1. Synchronous circadian rhythms of mRNA levels and activities of cholesterol 7 alpha-hydroxylase in the rabbit and rat.

Abstract

Cholesterol 7 alpha-hydroxylase, the key enzyme in a series of metabolic changes for the production of bile acids from cholesterol, shows circadian rhythms. The mechanism of the circadian rhythms is the subject of controversy; moreover, few pieces of information are presently available. Because the rabbit is a good animal model for the study of human cholesterol-bile acid metabolism, in the present study we isolated a complete cDNA encoding rabbit cholesterol 7 alpha-hydroxylase (3022 base pairs (bp) long and 1503 bp open-reading frame encoding 501 amino acid residues). Using this isolate rabbit cDNA and the rat cDNA as probes, the circadian rhythms of this enzyme were studied in normal male rabbits and rats. The animals were maintained on a 12-h light-dark cycle. Three subjects each were then killed every 3 h for 1 day. The livers were harvested, and the mRNA levels and the activities of the enzyme were measured. Both mRNA levels and activities of the enzyme showed remarkable circadian rhythms, with higher values during the dark phase and lower values reaching minimum during the light phase. The enzyme activities correlated well with the mRNA levels. There was no shift in the cycles of the enzyme activities and the mRNA levels. These findings suggest that pretranslational regulation is the most likely mechanism for the circadian rhythms of cholesterol 7 alpha-hydroxylase. Because the rhythms of mRNA levels and activities of this enzyme are synchronous, the turnover of the enzyme should be as fast as that of the mRNA, and we can hereafter measure the mRNA levels instead of the enzyme.

Abstract

Background: The Global Anticoagulant Registry in the FIELD–Atrial Fibrillation (GARFIELD-AF) is an ongoing prospective noninterventional registry, which is providing important information on the baseline characteristics, treatment patterns, and 1-year outcomes in patients with newly diagnosed non-valvular atrial fibrillation (NVAF). This report describes data from Indian patients recruited in this registry. Methods and results: A total of 52,014 patients with newly diagnosed AF were enrolled globally; of these, 1388 patients were recruited from 26 sites within India (2012–2016). Abstract

The magnetocaloric effect (MCE), magnetization, specific heat, and magnetostriction measurements were performed in both pulsed and steady high magnetic fields to investigate the magnetocaloric properties of Heusler alloys ${\mathrm{Ni}}_{50\ensuremath{-}x}{\mathrm{Co}}_{x}{\mathrm{Mn}}_{31.5}{\mathrm{Ga}}_{18.5}$ $(x=9 \mathrm{and} 9.7)$. From direct MCE measurements for ${\mathrm{Ni}}_{41}{\mathrm{Co}}_{9}{\mathrm{Mn}}_{31.5}{\mathrm{Ga}}_{18.5}$ up to 56 T, a steep temperature drop was observed for magnetic-field-induced martensitic transformation (MFIMT), designated as inverse MCE. Remarkably, this inverse MCE is apparent not only with MFIMT, but also in the magnetic-field-induced austenite phase. Specific heat measurements under steady high magnetic fields revealed that the magnetic field variation of the electronic entropy plays a dominant role in the unconventional magnetocaloric properties of these materials. First-principles based calculations performed for ${\mathrm{Ni}}_{41}{\mathrm{Co}}_{9}{\mathrm{Mn}}_{31.5}{\mathrm{Ga}}_{18.5}$ and ${\mathrm{Ni}}_{45}{\mathrm{Co}}_{5}{\mathrm{Mn}}_{36.7}{\mathrm{In}}_{13.3}$ revealed that the magnetic-field-induced austenite phase of ${\mathrm{Ni}}_{41}{\mathrm{Co}}_{9}{\mathrm{Mn}}_{31.5}{\mathrm{Ga}}_{18.5}$ is more unstable than that of ${\mathrm{Ni}}_{45}{\mathrm{Co}}_{5}{\mathrm{Mn}}_{36.7}{\mathrm{In}}_{13.3}$ and that it is sensitive to slight tetragonal distortion. We conclude that the inverse MCE in the magnetic-field-induced austenite phase is realized by marked change in the electronic entropy through tetragonal distortion induced by the externally applied magnetic field.

