The objective of this study was to assess whether the package inserts of industrialized phytomedicines are in line with current Brazilian legislation or not. Forty-one package inserts of phytomedicines containing artichoke, horse chestnut, ginkgo and senna were accessed in the respective proprietary websites and then analyzed. The analysis aimed to verify the presence of items required by the normative RDC 47/2009 and the usage indication, dosage and side effects compared to standard package inserts and to the scientific literature. Quantitative analysis proved that the package inserts are in accordance with the current legislation. However, in qualitative analysis, all phytomedicines analyzed in this study had at least one error related to dosage, posology or adverse reactions. A gradual improvement of the package inserts of phytomedicines has been noticed but, even with the availability of standard package inserts, some mistakes persist, which can induce the patient to incorrect use. O objetivo deste trabalho foi avaliar se as bulas de medicamentos fitoterápicos industrializados estão de acordo com a legislação nacional vigente. Foram analisadas 41 bulas dos medicamentos fitoterápicos compostos por alcachofra, castanha da Índia, ginkgo e sene, adquiridas nos sites das empresas titulares dos registros. A análise foi feita quanto à presença de itens segundo a RDC nº 47 de 2009 e ao conteúdo de indicação de uso, posologia e reações adversas comparada às bulas padrão e à literatura científica. A análise quantitativa provou que as bulas têm cumprido o que é exigido pela legislação vigente. No entanto, quanto à análise qualitativa, todos os fitoterápicos analisados neste trabalho possuíam pelo menos uma bula com erro relacionado à posologia, indicações de uso e reações adversas. Houve uma melhora gradativa das bulas dos medicamentos fitoterápicos, porém mesmo com as bulas padrão ainda existem erros, que podem induzir o paciente ao uso incorreto do medicamento.

Mandevilla guanabarica is an endemic plant from Brazil, with pharmacological and ornamental potential, both unexplored. This study established the best culture medium for in vitro plant maintenance, efficient protocol for its regeneration, and callogenesis from different explants excised from in vitro-grown plants. Woody plant medium with double boron concentration (WPM B) plus 2.27 µM thidiazuron or 0.49 µM 2-isopentenyladenine provided multiplication rates higher than 1:6. Shoots were 100% rooted on WPM B. After acclimatization, plants showed 83% survival. For callogenesis, the use of MS media supplemented with high concentrations of picloram or 2,4-dichlorophenoxyacetic acid produced, respectively, friable or compact non-morphogenic calluses from different types of explants. This micropropagation protocol allows the production of seedlings of M. guanabarica for ornamental or commercial uses, and for conservation purposes; calluses can be used to establish suspension cultures in prospecting for bioactive compounds. Mandevilla guanabarica é uma planta endêmica do Brasil, com potencial farmacológico e ornamental inexplorados. Este estudo estabeleceu um eficiente meio de cultura para manutenção da planta in vitro e eficazes protocolos de regeneração e calogênese a partir de diferentes explantes. O meio para plantas lenhosas suplementado com o dobro da concentração de boro (WPM B) e com 2,27 µM de thidiazuron ou 0,49 µM 2-isopenteniladenina proporcionou taxas de multiplicação maiores que 1:6. Os brotos foram 100% enraizados em WPM B, com 83% de sobrevivência após aclimatização. Na calogênese, o meio MS suplementado com altas concentrações de picloram ou ácido 2,4-diclorofenoxiacético produziu, respectivamente, calos não morfogênicos friáveis ou compactos. Este protocolo de micropropagação permite a produção de mudas de M. guanabarica para uso ornamental, comercial e com finalidades de conservação; os calos podem ser utilizados para o estabelecimento de culturas em suspensão na prospecção de compostos bioativos.

