- Abstract
The objective of this study was to characterize surveyed coaches and elucidate the practices of physical fitness assessment and monitoring for both male and female athletes across three countries. A total of 165 coaches participated by completing a comprehensive 32-question survey. Pre-season assessments are a priority for coaches, with a significant range from 60.5% to 87.7% in Romania, while Portuguese and Spanish coaches tend to prefer testing during the competition (26.3% and 16.9%, respectively). Portuguese and Spanish coaches predominantly favor aerobic tests (50% and 46.8% respectively), whereas Romanian coaches exhibit a preference for sprint (56.9%) and skill tests (52.3%). Notably, change of direction tests are less commonly employed, ranging from 10.5% to 21% across the countries. In terms of exercise intensity determination, Portuguese coaches predominantly employ maximal heart rate (31.6%), while Spanish coaches often rely on the 220-age formula or perceived exertion (27.4%). For strength assessment, Portuguese coaches lean towards direct (34.2%) or estimated (31.6%) maximal repetition methods. When it comes to maximal speed sprint, Portuguese and Romanian coaches show preference (50% and 43.1% respectively), while Spanish coaches exhibit a relative lack of emphasis on individualized speed measures (37.1%). Perceptual scales are the preferred method for recovery monitoring, with adoption rates of 57.9% in Portugal, 53.2% in Spain, and 44.6% in Romania. In summary, this study underscores the distinct assessment and monitoring practices employed by coaches in Portugal, Spain, and Romania. These findings are in alignment with established literature standards, highlighting the diversity of approaches used in different countries. [ABSTRACT FROM AUTHOR]

- Abstract
This study examines the relationships between body composition, anthropometry and physical fitness measures, and internal and external load (match physical and running demands) during a congested period of an overseas futsal training camp. Eleven under-20 national futsal players participated in a eleven-day training camp. During the matches, exercise heart rate (HR) and locomotion profiles were recorded via a Polar Team Pro system as the players' internal and external load. The friendly matches were scheduled on the training camp's 2nd, 4th, 6th, 8th, and 10th days. A repeated-measures analysis of variance (ANOVA) and Pearson's correlation coefficient were used for statistical analysis. The results revealed significant differences in sprint ( F = 2.74, p = 0.04, η² = 0.21), accelerations ( F = 3.63, p = 0.04, η² = 0.27), and decelerations ( F = 2.73, p = 0.04, η² = 0.21) performance between the five matches ( p < 0.05). Also, the overall body composition and physical fitness measures had large to very large correlations with match running demands during congested periods of futsal competition ( p < 0.05). In conclusion, the baseline body composition and the overall physical fitness measures may be essential factors in match running demands during a congested futsal match period. Futsal players who perform better in the 30--15 Intermittent Fitness Test can sustain a greater volume of highintensity accelerations during a congested period. [ABSTRACT FROM AUTHOR]

