Abstract

A Round Robin study has been carried out to estimate the impact of the human element in small-angle scattering data analysis. Four corrected datasets were provided to participants ready for analysis. All datasets were measured on samples containing spherical scatterers, with two datasets in dilute dispersions, and two from powders. Most of the 46 participants correctly identified the number of populations in the dilute dispersions, with half of the population mean entries within 1.5% and half of the population width entries within 40%, respectively. Due to the added complexity of the structure factor, much fewer people submitted answers on the powder datasets. For those that did, half of the entries for the means and widths were within 44% and 86% respectively. Abstract

We propose a versatile software package in the form of a Python extension, named CDEF (Computing Debye's scattering formula for Extraordinary Formfactors), to approximately calculate scattering profiles of arbitrarily shaped nanoparticles for small-angle X-ray scattering (SAXS). CDEF generates a quasi-randomly distributed point cloud in the desired particle shape and then applies the open source software DEBYER for efficient evaluation of Debye's scattering formula to calculate the SAXS pattern. If self-correlation of the scattering signal is not omitted, the quasi-random distribution provides faster convergence compared to a true random distribution of the scatterers, especially at higher momentum transfer. The usage of the software is demonstrated for the evaluation of scattering data of Au nanocubes with rounded edges, which were measured at the four-crystal monochromator beamline of PTB at the synchrotron radiation facility BESSY II in Berlin. Our implementation is fast enough to run on a single desktop computer and perform model fits within minutes. The accuracy of the method was analyzed by comparison with analytically known form factors and verified with another implementation, the SPONGE, based on a similiar principle with fewer approximations. Abstract

Engineered nanomaterials have been found to induce oxidative stress. Cellular oxidative stress, in turn, can result in the induction of antioxidant and detoxification enzymes which are controlled by the nuclear erythroid 2-related factor 2 (NRF2) transcription factor. Here, we present the results of a pre-validation study which was conducted within the frame of BIORIMA (“biomaterial risk management”) an EU-funded research and innovation project. For this we used an NRF2 specific chemically activated luciferase expression reporter gene assay derived from the human U2OS osteosarcoma cell line to screen for the induction of the NRF2 mediated gene expression following exposure to biomedically relevant nanobiomaterials. Specifically, we investigated Fe3O4-PEG-PLGA nanomaterials while Ag and TiO2 “benchmark” nanomaterials from the Joint Research Center were used as reference materials. The viability of the cells was determined by using the Alamar blue assay. We performed an interlaboratory study involving seven different laboratories to assess the applicability of the NRF2 reporter gene assay for the screening of nanobiomaterials. The latter work was preceded by online tutorials to ensure that the procedures were harmonized across the different participating laboratories. Fe3O4-PEG-PLGA nanomaterials were found to induce very limited NRF2 mediated gene expression, whereas exposure to Ag nanomaterials induced NRF2 mediated gene expression. TiO2 nanomaterials did not induce NRF2 mediated gene expression. The variability in the results obtained by the participating laboratories was small with mean intra-laboratory standard deviation of 0.16 and mean inter laboratory standard deviation of 0.28 across all NRF2 reporter gene assay results. We conclude that the NRF2 reporter gene assay is a suitable assay for the screening of nanobiomaterial-induced oxidative stress responses.

Abstract

International audience; The unambiguous correlation of possible health and sustainability risks to nanoparticle size must be enabled by reliable measurement of nanoparticle size, to ensure comparability and compatibility between results measured under different methods. The NPSIZE project funded by European Metrology Program (EMPIR) develop methods, reference materials and modelling to improve the traceability chain,comparability and compatibility of nanoparticle size measurements.In this work, we present how spherical silica nanoparticles are synthetized with controlled monomodal or bimodal dispersion to be use as reference materials and international round-robin. Improving the fabrication requires a fine understanding of synthesis (1), coupled with an expertise of in-situ or ex-situ analysis methods. This is a new challenge for the analysis : determining not only average characteristics (size, chemical composition and shape ...) but also the concentration and the distribution over the population studied (2).Small-Angle X-ray Scattering (3) allows very precise measurements of the nanoparticles size and concentration that can be directly link to the metric system (4) (metrological traceability) . We developed a SAXS laboratory instrument dedicated to the in-situ characterization of nanoparticles, which enable fast measurements, and the monitoring of the synthesis parameters. 22. Impact of Cooling Profile on Refined Palm Oil Crystallization: Microscopic and Small and Wide‐Angle X‐Ray Scattering Investigations.

