Background and Aim of Study: There are two different phases of untreated chronic myeloid leukemia – chronic phase, and blast crisis – according to the World Health Organization classification of Hematolymphoid tumors. The blast cells in the chronic myeloid leukemia blast crisis can express myeloid, lymphoid, bi-phenotypic, monocytic, megakaryocytic, and erythroid phenotypes. The immunophenotype of blast population determines how chronic myeloid leukemia – blast crisis patients respond to treatment, hence flowcytometric examination is required. The aim of the study: to assess immunophenotyping outcomes of flowcytometry performed on the chronic myeloid leukemia – blast crisis. Material and Methods: A five-year retrospective descriptive analysis was carried out in Pathology Department at King George’s Medical University Lucknow, India (2017-2021). The patient’s peripheral blood and bone marrow aspirate samples were analyzed. Clinical, hematological, and immunophenotypic data were retrieved. The flow cytometry samples were prepared using the standardized “lyse-stain-wash” method. Results: A total of 43 cases of chronic myeloid leukemia – blast crisis were retrieved from the departmental archive in 5 years. The mean age of study population was 39.62±14.86 years. There were 24 males and 19 females. 27 patients were diagnosed with myeloid blast crisis, 14 cases of B-lymphoid blast crisis and 2 cases of mixed phenotypic acute leukemia. Conclusions: Identification of the blast lineage of patients with chronic myeloid leukemia – blast crisis is crucial since the existence of atypical blast phenotypes influences the disease treatment and prognosis.

Background: Breast cancer is the most common cancer in women worldwide and in India. There are very few cases of breast cancer with peripheral blood dissemination having acute leukemia-like blast cells. Case presentation: Here, we present a 44-year-old breast cancer follow-up patient presenting with leukemia-like blast cells in peripheral blood. The specific cytochemical staining and immunophenotyping assays on bone marrow biopsy revealed that atypical cells in her peripheral blood were from breast cancer metastasis. A comprehensive workup confirmed carcinocythemia in the present case and systemic chemotherapy and hormonal therapy were planned. However, she died 15 days after the carcinocythemia presentation. Carcinocythemia patients have a poor survival. Conclusion: Because carcinocythemia cells can mimic leukemic blasts and share morphological features, the patient's medical history and a careful analysis of the feathery border of the smear are needed to make a definitive diagnosis. Background: Paediatric rotary file systems have recently been developed for primary teeth use. Aim: To study the cleaning efficacies of two paediatric rotary endodontic files, the Prime PedoTM, and the Kedo‐SG BlueTM against the standard H files. Design: This in vitro study included 54 freshly extracted primary molars, which were randomised into three groups (n = 18 each) and were prepared using either Kedo‐SG BlueTM, Prime PedoTM or hand H files after injecting methylene blue dye into the canals. Pre‐ and post‐operative cone beam computerised tomography (CBCT) was performed to assess change in root canal volumes. Methylene blue dye removal from canals was assessed using stereomicroscopy, and canal cleanliness was examined by scanning electron microscopy (SEM). Results: Both Prime PedoTM and Kedo‐SG BlueTM files reduced significantly less dentine when compared with conventional hand filing with Prime PedoTM removing the least amount of dentine. No significant difference was found in median SEM scores among the groups in the cervical, middle and apical thirds of the roots. Stereomicroscopic assessment of root canal cleanliness using dye removal technique shows a statistically significant difference existing between Kedo‐SG BlueTM and hand H files groups. Conclusion: Prime PedoTM removed the least amount of dentine. Kedo‐SG BlueTM performed significantly better than conventional hand filing with H files when the root canal cleanliness was assessed.
