Background and study aims Infected necrotic collections extending deep into the retroperitoneum may not be amenable solely to endoscopic necrosectomy. Rendezvous transgastric and percutaneous sinus tract endoscopic necrosectomy was conceived to maximize debridement and obviate the need for open necrosectomy or video-assisted retroperitoneal debridement. Patients and methods Patients who underwent simultaneous rendezvous transgastric and sinus tract endoscopic transluminal necrosectomy at the same session were identified from a prospectively maintained database. Demographic data, clinical outcomes, immediate and late complications were evaluated. Results Among 415 patients with necrotizing pancreatitis, four patients (three males, median age 47 years) underwent this intervention after a median 29.5 days following placement of percutaneous drain. Intra-procedural bleeding following dilation of percutaneous tract required placement of esophageal stent for tamponade. No patients required rescue open necrosectomy or video assisted retroperitoneal debridement. Complete removal of percutaneous drains was accomplished in all patients after a median of 78.5 days. Conclusions Our novel approach is safe and effective and can expand the available armamentarium for management of large necrotic collection with deep retroperitoneal extension.

Background and aims: Endoscopic ultrasound with fine needle aspiration (EUS-FNA) has become the standard of care in the evaluation of solid pancreatic lesions. Limited data exist on interobserver agreement (IOA) among cytopathologists in assessing solid pancreatic EUS-FNA specimens. This study aimed to evaluate IOA among cytopathologists in assessing EUS-FNA cytology specimens of solid pancreatic lesions using a novel standardized scoring system and to assess individual clinical and cytologic predictors of IOA. Methods: Consecutive patients who underwent EUS-FNA of solid pancreatic lesions at a tertiary care referral center were included. EUS-FNA slides were evaluated by four blinded cytopathologists using a standardized scoring system that assessed final cytologic diagnosis and quantitative (number of nucleated/diagnostic cells) and qualitative (bloodiness, inflammation/necrosis, contamination, artifact) cytologic parameters. Final clinical diagnosis was based on final cytology, surgical pathology, or 1-year clinical follow-up. IOA was calculated using multi-rater kappa (κ) statistics. Bivariate analyses were performed comparing cases with and without uniform agreement among the cytopathologists followed by logistic regression with backward elimination to model likelihood of uniform agreement. Results: Ninety-nine patients were included (49 % males, mean age 64 years, mean lesion size 26 mm). IOA for final diagnosis was moderate (κ = 0.45, 95 % confidence interval (CI) 0.4 – 0.49) with minimal improvement when combining suspicious and malignant diagnoses (κ = 0.54, 95 %CI 0.49 – 0.6). The weighted kappa value for overall diagnosis was 0.65 (95 %CI 0.54 – 0.76). IOA was slight to fair (κ = 0.04 – 0.32) for individual cytologic parameters. A final clinical diagnosis of malignancy was the most significant predictor of agreement [OR 3.99 (CI 1.52 – 10.49)]. Conclusions: Interobserver agreement among cytopathologists for pancreatic EUS-FNA specimens is moderate-substantial for the final cytologic diagnosis. The final clinical diagnosis of malignancy was the strongest predictor of agreement. These results have significant implications for patient management and need to be validated in future trials.

Abstract Background Randomised, placebo-controlled trials are needed to provide evidence demonstrating safe, effective interventions that reduce falls and fractures in the elderly. The quality of a clinical trial is dependent on successful recruitment of the target participant group. This paper documents the successes and failures of recruiting over 2,000 women aged at least 70 years and at higher risk of falls or fractures onto a placebo-controlled trial of six years duration. The characteristics of study participants at baseline are also described for this study. Methods The Vital D Study recruited older women identified at high risk of fracture through the use of an eligibility algorithm, adapted from identified risk factors for hip fracture. Participants were randomised to orally receive either 500,000 IU vitamin D3 (cholecalciferol) or placebo every autumn for five consecutive years. A variety of recruitment strategies were employed to attract potential participants. Results Of the 2,317 participants randomised onto the study, 74% (n = 1716/2317) were consented onto the study in the last five months of recruiting. This was largely due to the success of a targeted mail-out. Prior to this only 541 women were consented in the 18 months of recruiting. A total of 70% of all participants were recruited as a result of targeted mail-out. The response rate from the letters increased from 2 to 7% following revision of the material by a public relations company. Participant demographic or risk factor profile did not differ between those recruited by targeted mail-outs compared with other methods. Conclusion The most successful recruitment strategy was the targeted mail-out and the response rate was no higher in the local region where the study had extensive exposure through other recruiting strategies. The strategies that were labour-intensive and did not result in successful recruitment include the activities directed towards the GP medical centres. Comprehensive recruitment programs employ overlapping strategies simultaneously with ongoing assessment of recruitment rates. The ability to investigate hypervalent iodine(V) fluorides has been limited primarily by their difficult preparation traditionally using harsh fluorinating reagents such as trifluoromethyl hypofluorite and bromine trifluoride. Here, we report a mild and efficient route using Selectfluor to deliver hypervalent iodine(V) fluorides in good isolated yields (72-90%). Stability studies revealed that bicyclic difluoro(aryl)-λ 5 -iodane 6 was much more stable in acetonitrile- d 3 than in chloroform- d 1 , presumably due to acetonitrile coordinating to the iodine(V) centre and stabilising it via halogen bonding., (Copyright © 2024, Dearman et al.)

