node metastasis in clinical T1 stage patients based on non-invasive and easily accessible indicators. Methods From October 2019 to June 2022, the data of 84 consecutive cT1 NSCLC patients who had undergone PET/CT examination within 30 days before surgery were retrospectively collected. Univariate and multivariate logistic regression analyses were performed to identify the risk factors of lymph node metastasis. A nomogram based on these predictors was constructed. The area under the receiver operating characteristic (ROC) curve and the calibration curve was used for assessment. Besides, the model was confirmed by bootstrap resampling. Results Four predictors (tumor SUVmax value, lymph node SUVmax value, consolidation tumor ratio and platelet to lymphocyte ratio) were identified and entered into the nomogram. The model indicated certain discrimination, with an area under ROC curve of 0.921(95%CI 0.866–0.977). The calibration curve showed good concordance between the predicted and actual possibility of lymph node metastasis. Conclusions This nomogram was practical and effective in predicting lymph node metastasis for patients with cT1 NSCLC. It could provide treatment recommendations to clinicians.

E-20-monooxygenase (E20MO) is an enzymatic product of the shade (shd) locus (cytochrome p450, E20MO). Initially discovered in Drosophila, E20MO facilitates the conversion of ecdysone (E) into 20-hydroxyecdysone (20E) and is crucial for oogenesis. Prior research has implicated 20E in growth, development, and insecticide resistance. However, little attention has been given to the association between the E20MO gene and DENV2 infection. The transcriptome of Ae. aegypti cells (Aag2 cells) infected with DENV2 revealed the presence of the E20MO gene. The subsequent quantification of E20MO gene expression levels in Aag2 cells post-DENV infection was carried out. A CRISPR/Cas9 system was utilized to create an E20MO gene knockout cell line (KO), which was then subjected to DENV infection. Analyses of DENV2 copies in KO and wild-type (WT) cells were conducted at different days post-infection (dpi). Plasmids containing E20MO were constructed and transfected into KO cells, with pre- and post-transfection viral copy comparisons. Gene expression levels of E20MO increased after DENV infection. Subsequently, a successful generation of an E20MO gene knockout cell line and the verification of code-shifting mutations at both DNA and RNA levels were achieved. Furthermore, significantly elevated DENV2 RNA copies were observed in the mid-infection phase for the KO cell line. Viral RNA copies were lower in cells transfected with plasmids containing E20MO, compared to KO cells. Through knockout and plasmid complementation experiments in Aag2 cells, the role of E20MO in controlling DENV2 replication was demonstrated. These findings contribute to our understanding of the intricate biological interactions between mosquitoes and arboviruses.

Phytoplasma disease is one of the most serious infectious diseases that affects the growth and development of mulberry. Long non-coding RNAs (lncRNAs) play an important role in plants’ defense systems; however, the contribution of lncRNAs in the response to phytoplasma infection in mulberry is still largely unknown. Herein, strand-specific RNA sequencing was performed to profile the mRNAs and lncRNAs involved in the response to phytoplasma infection in mulberry, and a total of 4169 genes were found to be differentially expressed (DE) between healthy and phytoplasma-infected leaves. Moreover, 1794 lncRNAs were identified, of which 742 lncRNAs were DE between healthy and infected leaves. Target prediction showed that there were 68 and 44 DE lncRNAs which may function as cis and trans-regulators, targeting 54 and 44 DE genes, respectively. These DE target genes are associated with biological processes such as metabolism, signaling, development, transcriptional regulation, etc. In addition, it was found that the expression of the antisense lncRNA (MuLRR-RLK-AS) of the leucine-rich repeat receptor-like protein kinase gene (MuLRR-RLK) was decreased in the phytoplasma-infected leaves. Interestingly, it was found that overexpression of MuLRR-RLK-AS can inhibit the expression of MuLRR-RLK. Moreover, it was found that the expression levels of PTI-related and MAPK genes in the transgenic MuLRR-RLK Arabidopsis plants were significantly higher than those in the wild-type plants when inoculated with pathogens, and the transgenic plants were conferred with strong disease resistance. Our results demonstrate that MuLRR-RLK-AS, as a trans-regulatory factor, can inhibit the expression of the MuLRR-RLK gene and is a negative regulatory factor for mulberry resistance. The information provided is particularly useful for understanding the functions and mechanisms of lncRNAs in the response to phytoplasma infection in mulberry.

