Pig producers in Europe adopt different production methods for male pig fattening. More than half of the animals are surgically castrated. The different interpretations of animal welfare in different countries lead to market differentiation and economically different production conditions, which do not restrict trade, but economically lead to drastic competitive changes for local producers. While the Netherlands has already implemented surgical castration for the export market, using CO2 narcosis (NL), Denmark and Germany are each introducing their strategies with local anesthesia (DK) and isoflurane anesthesia (DE), respectively. Using typical pig farms from the agri benchmark Pig Network, the additional costs and economic impacts of animal welfare regulations are calculated. In Germany, isoflurane anesthesia increases costs by EUR 28.54 to EUR 49.86 per sow, or EUR 1.93 to EUR 3.81 per male piglet. This corresponds to a cost increase of around 5% per piglet. In Denmark, the costs of local anesthesia with procaine increase more moderately by EUR 3.55 to EUR 5.05 per sow, or around EUR 0.30 per male piglet. The cost increase here is less than 1% per piglet. The additional costs are leading to a loss of competitiveness for Denmark and Germany compared to the Netherlands. However, Germany is also losing profit significantly compared to Denmark. This study highlights that animal welfare regulations can negatively impact the competitiveness of the pig sector. It emphasizes the need for countries to implement such regulations carefully, ensuring that they do not lead to the loss of production or international competitiveness. Abstract Background Advanced prostate cancer leads to many symptoms, notably bone pain and lower urinary tract symptoms (LUTs); however, the degree and duration of pain relief, changes in LUTs severity and underlying factors associated with the extent of symptom relief remain inadequately understood. Surgical castration has proven effective in relieving both bone pain and urinary symptoms for metastatic prostate cancer patients. Objective To determine the extent and pattern of symptom relief in advanced prostate cancer patients following surgical castration at Muhimbili National Hospital (MNH). Methods We conducted a prospective cohort study for a period of 6 months involving men with advanced Prostate cancer (PCa) undergoing surgical castration at MNH and followed them for 30 days. The international prostate symptoms score tool was used to assess changes in LUTs, and the pain rating scale was used for assessing changes in bone pain symptoms before and after surgery. Logistic regression model was used to determine factors associated with complete bone pain relief. Results A total of 210 participants with a mean age of 72.3 years were recruited. The expression of genes involved in testis-derived hormone metabolism was altered following surgical castration, but the upstream regulatory factors and underlying mechanism remain unclear. In this study, we systematically profiled chromatin accessibility and transcriptional dynamics in liver tissue of castrated and intact full-sibling Yorkshire pigs. First, we identified 897 differentially expressed genes and 6864 differential accessible regions (DARs) using RNA- and ATAC-seq. By integrating the RNA- and ATAC-seq results, 227 genes were identified, and a significant positive correlation was revealed between differential gene expression and the ATAC-seq signal. We constructed a transcription factor regulatory network after motif analysis of DARs and identified a candidate transcription factor (TF) SP1 that targeted the HSD3B1 gene, which was responsible for the metabolism of androstenone. Subsequently, we annotated DARs by incorporating H3K27ac ChIP-seq data, marking 2234 typical enhancers and 245 super enhancers involved in the regulation of all testis-derived hormones. Among these, four typical enhancers associated with HSD3B1 were identified. Furthermore, an in-depth investigation was conducted on the androstenone-related enhancers, and an androstenone-related mutation was identified in a newfound candidatetypical enhancer (andEN) with dual-luciferase assays. These findings provide further insights into how enhancers function as links between phenotypic and non-coding area variations. The discovery of upstream TF and enhancers of HSD3B1 contributes to understanding the regulatory networks of androstenone metabolism and provides an important foundation for improving pork quality. Androgen from the testis and weak androgens from the adrenal cortex may interact with each other and affect their synthesis and secretion due to their similar functions. The purpose of this study was to investigate the compensatory effect of adrenal in rats after immunocastration and surgical castration, and the interaction between the hypothalamic–pituitary–testis (HPT) axis and the hypothalamic–pituitary–adrenal (HPA) axis. 24 male SD rats aged 8 weeks were randomly divided into three groups and accepted treatments: surgical castration group, immunocastration group and control group. In both surgical castration and immunocastration groups, the secretion of adrenocorticotropic hormone (ACTH) and dehydroepiandrosterone (DHEA) hormones was significantly increased compared with the control group (p < 0.05). In the HPT axis of the immunocastration group, the KISS1 expression was up-regulated, whereas GPR54, LH and LHR expression were down-regulated (p < 0.05). The expression levels of CRH, POMC and MC2R genes were also significantly up-regulated (p < 0.05). In addition, in the immunocastration group, the expression of adrenal LHR mRNA expression was decreased (p < 0.05). The expression of HPT axis genes and adrenal LHR were up-regulated in the surgical castration group (p < 0.05). These results show that in both immunocastration and surgical castration, adrenal androgen is increased, suggesting that the adrenal gland plays a compensatory role. Castration of male animals is intended to produce high-enhance quality of animal meat, prevent unpleasant taste, reduce aggressive behavior, and manage overbreeding. Over the years, Tranditional methods of mechanical and surgical castration have been employed over the years, but they fall short of meeting animal welfare requirements due to the associated risk of infection, pain, and stress. Immunocastration, specifically Gonadotropin-releasing hormone (GnRH)-immunocastration, targeting the hypothalamic–pituitary-testis (HPT) axis, has emerged as an animal-friendly alternative to surgical castration, effectively addressing these issues. This review seeks to systematically summarize the principles, development, current applications and challenges of GnRH-immunocastration, offering insights into its role in promoting animal welfare.