Objective: To evaluate the effects of a 4-week intervention combining smallsided games (SSGs) and high-intensity interval training (HIIT) on physical fitness in collegiate male soccer players. Methods: Twenty-one soccer players were randomly assigned to either the HIIT + SSGs group (n = 11) or a control group (n = 10). Physical fitness was assessed at baseline and 1-week post-intervention, including countermovement jump (CMJ), change of direction (COD) test, sprint test, repeated sprint ability (RSA) test, and 30-15 Intermittent Fitness Test (30-15IFT). The intervention comprised eight sessions over 4 weeks: four SSGs and four HIIT. Results: The intervention group showed small to moderate improvements: mean RSA improved by 4.5% (p = 0.07), CMJ increased by 3.2% (p = 0.12), and 30-15IFT scores enhanced by 6.8% (p = 0.09). Key predictors of group membership included heart rate load per minute (OR 1.602) and various GPS variables. Conclusion: The 4-week intervention combining SSGs with HIIT did not produce statistically significant improvements in most physical fitness variables compared to the control group. Although there were positive trends in variables such as RSA and 30-15IFT, these changes were modest and not statistically significant. The results suggest that while the combined SSGs and HIIT approach shows potential, its impact on physical fitness over a 4-week period is limited, with some variables, like CMJ, even showing decreases. Featured Application: Between the ages of 13–15 it is important to modulate the training load bearing in mind that physical improvements can vary significantly between individuals, depending on factors such as genetics, diet, sleep and the specific type and intensity of training. Monitoring of these variables must be done with care, keeping the health of the players as a top priority. In addition, coaches and trainers should encourage a balanced view of these metrics, encouraging players to focus on skill development and enjoyment of the sport rather than solely on physical changes. The purpose of this study was to compare the change of Body Composition (BC) (height, weight, body fat percentage and muscle mass) as a function of the trimester and category in a sample of young soccer players. Data collection was performed in five consecutive seasons (2016–2021). The sample consisted of 741 young male football players of different categories (Under 14 year old (U14), U15, U16, U17 and U18) belonging to a high-performance football academy. Considering the trimestral change of all the raw anthropometrics variables a set of new variables called the trimestral change in percentage (TC) of each raw variable was computed. Two-way repeated measures ANOVA (including the raw anthropometric variables as dependent and trimester and the age-category as independent) revealed differences for the anthropometric variables (p value < 0.001 in all cases), concluding that the effect of trimester reaches conventional levels of statistical significance. The trimester by age in contrast was significant (p < 0.05) in all raw variables except for the height. Considering the TC variables, the variable height-TC showed an increase (p value < 0.05) while the variable muscle mass-TC was near the significative value (p = 0.09). In this case the interaction trimester by age category was not significative (p > 0.05 in all cases). It seems that height suffers more changes in the first trimester but the weight, body fat percentage and muscle mass changes more in the second and third trimester. It is important to modulate the training load according to the trimester-specific response, although these improvements may vary according to factors such as genetics, diet, sleep and the specific training. Astrophysics - High Energy Astrophysical Phenomena ,Astrophysics - Instrumentation and Methods for Astrophysics ,Astrophysics - Solar and Stellar Astrophysics - Abstract
X-ray polarimetry, sometimes alone, and sometimes coupled to spectral and temporal variability measurements and to imaging, allows a wealth of physical phenomena in astrophysics to be studied. X-ray polarimetry investigates the acceleration process, for example, including those typical of magnetic reconnection in solar flares, but also emission in the strong magnetic fields of neutron stars and white dwarfs. It detects scattering in asymmetric structures such as accretion disks and columns, and in the so-called molecular torus and ionization cones. In addition, it allows fundamental physics in regimes of gravity and of magnetic field intensity not accessible to experiments on the Earth to be probed. Finally, models that describe fundamental interactions (e.g. quantum gravity and the extension of the Standard Model) can be tested. We describe in this paper the X-ray Imaging Polarimetry Explorer (XIPE), proposed in June 2012 to the first ESA call for a small mission with a launch in 2017 but not selected. XIPE is composed of two out of the three existing JET-X telescopes with two Gas Pixel Detectors (GPD) filled with a He-DME mixture at their focus and two additional GPDs filled with pressurized Ar-DME facing the sun. The Minimum Detectable Polarization is 14 % at 1 mCrab in 10E5 s (2-10 keV) and 0.6 % for an X10 class flare. The Half Energy Width, measured at PANTER X-ray test facility (MPE, Germany) with JET-X optics is 24 arcsec. XIPE takes advantage of a low-earth equatorial orbit with Malindi as down-link station and of a Mission Operation Center (MOC) at INPE (Brazil)., Comment: 49 pages, 14 figures, 6 tables. - Abstract
This descriptive study aimed to investigate the current practices of Portuguese and Brazilian soccer coaches in the design and implementation of small-sided games (SSGs) in soccer. A total of 187 male coaches participated in the online survey, consisting of 82 Portuguese and 105 Brazilian individuals. These coaches held various positions within the technical staff, with 63 serving as head coaches, 38 as assistant coaches, 38 as physical trainers, and 48 in other roles related to the technical staff. Additionally, the participants represented both youth (n = 102) and adult competitive levels (n = 59), along with some who were not currently associated with a specific group. The survey consisted of 32 questions divided into three main sections: (i) the timing of SSG application, (ii) the methods used for applying SSGs, and (iii) the reasons for applying SSGs. The Chi-square test revealed a statistically significant association between nationality and the frequency of SSGs used in training sessions during the pre-season (p = 0.039) and in-season (p < 0.001). Moreover, significant association between nationality and the time allocated to employing SSGs for targeting aerobic training (p < 0.001) was found. There was a significant association between nationality and the weekly frequency of SSGs use for targeting sprint training (p = 0.019). The Chi-square test identified significant associations between nationality and the use of SSGs for targeting technical training (p = 0.002), as well as for tactical training (p = 0.002). In summary, this study underscores that SSGs are primarily employed to enhance aerobic fitness, change of direction, technical skills, and tactical behaviors. Coaches generally favor employing SSGs two to three times a week, with typical sessions lasting between 16 to 30 minutes. Notably, the major discrepancies between nationalities lie in the importance assigned to the use of SSGs. However, in practice, the formats and objectives for implementing SSGs remain relatively similar. [ABSTRACT FROM AUTHOR]

- Abstract
The aim of the present study was twofold: (i) to analyze the relationship between weekly variations in ACWR, training monotony (TM), training strain (TS), s-RPE and the number of jumps; and (ii) to explore the association between tests based on the external and internal training intensity (TI) measures across weeks. An observational cohort study was conducted. Ten female elite volleyball players (two setters, three middle-blockers, three outside hitters, and two opposites) of a first Division Portuguese team (24.1 ± 6.12 years old; 66.48 ± 6.85 kg; and 175.4 ± 6.52 cm) participated in this study. Players were monitored for external and internal TI measures. The correlation coefficients between internal and external TI measures were determined using Pearson's r. The results showed a moderate relationship between weekly variations of ACWR, TM, TS, s-RPE, and the number of jumps. Additionally, a moderate association between external and internal TI measures were detected. In conclusion, s-RPE seems to be a more sensible measure than the number of jumps, since its percentage of variation was higher. [ABSTRACT FROM AUTHOR]

- Abstract
The primary objective of this study was to systematically investigate the physiological and physical fitness adaptations resulting from small-sided games (SSGs) training programs in basketball players competing at youth competitive levels, as compared to other training approaches and/or control groups. To achieve this, we conducted a literature search on PubMed, Scopus, SPORTDiscus, and Web of Science, adhering to the Preferred Reporting Items for Systematic Reviews and Meta-Analyses guidelines. From the initial 626 studies retrieved, five were considered eligible for the current study. Among the five included articles, four conducted comparisons between the effects of SSGs and running-based high-intensity interval training. Regarding this, the four studies revealed a significant improvement in the final velocity during the 30–15 Intermittent Fitness Test, ranging from 4.07% to 7.29% following SSG-based interventions. This improvement was not significantly different from the comparator group. Additionally, two studies indicated that the SSGs group showed a significant advantage in change-of-direction time, with improvements ranging from −2.11% to 6.69% after interventions, and these results were not significantly different from the comparator group. However, the effects on repeated sprint ability yielded contradictory findings; two studies reported significant improvements ranging from −5.00% to −2.16%, while two others did not show significant effects following SSGs-based interventions. Similarly, in the linear sprint, the results of SSGs-based interventions were inconsistent. In summary, based on the available research, it can be concluded that SSG-based training is effective in significantly enhancing aerobic performance and change of direction, comparable to alternative approaches. However, the effects on repeated sprint ability and sprint performance are not consistently demonstrated. [ABSTRACT FROM AUTHOR]

