Prolonged exposure to hypoxic conditions can lead to reduced appetite, stunted growth, systemic inflammation, and pulmonary hypertension. Previous studies have indicated a correlation between gut dysbiosis and the development of hypoxia-related hazards. We designed an experiment to investigate the effect of microbiota on mitigating hypoxic damage. Gut microbiota from high-altitude-adapted species (Ochotona curzoniae) were transplanted into Sprague Dawley (SD) rats, which were then housed in a simulated 6000 m altitude environment for 30 days. After the experiment, we conducted analyses on average daily weight gain (ADG), feed conversion ratio (FCR), mean pulmonary artery pressure (mPAP), gut flora, and fecal metabolism. The results demonstrated that the ADG in the transplantation group (2.98 ± 0.17 g) was significantly higher than in the control groups (2.68 ± 0.19 g and 2.26 ± 0.13 g) (p < 0.05). The FCR was reduced in the transplantation group (6.30 ± 0.33 g) compared to the control groups (8.20 ± 1.15 g and 8.83 ± 0.45 g) (p < 0.05). The mPAP was decreased in the transplantation group (38.1 ± 1.13 mmHg) compared to the control groups (43.4 ± 1.30 mmHg and 43.5 ± 1.22 mmHg) (p < 0.05). Multi-omics analysis revealed that Lachnospiraceae, Desulfovibrionaceae, and specific amino acid metabolic pathways play crucial roles in hypoxia and are associated with both inflammation and nutritional metabolism. This study proposes a novel approach to the treatment of hypoxic pulmonary hypertension and holds potential significance for improving high-altitude developmental potential.

Abstract Immune cells-enhanced immunotherapy exhibits unprecedented overall survival-prolongation even curable in some cancer patients. Although so, most of the patients show no response. Tumor microenvironment (TME) where immune cells settle down has multi-faceted influences, but usually creates an immunosuppressive niche that facilitating tumor cells escape from immune attack. The metabolites and malnutrition of TME exert enormous effects on the resident immune cells, but the underlying mechanism is largely unknown. The stromal interaction molecules 2 (STIM2) is an endoplasmic reticulum (ER) calcium (Ca2+) sensor to maintain Ca2+ homeostasis. Notably, the cytosol STIM2 C-terminus is long with various domains that are available for the combination or/and molecular modification. This distinct structure endows STIM2 with a high susceptibility to numerous permeable physico-chemical molecules or protein interactions. STIM2 and its variants are extensively expressed in various immune cells, especially in T immune cells. STIM2 was reported closely correlated with the function of immune cells via regulating Ca2+ signaling, energy metabolism and cell fitness. Herein, we sum the latest findings on the STIM2 structure, focusing on its distinct characteristics and profound effect on the regulation of Ca2+ homeostasis and multi-talented functionality. We also outline the advancements on the underlying mechanism how STIM2 anomalies influence the function of immune cells and on the turbulent expression or/and amenably modification of STIM2 within the tumor niches. Then we discuss the translation of these researches into antitumor approaches, emphasizing the potential of STIM2 as a therapeutic target for direct inhibition of tumor cells or more activation towards immune cells driving to flare TME. This review is an update on STIM2, aiming to rationalize the potential of STIM2 as a therapeutic target for immunomodulation, engaging immune cells to exert the utmost anti-tumor effect.

Abstract Background HBP, a novel biomarker released from neutrophils, may induce inflammatory responses and exacerbate vascular permeability, representing the pathophysiological characteristics of sepsis and septic shock. However, it remains uncertain whether the combination of HBP with other biomarkers yields enhanced diagnostic capacity for sepsis. We hypothesized that measurements included IL-6·IL-8·HBP, IL-6·IL-8·HBP/ALB and HBP/ALB which based on HBP will improve its diagnostic efficacy and even better than the traditional infection biomarkers. Methods Between July 2021 and June 2022, we carried out a comprehensive, multi-center, observational cohort study spanning six leading tertiary hospitals located in Heilongjiang Province, China. Patients were stratified into three categories based on the severity of infection: non-sepsis, sepsis, and septic shock. We collected clinical and laboratory data, along with infection and inflammation biomarkers, for analysis. Results A total of 195 patients were enrolled. Among the three groups, patients with septic shock (n = 75, 38.5%) had significantly higher baseline levels of HBP, WBC, Lac, CRP, PCT, IL-6, IL-8, and IL-10 compared to non-sepsis patients (n = 43, 22.0%) and sepsis patients (n = 77, 39.5%), with statistically significant differences (p