The bioassay-guided fractionation of the hexane extract obtained from the medicinal plant Myroxylon balsamum (red oil) was conducted in preparative thin layer chromatography on silica gel. The obtained fractions and some terpenoids and phenylpropanoids were assayed as larvicidal on third instar Aedes aegypti larvae, NPPN colony. The results indicate that the sesquiterpene nerolidol was the active constituent in the extract and that the sesquiterpenes were more active than the monoterpenes and phenylpropanoids utilized in this study. Lipophilicity seems to be an important property for the activity since the compounds with hydroxyl, carbonyl and methoxyl groups were less active. The results confirm also that essential oils can be a good tool for the control of dengue.

We compared the efficiency of three different methodologies (conventional Maceration [CM], ultrasound [UAE] and microwave [MAE] assisted extractions) concerning total phenolics (TP) and resveratrol (t-RSV) content, followed by antioxidant activity (AA) evaluation. By CM, at 1 h, it was found the highest RSV content (1.024 ± 0.036 mg / L) and, correspondly, the highest DPPH capture (23.90 ± 0.04%). The TP contents, at 1h, presented the highest value (27.26 ± 0.26 mg/g GAE). By the UAE, the maximum yield of TP (357.18 mg/g GAE) and RSV (2.14 mg/L), as well as, the highest AA (70.95%), were obtained by 5 min after a maceration pre-treatment, on the solid-solvent ratio 1:40 w/v. For MAE, a central composite rotatable design (CCRD) was applied followed by the FFD design in order to evaluate the statistical effects of four independent variables on the extraction of RSV. The optimal conditions established for obtaining the highest recovery (2.516 mg/g) were: 20 min; 90% MeOH aq.; 120 rpm; 60°C and, solid-solvent ratio: 1:35 w/v. Relevant correlations were established considering the TP and RSV contents, as well as, the AA, corroborating obvious advantages of such techniques in terms of high extraction efficiency in shorter times. The identification of the constituents by GC / MS revealed the presence of novel allelochemical substances in the species. Substances isolated in mixture or do not indicate the presence of steroids such as sitosterol, stigmasterol and lupeol in the hexanic extract of A. repens stems. These substances are attributed to the allelopathic activity known in the literature. The dichloromethane partition of the methanolic extract of stems and leaves showed an inhibitory allelopathic effect for both hypocotyls as in L. sativa radicles and in the weeds Commelina benghalenses and Panicum maximum Arachis repens, vulgarmente conhecido como grama-amendoim, e pela primeira vez detectamos a presença do composto bioativo trans-resveratrol (t-RSV). Comparamos a eficiência de três metodologias de extração diferentes: Maceração (MC), ultra-som (EAU) e micro-ondas em relação ao teor de fenóis totais (FT) e resveratrol, posteriormente avaliou-se a atividade antioxidante. A MC pelo período de 1 h possibilitou a maior recuperação de RSV (1,024 ± 0,036 mg / L) e, correspondentemente, a maior captura de DPPH (23,90 ± 0,04%) e o conteúdo de FT com (27,26 ± 0,26 mg / g de EAG). Na EAU, o rendimento máximo de FT (357,18 mg / g de GAE) e RSV (2,14 mg / L), bem como o AA mais efetiva (70,95%), foram obtidos com 5 min de exposição após um período de 5 min. de pré-tratamento por maceração, na proporção sólido-solvente 1:40 p / v. Para o EAM, aplicou-se um delineamento do composto central rotacional (DCCR) para avaliar os efeitos estatísticos de quatro variáveis independentes na extração de RSV. As condições ideais estabelecidas para obter a maior recuperação de RSV (2,516 mg / g) foram: 20 min; 90% MeOH aq.; 120 rpm; 60 ° C e relação sólido-solvente: 1:35 p / v. Correlações relevantes foram