Abstract

The results of optical measurements in the long wavelength infra-red (IR) range on the antiferromagnetic Heusler alloy Ru2CrGe with a Neel temperature of 13 K are reported. At 9 K there is absence of a Drude component in the absorption coefficient indicating the insulating nature of Ru2CrGe at this temperature. The absorption coefficient was observed to increase steeply around 50–70 cm−1 at all temperatures measured in this study.

Abstract

We have developed downsizing a piston-cylinder clamp-cell to measure a specific heat under pressure using a commercial Differential Scanning Calorimeter (Shimadzu DSC-60 plus). The downsized cell with a diameter of 6.0 mm and a height of 5.0 mm has been achieved by a simplification of the cell design and using a high-yield-strength alloy of β-titanium (KOBELCO, KS15-5-3). The cell allows us to achieve pressures of 0.7 GPa at room temperature as our designed value. As a demonstration of the cell, we have measured the specific heat anomaly due to the first-order transition at around 357 K in Ca2RuO4. The pressure variation of the transition temperature indicates that our cell allows us to achieve pressure of ~0.3 GPa. Moreover, we have estimated that a heat capacity of ~0.6 mJ/K is fully possible to measure within β-Titanium cell as absolute accuracy.

Abstract

The thermal expansion and lattice constants of Eu 0.58 Sr 0.42 MnO 3 perovskite have been measured at high pressure. It is found that an anomalous expansion due to metal–insulator transition appeared on heating in the temperature dependence of the thermal expansion and its magnitude decreases with increasing pressure. The metal–insulator transition temperature, T M-I , increases with increasing pressure. The lattice constants, a , b , and c are found to decrease with increasing pressure but there is a large anisotropy in the lattice compression. The linear com are extracted to be 1.5, 1.9 and 0.8 × 1 0 − 3 GPa − 1 for a , b and c -axis, respectively.

Abstract

A high-pressure apparatus which we can use at low temperature and high magnetic field has been designed for making measurements of thermal and transport properties of condensed matter. The details of the apparatus and some examples of the measurements are described and briefly discussed. It is proved that the apparatus is suitable for use up to hydrostatic pressure of ~ 3.5 GPa, down to ~ 2.0 K and up to ~ 11 T.

Abstract

A new hybrid ruthenate-cuprate superconductor RuSr 2 GdCu 2 O 5 has been synthesized. This compound exhibits superconductivity and magnetic ordering around 40 K (=T c ) and 130 K (=T m ). The effect of pressure on the electrical resistance has been measured up to 2.1 GPa in order to make clear the interplay between the magnetism and the superconductivity. It is found that both T c and T m increase with pressure with rates of dT c /dP = 1.9 K GPa -1 and dT m /dP = 5.7 K GPa -1 , which indicates that the superconductivity does not compete with the magnetic ordering. An x-ray diffraction experiment under pressures up to 13 GPa has been carried out in order to clarify the relation between the electronic properties and the lattice compression.

Abstract

Angle-dispersive x-ray diffraction measurements using CuGeO3 (I) and CuGeO3 (III) as the starting materials were carried out to 81 and 31 GPa, respectively, at room temperature. Data for phase (I) show that phase transitions occur at ~7, ~14, and ~22 GPa, respectively, corresponding to (I) → (II), (II) → (II'), and (II') → (VI) transitions, as reported previously. The tetragonal phase (VI) was found to be stable up to 81 GPa, the highest pressure determined in this study. The volume changes at the transition pressures are estimated to be of ~5%, ~0%, and ~14% for (I) → (II), (II) → (II'), and (II') → (VI) transitions, respectively. Data from measurements where phase (III) was the starting material show that phase (III) first changes to phase (IV) at ~7 GPa and then to (IV') at 13.5 GPa, and finally to phase (V) at ~18 GPa, with volume changes of 1.5%, 0%, and 20%, respectively, at the transition pressure. The volume change of 20% at 18 GPa is consistent with the pyroxene–perovskite transition.