In this paper, the remelting process is applied to Laser Directed Energy Deposition (L-DED) to repair single crystal superalloys. The main formation modes of stray grains in single crystal epitaxial growth were analyzed, and the inhibitory effect of remelting process on stray grains in single crystal epitaxial growth using L-DED was studied. Comparative experiments between remelting and non-remelting processes have been conducted. Three process parameters in the remelting process: power, z-axis lift and cladding-remelting interval time were studied. The impact of different remelting process parameters on single crystal epitaxial growth and the formation of stray grains has been studied. Remelting has been shown to effectively prevent the formation of stray grains during the solidification of the melt pool, which are otherwise caused by the presence of unmelted powder during the powder feeding phase of the L-DED process. A cladding layer without stray grains on both sides is obtained. After remelting, the epitaxial growth structure can achieve larger cellular and columnar dendritic zones and smaller primary dendrite arms. A favorable epitaxial growth structure was obtained in this paper, under the optimized remelting process window.

To evaluate the impact of different roughages on the intestinal microbiota of yak calves, we fed them oat hay in substitution of alfalfa hay, in addition to milk replacer and starter powder. Twenty-one 45-day-old male yak calves were selected and randomly assigned to three groups: the milk replacer + starter + alfalfa hay group (AH), the milk replacer + starter + oat hay group (OH), and the milk replacer + starter + mixed hay group (AO), in which the alfalfa hay and oat hay were administered in a 1:1 ratio. All calves in the three groups were fed the same milk replacer and an equivalent amount of dry matter. The formal experiment commenced after a 21-day pre-test period and lasted for 120 days. Following the experiment, the contents of the jejunum and colon were collected to investigate the intestinal microbiota and metabolites using 16S rRNA sequencing and LC-MS metabolomics. The result showed that the AO group had greater final body weights overall than the AH group and OH group (p < 0.05). The AH group and OH group had considerably greater feed-to-gain ratios than the AO group (p < 0.05). At the phylum level, the OH group exhibited an increased relative abundance of Bacteroidota and Spirochaetota in the jejunum (p < 0.05). The relative abundance of Actinobacteriota in the colon was increased in the AO group (p < 0.05). At the genus level, the AO group exhibited a decreased abundance of Clostridium sensu_stricto_1 (p < 0.05), and the OH group showed an increased abundance UCG-005 and Alistipes in the jejunum. There were many differential metabolites in the OH group and AO group compared to the AH group, and the different metabolites of the OH group were associated with the metabolic pathways of the nervous system, sensory system, amino acid metabolism, and lipid metabolism in the jejunum and with lipid metabolism, amino acid metabolism, and the nervous system in the colon. In the AO group, these metabolites were associated with the digestive system and the translation and metabolism of cofactors in the jejunum and with the metabolism of cofactors and vitamins in the colon. In summary, it is feasible to replace alfalfa hay with oat hay based on milk replacer and starter. The combination of the two forages enhanced nutrient absorption, improved immune function, maintained the internal homeostasis of yak calves, and was more beneficial to their growth and development.

This study investigates the effect of altitude on the fatty acid composition of pasture grass, yak milk, and yak ghee on the Qinghai–Tibet Plateau, aiming to understand how environmental factors influence the nutritional quality of these products. Samples were collected from four different altitudes and analyzed for fatty acid profiles using gas chromatography. The analysis reveals that higher altitudes are associated with an increased prevalence of beneficial unsaturated fatty acids, such as oleic acid (C18:1) and linoleic acid (C18:2n6c). These findings highlight the significant influence of altitude on yak lipid metabolism, ultimately enhancing the nutritional value of dairy products. This adaptation not only supports the health and resilience of yaks, but also provides vital nutritional benefits to residents in high-altitude regions. The research underscores the importance of further investigations to optimize dairy production practices, ensuring improved food security and health outcomes for residents of the plateau.