Forecasting blood glucose (BG) levels with routinely collected data is useful for glycemic management. BG dynamics are nonlinear, complex, and nonstationary, which can be represented by nonlinear models. However, the sparsity of routinely collected data creates parameter identifiability issues when high-fidelity complex models are used, thereby resulting in inaccurate forecasts. One can use models with reduced physiological fidelity for robust and accurate parameter estimation and forecasting with sparse data. For this purpose, we approximate the nonlinear dynamics of BG regulation by a linear stochastic differential equation: we develop a linear stochastic model, which can be specialized to different settings: type 2 diabetes mellitus (T2DM) and intensive care unit (ICU), with different choices of appropriate model functions. The model includes deterministic terms quantifying glucose removal from the bloodstream through the glycemic regulation system and representing the effect of nutrition and externally delivered insulin. The stochastic term encapsulates the BG oscillations. The model output is in the form of an expected value accompanied by a band around this value. The model parameters are estimated patient-specifically, leading to personalized models. The forecasts consist of values for BG mean and variation, quantifying possible high and low BG levels. Such predictions have potential use for glycemic management as part of control systems. We present experimental results on parameter estimation and forecasting in T2DM and ICU settings. A small proportion of the thousands of pesticides on the market today are associated with a disproportionately high incidence of severe acute pesticide poisoning and suicide. Paraquat stands out as one of the most lethal pesticides in common use, frequently involved in fatal incidents due to suicides or accidental exposure. Even though paraquat has been banned in over 67 countries, it is still widely used in many others, particularly in Asia and Latin America. Based on a literature review and consultations, this paper identifies options for replacing paraquat and distils practical lessons from numerous successes around the world. Our aim is to support regulators, policymakers, agronomists and the supply chain sector with practical information related to phasing out paraquat. Production data consistently failed to show any negative effects of banning paraquat on agricultural productivity. A wide range of alternative approaches to weed management and crop defoliation are available, many of which do not rely on herbicides. Over 1.25 million farmers in low- and middle-income countries (LMICs) successfully produce a range of crops for private voluntary standards (PVS) in food and fiber supply chains which prohibit paraquat use. We conclude from the findings of this study that eliminating paraquat will save lives without reducing agricultural productivity. Less hazardous and more sustainable alternatives exist. The rice field rat, Rattus argentiventer, is a significant pest of rice in Southeast Asia. Fertility control methods have the potential to provide safe and effective alternatives to control methods that often include indiscriminate use of rodenticides or electric barriers. The aim of this laboratory study was to assess uptake of bait coated with different concentrations of the contraceptive hormones, quinestrol (E) and levonorgestrel (P), delivered alone and in combination (i.e. EP-1) and determine the short-term effects on reproductive parameters of adult male and female R. argentiventer. In Experiment 1, 2 concentrations of E, P, and EP-1 (10, 20 ppm) were fed to groups of wild-caught rats for 7 days. In females, both E and EP-1 induced uterine edema. In males, EP-1 reduced epididymis and seminal vesicle weights and lowered sperm motility. However, these responses were inconsistent due to low bait acceptance, especially with increasing concentrations. In Experiment 2, EP-1 (0, 20, 50, 100 ppm) was administered by oral gavage daily for 7 days to male R. argentiventer. There were significant reductions in epididymal and seminal vesicle weights for all oral doses of EP-1, in sperm counts for the 50 ppm dose, and in sperm motility for the 20 and 50 ppm doses compared to the control group. To select the optimum dose of EP-1, we must address the poor acceptance of contraceptive-coated baits by rice field rats. Background: Trapping is a key method for monitoring small mammals and is also one of a number of methods recommended under an ecologically-based rodent management program to control rodent pest populations. Live-traps are widely used globally for studying small mammal populations. In Asia where rodents are major pests of rice, single capture traps typically provide low trap success. We compared the trap success between two types of live-traps in rice fields in Indonesia and the Philippines., Results: Multiple-capture traps (MCTs) in conjunction with a linear trap barrier were significantly more effective in catching rodent pest species than single-capture traps (SCTs) in Indonesia and the Philippines. In Indonesia, MCTs captured more individuals with a mean (±SE) percent trap success rate of (15.54 ± 4.29) compared to SCTs (3.88 ± 1.58). In the Philippines, MCTs captured more species of rodents and had a significantly higher recapture rate (1.96 ± 0.79), than SCTs (0.58 ± 0.32)., Conclusion: Multiple-capture traps with a linear trap-barrier were more effective for capturing Rattus argentiventer and Rattus tanezumi in rice field ecosystems compared to single-capture traps. MCTs captured more species of rodent pests in the Philippines and recaptured more individuals of each species. These results indicate that rodent populations can be more effectively monitored and controlled by using a multi-capture trap with barrier system than the use of single capture traps on their own. Southeast Asia is a major rice-producing region with a high level of internal consumption and accounting for 40% of global rice exports. Limited land resources, climate change and yield stagnation during recent years have once again raised concerns about the capacity of the region to remain as a large net exporter. Here we use a modelling approach to map rice yield gaps and assess production potential and net exports by 2040. We find that the average yield gap represents 48% of the yield potential estimate for the region, but there are substantial differences among countries. Exploitable yield gaps are relatively large in Cambodia, Myanmar, Philippines and Thailand but comparably smaller in Indonesia and Vietnam. Continuation of current yield trends will not allow Indonesia and Philippines to meet their domestic rice demand. In contrast, closing the exploitable yield gap by half would drastically reduce the need for rice imports with an aggregated annual rice surplus of 54 million tons available for export. Overuse of seed and chemical inputs is a major constraint for sustainable rice production in Vietnam. In this study, two seasons of field trials were conducted to compare different crop establishment practices for rice production in the Mekong River Delta using environmental and economic sustainability performance indicators. The indicators including energy efficiency, agronomic use efficiency, net income, and greenhouse gas emissions (GHGEs) were quantified based on four treatments including manual broadcast-seeding, blower seeding, drum seeding, and mechanized transplanting. Across the four treatments, yields ranged from 7.3-7.5 Mg ha -1 and 6.2-6.8 Mg ha -1 in the Winter-Spring (WS) and Summer-Autumn (SA) seasons, respectively. In comparison with direct seeding methods, mechanized transplanting decreased the seed rate by 40%. It also led to a 30-40% reduction in pesticide use during the main crop season (WS). Mechanized transplanting required higher inputs, including machine depreciation and fuel consumption, but its net energy balance, net income and GHGE were at a similar level as the other non-mechanized planting practices. Future rice systems must produce more grain while minimizing the negative environmental impacts. A key question is how to orient agricultural research & development (R&D) programs at national to global scales to maximize the return on investment. Here we assess yield gap and resource-use efficiency (including water, pesticides, nitrogen, labor, energy, and associated global warming potential) across 32 rice cropping systems covering half of global rice harvested area. We show that achieving high yields and high resource-use efficiencies are not conflicting goals. Most cropping systems have room for increasing yield, resource-use efficiency, or both. In aggregate, current total rice production could be increased by 32%, and excess nitrogen almost eliminated, by focusing on a relatively small number of cropping systems with either large yield gaps or poor resource-use efficiencies. Rodents present a major problem for food security in Asia where smallholder farming families are particularly vulnerable. We review here recent developments in the biology and management of rodent pests in cereal cropping systems in Asia. The past decade has seen a strong focus on ecologically-based rodent management (EBRM), its adoption in field studies significantly increased rice yields (6-15%) and income (>15%) in seven Asian countries. EBRM principles have also been successfully applied to maize in China. We provide case studies on EBRM in Cambodia, on interactions between rodent pests and weeds, and on the importance of modified wetlands for biodiversity and rodent pest management. Knowledge on post-harvest impacts of rodents is increasing. One research gap is the assessment of human health impacts from a reduction of rodent densities in and around houses. We identify 10 challenges for the next decade. For example, the need for population modelling, a valuable tool missing from our toolbox to manage rodent pests in cereal systems. We also need to understand better the interactive effects of cropping intensification, conservation agriculture and climate change. Finally, new management approaches such as fertility control are on the horizon and need to be considered in the context of smallholder cereal farming systems and mitigating health risks from zoonotic diseases associated with rodents. © 2021 The Authors. A new and efficient strategy for the rapid formation of novel fluorinated tetrahydropyridazines and dihydrooxazines has been developed by fluorocyclisation of β,γ-unsaturated hydrazones and oximes with the fluoroiodane reagent. Mechanochemical synthesis delivered fluorinated tetrahydropyridazines in similar excellent yields to conventional solution synthesis, whereas fluorinated dihydrooxazines were prepared in much better yields by ball-milling.