GNBPB6, a beta-1,3-glucan-binding protein, was identified in the transcriptome of Aedes aegypti (A. aegypti) with dengue (DENV), Zika (ZIKV), and chikungunya viruses (CHIKV). In this study, we not only clarified that DENV2 and ZIKV regulate the changes in GNBPB6 expression but also identified the relationship of this gene with viral infections. The changes in GNBPB6 expression were quantified and showed a decrease in A. aegypti cells (Aag2 cells) at 2 dpi and 3 dpi and an increase at 4 dpi and 5 dpi (p < 0.05). A significant increase was observed only at 5 dpi after DENV2 infection. Subsequently, a GNBPB6 knockout (KO) cell line was constructed using the CRISPR/Cas9 system, and the DENV2 and ZIKV RNA copies, along with cell densities, were quantified and compared between the KO and wild type (WT) cells at different dpi. The result showed that DENV2 and ZIKV RNA copies were significantly increased in the KO cell line with no significant change in cell growth. Finally, DENV2 copies decreased after GNBPB6 was complemented in the KO. In conclusion, GNBPB6 knockout and complementation in Aag2 cells revealed that GNBPB6 can inhibit the replication of both DENV2 and ZIKV. These results contribute to subsequent research on mosquito–virus interactions.

Abstract Mosquito transmit numbers of parasites and pathogens resulting in fatal diseases. Species identification is a prerequisite for effective mosquito control. Existing morphological and molecular classification methods have evitable disadvantages. Here we introduced Deep learning techniques for mosquito species identification. A balanced, high-definition mosquito dataset with 9900 original images covering 17 species was constructed. After three rounds of screening and adjustment-testing (first round among 3 convolutional neural networks and 3 Transformer models, second round among 3 Swin Transformer variants, and third round between 2 images sizes), we proposed the first Swin Transformer-based mosquito species identification model (Swin MSI) with 99.04% accuracy and 99.16% F1-score. By visualizing the identification process, the morphological keys used in Swin MSI were similar but not the same as those used by humans. Swin MSI realized 100% subspecies-level identification in Culex pipiens Complex and 96.26% accuracy for novel species categorization. It presents a promising approach for mosquito identification and mosquito borne diseases control.

Abstract Background Both Culex quinquefasciatus and Cx. pipiens molestus are sibling species within Cx. pipiens complex. Even though they are hard to distinguish morphologically, they have different physiological behaviors. However, the molecular mechanisms underlying these differences remain poorly understood. Methods Transcriptome sequencing was conducted on antennae of two sibling species. The identification of the differentially expressed genes (DEGs) was performed by the software DESeq2. Database for Annotation, Visualization and Integrated Discovery was used to perform GO pathway enrichment analysis. The protein–protein interaction (PPI) network was constructed with Cytoscape software. The hub genes were screened by the CytoHubba plugin and Degree algorithms. The identified genes were verified by quantitative real-time PCR. Results Most annotated transcripts (14,687/16,005) were expressed in both sibling species. Among 15 identified odorant-related DEGs, OBP10 was expressed 17.17 fold higher in Cx. pipiens molestus than Cx. quinquefasciatus. Eighteen resistance-related DEGs were identified, including 15 from CYP gene family and three from acetylcholinesterase, in which CYP4d1 was 86.59 fold more highly expressed in C. quinquefasciatus. Three reproductive DEGs were indentified with the expression from 5.01 to 6.55 fold. Among eight vision-related DEGs, retinoic acid receptor RXR-gamma in Cx. pipiens molestus group was more expressed with 214.08 fold. Among the 30 hub genes, there are 10 olfactory-related DEGs, 16 resistance-related DEGs, and four vision-related DEGs, with the highest score hub genes being OBP lush (6041148), CYP4C21 (6044704), and Rdh12 (6043932). The RT-qPCR results were consistent with the transcriptomic data with the correlation coefficient R = 0.78. Conclusion The study provided clues that antennae might play special roles in reproduction, drug resistance, and vision, not only the traditional olfactory function. OBP lush, CYP4C21, and Rdh12 may be key hints to the potential molecular mechanisms behind the two sibling species' biological differences. Graphical Abstract