Gallbladder cancer (GBC) is the most common malignant tumor of the biliary system with the worst prognosis. Even after radical surgery, the majority of patients with GBC have difficulty achieving a clinical cure. The risk of tumor recurrence remains more than 65%, and the overall 5-year survival rate is less than 5%. The gut microbiota refers to a variety of microorganisms living in the human intestine, including bacteria, viruses and fungi, which profoundly affect the host state of general health, disease and even cancer. Over the past few decades, substantial evidence has supported that gut microbiota plays a critical role in promoting the progression of GBC. In this review, we summarize the functions, molecular mechanisms and recent advances of the intestinal microbiota in GBC. We focus on the driving role of bacteria in pivotal pathways, such as virulence factors, metabolites derived from intestinal bacteria, chronic inflammatory responses and ecological niche remodeling. Additionally, we emphasize the high level of correlation between viruses and fungi, especially EBV and Candida spp., with GBC. In general, this review not only provides a solid theoretical basis for the close relationship between gut microbiota and GBC but also highlights more potential research directions for further research in the future.

Abstract Polyethylene glycol-coated magnetic nanoparticles (PEGylated MNPs) have demonstrated prominent advantages in cancer diagnosis and hyperthermia therapy. However, there is currently lack of standard mode and sufficient toxicity data for determining the delayed risk of PEGylated MNPs. Nevertheless, the toxicity potentials, especially those associated with the oxidative stress, were ubiquitously reported. In this study, PEGylated MNPs and p-PEGylated MNPs were administrated to SD (Sprague Dawley) rats by single intravenously injection, and various toxicity indicators were monitored till 56 days post-administration for a comprehensive toxicity evaluation. We revealed that both nanoparticles could be rapidly cleared from plasma and enter tissues, such as, liver, kidneys and spleen, and p-PEGylated MNP is less prone to be accumulated in the tissues, indicating a lower toxicity risk. PEGylated MNPs were more likely to up-regulate the expression levels of Th2 type cytokines and trigger inflammatory pathways, but no related pathological change was found. Both MNPs are not mutagenic, while recoverable mild DNA damage associated with the presence of nanoparticles might also be observed. This study demonstrated a research approach for the non-clinical safety evaluation of nanoparticles. It also provided comprehensive valuable safety data for PEGylated and p-PEGylated MNPs, for promoting the clinical application and bio-medical translation of such MNPs with PEG modifications in the cancer diagnosis and therapy.

This study proposes an approach to combat construction waste in the architecture, construction, and engineering (ACE) industry by developing a disassemblable brick partition wall. Brick reuse is severely restricted by the presence of mortar; innovative approaches need to be explored. An existing strategy, utilizing mortarless interlocking, relies on non-standardized bricks. It is worth noting that these methods are not specifically created for disassembly, despite the fact that they theoretically could be. A relatively innovative technique for tightening and stabilizing brick units emerged in recent years, involving the utilization of metal components. Despite its potential, there are limited case studies of this approach. By drawing on two typical examples of pros and cons, MIFA 1862 and the UMAR Unit, we propose a new strategy and examine it from multiple perspectives. The findings of the analysis demonstrate how adaptable and versatile the proposed system is, allowing it to be modified into a variety of sizes and forms. Additionally, the system has proven to have considerable advantages in terms of construction speed, and energy efficiency throughout the structure's service time and in future use phases.