Abstract

The results of a study of resistivity and magnetoresistance of the polycrystalline Ce2Fe17 compound under pressure up to 10 kbar at magnetic field up to 9 T are presented. The giant magnetoresistance observed under pressure at 4.2 K is a consequence of metamagnetic transition from antiferromagnetic to ferromagnetic state. It reaches the value of 42% under pressure of 5 kbar.

Abstract

A complex temperature dependence of a.c. susceptibility of Y 2 Fe 17 under high pressures together with recent neutron diffraction studies under pressure proved the instability of the collinear ferromagnetic structure and the development of a non-collinear magnetic arrangement of Fe moments in Y 2 Fe 17 . To study the correlation between magnetic structure and volume in more detail we performed compressibility and linear thermal expansion studies under high pressures up to 100 kbar and 10 kbar, respectively. The compressibility in the paramagnetic state s P (above 10 kbar at room temperature) was determined from the Murnaghan equation of state using the X-ray data, \kappa_{P}=0.80\ {\rm Mbar}^{-1} . The linear thermal expansion and compressibility in the ferromagnetic state at low temperatures are highly anisotropic. As a consequence, the c/a ratio decreases with increasing pressure. The magnetic phase diagram of Y 2 Fe 17 compound was compiled up to 20 kbar.

Abstract

UPtAl exhibits ferromagnetic ordering (T C = 42.5 K) of uranium 5f-electron moments. The magnetic ordering transition is accompanied by an electrical-resistivity (ρ) anomaly at Curie temperature (T C). We report on resistivity measurements performed in a wide temperature range on a UPtAl single crystal under hydrostatic pressures p ≤ 8 GPa. The initial increase of T C with p becomes gradually reduced for p > 2 GPa. The maximum T C value is reached between 4 and 6 GPa that is followed by a decrease of T C beyond 6 GPa. The latter result is attributed to the approaching collapse of U 5f moment ferromagnetism.

Abstract

The effect of pressure on the lattice parameters of R Ni 2 B 2 C ( R = Y, Ho, Er, Tm, Lu) has been measured in order to investigate the relation of T C to axial ratio c/a in the tetragonal lattice or the cohesive properties of these materials. The large anisotropies are observed in the lattice compression curves especially for R =Ho and Tm; the c -axis is more compressible than the a -axis. On the other hand, the anisotropy in the lattice compression is found to be relatively small for R =Y and Lu. The close relation between T C and c/a is pointed out particularly for R =Ho.

Abstract

UPtAl exhibits a ferromagnetic ordering of U magnetic moments at temperatures below T C =42.5 K. The magnetic-ordering transition is accompanied by an anomaly in the temperature dependence of electrical resistivity. This allows us to determine the value of Curie temperature from „ vs. T data that can be measured at very high pressures, at which reliable magnetization measurements are difficult. We report on resistivity measurements performed on an UPtAl single crystal under hydrostatic pressures p h 8 GPa. It was observed that the initial increase of T C with p becomes gradually reduced for p >2 GPa until the maximum T C value of , 48 K is reached between 4 and 6 GPa that is followed by a progressively developing downturn of the T C ( p ) curve. The latter result is attributed to the approaching collapse of the U 5 f -moment ferromagnetism. Low-temperature resistivity data point to a rapidly reduced magnetic anisotropy at highest pressures.

Abstract

We studied the electronic state of UGa3 by measuring the electrical resistivity under pressure. When pressure is applied to UGa3, the Neel temperature TN = 67 K decreases steeply and becomes zero around 2.6 GPa. Correspondingly, the coefficient A in the electrical resistivity ρ = ρ0 + AT2, which is characteristic for Fermi liquid, increases steeply with increasing pressure. Around a magnetic quantum critical point of 2.6 GPa, a non-Fermi-liquid behaviour is found, showing a linear T-dependence of the resistivity, although the Fermi-liquid behaviour recovers above 4 GPa and the A-value decreases with increasing pressure. Furthermore, a resistivity drop, which might be related to superconductivity, was found below 0.3 K at 1.5 GPa, where the residual resistivity ρ0 has a maximum.