Simple Summary: The use of boars for breeding purposes is common on small farms in rural regions of Italy. These animals are butchered at the end of their reproductive life for the production of charcuterie. In this regard, orchiectomy is required to remove boar taint caused by testosterone metabolites. The purpose of this study is to validate the efficacy of a safe, rapid and inexpensive castration procedure. To accomplish this objective, 91 boars were orchiectomised using the suture ligation technique. Following castration, characteristics of the severed ductus deference were evaluated in order to understand how much force would be required to tie knots without breaking the deferens and achieving proper haemostasis. After identifying a correlation between tensile strength and the age and weight of boars, a sutureless technique was successfully applied to 20 animals. The heterogeneity of Italian manufacturing processes results in the production of a large variety of pork products. In Sardinia, boars are raised and butchered to produce charcuterie. These animals are castrated before slaughter as androstenone would otherwise taint the meat, rendering it unfit for human consumption. However, to date, the literature concerning surgical orchiectomy in adult boars is limited. The goal of this study is to assess whether a sutureless swine orchiectomy procedure is feasible. Additionally, this study aims to determine the appropriate traction force needed to tie knots in the deferens duct of pigs of different weights and ages. Two groups were created: the first (n = 91) underwent orchiectomy by suture ligation; the second (n = 20) was castrated using the sutureless technique. Deferens ducts of animals in the first group (n = 182) were collected following castration, and their tensile strength was measured. Pearson's linear correlation was used to determine the relationship between the maximum traction force and weight and age groups. A correlation of 0.99 and 0.96 was shown between traction force and age and traction force and weight, respectively. In accordance with these results, sutureless castration was performed on 20 animals, calibrating the pulling force needed according to the age and weight of the boars. Unesp-Botucatu Cattle Pain Scale (UCAPS) is widely used in experimental settings, however the high number of UCAPS behaviors might represent a barrier to its implementation in the farm's or hospital's routine. We aimed to identify a smaller combination of UCAPS behaviors that could be used as behavioral red flags for optimizing the acute pain diagnosis in cattle. We hypothesize that a specific set of UCAPS behaviors might be used as behavioral red flags for pain. This would represent a quick and simple pain evaluation and might optimize the acute pain assessment in large-scale systems. Data from two previous studies regarding UCAPS assessments before (pain free condition) and after (painful condition) surgical castration of 59 male cattle was used. We fitted a decision tree, resulting in a logic with two behaviors that we used as behavioral red flags. The logic adapted from the decision tree considered the painful diagnosis when the Activity was scored 2. When Activity was scored less than 2, but Locomotion was scored 1 or 2, the diagnosis was also considered positive for pain. When Activity was below 2 and Locomotion was 0, the diagnosis was considered free pain. Behavioral red flags had an area under the curve of 95.95 % for predicting UCAPS diagnosis and 94.13 % for predicting overall pain free and painful conditions. In the United States (US), surgical castration of male piglets is typically performed without any form of analgesia. This may raise concerns with the public; however, there is no information regarding current public knowledge on swine industry practices in the US. In this study we gained insight into public knowledge and perception on castration with and without analgesia in comparison to knowledge of industry stakeholders on these same topics. Through an online survey, 119 respondents were asked four questions about castration in the US swine industry. Industry respondents were contacted via social media and networking. The general public sample was accessed through Mechanical Turk. Survey responses were categorised by experience (industry vs public). Industry respondents were more aware of practices compared to the general public. Most public respondents were unaware of castration practices and the lack of analgesia use. Respondents from rural communities were more aware of castration practices than (sub)urban communities and more aware of analgesia use than those from urban communities. Those with more education had greater awareness of castration practices (occurrence not frequency). Based on the results from this first US sample, knowledge on industry practices was especially lacking for public respondents, but also for a minority of industry respondents, indicating opportunities for education and further research on the topic.