The present study aimed to investigate the effects of a 10-week online high-intensity interval training (HIIT) program on body composition and aerobic and aerobic performance in physically sedentary women. A parallel, two-group, longitudinal (pre, post) design was used with physical tests performed before (preintervention) and after (postintervention) the 10-week intervention period. A total of forty-eight healthy and physically sedentary women (defined as an individual who lacks regular exercise or a structured fitness routine) were recruited to participate in this study. The participants were distributed in two groups: the experimental group (EG) with 24 women (mean ± SD: age 21.21 ± 2.15 years; weight: 61.16 ± 8.94 kg; height: 163.96 ± 4.87 cm; body mass index (BMI): 22.69 ± 2.49 kg/m
2 ) and the control group (CG) with another 24 women (mean ± SD: age 20.50 ± 1.29 years; weight: 62.0 ± 6.65 kg; height: 163.92 ± 4.89 cm; body mass index: 23.04 ± 1.74 kg/m2 ). The EG performed an online HIIT program for 10 weeks, while the CG continued with their daily life routines. The repeated measures ANCOVA indicated a significant effect in the within-group analysis for weight (p = 0.001; d = −0.96) and for BMI (p = 0.001; d = 0.24), with a significant decrease in the experimental group (EG). The control group (CG) did not show any significant decrease in either body weight or BMI. Regarding the maximal oxygen uptake (VO2 max) values, the EG exhibited a significant improvement (p = 0.001; d = −1.07), whereas the CG did not demonstrate a significant improvement (p = 0.08; d = −0.37). The EG's power output (W) (p = 0.001; d = −0.50) and power output standardized by body weight (W/kg) (p = 0.001; d = −0.96) were significantly improved. The CG did not show a significant improvement in either power output (W/kg) or power output. Lastly, the within-group analysis with load revealed that the EG significantly improved (p = 0.001; d = −0.50), while CG did not show a significant improvement in load (p = 0.10.; d = −0.10). The present study showed that 10 weeks of HIIT in an online environment during the COVID-19 lockdown significantly improved maximum oxygen consumption and caused weight loss and a significant decrease in body mass index in physically sedentary women. These results suggest that HIIT may be used as a time-efficient strategy to improve body composition and cardio-respiratory fitness in sedentary women. [ABSTRACT FROM AUTHOR]- Published

This study had two objectives: (i) to analyze the between-position differences in training:match load ratios and (ii) to test the relationships between the weekly training and match demands of male professional football players over a season. A cohort study lasting 43 weeks was performed. Nineteen professional football players (age: 27.5 ± 4.6 years old) used a 15-Hz global positioning system (GPS) unit integrating a 100-Hz tri-axial accelerometer. Total distance (TD), metabolic power average (MPA), new body load (NBL), accelerations (ACC), and decelerations (DEC) were considered. The training:match ratio was obtained for all the external load measures. Significant between-position differences were found only for DEC. Moderate correlations between the weekly training and match demands were found for NBL (r = 0.343 (0.19; 0.48); p < 0.008) and DEC (r = 0.472 (0.327; 0.595); p < 0.001). Moderate correlations between the mean training intensity and match demands of the same week were found for NBL (r = 0.454 (0.313; 0.575); p < 0.001) and DEC (r = 0.451 (0.304; 0.577); p < 0.001). This study did not show significant position differences for the overall training:match ratios. Significant position differences were revealed for left-back players compared to all other positions. Fullbacks performed four times more DEC during training sessions than during matches. It was revealed small to moderate associations between both the volume and intensity of the overall external load measures and their respective match running demands. However, such correlations are too weak to suggest a cause-and-effect relationship.

Quanto ao teor em ortofosfatos, o valor médio nas amostras de água subterrânea é de 0,25 mg PO4 3-/L, inferior ao VMA de 1,533 mg PO4 3-/L., The Douro River watershed plan warns of a problem of eutrophication in the Torrão reservoir, located in the Tâmega river watershed, relating it to the surroundings land usage. This alert was reiterated by the Portuguese Environmental Agency (APA), considering, amongst other sources of contamination, the agricultural sector as the main responsible for the production of nitrate (NO3 - ) and orthophosphate (PO4 3- ) rich effluents. This thesis aims to build a groundwater monitoring network for the Tâmega watershed in order to evaluate if the agricultural activities present in this area are in fact having an impact on nitrate and orthophosphate concentration in these waters. The methodology used to produce this network is based on the use of geographical and biophysical data, with particular influence in nutrient concentration and water infiltration capabilities, of the study area. In common engineering practice, there is always a concern regarding the stress control as it prevents material failure. The well-known compliance-based formulations result in stiff- ness-oriented optimal designs. However, strength-oriented designs are crucial in engineering practice. The present work addresses multi-material stress-based topology optimization, not well covered in the literature. More specifically, it aims to minimize the von Mises stress peak at the macrostructural level, through an appropriate interpolation law, where the performance of functionally graded materials (FGMs) in stress mitigation is investigated. These are ob- tained as an extensive smooth variation of material properties on account of varying compo- sition’s volume fractions of two isotropic solids throughout the design domain. Furthermore, one uses a density-based method, combined with other well-established techniques, to over- come the stress control difficulties in optimization problems. Three benchmarks problems are revisited here, plate with a hole under hydrostatic load, plate with a hole under shear load and MBB-Beam. Firstly, solving both the compliance and stress-based formulations in a single-material setting, one obtains the same design, i.e., the stiffest and strongest designs coincide. Ultimately, extending the stress-based formulation to graded materials, lower stress peaks are achieved in these solutions when compared to the single-material ones. The FGM solutions nearly attain fully stressed designs, optimal for stress mitigation.

