Abstract Background National Malaria Programmes (NMPs) monitor the durability of insecticide-treated nets (ITNs) to inform procurement and replacement decisions. This is crucial for new dual active ingredients (AI) ITNs, for which less data is available. Pyrethroid-only ITN (Interceptor®) and dual AI (Interceptor® G2, and PermaNet® 3.0) ITNs were assessed across three health districts over 36 months in southern Burkina Faso to estimate median ITN survival, insecticidal efficacy, and to identify factors contributing to field ITN longevity. Methods Durability was monitored through a prospective study of a cohort of nets distributed during the 2019 mass campaign. Three health districts were selected for their similar pyrethroid-resistance, environmental, epidemiological, and population profiles. Households were recruited after the mass campaign, with annual household questionnaire follow-ups over three years. Each round, ITNs were withdrawn for bioassays and chemical residue testing. Key measures were the percentage of cohort ITNs in serviceable condition, insecticidal effectiveness, and chemical residue content against target dose. Cox proportional hazard models were used to identify determinants influencing ITN survival. Results At endline, the median useful life was 3.2 (95% CI 2.5–4.0) years for PermaNet® 3.0 ITNs in Orodara, 2.6 (95% CI 1.9–3.2) years for Interceptor® G2 ITNs in Banfora and 2.4 (95% CI 1.9–2.9) years for Interceptor® ITNs in Gaoua. Factors associated with ITN survival included cohort ITNs from Orodara (adjusted hazard ratio (aHR) = 0.58, p = 0.026), households seeing less rodents (aHR = 0.66, p = 0.005), female-headed households (aHR = 0.66, p = 0.044), exposure to social behavior change (SBC) messages (aHR = 0.52, ≤ 0.001) and folding nets when not in use (aHR = 0.47, p

Abstract Background Mass distribution of insecticide-treated nets (ITNs) is the principal malaria vector control strategy adopted by Niger. To better inform on the most appropriate ITN to distribute, the National Malaria Control Programme (NMCP) of Niger and its partners, conducted insecticide resistance monitoring in selected sites across the country. Methods The susceptibility of Anopheles gambiae sensu lato (s.l.) to chlorfenapyr and pyrethroid insecticides was investigated in a total of sixteen sites in 2019 and 2020, using 2–5-day-old adults reared from wild collected larvae per site. The susceptibility status, pyrethroid resistance intensity at 5 and 10 times the diagnostic concentrations, and piperonyl butoxide (PBO) synergism with diagnostic concentrations of deltamethrin, permethrin and alpha-cypermethrin were assessed using WHO bioassays. Two doses (100 and 200 µg/bottle) of chlorfenapyr were tested using the CDC bottle assay method. Species composition and allele frequencies for knock-down resistance (kdr-L1014F and L1014S) and acetylcholinesterase (ace-1 G119S) mutations were further characterized using polymerase chain reaction (PCR). Results High resistance intensity to all pyrethroids tested was observed in all sites except for alpha-cypermethrin in Gaya and Tessaoua and permethrin in Gaya in 2019 recording moderate resistance intensity. Similarly, Balleyara, Keita and Tillabery yielded moderate resistance intensity for alpha-cypermethrin and deltamethrin, and Niamey V low resistance intensity against deltamethrin and permethrin in 2020. Pre-exposure to PBO substantially increased susceptibility with average increases in mortality between 0 and 70% for tested pyrethroids. Susceptibility to chlorfenapyr (100 µg/bottle) was recorded in all sites except in Tessaoua and Magaria where susceptibility was recorded at the dose of 200 µg/bottle. Anopheles coluzzii was the predominant malaria vector species in most of the sites followed by An. gambiae sensu stricto (s.s.) and Anopheles arabiensis. The kdr-L1014S allele, investigated for the first time, was detected in the country. Both kdr-L1014F (frequencies [0.46–0.81]) and L1014S (frequencies [0.41–0.87]) were present in all sites while the ace-1 G119S was between 0.08 and 0.20. Conclusion The data collected will guide the NMCP in making evidence-based decisions to better adapt vector control strategies and insecticide resistance management in Niger, starting with mass distribution of new generation ITNs such as interceptor G2 and PBO ITNs.