Thymic atrophy affects T cell generation and migration to the periphery, thereby affecting T cell pool diversity. However, the mechanisms underlying thymic atrophy have not been fully elucidated. Here, gonadotropin-releasing hormone (GnRH) immunization and surgical castration did not affect thymocyte proliferation, but significantly reduced the apoptosis and increased the survival rate of CD4-CD8-, CD4+CD8+, CD4+CD8-, and CD4-CD8+ thymocytes. Following testosterone supplementation in rats subjected to GnRH immunization and surgical castration, thymocyte proliferation remained unchange, but the apoptosis of CD4-CD8-, CD4+CD8+, CD4+CD8-, and CD4-CD8+ thymocytes significantly increased. Transcriptome analyses of the thymus after GnRH immunization and surgical castration showed a significant reduction in the thymus's response to corticosterone. Cholesterol metabolism and the synthesis and secretion of corticosterone were significantly reduced. Analysis of the enzyme levels involved in the corticosterone synthesis pathway revealed that corticosterone synthesis in thymocytes was significantly reduced after GnRH immunization and surgical castration, whereas exogenous testosterone supplementation relieved this process. Testosterone promoted thymocyte apoptosis in a concentration-dependent manner, and induced corticosterone secretion in vitro. Blocking the intracellular androgen receptor (AR) signaling pathway did not significantly affect thymocyte apoptosis, but blocking the glucocorticoid receptor (GR) signaling pathway significantly reduced it. Our findings indicate that testosterone regulates thymus remodeling by affecting corticosterone synthesis in thymocytes, which activates GR signal transduction and promotes thymocyte apoptosis. • Castration reduces thymocyte apoptosis and decreases the expression of CS-related proteins in thymocytes. • Testosterone promotes the apoptosis of thymocytes and increases the expression of CS-related proteins. • Testosterone induces thymocyte apoptosis in a concentration-dependent manner in vitro. • Testosterone promotes CS secretion, which is dependent on GR signal transduction to induce thymocyte apoptosis in vitro. In order to determine the influence of the castration method on the indicators of the meat cuts of pig carcasses, two groups of 74 boars were selected. Surgical castration was carried out in one of them, and immunological vaccination method was used in the other group. Both surgically castrated and immunocastrated boars were reared and fattened under the same conditions. And at the end of fattening, they were slaughtered and their carcasses evaluated for weight and proportion of meat cuts separately in the cervical-scapular, back-lumbar and pelvicfemoral thirds. The result of the assessment was the establishment of a significantly higher value of the weight indicators of such meat cuts as single-grade pork from the neck by 0.3 kg or 20.0% (P <0.01) and the weight of lard with skin by 0.6 kg or 13.64 % (P <0.05) in the shoulder-scapular third of the carcass and single-grade pork per 0.2 kg or 15.38% (P <0.05) in the pelvic-femoral third of the carcass in immunocastrated pigs. A significant difference in the weight and content of other meat cuts of carcasses was not established with the use of immuno- and surgical castration of pigs. [ABSTRACT FROM AUTHOR]

The clinical efficacy of the use of immunobiological castration was studied in comparison with boars in which orchidectomy was performed by open method «on a ligature», as well as in non-castrated animals. It was found that the disadvantage of surgical castration is the risk of bleeding, the development of purulent inflammatory process (edges of the surgical wound, scrotal tissues, stump, common vaginal membrane), as well as intra- and postoperative hernias, which are accompanied by prolapse of the omentum, intestines, and bladder. The low incidence of these complications is offset by the high death rate caused by them. A relatively high level of cryptorchidism was established on the farm, among which bilateral was 27.6%. As a result of the use of immunological castration, in comparison with surgical castration, and with non-castrated boars, the average daily gains were increased, the fattening period was reduced and feed costs were reduced against the background of feed conversion increase by 14 – 27%. The use of Improvac® (Pfizer Ltd) led to better assimilation of feed, an increase in the amount of meat on the carcass, in particular, lean meat by 12 – 16%, lumbar depth, the protein content in meat and a decrease of the fat percentage and fat thickness, eliminating specific odor, and reduction in the fattening period, as well as costs for feeding. In particular, compared to non-castrated boars, the weight of chilled half carcasses increased by 1.2 times (P < 0.05). Immunological castration led to atrophy of the testicles: their weight, length and width were smaller compared to non-castrated boars, which again confirms a decrease in the level of boars’ sex hormones. It is important to use Improvac® in breeding boars that have exhausted their potential, eliminating the need to keep them for about three months to reduce their meat and lard smell. Reducing the period of disappearance of the odor of products obtained from boars led to a reduction in the cost of feeding, maintenance, and veterinary care. Production indicators justify the economic feasibility of using immunological castration with Improvac® in order to obtain additional income, given the absence of probable complications associated with the conventional surgical procedure.