estabelecidas considerando os teores de FT, RSV e AA, evidenciando as vantagens destas técnicas em termos de eficiência de extração. A identificação dos constituintes por GC / MS revelou a presença de novas substâncias aleloquímicas na espécie. Substâncias isoladas na mistura ou não no extrato hexânico de caules de A. repens indicam a presença de esteróides como o sitosterol, estigmasterol e lupeol. A estas substancias atribui-se a atividade alelopática conhecida na literatura. A partição de diclorometano do extrato metanólico de caules e folhas mostrou um efeito alelopático inibitório tanto para hipocótilo como em radículas L. sativa e nas ervas daninhas Commelina benghalenses e Panicum máximo

The AEEm was obtained by infusion in distilled water (50 g d.w./L, 100°C), followed by filtration and lyophilization. The AEEm was fractionated on Sephadex LH 20, generating Fr20 and Fr40 fractions. The chromatographic profiles of fractions were analyzed by HPLC-DAD in a reverse phase C18 column. The chromatographic profile of HEEm was analyzed by GC-MS using a DB 5 (30 m x 0.25 mm x 0.25 µm) column. We used Swiss Webster mice (2- 4 months of age (n= 5/group)) for biological tests (CEUA 05/2009 e 07/2013). The antinociceptive effects of HEEm, AEEm, Fr20 and Fr40 were evaluated using hyperalgesia models induced by intraperitoneal (i.p.) injection of acetic acid, formalin test and tail immersion in a bath at 50°C. Treatment (p.o.) with AEEm and HEEm showed higher inhibition of the contortions number of 75% and 52% respectively at a dose of 25 mg/kg. The Fr20 (5 mg/kg) inhibited 39% of the contortions, and Fr40 treatment (25 mg/kg) showed 96% inhibition when compared to control. Treatment with dipyrone inhibited contortions induced by acetic acid by 51% on average in relation to the control group. In the tail immersion model in a bath at 50°C, treatment with AEEm showed no antinociceptive activity. In this model, the EHEm showed antinociceptive activity by treatment with 50 mg/kg, at 60min and 90 min and the Fr20 increased the latency time, 90 min after treatment with the three doses (25, 50 and 100 mgkg), when compared with the control. Morphine, used as standard drug showed antinociceptive effect after treatment (i.p.) in all time analyzed (30-180 min). In the MTT assay with NEURO-2A cells, AEEm exhibited the higher inhibition of the mitochondrial reducing activity (MRA) in a concentration dependent manner (IC50 of 36.15 mg/mL). HEEm showed MRA reduction reaching 49% at 100 mg/mL concentration and morphine exhibited maximum reduction of 23%. Fr20 fraction showed inhibition of 38.1% of MRA, and FR40 did not interfere with MRA in most concentrations tested. The precise mechanisms involved in the antinociceptive response of the Fr40 fraction from E. macrophyllus are not completely understood, but the presence of flavonoids and other aromatic compounds, suggests the possible contribution of NO adrenergic activation. By the analysis of the Fr40 compounds by HPLC-DAD and standards co-injection, and TLC evaluation isovitexin and isoorientin were identified in this fraction. Neste estudo avaliou-se o potencial antinociceptivo do extrato hexânico (EHEm), do extrato aquoso (EAEm) e frações (Fr) de folhas de Echinodorus macrophyllus (Kunth) Mich., conhecido no Brasil como chapéu de couro , em diferentes respostas nociceptivas geradas por um produto químico ou por estímulo nocivo térmico e realizar a análise fitoquímica. O EAEm foi obtido por infusão em água destilada (50 g p.s./L, 100°C), seguido de filtração e liofilização. O EAEm foi fracionado em Sephadex LH-20, gerando as frações Fr20 e Fr40. Os perfis cromatográficos das frações foram analisados por HPLC-DAD em coluna de fase reversa C18. O perfil cromatográfico do EHEm foi analisado por GC-MS utilizando-se coluna DB 5 (30 m x 0,25 mm x 0,25 μm). Utilizou-se camundongos Swiss Webster machos (2 a 4 meses de idade (n=5/grupo)) para os ensaios biológicos (CEUA 05/2009 e 07/2013). O efeito antinociceptivo do EHEm, EAEm, Fr20 e Fr40 foi verificado utilizando-se os modelos de hiperalgesia induzida por injeção intraperitoneal (i.p.) de ácido acético, formalina e de imersão da cauda em banho a 50ºC. O tratamento (v.o.) com o EAEm e EHEm apresentou maior inibição no número de contorções de 75% e 52%, respectivamente na dose de 25 mg/kg. A Fr20 (5 mg/kg) inibiu 39% das contorções e o tratamento com a Fr40 (25 mg/kg) apresentou uma inibição de 96%, quando comparados ao controle. O tratamento com dipirona inibiu em média 51% as contorções induzidas por ácido acético em relação ao grupo controle. No modelo de imersão da cauda em banho a 50ºC, o tratamento com EAEm não apresentou atividade antinociceptiva. Neste modelo, o EHEm mostrou atividade antinociceptiva no tratamento com a dose de 50 mg/kg nos tempos 60 e 90 min e a Fr20 aumentou o tempo de latência 90 min após tratamento com as três doses administradas (25, 50 e 100 mg/kg), quando comparados com o controle. A morfina utilizada como droga padrão, apresentou efeito antinociceptivo após o tratamento (i.p.) em todos os tempos analisados (30 a 180 min). No ensaio do MTT utilizando células NEURO-2A, o EAEm apresentou a maior inibição da atividade redutora mitocondrial (ARM) em função da concentração (IC50 de 36,15 µg/mL). O EHEm apresentou uma redução da ARM atingindo 49% na concentração de 100 mg/mL e a morfina apresentou redução máxima de 23% da ARM. A fração Fr20 apresentou inibição de 38,1% da ARM e a Fr40 não interferiu na ARM da maioria das concentrações testadas. Os mecanismos precisos envolvidos na resposta antinociceptiva da fração Fr40 de E. macrophyllus não estão completamente compreendidos, mas pela presença de flavonoides e outros compostos aromáticos, sugere-se a possível contribuição do NO e ativação adrenérgica. A análise dos compostos da Fr40 por HPLC-DAD e co-injeção de padrões, além de TLC identificou isovitexina e isoorientina nesta fração.

CAPES - Coordenação de Aperfeiçoamento de Pessoal de Nível Superior CNPq - Conselho Nacional de Desenvolvimento Científico e Tecnológico As espécies do gênero Andira (Fabaceae), popularmente conhecidas como “angelins”, destacam-se pelo uso popular como febrífugas, vermífugas e antimaláricas. Apesar das evidências da atividade antimalárica das espécies do gênero, existe uma carência de estudos químicos e biológicos em torno da espécie Andira nitida. Desta forma, este trabalho objetivou estabelecer o perfil químico e avaliar o potencial biológico de A. nitida, a fim de encontrar uma possível opção de novos alvos terapêuticos para o tratamento da malária. O perfil químico foi estabelecido por cromatografia em camada delgada (CCD) e cromatografia em fase líquida de alta eficiência (CLAE), após a análise dos cromatogramas constatou-se a presença de derivados fenólicos, especialmente, flavonoides como as isoflavonas e de derivados de catequina. O conteúdo de derivados fenólicos totais variou de 100-320 mg de equivalente em catequina/g amostra e de 90-280 mg de equivalente em ácido gálico/ g amostra, já o de isoflavonoides de 6,24-11,15 mg de equivalente em biochanina A/g amostra e o de flavonoides totais de 1,20-4,55 mg de equivalente em quercetina/g amostra. As CE50 para o método DPPH variaram de 5,99-31,90 μg/mL. Os valores da atividade antioxidante equivalente ao Trolox (TEAC) para as amostras foram de 0,161-0,693. Foi encontrada forte correlação entre atividade antioxidante por DPPH e o conteúdo de derivados fenólicos (Pearson > 0,9) e entre TEAC e o teor de flavonoides expressos em quercetina (Pearson > 0,83). A fração em hexano e em clorofórmio apresentaram CE50 de 40,5 