Abstract Purpose of the study Previous literature has examined the relationship between high school students’ postsecondary STEM major choices and their prior interest and perceived ability in mathematics. Yet, we have limited understanding of whether and how perceived ability and interest in science and mathematics jointly affect students’ STEM major choices. Results Using the most recent nationally representative longitudinal cohort of U.S. secondary school students, we examine the degree to which students’ perceived mathematical and scientific abilities and interests predict their STEM major choices, employing logistic regression and a series of interaction analyses. We find that while both mathematics and science perceived ability positively influence STEM major selection, academic interest in these subjects is a weaker predictor. Moreover, across a series of analyses, we observe a significant gender gap—whereby women are less than half as likely to select STEM majors—as well as nuanced distinctions by self-identified race. The relationships among perceived ability, interest, and STEM major choice are not found to meaningfully vary by race nor consistently by gender. However, perceived ability has a more positive effect for men than women who are pursuing Computing/Engineering majors and a more positive effect for women than men who are pursuing other STEM majors, including less applied Social/Behavioral, Natural, and Other Sciences. Implications These findings suggest potential opportunities to enhance their perceived mathematical and scientific abilities in high school, positioning them to potentially enter STEM fields. School sites with more resources to support the ambitions of STEM students of all backgrounds may be better positioned to reduce postsecondary disparities in STEM fields. Given existing opportunity gaps and resource differentials among schools, corresponding recommendations are suggested.

Introduction: Since Aedes aegypti invaded Yunnan Province in 2002, its total population has continued to expand. Shi et al. used microsatellite and mitochondrial molecular markers to study the Ae. aegypti populations in Yunnan Province in 2015 and 2016, found that it showed high genetic diversity and genetic structure. However, there are few studies on the population genetic characteristics of Ae. aegypti in Yunnan Province under different levels of human intervention. This study mainly used two common types of molecular markers to analyze the genetic characteristics of Ae. aegypti, revealing the influence of different input, prevention and control pressures on the genetic diversity and structure of this species. Understanding the genetic characteristics of Ae. aegypti populations and clarifying the diversity, spread status, and source of invasion are essential for the prevention, control and elimination of this disease vector.Methods: We analyzed the genetic diversity and genetic structure of 22 populations sampled in Yunnan Province in 2019 and 17 populations sampled in 2020 through nine microsatellite loci and COI and ND4 fragments of mitochondrial DNA. In 2019, a total of 22 natural populations were obtained, each containing 30 samples, a total of 660 samples. In 2020, a total of 17 natural populations were obtained. Similarly, each population had 30 samples, and a total of 510 samples were obtained.Results: Analysis of Ae. aegypti populations in 2019 and 2020 based on microsatellite markers revealed 67 and 72 alleles, respectively. The average allelic richness of the populations in 2019 was 3.659, while that in 2020 was 3.965. The HWE analysis of the 22 populations sampled in 2019 revealed significant departure only in the QSH-2 population. The 17 populations sampled in 2020 were all in HWE. The average polymorphic information content (PIC) values were 0.546 and 0.545, respectively, showing high polymorphism. The average observed heterozygosity of the 2019 and 2020 populations was 0.538 and 0.514, respectively, and the expected average heterozygosity was 0.517 and 0.519, showing high genetic diversity in all mosquito populations. By analyzing the COI and ND4 fragments in the mitochondrial DNA of Ae. aegypti, the populations sampled in 2019 had a total of 10 COI haplotypes and 17 ND4 haplotypes. A total of 20 COI haplotypes were found in the populations sampled in 2020, and a total of 24 ND4 haplotypes were obtained. STRUCTURE, UPGMA and DAPC cluster analyses and a network diagram constructed based on COI and ND4 fragments showed that the populations of Ae. aegypti in Yunnan Province sampled in 2019 and 2020 could be divided into two clusters. At the beginning of 2020, due to the impact of COVID-19, the flow of goods between the port areas of Yunnan Province and neighboring countries was reduced, and the sterilization was more effective when goods enter the customs, leading to different immigration pressures on Ae. aegypti population in Yunnan Province between 2019 and 2020, the source populations of the 2019 and 2020 populations changed. Mantel test is generally used to detect the correlation between genetic distance and geographical distance, the analysis indicated that population geographic distance and genetic distance had a moderately significant correlation in 2019 and 2020 (2019: p < 0.05 R2 = 0.4807, 2020: p < 0.05 R2 = 0.4233).Conclusion:Ae. aegypti in Yunnan Province maintains a high degree of genetic diversity. Human interference is one reason for the changes in the genetic characteristics of this disease vector.