Integrated data analysis shown that there existed a selective hyperfunction of SMG ribosome and mitochondrion in ORC but not in IM, which might be associated with more severe degeneration of SMG in ORC than in IM. Conclusions: Our findings suggested that both surgical castration and immunocastration caused SMG degeneration by disrupting epithelial cell development, angiogenesis, anatomical structure morphogenesis and enhancing cell death. But, surgical castration selectively induced hyperfunction of SMG ribosome and mitochondrion, thus causing more severe degeneration of SMG than immunocastration. [ABSTRACT FROM AUTHOR]

Abstract Background Castration is a husbandry practice raising important questions on the welfare and physiological status of farm animals. Searching for effective castration methods that minimally compromise the body physiology is worthy of attention. Therefore, this study aimed to evaluate the differential response of biological systems in donkeys to surgical castration versus the chemical one by CaCl2 with special emphasis on stress, lipid profile, and oxidative stress biomarkers. Donkeys were divided randomly and equally into two groups; the chemical (Ch) and surgical (S) groups (n = 6). The Ch group was chemically castrated by intratesticular injection of 20% CaCl2 dissolved in absolute ethanol. Blood samples were collected prior to castration and at 15, 30, 45, and 60 days after the beginning of experiment. Results Surprisingly, the Ch group at the end of the experiment was characterized by significantly higher cortisol level compared to the S group. TC and LDL-C levels in the S group significantly decreased at day 45, while TG levels significantly increased at days 45 and 60 in comparison with day 0. HDL-C levels at days 30 and 60 in the Ch group significantly increased in comparison with day 0. At day 30 post-castration, HDL-C was significantly higher and LDL-C was significantly lower in the Ch group than the S group. A significant elevation in TC and LDL-C was observed at day 45 and in HDL-C at the end of experimental duration in the Ch group when compared with the S group. TPX level was significantly lower and TAC was significantly higher in the Ch group at day 45 than the S group. Conclusion Surgical castration evoked less stress and minor changes in lipid profile and oxidant/antioxidant balance relative to chemical castration by intratesticular 20% CaCl2 dissolved in absolute ethanol.

Background: Surgical castration is one of the most frequently performed surgeries in horses and is associated with various post‐surgical complications ranging in severity. Objectives: We investigated the effects of administering an anti‐GnRH vaccine to colts for reduction of testis size on the incidence of complications associated with their subsequent surgical castration. Study design: Randomised open controlled trial. Methods: Nineteen colts were randomly assigned to one of the three groups. Treatment Groups V1‐57 and V2‐100 received two treatments of Improvac® (Zoetis, South Africa) before their castration in the field on either Day 57 or Day 100 following first treatment, respectively. Controls similarly received placebo treatments followed by castration on Day 57. Serial measurements of testicular dimensions and serum samples for measuring serum testosterone concentrations (STCs) and anti‐GnRH antibody titres (ABTs) were obtained pre‐ and post‐vaccination and on the day of castration. Clinical data recorded for 10 days determined post‐surgical complications. Results: All vaccinated colts showed a baseline STC concentration at castration with a strong ABT response. Mean testicular volume of Groups V1‐57 and V2‐100 reduced by 49.7% and 30.8%, respectively, on Day 57 and this further reduced in Group V2‐100 by 63.9% at castration on Day 100. Testis size was significantly correlated with post‐surgical preputial (P = 0.001) and scrotal (P = 0.025) swelling. Main limitations: A study population of young light horse breed colts and a relatively small sample size limited this pilot study. Conclusions: Administering two doses of Improvac® in colts 28 days apart prior to castration effectively reduced their testicular volumes with associated significant post‐surgical improvement in both the incidence and degree of local swelling. Abstract Background To explore whether a polypropylene mesh is suitable for application as a new material for testicular prostheses. Methods The data of 65 patients with advanced prostate cancer who underwent surgical castration in hospital were collected and analyzed. Patients who preferred to undergo traditional orchidectomy (n = 16) were assigned to the control group, and patients who underwent subcapsular orchiectomy plus implantation of a polypropylene mesh testicular prosthesis (n = 49) were assigned to the experimental group. The presence of hematoma, infection, and other complications in patients in these two groups were investigated at 3 and 12 months following the surgery. The patients were also followed up using a self-designed testicular castration satisfaction questionnaire. Results A higher score indicated greater satisfaction. The mean score was 15.33 ± 2.85 in the experimental group and 4.63 ± 1.45 in the control group at 3 months after the surgery. Metastatic choroidal tumors derived from prostate cancer are rare. In this study, we report a patient who manifested a choroidal tumor as the initial presenting sign of prostate cancer and review 23 patients with choroidal metastasis of prostate cancer in the literature to answer a clinical question how the choroidal metastases would respond to hormonal therapy. A 73-year-old man presented with a choroidal tumor in the right eye. He was in good health and had no previous history except for current hemodialysis in 3 years due to chronic renal failure as a sequel to glomerulonephritis. With the diagnosis of a probable metastatic tumor, positron emission tomography was performed to disclose high-uptake