Na busca pela melhoria do compartilhamento do conhecimento operário em empresas de um setor representativo da economia brasileira, a presente pesquisa identificou e avaliou fatores relativos à gestão da produção, gestão do trabalho e gestão do conhecimento em empresas do setor automotivo. Os dados coletados foram obtidos por um questionário aplicado presencialmente com gestores de produção de três empresas (uma montadora, uma fabricante de autopeças e outra de motores) a fim de se avaliar a importância dos fatores estudados pela ótica gerencial. A avaliação das respostas usou o método Analytic Hierarchy Process - Incomplete Pairwise Comparison, que possibilitou evidenciar maior importância da conversa técnica-sistêmica entre os operários, o papel da instrução de trabalho e do treinamento nos processos de compartilhamento do conhecimento. A pesquisa contribui para a qualificação e o aprendizado de novos operários, além da investigação de um contexto favorável para criação e compartilhamento do conhecimento no “chão-de-fábrica”.

Bovine kobuvirus (BKV) is an infectious agent associated with neonatal calf diarrhoea (NCD), causing important economic losses to dairy and beef cattle herds worldwide. Here, we present the detection rate and characterize the genome of BKV isolated from diarrhoeic calves from a Central Italy herd. From January to December 2021, we collected blood samples and nasal and rectal swabs from 66 calves with severe NCD between 3 and 20 days of age. After virological (bovine coronavirus, bovine viral diarrhoea virus, and bovine rotavirus), bacteriological (Escherichia coli spp. and Salmonella spp.), and parasitological (Cryptosporidium spp., Eimeria spp., and Giardia duodenalis) investigations, we detected BKV using the metagenomic analysis. This result was confirmed using a specific polymerase chain reaction assay that revealed the number of BKV-positive nasal (24.2%) and rectal swabs (31.8%). The prevalence of BKV was higher than that of BCoV. Coinfection with BKV and BCoV was detected in 7.5% of the rectal swabs, highlighting the involvement of another infectious agent in NCD. Using next generation sequencing (NGS) approach, it was possible to obtain the complete sequence of the BKV genome from other two rectal swabs previously analysed by real-time PCR. This is the first report describing the whole genome sequence (WGS) of BKV from Italy. The Italian BKV genomes showed the highest nucleotide sequence identity with BKV KY407744.1, identified in Egypt in 2014. The sequence encoding VP1 best matched that of BKV KY024562, identified in Scotland in 2013. Considering the small number of BKV WGSs available in public databases, further studies are urgently required to assess the whole genome constellation of circulating BKV strains. Furthermore, pathogenicity studies should be conducted by inoculating calves with either only BKV or a combination with other enteric pathogens for understanding the probable role of BKV in NCD.

Infectious bovine rhinotracheitis (IBR), caused by Bovine alphaherpesvirus 1 (BoHV-1), is a disease of cattle responsible for significant economic losses worldwide. IBR is under certain communitarian regulations. Every member state can approve its own national IBR control program for the entire territory – or part of it – and can demand additional guarantees for bovids destined to its territory; therefore, every member state can be officially declared as entirely or partly IBR-free. The aim of this review is to provide an overview of IBR control and eradication programs in European countries. BoHV-1 control schemes were first introduced in the late 1970s, mainly in Northern and Central Europe. Depending on the seroprevalence rate, control strategies rely on identification and removal of seropositive animals or the use of glycoprotein E (gE)-deleted marker vaccines in infected herds. The implementation of a novel law for disease eradication at the EU level and of a European IBR data flow could make the goal of IBR eradication in all European countries easier to achieve.

