Food and dairy industries play a very important role in the economy of every country. Aspartic proteases are important enzyme of dairy industry and is used in cheese making. Previously main sources of protease enzyme were plants, animal or fungi, but due to increased demand globally they are now mostly isolated from bacteria. Objectives: To isolate the milk clotting bacteria from the soil collected from dumping site of Mehmood Booti and produce aspartic protease from them. Methods: Soil sample was collected from Mahmood Booti dumping site near ring road, Lahore. After serial dilutions, sample was inoculated on nutrient agar plates. After 24 hours at 37°C temperature, opaque, round and cream-colored colonies were observed which were sub cultured in LB agar. From there colonies were grown on selective medium made of K2HPO4, (NH4)2 HPO4, casein, MgCl2, yeast extract and agar. After incubation, a colony with clear zone was selected and grown in LB broth for enzyme production. After incubation, broth was centrifuged and supernatant was isolated. While performing protease assay, 3 mL of 5% TCA was added in the mixture. Results: The mixture remained clear which depicted the hydrolysis of casein by protease. While the test tube containing water as blank showed precipitation of casein after the addition of TCA because in this enzyme was not present. Conclusions: This shows that the isolated bacteria had the ability to produce protease which was evident from the protease activity assay and that such bacteria are abundant in dumping site

Bioremediation involves the use of natural microorganisms for the purpose ofdegrading numerous types of industrial and environmental waste. Microorganismsrequire carbon, nutrients, and energy to live and multiply as all living organisms do. Inorder to obtain energy, such microorganisms break down organic pollutants into simplerorganic compounds like carbon, salts, water, and similar harmless products. Thisapproach of degrading contaminants using microorganisms has proved much beneficialand has been proven to be cost-effective and efficient. There are a lot of naturally occurringmicroorganisms that have been reported essential in the degradation of organic pollutants.Different industries use different types of bioremediation methods. Specificenvironmental conditions may be required for optimal functioning of microbes e.g., pH,temperature, humidity, etc. Bioremediation has been proven as an environment-friendlyand cost-effective approach to deal with industrial contaminants. Descriptive informationof microbes involved in bioremediation has been explained in this review.

Autoimmune disorder is a chronic immune imbalance which is developed through a series of pathways. The defect in B cells, T cells, and lack of self-tolerance has been greatly associated with the onset of many types of autoimmune complications including rheumatoid arthritis, systemic lupus erythematosus (SLE), multiple sclerosis, and chronic inflammatory demyelinating polyneuropathy. The SLE is an autoimmune disease with a common type of lupus that causes tissue and organ damage due to the wide spread of inflammation. In the current study, twenty anti-inflammatory peptides derived from plant and animal sources were docked as ligands or peptides counter to proinflammatory cytokines. Interferon gamma (IFN-γ), interleukin 3 (IL-3), and tumor necrosis factor alpha (TNF-α) were targeted in this study as these are involved in the pathogenesis of SLE in many clinical studies. Two docking approaches (i.e., protein-ligand docking and peptide-protein docking) were employed in this study using Molecular Operating Environment (MOE) software and HADDOCK web server, respectively. Amongst docked twenty peptides, the peptide DEDTQAMMPFR with S -score of -11.3018 and HADDOCK score of − 10.3 ± 2.5 kcal/mol showed the best binding interactions and energy validation with active amino acids of IFN-γ protein in both docking approaches. Depending upon these results, this peptide could be used as a potential drug candidate to target IFN-γ, IL-3, and TNF-α proteins to control inflammatory events. Other peptides (i.e., QEPQESQQ and FRDEHKK) also revealed good binding affinity with IFN-γ with S -scores of -10.98 and -10.55, respectively. Similarly, the peptides KHDRGDEF, FRDEHKK, and QEPQESQQ showed best binding interactions with IL-3 with S -scores of -8.81, -8.64, and -8.17, respectively.