sites in multiple bones, lymph nodes, and the prostate, together with multiple nodular lesions in bilateral lungs on computed tomography (CT) scan. Serum prostate-specific antigen (PSA) was elevated to 541 ng/mL, which supported prostate cancer as the primary site. He had degarelix injection, and the choroidal tumor resolved rapidly and became flat degeneration in a month. Prostate biopsy showed poorly differentiated adenocarcinoma, and he underwent surgical castration. He had no medication until 3 years later when he showed gradual increase of serum PSA up to 6.05 ng/mL and multiple bony metastases on CT scan. Bicalutamide, switched to enzalutamide and then to abiraterone, led to the undetectable level of serum PSA until the last visit with no relapse of the choroidal metastasis, 6.8 years after the initial visit. In the literature review of 24 patients with choroidal metastasis of prostate cancer, including this patient, 8 patients presented a choroidal tumor as the initial sign and the choroidal lesions mostly showed complete response to hormonal therapy. Among 13 patients who were frequently in the course of hormonal therapy, choroidal metastases showed complete or partial response to external beam radiation to the eye in 11 patients and episcleral plaque radiotherapy in 2 patients. In conclusion, metastatic choroidal tumors of prostate cancer would show good response to hormonal therapy when the therapy has not been initiated. Neutering male piglets by surgical procedures without anaesthesia, with analgesia and/or anaesthesia and, recently, immunological-chemical castration are practices to avoid unwanted or aggressive sexual behaviour, and to prevent the development of meat boar taint. This exploratory study aims to investigate Portuguese consumer's awareness, beliefs and attitudes in issues like boar taint, piglet's castration and pork meat quality, observing possible demographic trends. It is also intended to identify clusters of consumers with similar attitudes, crossing them with demographic data to verify the existence of patterns in Portugal related to these issues. To attain this objective, a consumer's survey was performed through an online questionnaire open for 30 days. A total of 158 respondents completed the survey. Almost a half (46%) of respondents stated their unknowledge about boar taint. Surgical castration and its effects are topics with which older consumers with a rural background are more familiar with, while immunological-chemical castration is still unknown to most consumers: 65% of consumers said they were not aware of this method, and 75% did not know whether it is an effective method for eliminating boar taint. Hierarchical clustering followed by K-means analysis segmented consumers into three clusters characterized according to their opinions, mainly divided by ethical and chemical-free orientations and by a more conservative meat quality and flavour-oriented attitudes, generally independent of prevailing demographics. In general, there were no defined opinions about the subjects under study, due mainly to the lack of information or knowledge. Nevertheless, cluster classification revealed differences in consumer's opinions, especially regarding the reasons for castration and the pain inflicted, about meat quality and the willingness to buy pork from entire males or to pay more for this type of product. Castration prevents the boar taint and minimises the aggressive behaviour of the piglets. Castration under anaesthesia maintains the welfare of animals. The aim of this study was to see the influence of castration, under local anaesthesia, on the growth rate of piglets. Total 32 piglets of similar age and body weight were selected and were randomly divided in to two groups: A and B. The piglets of Group B were castrated after applying local intratesticular anaesthesia and group A act as control. Their weights were measured after 7, 14, 21 and 28 days of time intervals. The mean average weight gain by group B piglets during the time period was statistically significantly higher than the group A piglets (P<0.5). The average body weight of Group A at the age of 1, 2, 3 and 4 week were 2.51±0.18 kg, 3.77±0.13 kg, 5.07±0.13 kg and 6.72±0.17 kg, while that of Group B were 2.50±0.13 kg, 4.82±0.11 kg, 7.15±0.16 kg and 9.13±0.17 kg, respectively. The weekly gain in body weight at 2nd, 3rd and 4th week were 1.26, 1.30 and 1.65 Kg, respectively in the Group A piglets, while the same in the Group B piglets was found to be 2.31, 2.33 and 1.98 kg, respectively. There was positive weight gain in case of castrated piglets in comparison to the uncastrated piglets and it was economical to adopt this practice by the pig farmers. As growing criminal offences, sexual offences defined by any sexual behavior lacking clear consent, creates significant morbidity, psychiatric comorbidities, adjustment disorders and mortality worldwide. Criminal laws vary between countries while use of chemical castration methods such as hormonal therapies or surgical castration techniques has been implemented in multiple states and countries while number of countries implemented castration methods have increased rapidly in recent years. Despite such trend no comprehensive randomized controlled trial or meta-analysis study has been performed yet. Additionally, multiple ethical questions arise regarding such castration techniques in sexual offenders. In this study our aim is to demonstrate efficiency and possible adverse effects of hormonal and surgical castration techniques, variations among countries and states, and ethical issues. Objective: To determine the effect of surgical castration and immunocastration on productive parameters, carcass quality, as well as the physicochemical characteristics of the meat of fattening pigs. Design/Metodology/Aproach: Ten male pigs of the York/Pietrain/Landrace breed with a body weight of 25±5 kg were used per treatment. They were housed in individual pens and fed diets