Abstract Background The interest in larvicides of plant origin is generally renewed in vector control because of their safety compared to synthetic larvicides. However, there are concerns about the relative safety dose of these phytochemicals on non-target organisms which led to the development of plant derived nanoparticles. In this study, we examined the bioefficacy of low doses of two green synthesized nanoparticles on immature stages of Anopheles mosquitoes in Nigeria. Aqueous plants (Moringa oleifera and Ficus exasperata) extracts were used in the biosynthesis. The prepared Ag-NPs were characterizations using Fourier-transform infrared spectroscopy (FT-IR), UV-Vis spectroscopy, and scanning electron microscopy (SEM). Third and early fourth instars of known susceptible laboratory strains of Anopheles gambiae s.s. (KISUMU strains) and pyrethroid resistant field strain of An. gambiae were exposed to serial dilutions of 0.25, 0.5, 0.75, 1.0 and 2.5 ppm of each phyto nanoparticles. Moribund and dead larvae were observed after 24 and 48 h post exposure, and the results were analysed with descriptive statistics. Results With the laboratory mosquitoes, Moringa oleifera AgNP effected high mortalities of 88–100% (LC50 = 0.39 ppm; LC95 = 0.62 ppm) at 24 h post exposure except at the lowest concentration, while Ficus exasperate AgNP induced a 32–100% mortality (LC50 = 0.51 ppm; LC95 = 1.15 ppm) except at the lowest concentration. In the field populations, mortality in Moringa oleifera and Ficus exasperata was 23–93% (LC50 = 0.65 ppm; LC95 = 2.28 ppm) and 37–50% (LC50 = 1.51 ppm; LC95 = 391.64 ppm) respectively. Abstract Background Aedes aegypti transmits several arboviral diseases of global public health threat such as dengue, zika, and yellow fever. Recently, a series of yellow fever outbreaks have been reported in different parts of the country. Globally, the control of mosquito-borne diseases has relied heavily on insecticides based vector control efforts. Therefore, we assessed the susceptibility status to three different classes of WHO approved insecticides and evaluated the importance of detoxifying enzymes on insecticides resistance in Aedes aegypti collected from Lagos State, Nigeria. Aedes aegypti immature stages were collected from suitable habitats in peri-domestic areas of Alimosho, Kosofe, Badagry, and Ibeju-Lekki Local Government Areas (LGAs) of Lagos State, 2–5 days old glucose-fed adult female mosquitoes were exposed to discriminating doses of DDT, permethrin, bendiocarb, and PBO synergist using WHO standard procedure and kits. Identification was through morphological means only. Results Resistance to DDT and permethrin was recorded in all the LGAs, suspected resistance to bendiocarb was recorded in Aedes aegypti from Alimosho while full susceptibility was recorded in other LGAs. PBO synergist significantly increases the 24 h mortality of DDT and permethrin-resistant Aedes aegypti in all the LGAs. The activities of both GSTs and cytochrome P450s increase with decreasing mortality for Aedes aegypti, regression value (R 2) ranges from 0.8889 to 0.6224 for GSTs and 0.4379 to 0.616 for cytochrome P450s. Conclusions Aedes aegypti population from Lagos State, Nigeria, have developed resistance to DDT and permethrin, the activities of P450s and GST were implicated in DDT and permethrin resistance in this study. Therefore, a need for regular insecticides monitoring and PBO should be incorporated into Aedes aegypti insecticides control strategies to forestall development of resistance to DDT and pyrethriods.