Abstract

We studied the crystal structure of CrTe at pressures up to 51 GPa in the temperature range of 35∼300 K by in situ angle-dispersive X-ray diffraction using synchrotron radiation. A pressure-induced transition from the NiAs-type structure (space group P6 3 /mmc ) to the MnP-type one ( Pbnm ) took place at pressures between 13 and 14 GPa. The values of the bulk moduli at room temperature for the low-pressure phase and the high-pressure phase are 43.8(5) and 45.8(5) GPa, respectively. The pressure derivative of the axial ratio c h / a h is determined to be −2.5×10 −3 GPa −1 at room temperature in the low-pressure phase. The relationship between the disappearance of ferromagnetism and structural phase transition is also discussed.

Magnetostriction and magnetization of Eu 0.58 Sr 0.42 MnO 3 were measured under hydrostatic pressures up to 2.0 GPa in order to investigate the effect of pressure on the metal–insulator (M–I) transition, recently investigated by resistivity measurements. At ambient pressure, around 2 T and 6 K, where the field-induced M–I transition was previously reported, the observed magnetostriction was estimated to be 0.04%. With increasing pressure, the saturation magnetization increased at a field of 0.5 T, whereas it decreased at fields of 1.5 and 5.0 T.

Abstract

Pressure-induced structural transitions in MnRhP and MnRhAs were studied by in situ angle-dispersive X-ray diffraction using synchrotron radiation and a diamond anvil cell. A transition from a hexagonal Fe 2 P-type structure to an orthorhombic one was observed in MnRhP at a pressure of approximately 34 GPa. The first-nearest neighboring Mn–Mn interatomic distance at this transition pressure is close to the distance at which the sign of the effective exchange interaction coefficient between Mn atoms turns from positive to negative. A structural phase transition was also observed in MnRhAs at around 26 GPa, although the crystal system of the high-pressure phase was undetermined.

The effect of pressure on the lattice parameters of NiO with a rhombohedral distorted $B1$ structure was investigated up to 141 GPa by in situ angle-dispersive x-ray diffraction using synchrotron radiation, an imaging plate, and a diamond anvil cell. The lattice constants a and c, expressed in the hexagonal lattice, decrease monotonically with increasing pressure. The axial ratio $c/a$ also decreases monotonically with increasing pressure, indicating that the distortion in the rhombohedral [111] direction becomes larger as pressure increases. No phase transition was observed up to 141 GPa. A significant change of pressure coefficient of $c/a$ above 60 GPa, as suggested by a recent theoretical calculation, was not observed.

Abstract

The crystal structure of silicon has been examined by means of X-ray diffraction according to the Pendellosung method. Measurements of the fringe pattern were made for the 111, 220, 113, 004, 331, 224 and 333 diffractions in a temperature range 17–300 K. It is found that the value of the Debye temperature determined from the temperature dependence of the period of Pendellosung fringes is in good agreement with that in the literature except for the first three diffractions; for these with shorter scattering vectors, the value depends on specimen and, in some cases, is smaller than that in the literature and depends on the length of the scattering vector. It is also found that the fringe pattern is stress sensitive, and that the stress can be released at lowest temperatures. This phenomenon of a kind of aging is discussed in connection with a similar effect observed in the high-resolution measurement of the lattice spacing.