according to their physiological state. The variables evaluated were analyzed with a student's t-test for independent samples. Results: Results do not show differences (p>0.05) in the productive variables, quality of the carcass or physicochemical characteristics. Study Limitations/Implications: The study did not consider qualitative variables such as flavor and smell of the treatments. Findings/Conclusions: Immunocastration is an alternative to surgical castration since the quality of the carcass, the productive variables and the physicochemical characteristics of the meat are not affected, and it favors animal welfare. This study was conducted aiming at measuring the performance and behavior of beef cattle surgically castrated, immunocastrated or non-castrated, finished at 18 months old in Aruana pasture and with energy supplementation. Thirty-nine male bovines with an initial average body weight and age of 284.1 ± 31.4 kg and 14 months old, respectively, were used. The bromatological composition analysis and productive parameters of the pasture did not differ between treatments. The performance of young cattle was not changed by the sexual condition. When evaluating the agonistic behavior, the non-castrated bovine presented a higher number of activities, such as threats and fights, in relation to those immunocastrated. The time spent on the feeder by non-castrated young cattle (56.20 minutes) was higher than that observed for surgically castrated or immunocastrated (41.43 and 32.38 minutes, respectively). The combination between the correct management of Aruana pasture and use of supplementation showed to be promising for finishing bovines. Both surgically castrated and immunocastrated steers demonstrated to be equivalents in the performed evaluations. Therefore, if the farmer chooses castration, the use of immunocastration is suggested. This practice is less invasive, preserves animal welfare more than surgical castration, and has a lower cost.

Follicle-stimulating hormone (FSH) was recently implicated as a novel regulator of fat accumulation. Surgical castration causes high FSH concentrations and increases fat accumulation, whereas immunocastration results in low FSH concentrations and less fat in immunocastrated boars versus barrows. However, detailed information regarding the role of FSH in regulation of fat accumulation in male pigs is unclear. First, expression of FSH receptor was confirmed (real-time quantitative PCR) in subcutaneous and visceral adipose tissues (SAT and VAT, respectively) of boars. Then, surgical castration (high FSH model) was compared to immunocastration (low FSH model) to investigate potential roles of FSH in adipogenesis and fat accumulation. High FSH concentrations after surgical castration activated PPARγ signaling by upregulating expression of CREB (P < 0.05), and then recruited an array of PPARγ target adipogenic genes, including transcription factor (C/EBPα), long-chain fatty acid uptake (LPL), fatty acid de novo synthesis (FASN , ACACA) and lipid droplet formation (PLIN1) in both SAT and VAT, promoting fat accumulation in barrows. In contrast, much lower serum FSH concentrations in immunocastrates attenuated (P < 0.05) expressions of PPARγ and PPARγ target genes in both SAT and VAT, resulting in less fat accumulation in immunocastrated boars versus barrows. We concluded that the substantially elevated FSH concentrations in barrows promoted fat accumulation by activating the PPARγ signaling pathway in adipose tissues, whereas immunocastrates accumulated less fat due to low FSH. • The sharply elevated FSH in boars after surgical castration plays a great role in promoting fat accumulation. • Immunocastrated boars are with low FSH levels thus accumulate less carcass fat than barrows. • FSH promotes fat accumulation in boars through activating PPARγ signaling in adipose tissues. The objective was to estimate the metabolizable energy requirement (ME) for male pigs immunocastrated in the finishing phase. The experiment was conducted at the Universidade Estadual Norte Fluminense Darcy Ribeiro. 60 male pigs were used in a randomized block design, with 50 immunocastrated pigs - IM (treatment 1 to 5) and 10 surgically castrated - CC (treatment 3b). The treatments differed according to the level of MS used, being T1 - 3030, T2 - 3130, T3a - 3230, T3b - 3230, T4 - 3330 and T5 - 3430 Kcal of EM /Kg. Performance characteristics were evaluated in different periods (A - interval between the first and second immunizations, B - interval between the second immunization and slaughter and T - total period, from the first immunization to slaughter) and carcass, in addition to sensory analysis pig meat. The average data obtained at the different energy levels were subjected to analysis of variance and linear regression analysis at a level of significance of 5%. The analysis was performed with the aid of the SAEG statistical analysis program. Increasing linear effect (P <0.05) was observed for daily feed intake, daily weight gain, daily energy consumption and daily lysine consumption in different periods. There were significant results (P <0.01) for daily energy consumption and lysine consumption in different periods. There was no difference (P> 0.05) between the carcass characteristics evaluated. In the sensory analysis, although the evaluators detected differences in the odor of pork from CC and IM pigs, such differences were not due to sexual odor. In spite of a long-lasting criticism by several international human rights bodies, the Czech Republic is the last EU member state which still perform therapeutic surgical castration of sex offenders and men suffering from certain paraphilias. While critics consider the practice medically unnecessary and inhuman, proponents claim that therapeutic surgical castration offers the last hope to patients in whom other treatment options failed. This paper analyses the question of whether and under what circumstances can surgical castration