A nine-step asymmetric total synthesis of (+)-tubingensin A has been realized.

An array of spiro[benzofuran-2,3′-pyrazol]-3-imines with diverse functional groups were readily assembled from aurone-derived azadienes and in-situ generated nitrilimines under mild conditions. The transformation was performed in a regio- and diastereoselective fashion, in which a synergetic [2+3] cycloaddition pathway was likely involved. This novel methodology has extended the synthetic application of aurone-derived azadienes as a kind of two-atom synthon.

The first asymmetric total synthesis of three picrinine-type akuammiline alkaloids, (-)-picrinine, (-)-scholarisine C, and (+)-5-β-methoxyaspidophylline, has been accomplished. The synthesis features an efficient acid-promoted oxo-bridge ring-opening and further carbonyl O-cyclization to assemble the furoindoline scaffold, an unusual Dauben-Michno oxidation to construct the requisite α,β-unsaturated aldehyde functionality, and a nickel-mediated reductive Heck reaction to forge the [3.3.1]-azabicyclic core.

A formal (3+2) annulation reaction between isatin-derived N-tosylhydrazones and o-(trimethylsilyl)aryl triflates allows facile synthesis of spiro[indazole-3,3′-indolin]-2′-ones in high to excellent yields. These two types of 1,4-dihydrobenzofuro[3,2-b]pyridines could be aromatized in the presence of DBU to afford benzofuro[3,2-b]pyridines, which could also be accessed via a one-pot procedure.

A facile access to thiazolidin-4-ones from α-enolic dithioesters and α-halohydroxamates in situ derived active 1,3-dipolar aza-oxyallyl cations was achieved under mild conditions. These five-membered heterocycles with Z-exocyclic double bonds and diverse substituents can be readily obtained via a regio- and stereoselective formal [3+2] cycloaddition pathway.

A cobalt-catalyzed direct C–H/N–H functionalization of thiophene-2-carbohydrazides with maleimides by utilizing 2-(1-methylhydrazinyl)pyridine (MHP) as an easily removable bidentate directing group has been developed. This formal [4+2] cycloaddition has been achieved for the first time, and it provides an alternative and versatile approach to construct thiophene-fused pyridones using an inexpensive cobalt catalyst. The C–H/N–H activation cascade protocol showed a high efficiency and a broad substrate scope, and the products were obtained in good to excellent yields.

A novel and practical protocol for the synthesis of fully substituted pyrazoles from pyridinium 1,4-zwitterionic thiolates and hydrazonoyl chlorides in excellent yields under mild conditions is described. A novel [4+3] cascade cyclization reaction of pyridinium 1,4-zwitterionic thiolates with azoalkenes derived from α-halo hydrazones in situ has been developed. This reaction was found to allow expedient access to an array of 2,5-dihydro-1,4,5-thiadiazepines. Libraries of highly functionalized sulfoxide and sulfone analogues of 2,5-dihydro-1,4,5-thiadiazepines were also obtained via selective oxidation with m-CPBA.

The synthesis of pyrazolotetrazepin-7-ones from 1,3,5-triazinanes with N,N′-cyclic azomethine imines was achieved via a formal (5 + 2) pathway under thermal conditions. The annulation reaction is catalyst-free and additive-free.

A 1,3-dipolar cycloaddition reaction of arynes with 3-diazoindolin-2-ones under mild conditions in excellent yields has been developed, which allows facile access to an library of labile spiro[indazole-3,3′-indolin]-2′-ones. Spiro[indazole-3,3′-indolin]-2′-imines could be obtained as well following the same protocol. The isomerization reaction of spiro[indazole-3,3′-indolin]-2′-ones under thermal or acidic conditions has been efficiently achieved to afford a wide range of indazolo-[2,3-c]quinazolin-6(5H)-ones and the one-pot synthesis of indazolo-[2,3-c]quinazolin-6(5H)-ones from arynes and 3-diazoindolin-2-ones is also described. Whereas, spiro[indazole-3,3′-indolin]-2′-imines could not undergo the same rearrangement.

A cobalt-catalyzed oxidative coupling of benzoic hydrazides with maleimides by utilizing 2-(1-methylhydrazinyl)pyridine as a bidentate directing group has been developed. This C–H functionalization/spirocyclization cascade protocol shows high efficiency and remarkable functional group tolerance, and the ubiquitous spirosuccinimides were obtained in good to excellent yields with high regioselectivity. This strategy also provides a novel and efficient access to diverse symmetric and unsymmetrical bisspirosuccinimides.

A chemoselective N-arylation reaction of 2-aminopyridine derivatives with arynes in good to excellent yields has been described. The N-arylation products could be further applied to the facile construction of benzoisoquinuclidines and isoquinuclidines as well as pyrido[1,2-a]benzimidazoles.

An asymmetric total synthesis of (−)-aspidophylline A has been accomplished. The key transformations include an asymmetric hydride transfer hydrogenation of an α,β-acetylenic ketone and a cationic gold(I)-catalyzed 6-exo-dig cyclization involving an alkynylindole/aminal formation cascade, which enables stereoselective establishment of the requisite quaternary carbon center.