Abstract

Among the different possible strategies, thermal control is a major lever to control fat structure (i.e., triacylglycerols (TAGs) molecules organization and crystals morphology). In this article, the impact of thermal treatments on TAGs packing tendency contained in refined palm oil (RPO) is determined. Three cooling profiles are considered: slow, intermediate, and fast. Slow cooling induces the formation of β' polymorphic form in sequence as 2Lβ' then 3Lβ' configurations. RPO rapidly cooled first crystallizes in α form before turning into the 2Lβ' structure. At intermediate cooling profile, 2Lβ' and 3Lβ' are observed and another original phase ϕ is formed around 0°C. This phase has similarities with the sub‐α phase observed in cocoa butter which corresponds to a β' subcell and presents the capacity of adapting its composition during cooling, leading to an increase of its interplanar distances by decreasing temperature. Practical applications: In case of fats in food products, suitable cooling profile is a compromise between rheological properties of final product (thus, sensorial properties) and relative stability during storage. In this article, three different cooling profiles applied to RPO, leading to various crystal lattices are studied. These lead to systems with varying sensory properties and stability. Abstract

International audience; Size measurement of nanoparticles (NP) becomes a challenging analytical problem when non-spherical shapes must be traceably measured [1]. However, most industrial NPs have irregular shapes and broad size distribution making it often more complex to follow European regulatory to identify a material as a nanomaterial according to which accurate measurement of the smallest dimension and its size distribution is necessary.The European research project nPSize - Improved traceability chain of nanoparticle size measurements aims to fill this gap by developing potential non-spherical reference nanoparticles, measurement procedures and physical modelling to improve the traceability chain, comparability and compatibility for NP size measurements between different methods. Therefore, new model NP with well-controlled shape has been synthesized and are supposed to be systematically characterized using the traceable methods scanning/transmission electron microscopy, atomic force microscopy and small angle X-ray scattering. Following NP candidates are under investigation with respect to their homogeneity and stability: (i) titania nanoplatelets (10-15 nm thickness x 50-100 nm lateral) [2], (ii) titania bipyramides (~60 nm length x 40 nm width) [3], (iii) titania acicular particles (100 nm length x 15-20 nm width; aspect ratio 5.5/6) [4], (iv) gold nanorods (~10 nm width x 30 nm length) [5], and (v) gold nanocubes (~55 nm x 55 nm x 55 nm) [6,7]. In addition, sample preparation procedures as well as measurement analysis procedures with evaluation of appropriate measurands and descriptors for each material class and method are being developed to support standardization. To underpin the traceability of the size measurement of non-spherical NP, physical modelling of the signals in e.g. electron microscopy techniques [8] will be used and in combination, the implementation of machine learning is aimed to facilitate measurement analysis procedures, especially regarding the accurate thresholding/segmentation of the NPs.

Abstract

International audience; Small Angle X-rays Scattering Spectroscopy (SAXS) is the method of choice for nanopar-ticle diameter and concentration determination. It is metrologically traceable for spherical nanoparticle mean diameter determination and does not require any sample preparation or calibration. On the other hand, Single Particle ICPMS (SPICPMS) is still under developments and requires involved process clarification and accuracy improvement. The strategy of this presentation based on 6 spherical gold nanoparticle suspensions distributed over a large size range (30, 50, 60, 80,100 and150 nm), is the comparison of the two techniques to study comprehensively SPICPMS performance and observe phenomena: Potential matrix effect are eliminated by stabilizing nanoparticles with chitosan in HCL. Chitiosan encapsulates nanoparticles, stabilizes their dispersion and protects them from dissolution. Detection counting/analog threshold and timeout appear as the relevant parameters for transient signals. They show an influence not only on mean signal but also on signal distribution. The detection tuning proposed allow to linearly calibrate the nanoparticle distribution signal to cubed diameter over the entire range studied with no sensitivity diminution. Comparing the 3 classical transport efficiency methods (number, size and waste), size transport efficiency is shown as the most accurate. A procedure is proposed, it is validated analyzing three gold nanoparticles suspensions (135, 40 and 50nm). The results are consistent with SAXS measurements.