be considered permissible. After that, the legal regulation in the Czech Republic and the relevant international criticism are dealt with. Background: Castration is a husbandry practice raising important questions on the welfare and physiological status of farm animals. Searching for effective castration methods that minimally compromise the body physiology is worthy of attention. Therefore, this study aimed to evaluate the differential response of biological systems in donkeys to surgical castration versus the chemical one by CaCl2 with special emphasis on stress, lipid profile, and oxidative stress biomarkers. Donkeys were divided randomly and equally into two groups; the chemical (Ch) and surgical (S) groups (n = 6). The Ch group was chemically castrated by intratesticular injection of 20% CaCl2 dissolved in absolute ethanol. Blood samples were collected prior to castration and at 15, 30, 45, and 60 days after the beginning of experiment. Results: Surprisingly, the Ch group at the end of the experiment was characterized by significantly higher cortisol level compared to the S group. TC and LDL-C levels in the S group significantly decreased at day 45, while TG levels significantly increased at days 45 and 60 in comparison with day 0. HDL-C levels at days 30 and 60 in the Ch group significantly increased in comparison with day 0. At day 30 post-castration, HDL-C was significantly higher and LDL-C was significantly lower in the Ch group than the S group. A significant elevation in TC and LDL-C was observed at day 45 and in HDL-C at the end of experimental duration in the Ch group when compared with the S group. TPX level was significantly lower and TAC was significantly higher in the Ch group at day 45 than the S group. Conclusion: Surgical castration evoked less stress and minor changes in lipid profile and oxidant/antioxidant balance relative to chemical castration by intratesticular 20% CaCl2 dissolved in absolute ethanol. Brain metastases from prostate cancer are uncommon, occurring in fewer than 1 % of cases of metastatic prostate cancer. Brain metastasis can cause cerebral edema, neurologic symptoms, and may be misdiagnosed as primary brain tumors on imaging if thorough investigations are not done. It is difficult to identify and diagnose brain metastasis from prostate cancer since the intracranial metastatic process and presentation are poorly understood and limited to case studies. Most patients with brain metastases from prostate cancer exhibit a variety of metastatic symptoms; however, this patient's presentation was defined by only isolated intense headache. Our goal is to draw attention to the uncommon instance of brain metastases from prostate cancer in addition to reviewing the literature on the advances in treatment for prostatic cancer with metastasis to the brain. We report the case of a 67-year-old male with metastatic prostate adenocarcinoma into the brain, presenting with isolated severe headache with no prostate cancer symptoms. Following extensive radiologic examination, metastatic deposits were detected in the left side of the brain with multiple intracerebral and cerebellar vermis lesions. Multiplanar T2 weighted abdominal pelvic MRI visualized the primary lesion in the prostate which was confirmed by histology. After a month following surgical castration, the patient reported resolved headache and resumed his daily activities. The patient's serum PSA decreased from 7.8 ng/ml to 0.3 ng/ml during a 12-months follow-up with no neurological symptoms. Prostate cancer rarely causes brain metastases, and the percentage of all brain metastases that originate from prostate cancer is seldom updated. It can be difficult to distinguish between primary brain lesions and metastatic brain prostate cancer, particularly when there is just one lesion present. Despite the recently developed diagnostic approaches, symptomatic patients exhibit a variety of clinical manifestations that vary depending on the location of the metastatic focus. These manifestations include headache, seizures, and focal neurological deficits, in addition to some common non-focal manifestations like confusion and memory deficits. Our patient had a PSA of 7.8 ng/ml at the beginning and the DRE results were normal, clinically prostate cancer was not thought to be the main cause of brain metastasis. Abdominal pelvic MRI was performed to investigate the primary lesion and confirmed the presence of prostate cancer with extra prostatic extensions. Adenocarcinoma prostate cancer was found to be the main cause when histopathology was done. This report reviews the literature on brain metastases from prostate cancer and points out that while very rare, brain metastases from prostatic cancer do occur and should not be overlooked, particularly in light of the recent advancements in prostatic cancer therapies that may extend the patient's survival. Abstract Background Surgical castration is still practiced in many EU countries to avoid undesirable aggressive behavior and boar taint in male pigs. However, evidence shows that castration is painful and has a detrimental influence on pig health. This study investigated the clinical and productive effects of surgical castration in the suckling period. A total of 3696 male pigs, 3 to 6 days old, comprising of 721 litters from two different farms were included in the study. Within each litter, half of the males were kept as intact males (IM) and half were surgically castrated (CM). Surgical castration was conducted by a trained farmer. Average daily gain (ADG), body weight at weaning (BWW), percentage of pre-weaning mortality (PWM) and antibiotic usage were measured. Pig major acute phase protein (PigMAP) serum concentrations were analyzed prior to castration, and on days 1 and 10 after castration. Productive performance data were analyzed using a linear mixed model. Mortality and percentage of pigs treated with antibiotics were analyzed using the Fisher’s exact test. Results No overall differences in BWW and ADG were observed between the two groups. However, differences were observed when the same effects were analyzed in the 25% lightest, 50% medium and 25% heaviest pigs at birth. PWM was higher in CM than in IM groups (6.3% vs 3.6%; p