Abstract

The Vibrio cholerae fur gene was previously cloned and sequenced. A putative Fur box was identified in the divergent promoters of irgA, a virulence factor of V. cholerae, and irgB, a transcriptional activator of irgA. In this work, V. cholerae Fur was overexpressed in Escherichia coli and purified to approximately 95% homogeneity. The purified protein bound a DNA fragment containing the irgA-irgB promoter in a gel shift assay. The purified protein was used to raise monoclonal and polyclonal antibodies to V. cholerae Fur, and a Fur sandwich enzyme-linked immunosorbent assay was developed to estimate the intracellular abundance of Fur under a variety of growth conditions. The number of Fur molecules per cell during exponential growth was approximately 2,500, which is higher than most measurements for other bacterial repressors but comparable to the intracellular concentration of the leucine-responsive regulatory protein. The number of Fur molecules per cell increased in the late logarithmic and stationary phases. Growth of V. cholerae in low-iron medium did not alter the intracellular abundance of Fur significantly. Growth under microaerophilic conditions resulted in a significant, approximately twofold decrease in the intracellular levels of Fur. The measurements of intracellular Fur abundance indicate that a large amount of this repressor is produced constitutively and that the concentration of Fur in the cell varies by less than a factor of 2 under the conditions studied. We hypothesize that the high constitutive expression of Fur is necessary for its role as an iron-responsive regulator.

Abstract

To clarify whether significant interrelations between hyperinsulinemia and cardiovascular diseases exist in Japanese, we analyzed serum insulin levels, fasting and after oral glucose load, in relation to blood pressure (BP), serum lipids, and incidence of acute myocardial infarction (MI) and stroke in 584 male employees of a railroad company with borderline to mild hypertension (age: 49 +/- 7 years, BP: 139 +/-13/90 +/- 8 mmHg, body mass index: BMI 24.0 +/- 2.8, mean +/- SD). Those who were taking antihypertensive drugs and/or hypoglycemic agents were excluded. The subjects were classified into five groups of comparable size according to sigma IRI (area composed by insulin levels at 0, 1, 2 hours post glucose load). Although averaged age, levels of fasting glucose and HbA1c were not significantly different among quintiles, the hyperinsulinemic groups had higher levels of fasting IRI and sigma glucose. Multiple regression analysis revealed that sigma IRI was associated positively with BMI, sigma glucose, systolic BP, total cholesterol, and negatively with HDL-cholesterol. During 7-year follow-up 7 cases with MI and 15 cases with stroke were registered. The subjects with MI, but not with stroke were involved in higher deciles of insulin levels. These results indicate that the insulin resistance syndrome exists in Japanese middle-aged men with borderline to mild hypertension. The association with cardiovascular morbid events appeared to be evident in coronary heart disease but not in cerebrovascular disease.

Aim To assess blood glucose control over 24 h and the safety of teneligliptin 10 and 20 mg, a novel dipeptidyl peptidase-4 inhibitor, in Japanese patients with type 2 diabetes mellitus inadequately controlled with diet and exercise. Methods Ninety-nine patients were administered teneligliptin 10 or 20 mg or placebo before breakfast for 4 weeks in a randomized, double-blind, placebo-controlled, parallel-group study. Results Both teneligliptin-treated groups showed significantly smaller 2-h postprandial glucose (2-h PPG), 24-h mean glucose and fasting plasma glucose values than the placebo group. A new hybrid ruthenate–cuprate superconductor RuSm1.4Ce0.6Sr2Cu2O10 was synthesized. This compound also exhibits magnetic ordering around 130 K . Electrical resistance measurement under high pressure has been carried out in order to clarify the interplay between magnetism and superconductivity. It is found that Tc increases but the magnetic ordering temperature decreases with pressure, i.e., the superconductivity competes with magnetic ordering.