The knowledge of the ecology and abundance of mosquito is important in determining the potential risk of exposure to mosquito-borne diseases and the appropriate control measure to effectively reduce the mosquito population. A survey was carried out in four Local Government Areas (LGAs) of Lagos State, Nigeria for one year to determine the abundance, diversity, and larval habitat characteristics of mosquito. Larvae were collected using WHO standard methods, the physiochemical characteristics of the habitats were recorded, the larvae were reared to adults and identified using morphological identification keys and molecular assays. Adult mosquitoes were also collected outdoor using a trap with octanol attractant. Five species belonging to 3 genera were collected and identified as Anopheles gambiae complex, Anopheles funestus complex, Culex quinquefasciatus, Aedes aegypti, and Aedes albopictus. Cx quinquefasciatus (37.7%) is the most abundant followed by Ae aegypti (31.3%) and An. gambiae s.l. (26.9%) while Ae. albopictus (1.9%) is the least abundant. Abstract Background The decline in the efficacy of artemisinin-based combination treatment (ACT) in some endemic regions threatens the progress towards global elimination of malaria. Molecular surveillance of drug resistance in malaria-endemic regions is vital to detect the emergence and spread of mutant strains. Methods We observed 89 malaria patients for the efficacy of artemether-lumefantrine for the treatment of uncomplicated Plasmodium falciparum infections in Lagos, Nigeria and determined the prevalence of drug resistant strains in the population. Parasite clearance rates were determined by microscopy and the highly sensitive var gene acidic terminal sequence (varATS) polymerase chain reaction for 65 patients with samples on days 0, 1, 3, 7, 14, 21 and 28 after commencement of treatment. The genomic finger print of parasite DNA from pre- and post-treatment samples were determined using 24 nuclear single nucleotide polymorphisms (SNP) barcode for P. falciparum. Drug resistance associated alleles in chloroquine resistance transporter gene (crt-76), multidrug resistance genes (mdr1–86 and mdr1–184), dihydropteroate synthase (dhps-540), dihydrofolate reductase (dhfr-108) and kelch domain (K-13580) were genotyped by high resolution melt analysis of polymerase chain reaction (PCR) fragments. Results By varATS qPCR, 12 (18.5%) of the participants had detectable parasite DNA in their blood three days after treatment, while eight (12.3%) individuals presented with genotypable day 28 parasitaemia. Complexity of infection (CoI) was 1.30 on day 0 and 1.34 on day 28, the mean expected heterozygosity (HE) values across all barcodes were 0.50 ± 0.05 and 0.56 ± 0.05 on days 0 and 28 respectively. Barcode (π) pairwise comparisons showed high genetic relatedness of day 0 and day 28 parasite isolates in three (37.5%) of the eight individuals who presented with re-appearing infections. Crt-76 mutant allele was present in 38 (58.5%) isolates. The mdr1–86 mutant allele was found in 56 (86.2%) isolates. No mutation in the K-13580 was observed. Conclusions Persistence of DNA-detectable parasitaemia in more than 18% of cases after treatment and indications of genetic relatedness between pre- and post-treatment infections warrants further investigation of a larger population for signs of reduced ACT efficacy in Nigeria.

Background: Timely entomological and insecticide resistance monitoring is a key to generating relevant data for vector management. We investigated the insecticide susceptibility status of Anopheles gambiae s.l. in eight rural farming communities in Southern Gombe, Nigeria. Methods: Overall, 3–5 days-old adult female Anopheles mosquitoes reared from field-collected immature stages between September and November, 2014 were exposed to the diagnostic doses of pyrethroids, organophosphate and carbamate insecticides using the Center for Disease Control Bottle bioassay. The observatory knockdown time from exposure to each insecticide was recorded up to two hours. The dead mosquitoes were then identified morphological­ly and by molecular assays. Results: Mortality results showed resistance in An. gambiae s.l. populations to bendiocarb (2.3–100%), deltamethrin (39–70%), pirimiphos-methyl (65-95%), dichloro-diphenyl-trichloroethane (0–38.1%), permethrin (0–46.3%) and lambda-cyhalothrin (42.5–86.4%). The few cases of full susceptibility were observed from lamdacyhalothrin exposed population of An. gambiae s.l. in Banbam and Pantami respectively. An. gambiae 177 (45%) was significantly higher (P< 0.05) than An. arabiensis 64 (16.3%), An. coluzzii 34 (8.7%) and An. gambiae/An. coluzzii hybrid 78 (19.8%). Conclusion: A strong evidence of widespread resistance in the major malaria vector species in Southern Gombe to all common classes of insecticides is a justification for the State Malaria Elimination Programme to consciously con­sider incorporating insecticide resistance management strategies into control programs in order to sustain the future of current control interventions.