Friction stir welding (FSW) in butt joint configuration between 6.35 mm thick plates of AA2024-T3 and 2.5 mm thick plates of AA7475-T7 dissimilar alloys is addressed using T4 tool steel with a tapered cylindrical pin having scrolled shoulder. In this experimental study, the effect of double-pass FSW on joint properties has been investigated in detail. The weld microstructure and mechanical properties of FSWed joints were characterized. Scanning electron microscopy and x-ray energy-dispersive spectroscopy were employed to analyze the distribution and nature of precipitates. The temperature rise and traverse load during the FSW process were evaluated. Experimental results indicated the significant increase in grain size by double-pass FSW. Moreover, the hardness value decreases with the second pass, and the hardness dropped significantly in the heat-affected zone due to grains coarsening and change in precipitate morphology caused by increased heat input by double-pass FSW. The overall reduction in joint strength was found to be 4.8% by second pass FSW.

Although the various advantages of novel Friction stir welding (FSW) process; the weld surfaces are subjected to various serious problems such as lower corrosion resistance, high susceptibility to stress corrosion cracking and poor joint fatigue strength due to complex material flow and severe plastic deformation during the welding process. Corrosion behaviour of friction stir welded (FSWed) precipitate strengthening Al alloys have significant impact on metallurgical and electrochemical properties of structures. In FSW of precipitate strengthening Al alloys the localized heat input and severe plastic deformation creating appreciable changes in microstructure and modifies the microchemistry and metallurgical characteristics of precipitates. The heterogeneous distribution of precipitates and precipitate free zones (PFZs) along the grain boundaries leads to variation in electrochemical behaviour across the weld zones and hence increasing the susceptibility of weld surface to various corrosion attacks such as intergranular corrosion, pitting corrosion, exfoliation corrosion, stress corrosion cracking and galvanic corrosion. However, the corrosion resistance of FSWed joints can be improved either by reducing the size or redissolve the coarsened precipitates within the joint or modifying the microchemistry by controlling the size, location and distribution of precipitates which largely determine the corrosion rate of the weld surfaces. Consequently, it is imperative to address the influence of material modifications during FSW on corrosion behaviour of weld surface. This review paper addresses the precipitate dependent corrosion behaviour of FSWed joints of heat treatable/precipitate strengthening Al alloys and the various effective methods either to reduce or eliminate the effect of corrosion attack in FSWed joints of heat treatable Al alloys.

Women in Pakistan are obsessed with the concept of glowing whitish complexion to get good marriage proposals, jobs and desired status in society. Most of these fairness creams contain very high amount of mercury and hydroquinone. These act by inhibiting the tyrosinase enzyme responsible for melanin production. The use of skin fairness products that frequently contain toxic ingredients is associated with significant adverse health side effects, resulting in immense challenges for dermatologists. Despite current regulations, lightening agents continue to dominate the cosmetic industry. In this review, our international team of dermatologists tackles the topic of skin lightening as a global public health issue, one of great concern for both women’s health and racial implications. We aim to inspire a global discourse on how modern dermatologists can utilize scientific evidence and cultural competency to serve and protect patients of diverse skin types and backgrounds. In doing so, we hope to promote healthy skin and inclusive concepts of beauty in our patients and society.

The junk DNA "pseudogenes," known as genomic fossils, are characterized by their ubiquitousness and abundance within the genomic structure. These genomics sets are recognized by the potential activity of meta-regulating the parent genes; these are transcribed into interfering RNA, consequently acting on miRNA concentration, thereby shedding light on the crosstalk of the pseudogenes' miRNA, siRNA, lncRNA/tumor therapy co-relationship. Moreover, an upcoming visualization regarding pseudogenes is under investigation, which describes the potentiality of pseudogenes as a fundamental component of cancerous evolutionary processing tools. Accordingly, here is a systematic review covering pseudobirth, pseudosignatures, and functional properties of pseudogenes, concluding that these pseudogenes are hypothetically predictive tumor therapies.