Abstract

The disulphate ester of ursodeoxycholyl-p-aminobenzoic acid (PABA-UCDA) was synthesised and compared with PABA-UDCA for its use in detection of intestinal bacteria. This compound, PABA-UDCA disulphate, had characters in common with PABA-UDCA in that it was deconjugated by cholylglycine hydrolase to release free PABA and bacteria that split glycocholic acid deconjugated PABA-UDCA disulphate. Further, in rat experiments urinary excretions of PABA were measured for six hours after oral administration of 15 mg PABA-UDCA disulphate. Ten control rats excreted (mean (SE) 188.2 (13.6) micrograms of PABA; 10 rats with an intestinal stagnant loop excreted more (530.1 (30.1) micrograms; p < 0.001): whereas 10 rats in each of three groups pretreated by oral administration of various antibiotics excreted less (polymixin B+tinidazole, 4.9 (1.6) micrograms; kanamycin, 31.0 (4.7) micrograms; clindamycin 40.9 (5.5) micrograms; p < 0.001). By contrast with PABA-UDCA, PABA-UDCA disulphate was not actively absorbed from any part of the small intestine in everted gut sac experiments, and showed poor recovery from bile after its intraileal instillation in rats. This indicated that PABA-UDCA disulphate is a single pass type substance in the gut and its oral administration test reflects the sum of the activities of bacteria in the small intestine and colon. The disulphate was easily soluble in water and this allowed its application in an in vitro test involving PABA-UDCA disulphate incubation with intraperitoneal pus (PABA-UDCA disulphate incubation test) from patients with peritonitis. This test was carried out on six patients with peritonitis, and the severity of bacterial peritonitis was expressed quantitatively. From the results obtained PABA-UDCA disulphate was considered a good material to detect intestinal bacteria.

The influence of hydrostatic pressure on electrical resistance of a perovskite manganese oxide (La 0.1 Ce 0.4 Sr 0.5 )MnO 3 has been investigated under pressures up to 1.8 GPa in the temperature range of 77–320 K. With application of pressure, Curie temperature ( T C ) increases at a rate of d T C /d p =+1.9 K/GPa . It is also found that the coefficient of −log T term originated from Kondo-like effect decreases with increasing pressure and the resistance maximum temperature ( T max ) shifts largely to lower temperature at a rate of d T max /d p =−86 K/GPa .

Abstract

Although the number of patients undergoing laparoscopy-assisted distal gastrectomy (LADG) has been increasing, a prospective study with a sample size sufficient to investigate the benefit of LADG has never been reported. We conducted a multi-institutional phase II trial to evaluate the safety of LADG with nodal dissection for clinical stage I gastric cancer patients.The subjects comprised patients with clinical stage I gastric cancer who were able to undergo a distal gastrectomy. LADG with D1 plus suprapancreatic node dissection was performed. The primary endpoint was the proportion of patients who developed either anastomotic leakage or a pancreatic fistula. The secondary endpoints included surgical morbidity and short-term clinical outcome.Between November 2007 and September 2008, 176 eligible patients were enrolled. The proportion of patients who developed anastomotic leakage or a pancreatic fistula was 1.7%. The overall proportion of in-hospital grade 3 or 4 adverse events was 5.1%. The short-term clinical outcomes were as follows: 43.2% of the patients requested an analgesic on postoperative days 5-10; the median time from surgery until the first episode of flatus was 2 days; and 88 patients (50.0%) had a body temperature of 38 °C or higher during their hospital stay.This trial confirmed the safety of LADG performed by credentialed surgeons in terms of the incidence of anastomotic leakage or pancreatic fistula formation. A phase III trial (JCOG 0912) to confirm the noninferiority of LADG to an open gastrectomy in terms of overall survival is ongoing.

Abstract

We report a 29-year-old Japanese female with severe hypertension and unilateral renal dysplasia, which is rarely found in adults. The involved kidney was surgically removed and then the blood pressure fell to borderline levels without any changes in plasma renin activity or plasma aldosterone concentration. Histological examinations revealed typical renal dysplasia without hyperplasia of the juxtaglomerular apparatus. Unilateral renal dysplasia may be a cause of secondary hypertension in adults, though the mechanisms are still unknown.