Activated aromatics were reacted with two different fluoroidoane reagents 1 and 2 in the presence of triflic acid to prepare only the para-substituted diaryliodonium salts. With fluoroiodane 1 the unsymmetrical diaryliodonium salts contained an ortho-propan-2-ol sidearm, whereas the alcohol sidearm was eliminated to form an ortho-styrene sidearm in the reaction with fluoroiodane 2. Only the diaryliodonium salts containing a styrene sidearm were fluorinated successfully to deliver para-fluorinated aromatics in good yields.

Background: Regular methamphetamine use is associated with increased rates of psychiatric symptoms. Although there has been a substantial body of research reporting on the effectiveness of psychological treatments for reducing methamphetamine use, there is a paucity of research examining the effects of these treatments on co-occurring psychiatric symptoms. We addressed this gap by undertaking a systematic review of the evidence of the effectiveness of psychological treatments for methamphetamine use on psychiatric symptom outcomes in randomized controlled trials., Methods: A narrative synthesis of studies was conducted following the Cochrane Handbook for Systematic Reviews of Interventions and the Preferred Reporting Items for Systematic Reviews and Meta-Analysis (PRISMA) statement to inform methodology. Eight electronic peer-reviewed databases were searched. Ten eligible studies were assessed., Results: Most studies found an overall reduction in levels of methamphetamine use and psychiatric symptoms among samples as a whole. Although brief interventions were effective, there is evidence that more intensive interventions have greater impact on methamphetamine use and/or psychiatric symptomatology. Intervention attendance was variable., Conclusions: The evidence suggests that a variety of psychological treatments are effective in reducing levels of methamphetamine use and improving psychiatric symptoms. Future research should consider how psychological treatments could maximize outcomes in the co-occurring domains of methamphetamine use and psychiatric symptoms, with increasing treatment attendance as a focus. Integrated biodiversity management aims to conserve the beneficial species components of production ecosystems and reduce the impacts of pests. In 2011 and 2013, experiments were conducted at Los Baños, Laguna, Philippines, to compare arthropod communities in rice plots and on levees with and without vegetation strips. Vegetation strips included spontaneous weeds, sesame and okra (2011), or mung bean (2013). The plots were treated with one of three nitrogen levels and in one experiment were planted with planthopper-resistant (IR62) and susceptible (IR64) rice varieties. Parasitoids and predators of lepidopteran pests and of the ricebug, Leptocorisa oratorius, were more abundant in high-nitrogen rice plots where their prey/hosts also had highest densities. Planthoppers and leafhoppers were more abundant in low-nitrogen plots. Weedy and sesame/okra bunds provided habitat for a range of natural enemies including spiders, parasitoids and predatory bugs, but did not have higher pest numbers than cleared bunds. Higher abundances of the predator Cythorhinus lividipennis and higher parasitism of planthopper ( Nilaparvata lugens ) eggs by Anagrus sp. were associated with sesame/okra bunds in late season rice plots. Mung bean also provided habitat for key predators and parasitoids that spilled over to adjacent rice; however, mung bean was also associated with higher numbers of lepidopteran and grain-sucking pests in the adjacent rice, albeit without increased damage to the rice. For ricebug in particular, damage was probably reduced by higher parasitoid:pest ratios adjacent to the vegetation strips. Varietal resistance and mung bean strips had an additive effect in reducing abundance of the planthopper Sogatella furcifera and the leafhopper Nephotettix virescens. Reduced numbers of these latter pests close to vegetation strips were often compensated for by other plant-sucking bugs, thereby increasing the intensity of potentially stabilizing interspecific interactions such as competition. We highlight the benefits of diversifying rice landscapes and the need to optimize vegetation strips, e.g., by including lepidopteran trap-plants, for intensive rice production systems., Competing Interests: The authors declare no conflict of interest.

Hexafluoroisopropan-2-ol (HFIP) is an excellent solvent for promoting fluorinations with the hypervalent fluoroiodane reagent 1 and crucially, it removes the need for transition metals or TREAT-HF activators. The fluoroiodane reagent 1 was used in HFIP to monofluorinate 1,3-ketoesters and to fluorocyclise unsaturated carboxylic acids in excellent yields under mild reaction conditions.

The Metropolis-Adjusted Langevin Algorithm (MALA) is a Markov Chain Monte Carlo method which creates a Markov chain reversible with respect to a given target distribution, π N , with Lebesgue density on R N ; it can hence be used to approximately sample the target distribution. When the dimension N is large a key question is to determine the computational cost of the algorithm as a function of N . The measure of efficiency that we consider in this paper is the expected squared jumping distance (ESJD), introduced in Roberts et al. (Ann Appl Probab 7(1):110-120, 1997). To determine how the cost of the algorithm (in terms of ESJD) increases with dimension N , we adopt the widely used approach of deriving a diffusion limit for the Markov chain produced by the MALA algorithm. We study this problem for a class of target measures which is not in product form and we address the situation of practical relevance in which the algorithm is started out of stationarity. We thereby significantly extend previous works which consider either measures of product form, when the Markov chain is started out of stationarity, or non-product measures (defined via a density with respect to a Gaussian), when the Markov chain is started in stationarity. In order to work in this non-stationary and non-product setting, significant new analysis is required. In particular, our diffusion limit comprises a stochastic PDE coupled to a scalar ordinary differential equation which gives a measure of how far from stationarity the process is. The family of non-product target measures that we consider in this paper are found from discretization of a measure on an infinite dimensional Hilbert space; the discretised measure is defined by its density with respect to a Gaussian random field. The results of this paper demonstrate that, in the non-stationary regime, the cost of the algorithm is of O ( N 1 / 2 ) in contrast to the stationary regime, where it is of O ( N 1 / 3 ) .

