Dermal adipocyte lineage cells are highly plastic and can undergo reversible differentiation and dedifferentiation in response to various stimuli. Using single-cell RNA sequencing of developing or wounded mouse skin, we classify dermal fibroblasts (dFBs) into distinct non-adipogenic and adipogenic cell states. Cell differentiation trajectory analyses identify IL-1-NF-κB and WNT-β-catenin as top signaling pathways that positively and negatively associate with adipogenesis, respectively. Upon wounding, activation of adipocyte progenitors and wound-induced adipogenesis are mediated in part by neutrophils through the IL-1R-NF-κB-CREB signaling axis. In contrast, WNT activation, by WNT ligand and/or ablation of Gsk3, inhibits the adipogenic potential of dFBs but promotes lipolysis and dedifferentiation of mature adipocytes, contributing to myofibroblast formation. Finally, sustained WNT activation and inhibition of adipogenesis is seen in human keloids. These data reveal molecular mechanisms underlying the plasticity of dermal adipocyte lineage cells, defining potential therapeutic targets for defective wound healing and scar formation.

Marine fungi represent a treasure trove of bioactive secondary metabolites, with benzopyran compounds emerging as a significant class of these natural products. This review delves into the structural diversity, biological activities, and sources of benzopyran compounds, highlighting their isolation from marine fungi inhabiting diverse environments such as sponges, marine sediments, algae, mangroves, and corals. Our literature search, conducted from 2000 to 2023, has identified a wealth of benzopyran compounds, showcasing their potential as lead compounds in drug development. The characteristics of benzopyran from marine fungi are explored, encompassing various subclasses such as chromones, isocoumarins, citrinins, and other related compounds. These compounds exhibit a remarkable chemical diversity, which is crucial for their diverse biological activities. The potential of benzopyran compounds in drug development is also discussed, emphasizing their roles in anti-tumor, antibacterial, anti-inflammatory, and enzyme inhibitory activities. In recent years, a remarkable 210 bioactive benzopyran compounds have been isolated from the secondary metabolites of marine fungi. These findings underscore the importance of marine fungi as a source of novel bioactive compounds, offering a plethora of potential lead compounds for the development of marine-derived drugs. This article explores the role of a circular RNA called circLIPH in the progression of pancreatic cancer. The study reveals that circLIPH is increased in pancreatic tumor tissue and is associated with tumor size and stage. It also demonstrates that circLIPH promotes the growth, migration, and invasion of pancreatic cancer cells. The researchers identify miR-769-3p as a target of circLIPH and show that circLIPH increases the expression of GOLM1, a gene involved in cancer progression. These findings suggest that circLIPH could potentially be used as a biomarker and therapeutic target for pancreatic cancer. The study further investigates the role of circLIPH in pancreatic cancer progression and finds that overexpression of miR-769-3p inhibits the growth, proliferation, migration, and invasion of pancreatic cancer cells. Conversely, treatment with a miR-769-3p inhibitor has the opposite effects. The target genes of miR-769-3p are enriched in the mTOR signaling pathway. Aberrant activation of dermal fibroblasts during wound healing often leads to debilitating fibrotic changes in the skin, such as scleroderma and keloids. However, the underlying cellular and molecular mechanisms remain elusive. Here, we established a wound‐induced skin fibrosis (WISF) mouse model in mature adult mice, characterised by excessive deposition of collagen bundles, loss of dermal adipocytes, and enrichment of DPP4+Ly6A+THY1+ hypodermal interstitial adipocyte progenitors (HI‐APs) and pericytes, resembling human fibrotic skin diseases. This WISF model exhibited an age‐dependent gain of fibrotic characteristics, contrasting with the wound‐induced hair neogenesis observed in younger mice. Through comprehensive analyses of the WISF, we delineated a trajectory of fibroblast differentiation that originates from HI‐APs. These progenitors highly expressed several extracellular matrix (ECM) genes and exhibited a TGFβ pathway signature. TGFβ was identified as the key signal to inhibit the adipogenic potential and maintain the fibrogenic potential of dermal APs. Additionally, administering a TGFβ receptor inhibitor to wound scar reduced the abundance of ECM‐producing APs. Finally, analysis of human scleroderma skin tissues revealed a negative correlation between the expression of AP‐, ECM‐, and TGFβ pathway‐related genes and PPARG. Overall, this study establishes a wound‐induced skin fibrosis mouse model and demonstrates that TGFβ‐mediated blockage of HI‐AP differentiation is crucial for driving fibrotic pathology. The segmentation accuracy of bridge crack images is influenced by high‐frequency light, complex scenes, and tiny cracks. Therefore, an integration–competition network (complex crack segmentation network [CCSNet]) is proposed to address these problems. First, a grayscale‐oriented adjustment algorithm is proposed to solve the high‐frequency light problem. Second, an integration–competition mechanism is proposed to detach complex backgrounds and grayscale features of cracks. Finally, a tiny attention mechanism is proposed to extract the shallow features of tiny cracks. CCSNet outperforms seven state‐of‐the‐art crack segmentation methods in both generalization and comparison experiments on self‐built dataset and four public datasets. It also achieved excellent performance in practical bridge crack tests. Therefore, CCSNet is an effective auxiliary method for lowering the cost of bridge safety detection. [ABSTRACT FROM AUTHOR]

Green remediation of severely contaminated soils around mining sites can be achieved using suitable woody plants such as Quercus species, but their phytoremediation potential has not been well evaluated yet. Six Quercus species, which were popular in ecological restoration and landscape application in east China, were selected and evaluated for their phytoremediation potential of metal polluted soil using a pot experiment that lasted for 150 d. The results suggested that Quercus species exhibited high tolerance to multi-metal contamination of Cu (9839 mg · kg−1), Cd (8.5 mg · kg−1), and Zn (562 mg · kg−1) with a tolerance index (TI) ranging from 0.52 to 1.21. Three Quercus (Q. pagoda, Q. acutissima, and Q. nuttallii) showed relatively higher tolerance with TIs of 1.08, 1.09, and 1.21, respectively. Above-ground tissues accounted for most of the total biomass in T1 (mixture of clean and polluted soil, 50%) and T2 (100% polluted soil) treatments for most species. The Cu contents in plant tissues were in the order of root > leaf > stem, whereas Zn exhibited the order of leaf > stem > root, and Cd showed divergent mobility within the Quercus species. All the Quercus species exhibited higher capacity for Zn phytoextraction with translocation factor (TF) over 1 and Cu/Cd phytostabilization with TFs lower than 1. The analytic hierarchy process-entropy weight model indicated that Q. virginiana and Q. acutissima were two excellent species with evident phytoremediation capacity of Cu, Cd, and Zn co-contaminated soil. Taken together, Quercus species showed great potential for phytoremediation of soils severely polluted by Cu, Cd, and Zn around historic mining sites. Application of Quercus species is a green remediation option with low-maintenance cost and prospective economic benefit for phytomanagement of historic mining sites. [ABSTRACT FROM AUTHOR]