The increasing industrial demand to reduce structural weight, optimize structural strength, and high performance has led to the increasing use of tailors welded blanks. However, the joining of tailors welded blanks is an issue of concern to researchers owing to the difference in thickness between the materials in addition to the difference in physical and chemical properties in the case of dissimilar materials. These challenges in joining of tailors welded blanks are conquered by a recent solid phase joining process known as friction stir welding. This review work covers different methods of joining unequal thickness similar/dissimilar (or tailors welded blanks) aircraft materials in different joint configurations along with the challenges and failure modes encountered in these joining processes. From this article researchers and industrialists can take advantage and conquer relative limitations in order to meet the future challenges for joining of similar/dissimilar aircraft materials with unequal thickness

Friction stir welding between plates of unequal thickness, which are made from similar or dissimilar materials, finds wide range of applications in the aerospace and automotive sectors. Friction stir welding of plates made from dissimilar materials having unequal thicknesses is challenging. One of the major challenges is the control of rapid tool degradation which occurs during welding. This work reports a maiden study on tool degradation of high thickness ratio unequal thickness dissimilar material joints made between 6.3 mm thick AA2024-T3 and 2.5 mm thick AA7475-T7 plates. The degradation of friction stir welding tool made of T4 tool steel having tapered cylindrical pin and scrolled shoulder was analyzed. The geometry of tool (before and after welding) was compared; the degradation was categorized, characterized, and analyzed in the light of measured welding temperature, process forces, and process parameters. It was found that the pin undergoes significant degradation in the form of wear and deformation compared to the tool shoulder. The experimental results demonstrated that lower flow stresses caused by higher process temperature leads to lower tool wear and deformation, and vice versa. In addition to temperature and process forces, the surface tilt angle was found to significantly affect the pin deformation. The higher surface tilt angle caused an increase in tool wear and deformation.

Material flow and heat generation by tool shoulder during Friction Stir Welding (FSW) significantly alters the microstructural and thermomechanical behavior of joints. The effect of shoulder size on mechanical properties of joints has not yet been reported in the FSW of Tailor-Welded Blanks (TWBs). This article reports the effect of shoulder size on joint quality in FSW of TWBs between 6.35 mm thick plates of AA2024-T3 and 2.5 mm thick plates of AA7475-T7 alloys in butt joint configuration fabricated under shoulder sizes: 18 mm, 20 mm, and 22 mm. Microstructural evaluation of FSWed joints reveals a significant increase in grain size with shoulder size. The X-ray energy-dispersive spectroscopic (EDS) elemental maps reveal the presence of fine second phase particles in the stir zone. The progressive elimination of void defects with the increase in shoulder size was observed. The tensile testing reveals the highest strength of joints fabricated via shoulder size of 18 mm. Fractographic analyses of broken tensile specimens showed the mixed mode of failure in all the welded specimens.

Background: When an object traverses through the cranium leaving behind both an entry and exit wound, it is called perforating brain injury. Perforating open brain injury is rare. A paucity of published literature on such cases and a lack of a standard management protocol pose significant challenges in managing such cases. Case Description: We present a case of a 24-year-old man who worked as a carpenter at the construction site. He slipped while working and fell from a height of 13 feet onto a rusty, vertically placed 3 feet iron rod located on the ground. Iron rod entered his body from the right upper chest, came out from the neck, and again re-entered through the right upper neck medial to the angle of the mandible and finally came out from the posterosuperior surface of the right side of the head. He presented to the emergency department in a conscious state, but his voice was heavy and slow-paced, and he showed signs of lower cranial nerve palsy on the right side. He underwent numerous radiological investigations. The iron rod was removed in the operation theater under strict aseptic precautions. On day 7 after surgery, he developed right lobar pneumonia, and on day 21, he developed an altered sensorium, followed by a loss of consciousness. He did not regain consciousness and, unfortunately, succumbed after 30 days of sustaining the injuries. Conclusion: Perforating open brain injuries are rare, especially in civilian society, and are usually associated with significant morbidity and mortality. Due to a lack of standard guidelines for managing such severe injuries and limited knowledge, many patients with these injuries do not survive. Although each case presents differently, certain management principles must be followed.