Bovine respiratory syncytial virus (BRSV) is an economically significant pathogen in cattle production worldwide. Usually, it is detected in outbreaks of respiratory disease, most often during the winter period. During the middle of October 2018, a serious outbreak of respiratory disease occurred in a cattle farm comprising about 300 heads located in Central Italy. The herd was affected by a severe flu‐like syndrome unresponsive to any antibiotic treatment. Within 3 weeks, 39 adult animals died, and 12 abortions occurred. Direct and indirect laboratory tests were performed to detect the main pathogens causing the respiratory disease of the affected cattle. The results of laboratory investigations provided evidence of an acute and severe BRSV syndrome characterized by unusual mortality. In order to investigate the molecular underpinnings of this syndrome, phylogenetic analysis of the BRSV strain detected from the outbreak was carried out. The sequence analysis showed that the strain was genetically divergent from BRSV strains previously identified in Italy, as it showed high sequence similarity of more than 97% with strains isolated during a major BRSV epizootic that occurred in Sweden, Norway and Denmark during 2010–2011. The infection of the herd in Italy with this BRSV strain was likely due to the introduction of animals imported into Italy from abroad., Bovine respiratory syncytial virus (BRSV) is an economically significant pathogen in cattle production. In Italy, although respiratory problems due to BRSV are frequently described, major outbreaks have not yet been reported. Here, we report an outbreak caused by a BRSV strain with high mortality that occurred in a dairy farm in Central Italy; this strain shows a sequence similarity of >97% to the strains isolated during a major BRSV epizootic that occurred in Sweden and Norway during 2010–2011. Epidemiological analysis indicated that recently introduced heifers from Germany were the likely carriers of the virus.

Enzootic Bovine Leukosis (EBL), caused by the bovine leukemia virus (BLV), has been eradicated in over 20 countries, most of which are in Western Europe. The European Commission, in 2017, declared Italy to be an officially EBL-free country by means of Commission Implementing Decision (EU) 2017/1910, despite the presence of some infection clusters located in four regions of Central-Southern Italy. As a consequence of persisting infection, the Italian Ministry of Health established specific eradication measures in these areas. In collaboration with the National Reference Laboratory for the Study of Ruminant Retroviral Infectious Diseases, the Ministry of Health employed data from the veterinary information system digital platform, combined with a gap analysis exercise, to monitor and verify the progress of control activities within infection clusters during the period 2018–2021. Our aim was to identify any remaining gaps and, consequently, specific measures to eliminate the factors favouring EBL persistence, on the basis of a description and analysis of the current data regarding epidemiological trends in Italian clusters. The final goal is to achieve the implementation of a less expensive surveillance plan in these areas, as well. The results of comprehensive analysis showed that the eradication activities had been effectively implemented by official local veterinary services, resulting in a drastic reduction of EBL outbreaks in most territories during the period 2018–2021.

The proposed model maximizes 14 objectives and 14 constraints by referring in a generalized way to lane comfort, safety, path objectivity, network connectivity, intermodality with bus service, and monetary budgets. In the context of a developing country, we believe that this model can address the transit network design problem (TNDP) as an innovative proposal for integration between alternative modes of transport and buses. A case study in Sao Jose dos Campos city, Sao Paulo state, Brazil, is conducted, and exact solutions are obtained with partially connected networks (first approach) and fully connected networks (second approach). A scenario analysis enables verification of the integrated transportation system performance and, for the case study, prioritizes the least expensive bikeway type. Border disease virus (BDV) belongs to the genus Pestivirus of the family Flaviviridae. Interspecies transmission of BDV between sheep, cattle, and pigs occurs regularly, sometimes making diagnosis a challenge. BDV can yield substantial economic losses, including prenatal and postnatal infections in lambs, which are the primary source of infection and maintenance of the virus in the population. Since BDV is antigenically and genetically related to bovine viral diarrhea virus (BVDV), it might pose a significant risk to cattle, influencing BVDV eradication campaigns. Similarly, the presence of BDV in swine herds due to pestivirus spillover between small ruminants and pigs might cause uncertainty in classical swine fever virus (CSFV) diagnostics. Therefore, knowledge of BDV epidemiology in different geographical regions will help prevent its spread and optimize control measures. Previous epidemiological studies have shown that various BDV genotypes are predominant in different countries. This review provides an overview of the spread of BDV world-wide in different host species.

In this study, we demonstrated for the first time in Italy, the serological cross-reactivity between Bovine alphaherpesvirus 2 (BoHV-2) and Bovine alphaherpesvirus 1 (BoHV-1). Five months after arriving at a performance test station in Central Italy, a 6-month-old calf, which was part of a group of 57 animals, tested positive for BoHV-1 in a commercial gB-ELISA test. It was immediately transferred to the quarantine unit and subjected to clinical observation and serological and virological investigations. During this period, the calf showed no clinical signs. The results from laboratory investigations demonstrated the presence of antibodies via competitive glycoprotein B (gB) ELISAs, indirect BoHV-1 ELISAs, and indirect BoHV-2 ELISAs. Furthermore, the plaque reduction assay provided evidence for the presence of antibodies only for BoHV-2, whereas the virus neutralization test showed negative results for both BoHV-1 and BoHV-5. These findings strongly suggest the occurrence of a serological cross-reactivity between BoHV-2 and BoHV-1. Interference of BoHV-2 antibodies in serological BoHV-1 diagnostics should be considered during routine IBR tests, especially when animals are kept in a performance test station.

