O objetivo deste estudo foi propor um método de estratificação em classes homogêneas de estoque volumétrico da floresta ombrófila densa de terra firme não explorada, empregando-se as técnicas de análises de agrupamento e discriminante. A pesquisa foi conduzida na Unidade de Manejo Florestal (UMF) da Fazenda Tracajás (02º35'53"S e 47º47'10"W), pertencente à empresa Nova Era Agroflorestal, de propriedade do Grupo Rosa Madeireira, Município de Paragominas, Estado do Pará. Foi realizado um censo (100%), no qual se estimaram os volumes do fuste comercial das árvores com dap > 45 cm de 55 espécies comerciais em 49 talhões de exploração com 10 ha cada um. Reuniram-se os volumes estimados do fuste comercial das árvores individuais por talhão em ordem crescente. Elaborou-se uma matriz X de dados desses volumes, em que cada variável x ij representou o i-ésimo volume classificado no j-ésimo talhão. A matriz X foi utilizada como input nas análises de agrupamento e discriminante. A aplicação da análise de agrupamento, método de Ward, resultou em agrupamentos hierárquicos dos talhões em classes de estoques. A análise do dendrograma permitiu estratificar o povoamento em três grupos homogêneos e distintos, denominados classes I, II e III de estoques volumétricos. A análise discriminante, método de Fisher, indicou que 100% dos talhões foram corretamente classificados. A classificação multivariada da floresta em classes de estoques volumétricos mostrou-se um método eficiente na estratificação de áreas homogêneas de florestas ineqüiâneas, as quais podem se constituir em estratos, compartimentos, classes de sítio e unidades de produção anual.

Este trabalho teve como objetivos desenvolver e testar um algoritmo com base na metaheurística busca tabu (BT), para a solução de problemas de gerenciamento florestal com restrições de inteireza. Os problemas avaliados tinham entre 93 e 423 variáveis de decisão, sujeitos às restrições de singularidade, produção mínima e produção máxima periódicas. Todos os problemas tiveram como objetivo a maximização do valor presente líquido. O algoritmo para implementação da BT foi codificado em ambiente delphi 5.0 e os testes foram efetuados em um microcomputador AMD K6II 500 MHZ, com memória RAM de 64 MB e disco rígido de 15GB. O desempenho da BT foi avaliado de acordo com as medidas de eficácia e eficiência. Os diferentes valores ou categorias dos parâmetros da BT foram testados e comparados quanto aos seus efeitos na eficácia do algoritmo. A seleção da melhor configuração de parâmetros foi feita com o teste L&O, a 1% de probabilidade, e as análises através de estatísticas descritivas. A melhor configuração de parâmetros propiciou à BT eficácia média de 95,97%, valor mínimo igual a 90,39% e valor máximo igual a 98,84%, com um coeficiente de variação de 2,48% do ótimo matemático. Para o problema de maior porte, a eficiência da BT foi duas vezes superior à eficiência do algoritmo exato branch and bound, apresentando-se como uma abordagem muito atrativa para solução de importantes problemas de gerenciamento florestal.

No presente trabalho foram analisados a dinâmica de sucessão, o crescimento e a produção de uma Floresta Ombrófila Densa secundária, após corte de cipós. O tratamento silvicultural foi executado com o objetivo de promover o rápido retorno da floresta às suas condições primárias ou, no mínimo, diminuir o intervalo de tempo entre dois ciclos de corte sucessivos. Os dados foram provenientes de um experimento implantado em 1989 e medido bienalmente na Reserva Natural da Companhia Vale do Rio Doce S.A., localizada entre os municípios de Linhares e Jaguaré, ES, Brasil. Foi empregada amostragem sistemática, tendo sido considerados como indivíduos da regeneração natural todos aqueles com dap < 5,0 cm. Foram analisadas as estimativas dos parâmetros da composição florística, a diversidade de espécies e a estrutura da regeneração natural, em cada ocasião de monitoramento. As análises permitiram concluir que, para todas as categorias analisadas, houve grandes variações positivas nas taxas de regeneração natural e aumento da área basal desses indivíduos ao longo do período de monitoramento, indicando ser este compartimento da floresta o mais beneficiado pela aplicação do tratamento e, indiretamente, o estrato arbóreo, ao receber o ingresso desses indivíduos. Assim, o corte de cipós favoreceu a dinâmica de sucessão secundária, diminuindo a concorrência por espaço, nutrientes e luz, o que proporciona importante desenvolvimento da regeneração natural e grande elevação da taxa de ingresso de novos indivíduos no estrato arbóreo.

The adsorption of H and S2- species on Pd (100) has been studied with ab initio, density-functional calculations and electrochemical methods. A cluster of five Pd atoms with a frozen geometry described the surface. The computational calculations were performed through the GAUSSIAN94 program, and the basis functions adapted to a pseudo-potential obtained by using the Generator Coordinate Method adapted to the this program. Using the cyclic voltammetry technique through a Model 283 Potentiostat/Galvanostat E.G.&G-PAR obtained the electrochemical results. The calculated chemisorption geometry has a Pd-H distance of 1.55Å, and the potential energy surface was calculated using the Becke3P86//(GCM/DFT/SBK) methodology. The adsorption of S2- ions on Pd surface obtained both through comparison between the experimental and theoretical results, at MP2 level, suggest a S2- absorption into the metallic cluster. The produced Pd-(S2-) system was show to be very stable under the employed experimental conditions. The paper has shows the powerful aid of computational methods to interpret adsorption experimental data.

