Importance: Acupoint autohemotherapy (AA), a therapeutic technique involving the subcutaneous injection of autologous blood into acupoints, has been empirically validated as safe and effective for treating asthma by alleviating symptoms and decreasing acute attacks, though its mechanism is not well understood.Objective: The role of heme oxygenase-1 (HO-1) in AA-induced suppression of asthmatic airway inflammation is examined. Methods: Twenty rats were assigned randomly to four groups, namely the Control, OVA, OVA + AA, and (OVA + Snpp) + AA. Rats in the OVA + AA and (OVA + Snpp) + AA received autologous blood injections into acupoints (BL13 and BL23) following OVA challenge. Rats in the (OVA + Snpp) + AA were concurrently subjected to intraperitoneal injections of Snpp, a inhibitor of HO-1. Airway inflammation was evaluated through HE staining, while the concentrations of cytokines in BALF were quantified using ELISA. The mRNA and protein levels of RORγt (Th17-specific transcription factor), Foxp3 (Treg-specific transcription factor), and HO-1 in lung tissue were assessed through qRT-PCR and WB. Results: HE staining indicated that airway inflammation was alleviated in the OVA + AA. The OVA + AA displayed significantly lower counts of total cells and eosinophils in the BALF compared to both the OVA and (OVA + Snpp) + AA. The ELISA demonstrated a significant decrease in levels of pro-inflamatory cytokines (IL-4, IL-17A), and an increase in levels of anti-inflamatory cytokines (IFN-γ, IL-10), in the OVA + AA when compared to both OVA and (OVA + Snpp) + AA. The qRT-PCR and WB analyses revealed an upregulation of HO-1 and Foxp3 expression, and a downregulation of RORγt expression, in the OVA + AA when compared to OVA and (OVA + Snpp) + AA. Conclusions and Relevance: The involvement of HO-1 in the underlying mechanism responsible for the anti-inflammatory effects of AA is evident.

High-temperature molten salt capture and electrolysis technology of CO2 for the effective preparation of elemental carbon materials are considered to be an exceedinglyare gaining traction as promising carbon capture utilization and storage technology. The reason is their excellent selectivity and ease of operation, making them suitable for the effective preparation of elemental carbon materials from CO2. Nonetheless, the current research on the kinetic mechanism of CO2 capture in high-temperature molten salt systems is inadequate. Furthermore, the energy consumption and production costs associated with the process of preparing elemental carbon materials through CO2 molten salt electrolysis are relatively high. Therefore, achieving efficient CO2 capture in high-temperature molten salts and low-energy consumption electrolysis is very crucial. This paper delves into the thermodynamics of CO2 capture by various alkali metals/alkaline earth metal oxides. It also compares the theoretical decomposition voltage and energy consumption of various alkali metals/alkaline earth metal carbonates. The capture behavior of CO2 in typical binary chloride molten salts is also examined. The study further explores the thermodynamics of CO2 capture in molten salts to carbonates and calculates the theoretical voltage and power consumption during carbonate electrolysis in a high-temperature molten salt system. The kinetics of CO2 capture in typical CaCl2-based molten salts are examined by advanced technologies such as online gas mass spectrometry, carbon–sulfur analyzer, X-ray diffraction, and energy spectroscopy. The effects of temperature and voltage on the electrolysis process, electrolytic energy consumption, and product structure of elemental carbon materials prepared by CO2 electrolysis are investigated in typical ternary Na2CO3–K2CO3–Li2CO3 molten salts and ternary CaCl2–NaCl–CaO molten salts using scanning electron microscopy (SEM), Raman spectroscopy, and constant voltage electrolysis. The results indicate that CaO outperforms alkali metal oxide as a CO2 capture agent, and CaCO3 obtained by energy and conversion exhibits the lowest theoretical decomposition voltage and electrolytic energy consumption. In a typical CaCl2–3.0% CaO (mass fraction) molten salt, the CO2 capture capacity is 0.016 gCO2·g−1. Compared with the Na2CO3–K2CO3–Li2CO3 molten salt, the CaCl2–NaCl–CaO molten salt has a lower electrolysis voltage and energy consumption. Moreover, the current efficiency of the electrolytic elemental carbon material reaches up to 95.3%, with the minimum electrolytic energy consumption being only 14.1 kW·h·kg−1 under the optimal conditions of 750 ℃ and 1.5 V. Therefore, this study provides a foundation for exploring CO2 molten salt capture materials that combine excellent performance, low cost, and environmental friendliness. It also offers theoretical and data support for optimizing the process and reducing the cost of the actual electrolysis.

Abstract Background Polycystic ovary syndrome (PCOS) is a unity of endocrine and metabolic disorders, associated with PI3K/AKT/mTOR, autophagy, and gut microbiota. Quinoa is a valuable food source, which contains rich minerals, unsaturated fatty acids, and has a positive modulating effect on metabolic diseases. However, its effects and potential mechanisms on PCOS have not been reported yet. Therefore, the purpose of this study is to investigate the effect of quinoa on PCOS rats by regulating PI3K/AKT/mTOR, autophagy, and gut microbiota. Methods Ten–week-old female Sprague-Dawley (SD) rats have received letrozole for 24 days for induction of PCOS and subsequently were treated with a quinoa diet for 8 weeks. Vaginal smears were used to analyze the estrous cycle of rats. Hormone and biochemical indexes were analyzed by kit assays and glucometer. The pathological changes of ovary, pancreas, duodenum and colon were observed by HE staining. PI3K, AKT, mTOR and autophagy-related proteins in the ovary and colon were measured by western blot and immunohistochemistry staining. Tight junction proteins in colon were measured by immunohistochemistry staining. 16 s rDNA sequencing was used to detect the changes of intestinal microbiota in rats. Network pharmacology and molecular docking were used to study the possible targets and mechanisms of quinoa on PCOS. Spearman correlation analysis was used to study the relationship between intestinal microbial abundance and hormone levels of PCOS rats at the phylum and genus level. Results Quinoa significantly improved estrous cycle and biochemical parameters of PCOS-like rats, and the pathological state of ovary, pancreas, duodenum and colon tissues. Especially, quinoa significantly regulated the expression of PI3K, AKT, mTOR and autophagy-related proteins in the ovary. Quinoa may repair the intestinal barrier by upregulating the expression of tight junction proteins in the colon, and regulate autophagy-related factors in colon. Additionally, quinoa increased the abundance of Lactobacillu, Bacteroides and Oscillospira, and decreased the Firmicutes/Bacteroidetes ratio and the Blautia, and Prevotella, reversing the dysregulation of the gut microbiota. Correlation analysis showed that there is a strong correlation between gut microbiota with significant changes in abundance and hormone related to PCOS. Conclusion Our result indicated that effect of quinoa on PCOS maybe associated with activation of the PI3K/AKT/mTOR signaling pathway, inhibition of autophagy, and regulation of intestinal flora.

Abstract Background Aging negatively impacts tissue repair, particularly in skeletal muscle, where the regenerative capacity of muscle stem cells (MuSCs) diminishes with age. Although aerobic exercise is known to attenuate skeletal muscle atrophy, its specific impact on the regenerative and repair capacity of MuSCs remains unclear. Methods Mice underwent moderate‐intensity continuous training (MICT) from 9 months (aged + Ex‐9M) or 20 months (aged + Ex‐20M) to 25 months, with age‐matched (aged) and adult controls. Histological examinations and MuSC transplantation assays assessed aerobic exercise effects on MuSC function and muscle regeneration. Abstract Purpose This study utilizes a meta-analytic approach to investigate the effects of cryoablation and robot-assisted partial nephrectomy on perioperative outcomes, postoperative renal function, and oncological results in patients. Methods This study followed the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) statement. Four electronic databases (PubMed, Embase, Web of Science, and the Cochrane Library database) were systematically searched to identify relevant studies published in English up to November 2023. The primary outcomes were perioperative results, complications, postoperative renal function and oncologic outcomes. Review Manager 5.4 was used for this analysis. Results This study included a total of 10 studies comprising 2,011 patients. Abstract Wearable ultrasound imaging technology has become an emerging modality for the continuous monitoring of deep-tissue physiology, providing crucial health and disease information. Fast volumetric imaging that can provide a full spatiotemporal view of intrinsic 3D targets is desirable for interpreting internal organ dynamics. However, existing 1D ultrasound transducer arrays provide 2D images, making it challenging to overcome the trade-off between the temporal resolution and volumetric coverage. In addition, the high driving voltage limits their implementation in wearable settings. With the use of microelectromechanical system (MEMS) technology, we report an ultrasonic phased-array transducer, i.e., a 2D piezoelectric micromachined ultrasound transducer (pMUT) array, which is driven by a low voltage and is chip-compatible for fast 3D volumetric imaging. By grouping multiple pMUT cells into one single drive channel/element, we propose an innovative cell–element–array design and operation of a pMUT array that can be used to quantitatively characterize the key coupling effects between each pMUT cell, allowing 3D imaging with 5-V actuation. The pMUT array demonstrates fast volumetric imaging covering a range of 40 mm × 40 mm × 70 mm in wire phantom and vascular phantom experiments, achieving a high temporal frame rate of 11 kHz. The proposed solution offers a full volumetric view of deep-tissue disorders in a fast manner, paving the way for long-term wearable imaging technology for various organs in deep tissues.