IntroductionThe dietary protein level plays a crucial role in maintaining the equilibrium of rumen microbiota in yaks. To explore the association between dietary protein levels, rumen microbiota, and muscle metabolites, we examined the rumen microbiome and muscle metabolome characteristics in yaks subjected to varying dietary protein levels.MethodsIn this study, 36 yaks were randomly assigned to three groups (n = 12 per group): low dietary protein group (LP, 12% protein concentration), medium dietary protein group (MP, 14% protein concentration), and high dietary protein group (HP, 16% protein concentration).Results16S rDNA sequencing revealed that the HP group exhibited the highest Chao1 and Observed_species indices, while the LP group demonstrated the lowest. Shannon and Simpson indices were significantly elevated in the MP group relative to the LP group (P < 0.05). At the genus level, the relative abundance of Christensenellaceae_R-7_group in the HP group was notably greater than that in the LP and MP groups (P < 0.05). Conversely, the relative abundance of Rikenellaceae_RC9_gut_group displayed an increasing tendency with escalating feed protein levels. Muscle metabolism analysis revealed that the content of the metabolite Uric acid was significantly higher in the LP group compared to the MP group (P < 0.05). The content of the metabolite L-(+)-Arabinose was significantly increased in the MP group compared to the HP group (P < 0.05), while the content of D-(-)-Glutamine and L-arginine was significantly reduced in the LP group (P < 0.05). The levels of metabolites 13-HPODE, Decanoylcarnitine, Lauric acid, L-(+)-Arabinose, and Uric acid were significantly elevated in the LP group relative to the HP group (P < 0.05). Furthermore, our observations disclosed correlations between rumen microbes and muscle metabolites. The relative abundance of NK4A214_group was negatively correlated with Orlistat concentration; the relative abundance of Christensenellaceae_R-7_group was positively correlated with D-(-)-Glutamine and L-arginine concentrations.DiscussionOur findings offer a foundation for comprehending the rumen microbiome of yaks subjected to different dietary protein levels and the intimately associated metabolic pathways of the yak muscle metabolome. Elucidating the rumen microbiome and muscle metabolome of yaks may facilitate the determination of dietary protein levels.

Background and objective:. The incidence of pancreatic neuroendocrine neoplasms (pNENs) has been increasing year by year, and pancreatic steatosis has been paid more and more attention to by clinicians. Our study aims to reveal the correlation between pancreatic steatosis, clinicopathological parameters, and the prognosis of patients with pNENs. Methods:. The clinicopathological data of patients with pNENs who underwent surgical resections in our institution from January 2013 to May 2022 were retrospectively analyzed. The mean computed tomography (CT) values of the pancreas and spleen were used to assess the severity of the pancreatic steatosis. Pearson Chi-square test, T test, Mann Whitney U test, Univariate Cox regression, and Multivariate Cox regression were used to explore the relationship between pancreatic steatosis and age, sex, body mass index, tumor type, tumor location, tumor stage, blood lipid, prognosis, and other factors. Results:. A total of 122 patients were included in our study, and 19.67% have pancreatic steatosis. The incidence of pancreatic steatosis was significantly higher in patients with fatty liver than in patients without fatty liver (36.8% vs 16.5%, P = .04). In insulinoma, the incidence of pancreatic steatosis in elderly patients was significantly higher than in young and middle-aged patients (41.7% vs 9.3%, P = .025). There was no significant difference in lipid levels between the pancreatic steatosis group and the non-pancreatic steatosis group (P > .05). Kaplan-Meier curves show that the prognosis of stage III/IV patients was significantly worse than that of stage I/II patients (P < .001). However, there was no significant difference in prognosis between patients with and without pancreatic steatosis (P = .404). Conclusion:. The incidence of pancreatic steatosis was significantly higher in those with fatty liver than those without fatty liver. There is a high incidence of pancreatic steatosis in elderly patients with insulinoma. American Joint Committee on Cancer (AJCC) stage, but not pancreatic steatosis, significantly affected the prognosis of patients with pNENs.

To overcome the serious situation of electromagnetic pollution at present stage, herein the hierarchical carbon/CoFe2O4/Co3Fe7 composites were prepared via an innovative ball milling assistant chemical precipitation and subsequent annealing technique. By changing the solvent ratio of DW/DMF and the additive amount of milling balls, the micro-nano structures, electromagnetic parameters and response behavior of hybrids can be adjusted, in which the N-6 composite obtains a maximal RL of −42.70 dB at 16.42 GHz with the broadest EAB of 8.68 GHz for only 2.38 mm. Based on the well matched impedance within wide frequency region, the excellent attenuation properties of this composite generated from the strong dielectric polarizations, magnetic resonances, eddy current and structure-scattering effect could play roles to absorb incident electromagnetic wave. In short, this investigation offers an innovative route to synthesize the hierarchical carbon decorated with CoFe2O4 and Co3Fe7 nanoparticles for super electromagnetic absorption.