Abstract

Some studies have demonstrated the benefit of blood glucose control as close as possible to physiological conditions. Not enough reports have investigated in detail the 24-h plasma profiles of insulin/glucose/C-peptide. Here we investigated the 24-h plasma profiles of physiological insulin/glucose/C-peptide in healthy Japanese adults.In order to evaluate the 24-h profiles of physiological insulin/glucose/C-peptide profiles, 42 blood samples were taken from each subject in our group of healthy Japanese volunteers to measure the 24-h profile with three standardized meals.Plasma glucose and insulin increased rapidly followed by a rapid decrease after each meal with little variation at night. The average peak values of insulin after each meal were as follows: 426.20 pmol/L (breakfast), 373.75 pmol/L (lunch), and 410.28 pmol/L (dinner). The average times to peak insulin were 0.651 h (breakfast), 0.677 h (lunch), and 0.689 h (dinner). The corresponding average maximum postprandial plasma glucose levels were 8.39 mmol/L (breakfast), 8.77 mmol/L (lunch), and 8.74 mmol/L (dinner). The average times to peak glucose were 0.738 h (breakfast), 0.650 h (lunch), and 0.625 h (dinner). The average maximum postprandial C-peptide levels were 2.64 nmol/L (breakfast), 2.55 nmol/L (lunch), and 2.67 nmol/L (dinner). No major differences were found in these parameters between the Caucasian and Japanese populations.This is the first investigation to measure the 24-h profiles of insulin/glucose/C-peptide in healthy Japanese volunteers with standardized meals. It is hoped this information will provide useful reference for future research and clinical management.

Abstract

The value of inversion-recovery (IR) sequences in the diagnosis and staging of prostatic carcinoma with magnetic resonance (MR) imaging was studied. Twenty-six patients with carcinoma of the prostate were imaged at 1.5 T with an endorectal surface coil and with a variety of IR sequences and a set of spin-echo (SE) sequences for comparison. Ex vivo prostate specimens were imaged again at the same field strength. The two images were correlated with histologic sections. Cancer was identified with MR imaging in 96% of patients. Of the tumors more than 4 mm in diameter, 87% were identified on T2-weighted SE images, whereas only 26% were identified on IR images. However, IR images may be more useful in local staging of carcinoma. Gross capsular infiltration was present in only two patients; however, it was detectable (and excluded in five other patients) by means of IR images. It was not detectable on SE images. The high quality of images obtained with the endorectal coil was confirmed. The authors conclude that addition of the IR sequence to MR imaging with the endorectal coil may improve the usefulness of this examination.

Abstract

Thrombus propagation on disrupted plaque is a major cause of acute coronary events and serious complication after coronary intervention. 5-Hydroxytryptamine (5-HT) is a potent vasoactive and platelet-aggregating substance that is predominantly mediated by 5-HT2A receptor. However, the roles of 5-HT2A receptor in occlusive thrombus formation on disrupted plaque remain obscure.We investigated the role of 5-HT2A receptor in thrombus formation using a rabbit model of repeated balloon-injury.Three weeks after a first balloon-injury of the femoral arteries, luminal diameter, neointimal growth, and vasoconstriction by 5-HT in vitro were examined. Thrombus propagation and the role of 5-HT2A receptor after a second balloon-injury were evaluated using sarpogrelate, a selective 5-HT2A receptor antagonist.Three weeks after the first balloon-injury, luminal stenosis was evident in the femoral arteries, where the neointima expressed tissue factor and 5-HT2A receptor. The hypercontractile response of the stenotic arteries to 5-HT was significantly reduced by sarpogrelate. Balloon-injury of the neointima with substantially reduced blood flow promoted the formation of occlusive thrombus that was immunoreactive against glycoprotein IIb-IIIa, 5-HT2A receptor and fibrin. Intravenous injection of sarpogrelate significantly inhibited ex vivo platelet aggregation induced by adenosine 5'-diphosphate, thrombin and collagen alone as well as with 5-HT, and significantly prevented occlusive thrombus formation in vivo.The 5-HT2A receptor appears to play a crucial role in occlusive thrombus formation in diseased arteries via platelet aggregation and vasoconstriction. Inhibition of 5-HT2A receptor might help reduce the onset of acute coronary events and of acute coronary occlusion after the intervention.