Ab initio Hartree-Fock (HF), Density Functional (B3LYP) and electron correlation (MP2) methods have been used to caracterize the aqueous medium intramolecular hydrogen bond in a-alanine. The 6-31G* and 6-31++G** were taken from Gaussian94 library. We were concerned on the structure of three conformers of a-alanine, in their neutral form plus on the structure of the zwitterionic form (Z). The Z structure is a stationary point at the HF/6-31G* level but it is not when diffuse functions and electron correlation are included. This results shows that the Z form does not exist in the gas phase. The inclusion of solvent effects changed significantly the results obtained in gas phase, therefore this inclusion make the Z form a stationary point within all level of theory, and the relative energy depends dramatically on the level of calculation.

Sepsis poses a significant challenge due its lethality, involving multiple organ dysfunction and impaired immune responses. Among several factors affecting sepsis, monocytes play a crucial role; however, their phenotype, proteomic profile, and function in septic shock remain unclear. Our aim was to fully characterize the subpopulations and proteomic profiles of monocytes seen in septic shock cases and discuss their possible impact on the disease. Peripheral blood monocyte subpopulations were phenotype based on CD14/CD16 expression by flow cytometry, and proteins were extracted from the monocytes of individuals with septic shock and healthy controls to identify changes in the global protein expression in these cells. Analysis using 2D-nanoUPLC-UDMSE identified 67 differentially expressed proteins in shock patients compared to controls, in which 44 were upregulated and 23 downregulated. These proteins are involved in monocyte reprogramming, immune dysfunction, severe hypotension, hypo-responsiveness to vasoconstrictors, vasodilation, endothelial dysfunction, vascular injury, and blood clotting, elucidating the disease severity and therapeutic challenges of septic shock. This study identified critical biological targets in monocytes that could serve as potential biomarkers for the diagnosis, prognosis, and treatment of septic shock, providing new insights into the pathophysiology of the disease.

Depression is a common mood disorder and many patients do not respond to conventional pharmacotherapy or experience a variety of adverse effects. This work proposed that riparin I (RIP I) and riparin II (RIP II) present neuroprotective effects through modulation of astrocytes and microglia, resulting in the reversal of depressive-like behaviors. To verify our hypothesis and clarify the pathways underlying the effect of RIP I and RIP II on neuroinflammation, we used the chronic unpredictable mild stress (CUMS) depression model in mice. Male Swiss mice were exposed to stressors for 28 days. From 15 th to the 22 nd day, the animals received RIP I or RIP II (50 mg/kg) or fluoxetine (FLU, 10 mg/kg) or vehicle, by gavage. On the 29 th day, behavioral tests were performed. Expressions of microglia (ionized calcium-binding adaptor molecule-1 - Iba-1) and astrocyte (glial fibrillary acidic protein - GFAP) markers and levels of cytokines tumor necrosis factor alfa (TNF-α) and interleukin 1 beta (IL-1β) were measured in the hippocampus. CUMS induced depressive-like behaviors and cognitive impairment, high TNF-α and IL-1β levels, decreased GFAP, and increased Iba-1 expressions. RIP I and RIP II reversed these alterations. The biometric differences between fruits and seeds are useful characteristics that can provide important data for the investigation and preservation of the species and may be linked to environmental and genetic influences. In this sense, considering the importance of this species and the need for conservation, the objective was to carry out physical characterization of the fruits and seeds of Hymenaea martiana as well as to determine the seed imbibition curve. The experiment was conducted at the Seed Analysis Laboratory at the Agricultural Sciences Center at the Federal University of Paraíba in Areia, PB. The evaluations carried out were as follows: biometry of fruits and seeds, number of seeds per fruit, colorimetry of fruits and seeds, percentage of damaged seeds, weight of a thousand seeds, seed water content, mass and imbibition curve. The biometric data were subjected to descriptive analysis to determine the minimum, maximum, average value, standard deviation, asymmetry, and kurtosis of the fruits and seeds. In terms of the biometric characteristics of the fruits and seeds of H. martiana, there was a marked variation, with average fruit lengths of 90.28 mm, widths of 46.83 mm, thicknesses of 34.69 mm, weights of 65.86 g and four seeds per fruit. The average length, width, thickness and weight of the seeds were 23.75 mm, 18.34 mm, 12.71 g and 4.13 g, respectively. The fruits were darker than the seeds, and both the fruits and seeds had red tones. Compared with nonscarified seeds, scarified seeds absorb a greater amount of water.