Purpose This study compared the efficacy of heat insulation between 5% dextrose and 0.9% saline in radiofrequency ablation (RFA). Accordingly, temperature variations and maximum temperatures were assessed at identical distances and heat field distributions. Methods Cubes of porcine liver tissue, measuring 10 mm across, were selected to precisely align the ablation boundary with the tissue boundary. An 18-gauge electrode with a 7-mm tip was inserted into each cube (10 per group) in a stainless-steel cup containing 40 mL of 5% dextrose or 0.9% saline. Fixed ablation was performed for 3 minutes using continuous mode at 30 W, simulating the typical thermal environment during thyroid RFA. Real-time temperature measurements were recorded by sensors positioned 0, 1, 3, and 5 mm from the cube’s edge. A comparative analysis was conducted to assess the maximum temperature, temperature variation, and duration of temperatures exceeding 42℃. Results In both groups, the temperature curve declined with increasing distance from the edge of the ablated tissue. However, 0.9% saline exhibited higher maximum temperatures at 1, 3, and 5 mm compared to 5% dextrose (1 mm: 44.55°C±5.25°C vs. 34.68°C±3.07°C; 3 mm: 39.64°C±2.53°C vs. 29.22°C±2.21°C; 5 mm: 38.86°C±2.14°C vs. 28.74°C±2.51°C; all P

Objective: To evaluate whether improved progression-free survival (PFS) from radiotherapy (RT) translates into an overall survival (OS) benefit for diffuse large B-cell lymphoma (DLBCL). Methods: A systematic literature search identified randomized controlled trials (RCTs) and retrospective studies that compared combined-modality therapy (CMT) with chemotherapy (CT) alone. Weighted regression analyses were used to estimate the correlation between OS and PFS benefits. Cohen's kappa statistic assessed the consistency between DLBCL risk-models and PFS patterns. Furthermore, the benefit trend of RT was analyzed by fitting a linear regression model to the pooled hazard ratio (HR) according to the PFS patterns. Results: For both 7 RCTs and 52 retrospective studies, correlations were found between PFS HR (HRPFS) and OS HR (HROS) at trial level (r = 0.639–0.876), and between PFS and OS rates at treatment-arm level, regardless of CT regimens (r = 0.882–0.964). Incorporating RT into CT increased about 18% of PFS, and revealed a different OS benefit profile. Patients were stratified into four CT-generated PFS patterns (>80%, >60–80%, >40–60%, and ≤40%), which was consistent with risk-stratified subgroups (kappa > 0.6). Absolute gain in OS from RT ranged from ≤5% at PFS >80% to about 21% at PFS ≤40%, with pooled HROS from 0.70 (95% CI, 0.51–0.97) to 0.48 (95% CI, 0.36–0.63) after rituximab-based CT. The OS benefit of RT was predominant in intermediate- and high-risk patients with PFS ≤ 80%. Conclusion: We demonstrated a varied OS benefit profile of RT to inform treatment decisions and clinical trial design.

The bio-corrosion behavior and bio-compatibility of TiNbZrTa and TiNbZrTaMo high entropy alloys (HEAs) are crucial for their efficient maintenance during biological implantation. In this work, TiNbZrTa and TiNbZrTaMo HEAs were successfully prepared via vacuum melting, with a β-type titanium alloy Ti35Nb7Zr5Ta, exhibiting good biocompatibility used as a comparison. Due to the high entropy effect of equal atomic ratios, both TiNbZrTa and TiNbZrTaMo HEAs exhibit body-centered cubic (BCC) structure and lattice distortion compared to the β titanium alloy Ti35Nb7Zr5Ta. Mo elements contribute to the formation of a new BCC phase, resulting in a white matrix rich in Ta and Mo, and a dark second phase rich in Ti, Zr, and Nb. Electrochemical test results at 37 °C shown that TiNbZrTa has a wider and more stable passivation area than TiNbZrTaMo. The XPS results shown that formation of the passive film is related to the main elements added. Ti, Nb, Ta and Mo formed oxides via the solid-liquid interface and migrate inward, while Zr is formed at the passivation film/metal substrate interface and migrates outward, finally forming the layered structure of the passivation film. Additionally, the cytotoxicity test of mouse fiber cells was carried out to evaluate the biocompatibility of the HEAs. The results shown that the cell proliferation rate of TiNbZrTa and TiNbZrTaMo HEAs reached 0 and 1, respectively, with TiNbZrTa exhibiting better biocompatibility, as adding Mo reduced the cell proliferation rate. These findings may provide a method for predicting the bio-corrosion behavior and bio-compatibility of HEAs used for biological implantation.

The transition period for dairy cows usually refers to the 3 weeks pre-calving to the 3 weeks post-calving. During this period, dairy cows undergo metabolic and physiological adaptations because of their susceptibility to metabolic and infectious diseases. Poor feeding management under these circumstances may adversely affect the health and subsequent production performance of the cows. Owing to long-term adaptation and evolution, the rumen has become a unique ecosystem inhabited by a complex microbial community closely associated with its natural host. Dietary components are metabolized by the rumen microbiota, and volatile fatty acids and microbial protein products can be used as precursor substances for synthesizing meat and milk components. The successful transition of perinatal dairy cows includes changes in diet, physiology, and the rumen microbiota. Rumen microbial profiles have been confirmed to be heritable and repairable; however, adverse circumstances affect rumen microbial composition, host digestion and metabolism, as well as postpartum production traits of dairy cows for a certain period. Preliminary evidence indicates a close relationship between the rumen microbiota and animal performance. Therefore, changes in rumen microbes during the transition period and the intrinsic links between the microbiota and host postpartum phenotypic traits need to be better understood to optimize production performance in ruminants.

Aiming at the problem that there exist lacks of analysis of the elastic properties of screen panel materials and research on the influence of mineral impact on the deflection changes of screen panel in response to the theory of large deflection deformation of flip-flow screen surface. Considering the scalability of the flip-flow screen panel material, a theoretical model of large deflection nonlinear elastic deformation of the screen panel is established. Combining with the external excitation sine displacement function, the time-domain expression of the screen panel with different amplitudes and frequencies is proposed for the first time. A testing platform for the dynamic characteristics of the flip-flow screen panel is built to verify the accuracy of the theoretical model in describing the deflection change process of the screen panel, and the optimal vibration parameters of the large deflection elastic deformation theory in describing the dynamic characteristics of the screen panel is clarified using the response surface optimization method. Based on the theory of large deflection elastic deformation of unloaded screen panel, a deflection variation theory of screen panel when particles collide with screen panel is proposed, and the deflection curve of screen panel with different motion states under particle impact force is calculated. Then the influence of different physical parameters on the local deformation degree of the collision point between the screen panel and mineral particles is revealed, and the accuracy of the theoretical expression of the deflection change of the screen panel under the collision of particles is verified through simulation experiments in describing the dynamic characteristics of the screen panel using different initial constraints. The research results show that the error rate of the experiments and theory of large deflection elastic deformation of screen panel under dynamic conditions is below 6%. The theoretical and experimental error rates of large deflection elastic deformation of the screen panel under dynamic conditions decrease with the increase of amplitude and frequency, but this change is nonlinear. When the amplitude is 7.33 mm and the frequency is 11.75 Hz, the error rate between the theoretical and experimental values at each measuring point reaches the lowest, and the average error rate at the midpoint of the screen panel is less than 3%. The incident angle of particles, the position of the impact point, the installation angle of the screen panel, and the operating state are important factors that affect the degree of local deformation of the screen panel during impact. Theoretical proof is shown that when the inclination angle and particle incidence angle of the screen surface installation are large, and the screen panel near the midpoint position move to the highest point, the collision between particles and the screen panel can obtain the maximum initial kinetic energy, and there is an average error of less than 4 mm between deflection deformation theory of the screen panel after collision in describing the impact point position and simulation results.