Rodents are globally important pre-harvest pests of rice. In Southeast Asia, rodent damage to growing rice crops is commonly concentrated towards the center of rice fields, away from the field edge, resulting in a clear pattern known as the "stadium effect." To further understand this behavior of rodent pests and to develop recommendations for future research and management, we examined the relation between giving-up densities (GUDs) and damage patterns. In Tanay, Luzon, Philippines, GUD trays containing pieces of coconut in a matrix of sand were placed at 4 different distances from the field edge to quantify the perceived risk of predation in a rice field pest, Rattus tanezumi. GUDs were recorded during a dry and wet season crop at the reproductive and ripening stages of rice. In addition, assessments of active burrows, tracking tile activity and rodent damage to the rice crop, were conducted in the dry season. GUDs were significantly lower in the center of the rice fields than on the field edges, suggesting that rodent damage to rice is greater in the middle of rice fields due to a lower perceived predation risk. Furthermore, this perception of predation risk (or fear) increases towards the field edge and was greatest on the rice bund, where there was no vegetation cover. We discuss the implications for rodent management and rodent damage assessments in rice fields. Background: Little is known about native and non-native rodent species interactions in complex tropical agroecosystems. We hypothesised that the native non-pest rodent Rattus everetti may be competitively dominant over the invasive pest rodent Rattus tanezumi within agroforests. We tested this experimentally by using pulse removal for three consecutive months to reduce populations of R. everetti in agroforest habitat, and assessed over 6 months the response of R. tanezumi and other rodent species., Results: Following removal, R. everetti individuals rapidly immigrated into removal sites. At the end of the study period, R. tanezumi were larger and there was a significant shift in their microhabitat use with respect to the use of ground vegetation cover following the perturbation of R. everetti. Irrespective of treatment, R. tanezumi selected microhabitat with less tree canopy cover, indicative of severely disturbed habitat, whereas R. everetti selected microhabitat with a dense canopy., Conclusion: Our results suggest that sustained habitat disturbance in agroforests favours R. tanezumi, while the regeneration of agroforests towards a more natural state would favour native species and may reduce pest pressure in adjacent crops. A new class of fluorinated lactones was prepared by the intramolecular fluorocyclizations of unsaturated carboxylic acids by using the stable fluoroiodane reagent in combination with AgBF4. This unique reaction incorporates a cyclization, an aryl migration, and a fluorination all in one step. The fluoroiodane reagent, prepared easily from fluoride, can also be used without a metal catalyst to give moderate yields within just 1 hour, thus demonstrating that it is a suitable reagent for developing new (18)F-labelled radiotracers for PET imaging., (© 2015 WILEY-VCH Verlag GmbH & Co. Aqueous microdroplets with a volume of a few femtoliters are an ideal sample size for single-molecule fluorescence experiments. In particular, they enable prolonged measurements to be made on individual molecules that can diffuse freely in the surrounding medium. However, the rapid production of monodisperse droplets in a hydrodynamic flow, such as microfluidic flow focusing, will often involve volumes that are typically too large (>0.5 pL) for single-molecule studies. Desired volumes of a few femtoliters, or smaller, can be produced by either tip streaming or step emulsification in a flow-focusing device; however, in both of these methods, the aqueous droplets are dispersed in a large volume of the continuous phase, where individual droplets can diffuse perpendicular to the flow direction, and the monodispersity of droplet size produced by tip streaming is difficult to sustain for more than transient time scales. We show here that the optimized design and fabrication of microfluidic devices with shallow channel depths can result in the reliable production of stable droplets of a few femtoliters at a high rate in the dripping regime of flow focusing. Furthermore, the generated microdroplets are localized in a two-dimensional plane to enable immediate analysis. We have demonstrated the fluorescence monitoring of single molecules of encapsulated green fluorescent protein. The apparatus is straightfoward, inexpensive, and readily assembled within an ordinary laboratory environment.