The State University of North Fluminense Darcy Ribeiro (UENF) has been developing for fifteen years a breeding program that aims at the development of new cultivars of elephant grass due to its high potential and the low availability of cultivars developed by genetic breeding programs that meet the needs of producers in the State of Rio de Janeiro. In this sense, inbred families were also obtained as a way of fixing potential alleles for traits related to production, as the inbreeding process apparently does not strongly affect elephant grass in aspects related to inbreeding depression. This study aimed to estimate genetic diversity, variance components and prediction of genotypic values in 11 (S 1 ) elephant grass families, and perform the truncation and simultaneous selection of traits using the selection index, by mixed models. The experimental design consisted of randomized blocks with 11 (S 1 ) families, three replications, and six plants per plot. For variables dry matter production, percentage of dry matter, plant height, stem diameter, number of tillers and leaf blade width, was performed the estimation of genetic parameters and selection of the best genotypes based selection index using mixed model. The descriptors were subjected to correlation analysis, distance matrices were generated by the Mahalanobis method, and individuals were grouped by the UPGMA method. In the selection via mixed models (REML/BLUP), families 6, 11, 8, 1, 3, 7, and 9 contributed most of the genotypes selected for the evaluated traits, indicating their high potential to generate superior genotype. The selection indices via mixed models indicated that the multiplicative index presented a greater selection gain.The phenotypic correlations showed the possibility of performing an indirect selection from six evaluated traits.The genotypes were separated into 18 groups by the Mahalanobis distance, allowing the observation of a wide genetic diversity. Home office (HO) stands out as one of the most promising and popular forms of teleworking, especially after the COVID-19 pandemic. Therefore, many companies want to implement or maintain this working method, given its numerous advantages. However, there are adverse effects that are mainly related to physical and mental health. This article presents ergonomic analyses of HOs in neighborhoods considered heat islands. Temperature levels, extreme low-frequency non-ionizing radiation (ELF-NIR), illuminance, physical layout characteristics, and physiological parameters of teleworkers were measured. The results reveal that 92% of these professionals work 6 to 8 h daily with an ambient temperature between 25 and 30 °C, illumination levels in the range 11.20-290 Lux, and ELF-NIR > 0.4 µT. The majority of teleworkers are overweight (BMI > 24.9), and some of them have blood pressure higher than average values (129 mmHg for systolic and 84 mmHg for diastolic) in addition to a reduction in the number of red blood cells and hematocrits. Symptoms such as burning sensation, dryness, tired eyes, redness, itching, and photophobia (light sensitivity) show a 68.95% similarity. These HOs do not meet the required ergonomic and health standards.

Fungi of the genus Trichoderma spp have been related to the production of hormones or correlated with growth factors, promoting greater efficiency in the use of some nutrients, thus allowing greater availability and absorption by plants. In this context, the objective of this study was to determine the dose of organomineral fertilizer from cupuaçu (Theobroma grandiflorum) residues and the efficiency of Trichoderma harzianum on the initial growth and morphophysiological quality of Mezilaurus itauba seedlings in the northern Amazon. Dose of 50% of the organomineral fertilizer from cupuaçu residues (ORFCup) with Trichoderma harzianum promotes better quality and robustness in Mezilaurus itauba seedlings. The presence of Trichoderma harzianum + 50% ORFCup promotes positive gains in the root biomass of Mezilaurus itauba seedlings. The presence of Trichoderma harzianum promotes an increase in chlorophylls a and b contents in Mezilaurus itauba seedlings. For the production of Mezilaurus itauba seedlings, it is recommended to use Trichoderma harzianum + 50% ORFCup, as it promoted increments in all physiological and morphological indices under the conditions of the present study.

Common bean provides diet rich in vitamins, fiber, minerals, and protein, which could contribute into food security of needy populations in many countries. Developing genotypes that associate favorable agronomic and grain quality traits in the common bean crop could increase the chances of adopting new cultivars black bean. In this context, the present study aimed at selection of superior black bean lines using multi-variate indexes, Smith-Hazel-index, and genotype by yield*trait biplot analysis. These trials were conducted in Campos dos Goytacazes - RJ, in 2020 and 2021. The experimental design used was randomized blocks, with 28 treatments and three replications. The experimental unit consisted of four rows 4.0 m long, spaced at 0.50 m apart, with a sowing density of 15 seeds per meter. The two central rows were used for the evaluations. The selection of superior genotypes was conducted using the multiple trait stability index (MTSI), multi-trait genotype-ideotype distance index (MGIDI), multi-trait index based on factor analysis and genotype-ideotype distance (FAI-BLUP), Smith-Hazel index, and Genotype by Yield*Trait Biplot (GYT). The multivariate indexes efficiently selected the best black bean genotypes, presenting desirable selection gains for most traits. The use of multivariate indexes and GYT enable the selection of early genotypes with higher grain yields. The present study was conducted to determine the efficiency of organomineral fertilizer from cupuaçu residues (ORFCup) and dose of maximum technical efficiency of Azospirillum brasilense on the initial growth and morphophysiological quality of Mezilaurus itauba seedlings in the northern Amazon. The variables evaluated were: shoot height (H, cm), stem diameter (SD, mm), shoot dry mass (SDM, g plant-1), root dry mass (RDM, g plant-1) total dry mass (TDM, g plant-1), Dickson quality index (DQI), net assimilation rate (NAR, g m-2 day-1), leaf relative growth rate (RGR, g m-2 day-1), leaf area ratio (LAR, m2 g-1), leaf relative growth rate (RGR, g m-2 day-1), leaf area ratio (LAR, m2 g-1), specific leaf area (SLA, cm2 g-1), and leaf mass ratio (LMR, g g-1). Organomineral fertilizer from cupuaçu residues promotes better quality and robustness in M. itauba seedlings at the dose of maximum technical efficiency of 0.45 mL. L-1 of A. brasilense.