IntroductionN-methyl-D-aspartate receptor (NMDAR) is one of the main receptor of the excitatory neurotransmitter glutamate in the brain, which is the key determinant of the excitatory/inhibitory balance of neural network. GluN2A/GRIN2A is one of the subunits of NMDAR and plays an important role in epilepsy. Approximately 78% of patients with GluN2A/Grin2a mutations have epilepsy, and the underlying mechanism of this association is not well characterized.MethodsWe constructed a mouse model of hyperthermic seizure, and conducted in vitro and in vivo electrophysiological and behavioral studies to clarify the pathogenic characteristics and mechanism of GluN2A/GRIN2A-V685G mutation. In addition, the drug efavirenz (EFV), which is used to treat HIV infection, was administrated to mutant animals to assess whether it can restore the loss of function.ResultsMutant mice showed no significant change in the mRNA or protein expressions of NMDAR compared with wild type (WT) mice. Mice with GluN2A/GRIN2A-V685G mutation exhibited shorter latency to seizure, increased frequency of seizure-like events, decreased peak current and current area of NMDAR excitatory postsynaptic current, and decreased event frequency of micro-inhibitory postsynaptic current, compared to WT mice. They also exhibited decreased threshold, increased amplitude, increased input resistance, and increased root number of action potential. EFV administration reversed these changes. The loss-of-function (LoF) mutation of NMDAR changed the excitatory/inhibitory balance of neural network, rendering animal more prone to seizures.DiscussionEFV was indicated to hold its potential in the treatment of inherited epilepsy.

Abstract Background Aedes aegypti is one of the most important vectors of zoonotic diseases worldwide, and its survival and reproductive processes depend heavily on its olfactory system. In this study, the expression levels of all odorant receptor (OR) genes of Ae. aegypti were explored during different physiological periods to identify olfactory genes that may be associated with mosquito blood-feeding and the search for oviposition sites. Methods Four experimental groups, consisting of Ae. aegypti males, pre-blood-feeding females, post-blood-feeding females and post-oviposition females, were established. A total of 114 pairs of primers targeting all messenger RNA encoded by OR genes were designed based on the whole genome of Ae. aegypti. The expression of OR genes was evaluated by real-time fluorescence quantitative PCR for relative quantification and the comparison of differences between groups. Results A total of 53 differentially expressed OR genes were identified between males and females in Ae. aegypti antennae. Also, eight, eight and 13 differentially expressed OR genes were identified in pre- versus post-blood-feeding females, in pre- versus post-oviposition females and in post-blood-feeding versus post-oviposition females, respectively. In addition, 16 OR genes were significantly differentially expressed in multiple physiological periods of the mosquitoes. Conclusions A large number of ORs with significant intergroup differences and high expression levels were screened in this study. Some of these genes are reported for the first time, providing possible targets for the development of mosquito control pathways based on the olfactory system. Graphical Abstract

Background: Parathyroid carcinoma (PC), often misdiagnosed as a parathyroid adenoma (PA), is prone to local relapse due to the initial surgery being restricted to parathyroid lesions instead of en bloc resection of parathyroid lesions with negative incision margins. However, it is very challenging to distinguish PC from PA preoperatively; hence, this study investigated an effective biomarker for increasing accurac y in PC diagnosis. Method: First, the differentially expressed circular RNAs between three PC tissues and three PA tissues were screened by high-throughput circular RNA sequencing, and the expression of hsa_circ_0005729 was verified by qRT-PCR in 14 patients with PC and 40 patients with PA. Secondly, the receiver operating characteristic curve and the area under the curve (AUC) were used to analyze the diagnostic efficie ncy of hsa_circ_0005729 in PC by combining with laboratory data. Thirdly, RNF138 mRNA, the corresponding linear transcript of hsa_circ_0005729, was measured, and the relationship between hsa_ circ_0005729 and RNF138 mRNA was analyzed in patients with PA and patients with PC. Results: Hsa_circ_0005729 expression was significantly higher in patients with PC than in patients with PA. Serum calcium (P = 0.045), alkaline phosphatase (ALP) (P = 0.048), and creatinine levels (P = 0.036) were significantly higher in patients with PC than in pa tients with PA. The AUC increased to 0.86 when hsa_circ_0005729 combined with serum calcium, creatinine, and ALP. In addition, hsa_circ_0005729 was positively correlated with RNF138 mRNA in patients with PA but not in patients with PC. Conclusion: The novel circular RNA hsa_circ_0005729 was found to have a higher expression in patients with PC, indicating its usefulness for distinguishing PC from PA.