Background: Reduction of vegetation height is recommended as a management strategy for controlling rodent pests of rice in South-east Asia, but there are limited field data to assess its effectiveness. The breeding biology of the main pest species of rodent in the Philippines, Rattus tanezumi, suggests that habitat manipulation in irrigated rice-coconut cropping systems may be an effective strategy to limit the quality and availability of their nesting habitat. The authors imposed a replicated manipulation of vegetation cover in adjacent coconut groves during a single rice-cropping season, and added artificial nest sites to facilitate capture and culling of young., Results: Three trapping sessions in four rice fields (two treatments, two controls) adjacent to coconut groves led to the capture of 176 R. tanezumi, 12 Rattus exulans and seven Chrotomys mindorensis individuals. There was no significant difference in overall abundance between crop stages or between treatments, and there was no treatment effect on damage to tillers or rice yield. Only two R. tanezumi were caught at the artificial nest sites., Conclusion: Habitat manipulation to reduce the quality of R. tanezumi nesting habitat adjacent to rice fields is not effective as a lone rodent management tool in rice-coconut cropping systems., (© 2013 Society of Chemical Industry.)

The air and moisture stable fluoroiodane 8, readily prepared on a 6 g scale by nucleophilic fluorination of the hydroxyiodane 7 with TREAT-HF, has been used as an electrophilic fluorinating reagent for the first time to monofluorinate 1,3-ketoesters and difluorinate 1,3-diketones in good isolated yields.

Two approaches have been developed for the enantioselective Reformatsky reaction of ethyl iododifluoroacetate with ketones to form a quaternary carbon centre using (1R,2S)-1-phenyl-2-(1-pyrrolidinyl)-1-propanol as the chiral ligand. Good yields and high enantioselectivities (80-91% ee) were achieved with a range of alkyl aryl ketones in a convenient one-pot protocol using ethyl iododifluoroacetate and diethylzinc to form the difluorinated Reformatsky reagent homogeneously. In the traditional two-step Reformatsky reaction using the preformed Reformatsky reagent generated from ethyl iododifluoroacetate and zinc dust, good yields and good enantioselectivities (75-84% ee) were also obtained., (This journal is © The Royal Society of Chemistry 2012)

The first enantioselective Reformatsky-type reaction of ethyl iodofluoroacetate has been accomplished with alkyl aryl ketones. High diastereoselectivities and excellent enantioselectivities for the major diastereomer (93-95% ee) were achieved with large alkyl groups. For smaller alkyl groups the diastereoselectivities were moderate, but excellent enantioselectivities were obtained for both diastereomers (79-94% ee)., (This journal is © The Royal Society of Chemistry 2012)

A new fluorous PCP pincer ligand has been coordinated to Ni(II), Pd(II) and Pt(II). The air stable palladium complex, which promotes Heck reactions between methyl acrylate and either aryl bromides or iodides, can be recovered intact by fluorous solid-phase extraction and was reused four times in the Heck reaction between methyl acrylate and 4-bromoacetophenone without loss in catalytic activity.

Purpose: Multidrug-resistant (MDR) Salmonella strains are associated with excess bloodstream infections, hospitalizations, and deaths compared with pansusceptible strains. Bovine products are sometimes a source of MDR Salmonella. To generate hypotheses for regional differences in risk factors for human infection, we analyzed distributions of the two most prevalent MDR Salmonella phenotypes in the United States, 2003-2005: (i) MDR-ACSSuT (resistant to at least ampicillin, chloramphenicol, streptomycin, sulfonamides, and tetracycline) Typhimurium; (ii) MDR-AmpC (resistant to at least ampicillin, chloramphenicol, streptomycin, sulfonamides, tetracycline, amoxicillin/clavulanic acid, and ceftiofur, and with decreased susceptibility to ceftriaxone) Newport., Materials and Methods: Participating public health laboratories in all states forwarded every 20th Salmonella isolate from humans to the National Antimicrobial Resistance Monitoring System for Enteric Bacteria for antimicrobial susceptibility testing. Among the serotypes Typhimurium and Newport isolates submitted 2003-2005, pansusceptible, MDR-ACSSuT Typhimurium, and MDR-AmpC Newport were identified. Patterns of resistance, demographic factors, and cattle density were compared across regions., Results: Of 1195 serotype Typhimurium isolates, 289 (24%) were MDR-ACSSuT. There were no significant differences in region, age, or sex distribution for pansusceptible versus MDR-ACSSuT Typhimurium. Of 612 serotype Newport isolates, 97 (16%) were MDR-AmpC, but the percentage of MDR-AmpC isolates varied significantly across regions: South 3%, Midwest 28%, West 32%, and Northeast 38% (p < 0.0001). The South had the lowest percentage of MDR-AmpC Newport isolates and also the lowest density of milk cows. More Newport isolates were MDR-AmpC in the 10 states with the highest milk cow density compared with the remaining states. Overall, 22% of pansusceptible Newport isolates but only 7% of MDR-AmpC Newport isolates were from patients <2 years of age. For both serotypes, MDR phenotypes had less seasonal variation than pansusceptible phenotypes., Conclusion: This is the first analysis of the distribution of clinically important MDR Salmonella isolates in the United States. MDR-ACSSuT Typhimurium was evenly distributed across regions. However, MDR-AmpC Newport was less common in the South and in children <2 years of age. Information on individuals' exposures is needed to fully explain the observed patterns.