Here, in a multi-ancestry genome-wide association study meta-analysis of kidney cancer (29,020 cases and 835,670 controls), we identified 63 susceptibility regions (50 novel) containing 108 independent risk loci. In analyses stratified by subtype, 52 regions (78 loci) were associated with clear cell renal cell carcinoma (RCC) and 6 regions (7 loci) with papillary RCC. Notably, we report a variant common in African ancestry individuals ( rs7629500 ) in the 3' untranslated region of VHL, nearly tripling clear cell RCC risk (odds ratio 2.72, 95% confidence interval 2.23-3.30). In cis-expression quantitative trait locus analyses, 48 variants from 34 regions point toward 83 candidate genes. Enrichment of hypoxia-inducible factor-binding sites underscores the importance of hypoxia-related mechanisms in kidney cancer. Our results advance understanding of the genetic architecture of kidney cancer, provide clues for functional investigation and enable generation of a validated polygenic risk score with an estimated area under the curve of 0.65 (0.74 including risk factors) among European ancestry individuals., (© 2024. The use of biomass for energy production constitutes a promising strategy that warrants the search for new sources of biomass. Elephant grass has been gaining notoriety due to its high dry matter yield and rapid growth. The present study was carried out to quantify the genetic divergence of nine elephant grass half-sib families in order to identify genotypes with greater genetic divergence and productive potential for hybridization, using the hierarchical clustering methodology based on principal components. Half-sib families were generated using genotypes from the Active Germplasm Bank of Elephant Grass. The experiment was laid out in a randomized-block design with nine half-sib families, three replicates, and eight plants per plot. A total of 216 genotypes of elephant grass were evaluated. Principal component (PC), biplot, and hierarchical clustering analyses for diversity estimation were conducted using R software. The first two PCs of biplot analysis accounted for 64% of the cumulative variation. Dry matter yield was the most important trait for genotype discrimination (0.89), followed by plant height (0.67) and stem diameter (0.61) in PC1. In this analysis, the distances between accessions were considered and there were no family links, which indicates the existence of wide variability within the evaluated families, since genotypes belonging to the same family were not grouped together, but rather distributed into different groups. Crosses between genotypes of group three and genotypes of groups one and two are recommended for the development of high-yielding genotypes when aiming at energy production.

The early diagnosis of leprosy serves as an important tool to reduce the incidence of this disease in the world. Phage display (PD) technology can be used for mapping new antigens to the development of immunodiagnostic platforms. Our objective was to identify peptides that mimic Mycobacterium leprae proteins as serological markers using phage display technology. The phages were obtained in the biopanning using negative and positive serum from household contacts and leprosy patients, respectively. Then, the peptides were synthesized and validated in silico and in vitro for detection of IgG from patients and contacts. To characterize the native protein of M. leprae, scFv antibodies were selected against the synthetic peptides by PD. The scFv binding protein was obtained by immunocapture and confirmed using mass spectrometry. We selected two phase-fused peptides, MPML12 and MPML14, which mimic the HSP60 protein from M. leprae. The peptides MPML12 and MPML14 obtained 100% and 92.85% positivity in lepromatous patients. MPML12 and MPM14 detect IgG, especially in the multibacillary forms. The MPML12 and MPML14 peptides had positivity of 11.1% and 16.6% in household contacts, respectively. There was no cross-reaction in patient's samples with visceral leishmaniasis, tuberculosis and other mycobacteriosis for both peptides. Given these results and the easy obtainment of mimetic antigens, our peptides are promising markers for application in the diagnosis of leprosy, especially in endemic and hyperendemic regions., (© 2023. Familial hypercholesterolemia (FH) is a monogenic disease characterized by high plasma low-density lipoprotein cholesterol (LDL-c) levels and increased risk of premature atherosclerotic cardiovascular disease. Mutations in FH-related genes account for 40% of FH cases worldwide. In this study, we aimed to assess the pathogenic variants in FH-related genes in the Brazilian FH cohort FHBGEP using exon-targeted gene sequencing (ETGS) strategy. FH patients (n = 210) were enrolled at five clinical sites and peripheral blood samples were obtained for laboratory testing and genomic DNA extraction. ETGS was performed using MiSeq platform (Illumina). To identify deleterious variants in LDLR, APOB, PCSK9, and LDLRAP1, the long-reads were subjected to Burrows-Wheeler Aligner (BWA) for alignment and mapping, followed by variant calling using Genome Analysis Toolkit (GATK) and ANNOVAR for variant annotation. The variants were further filtered using in-house custom scripts and classified according to the American College Medical Genetics and Genomics (ACMG) guidelines. A total of 174 variants were identified including 85 missense, 3 stop-gain, 9 splice-site, 6 InDel, and 71 in regulatory regions (3'UTR and 5'UTR). Fifty-two patients (24.7%) had 30 known pathogenic or likely pathogenic variants in FH-related genes according to the American College Medical and Genetics and Genomics guidelines. Fifty-three known variants were classified as benign, or likely benign and 87 known variants have shown uncertain significance. Four novel variants were discovered and classified as such due to their absence in existing databases. Melaleuca alternifolia essential oil (MaEO) is a green antimicrobial agent suitable for confection eco-friendly disinfectants to substitute conventional chemical disinfectants commonly formulated with toxic substances that cause dangerous environmental impacts. In this contribution, MaEO-in-water Pickering emulsions were successfully stabilized with cellulose nanofibrils (CNFs) by a simple mixing procedure. MaEO and the emulsions presented antimicrobial activities against Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli). Moreover, MaEO deactivated the SARS-CoV-2 virions immediately. FT-Raman and FTIR spectroscopies indicate that the CNF stabilizes the MaEO droplets in water by the dipole-induced-dipole interactions and hydrogen bonds. The factorial design of experiments (DoE) indicates that CNF content and mixing time have significant effects on preventing the MaEO droplets' coalescence during 30-day shelf life. The bacteria inhibition zone assays show that the most stable emulsions showed antimicrobial activity comparable to commercial disinfectant agents such as hypochlorite. We review the recent progress that have led to the development of porous materials based on cellulose nanostructures found in plants and other resources. In light of the properties that emerge from the chemistry, shape and structural control, we discuss some of the most promising uses of a plant-based material, nanocellulose, in regenerative medicine. Following a brief discussion about the fundamental aspects of self-assembly of nanocellulose precursors, we review the key strategies needed for material synthesis and to adjust the architecture of the materials (using three-dimensional printing, freeze-casted porous materials, and electrospinning) according to their uses in tissue engineering, artificial organs, controlled drug delivery and wound healing systems, among others. For this purpose, we map the structure-property-function relationships of nanocellulose-based porous materials and examine the course of actions that are required to translate innovation from the laboratory to industry. Such efforts require attention to regulatory aspects and market pull. Chitosan is a cationic polysaccharide with intrinsic antimicrobial properties that can be used as an ecological alternative to develop functional materials to inhibit the proliferation of microorganisms. This work evaluates chitosan nanocapsules (CNs) as a self-disinfecting agent to provide bactericidal activity on cotton fabrics (CF), using polyacrylate to bind the CNs on the CF surface. The fabrics were characterized by Fourier-transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), contact angle (CA), moisture retention, and antimicrobial tests against Escherichia coli and Bacillus subtilis . The FTIR results showed new peaks related to chitosan structure, indicating the adequate fixation of the CNs on the cotton fibers. SEM images corroborated the polyacrylate binder's efficient adhesion, connecting the CNs and the cotton fiber surface. We postulated that dimethyl fumarate (DMF) exerts neuroprotective effects against depression-like behaviors through astrocytes and microglia modulation. To ascertain our hypothesis and define the mechanistic pathways involved in effect of DMF on neuroinflammation, we used the depression model induced by chronic unpredictable mild stress (CUMS), in which, the mice were exposed to stressful events for 28 days and from the 14th day they received DMF in the doses of 50 and 100 mg/kg or fluoxetine 10 mg/kg or saline. On the 29th day, the animals were subjected to behavioral tests. Microglia (Iba1) and astrocyte (GFAP) marker expressions were evaluated by immunofluorescence analyzes and the cytokines TNF-α and IL-Iβ by immunoenzymatic assay. In addition, computational target prediction, 3D protein structure prediction, and docking calculations were performed with monomethyl fumarate (DMF active metabolite) and the Keap1 and HCAR2 proteins, which suggested that these could be the probable targets related protective effects. CUMS induced anxiety- and depressive-like behaviors, cognitive deficit, decreased GFAP, and increased Iba1, TNF-α, and IL-Iβ expression in the hippocampus. These alterations were reversed by DMF. Thus, it is suggested that one of the mechanisms involved in the antidepressant effect of DMF is neuroinflammatory suppression, through the signaling pathway HCAR2/Nrf2. Alzheimer's disease (AD) is characterized by progressive, irreversible loss of memory and cognitive function. Drosophila melanogaster and other animal models are used to study several diseases, in order to elucidate unknown mechanisms and develop potential therapies. Molecular studies require biological samples and, for neuropathologies such as AD biopsy of the human brain, are invasive and potentially damaging. The solution is to use animal models, such as D. melanogaster, which is a model organism that can replace mammalian organisms in such studies. In this study, we evaluated the climbing ability and differential gene expression during AD progression due to the amylodoigenic pathway using RNA-seq, and we performed an in silico analysis of a fruit fly AD-like GFP (Green Fluorescent Protein) model with GFP expression in the pan-neural elav driver. A total of 1388 genes were differentially expressed in all analyzed groups. The main pathways related to those Differentially Expressed Genes (DEGs) during aging and AD progression were evaluated using the fly genes and human orthologs, in order to link genomic information to higher-order functional information with gene pathway mapping. We identified pathways present in all analyzed groups, such as metabolic pathways, ribosomal pathways, proteasome pathways and immune system pathways. Some of the genes were validated by qPCR. Knockdown of CG17754 gene by RNAi promoted degeneration in the fly eye, validating these findings in vivo. The successful development of multifunctional cotton fabrics with antimicrobial and antiviral activities is essential to prevent the proliferation of microorganisms and transmission of coronavirus virions today, especially with the emergence of new variants of SARS-CoV-2. In this work, we developed antimicrobial cotton fabrics with Ag/TiO 2 nanoparticles synthesized via sonochemistry. Here, we show that more than 50% of infectious SARS-CoV-2 remain active after prolonged direct contact self-disinfecting materials capable of inhibiting the proliferation of Escherichia coli and Staphylococcus aureus . Thermoplastic starch (TPS) films filled with chitosan nanocapsules (CN) containing essential oils (EO) were prepared aiming active packaging. Two different EOs were studied: Ho wood (H) and Cinnamon (C). Besides, different capsules concentrations were investigated (1, 3, and 5 wt%), and the films were evaluated by chemical structure, thermal stability, crystallinity, water vapor permeability, antimicrobial assays, and potential application for strawberry packaging. The TPS/CN-Ho wood films showed a strong interaction between chitosan-starch, mainly for 3 and 5 wt%, confirmed by XRD. The FT-Raman spectra of TPS/CN-Cinnamon film indicated that Cinnamon EO quickly migrated to starch films, probably due to the new crystal structure, named C-type, affecting the film's water permeability. The addition of 1 and 3 wt% CN loaded with Ho wood or Cinnamon EO to the films decreased the water permeability. 