Abstract

The effects of hydrostatic pressure and magnetic field on the electrical resistance of the Kondo-like perovskite manganese oxide, ${\mathrm{La}}_{0.1}{\mathrm{Ce}}_{0.4}{\mathrm{Sr}}_{0.5}\mathrm{Mn}{\mathrm{O}}_{3}$ with a ferrimagnetic ground state, have been investigated up to $2.1\phantom{\rule{0.3em}{0ex}}\mathrm{GPa}$ and $9\phantom{\rule{0.3em}{0ex}}\mathrm{T}$. In this compound, the Mn moments undergo a double exchange mediated ferromagnetic ordering at ${T}_{C}\ensuremath{\sim}280\phantom{\rule{0.3em}{0ex}}\mathrm{K}$ and there is a resistance maximum, ${T}_{\mathrm{max}}$ at about $130\phantom{\rule{0.3em}{0ex}}\mathrm{K}$, which is correlated with an antiferromagnetic ordering of cerium with respect to the Mn-sublattice moments. Under pressure, the ${T}_{\mathrm{max}}$ shifts to lower temperatures at a rate of $d{T}_{\mathrm{max}}∕dP=\ensuremath{-}162\phantom{\rule{0.3em}{0ex}}\mathrm{K}∕\mathrm{GPa}$ and disappears at a critical pressure ${P}_{c}\ensuremath{\sim}0.9\phantom{\rule{0.3em}{0ex}}\mathrm{GPa}$. Further, the coefficient, $m$ of $\ensuremath{-}\mathrm{ln}\phantom{\rule{0.2em}{0ex}}T$ term due to Kondo scattering decreases linearly with increase of pressure showing an inflection point in the vicinity of ${P}_{c}$. These results suggest that cerium undergoes a transition from ${\mathrm{Ce}}^{3+}$ state to a ${\mathrm{Ce}}^{4+}∕{\mathrm{Ce}}^{3+}$ mixed valence state under pressure. In contrast to the pressure effect, the applied magnetic field shifts ${T}_{\mathrm{max}}$ to higher temperatures, presumably due to enhanced ferromagnetic Mn moments.

Abstract

Adrenomedullin is a recently discovered peptide that was first purified from phaeochromocytoma tissue and has marked vasodilatory activity, causing hypotension. In thyrotoxicosis, various haemodynamic changes are observed, including an increase in cardiac output and heart rate with a concomitant decrease in peripheral vascular resistance. To evaluate the mechanism underlying these haemodynamic changes in thyrotoxicosis, we measured the plasma adrenomedullin concentration in thyrotoxic patients with Graves' disease. The plasma concentration of adrenomedullin was elevated in hyperthyroid patients (14.7 +/- 5.7 pmol L-1) compared with euthyroid control subjects (5.6 +/- 1.3 pmol L-1) (P < 0.001). The correlation between the plasma adrenomedullin concentration and serum free thyroid hormone levels was marginally significant. The mean blood pressure was relatively low in the face of an elevated plasma adrenomedullin level. Adrenomedullin may therefore be responsible for the vasodilatation observed in thyrotoxicosis.

Abstract

Angiosarcomas rarely develop within a peripheral nerve or a peripheral nerve sheath tumor. A 68-year-old woman was admitted for right hemothorax. She had suffered from the left thoracic empyema and the right chest wall tumor which had been regarded as schwannoma clinically. Anemia got serious due to continuous bloody effusion. Although bleeding point was not detected by thoracotomy. The right chest wall tumor was resected. Postoperatively bloody effusion still drained continuously, she died ten days after the operation. Resected tumor was almost necrotized. The tumor was diagnosed as angiosarcoma pathologically, in which a part of schwannoma component resided. It is indicated that angiosarcoma in this case arised from degenerated schwannoma. It is necessary to consider angiosarcoma, rarely arised from degenerated schwannoma, during conservative, long-term observation on the care of schwannoma.