A series of N,N'-dialkyl-4,13-diaza-18-crown-6 lariat ethers possessing two C8H17 (2), (CH2)3C8F17 (3), (CH2)3C10F21 (4), and (CH2)2C8F17 (5) side arms were synthesized in good yields by N-alkylation of 4,13-diaza-18-crown-6. Potassium picrate could be extracted from an aqueous solution into an organic phase by all of the perfluoroalkylated macrocycles demonstrating their potential to be used as phase-transfer catalysts, and preliminary studies on a classical nucleophilic substitution established that they each gave higher catalytic activities under solid-liquid than under liquid-liquid phase-transfer conditions. The light fluorous macrocycles gave similar, if not better, catalytic activity compared to the parent, non-fluorinated phase-transfer catalyst 2 under solid-liquid conditions in conventional organic solvents in both an aliphatic and an aromatic nucleophilic substitution. N,N'-Bis(1H,1H,2H,2H,3H,3H-perfluoroundecyl)-4,13-diaza-18-crown-6 (3) was recycled six times in the iodide displacement reaction of 1-bromooctane and four times in the fluoride displacement reaction of 2,4-dinitrochlorobenzene using fluorous solid-phase extraction without any loss in activity.

Highly fluorophilic phosphines incorporating at least one aromatic ring containing two directly attached perfluoroalkyl groups have been synthesised, their partition coefficients (organic phase : fluorous phase) measured and their electronic properties probed using (1)J(PtP) data for their trans-[PtCl(2)L(2)] complexes. These phosphines have been used as modifying ligands for the rhodium catalysed hydroformylation of 1-octene in perfluorocarbon solvents. Catalyst activity, regioselectivity and the levels of rhodium leaching to the product phase vary with the substitution patterns of the modifying ligands that do not correlate with the electronic properties or partition coefficients of these ligands, but can be interpreted in terms of differences in the resting states of the catalysts.

We consider the dynamics of systems in the presence of inertia and colored multiplicative noise. We study the limit where the particle relaxation time and the correlation time of the noise both tend to zero. We show that the limiting equation for the particle position depends on the magnitude of the particle relaxation time relative to the noise correlation time. In particular, the limiting equation should be interpreted either in the Itô or Stratonovich sense, with a crossover occurring when the two fast-time scales are of comparable magnitude. At the crossover the limiting stochastic differential equation is neither of Itô nor of Stratonovich type. This means that, after adiabatic elimination, the governing equations have different drift fields, leading to different physical behavior depending on the relative magnitude of the two fast-time scales. Our findings are supported by numerical simulations.

Highly efficient separation, recovery and recycling of the fluorous-tagged catalyst, [Ni[F13C6C(O)CHC(O)C6F13]2], can be achieved with fluorous solid-phase extraction, after the Lewis acid catalysed synthesis of enaminodiones in conventional organic solvents.

Triaryl-phosphines and -phosphites bearing fluorous ponytails give high rates, good linear selectivity and good retention of catalyst in the fluorous phase during hydroformylation of alkenes in fluorous solvents.

This study compared the efficacy and safety of the fluticasone propionate 125 microg pressurized metered dose inhaler (pMDI) propelled by either hydrofluoroalkane (HFA) 134a or chlorofluorocarbon (CFC) propellants, in adult patients with asthma. HFA 134a is a non-ozone depleting propellant used as a replacement for the CFC propellants 11 and 12 which are being phased out in accordance with the Montreal Protocol. Three hundred and eighty patients with mild to moderate asthma and 'room for improvement' in their treatment were randomized to receive fluticasone propionate 250 microg twice daily via pMDIs propelled by either CFC propellants 11 and 12 (n = 195) or HFA 134a (n = 185). Fluticasone propionate significantly improved lung function over the 4-week treatment period in both treatment groups. The improvement in mean morning peak expiratory flow (PEF) after 7 days of treatment was approximately 12 l min(-1) in both groups, rising to approximately 22 l min(-1) at the end of the 4-week treatment period. The adjusted mean difference between the two formulations over weeks 1-4 was -1 l min(-1) (90% confidence interval: -7, 5 l min(-1)), confirming their equivalence. Clinical comparability was also demonstrated with respect to secondary efficacy variables, including daily symptom scores, evening PEF and clinic visit expiratory measurements. There were no clinically relevant differences in adverse events or serum cortisol levels between the two groups. The fluticasone propionate 125 microg HFA 134a pMDI is an effective and well tolerated product and is a suitable replacement for the fluticasone propionate 125 microg CFC pMDI at a microgram equivalent dose.