The issue of opacity within data-driven artificial intelligence (AI) algorithms has become an impediment to these algorithms’ extensive utilization, especially within sensitive domains concerning health, safety, and high profitability, such as chemical engineering (CE). In order to promote reliable AI utilization in CE, this review discusses the concept of transparency within AI utilizations, which is defined based on both explainable AI (XAI) concepts and key features from within the CE field. This review also highlights the requirements of reliable AI from the aspects of causality (i.e., the correlations between the predictions and inputs of an AI), explainability (i.e., the operational rationales of the workflows), and informativeness (i.e., the mechanistic insights of the investigating systems). Related techniques are evaluated together with state-of-the-art applications to highlight the significance of establishing reliable AI applications in CE. Furthermore, a comprehensive transparency analysis case study is provided as an example to enhance understanding. Overall, this work provides a thorough discussion of this subject matter in a way that—for the first time—is particularly geared toward chemical engineers in order to raise awareness of responsible AI utilization. With this vital missing link, AI is anticipated to serve as a novel and powerful tool that can tremendously aid chemical engineers in solving bottleneck challenges in CE.

Abstract Colorectal cancer is one of the most common cancers around the world, which is a severe threat to people’s health. SMAD4 belongs to the dwarfin/SMAD family, which plays a crucial role in TGF-β and BMP signal pathways. As the molecular characterization of colon cancer patients following SMAD4 mutations remains unclear, we integrated multi-omics data of SMAD4 mutant patients to reveal the profile of molecular characterization of SMAD4 mutation. A missense mutation is the most common mutant type of SMAD4. Patients with SMAD4 mutation had worse survival. Tumor tissues from patients carrying the SMAD4 mutation showed a reduction in various immune cells, such as CD4 + memory T cells and memory B cells. Many differential genes were identified compared to the SMAD4 mutation-free group and could be significantly enriched for tumor- and immune-related signaling pathways. In addition, the mutant group had different drug sensitivities than the non-mutant group.

Abstract Revealing key factors that modulate the regioselectivity in heterogeneous hydroformylation requires identifying and monitoring the dynamic evolution of the truly active center under real reaction conditions. However, unambiguous in situ characterizations are still lacking. Herein, we elaborately construct a series of Rh-POPs catalysts for propylene hydroformylation which exhibited tunable regioselectivity. Multi-technique approaches reveal the unique microenvironment of the diverse HRh(CO)(PPh3-frame)2 sites with distinct P-Rh-P bite angles ranging from 90° to 120° and 158° to 168°, respectively. In situ time-resolved XAFS, FT-IR, and quasi-in situ Solid-state NMR experiments combined with DFT calculations explain the dynamic evolution of the electronic and coordinate state of the distinct active sites induced by hemilabile PPh3-frame ligands and further disclose the regulatory mechanism of regioselectivity. These state-of-the-art techniques and multiscale analysis advance the understanding of how hemilabile coordination influences regioselectivity and will provide a new thought to modulate the regioselectivity in future industrial processes.

The traditional evaporator of water source heat pump system is easy to freeze when the water source temperature is too low in winter. A novel ice source heat pump system is proposed, which can use low temperature surface water to supply heat to buildings. One of the key problems for safe transportation is velocity range of ice slurry in ice outlet pipeline. In order to study the critical flow velocity of ice slurry in the pipe, an Euler model of horizontal straight pipe and 90° elbow pipe were established by Fluent software. The influence of five factors on the critical flow velocity of two kinds of pipes, including initial ice packing fraction, ice particle size, pipe diameter, length of straight pipe and radius curvature of elbow was studied. It found that the initial ice content should be controlled from 10% to 15%. The inlet flow rate was 20% higher than the critical flow rate under the most unfavorable condition. Ice particle size was less than 0.2 mm. The total length of ice discharge pipeline was less than 50 m. Curvature radius of the bend should be met the minimum requirements of the specification of the pipe used.

Mesenchymal stem cells (MSCs), as a stem cell type with multiple differentiation potentials and immune regulatory abilities, have shown broad prospects in the treatment of ischemic stroke in recent years. The main characteristics of MSCs include their self-renewal ability, differentiation potential for different types of cells, and the ability to secrete various bioactive factors such as cytokines, chemokines, and growth factors, which play a key role in tissue repair and regeneration. In the treatment of ischemic stroke, MSCs exert therapeutic effects through various mechanisms, including promoting vascular regeneration of damaged brain tissue, reducing inflammatory responses, and protecting neurons from damage caused by apoptosis. Research have shown that MSCs can promote the repair of ischemic areas by releasing neurotrophic factors and angiogenic factors, while inhibiting immune responses triggered by ischemia, thereby improving neurological function. With the in-depth study of its biological mechanism, MSCs have gradually shown good safety and effectiveness in clinical applications. Therefore, fully exploring and utilizing the potential of MSCs in the treatment of ischemic stroke may provide new ideas and solutions for future neural repair and regenerative medicine.

Abstract Purpose The aim of study was to screen factors associated with the overall survival of colorectal cancer patients with lymph nodes metastasis who received neoadjuvant therapy and construct a nomogram model. Methods All enrolled subjects of the SEER database were randomly assigned to the training and testing group in a ratio of 3:2. The patients of Tangdu Hospital were seemed as validation group. Univariate cox regression analysis, lasso regression and random forest survival were used to screen variables related to the survival of advanced CRC patients received neoadjuvant therapy in the training group. Area under curves were adopted to evaluate the 1,3,5-year prediction value of the optimal model in three cohorts. Calibration curves were drawn to observe the prediction accuracy of the nomogram model. Decision curve analysis was used to assess the potential clinical value of the nomogram model. Results A total of 1833 subjects were enrolled in this study. After random allocation, 1055 cases of the SEER database served as the training group, 704 cases as the testing group and 74 patients from our center as the external validation group. Variables were screened by univariate cox regression used to construct a nomogram survival prediction model, including M, age, chemotherapy, CEA, perineural invasion, tumor size, LODDS, liver metastasis and radiation. The AUCs of the model for predicting 1-year OS in the training group, testing and validation group were 0.765 (0.703,0.827), 0.772 (0.697,0.847) and 0.742 (0.601,0.883), predicting 3-year OS were 0.761 (0.725,0.780), 0.742 (0.699,0.785), 0.733 (0.560,0.905) and 5-year OS were 0.742 (0.711,0.773), 0.746 (0.709,0.783), 0.838 (0.670,0.980), respectively. The calibration curves showed the difference between prediction probability of the model and the actual survival was not significant in three cohorts and the decision curve analysis revealed the practice clinical application value. And the prediction value of model was better for young CRC than older CRC patients. Conclusion A nomogram model including LODDS for the prognosis of advanced CRC received neoadjuvant therapy was constructed and verified based on the SEER database and single center practice. The accuracy and potential clinical application value of the model performed well, and the model had better predictive value for EOCRC than LOCRC.

Abstract This study aimed to address the issue of larger prediction errors existing in intelligent predictive tasks related to Alzheimer’s disease (AD). A cohort of 487 enrolled participants was categorized into three groups: normal control (138 individuals), mild cognitive impairment (238 patients), and AD (111 patients) in this study. An improved multifeature squeeze-and-excitation-dilated residual network (MFSE-DRN) was proposed for two important AD predictions: clinical scores and conversion probability. The model was characterized as three modules: squeeze-and-excitation-dilated residual block (SE-DRB), multifusion pooling (MF-Pool), and multimodal feature fusion. To assess its performance, the proposed model was compared with two other novel models: ranking convolutional neural network (RCNN) and 3D vision geometrical group network (3D-VGGNet). Our method showed the best performance in the two AD predicted tasks. For the clinical scores prediction, the root-mean-square errors (RMSEs) and mean absolute errors (MAEs) of mini-mental state examination (MMSE) and AD assessment scale–cognitive 11-item (ADAS-11) were 1.97, 1.46 and 4.20, 3.19 within 6 months; 2.48, 1.69 and 4.81, 3.44 within 12 months; 2.67, 1.86 and 5.81, 3.83 within 24 months; 3.02, 2.03 and 5.09, 3.43 within 36 months, respectively. At the AD conversion probability prediction, the prediction accuracies within 12, 24, and 36 months reached to 88.0, 85.5, and 88.4%, respectively. The AD predication would play a great role in clinical applications.