The increasing aggression in the population, which is undoubtedly associated with the onset of narcissism, also entails an increase in the number of assaults in society. Of course, the number of clashes when a Weapon is used is also increasing. In most cases, these are weapons classified as weapons of category D (according to the Czech legislation), especially air guns, expansion weapons, and airsoft guns. This fact needs to be addressed, not by impulsive bans under duress, but by subjecting these weapons to the investigation, especially in the field of wounding ballistics, which answers the questions about the wounding potential of these weapons. This paper deals with one of these weapons, specifically airsoft gun with used ammunition lead shot. The depth of the shot and the size of the temporary and permanent cavity in relation to the distance were investigated, the contact distance was measured, and then the distance of 5 m.

This research was focused on the development of a project that aimed the analysis of an industrial process and presentation of improvement actions to reduce out-of-specification product. The project was developed in the hot preparation department at Continental Mabor, in the process of wedge application (APEX). It was followed a DMAIC approach and quality tools were used for the process characterization and analysis. The output of the process must comply with the specified values and if it’s not according these values, it results in waste, output loss and possible internal and external complaints. The identification and elimination of the causes of non-conforming product will prevent the creation of waste and potential non conformities. The company and the process of wedge application were characterized in detail and it was presented an overview of the current status. This information was analyzed, improvement actions were proposed and it was provided more information for future actions and decisions. Continental Mabor has an established quality culture that reinforces the use of quality methodologies and tools in problem solving. Unlike other processes, the data about this process was almost non-existent which made it difficult, at an early stage, to understand the current state of the process and to prioritize the project's objectives. The completion of this project allowed the identification of several factors influencing the process performance and the presentation of improvement actions to reduce the impact of these factors. It also allowed to obtain updated data about the process, which will help in future actions, decisions and projects.

The aim of this study was two-fold: (i) to analyze the variations of training intensity and well-being measures of youth football player over a 38 week period; and (ii) to test the relationships between training intensity and well-being variations throughout a youth football season. This study followed a longitudinal design, lasting 38 weeks. Twenty-five players (age: 15.0 ± 0.4 years) participated in this study. Participants were monitored daily to quantify the training intensity (TI) using the session-rate of perceived exertion (s-RPE) and its related indices: training monotony (TM), weekly training intensity (wTI), mean training intensity (mTI), and 5-day average (5d-AVG). A four-item questionnaire was collected daily to quantify the well-being status of each player. Fatigue, stress, delayed onset muscle soreness (DOMS), sleep quality, and the Hooper Index (HI) measures were analyzed. Kruskal-Wallis H test revealed significant differences for TM, mTI, wTI and 5d-AVG (H = 359.53, p = 0.001, η2 = 0.35; H = 414.97, p = 0.001, η2 = 0.41, H = 258.42, p = 0.001, η2 = 0.24 and H = 282.18, p = 0.001, η2 = 0.26). A positive large correlation was found between TM and sleep quality (r = 0.65, p = 0.05) and a negative large correlation between mTI and sleep quality (r = −0.69, p = 0.04). In conclusion, both TI and well-being measures present significant between-week differences at different periods of the season. Also, the variations of sleep quality seem to depend on TM and mTI variations across a youth football season. [ABSTRACT FROM AUTHOR]

The use of online commerce as a means of making purchases has been on the rise around the world. Companies such as Amazon and Alibaba have been the big drivers in this market, which quickly went from being a bet for the future to become a present market where it is possible to present significant gains. Aware of these gains and trying to get their share, companies around the world have increased investment in Marketplace and own sites. Even with this great growth in online sales, fresh products are still a product that has not been able to impose itself in this market, being even one of the less purchased types of products in online commerce. Thus, this study aimed to better understand the consumption of fresh products online and in particular to identify the main factors influencing the purchase of this type of products. After the research we identified eight factors, namely price, perceived usefulness, perceived ease of use, perceived risk, credibility attributes, demand attributes, and quality. The research conducted was based on studying the differences in these eight factors between two distinct groups of consumers, those who had already made an online purchase of fresh produce and those who had not yet made any online purchases. For this purpose, an online questionnaire was applied to a sample of 305 consumers, where 104 belong to the buyer’s group and the remaining 201 belong to the non-buyers group. The results indicate that there are differences between the two groups about price and perceived utility, with the buyer group placing a higher value on perceived utility and a lower value on price than the non-buyer group, that is, the buyer group does not mind paying more if they can identify the utility of the purchase. There were no statistically significant differences in the other determinants studied. These results suggest several recommendations for the online marketing of fresh products with emphasis on a greater focus on improvements that increase the perceived value of the online purchase by the customer throughout the different stages of the customer journey, from purchase to delivery, such as greater flexibility in delivery and facilitating the same benefits that they would have if they purchased in the offline channel.