3 wt% CN was the optimum concentration to inhibit the Escherichia coli or Bacillus subtillis growth on the films, confirming their biological activity. The films' preservation properties were evaluated using strawberries, and films with 1 or 3 wt% loaded-CN could extend the strawberries' shelf life without fungi contamination. Aims: AMPK plays a critical role regulating cell metabolism, growth and survival. Interfering with this enzyme activity has been extensively studied as putative mechanism for cancer therapy. The present work aims to identify a specific AMPK activator for cancer cells among a series of novel heterocyclic compounds., Materials and Methods: A series of novel hybrid heterocyclic compounds, namely naphtoquinone-4-oxoquinoline and isoquinoline-5,8-quinone-4-oxoquinoline derivatives, were synthesized via Michael reaction and their structures confirmed by spectral data: infrared; 1 H and 13 C NMR spectroscopy (COSY, HSQC, HMBC); and high-resolution mass spectrometry (HRMS). The novel compounds were screened and tested for antitumoral activity and have part of their mechanism of action scrutinized., Key Findings: Here, we identified a selective AMPK activator among the new hybrid heterocyclic compounds. This new compound presents selective cytotoxicity on breast cancer cells but not on non-cancer counterparts. Biopolymers are promising materials for water treatment applications due to their abundance, low cost, expandability, and chemical structure. In this work, gelatin hydrogels filled with cellulose in the form of pristine eucalyptus residues (PER) or treated eucalyptus residues (TER) were prepared for adsorption and chromium removal in contaminated water. PER is a lignocellulosic compound, with cellulose, hemicellulose, and lignin, while TER has cellulose as a major component. FT-Raman Spectroscopy and FTIR analysis confirmed the crosslink reaction with glutaraldehyde and indicated that fillers altered the gelatin molecular vibrations and formed new hydrogen bonds, impacting the hydrogels' crystalline structure. The hydrogen bond energy was altered by the cellulosic fillers' addition and resulted in higher thermal stability (~10 °C). Hydrogels presented a Fickian diffusion, where gelatin hydrogel showed the highest swelling ability (466%), and composites showed lower values with the filler content increase. The chromium adsorption capacity presented values between 12 and 13 mg/g, i.e., featuring an excellent removal capacity which is related with hydrogel crosslinked structure and fibers surface hydroxyl groups, highlighting gelatin hydrogel TER 5% with better removal capacity. This work aimed to prepare surfactant modified-PBAT (poly(butylene adipate-co-terephthalate)) sheets with superior properties to increase the PBAT applicability and be a possible solution for plastic disposal environmental problems. Three different surfactant contents (0, 1, 5, and 10 wt%) were investigated, and their effects on PBAT chemical structure, mechanical and morphological properties, wettability, and water absorption were investigated. Modified-PBAT samples showed high hydrogen bond coefficients (0.57) than the pristine PBAT (0.54), indicating an excellent electrostatic interaction between both components and the formation of a rigid hydrogen-bonded network, as confirmed by mechanical tests, where the elastic modulus values for PBAT and PBAT+10% surfactant were 44 and 60 MPa. SEM images and roughness measurements showed changes in PBAT morphology after surfactant addition, improving the roughness and wettability by the voids and polar groups presence, altering the water absorption (WA) behavior. The higher water affinity resulted in high water absorption for PBAT-10%S (17%) compared to the pristine PBAT (2%), which improves hydrolysis tendency, which is the initial step to biodegradation. Biodegradation results indicated that the roughness and WA behavior influenced the biodegradation rate, facilitating hydrolysis and microbial attack, and accelerating modified samples weight loss. Our results suggested developing a material with superior mechanical properties, mainly for PBAT-10%S, that can be applied in several applications, such as packaging and furniture. In this work, we prepared PU-composites with Australian palm residues (PR) in different contents (5, 10, 15, and 20 wt%) and granulometry (28 and 35 mesh) to improve the oil (crude oil and S500 Diesel) sorption capacity. The foams were characterized by life cycle assessment (LCA), scanning electron microscopy, oil sorption, desorption, and Langmuir, Freundlich, and Temkin sorption isotherms. LCA indicated that higher PR contents decreased the foam environmental impacts than the classical residue handling, indicating that 20 wt% PR is the better environmental option, independent of the residues granulometry. The PR incorporation into PU foams resulted in smaller pore sizes, with a higher number of homogeneous open-cells. The PU composites exhibited higher oil adsorption capacity than the pristine foam. The PU sample showed maximum absorption capability of 6.1 and 6.7 g g -1 for diesel S500 and crude oil, and the composites showed increased values of ∼18 g g -1 and ∼24 g g -1 . The Langmuir model presented the best fit and predicted a maximum adsorption capacity of 30.39 and 25.57 g g -1 for PU-20% PR 28 and 35 mesh, respectively. The composites presented excellent reusability with PU-20% PR (28 mesh) and PU-20% PR (35 mesh), showing removal efficiency after 16 and 9 cycles, respectively. Immune-inflammatory mechanisms are involved in the pathophysiology of bipolar disorder. Tetracyclines present neuroprotective actions based on their anti-inflammatory and microglia suppressant effects. Doxycycline (DOXY) is a tetracycline that demonstrates a better