Abstract Microbial cell factories serve as pivotal platforms for the production of high-value natural products, which tend to accumulate on the cell membrane due to their hydrophobic properties. However, the limited space of the cell membrane presents a bottleneck for the accumulation of these products. To enhance the production of intracellular natural products and alleviate the burden on the cell membrane caused by product accumulation, researchers have implemented various membrane engineering strategies. These strategies involve modifying the membrane components and structures of microbial cell factories to achieve efficient accumulation of target products. This review summarizes recent advances in the application of membrane engineering technologies in microbial cell factories, providing case studies involving Escherichia coli and yeast. Through these strategies, researchers have not only improved the tolerance of cells but also optimized intracellular storage space, significantly enhancing the production efficiency of natural products. This article aims to provide scientific evidence and references for further enhancing the efficiency of similar cell factories.

Abstract Purpose The purpose of this study was to compare the learning in the implant dentistry hands-on course to that of the flipped classroom (FC) and the traditional lecture cohorts (control). Materials and methods In this study,80 students were enrolled for the first time in an implant dentistry program. Subsequently, they were divided into two groups. The first, the FC group, which had free access to a video with a PowerPoint presentation on the Chaoxing-WHU-MOOC platform about the implant placement on first molar sites before class. The second, the control group, which attended a didactic lecture describing implant practice on the first molar site via a bidirectional multimedia interactive teaching demonstration and then operated on a simulation model. Cone beam computed tomography (CBCT) and the deviation gauge were utilized to analyze the accuracy of the implant placement in the students’ models. An online satisfaction questionnaire was distributed to both groups one week after the class. Results The linear deviation of the CBCT examination did not show any statistical difference between the two groups concerning cervical, apex, and angular. A significant buccal deviation was observed in the control group compared with the FC group (mean: 0.7436 mm vs. 0.2875 mm, p = 0.0035), according to the restoration-level deviation gauge. A total of 74.36% of students in the FC group placed implant within 0.5 mm buccal-to-lingual deviations, but only 41.03% of students in the control group reached within 0.5 mm buccal-to-lingual deviation ranges. Additionally, 91.67% of the students in the FC group and 97.5% of the students in the control group were satisfied with the practical implant class. Conclusion FC was more effective than a didactic lecture for implant dentistry practical skill acquisition.

Abstract Asymptomatic Leucine-Rich Repeat Kinase 2 Gene (LRRK2) carriers are at risk for developing Parkinson's disease (PD). We studied presymptomatic substantia nigra pars compacta (SNc) regional neurodegeneration in asymptomatic LRRK2 carriers compared to idiopathic PD patients using neuromelanin-sensitive MRI technique (NM-MRI). Fifteen asymptomatic LRRK2 carriers, 22 idiopathic PD patients, and 30 healthy controls (HCs) were scanned using NM-MRI. We computed volume and contrast-to-noise ratio (CNR) derived from the whole SNc and the sensorimotor, associative, and limbic SNc regions. An analysis of covariance was performed to explore the differences of whole and regional NM-MRI values among the groups while controlling the effect of age and sex. In whole SNc, LRRK2 had significantly lower CNR than HCs but non-significantly higher volume and CNR than PD patients, and PD patients significantly lower volume and CNR compared to HCs. Inside SNc regions, there were significant group effects for CNR in all regions and for volumes in the associative region, with a trend in the sensorimotor region but no significant changes in the limbic region. PD had reduced volume and CNR in all regions compared to HCs. Asymptomatic LRRK2 carriers showed globally decreased SNc volume and CNR suggesting early nigral neurodegeneration in these subjects at risk of developing PD.

Abstract The North China Plain (NCP) is one of the three great plains in China and also serves as a vital region for grain, cotton, and oil production. Under the influence of regional hydrothermal changes, groundwater overexploitation, and seawater intrusion, the vegetation coverage is undergoing continuous alterations. However, a comprehensive assessment of impacts of precipitation, temperature, and groundwater on vegetation in marine sedimentary regions of the NCP is lacking. Heilonggang Basin (HB) is located in the low-lying plain area in the east of NCP, which is part of the NCP. In this study, the HB was chosen as a typical area of interest. We collected a series of data, including the Normalized Difference Vegetation Index (NDVI), precipitation, temperature, groundwater depth, and Total Dissolved Solids (TDS) from 2001 to 2020. Then the spatiotemporal variation in vegetation was analyzed, and the underlying driving mechanisms of vegetation variation were explored in this paper. The results show that NDVI experiences a rapid increase from 2001 to 2004, followed by stable fluctuations from 2004 to 2020. The vegetation in the HB has achieved an overall improvement in the past two decades, with 76% showing improvement, mainly in the central and eastern areas, and 24% exhibiting deterioration in other areas. From 2001 to 2020, NDVI correlates positively with precipitation, whereas its relationship with temperature fluctuates between positive and negative, and is not statistically significant. There is a threshold for the synergistic change of NDVI and groundwater depth. When the groundwater depth is lower than 3.8 m, NDVI increases sharply with groundwater depth. However, beyond this threshold, NDVI tends to stabilize and fluctuate. In the eastern coastal areas, NDVI exhibits a strong positive correlation with groundwater depth, influenced by the surface soil TDS controlled by groundwater depth. In the central regions, a strong negative correlation is observed, where NDVI is primarily impacted by soil moisture under the control of groundwater. In the west and south, a strong positive correlation exists, with NDVI primarily influenced by the intensity of groundwater exploitation. Thus, precipitation and groundwater are the primary driving forces behind the spatiotemporal variability of vegetation in the HB, while in contrast, the influence of temperature is uncertain. This study has elucidated the mechanism of vegetation response, providing a theoretical basis for mitigating adverse factors affecting vegetation growth and formulating rational water usage regulations in the NCP.

We presented a case of a 66-year-old female whose initial symptom was headache without obvious inducement. The patient's condition progressed rapidly to a semi-coma state after symptomatic treatment. The 18F-FDG PET/CT scan revealed circular FDG hypermetabolism and central metabolic defect of the pons and left frontal lobe lesions. The combination of clinical findings, MRI, and Metagenomic next-generation sequencing (NGS) of cerebrospinal fluid led to the diagnosis of Balamuthia mandrillaris encephalitis. The patient died 5 days after discharge.

Programmed cell death protein-1 (PD-1) inhibitors are increasingly utilized in the treatment of lung cancer (LC). Combination therapy has recently gained popularity in treating LC. This study aimed to assess the efficacy of combining Astragaloside IV (AS-IV) and anti-PD-1 in LC. C57BL/6J mice were subcutaneously injected with Lewis lung carcinoma (LLC) cells. After 3 weeks, the animals were sacrificed, and the tumors were harvested for analysis. Ki-67 immuno-labeling and TUNEL assay were used for evaluating cell proliferation and apoptosis in tumor tissues. In addition, anti-cleaved caspase 3 was used for immunolabelling of apoptotic cells. Immune cell infiltration (macrophages and T cells) and gene expression in tumor tissues were also investigated by using immunofluorescence staining. Compared to treatment with anti-PD-1 or AS-IV, the combination of AS-IV and anti-PD-1 notably reduced tumor volume and weight of LLC-bearing mice. Additionally, the combination treatment strongly induced the apoptosis and suppressed the proliferation in tumor tissues through inactivating PI3K/Akt and ERK signaling pathways, compared to single treatment group. Moreover, the combination treatment elevated levels of the M1 macrophage marker mCD86, reduced levels of the M2 macrophage marker mCD206, as well as upregulated levels of the T cell activation marker mCD69 in tumor tissues. Collectively, the combination treatment effectively inhibited tumor growth in LLC mice through promoting M1 macrophage polarization and T cell activation. These findings showed that combining AS-IV with anti-PD-1 therapy could be a promising therapeutic approach for LC.