- Abstract
Tese de Doutoramento em Engenharia Química e Biológica, Industrial biotechnology is a segment of the bioeconomy in which microorganisms are considered core assets, playing a central role as cell factories in fermentation processes. Despite numerous proof-of-concept studies performed in industrial models, many commercial bioprocesses are based on non-conventional microorganisms with optimized innate metabolic potential. In fact, the choice between engineering microbial models or improving the innate metabolism of non-conventional microorganisms has been a major question in the field. Ashbya gossypii is a filamentous fungus that stands in the intersection between industrial models and non-conventional microorganisms. It is considered a paradigm of the industrial biotechnology owing to the success of the bioprocess implemented almost 30 years ago that makes use of its overwhelming capacity to overproduce riboflavin (vitamin B2). Until the beginning of this decade, its biotechnological relevance remained restricted to this application. Since then, interesting potentialities of its natural metabolism have been unravelled. Therefore, with the objective of developing A. gossypii as an alternative cell factory and of identifying novel biotechnological products, this thesis focused on the systematic exploration of this fungus natural metabolism through its physiological and molecular characterization. The overproduction of riboflavin is the most distinctive trait of this hemiascomycete. It has been proposed to act as an ecological defence, as it is triggered by environmental stress. Envisioning a better understanding of this trait, a presumed genotoxic effect associated with the overproduction of riboflavin was investigated. For assessing that, the Ashbya Comet Assay was developed, which was able to reproducibly measure DNA damage. Using this protocol, exposure to sun-mimicking light during growth was found to significantly increase DNA damage in riboflavin-overproducing cells, but not in non-overproducing ones. In overproducing cells, light induced the intracellular accumulation of reactive oxygen species and increased the production of riboflavin 1.5-fold. These results draw attention for the importance of controlling the exposure to light of riboflavin biotechnological processes. Proceeding with the influence of riboflavin overproduction on the global metabolism of A. gossypii, the molecular basis for the unusual sensitivity to uracil displayed by the A. gossypii Agura3 pyrimidine auxotroph was identified. Uracil phosphoribosyltransferase (UPRTp) is the pyrimidine salvage pathway enzyme responsible for converting uracil to uridine monophosphate in the presence of phosphoribosyl pyrophosphate (PRPP). Characterization of the A. gossypii UPRTp, produced and purified from Escherichia coli, revealed that uracil concentrations above 1 mM negatively affected its activity, thus explaining the hypersensitivity of the Agura3 mutant to uracil. Decreased UPRT activity ultimately favours the preservation of PRPP, which may be directed to promote riboflavin production. Thus, this UPRTp modulation reveals a putative means of saving precursors essential for riboflavin overproduction. The A. gossypii Agura3 mutant, which displays a riboflavin-overproducing phenotype, was further identified as a suitable host for the production of a novel high-value compound, orotic acid. This strain was found to accumulate and excrete orotic acid to culture supernatants. Evidences are discussed indicating that the metabolic feature crucial for this phenotype is a putative mitochondrial dihydroorotate dehydrogenase encoded by AgURA9. Further engineering of this strain with the overexpression of a gene involved in glycerol metabolism (AgGUP1) improved its glycerol consumption profile, as well as its glycerol-dependent hyperosmotic tolerance. Thus, a flexible and robust strain for the production of high-value chemicals was constructed, which is able to sustain their production from a medium completely based on industrial by-products. Finally, an alternative production process for γ-lactones is presented. The biotechnological exploration of these valuable fragrances relied solely in biotransformation processes, which limited their production from more sustainable substrates. Here, the innate ability of A. gossypii for de novo biosynthesis of γ-lactones from glucose was uncovered and improved. Characterization of nine strains of this fungus revealed the presence of seven chemically different γ-lactones. To understand this biosynthetic process, metabolic engineering strategies were applied to the fatty acid biosynthesis and the β-oxidation pathways. Overexpression of AgDES589, encoding a desaturase for the conversion of oleic acid into linoleic acid, and deletion of AgELO624, which encodes an elongase that catalyzes the formation of longer fatty acids increased the production of γ-lactones up to 6.4-fold. Further substitution of AgPOX1 by a codon-optimized POX2 gene from Yarrowia lipolytica fine-tuned the biosynthesis of γ-decalactone. By means of metabolic engineering, this study elucidated key steps in the biosynthesis of these compounds, and demonstrated the potential of A. gossypii as a model and future platform for the de novo biosynthesis of γ-lactones. In sum, this thesis contributed to the goal of establishing A. gossypii as an alternative cell factory by disclosing two novel biotechnological applications that emerged from the exploration and understanding of this fungus natural metabolism., A biotecnologia industrial é um segmento da bioeconomia no qual os microrganismos são considerados ativos nucleares, desempenhando um papel central como fábricas celulares em processos de fermentação. 30. ESSAYS ON EQUILIBRIUM SELECTION AND GAME THEORY