usage profile with protective actions against inflammation and CNS injury. Here, we investigated the effects of DOXY against behavioral, neuroinflammatory, and pro-oxidative changes induced by the d-amphetamine mania model. Adult mice were given d-amphetamine 2.0 mg/kg or saline for 14 days. Between days 8 and 14, received lithium, DOXY (25 or 50 mg/kg), or their combination (lithium+DOXY) on both doses. We collected the brain areas prefrontal cortex (PFC), hippocampus, and amygdala to evaluate inflammatory and oxidative alterations. D-amphetamine induced hyperlocomotion and impairment in recognition and working memory. Lithium reversed hyperlocomotion but could not restore cognitive alterations. DOXY alone (at both doses) or combined with lithium reversed d-amphetamine-induced cognitive changes. DOXY, better than lithium, reversed the d-amphetamine-induced rise in TNFα, MPO, and lipid peroxidation. DOXY reduced the hippocampal expression of Iba1 (a marker of microglial activation), inducible nitric oxide synthase (iNOS), and nitrite. Combined with lithium, DOXY increased the phosphorylated (inactivated) form of GSK3β (Ser9). Therefore, DOXY alone or combined with lithium reversed cognitive impairment and neuroinflammation induced by the mice's d-amphetamine model. This study points to DOXY as a promising adjunctive tool for bipolar disorder treatment focused on cognition and neuroimmune changes. In the present research, soybean straw was used to prepare nanocellulose (NC) via a ball mill, in different milling times (6, 9, and 12 h) and in-situ modified with an anionic surfactant. NCs were characterized for their chemical structure, surface composition, dimension and stability, morphology, crystalline structure, and thermal stability. Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy results indicated a cellulosic structure for NCs and a physical interaction due to the electronic attractions between nanocellulose hydroxyls and surfactant end chain groups. The dynamic light scattering, Zeta potential, and transmission electron microscopy indicated that the in situ modified samples showed smaller sizes and good electrostatic stability. Besides, while ball mill resulted in nanofibers, the in situ modified-NC showed a nanocrystal shape, indicating that the surfactant alters the milling process and cellulose scale reduction. The modified-NC showed lower crystallinity and crystal size than unmodified nanocelluloses due to the surfactant chains' addition and influence during the milling process. The modified-NC showed slightly superior thermal stability. The NC-12S showed smaller particle sizes, high electrostatic, and thermal stability and indicated that 12 h is adequate to prepare modified nanocellulose via in situ modification. Cell-free DNA is present in different biological fluids and when released by tumor cells may contribute to pro-tumor events such as malignant transformation of cells adjacent to the tumor and metastasis. Thus, this study analyzed the effect of tumor cell-free DNA, isolated from the blood of prostate cancer patients, on non-tumor prostate cell lines (RWPE-1 and PNT-2). To achieve this, we performed cell-free DNA quantification and characterization assays, evaluation of gene and miRNA expression profiling focused on cancer progression and EMT, and metabolomics by mass spectrometry and cellular migration. The results showed that tumor-free cell DNA was able to alter the gene expression of MMP9 and CD44, alter the expression profile of nine miRNAs, and increased the tryptophan consumption and cell migration rates in non-tumor cells. Therefore, tumor cell-free DNA was capable of altering the receptor cell phenotype, triggering events related to malignant transformation in these cells, and can thus be considered a potential target for cancer diagnosis and therapy.

This work evaluated the synergic effect of carvacrol essential oil (EO) and montmorillonite (MMT) (named hybrid compound), incorporated in thermoplastic starch (TPS) films in different contents (4.5, 9, and 15 wt%). TPS films were characterized by FTIR, FT-Raman, DSC, XRD, and antimicrobial assay. FTIR and FT-Raman indicated new hydrogen bonds between hybrid and starch, altering the thermal properties and requiring more energy to destroy the TPS-hybrid bonds. XRD analysis showed slight crystallinity changes, and TPS-15 hybrid showed an amorphous structure that allows the diffusion of EO thought the films. TPS-hybrid films showed antimicrobial behavior against E. coli due to the synergistic effect of MMT and EO, which resulted in strong antimicrobial effects due to the destabilization and partial destruction of the bacteria cell membrane. The hybrid probably changed the E. coli cell permeability, respiration, and electron transport. TPS-15 hybrid sample presented the lowest crystallinity, facilitating the film's oil diffusion, maximizing the bacterium inhibitory effect, and acting as a biocidal agent. Xylella fastidiosa colonizes the xylem of various cultivated and native plants worldwide. Citrus production in Brazil has been seriously affected, and major commercial varieties remain susceptible to Citrus Variegated Chlorosis (CVC). Collective cellular behaviors such as biofilm formation influence virulence and insect transmission of X. fastidiosa . The reference strain 9a5c produces a robust biofilm compared to Fb7 that remains mostly planktonic, and both were isolated from symptomatic citrus trees. This work deepens our understanding of these distinct behaviors at the molecular level, by comparing the cellular and secreted proteomes of these two CVC strains. Out of 1017 identified proteins, 128 showed differential abundance between the two strains. Different protein families were represented such as proteases, hemolysin-like proteins, and lipase/esterases, among others. Here we show that the lipase/esterase LesA is among the most abundant secreted proteins of CVC strains as well, and demonstrate its functionality by complementary activity assays. More severe symptoms were observed in Nicotiana tabacum inoculated with strain Fb7 compared to 9a5c. Our results support that systemic symptom development can be accelerated by strains that invest less in biofilm formation and more in plant colonization. This has potential application in modulating the bacterial-plant interaction and reducing disease severity.