The identification of cross-reactive monoclonal antibodies (mAbs) that recognize orthologous leukocyte differentiation molecules (LDM) in buffaloes has overcome a major impediment limiting research on the immune response to pathogens and development of vaccines. As reported, two pilot trials were conducted to accomplish two objectives: (1) demonstrate that multiparameter flow cytometry can be conducted equally well in buffalo with mAbs directly and indirectly labeled with fluorochromes in research and (2) flow cytometry can be used to compare and extend studies on diseases of economic importance to buffalo using bovine viral diarrhea virus (BVDV) as a model pathogen. Pregnant buffalo cows were infected with BVDV-1 at 81 (trial 1) and 203 (trial 2) days post artificial insemination and flow cytometric evaluations were performed at 0, 3, 4, and 14 days after infection (dpi). Fluorochrome conjugated mAbs were used in trial 1, and fluorochrome conjugated goat isotype specific anti-mouse antibodies were used to label mAbs in trial 2. Flow cytometric analysis revealed a transient lymphopenia occurs during the 1st days following infection similar to lymphopenia reported in cattle. In particular, significant differences were observed between pre- and post-infection absolute values of T lymphocytes (−56%, P < 0.01). CD21+ B lymphocytes (−65%, P = 0.04), and Natural Killer cells (−72%, P < 0.001). No significant differences were observed in monocytes and neutrophil absolute values, or the CD4:CD8 ratio. Animal health status was followed until 15 days after calving. No clinical signs of infection were observed during the evaluation period, however, animals in trial 1 developed complications later the infection. One cow aborted at 57 days post-infection, the second cow developed a prolapse a day after calving and died. These two animals also showed a more pronounced lymphopenia in comparison with animals infected at 203 days of pregnancy (e.g., −77 vs. −22% T lymphocytes at 3 dpi, respectively). The pilot studies have demonstrated that it is possible to use multicolour multiparameter flow cytometry to study the immune response to pathogens affecting the health of buffalo.

