However, patient adherence can be impaired by local tolerability in the first 1– 2 weeks of treatment. Mineral 89 Probiotic Fractions® (M89PF) containing Vichy volcanic mineralizing water®, probiotic fractions, hyaluronic acid, niacinamide and tocopherol was developed to fulfill the need for adjunctive products that can reinforce skin barrier and manage retinoid induced irritation.Patients and Methods: The study included 38 women, aged 44– 60 years, phototype II–VI, applying 0.025% tretinoin gel once nightly for 84 days. For 28 days, one hemi face was treated with M89PF and sunscreen SPF 50+ while other hemi face received sunscreen only. Then, M89PF application was changed to full face. Evaluations were performed at days 0, 7, 28 and 84. Erythema, dryness, fine lines, skin tone, radiance and pore appearance were assessed by a dermatologist. Tolerability was evaluated through self-assessment questionnaire. Skin hydration levels, inflammatory and oxidative stress biomarkers were analyzed by immunological assay: Interleukin(IL)-8, IL1-alpha, IL1-Receptor Antagonist (IL-1Ra), Prostaglandin E2 (PGE2), Catalase and Superoxide Dismutase (SOD).Results: Hemiface analysis showed that erythema, fine lines, skin tone, radiance, pore appearance, hydration, tightness, dryness, burning, itching and stinging sensations were improved (p< 0.05) on the M89PF side. At full face analysis on D84, erythema, fine lines, skin tone, radiance and pore appearance were improved compared to D0 (p< 0.001). Tightness, dryness, burning, itching and stinging were reduced when compared to D7 (p< 0.05). Dermatology Life Quality Index (DLQI) and Skindex 16 showed improvement in quality of life (p< 0.05). IL-1RA increased at D28 (p=0.003) and PGE2 decreased at D28 and D84 compared to D0 (p< 0.01).Conclusion: M89PF reduced retinoid induced irritation with a good tolerability profile and, used as an adjunct to topical tretinoin, significantly improved skin hydration, erythema, fine lines, skin tone, radiance and pore appearance.Keywords: skin barrier, irritation, Vichy volcanic mineralizing water®, hyaluronic acid, niacinamide, tocopherol

Abstract

The results of numerical and experimental research conducted in the Laboratory for Hydraulic Machinery Construction of Peter the Great St. Petersburg Polytechnic University are presented. The research is aimed at studying the effects of an impeller radial clearance and rim on the energy characteristic of low-pressure axial pumps of the specific speed ns≈600. It is shown that these design features of a flow duct have significant effects on stage parameters, and they have to be accounted for when verifying design and experimental characteristics of axial pumps.

Abstract

This paper proposes a methodology for calculating hydraulic shock, considering the occurrence of cavitation caverns in the open source OpenFOAM software package, using it cavitatingFoam solver. The proposed calculation method is verified by experimental data for different experimental cases: flow with no cavitation, limited cavitation, moderate cavitation, and severe cavitation. Also received results were compared with simulations in commercial software package FlowMaster, where the proposed method is compared with the existing method of calculation by the method of characteristics. Conducted investigations confirmed the possibility of using developed method to predict the effect of cavitation on network operation in different transition modes. [ABSTRACT FROM AUTHOR]

Abstract

We present a combined report on the results of three editions of the Cell Tracking Challenge, an ongoing initiative aimed at promoting the development and objective evaluation of cell segmentation and tracking algorithms. With 21 participating algorithms and a data repository consisting of 13 data sets from various microscopy modalities, the challenge displays today's state-of-the-art methodology in the field. We analyzed the challenge results using performance measures for segmentation and tracking that rank all participating methods. We also analyzed the performance of all of the algorithms in terms of biological measures and practical usability. Although some methods scored high in all technical aspects, none obtained fully correct solutions. We found that methods that either take prior information into account using learning strategies or analyze cells in a global spatiotemporal video context performed better than other methods under the segmentation and tracking scenarios included in the challenge.

31. Differential replication of a single, UV-induced lesion in the leading or lagging strand by a human cell extract: fork uncoupling or gap formation

Abstract

We have constructed simian virus 40 minireplicons containing uniquely placed cis,syn-thymine dimers (T <> T) for the analysis of leading- and lagging-strand bypass replication. Assaying for replication in a human cell-free extract through the analysis of full-size labeled product molecules and restriction fragments spanning the T <> T site resulted in the following findings: (i) The primary site of synthesis blockage with T <> T in either the leading or lagging strand was one nucleotide before the lesion. (ii) Replicative bypass of T <> T was detected in both leading and lagging strands. The efficiency of synthesis past T <> T was 22% for leading-strand T <> T and 13% for lagging-strand T <> T. (iii) The lagging-strand T <> T resulted in blocked retrograde synthesis with the replication fork proceeding past the lesion, resulting in daughter molecules containing small gaps (form II' DNA). (iv) With T <> T in the leading-strand template, both the leading and lagging strands were blocked, representing a stalled replication fork. Uncoupling of the concerted synthesis of the two strands of the replication fork was observed, resulting in preferential elongation of the undamaged lagging strand. These data support a model of selective reinitiation downstream from the lesion on lagging strands due to Okazaki synthesis, with no reinitiation close to the damage site on leading strands [Meneghini, R. & Hanawalt, P.C. Abstract