μg/mL e 136,2 μg/mL, respectivamente, no ensaio de quantificação de substâncias reativas ao ácido tiobarbitúrico (SRAT). As amostras mais ativas no ensaio antiplasmodial (Teste LDH) foram as frações em hexano e em clorofórmio com 31% de inibição do crescimento de P. falciparum. Já no ensaio de inibição da polimerização da ferriprotoporfirina (FBIT) as CE50 variaram de 0,41-1,12 mg/mL sendo as frações em hexano, clorofórmio e em acetato de etila ativas, juntamente com o extrato lipofílico. In vivo, estas amostras a 200 mg/kg/dia foram ativas no teste de Peters com inibições da parasitemia de 39-75% no sétimo dia após a infeccção. Os resultados indicaram que A. nitida apresenta perfil químico predominante em isoflavonas, corroborado pela quantificação de isoflavonoides, dessa forma ratifica-se o papel das isoflavonas como possíveis marcadores quimiossistemáticos do gênero Andira, além de responsáveis, em parte, pela atividade antimalárica das espécies estudadas até o presente momento. Além disso, as amostras provenientes dos galhos de A. nitida podem ser fontes de substâncias bioativas com pronunciada atividade antioxidante, além de interessante atividade antimalárica. The Andira species (Fabaceae), popularly known as "Angelins" stand out for popular use as febrifugue, vermifuge and antimalarial. Chemically they are characterized by the presence of phenolic derivatives such as isoflavones. However, despite evidence of antimalarial activity of the genus, there is a lack about chemical and biological studies of Andira nitida. So the present study aimed to establish the chemical profile and assess the biological potential of A. nitida, to find a possible option for new therapeutic targets for the malaria treatment. The chemical profile was established by thin layer chromatography (TLC) and high performance liquid chromatography (HPLC), after analysis of the chromatograms; they showed the predominant presence of phenolic derivatives, especially flavonoids such as isoflavones and catechin derivatives. The contents of phenolic derivatives ranged from 100 to 320 mg catechin equivalent/g sample and from 90 to 280 mg gallic acid equivalent/g sample. The isoflavonoids content ranged from 6.24 to 11.15 mg equivalent biochanin A/g sample and flavonoids content ranged from 1.20 to 4.55 mg quercetin equivalent/g sample. The EC50 for DPPH ranged from 5.99-31.90 μg/mL.Trolox equivalent antioxidant activity (TEAC) values for the samples were 0.161 to 0.693. Strong correlation was found between oxidant activity by DPPH and content of phenolic derivatives (Pearson > 0.9) and between TEAC and the flavonoids content expressed in quercetin (Pearson > 0.83). The fraction in hexane and in chloroform showed EC50 of 40.5 μg/mL and 136.2 μg/mL, respectively; in the quantification of thiobarbituric acid reactive substances assay (TBARS). The more active samples in antiplasmodial assay (LDH testing) were the fractions in hexane and in chloroform with 31% inhibition of P. falciparum growth. In the ferriprotoporphyrin polymerization inhibition assay (FBIT) the EC50 ranging from 0.41 to 1.12 mg/ml and the fractions in hexane, in chloroform and in ethyl acetate were actives along with the lipophilic extract. In vivo, these samples at 200 mg/kg/day (Peters testing) showed parasitaemia inhibitions of 39-75 % in day 7 after infection. The results indicated that A. nitida presents chemical profile composed of isoflavones, corroborated by quantification of isoflavones, and thus confirms the role of isoflavones as potential chemosystematic markers of Andira genus. Futhermore, the samples from A. nitida can be a source of bioactive substances with pronounced antioxidant activity, as well as interesting antimalarial activity thus can be an alternative for future therapeutic applications.