Hematology and serum biochemistry study may provide antique knowledge about the physical status of individuals, making them a valuable tool to differentiate healthy animals from affected animals. We aimed to investigate Steroid safety levels in birds through ex-situ studies at regular therapeutic doses. A total of 100 birds were used for hematology and serum biochemistry. This study was designed into two trials over the summer and winter, each comprised five, ten, fifteen, and twenty days. Each study group was based on five control group birds and 20 experimental group birds. A sum of two groups representing two different Steroids trial groups was treated with therapeutic doses t the stretch of five, ten, fifteen, and twenty days each season. A therapeutic dose of each of the Steroids was given at the rate of three drops two times a day to each bird. Analysis of data reveals that Steroids had severe effects on bird's (Coturnix coturnix) hematological parameters. In most trials, the hematological effects of Bromocriptine as mesylate showed an increase in Red blood cell count and White blood cell count. On the other hand, steroid Estradiol Valerate showed a decrease in these parameters. Effect of steroids on serum biochemistry profile indicate acute damage to vital organs, especially to liver and kidney, indicating an increase in cholesterol, total protein, albumin, urea, and uric acid. The overall effect of steroids on the bird's serum and biochemistry of quails were nearly similar but different only in their intensity.

Tailgut cyst is an uncommon developmental anomaly arising from the embryonic hindgut in the retrorectal space. The patient frequently is asymptomatic or has vague perineal complaints that pose a diagnostic dilemma. Moreover, the patient is often misdiagnosed and therefore mismanaged. MRI is the investigation of choice for diagnosis. After the diagnosis is established, complete surgical excision is required to alleviate patient discomfort and to prevent complications such as infection, malignant transformation, and recurrence. Proper clinical examination and imaging not only establish the accurate diagnosis but also help in determining the best surgical approach for the patient (anterior abdominal, posterior sacral perineal, or combined approach). We hereby report a case of recurrent tailgut cyst managed with a combined anterior laparoscopic and perineal approach.

A putative protease gene (aprE) from the thermophilic bacteriumCoprothermobacter proteolyticuswas cloned and expressed inBacillus subtilis. The enzyme was determined to be a serine protease based on inhibition by PMSF. Biochemical characterization demonstrated that the enzyme had optimal activity under alkaline conditions (pH 8–10). In addition, the enzyme had an elevated optimum temperature (60°C). The protease was also stable in the presence of many surfactants and oxidant. Thus, theC. proteolyticusprotease has potential applications in industries such as the detergent market.

Context: Head and neck surgeries are complex. Wound complications are associated with considerable morbidity and can result in delay in the adjuvant treatment. Identification of factors will help in formulating preventive guidelines. Aims: The aim of this study is to identify perioperative factors responsible for wound complications. Settings and Design: Prospective study of 186 head and neck patients. Subjects and Methods: Pre-operative, intraoperative and post-operative factors were recorded. Each patient was evaluated for minor and major wound complications twice daily during the hospital stay. Statistical Analysis: Chi-square test was used for univariate and log regression test was used for multivariate analysis. Results: The overall wound complication rate was 29% with 7% major and 22% minor complications. On univariate analysis, laryngeal and hypopharyngeal location, advanced T stage, poor oral hygiene, clean-contaminated surgery, low Karnofsky performance status (KPS), flap reconstruction, blood loss more than 1000 ml, perioperative blood transfusion, duration of surgery greater than 4.3 h and post-operative hemoglobin lesser than 11 g%, post-operative tracheostomy and resection of mandible were statistical significant factors. On multivariate analysis, post-operative tracheostomy, low KPS, post-operative serum albumin less than 3.7 g/dl and duration of surgery greater than 4.3 h were significant factors. Conclusion : Apart from unavoidable factors, it is essential to take care of certain factors viz nutrition, haemoglobin, oral hygiene, asepsis and repeating antibiotics in prolonged surgery.

An Industrial robot is reprogrammable, automatically controlled, multifunctional manipulator programmable in three or more axes, which may be either fixed in place or mobile for use in industrial automation applications. Technical innovations in robotic welding has facilitated manual welding processes in sever working conditions with enormous heat and fumes to be replaced with robotic welding. The robotic welding has greater capability to control robot motion, welding parameters and enhanced wrong detection and wrong correction. Major difficulties in robotic welding are joint edge inspection, weld penetration control, seam tracking of joints, and width or profile measurement of a joint. These problems can be more easily solved by use of sensory feedback signals from weld joint. Robotic welding system has intelligent and effective control system that can track the joint, monitor the joint in process and accounts for variation in joint location. Sensors play an important role in robotic welding systems with adaptive and intelligent control system features that can track the joint, account for variation in joint location and geometry monitor in-process quality of the weld. In this paper various aspects of robotic welding, robot programming, and problems associated with robot welding are undertaken.