In order to accomplish the transition from avascular to vascular growth, solid tumours secrete a diffusible substance known as tumour angiogenesis factor (TAF) into the surrounding tissue. Endothelial cells which form the lining of neighbouring blood vessels respond to this chemotactic stimulus in a well-ordered sequence of events consisting, at minimum, of a degradation of their basement membrane, migration, and proliferation. A model mechanism is presented which includes the diffusion of the TAF into the surrounding host tissue and the response of the endothelial cells to the chemotactic stimulus. The model accounts for the main observed events associated with the endothelial cells during the process of angiogenesis (i.e. cell migration and proliferation); the numerical results compare very well with experimental observations. The situation where the tumour (i.e. the source of TAF) is removed and the vessels recede is also considered.

Three-hundred and fifty patients with moderate to severe reversible obstructive airways disease (forced expiratory volume in 1 s or peak expiratory flow rate < or = 50% predicted, a 15% reversibility to inhaled salbutamol and symptomatic) were recruited into a multi-centre, multinational, double-blind, parallel-group randomized study. Two-hundred and eighty-three patients were randomized to receive 50 micrograms salmeterol twice daily or 100 micrograms salmeterol twice daily administered from a metered-dose inhaler for 3 months. Salbutamol (100 micrograms per metered actuation) was provided for symptomatic relief. Morning and evening peak expiratory flow rate (PEFR), day-time and night-time asthma symptoms and additional bronchodilator usage were recorded by the patient on a daily basis. Lung function and patient/physician assessment of treatment efficacy were recorded at scheduled clinic visits. Safety was determined by monitoring adverse events and standard biochemical, haematological and cardiovascular parameters. Salmeterol 100 micrograms twice daily was consistently superior to salmeterol 50 micrograms twice daily in morning and evening PEFR measurements (mean differences between the treatments: 10-14 l min-1 for morning, 95% CI-0, 22 l min-1, P = 0.047; and 10-15 l min-1 for evening, 95% CI 2, 22 l min-1, P = 0.023). The improvement in PEFR was independent of concurrent steroid usage, with the most marked improvement being seen in the more severe asthmatics requiring concurrent oral corticosteroids (mean differences between the treatments: 27-31 l min-1, 95% CI: 3,55 l m-1, P = 0.027).(ABSTRACT TRUNCATED AT 250 WORDS)

Unless they are furnished with an adequate blood supply and a means of disposing of their waste products by a mechanism other than diffusion, solid tumours cannot grow beyond a few millimetres in diameter. It is now a well-established fact that, in order to accomplish this neovascularization, solid tumours secrete a diffusable chemical compound known as tumour angiogenesis factor (TAF) into the surrounding tissue. This stimulates nearby blood vessels to migrate towards and finally penetrate the tumour. Once provided with the new supply of nutrient, rapid growth takes place. In this paper, a mathematical model is presented for the diffusion of TAF into the surrounding tissue. The complete process of angiogenesis is made up of a sequence of several distinct events and the model is an attempt to take into account as many of these as possible. In the diffusion equation for the TAF, a decay term is included which models the loss of the chemical into the surrounding tissue itself. A threshold distance for the TAF is incorporated in an attempt to reflect the results from experiments on corneal implants in test animals. By formulating the problems in terms of a free boundary problem, the extent of the diffusion of TAF into the surrounding tissue can be monitored. Finally, by introducing a sink term representing the action of proliferating endothelial cells, the boundary of the TAF is seen to recede, and hence the position and movement of the capillaries can be indirectly followed. The changing concentration gradient observed as the boundary recedes may offer a possible explanation for the initiation of anastomosis. Several functions are considered as possible sink terms and numerical results are presented. The situation where the tumour (i.e. the source of TAF) is removed is also considered.

The regional distribution of 5-hydroxytryptamine-1 receptors in the primate brain was studied by semi-quantitative autoradiographic analysis of tritiated ligand binding. Areas showing the highest density of 5-hydroxytryptamine-1 receptors (greater than 200 fmol [3H]5-hydroxytryptamine bound per mg tissue), included the cerebral cortex (laminae I-II), claustrum, posterior cell group of the basal nucleus of Meynert, the infracommissural part of the globus pallidus, cortical amygdaloid nucleus, hippocampal formation (CA1-subiculum region, the anterior CA2, CA3 and CA4 regions and the molecular layer of the dentate gyrus), thalamic nuclei (parafascicular, parataenial, paraventricular and superior central lateral nuclei), substantia nigra pars reticulata, dorsal raphe nucleus and choroid plexus. The distribution of 5-hydroxytryptamine-1 receptors is compared to the distribution of both 5-hydroxytryptamine receptors and terminal fields of serotonergic projections as previously described in subprimates.