A investigação dos critérios de infraestrutura que influenciam a demanda cicloviária e que deveriam compor a modelagem matemática foi realizada por meio da Revisão da Literatura e cinco critérios identificados, referentes ao: Conforto, Segurança, Objetividade, Intermodalidade e Continuidade. Cada critério é composto por uma série de subcritérios, dezesseis no total, classificados como parâmetros de projeto de entrada e/ou de saída para o modelo matemático, segundo funções objetivos e restrições. Uma aplicação do modelo foi realizada em um caso real para cidade de grande porte, em São José dos Campos – SP, e obtidas soluções exatas com redes parcialmente conexas (abordagem 1) e redes totalmente conexas (abordagem 2), integradas aos pontos de parada de ônibus e as linhas de ônibus da região delimitada, por meio da linguagem de modelagem General Algebric Modeling System (GAMS) e o solver CPLEX. Os resultados obtidos foram aderentes a realidade da macrozona de tráfego na região estudada e contribuem para o planejamento urbano como validado pela Secretaria de Mobilidade Urbana de São José dos Campos. A análise de diversos cenários para o modelo matemático, ponderando os objetivos do modelo, possibilitou verificar o comportamento dos resultados, que tende a priorizar os caminhos ciclísticos mais baratos e em seguida, aqueles de maior preferência relatada. No modelo, a abordagem 1 atende a todas as linhas de ônibus, enquanto na abordagem 2 o atendimento de todas linhas se mostrou incompatível com a conexidade total da rede, sendo assim, necessária a maximização do atendimento das linhas do transporte coletivo para essa abordagem. Essa pesquisa compreende ao primeiro modelo de Otimização Linear Multiobjetivo a lidar com o tema Transit Network Design Problem (TNDP), propondo soluções conexas para o projeto de redes integradas envolvendo modos alternativos de transporte e ônibus, sobretudo em uma realidade de um país em desenvolvimento. The present work created a mathematical model that contributes to design bikeway networks integrated to the public transport service by buses in urban areas, using the Network Multiobjective Optimization through Mixed by means of Mixed Integer Linear Programming (MILP or MIP). The investigation of the infrastructure criteria that influence cycling demand and that should set the mathematical modeling was carried out through the Literature Review and five criteria were identified, referring to: Comfort, Safety, Objectivity, Intermodality and Connectivity. Each criteria is composed of a subcriteria set, sixteen in total, which performed as input and/or output design parameters for the mathematical model, according to fitness functions and constraints. An application of the model was carried out in a real case in São Jose dos Campos city - Sao Paulo state, and exact solutions were obtained with partially connected networks (approach 1) and fully connected networks (approach 2), integrated to the stop points by bus and bus lines in the defined region, using the General Algebric Modeling System (GAMS) modeling language and the CPLEX solver. The results obtained were adherent to the reality of the traffic macrozone in the studied region and contribute to urban planning, as validated by the São José dos Campos Urban Mobility Secretariat. The analysis of several scenarios for the mathematical model, considering the model's objectives, made it possible to verify the behavior of the results, which tends to prioritize the cheapest cycling paths and then those of the most reported preference. In the model, approach 1 serves all bus lines, while in approach 2 the service of all lines proved to be incompatible with full network connectivity, therefore, it is necessary to maximize the service of public transportation lines for this approach. This research comprises the first Multiobjective Linear Optimization model to deal with the Transit Network Design Problem (TNDP) theme, proposing related solutions for the design of integrated networks involving alternative modes of transport and buses, especially in a reality in a developing country. In recent years, extracellular vesicles (EVs), cell-derived micro and nano-sized structures enclosed in a double-layer membrane, have been in the spotlight for their high potential in diagnostic and therapeutic applications. Indeed, they act as signal mediators between cells and/or tissues through different mechanisms involving their complex cargo and exert a number of biological effects depending upon EVs subtype and cell source. Being produced by almost all cell types, they are found in every biological fluid including milk. Milk EVs (MEVs) can enter the intestinal cells by endocytosis and protect their labile cargos against harsh conditions in the intestinal tract. In this study, we performed a metabolomic analysis of MEVs, from three different species (i.e., bovine, goat and donkey) by mass spectroscopy (MS) coupled with Ultrahigh-performance liquid chromatography (UHPLC). Metabolites, both common or specific of a species, were identified and enriched metabolic pathways were investigated, with the final aim to evaluate their anti-inflammatory and immunomodulatory properties in view of prospective applications as a nutraceutical in inflammatory conditions. In particular, metabolites transported by MEVs are involved in common pathways among the three species. These metabolites, such as arginine, asparagine, glutathione and lysine, show immunomodulating effects. Moreover, MEVs in goat milk showed a greater number of enriched metabolic pathways as compared to the other kinds of milk.

Infectious bovine rhinotracheitis (IBR)/infectious pustular vulvovaginitis (IPV) caused by Bovine alphaherpesvirus 1 (BoHV-1) is a significant disease in domestic and wild cattle. In June 2015, the Ministry of Agriculture, Food and Forestry in Italy approved a national surveillance plan to control and eradicate IBR in beef cattle breeds. The objective of this study was to evaluate the results of the first year of the IBR voluntary surveillance plan in Italy. The aim of the plan is to eradicate IBR in all bovines recorded in the National Herd Book for Italian beef cattle breeds over six years. Monetary incentives are used to encourage breeders to achieve the annual seroprevalence ranges stated in the plan. A Ministerial decree states that all bovines in breeding herds and aged older than 12 months should be serologically tested. Serum samples were tested for presence of the antibody to glycoprotein E of BoHV-1 using commercially available enzyme-linked immunosorbent assays. The national herd seroprevalence was 55.49% (95% confidence interval [CI] 52.01-58.92). Of 25,121 bovines tested for antibodies against BoHV-1, 8014 were positive. The seroprevalence in animals from autochthonous Italian cattle breeds was 31.89% (95% CI 31.31-32.47). Seroprevalence was highest in Podolica cattle (55.14%; 95% CI 54.07-56.21), lowest in Maremmana cattle (9.95%; 95% CI 7.99-12.31), and intermediate in Chianina (22.01%; 95% CI 21.03-23.01), Marchigiana (24.85%; 95% CI 23.52-26.23), and Romagnola (15.60%; 95% CI 14.62-16.64) cattle. These seroprevalence rates indicate a need for intervention to decrease the inevitable severe economic losses arising from BoHV-1 infection. Although some regions in Italy have a long history of combatting BoHV-1 infection, only the province of Bolzano has eradicated IBR.