Abstract

International audience; Small-Angles X-Ray Scattering (SAXS) has been established as a metrological method for the determination of nanoparticles size and size distribution. Modern SAXS Laboratory experiments 1 , by involving synchrotron-based instrumentation at lower price and very stable X-ray source, are more and more used in nanomaterials metrology. In the frame of the EURAMET Innanopart project, we have developed a methodology for the size, size distribution and concentration determination of spherical nanoparticles. This protocol involves a precise sample preparation, a rigorous procedure for the data acquisition, and a set of homemade software tools for the data processing-from the acquisition, the absolute scaling, to the analysis. The SAXS technique is very accurate for the characterization of simple case using nanoparticles : monodisperses, spheric form, unique composition. But it is more complicated in the case of nanoparticles size mixtures and in case of nanoparticles in complex media. Recent works in the lab shows it is possible to achieve precise measurement on mixture of spheric nanoparticles and nanoparticles in complex media (food additives) using different software or methodology approaches. In the frame of the Innanopart project we have develop a methodology for the size and concentration determination of spheric Silica nanoparticles. This protocol involves a precise sample preparation, and a set of homemade software tools for the data processing-from the acquisition, the absolute scaling, to the analysis. spICPMs is not a metrological traceable technique but has many strengths to become a useful complement of nanoparticle characterization methods such as SAXS and microscopy. It can also measure highly diluted nanoparticles suspensions which is not the case of Dynamic Light scattering (DLS) or SAXS. Finally, ICPMS analyzes inorganic ions in liquid solution in a very large range of concentration, which should allow linear diameter measurement range over at least 3 orders of magnitude by spICPMS. In this work, we confront spICPMS, as a counting technique, with SAXS in order to investigate the method and the developed protocols on a set of commercial Gold Nanoparticles.

Abstract

International audience; Despite the long-lasting interest in the synthesis control of nanoparticles (NPs) in both fundamental and applied nanosciences, the driving mechanisms responsible for their size and shape selectivity in an environment (solution) are not completely understood, and a clear assessment of the respective roles of equilibrium thermodynamics and growth kinetics is still missing. In this study, relying on an efficient atomistic computational approach, we decipher the dependence of energetics, shapes and morphologies of gold NPs on the strength and nature of the metal–environment interaction. We highlight the conditions under which the energy difference between isotropic and elongated gold NPs is reduced, thus prompting their thermodynamic coexistence. The study encompasses both monocrystalline and multi-twinned particles and extends over size ranges particularly representative of the nucleation and early growth stages. Computational results are further rationalized with arguments involving the dependence of facet and edge energies on the metal–environment interactions. We argue that by determining the abundance and diversity of particles nucleated in solution, thermodynamics may constitute an important bias influencing their final shape. The present results provide firm grounds for kinetic simulations of particle growth.

Abstract

International audience; Small Angle X-ray Scattering (SAXS) allows traceable measurements of size, size distribution , surface area, concentration and shape of nanomaterials in solutions, powders and in bulk materials. We present here a custom-made state-of-the-art SAXS laboratory instrument (see Figure 1) based on Mo X-ray generator. Its design has been thought and optimized for the wide angles (WAXS) for investigate nanostructured material in particular in the size range from below 1 nm to above 20 nm. The X-ray generator is a molybdenum (17 keV) rotating anode. A combination of a multilayer collimating mirror and a patented hybrid slits gives us a very sharp and high purity beam for a laboratory setup, with a size of 1x1 mm2 and a flux of 10 8 photons/s. The motorized sample holder can load 20 capil-laries, a temperature controlled system or a circulation environment system for kinetic studies. A vacuum chamber is placed behind whose output window diameter is designed for the 2D image plate Mar345 detector. The accessible q-range is qmin = 0.03 Å-1 to qmax = 3 Å-1. A special attention was given to the complete data treatment processing (absolute intensities, uncertainties, reference samples, calibration ,…), enable us to make traceable measurements. Combining molybdenum energy and the state-of-art experimental setup provide us with a very powerful tool for nanomaterial studies [2][3]. We present here some results obtained with this experiment for inter-laboratories characterization studies in particular of silver nanoparticles, and Si nanoparticles.

Abstract

The main objective of the nPSize project is to improve the measurement capabilities for nanoparticle size based on both measurement methods traceable to SI units and new reference materials. Two basic approaches have been used in order to develop measurement procedures resulting in traceable results of the nanoparticle size distribution: physical modelling for the methods used in the project (TSEM, SEM, AFM and SAXS) and machine learning. Physical modelling:In this part, the physical models associated with different shape measurements for the techniques TSEM, SEM, AFM and SAXS have been collected and further developed with the aim to simulate the resulting signal as measured by the individual methods. Uncertainties and traceability associated with each model were investigated and evaluated. In the following, the progress on these physical models is reported for each individual method. Machine Learning modelling:The aim of this part is to use machine learning to enable automatic measurement of nanoparticle shape from expert a-priori information only. No physical model will be used as a-priori information in this task. The accuracy and traceability of the size results obtained by each technique will be analyzed and compared with the physical modelling. A machine learning database will then be used to create automatic detection algorithms.