By using a human cell-free system capable of nucleotide excision repair, a synthetic substrate consisting of a plasmid containing four thymidine dimers at unique locations, and deoxyribonucleoside 5'-[alpha-thio]triphosphates for repair synthesis, we obtained DNA fragments containing repair patches with phosphorothioate linkages. Based on the resistance of these linkages to digestion by exonuclease III and their sensitivity to cleavage by I2, we were able to delineate the borders of the repair patch to single-nucleotide resolution and found an asymmetric patch with sharp boundaries. That the repair patch was produced by filling in a gap generated by an excision nuclease and not by nick-translation was confirmed by the finding that the thymidine dimer was released in a 27- to 29-nucleotide oligomer.

Abstract

1. Rapamycin is metabolically transformed in rat liver microsomes to 3,4- and 5,6-dihydrodiol metabolites under the influence of the cytochrome P-450 mixed function oxygenase system. These metabolites were produced from dexamethasone-induced as well as from non-induced rat liver microsomes. The comparison of the ion spray mass spectra of the 5,6-dihydrodiol with the 3,4-dihydrodiol of rapamycin shows clearly that dihydrodiols were formed in two distinct positions of rapamycin. 2. FAB mass spectra as well as electrospray mass spectra of two additional peaks isolated from the same chromatographic run confirm the presence of a 3,4-dihydrodiol metabolite of rapamycin as also strongly suggested by UV spectra. Hplc reinjection of each individual peak always resulted in chromatograms showing a combination of the same three peaks and therefore are to be considered as tautomers of the 3,4-dihydrodiol of rapamycin. 3. These tautomeric conformations were found to have no immunosuppressive potency, most probably due to important structural and stereochemical modifications of the rapamycin binding domain to the binding protein (FKBP-12) and/or to important metabolic structural modifications of rapamycin effector domain.

47. Characteristics That Promote Transmission of Staphylococcus aureusin German Nursing Homes

AbstractObjective:To determine factors that influence transmission of Staphylococcus aureusin nursing homes in the Rhine-Neckar region of southern Germany.Design:Ecologie study.Setting:Forty-seven nursing homes in the region.Participants:Residents of the approached nursing homes who agreed to participate.Methods:Personal data and swabs of the nares were collected from participants. Swabs were examined for growth of S. aureus.All S. aureusisolates were typed using pulsed-field gel electrophoresis (PFGE). Transmission rates were calculated by dividing the number of transmissions (ie, cases in which two inhabitants shared the same PFGE type) by the number of S. aureuscarriers. Characteristics of the nursing homes were correlated with a home's transmission rate.Results:In each nursing home, 12% to 54% of the residents were colonized with S. aureus.The transmission rates for the 47 nursing homes ranged from 0% to 70%. A linear regression model revealed that a stay in the nursing home of longer than 6 months and accommodation in a room with 3 or more beds were positively associated with the transmission rate. Receipt of antibiotics during the 4 weeks preceding the study was negatively associated with transmission.Conclusions:Stays beyond 6 months and accommodation in rooms with multiple beds are important for the transmission of S. aureus.One way to reduce transmission would be to design facilities with single and double rooms. However, the social needs of the residents must be evaluated and respected.

All swabs were examined for growth of MRSA All S. aureusisolates underwent oxacillin susceptibility testing and polymerase chain reaction for demonstration of the meek gene. All MRSA isolates were typed using pulsed-field gel electrophoresis after digestion with SmaI.Results:Swabs from 3,236 nursing home residents yielded 36 MRSA strains, contributing to a prevalence rate of 1.1%. Significant risk factors for MRSA carriage in the multivariate analysis were the presence of wounds or urinary catheters, limited mobility, admission to a hospital during the preceding 3 months, or stay in a medium-size nursing home. One predominant MRSA strain could be detected in 30 of the 36 MRSA carriers.Conclusions:The prevalence of MRSA in German nursing homes is still low. These residents seemed to acquire their MRSA in the hospital and transfer it to their nursing home. Apart from well-known risk factors for the acquisition of MRSA we identified the size of the nursing home as an independent risk factor. This might be due to an increased use of antimicrobials in nursing homes of a certain size.