Background: The use of noninvasive ventilation (NIV) during and after extubation is common. We designed this study to determine the optimal strategy to compensate for mask leaks and achieve effective ventilation during NIV by comparing commonly used operating room ventilator systems and a regular facemask. Methods: We tested four operating room ventilator systems (Dägger Zeus, Dägger Apollo, Dägger Fabius Tiro, and General Electric Healthcare Carestation 650) on a lung model with normal compliance and airway resistance and evaluated pressure control ventilation (PCV), volume control ventilation (VCV), and AutoFlow mode (VAF). We set the O2 flow at 10 L/min and the maximal flow at 13, 16, or 26 L/min. We simulated five leak levels, from no leak to over 40 L/min (I to V levels), using customized T-pieces placed between the lung model and the breathing circuit. We recorded the expired tidal volume (Vte) from the lung model and peak inspiratory pressure via two flow/pressure sensors that were placed distally and proximally to the T-pieces. Results: 1. Comparison of four ventilators: with any given ventilation mode, an increase in leak level caused a decrease in Vte. With PCV, only Zeus produced Vte larger than 150 ml at leak level V. 2. With the assistance of deep neural networks, a bifunctional metasurface (MS) was designed and optimized, i.e., a broadband absorber and a broadband polarization converter. The MS acts as a wide absorber when the vanadium dioxide (VO2) is in the metallic state and has an absorption bandwidth of 5.21 THz with an absorption rate ≥ 90%. In contrast, the MS acts as a linear–linear polarization converter when the top VO2 is in the insulating state and has a bandwidth of 3.7 THz with a conversion efficiency ≥ 90%. The bandwidth in both states is maximum compared to other bifunctional counterparts, while this bifunctional MS has good parametric and angular tolerance characteristics and low material cost. The proposed structure and design method of the bifunctional MS can provide a useful reference for the research of new multifunctional terahertz devices.

Common problems in engineering projects that involve artificial ground freezing of soil or rock include inadequate thickness, strength and continuity of artificial frozen walls. It is difficult to evaluate the freezing state using only a few thermometer holes at fixed positions or with other existing approaches. Here we report a novel experimental design that investigates changes in ultrasonic properties (received waveform, wave velocity Vp, wave amplitude, frequency spectrum, centroid frequency fc, kurtosis of the frequency spectrum KFS, and quality factor Q) measured during upward freezing, compared with those during uniform freezing, in order to determine the freezing state in 150 mm cubic blocks of Ardingly sandstone. Water content, porosity and density were estimated during upward freezing to ascertain water migration and changes of porosity and density at different stages. The period of receiving the wave increased substantially and coda waves changed from loose to compact during both upward and uniform freezing. The trend of increasing Vp can be divided into three stages during uniform freezing. During upward freezing, Vp increased more or less uniformly. The frequency spectrum could be used as a convenient and rapid method to identify different freezing states of sandstone (unfrozen, upward frozen, and uniformly frozen). The continuous changes in reflection coefficient rφ, refraction coefficient tφ and acoustic impedance field are the major reason for larger reflection and refraction during upward freezing compared with uniform freezing. Wave velocity Vp, wave amplitude Ah, centroid frequency fc and quality factor Q were adopted as ultrasonic parameters to evaluate quantitatively the temperature T of uniformly frozen sandstone, and their application within a radar chart is recommended. Determination of Vp provides a convenient method to evaluate the freezing state and calculate the cryofront height and frozen section thickness of upward frozen sandstone, with accuracies of 73.37%–99.23%.

Yaks living on the Qinghai-Tibetan Plateau for a long time have evolved a series of mechanisms to adapt to the unique geographical environment and climate characteristics of the plateau. Compared with other ruminants, yaks have higher energy utilization and metabolic efficiency. This paper presents possible mechanisms responsible for the efficient energy utilization, absorption and metabolism resulting from the unique evolutionary process of yaks. It is hoped that the information discussed in this review will give a better insight into the uniqueness and superiority of yaks in regards to energy metabolism and utilization compared with cattle and open new avenues for the targeted regulation of energy utilization pathways of other ruminants.