Chronic graft-versus-host disease (cGVHD) is one of the leading causes of mortality following allogeneic hematopoietic stem cell transplantation (HSCT), with only 50 % of patients responding to conventional corticosteroids with or without calcineurin inhibitors. Monoclonal antibodies (mAbs), such as CD20 mAbs, are the first drugs to demonstrate greater efficacy than corticosteroids in first-line treatment of cGVHD. This review provides a comprehensive overview of recent developments in the clinical utilization of mAbs to prevent and treat cGVHD. We also describe the application of drugs with target sites that are identical to or linked to those of cGVHD in autoimmune diseases as potential future treatments for cGVHD. In summary, we collected known evidence concerning the clinical research progress on the use of mAbs in the prevention and treatment of cGVHD and identified drugs with the same target sites in autoimmune diseases for potential future applications in cGVHD, with the goal of providing suggestions and inspiration for optimizing cGVHD treatment strategies and research directions.

As an important biomarker of neural aging, the brain age reflects the integrity and health of the human brain. Accurate prediction of brain age could help to understand the underlying mechanism of neural aging. In this study, a cross-stratified ensemble learning algorithm with staking strategy was proposed to obtain brain age and the derived predicted age difference (PAD) using T1-weighted magnetic resonance imaging (MRI) data. The approach was characterized as by implementing two modules: one was three base learners of 3D-DenseNet, 3D-ResNeXt, 3D-Inception-v4; another was 14 secondary learners of liner regressions. To evaluate performance, our method was compared with single base learners, regular ensemble learning algorithms, and state-of-the-art (SOTA) methods. The results demonstrated that our proposed model outperformed others models, with three metrics of mean absolute error (MAE), root mean-squared error (RMSE), and coefficient of determination (R2) of 2.9405 years, 3.9458 years, and 0.9597, respectively. Furthermore, there existed significant differences in PAD among the three groups of normal control (NC), mild cognitive impairment (MCI) and Alzheimer's disease (AD), with an increased trend across NC, MCI, and AD. It was concluded that the proposed algorithm could be effectively used in computing brain aging and PAD, and offering potential for early diagnosis and assessment of normal brain aging and AD.

The quality of strong-flavor Baijiu, a prominent Chinese liquor, is intricately tied to the choice of sorghum variety used in fermentation. However, a significant gap remains in our understanding of how glutinous and non-glutinous sorghum varieties comprehensively impact Baijiu flavor formation through fermentation metabolites. This study employed untargeted metabolomics combined with feature-based molecular networking (FBMN) to explore the unique metabolic characteristics of these two sorghum varieties during fermentation. FBMN analysis revealed 267 metabolites within both types of fermented sorghum (Zaopei) in the cellar. Further multidimensional statistical analyses highlighted sphingolipids, 2,5-diketopiperazines, and methionine derivatives as critical markers for quality control. These findings represent a significant advancement in our understanding and provide valuable insights for regulating the quality of Baijiu flavors.

Background: Extranodal natural killer/T-cell lymphoma (ENKTCL) has a unique treatment principle. However, the optimal combination of drugs along with radiotherapy (RT) is unknown. Design: Retrospective cohort study. Objectives: We screened multiple drug combinations to identify the most efficacious therapeutic combinations. Methods: We reviewed 3105 patients who received 40 chemotherapy regimens with different combinations of 9 drug classes and/or RT. Least absolute shrinkage and selection operator and multivariable Cox regression analyses were used to screen efficacious single drugs and identify optimal combinations for overall survival (OS). Inverse probability of treatment weighting (IPTW) and multivariable analyses were used to compare survival between treatment regimens. Results: Screening and validation revealed RT, asparaginase (ASP), and gemcitabine (GEM) to be the most efficacious single modality/drug. RT remained an important component of first-line treatment, whereas ASP was a fundamental drug of non-anthracycline (ANT)-based regimens. Addition of RT to non-ANT-based or ASP/GEM-based regimens, or addition of an ASP-drug into ANT-based or GEM/platinum-based regimens, improved 5-year OS significantly. Use of ASP/GEM-based regimens was associated with significantly higher 5-year OS (79.9%) compared with ASP/ANT-based (69.2%, p = 0.001), ASP/methotrexate-based (63.5%, p = 0.011), or ASP/not otherwise specified-based (63.2%, p

Background The main challenge against patients with cancer to derive benefits from immune checkpoint inhibitors targeting PD-1/PD-L1 appears to be the immunosuppressive tumor microenvironment (TME), in which IL-33/ST2 signal fulfills critical functions. However, whether IL-33 limits the therapeutic efficacy of anti-PD-L1 remains uncertain.Methods Molecular mechanisms of IL-33/ST2 signal on anti-PD-L1 treatment lewis lung carcinoma tumor model were assessed by RNA-seq, ELISA, WB and immunofluorescence (IF). A sST2-Fc fusion protein was constructed for targeting IL-33 and combined with anti-PD-L1 antibody for immunotherapy in colon and lung tumor models. On this basis, bifunctional fusion proteins were generated for PD-L1-targeted blocking of IL-33 in tumors. The underlying mechanisms of dual targeting of IL-33 and PD-L1 revealed by RNA-seq, scRNA-seq, FACS, IF and WB.Results After anti-PD-L1 administration, tumor-infiltrating ST2+ regulatory T cells (Tregs) were elevated. Blocking IL-33/ST2 signal with sST2-Fc fusion protein potentiated antitumor efficacy of PD-L1 antibody by enhancing T cell responses in tumor models. Bifunctional fusion protein anti-PD-L1-sST2 exhibited enhanced antitumor efficacy compared with combination therapy, not only inhibited tumor progression and extended the survival, but also provided long-term protective antitumor immunity. Mechanistically, the superior antitumor activity of targeting IL-33 and PD-L1 originated from reducing immunosuppressive factors, such as Tregs and exhausted CD8+ T cells while increasing tumor-infiltrating cytotoxic T lymphocyte cells.Conclusions In this study, we demonstrated that IL-33/ST2 was involved in the immunosuppression mechanism of PD-L1 antibody therapy, and blockade by sST2-Fc or anti-PD-L1-sST2 could remodel the inflammatory TME and induce potent antitumor effect, highlighting the potential therapeutic strategies for the tumor treatment by simultaneously targeting IL-33 and PD-L1.

Summary: Building a simple and efficient in vitro differentiation system is crucial for studying the regulatory mechanisms during the development of innate lymphoid cells (ILCs). Here, we present a protocol for generating ILC subsets from α4β7+ lymphoid progenitors (αLPs). We describe steps for murine cell isolation from fetal liver and adult bone marrow, flow cytometry sorting for αLPs, and cell culture. We then detail procedures for flow cytometry analysis of ILCs. Abstract MXene is investigated as an electrode material for different energy storage systems due to layered structures and metal‐like electrical conductivity. Experimental results show MXenes possess excellent cycling performance as anode materials, especially at large current densities. However, the reversible capacity is relatively low, which is a significant barrier to meeting the demands of industrial applications. This work synthesizes N‐doped graphene‐like carbon (NGC) intercalated Ti3C2Tx (NGC‐Ti3C2Tx) van der Waals heterostructure by an in situ method. The as‐prepared NGC‐Ti3C2Tx van der Waals heterostructure is employed as sodium‐ion and lithium‐ion battery electrodes. For sodium‐ion batteries, a reversible specific capacity of 305 mAh g−1 is achieved at a specific current of 20 mA g−1, 2.3 times higher than that of Ti3C2Tx. For lithium‐ion batteries, a reversible capacity of 400 mAh g−1 at a specific current of 20 mA g−1 is 1.5 times higher than that of Ti3C2Tx. Both sodium‐ion and lithium‐ion batteries made from NGC‐Ti3C2Tx shows high cycling stability. The theoretical calculations also verify the remarkable improvement in battery capacity within the NGC‐Ti3C2O2 system, attributed to the additional adsorption of working ions at the edge states of NGC. This work offers an innovative way to synthesize a new van der Waals heterostructure and provides a new route to improve the electrochemical performance significantly.