- Abstract
The problem of equilibrium selection has been a relevant and fundamental topic in game theory since the own definition of the Nash equilibrium concept. However, despite its importance, this topic does not appear any longer as a top priority in the game-theoretic research agenda; at least, considering the absence of relevant contributions in the last two decades. In its present state, the existent literature consists of an eclectic collection of methods, each one dependent on a particular methodology, or tailored merely to a specific class of problems. Hence, it becomes almost impossible to organize those approaches under the umbrella of one or two research programs. The current state is partially the product of the initial approach to the equilibrium multiplicity issue; that was the equilibrium refinement research program. Its purpose was the refinement of the equilibrium concept, seeking in the process to produce more reasonable predictions, and to reduce the number of possible solutions in a game. However, the result was a sequence of competing refinements and the absence of a consensual and unanimous concept. Even in extensive-form games, where the sequential equilibrium concept is possibly the most sensible candidate, we can find several variations that depend on the conditions for the consistency of beliefs, none of which being entirely satisfactory. The first approach aiming at the development of rational criteria for the selection of an equilibrium as the solution of a game emerges in the late 1980s, seen as the culmination of the refinement program and led by two of its main contributors, John Harsanyi, and Reinhard Selten. The subsequent interest resulted in some new methods and theories; however, none became utterly dominant. The slowdown that followed was also the product of an emerging paradigm, under which the information provided by the multiplicity of solutions is perhaps more useful than the prediction/prescription of a single one. However, in my opinion, game theory has reached a stage that justifies renewed attention over this topic. In the one hand, the information provided by the multiplicity of solutions does not prevent the selection of a single solution. On the other hand, the dissemination of game theoretical tools in applied research raises the necessity for sensible predictions. Those predictions do not have to correspond to an exact behavior code; instead, they may just have a prescriptive orientation, matching the limitations that real agents face. The absence of such criteria limits the impact of game-theoretical tools and raises questions as to the usefulness of the theory itself. The product of behavioral and experimental economics research also has increased the criticism on game theory, based on the argument that a theory that does not make consistent and recurrent predictions cannot be effectively useful in social sciences. In this dissertation, I treat the selection issue with a prescriptive orientation. Therefore, I propose an equilibrium selection method to static games with complete information and an extension of it to dynamic games with asymmetric information. In the first paper - equilibrium selection in static games - I define criteria of risk and payoff dominance, which I combine into a single measure. That measure - the premium of an equilibrium - represents the risk of an equilibrium to a player, given his perception about risk, and the expected payoff. Such measure helps to rationalize the available experimental evidence by adjusting the importance of each dominance criterion to the selection of an equilibrium according to the characteristics of the game, namely, the distribution of the payoffs across the game outcomes. The solution of a game is an equilibrium that minimizes the premium to the player, that is, which minimizes the risk to a player given their perception of risk and their expected payoff, conditional on the same being true for every opponent. I provide a brief axiomatic characterization of that measure, and show that the solution set is nonempty and that almost all games have a unique solution; therefore, the set of games with multiple solutions have null Lebesgue measure. In the second paper, I extend the method to dynamic games with asymmetric information. Considering the sequential nature of decision making, I show that a solution of an extensive-form game does not necessarily coincide with the solution of its reduced normal-form. I apply the method to the most basic version of Spence signaling game with just two types of worker. I obtain that the solution of such a game depends on the firms? prior concerning the players? types. In both dynamic and static games, I show that the method?s solutions respect certain invariance properties. I am then able to identify several directions of research. In one direction, we have the application of the selection method to specific problems in which the multiplicity of solutions play a critical role. Among such problems, I highlight bank-runs, and climate change negotiations. Additionally, I am interested in expanding the selection analysis of Spence game through the inclusion of additional types of worker. Another direction follows a different path, and focus on the identification and further characterization of the epistemic conditions of this selection method, and the comparison with the conditions in other selection methods and/or equilibrium refinement concepts.

This study aims to discover how to make the best possible use of the Army’s career officers in the reserves so that the Army can capitalise on and enhance the value of its human resources. The investigation used an inductive approach, which involves understanding specific aspects to make general inferences. To solve the proposed problem, the answers to two questionnaires were analysed and compared against the Human Capital Valuation model by Delgado (2015). Each questionnaire served a different purpose: the first ascertained the Army’s requirements in terms of these resources, and the second determined how to enhance their value. The surveys were delivered to respondents from the six bodies in the Army’s Command Structure that encompass all the Units, Establishments and Corps that make up the Portuguese Army. The findings revealed that: one body does not employ any reserve officers and two bodies would not increase these HR; all bodies recognise the value of these officers’ experience and technical expertise. Although the valuation model used in this study was not specifically tailored to the military context, it provides a basis for a future model. The study concluded that, in order to enhance the value of these officers, the Army must intervene on the variables financial well-being, entrepreneurial spirit, proactive work performance and level of qualification. info:eu-repo/semantics/publishedVersion

- Abstract
Glycerol is a main by-product of the biodiesel industry and thus has drawn attention as substrate for the production of high-value chemicals by microorganisms. Compared to sugars like glucose, it offers some advantages since it holds a higher degree of reduction. However, many industrial microbes have poor ability to use it. For instance, the eukaryotic model Saccharomyces cerevisiae has to be engineered to efficiently metabolize it. In contrast, the filamentous fungus Ashbya gossypii, used for industrial riboflavin production, owns a natural ability to utilize glycerol. Still, its potential to exploit it remains unexplored. Therefore, in this study, an engineered A. gossypii strain, harnessed for improved riboflavin production1 and orotic acid biosynthesis, was further modified for improved glycerol consumption. This strategy revealed to be efficient in improving the consumption profile, the glycerol-dependent hyperosmotic tolerance and the yield of biomass production of A. gossypii strains while effectively maintaining the metabolic flux towards increased riboflavin production and orotic acid accumulation/excretion. Furthermore, optimization of process conditions in shake-flask and bioreactor were performed to enhance the production of these compounds. Thus, we present A. gossypii as a suitable host for the production of high-value chemicals, particularly orotic acid and riboflavin, from glycerol feedstocks. 1Silva et al, J Biotechnol 193;37-40., info:eu-repo/semantics/publishedVersion

- Abstract
Water is probably the oldest factor receiving attention from the population, since the beginning of the human race. The evolution of water supply systems is directly related to the urban growth of the cities and their geographical location. This study focuses on SCADA (Supervisory Control And Data Acquisition) systems used to control and managing water distribution systems all around the world, highlighting their components and deeply approaching SCADA networks and communication protocols. Based on the need of protection and security on SCADA networks, this study aims to provide a review of the existing literature, through the case study of water distribution systems, identifying the different components of a SCADA system, as well as its communication architectures and protocols and some known attacks, to design a framework on current and old SCADA networks. Results show that SCADA systems are not just applied to water distribution and water waste systems, but also to many other industrial automated systems that control on crucial systems. The security of such systems is yet weak and faces many vulnerabilities and threats, where security mechanisms must be applied. For this purpose, the study of SCADA networks and communications is considered a crucial point for the correct development of security tools.