[Introduction] The purpose of this paper is to study the influence of real "wake effect" of adjacent offshore wind farms on generation loss. [Method] The method is established with the wake scene classification based on the actual arrangement of wind farms under different wind direction and the real wake power loss of adjacent wind farms (with a spacing of more than 20D) in operation are analyzed, based on the actual SCADA data of wind turbines in large offshore wind farms and the measured wind data of LIDAR in the same period. [Result] The results show that: for the large-scale offshore wind farms with regular arrangement, the power generation normalization of the actual SCADA data can better reflect the distribution characteristics of offshore wind energy resources and the difference of power generation capacity; Under the condition of highly centralized wind direction, the adjacent wind farms in the downwind are obviously affected by the "wake effect" of the upwind wind farm; The buffer zones with different distances of adjacent wind farms have an obvious effect on the recovery of wind speed which affected the power generating capacity. The power generating capacity can be improved but if the buffer zone can reach enough distance; In different scenes of this case, the buffer zone distance is between 23D and 44D, and the power loss of wake decreases by 27%~4%. [Conclusion] This work can provide guidance for the planning of offshore wind power base and the optimization design of large offshore wind frams.

Yak (Bos grunniens) is an important economic animal species on the Qinghai-Tibet Plateau. Yaks grazed in the cold season often suffer from nutritional stress, resulting in low production performance. This situation can be improved by properly feeding the grazing yaks in the cold season; however, there is still little information about the effect of different feeding levels on the intestinal microflora and metabolites of yaks. Therefore, this study aimed to explore the effect of feeding different doses of concentrate supplements on rumen bacterial communities and metabolites in grazing yaks during the cold season. Feed concentrate supplementation significantly improved the production performance and rumen fermentation status of grazing yaks during the cold season, and switched the type of ruminal fermentation from acetic acid fermentation to propionic acid fermentation. Ruminal fermentation parameters and ruminal bacterial abundance correlated strongly. At the phylum level, the abundance of Firmicutes increased with increasing concentrate supplementation, while the opposite was true for Bacteroidota. At the genus level, the abundance of Christensenellaceae_R-7_group, NK4A214_group, Ruminococcus, norank_f__Eubacterium_coprostanoligenes_group, norank_f__norank_o__ Clostridia_UCG-014, Lachnospiraceae_NK3A20_group, Acetitomaculum, and Family_XIII_AD3011_group increased with increasing concentrate supplementation, while the abundance of Rikenellaceae_RC9_gut_ group decreased. Dietary concentrate supplementation altered the concentration and metabolic mode of metabolites in the rumen, significantly affecting the concentration of metabolites involved in amino acid and derivative metabolism (e.g., L-aspartic acid, L-glutamate, and L-histidine), purine metabolism (e.g., guanine, guanosine, and hypoxanthine), and glycerophospholipid metabolism (e.g., phosphatidate, phosphatidylcholine, and phosphocholine), and other metabolic pathways. The strong correlation between yak rumen microorganisms and metabolites provided a more comprehensive understanding of microbial community composition and function. This study showed significant changes in the composition and abundance of bacteria and metabolites in the rumen of cool season grazing yaks fed with concentrate supplements. Changes in ruminal fermentation parameters and metabolite concentration also showed a strong correlation with ruminal bacterial communities. These findings will be helpful to formulate supplementary feeding strategies for grazing yaks in the cold season from the perspective of intestinal microorganisms.

This work provides an overview on micromirror arrays based on different material systems such as dielectrics, element silicon, compound semiconductors, metals, and novel 2D materials. The goal is to work out the particular strength of each material system to enable optimum performance for various applications. In particular, this review is intended to draw attention to the fact that MEMS micro-mirrors can be successful in many other material systems besides silicon. In particular, the review is intended to draw attention to two material systems that have so far been used less for MEMS micromirror arrays, that have been less researched, and of which fewer applications have been reported to date: metallic heterostructures and 2D materials. However, the main focus is on metallic MEMS micromirror arrays on glass substrates for applications like personalized light steering in buildings via active windows, energy management, active laser safety goggles, interference microscopy, and endoscopy. Finally, the different micromirror arrays are compared with respect to fabrication challenges, switching speed, number of mirrors, mirror dimensions, array sizes, miniaturization potential for individual mirrors, reliability, lifetime, and hinge methodology.