Metabolic and psychiatric disorders present a bidirectional relationship. GLP-1 system, known for its insulinotropic effects, has also been associated with numerous regulatory effects in cognitive and emotional processing. GLP-1 receptors (GLP-1R) agonists present neuroprotective and antidepressant/anxiolytic properties. However, the effects of GLP-1R agonism in bipolar disorder (BD) mania and the related cognitive disturbances remains unknown. Here, we investigated the effects of the GLP-1R agonist liraglutide (LIRA) at monotherapy or combined with lithium (Li) against D-amphetamine (AMPH)-induced mania-like symptoms, brain oxidative and BDNF alterations in mice. Swiss mice received AMPH 2 mg/kg or saline for 14 days. Between days 8-14, they received LIRA 120 or 240 μg/kg, Li 47.5 mg/kg or the combination Li + LIRA, on both doses. After behavioral evaluation the brain areas prefrontal cortex (PFC), hippocampus and amygdala were collected. AMPH induced hyperlocomotion, risk-taking behavior and multiple cognitive deficits which resemble mania. LIRA reversed AMPH-induced hyperlocomotion, working and recognition memory impairments, while Li + LIRA240 rescued all behavioral changes induced by AMPH. LIRA reversed AMPH-induced hippocampal oxidative and neurotrophic changes. Li + LIRA240 augmented Li antioxidant effects and greatly reversed AMPH-induced BDNF changes in PFC and hippocampus. LIRA rescued the weight gain induced by Li in the course of mania model. Therefore, LIRA can reverse some mania-like behavioral alterations and combined with Li augmented the mood stabilizing and neuroprotective properties of Li. The aim of this work was to synthesize pinus residue/pectin-based composite hydrogels for the immobilization of β-D-galactosidase. These hydrogels were synthesized via chemical crosslinking, and characterized by Fourier-transform infrared spectroscopy, scanning electron microscopy, thermal analysis, mechanical assays, X-ray diffraction, and swelling kinetics. The water absorption mechanism in the hydrogel networks occurs by non-Fickian transport. The β-D-galactosidase immobilization capacities of the hydrogels containing 0, 5 and 10% of pinus residue were respectively 242.08 ± 0.36, 181.27 ± 0.50 and 182.71 ± 0.36 mg enzyme per g dried hydrogel, at pH 4.0 and after 600 min. These values were 182.99 ± 0.41, 219.99 ± 0.47 and 218.56 ± 0.39 mg g -1 , respectively, at pH 5.6. The RNA aptamer A4 binds specifically to tumor prostate cells. A4 was modified (mA4) by adding deoxyribonucleotides to its ends to remove the reactive 2' hydroxyl groups of RNA's sugar at the ends of the aptamer and to make it more stable to widespread RNase contamination in laboratories. Thus, mA4 would be more suitable to use in the clinical settings of prostate cancer (PCa). We aimed to characterize this optimized oligonucleotide to verify its potential as a diagnostic tool. The sequences and structures of A4 and mA4 were compared through in silico approaches to corroborate their similarity. Then, the degradation of mA4 was measured in appropriate media and human plasma for in vitro tests. In addition, the binding abilities of A4 to prostate cells were contrasted with those of mA4. The effects of mA4 were assessed on the viability, proliferation, and migration of human prostate cell lines RWPE-1 and PC-3 in three-dimensional (3D) cell cultures. mA4 showed configurational motifs similar to those of A4, displayed a half-life in plasma substantially higher than A4, and exhibited a comparable binding capacity to that of A4 and unaltered viability, proliferation, and migration of prostatic cells. Therefore, mA4 maintains the crucial 3D structures of A4 that would allow binding to its target, as suggested by in silico and binding analyses. mA4 may be a good PCa reporter as it does not change cellular parameters of prostate cells when incubated with it. Its additional deoxyribonucleotides make mA4 inherently more chemically stable than A4, avoiding its degradation and favoring its storage and handling for clinical applications. Anaplasmosis is one of the most prevalent tick-borne diseases of cattle caused by Anaplasma marginale. MSP1a surface protein has been shown to be involved in eliciting immunity to infected cattle. Carbon nanotubes (CNTs) has been increasingly highlighted due to their needle like structure, which contain multiple attachment sites for biomolecules and may interact with or cross biological membranes, increasing antigen availability to immune system. Here, we have successfully designed a nanocomplex of a synthetic peptide noncovalently attached to multiwalled CNT (MWCNT). Peptide comprising the core motif of the MSP1a was efficiently adsorb onto the nanoparticle surface. The MWCNT-Am1 nanocomplex exhibited high stability and good dispersibility and in vivo immunization showed high levels of IgG1 and IgG2a, followed by increased expression of pro-inflammatory and anti-inflammatory cytokines. Electronic health records (EHRs), originally designed to facilitate health care delivery, are becoming a valuable data source for health research. EHR systems have two components, both of which have various components, and points of data entry, management, and analysis. The "front end" refers to where the data are entered, primarily by healthcare workers (e.g. physicians and nurses). The second component of EHR systems is the electronic data warehouse, or "back-end," where the data are stored in a relational database. EHR data elements can be of many types, which can be categorized as structured, unstructured free-text, and imaging data. The Sunrise Clinical Manager (SCM) EHR is one example of an inpatient EHR system, which covers the city of Calgary (Alberta, Canada). This system, under the management of Alberta Health Services, is now being explored for research use. The purpose of the present paper is to describe the SCM EHR for research purposes, showing how this generalizes to EHRs in general. Arabic gum-based composite hydrogels reinforced with eucalyptus and pinus residues were synthesized via free-radical reaction aiming to controlled phosphorus release. All hydrogels were characterized by swelling kinetics (SK), Fourier-transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM), X-Ray diffraction (XRD), thermogravimetric analysis (TGA), differential scanning calorimetry (DSC), and mechanical assays (MA). The water and solute transports through the hydrophilic three-dimensional networks of the hydrogels occur preferably by diffusion processes and macromolecular relaxation. Hemicellulose, lignin and cellulose fibers contained in eucalyptus and pinus residues affected the crosslinking density, crystalline structure, and water/solute diffusion due to reduction of free hydroxyl and amine groups in the hydrogel networks. Hence, the eucalyptus and pinus residues improved the mechanical and thermal resistances of the composite hydrogels. Finally, the Arabic gum-based hydrogel and Arabic gum-based composite hydrogels reinforced with eucalyptus and pinus residues demonstrated to be excellent alternatives for the controlled phosphorus release in agricultural nutrient-poor soils., (Copyright © 2019 Elsevier B.V.