Abstract Non‐alcoholic fatty liver disease (NAFLD) is a prominent cause of various chronic metabolic hepatic diseases with limited therapeutics. Rubicon, an essential regulator in lysosomal degradation, is reported to exacerbate hepatic steatosis in NAFLD mice and patients, indicating its probability of being a therapeutic target for NAFLD treatment. In this study, the therapeutic potential of Rubicon blockage is investigated. Lipid nanoparticles carrying Rubicon‐specific CRISPR‐Cas9 components exhibited liver accumulation, cell internalization, and Rubicon knockdown. A single administration of the nanoparticles results in attenuated lipid deposition and hepatic steatosis, with lower circulating lipid levels and decreased adipocyte size in NAFLD mice. Furthermore, the increase of phosphatidylcholine and phosphatidylethanolamine levels can be observed in the NAFLD mice livers after Rubicon silencing, along with regulatory effects on metabolism‐related genes such as CD36, Gpcpd1, Chka, and Lpin2. The results indicate that knockdown of Rubicon improves glycerophospholipid metabolism and thereby ameliorates the NAFLD progression, which provides a potential strategy for NAFLD therapy via the restoration of Rubicon.

This study aims to elucidate the mechanism underlying the reduced jejunal injury in PEDV-challenged piglets after oral administration of puerarin and poria cocos polysaccharides combination (PR + PCP, PP). Results showed that the PEDV challenge caused damage to the small intestine by injuring the villi and inducing redox imbalance which exacerbated the diarrhea of piglets, while PP administration trended to reduce the PEDV infection-caused diarrhea in piglets by alleviating the jejunal injury and restoring the redox balance. Further proteomics and qPCR results showed that PP could significantly reduce the decline of the protein and mRNA levels of gene SDSL inhibiting cellular autophagy in PEDV-infected jejunum of piglets. The immunofluorescence results showed that the autophagy levels of the jejunum expressing low SDSL caused by PEDV infection in the PEDV group were significantly higher than those of the jejunum expressing higher levels of SDSL caused by PP administration in the PP + PEDV group. These results showed that PEDV infection caused jejunal excessive autophagy in piglets by reducing the expression level of SDSL. However, the restoration of the expression level of SDSL significantly reduced the PEDV infection-caused jejunal excessive autophagy in piglets, leading to a decrease in jejunal injury. Therefore, PP administration can alleviate the excessive autophagy-caused jejunal injury by increasing SDSL levels in PEDV-infected piglets. This result suggests that PP can be used as a potential feed additive to prevent PEDV infection.

Hydrogen sulfide (H2S) has recently been recognized as an important gaseous transmitter with multiple physiological effects in various species. Previous studies have shown that H2S alleviated heat-induced ganoderic acids (GAs) biosynthesis, an important quality index of Ganoderma lucidum. However, a comprehensive understanding of the physiological effects and molecular mechanisms of H2S in G. lucidum remains unexplored. In this study, we found that heat treatment reduced the mitochondrial membrane potential (MMP) and mitochondrial DNA copy number (mtDNAcn) in G. lucidum. Increasing the intracellular H2S concentration through pharmacological and genetic means increased the MMP level, mtDNAcn, oxygen consumption rate level and ATP content under heat treatment, suggesting a role for H2S in mitigating heat-caused mitochondrial damage in G. lucidum. Further results indicated that H2S activates sulfide-quinone oxidoreductase (SQR) and complex III (Com III), thereby maintaining mitochondrial homeostasis under heat stress in G. lucidum. Moreover, SQR also mediated the negative regulation of H2S to GAs biosynthesis under heat stress. Furthermore, SQR might be persulfidated under heat stress in G. lucidum. Thus, our study reveals a novel physiological function and molecular mechanism of H2S signalling under heat stress in G. lucidum with broad implications for research on the environmental response of microorganisms.

Abstract Background Two-tiered plant immune responses involve cross-talk among defense-responsive (DR) genes involved in pathogen-associated molecular pattern (PAMP)-triggered immunity (PTI), effector-triggered immunity (ETI) and effector-triggered susceptibility (ETS). Bacterial leaf streak (BLS), caused by Xanthomonas oryzae pv. oryzicola (Xoc) is an important bacterial disease that causes serious threats to rice yield and quality. Transcriptomic profiling provides an effective approach for the comprehensive and large-scale detection of DR genes that participate in the interactions between rice and Xoc. Results In this study, we used RNA-seq to analyze the differentially expressed genes (DEGs) in susceptible rice after inoculation with two naturally pathogenic Xoc strains, a hypervirulent strain, HGA4, and a relatively hypovirulent strain, RS105. First, bacterial growth curve and biomass quantification revealed that differential growth occurred beginning at 1 day post inoculation (dpi) and became more significant at 3 dpi. Additionally, we analyzed the DEGs at 12 h and 3 days post inoculation with two strains, representing the DR genes involved in the PTI and ETI/ETS responses, respectively. Gene Ontology (GO) functional and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analyses were performed on the common DEGs, which included 4380 upregulated and 4019 downregulated genes and 930 upregulated and 1383 downregulated genes identified for the two strains at 12 h post inoculation (hpi) and 3 dpi, respectively. Compared to those at 12 hpi, at 3 dpi the number of common DEGs decreased, while the degree of differential expression was intensified. In addition, more disease-related GO pathways were enriched, and more transcription activator-like effector (TALE) putative target genes were upregulated in plants inoculated with HGA4 than in those inoculated with RS105 at 3 dpi. Then, four DRs were randomly selected for the BLS resistance assay. We found that CDP3.10, LOC_Os11g03820, and OsDSR2 positively regulated rice resistance to Xoc, while OsSPX3 negatively regulated rice resistance. Conclusions By using an enrichment method for RNA-seq, we identified a group of DEGs related to the two stages of response to the Xoc strain, which included four functionally identified DR genes.

Abstract Background Teaching assistants (TAs) play a crucial role in pedagogical practices, and the TA training has emerged as a vital strategy for enhancing teaching quality and fostering effective interactions. The self-efficacy of TAs can substantially impact their performance. Nevertheless, little research has focused on the change in TAs’ self-efficacy following their training. Methods A self-control quasi-experiment was conducted to examine shifts in the self-efficacy of Tas at Peking University before and after their TA training. A questionnaire was used to assess the change, and the reliability and validity of the questionnaire was also calculated. A paired data rank sum test was used to analysis the changes in TA self-efficacy before and after training. Results A total of 372 TAs from School of Basic Medicine (N = 173), School of Pharmacy (N = 112), School of Public Health (N = 69), and other schools (N = 18) submitted complete questionnaires. The questionnaire showed a good performance in internal reliability and validity test (Cronbach’s alpha index = 0.906, and KMO value was 0.903). Abstract Urban water bodies can effectively mitigate the urban heat island effect and thus enhance the climate resilience of urban areas. The cooling effect of different water bodies varies, however, the cooling heterogeneity of different sections of a single watercourse or river network is rarely considered. Based on various satellite images, geospatial approaches and statistical analyses, our study confirmed the cooling heterogeneity from spatial and seasonal perspectives of the Suzhou Outer-city River in detail in the urban area of Suzhou, China. The cooling effect of the river was observed in the daytime in four seasons, and it is strongest in summer, followed by spring and autumn, and weakest in winter. The combination of the width of the river reach, the width and the NDVI value of the adjacent green space can explain a significant part of the cooling heterogeneity of the different river sections in different seasons. Land surface temperature (LST) variations along the river are more related to the width of the river reach, but the variations of the cooling distance are more related to the adjacent green space. The cooling effect of a river reach could be enhanced if it is accompanied by green spaces. In addition, the cooling effect of a looping river is stronger on the inside area than on the outside. The methodology and results of this study could help orient scientific landscape strategies in urban planning for cooler cities.

Abstract Since the 1980s, low-permeability slurry trench cutoff walls have been widely constructed as barriers to retard the migration of contaminants. The thickness of the cutoff walls is a key determinant of the wall service life. Through a series of theoretical derivations, simplified methods for determining the flux limit and concentration limit were proposed to determine the thickness of cutoff walls for contaminated sites with constant pollutant flux. The relative errors of both the flux-based and concentration-based methods increase as the breakthrough criterion of the ratio between the specified limit concentration of the contaminant to the source concentration (C *) and the ratio of the limited value of contaminant flux to the constant source flux (F *) increases, with a given Peclet number P L. The maximum relative error reaches 4% and 6% when C * and F * are both 0.1, which covers most practical situations in cutoff wall design. Good agreements of wall thickness were obtained between the proposed simplified methods and analytical solutions via a clear example. The proposed method can efficiently simplify the design process of cutoff walls with high accuracy, providing a basis for containing contaminated sites.