Experiments such as microbial activation culture, subculture selection, and fire damage repair of cement mortar specimens were conducted to investigate the repairing effect of Sporosarcina pasteurii as a repair agent for fire-damaged cracks in cement mortar specimens. In addition, multi-scale parameters such as compressive strength and chloride ion migration coefficient of cement mortar specimens before and after restoration were compared. The effect of microbial mineralization technology on the repair of fire-damaged cracks in cement mortar specimens was investigated, and the microstructure and mineral composition of the products were analyzed. The results showed that the strong alkaline environment in the cracks of the cement mortar specimens after a high temperature of 500 °C inhibited the activity of bacteria and weakened the mineralization effect; the compressive strength of the repaired cement mortar specimens was 22.8% higher than that of the unrepaired fire-damaged specimens; the compressive strength of the repaired cement mortar reached 78.2% of the strength of the original cement mortar specimen without high temperature; after restoration, the chloride ion penetration resistance of the cement mortar specimens decreased by about 16.9% compared with that before restoration.

Lung cancer is a life-threatening malignant tumour that is prevalent worldwide. Here, the GCNT3 gene in lung adenocarcinoma was studied via public databases, and cytology and molecular biology experiments were performed to further explore the role of this gene in lung adenocarcinoma. In this study, abnormally high GCNT3 expression levels were observed in tumour tissues compared with normal tissues at both the mRNA and protein levels. In the pancancer analysis, abnormal GCNT3 expression was observed in many tumour types. Moreover, the survival analysis revealed that among patients receiving radiotherapy, those with high GCNT3 expression levels had a worse prognosis. Cell and molecular biology experiments showed that the proliferation, migration and invasion capabilities of the A549 cell line were decreased after knockdown of GCNT3, and epithelial-mesenchymal transformation was significantly inhibited. In subsequent studies, we found that the sensitivity of cells to radiotherapy was enhanced after GCNT3 knockdown. Overall, our findings reveal that GCNT3 is an important factor affecting the radiotherapy sensitivity of lung adenocarcinoma, and GCNT3 inhibition deserves further study as a radiotherapy sensitising strategy.

Background: To investigate the preventive effect of early multidisciplinary collaboration combined with cluster strategy on acquired frailty syndrome in ICU of patients with acute myocardial infarction (AMI). Methods: A total of 240 patients with myocardial infarction (MI) admitted to ICU in The Third Affiliated Hospital of Qiqihar Medical University, Qiqihar, China from March 2018 to March 2020 were selected. The patients were randomly divided into experiment group and control group equally according to random number method. The experimental group was treated with early multidisciplinary collaboration and cluster strategy, while the control group was treated with routine nursing mode. Duration of ventilation, total length of stay, length of ICU stay, New York College of Cardiology cardiac function classification, MRC-Score, BI and complications during hospitalization were recorded. Results: After intervention, there were statistically significant differences in ventilation duration, total length of stay, length of stay in ICU, Heart function classification of New York Cardiology Society, MRC-Score, BI index, SF-36 score and complications during hospitalization in 2 groups (P < 0.05), and the above indicators in the experimental group were better than those in the control group. Conclusion: Early multidisciplinary collaboration combined with cluster strategy can significantly reduce the incidence of AMI in ICU of MI patients.

BackgroundWolbachia is gram-negative and common intracellular bacteria, which is maternally inherited endosymbionts and could expand their propagation in host populations by means of various manipulations. Recent reports reveal the natural infection of Wolbachia in Aedes Aegypti in Malaysia, India, Philippines, Thailand and the United States. At present, none of Wolbachia natural infection in Ae. aegypti has been reported in China.MethodsA total of 480 Ae. aegypti adult mosquitoes were collected from October and November 2018 based on the results of previous investigations and the distribution of Ae. aegypti in Yunnan. Each individual sample was processed and screened for the presence of Wolbachia by PCR with wsp primers. Phylogenetic trees for the wsp gene was constructed using the neighbour-joining method with 1,000 bootstrap replicates, and the p-distance distribution model of molecular evolution was applied.Results24 individual adult mosquito samples and 10 sample sites were positive for Wolbachia infection. The Wolbachia infection rate (IR) of each population ranged from 0 - 41.7%. The infection rate of group A alone was 0%-10%, the infection rate of group B alone was 0%-7.7%, and the infection rate of co-infection with A and B was 0-33.3%.ConclusionsWolbachia infection in wild Ae. aegypti in China is the first report based on PCR amplification of the Wolbachia wsp gene. The Wolbachia infection is 5%, and the wAlbA and wAlbB strains were found to be prevalent in the natural population of Ae. aegypti in Yunnan Province.