Anopheles gambiae, An. coluzzii and An. arabiensis are the three major vectors of malaria in Nigeria. These mosquitoes have developed resistance to different insecticides. Insecticides resistance intensity assay was recently introduced to provide insight into the potential operational significance of insecticide resistance. Here, we present data on pyrethroids resistance intensity and resistance mechanisms from six vector surveillance sites (Lagos, Ogun, Edo, Anambra, Kwara and Niger) in Nigeria. Adult Anopheles reared from larval collections were tested using WHO insecticides susceptibility protocol with 1x concentration of permethrin and deltamethrin followed with intensity assays with 5x and 10x concentrations of both insecticides. Synergistic and biochemical assays were carried out and underlying resistance mechanisms determined following standard protocols. Anopheles gambiae constituted >50% samples tested in five sites. Permethrin and deltamethrin resistance was observed at all the sites. The Kdt50 varied from 15 minutes (CI = 13.6-17.2) in deltamethrin to 42.1 minutes (CI = 39.4-44.1) in permethrin. For both insecticides, Kdt95 was >30 minutes with 25% to 87% post exposure mortality at the different sites. The West Africa knock down resistance (kdr-w) mechanism was found at each site. Resistant An. gambiae from Lagos, Ogun and Niger synergized prior to permethrin or deltamethrin exposure showed significant mortality (89-100%) compared to unsynergized mosquitoes (Lagos, p = 0.031; Ogun, p = 0.025; Niger, p = 0.018). Biochemical analyses revealed significant increased levels of P450 enzymes in resistant Anopheles gambiae from Lagos (p = 0.038); Ogun (p = 0.042) and Niger (p = 0.028) in addition to GST in Lagos (p = 0.028) and Ogun (p = 0.033). Overall, the results revealed high pyrethroid resistance associated with increased activities of metabolic enzymes (P450 + GST) in An. gambiae and An. coluzzii from Lagos and Ogun. The presence of kdr + P450 conferred moderate resistance whereas low resistance was the case where kdr was the sole resistance mechanism. Findings thus suggests that elevated levels of cytochrome P450 enzymes together with GST were responsible for high or severe pyrethroid resistance.

Baited traps are potential tools for removal or surveillance of disease vectors. To optimize the use of counter-flow traps baited with human odor (nylon socks that had been worn for a single day) to capture wild mosquitoes in the Gambia, investigations were conducted at a field experimental site.Experiments employing Latin square design were conducted with a set of six huts to investigate the effects of the following on overnight mosquito trap catches: (1) placement of traps indoors or immediately outdoors, CO(2) supply, and presence of a human subject in the hut; (2) trap height for collecting mosquitoes immediately outdoors; (3) height and distance from hut; (4) interaction between multiple traps around a single hut and entry of mosquitoes into huts. A total of 106,600 adult mosquitoes (9.1% Anopheles gambiae s.l., 4.0% other Anopheles species) were collected over 42 nights. The high numbers of An. gambiae s.l. and other mosquitoes collected by odor-baited traps required CO(2) but were largely independent of the presence of a person sleeping in the hut or of trap placement indoors or outdoors. For outdoor collection that is considered less intrusive, traps opening 15 cm above the floor of the hut veranda were more highly effective than traps at other heights or further from the hut. There was no significant evidence of saturation or competition by the traps, with multiple traps around a hut each collecting almost as many mosquitoes as single traps and no effect on the numbers of mosquitoes entering the huts.The outdoor trapping protocol is convenient to compare attractiveness of different odors or synthetic chemicals to malaria vectors and other wild mosquitoes. The finding that such traps are reliably attractive in the presence or absence of a human volunteer encourages their potential development as standardised surveillance tools.