Abstract Fat loss predicts adverse outcomes in advanced heart failure (HF). Disrupted circadian clocks are a primary cause of lipid metabolic issues, but it's unclear if this disruption affects fat expenditure in HF. To address this issue, we investigated the effects of disruption of the BMAL1/REV-ERBα circadian rhythmic loop on adipose tissue metabolism in HF.50 Wistar rats were initially divided into control (n = 10) and model (n = 40) groups. The model rats were induced with HF via monocrotaline (MCT) injections, while the control group received equivalent solvent injections. After establishing the HF model, the model group was further subdivided into four groups: normal rhythm (LD), inverted rhythm (DL), lentivirus vector carrying Bmal1 short hairpin RNA (LV-Bmal1 shRNA), and empty lentivirus vector control (LV-Control shRNA) groups, each with 10 rats. The DL subgroup was exposed to a reversed light–dark cycle of 8 h: 16 h (dark: light), while the rest adhered to normal light–dark conditions (light: dark 12 h: 12 h). Histological analyses were conducted using H&E, Oil Red O, and Picrosirius red stains to examine adipose and liver tissues. Immunohistochemical staining, RT-qPCR, and Western blotting were performed to detect markers of lipolysis, lipogenesis, and beiging of white adipose tissue (WAT), while thermogenesis indicators were detected in brown adipose tissue (BAT). The LD group rats exhibited decreased levels of BMAL1 protein, increased levels of REV-ERBα protein, and disrupted circadian circuits in adipose tissue compared to controls. Additionally, HF rats showed reduced adipose mass and increased ectopic lipid deposition, along with smaller adipocytes containing lower lipid content and fibrotic adipose tissue. In the LD group WAT, expression of ATGL, HSL, PKA, and p-PKA proteins increased, alongside elevated mRNA levels of lipase genes (Hsl, Atgl, Peripilin) and FFA β-oxidation genes (Cpt1, acyl-CoA). Conversely, lipogenic gene expression (Scd1, Fas, Mgat, Dgat2) decreased, while beige adipocyte markers (Cd137, Tbx-1, Ucp-1, Zic-1) and UCP-1 protein expression increased. In BAT, HF rats exhibited elevated levels of PKA, p-PKA, and UCP-1 proteins, along with increased expression of thermogenic genes (Ucp-1, Pparγ, Pgc-1α) and lipid transportation genes (Cd36, Fatp-1, Cpt-1). Plasma NT-proBNP levels were higher in LD rats, accompanied by elevated NE and IL-6 levels in adipose tissue. Remarkably, morphologically, the adipocytes in the DL and LV-Bmal1 shRNA groups showed reduced size and lower lipid content, while lipid deposition in the liver was more pronounced in these groups compared to the LD group. At the gene/protein level, the BMAL1/REV-ERBα circadian loop exhibited severe disruption in LV-Bmal1 shRNA rats compared to LD rats. Additionally, there was increased expression of lipase genes, FFA β oxidation genes, and beige adipocyte markers in WAT, as well as higher expression of thermogenic genes and lipid transportation genes in BAT. Furthermore, plasma NT-proBNP levels and adipose tissue levels of NE and IL-6 were elevated in LV-Bmal1 shRNA rats compared with LD rats. The present study demonstrates that disruption of the BMAL1/REV-ERBα circadian rhythmic loop is associated with fat expenditure in HF. This result suggests that restoring circadian rhythms in adipose tissue may help counteract disorders of adipose metabolism and reduce fat loss in HF.

Abstract Named Entity Recognition (NER) in cybersecurity is crucial for mining information during cybersecurity incidents. Current methods rely on pre-trained models for rich semantic text embeddings, but the challenge of anisotropy may affect subsequent encoding quality. Additionally, existing models may struggle with noise detection. To address these issues, we propose JCLB, a novel model that Joins Contrastive Learning and Belief rule base for NER in cybersecurity. JCLB utilizes contrastive learning to enhance similarity in the vector space between token sequence representations of entities in the same category. A Belief Rule Base (BRB) is developed using regexes to ensure accurate entity identification, particularly for fixed-format phrases lacking semantics. Moreover, a Distributed Constraint Covariance Matrix Adaptation Evolution Strategy (D-CMA-ES) algorithm is introduced for BRB parameter optimization. Experimental results demonstrate that JCLB, with the D-CMA-ES algorithm, significantly improves NER accuracy in cybersecurity.

Abstract The growth of multimedia applications poses new challenges to software facilities in edge computing. Developers must effectively develop edge computing software to accommodate the rapid expansion of multimedia applications. Code search has become a prevalent practice to enhance the efficiency of the construction of edge software infrastructure. Researchers have proposed lots of approaches for code search, and employed deep learning technology to extract features from program representations, such as token, AST, graphs, method name, and API. Nevertheless, two prominent issues remain: 1) there are only a few studies on the effective use of graph representation for code search (especially in Java language), and 2) there is a lack of empirical study on the contributions of different program representations. To address these issues, we conduct an empirical study to explore program representations, especially program graphs. To the best of our knowledge, this is the first attempt to conduct code search with mixed graphs representation for Java language, containing the control flow graph and the program dependence graph. We also present a hybrid approach to capture and fuse the features of a program with representations of Token, AST, and Mixed Graphs (TAMG). The results of our experiment show that our approach possesses the best ability (R@1 with 37% and R@10 with 67.1%). Our graph representation exhibits a positive effect, and the token and AST also have a significant contribution to the code search. Our findings can aid developers in efficiently searching for the desired code while constructing the software infrastructure for edge computing, which is crucial for the rapid expansion of multimedia applications.

Abstract Two-dimensional (2D) magnetic materials are of not only fundamental scientific interest but also promising candidates for numerous applications. However, so far only a few intrinsic magnets with long-ranged order down to the 2D limit have been experimentally established. Here, we report that the intrinsic 2D ferromagnetism can be realized in van der Waals (vdW) Fe3GaTe2 nanoflake down to monolayer. By measuring the Hall resistance and magnetoresistance, we demonstrate that the Fe3GaTe2 monolayer exhibits 2D hard ferromagnetism with record-high Cure temperature (T c) of 240 K for the monolayer of known intrinsic ferromagnets. Both of square-shaped hysteresis loops with near-vertical jump in anomalous Hall effect (AHE) and the negative magnetoresistance (NMR) behavior with an applied out-of-plane magnetic field reveal robust perpendicular magnetic anisotropy (PMA) in Fe3GaTe2 nanoflakes down to the monolayer limit. Furthermore, we find the intrinsic mechanism that stems from the Berry curvature of electronic bands dominates AHE of nanoflakes in the low temperature range. Our results not only provide an excellent candidate material for next-generation spintronic applications, but also open up a platform for exploring physical mechanisms in 2D ferromagnetism.

Abstract Background Homegardens are small-scale land use systems with significant implications for local livelihoods, biodiversity conservation, and sustainable development due to their diverse flora and fauna. Conducting homegarden surveys enables researchers to gain insights into the traditional ecological knowledge of indigenous communities, preserve homegarden biodiversity, and promote sustainable livelihoods. This study has two objectives: first, to collect, record, and organize data on homegarden plants and related traditional knowledge from three communities in the Laershan Plateau, and second, to analyze the species and functional diversity of homegarden plants in the region while exploring the factors that contribute to the heterogeneous distribution of these plants among the communities of three townships. Methods This research employed field surveys in the Laershan Miao Autonomous Region in Xiangxi, China, from September 2021 to August 2023. Data were collected through observation and semi-structured interviews. The study utilized descriptive statistics and quantitative analysis, including the Relative Importance Value (RI), Use Value Index (UV), Jaccard Index (JI), and Comprehensive Index of Land Use Degree (La), to examine the diversity of local homegarden plants and related traditional knowledge, as well as community heterogeneity. Results The study area exhibited rich biodiversity, with 152 plant species recorded belonging to 62 families and 124 genera. These findings highlight the importance of homegarden plants in maintaining ecological balance and enhancing system resilience against disturbances. Homegarden plants serve multiple functions such as food, ornamental, medicinal, and fodder purposes, with edible and trade plants having the most abundant knowledge. Furthermore, this research revealed that communities with similar forest resource compositions may have similar homegarden plant compositions, demonstrating the connection between cultural exchange among different communities and patterns of plant species distribution. Conclusions The Laershan region boasts diverse homegarden plant species crucial for ecological balance and resilience. Their multifunctionality reflects human impact. Plant diversity varies with residents' lifestyles, needs, and cultural exchanges. The close connection between plants and local life is clear. Factors like resource distribution, cultural exchange, and lifestyle influence plant distribution. Further research is needed for conservation and sustainable development. Future research should focus on culturally valuable plant species and traditional knowledge applications.