The exploding mobile data traffic increase in recent years has been putting a high load on both mobile cells and core network, with operators facing the need to upgrade their networks. Nowadays, to do this upgrade, operators need to purchase new specialized equipment for network functions, having to cope with a high upgrade CAPEX. Furthermore, networks are deployed with a one size fits all approach, which in some cases might not satisfy the requirements of specific services. 5G aims to solve these problems by virtualizing network functions in datacenters, decoupling the software from the hardware for network functions and using general purpose hardware instead. To support this, Software Defined Networking (SDN) is introduced, which allows the network to have a higher degree of programmability, enabling new features such as higher flexibility and network slicing, where multiple virtual networks can be created and tailored to specific requirements. This thesis addresses an architecture that evolves the Evolved Packet Core (EPC) into a core network closer to 5G by virtualizing EPC’s network functions, introducing SDN and supporting 4G to Wi-Fi traffic offloading, helping to reduce the load on mobile cells by leveraging on the smartphone’s support for dual connectivity and high density of Wi-Fi access points already deployed worldwide. The proposed architecture is then evaluated and compared to a vanilla EPC whenever possible showing that, although there is an increase in latency at the virtual EPC, the bottleneck of the system resides in the air interface. Also, a use case for this architecture was defined and evaluated. The use case considered traffic offloading and dynamic Wi-Fi slice creation, with results showing that it can seamlessly offload a video stream from 4G to Wi-Fi without affecting the user’s Quality of Experience. - Abstract
[Introduction] The Comet Assay (Single Cell Gel Electrophoresis) is one of the most used techniques for measuring DNA damage1 due to its simplicity, sensitivity and versatility2. The theoretical scientific basis of this methodology consists on the increased electrophoretic mobility of damaged DNA. It has a broad range of applications such as: compounds’ genotoxicity/protective testing, DNA repair studies and ecological monitoring. With a huge amount of optimized protocols for superior eukaryotes, the Comet Assay has been poorly utilized among microorganisms. However, Azevedo et al. (2011)3 developed an optimized protocol for Saccharomyces cerevisiae that reinforce the utility and potential of this method for microbial systems. Therefore, in this work we created a reproducible and optimized Comet Assay protocol for use in Ashbya gossypii by adaptation of the Yeast Comet Assay3 (Fig. 1). This protocol allows the measuring of oxidative and non-oxidative DNA damage. Moreover, as riboflavin overproduction in A. gossypii has been associated with stress (mainly oxidative), we went to check if this trait was also associated with increased DNA damage. Therefore, we used this recently created protocol to investigate the DNA damage accumulation in riboflavin overproducing and non-overproducing A. gossypii wild strains., The authors thank the financial support from FCT, Portugal: strategic funding of UID/BIO/04469/2013 unit and COMPETE 2020 (POCI-01-0145-FEDER-006684) (Post-Doc fellowship to TQ Aguiar), BioTecNorte operation (NORTE-01-0145-FEDER-000004) funded by FEDER under the scope of Norte2020 - Programa Operacional Regional do Norte, and PhD grant to R Silva (PD/BD/113812/2015) through the FCT PhD Programme in Applied and Environmental Microbiology., info:eu-repo/semantics/publishedVersion

Glycerol ,Hyperosmotic stress ,Ashbya gossypii - Abstract
The worldwide scenario of strong investment in biodiesel production brings new economical and environmental concerns that need to be addressed. Among them, the most prominent lies on the accumulation of large amounts of a main by-product of the process, crude glycerol. Valorization of this biodiesel waste through its microbial conversion into high-value products is one of the most attractive and sustainable applications explored. Crude glycerol contains several impurities such as methanol, soaps and salts. After pre-treatment of this biodiesel waste, some impurities can disappear or be mitigated, however salts as KCl and NaCl continue to be present, which impose osmotic pressure to microorganisms. The filamentous fungus A. gossypii [1], used for more than 20 years for the commercial production of riboflavin, is able to metabolize glycerol and use it as its preferred solute to overcome hyperosmotic stress conditions [2]. However, A. gossypii exhibits great sensitivity to hyperosmotic stress when compared with other fungal species [3]. Here, we designed a strategy to improve the osmotic stress tolerance of A. gossypii by overexpressing native and heterologous genes involved in the uptake of glycerol. The cells were exposed to hyperosmotic stress in complex medium with glucose as carbon source and their colony radial growth was monitored. Different stress agents, ionic (NaCl and KCl) and non-ionic (sorbitol), were used in the presence or absence of glycerol to test if the overexpressing strains could efficiently utilize exogenous glycerol towards an improvement in hyperosmotic stress tolerance. In the presence of 0.8M and 1M KCl, A. gossypii overexpressing strains displayed significantly improved hyperosmotic stress tolerance compared to the control strain, being able to grow in these conditions due the presence of glycerol in the medium. These results contribute to the further development of A. gossypii as an environmental-friendly cell factory organism, contributing to its establishment in the biorefinery concept., Fundação para a Ciência e a Tecnologia, Portugal: Strategic Project CEB unit UID/BIO/04469/2013 and PhD grant PD/BD/113812/2015 to R Silva