We evaluated the use of oral fluid as an alternative to serum samples for Classical swine fever virus (CSFV) detection. Individual oral fluid and serum samples were collected at different times post-infection from pigs that were experimentally inoculated with CSFV Alfort 187 strain. We found no evidence of CSFV neutralizing antibodies in swine oral fluid samples under our experimental conditions. In contrast, real-time reverse transcription–polymerase chain reaction could detect CSFV nucleic acid from the oral fluid as early as 8 d postinfection, which also coincided with the time of initial detection in blood samples. The probability of CSFV detection in oral fluid was identical or even higher than in the corresponding blood sample. Our results support the feasibility of using this sampling method for CSFV genome detection, which may represent an additional cost-effective tool for CSF control.

Also, the paper raises issues and challenges involved for production organizations to engage in cross-national knowledge sharing, which remain relatively under-explored.Research method Drawing on the Analytic Hierarchy Process (AHP) approach, we conduct a comparative survey among the production managers in five automotive plants in China and Brazil. The AHP approach is based on Objective: Promote knowledge sharing among production workers; Criteria of knowledge conversion as Socialization, Externalization, Internalization; and Alternatives as Structure, Communication, Training, Incentive, Standard Operating Procedure, Problem Solving Methodology. The methodology focused on the 3 Brazilian and 2 Chinese automotive plants in our fieldwork.Main findings While many authors argue that socialization (dialogue) is the key form of worker knowledge sharing, our findings indicate that workers and managers perceive internalisation as more important in the automotive context. The significant differences between Chinese and Brazilian workers on the relative importance they attributed to knowledge sharing processes and corresponding enabling factors reinforce our understanding about the challenges of knowledge sharing across cultures and lends support to a more particularistic debate of knowledge management. While automotive workers in Brazil preferred to receive more training and better incentive schemes as essential knowledge sharing support mechanisms, Chinese workers valued more standardization through structure and standard operation procedures. The study reaffirms the role of knowledge sharing in continuous improvement and incremental innovation in shop floor operations.Implications for theory and practice In lieu of the gaps in the extant literature, this research aims to further identify and evaluate factors that influence cross-national knowledge sharing in automotive production contexts. Background. Equine adipose-derived mesenchymal stromal cells (e-AdMSC) exhibit attractive proregenerative properties strongly related to the delivery of extracellular vesicles (EVs) that enclose different kinds of molecules including RNAs. In this study, we investigated small RNA content of EVs produced by e-AdMSC with the aim of speculating on their possible biological role. Methods. EVs were obtained by ultracentrifugation of the conditioned medium of e-AdMSC of 4 subjects. Transmission electron microscopy and scanning electron microscopy were performed to assess their size and nanostructure. RNA was isolated, enriched for small RNAs (Results. Electron microscopy showed the presence of vesicles ranging in size from 30 to 300 nm and expressing typical markers. RNA analysis revealed that ribosomal RNA was the most abundant fraction, followed by small nucleolar RNAs (snoRNAs, 13.67%). Miscellaneous RNA (misc_RNA) reached 4.57% of the total where Y RNA, RNaseP, and vault RNA represented the main categories. miRNAs were sequenced at a lower level (3.51%) as well as protein-coding genes (1.33%). Pathway analyses on the protein-coding fraction revealed a significant enrichment for the “ribosome” pathway followed by “oxidative phosphorylation.” Gene Ontology analysis showed enrichment for terms like “extracellular exosome,” “organelle envelope,” “RNA binding,” and “small molecule metabolic process.” The miRNA target pathway analysis revealed the presence of “signaling pathways regulating pluripotency of stem cells” coherent with the source of the samples. Conclusion. We herein demonstrated that e-AdMSC release EVs enclosing different subsets of small RNAs that potentially regulate a number of biological processes. These findings shed light on the role of EVs in the context of MSC biology.