Abstract Forecasting all components in complex systems is an open and challenging task, possibly due to high dimensionality and undesirable predictors. We bridge this gap by proposing a data-driven and model-free framework, namely, feature-and-reconstructed manifold mapping (FRMM), which is a combination of feature embedding and delay embedding. For a high-dimensional dynamical system, FRMM finds its topologically equivalent manifolds with low dimensions from feature embedding and delay embedding and then sets the low-dimensional feature manifold as a generalized predictor to achieve predictions of all components. The substantial potential of FRMM is shown for both representative models and real-world data involving Indian monsoon, electroencephalogram (EEG) signals, foreign exchange market, and traffic speed in Los Angeles Country. FRMM overcomes the curse of dimensionality and finds a generalized predictor, and thus has potential for applications in many other real-world systems.

This paper investigated the improvement behaviors on dispersivity, water stability and mechanical properties of dispersive soil by calcined coal gangue (CCG) at 700 °C, and analyzed the modification mechanism. Dispersive soil specimens with different content of CCG (varying from 1 % to 10 %) were prepared and cured for 0–28 days. The dispersivity of the soil was determined by three different dispersivity determination tests. The tensile strength and compressive strength of the dispersive soil were determined by mechanical property tests. SEM, EDS, TG and XRD analytical methods were employed to reveal microstructure and mineral changes during modification. The results of the study show that the admixture of CCG and the prolongation of curing time contributed favorably to suppressing the dispersivity of the soil and enhancing the water stability, the compressive strength and tensile strength of the dispersive soil. With the increasing of CCG content and the prolongation of curing time, the dispersive soil gradually transforms into non-dispersive soil. Microstructural and mineral analysis indicate that CCG has pozzolanic activity, and the production of pozzolanic reaction products significantly increase the friction and cohesion among soil particles. The results show that the utilization of CCG as an admixture to improve the dispersive soil not only solves the disposal problem of waste gangue, but also optimizes the undesirable characteristics of the dispersive soil. And the modification effect of CCG on dispersive soil in practical engineering is confirmed by validation test.

P110 steel, renowed for its good comprehensive mechanical properties, is widely used as casing in oil and gas wells. However, it is seriously worn when using cemented carbide tools to drill it underwater. The study delves into the experimental exploration of the wear ratio of P110 steel during underwater drilling using three different kinds of super-hard abrasive grains and binders. The investigation includes tracking and observing the morphological changes of abrasive grains on the core drill's surface, along with the analysis of graphitization on the surface of diamond abrasive grains. The results show that under the same processing conditions, the end wear of the three different core drills is roughly the same. However, the side wear of the core drill using diamond abrasive particles is significantly less than that using cubic Boron Nitride (cBN) abrasive particles. Under the condition of underwater machining, the end face diamond abrasive grains will produce graphitization. Despite this, the diamond abrasive grains still maintain a certain cutting ability. The hardness of the bonding agent affects the exposure of abrasive particles, which results in the difference in the performance of the core drill under different working conditions.

Objective: Few studies use all nodule burdens to specify the prognosis of multinodular goiter (MNG) following radiofrequency ablation (RFA), so this study addresses this question for MNG after completely ablating dominant nodules. Methods: The RFA indications for MNG include 2–5 benign nodules with over 50% normal tissue on ultrasound, 1–3 well-defined benign dominant nodules on cytology, largest diameter ≥20 mm and/or with clinical complaints, and patient refusal or unable to undergo surgery. A retrospective study of 185 MNG patients with completely ablated dominant nodules in a single-session RFA was conducted. The efficacy and complications were evaluated at 1, 6, 12 months, and yearly thereafter. Based on retreatment risks, progressive disease (PD), stable disease (SD), and complete relief (CR) were introduced to assess all nodule load changes. PD was clarified as having new/non-target nodules that newly appeared to ACR TI-RADS≥4, or new/enlarged non-target nodules ≥1 cm. Results: The initial ablation ratios of target nodules were 100% at one month. During a mean 22.38 ± 13.75 months (range, 12–60 months), the volume reduction rate of ablated nodules was 98.25% at 24 months without regrowth. Cosmetic and symptomatic scores decreased to 1 and 0, respectively, after 48 months. Of the patients, 9.7% (18/185) had PD and the retreatment rate was 2.2% (4/185). The complication rate was 2.7% (5/185). Conclusion: RFA provides cosmetic and symptomatic relief for an average of two years. RFA is a useful minimally invasive treatment modality for selected MNG patients.

Background: Since the end of the COVID-19 pandemic, the potential roles of thyroid-inflammatory derangements in driving or being associated with the prognosis of COVID-19 remain controversial. We aimed to clarify the association between COVID-19 infection and thyroid dysfunction, and highlight the impacts of subsequent autoimmune thyroid disease (AITD) on the prognosis of COVID-19. Methods: The retrospective, multicenter, cohort study enrolled 2,339 participants with COVID-19 from three hospitals located in the north, middle, and south regions of Shaan Xi Province, China, between December 2022 and July 2023. 464 non-COVID-19 patients within the same period were supplemented, divided into groups with and without AITD. At hospital admission (baseline), 3- and 6-month follow-ups, we presented a dynamic description and correlation analysis of thyroid-inflammatory-autoimmune derangements in patients with AITD. Results: A total of 2,082 COVID-19 patients diagnosed with AITD and 257 cases without AITD were included in the study, and 464 non-COVID-19 patients were supplemented, dividing into 14 AITD and 450 non-AITD cases. We found that COVID-19 infection was closely associated with thyroid dysfunction (χ2 = 1518.129, p = 0.000). AITD patients with COVID-19 showed a higher prevalence of symptoms and comorbidities and longer hospital stays at baseline than non-AITD patients with COVID-19 (p = 0.000, p = 0.000, and p = 0.000). The baseline free triiodothyronine (FT3), free thyroxine, and radioactive iodine uptake at 24 h in AITD cases significantly decreased (p = 0.000, p = 0.000, and p = 0.000), while thyroid stimulating hormone, thyroglobulin, reverse triiodothyronine (rT3), and thyroid antibodies varying elevated from the baseline to the follow-up (baseline: p = 0.000, p = 0.000, p = 0.000, p = 0.000, p = 0.000, and p = 0.000; 3-month follow-up: p = 0.000, p = 0.000, p = 0.000, p = 0.000, p = 0.030, and p = 0.000). C-reactive protein, calcitonin, interleukin-6, -8, -10, and tumor necrosis factor-α rose significantly at baseline (p = 0.000, p = 0.000, p = 0.000, p = 0.000, p = 0.000, and p = 0.000) in AITD. Interferon-α and interferon-γ at baseline showed a significant decrease (p = 0.000 and p = 0.000), and remained at low levels after 6 months (p = 0.000 and p = 0.000). FT3 and rT3 were positively and negatively correlated with hospitalization, respectively (r = -0.208 and 0.231; p = 0.000 and p = 0.000). ROC curves showed that FT3 and rT3 had better robustness in predicting severe COVID-19 prognosis (AUC = 0.801 and 0.705). Ordered logistic regression revealed that ORs were 0.370, 0.048, and 0.021 for AITD [(subacute thyroiditis, Grave's disease, and Hashimoto's thyroiditis compared to non-thyroidal illness syndrome (NTIS)] with COVID-19 risk, indicating that NTIS was the predominant risk factor for the severity of COVID-19. Conclusions: A robust association has been identified, wherein COVID-19 infection is closely associated with thyroid dysfunction, and the subsequent AITD may aggravate the poor prognosis of COVID-19.