Dissertação de mestrado em Bioengenharia, The implementation of biorefineries has been proposed as a mean to increase the economic viability of the biobased industries. In its “conventional” form, a biorefinery makes use of by-products (e.g. crude glycerol) generated during the production of biodiesel to coproduce high-value products. For this, it is necessary to develop robust microorganisms, easily metabolically and genetically manipulated in order to develop tailor made cell factories. Ashbya gossypii is considered an example of the sustainable white biotechnology business model in what concerns the industrial production of riboflavin. Its biotechnological relevance has allowed the development of directed and based-knowledge methodologies for strain optimization for riboflavin production. The metabolic engineering strategies tested to create overproducing strains have focused on pathways strongly connected with riboflavin production. However, the A. gossypii pyrimidine pathway and its connection with the riboflavin biosynthetic pathway had not yet been assessed. We report that the blockage of the de novo pyrimidine biosynthetic pathway in the recently generated A. gossypii Agura3 uridine/uracil auxotrophic strain led to improved riboflavin production. Considering that the riboflavin and the pyrimidine pathways share the same precursors and that riboflavin overproduction may be triggered by stress, we suggest that overproduction of riboflavin by A. gossypii Agura3 may occur as an outcome of a nutritional stress response and/or of an increased availability in precursors for riboflavin biosynthesis, due to their reduced consumption by the pyrimidine pathway. Despite the recognized capabilities of A. gossypii to use industrial wastes as substrate for the production of riboflavin, glycerol had not yet been exploited. In this study we explore riboflavin production by A. gossypii from glycerol. Additionally, we designed a strategy to improve its glycerol consumption profile by overexpressing glycerol uptake proteins (GUP1) genes. A. gossypii strains overexpressing the native GUP1 gene (pRSAG) or GUP1 gene from Saccharomyces cerevisiae (pRSSG) under the control of the AgTEF promoter displayed significantly improved glycerol-dependent hyperosmotic stress tolerance and glycerol consumption profiles, indicating that our strategy successfully led to the improvement of glycerol utilization. These results contribute to the further development of A. gossypii as an environmentalfriendly cell factory organism, contributing to its establishment in the biorefinery concept., A implementação de biorrefinarias tem sido proposta como um meio para aumentar a viabilidade económica das bioindústrias.

- Abstract
The objectives of this study were twofold: (a) to analyze the variability of 3v3 and 5v5 small-sided games (SSG) formats in sedentary young adults, and (b) to compare the physiological and locomotor demands of 3v3 and 5v5 SSG formats while considering variations based on sex. The study followed a longitudinal design over 4 months. Thirty sedentary young adults with a mean age of 20.2 ± 1.0 years, height of 1.67 ± 0.06 m, and body mass of 86.3 ± 11.8 kg were included in the study. The participants engaged in 3v3 and 5v5 SSG formats, with each format being played 10 times per month. During each session, heart rate (HR), rate of perceived exertion (RPE), and total distance were measured and analyzed. The within-player variability for HR ranged from a minimum of 1.6% to a maximum of 2.8% (considering the levels at each month), while the between-players variability for HR ranged from a minimum of 1.4% to a maximum of 2.6% (considering the levels at each month). Similar variability patterns were observed for the other outcomes. In terms of comparisons between the formats, the 3v3 format resulted in higher RPE than the 5v5 format for both male (p= 0.006) and female (p= 0.628) participants, as well as for the other outcome measures. In summary, these findings highlight the reproducibility of physiological responses in 3v3 and 5v5 SSG among sedentary individuals. Notably, the 3v3 format consistently induced higher RPE levels. These findings underscore the importance of programming SSG based on sex and format preferences for optimizing exercise outcomes in sedentary.

- Abstract
At present, the designers of voltage converters can choose from a wide variety of custom integrated drivers operating at different principles. The authors of this paper aim to show that although the two different designs of a step-down voltage converters can achieve comparable nominal values, at least from the view of the user, due to the differences in the basic principles applied in their drivers they may exhibit different behaviour in the area of the electromagnetic compatibility. This can lead to considerable problems in design of more complex circuits these voltage converters are a part of. For the purposes of the hereby presented results, two different constructions of step-down converters have been chosen. The first one is based on a low-cost and randomly operating solution based on the driver MC 34063 while the second one employs more advanced driver marketed under the label AP 1501. Both converters were constructed and tested for the purposes of application in a power backup device for Power over the Ethernet. The nominal input voltage of both converters is 24 V while their nominal output voltage is 12 V, as used in common applications. Both converters were tested in the EMC laboratory of Tomas Bata University Zlin in order to obtain not only the static parameters, but also the information on how they can interfere with other electronic devices. Within this paper, the most interesting findings are published. © 2016, North Atlantic University Union. All rights Reserved.

In recent years, the Design field and professional designers have been acknowledged as a driver and enabler of innovation, and thus important to key business activities and to firms' performance and competitiveness. The complex nature of Design still raises obstacles to the comprehension of its activity and results. Research revealed that different firms can have different understanding of Design's value potential and that their perception and usage (design maturity) might play a decisive role in the way design activity is practiced in a firm. This research intended to investigate and provide insights regarding Portuguese business firms' perception and usage of Design. For that purpose, the current Design Maturity level of Portuguese innovation-driven firms was described, using the Danish Design Ladder (DDL) framework. A survey strategy was applied, by a self-administered on-line questionnaire based on the De.:SID survey, to a group of 226 innovation-driven SMEs - COTEC Portugal's Rede PME Inovação. A response rate of 33% was achieved and interesting insights were found about the importance Design can have in a business context. The main conclusion is that respondents' design maturity corresponds to the DDL's third stage: Design as Process. Moreover, data suggests a difference between firm's perception and the actual importance and usage of the Design role in their business, which reveals a certain lack of knowledge and experience in working with Design.