Castration is a common management procedure employed in North American cattle production and is known to cause a pain response. The present study was designed to investigate the effect of unmitigated surgical castration on the electroencephalography (EEG) responses and plasma substance P (SP) concentrations in calves of different ages under the same experimental conditions. Thirty male Holstein calves in three age categories [p = 0.004, p = 0.04, p = 0.04, p = 0.03, and p = 0.008, respectively). Following CAST, EEG total power decreased, and median frequency increased relative to SHAM in 6W and 3M calves only following treatment. For 6W and 3M calves, delta and beta absolute power increased at CAST and at later time points relative to SHAM. Marginal evidence for two-way interactions was noted between time and treatment and between age and treatment on the concentration of SP (p = 0.068 and p = 0.066, respectively). Substance P concentrations decreased in CAST treatment compared to SHAM at the later times (8 h: p = 0.007; 12 h: p = 0.048); 6W calves showed lower SP concentration at CAST relative to SHAM (p = 0.017). These findings indicate variation in EEG responses and in SP concentrations following unmitigated surgical castration in calves and that these responses may be age specific. These EEG findings have implications for supporting the perception of the pain associated with surgical castration in young calves and emphasize the urgency of pain mitigation strategies during routine husbandry practices such as castration, as typically implemented in North American cattle management.

The Czech Republic holds one of the highest numbers of men labelled as sexual delinquents worldwide who have undergone the irreversible process of surgical castration – a policy that has elicited strong international criticism. Nevertheless, Czech sexology has not changed its attitude towards 'therapeutic castration', which remains widely accepted and practised. In this paper, we analyse the negotiation of expertise supporting castration and demonstrate how the changes in institutional matrices and networks of experts (Eyal 2013) have impacted the categorisation of patients and the methods of treatment. Our research shows the great importance of historical development that tied Czech sexology with the state. Indeed, Czech sexology has been profoundly institutionalised since the early 1970s. In accordance with the state politics of that era, officially named Normalisation, sexology focused on sexual deviants and began creating a treatment programme that included therapeutic castration. This practice, the aim of which is to protect society from sex offenders, has changed little since. We argue that it is the expert–state alliance that enables Czech sexologists to preserve the status quo in the treatment of sexual delinquents despite international pressure. Our research underscores the continuity in medical practice despite the regime change in 1989. With regard to previous scholarship on state-socialist Czechoslovakia, we argue that it was the medical mainstream that developed and sustained disciplining and punitive features. Background: The aim of this study was to investigate the effects of surgical castration, particularly delaying the time to entrance of systemic chemotherapy, in castration-resistant prostate cancer (CRPC) patients who were refractory to initial combination androgen deprivation therapy. Materials and methods: We analyzed the clinical data of 14 CRPC patients diagnosed at Seoul National University Bundang Hospital (SNUBH) from November 2008 through May 2015. After exclusion of three patients, we finally analyzed the baseline characteristics of 11 CRPC patients. We also assessed the delaying time of docetaxel administration, which was defined as response duration, after surgical castration. Results: After bilateral orchiectomy, the treatment response rate was 45.4% and the median duration of response was 9 months (range 4–48 mo). Responders had less aggressive biopsy Gleason scores compared to nonresponders. Notably, responders showed the reducing pattern of serum prostate specific antigen levels, while nonresponders demonstrated increasing tendency after surgical castration. Moreover, responders also presented with a reduction pattern of serum testosterone levels, whereas nonresponders showed an increasing pattern of testosterone levels after bilateral orchiectomy. Conclusions: In summary, despite the limited number of cases for convincing evidence, our results shed light again on the clinical benefits of surgical castration prior to the systemic chemotherapy in some CRPC patients after initial hormone therapy.

At slaughter, live BW, liver weight as a percent of live BW, dissectible bone as a percent of cold carcass side, body protein and water contents and whole-body protein deposition decreased with time after surgical castration (linear;P <0.05), whereas dressing percentage, dissectible fat, probe fat depth and body fat content increased with time after surgical castration (linear;P <0.05). The IM had intermediate dressing percentage and dissected fat to EM and EC, whereas liver weight as a percent of live BW and body protein and lipid contents were not different from EM. Whole-body lipid deposition tended to be greater in IM than in EM between 14 and 20 weeks of age (373v . 286g/d;P = 0.051). In conclusion, castration of male pigs after 6 weeks of age has a lasting effect on physical and chemical body composition. The relationship between time after castration and body composition may be developed to predict carcass composition and can be used to determine the ideal immunization schedule aimed at specific markets in the future.