Abstract Background Tenderness is the main quality of meat products. However, the meat tenderness formation is a complex biological process, and pathways and proteins that affect the tenderness of yak meat are unknown. Methods Label‐free proteomics method was used to explore the effects of differentially expressed proteins on the tenderness of yak skeletal muscle (tenderloin) during post‐mortem storage (0, 3, and 7 days) at 3 ± 1°C. Results The tenderness of yak skeletal muscle improved significantly during storage. A total of 91 differentially expressed proteins of yak skeletal muscle during post‐mortem storage were identified by the following comparisons: day 3 versus 0, day 7 versus 0, and day 7 versus 3. NDUFS6, CYCS, COX6A2, LDB3, HSPB7, TPM4, TAGLN, COL1A1, LUM, MYH11, ACTC1, and MYOZ1 proteins showed a significant difference during yak skeletal muscle post‐mortem storage. Furthermore, bioinformatics analyses revealed that the identified proteins were related to carbon metabolism, citrate cycle, glycolysis, oxidative phosphorylation, and RNA degradation. Conclusion The results of the present study could provide proteomic insights into changes in yak skeletal muscle tenderness during storage and may be a valuable resource for future investigations.

Recently, the application of high performance concrete (HPC) is popular in inner lining of frozen shaft in water-rich stratum. The circular crack of inner lining concrete easily appears in the early age (0∼240 h) leading to water burst accidents in the service period. The early-age cracking potential of HPC in inner lining is investigated by Temperature Stress Test Machine (TSTM) in the laboratory. The complex boundary, temperature history and variable restraint degree, was performed simultaneously by TSTM in this study. The experimental analysis indicated that (1) The range of restraint degree is almost 40% to 89.3% under steel frame restrained condition which can almost realize actual constraint degree of inner lining; (2) The cracking potential increased slightly for Mixture C60, C70, and increased severely once the Mixture strength exceeded C70; (3) The integrated crack indicators of C60, C70 and C90 repeat such as cracking stress, crack age, cracking temperature and cracking thermal drop were fluctuant slightly with the concrete strength increased; (4) The increasing of concrete strength decreased the cracking potential of inner lining concrete under steel frame restrained condition, though the tensile strength is improved.

IntroductionManganese (Mn) is an essential trace element for livestock, but little is known about the optimal Mn source and level for yak.MethodsTo improve yak’s feeding standards, a 48-h in vitro study was designed to examine the effect of supplementary Mn sources including Mn sulfate (MnSO4), Mn chloride (MnCl2), and Mn methionine (Met-Mn) at five Mn levels, namely 35 mg/kg, 40 mg/kg, 50 mg/kg, 60 mg/kg, and 70 mg/kg dry matter (includes Mn in substrates), on yak’s rumen fermentation.ResultsResults showed that Met-Mn groups showed higher acetate (p < 0.05), propionate, total volatile fatty acids (p < 0.05) levels, ammonia nitrogen concentration (p < 0.05), dry matter digestibility (DMD), and amylase activities (p < 0.05) compared to MnSO4 and MnCl2 groups. DMD (p < 0.05), amylase activities, and trypsin activities (p < 0.05) all increased firstly and then decreased with the increase of Mn level and reached high values at 40–50 mg/kg Mn levels. Cellulase activities showed high values (p < 0.05) at 50–70 mg/kg Mn levels. Microbial protein contents (p < 0.05) and lipase activities of Mn-Met groups were higher than those of MnSO4 and MnCl2 groups at 40–50 mg/kg Mn levels.DiscussionTherefore, Mn-met was the best Mn source, and 40 to 50 mg/kg was the best Mn level for rumen fermentation of yaks.

Teacher collaboration has gradually become a common idea in the reform of teacher education all over the world. However, up to now, the current situation of teacher collaboration is still isolated and conservative to a certain extent. How to avoid being mere formality and promote the development of teachers’ cooperative practice effectively has become an urgent problem for educational researchers and practitioners. Based on the data of TALIS 2018, this study focuses on the impact of teacher cooperation on teaching practice in China and the United Kingdom, with teacher self-efficacy playing the mediating role. Above all, SPSS 23.0 was used for data preparation, descriptive statistics, and Pearson product difference correlation analysis. Then, maximum likelihood estimation and robust standard deviation (MLD) were applied to estimate confirmatory factor analysis and structural equation model through Mplus 7.4. In addition, bootstrap self-help sampling method was adopted to test the mediating effect. About the direct effects of teacher collaboration on teaching practices, the results indicated that exchange and co-ordination was positively related to clarity of instruction in both China and England while professional collaboration was not significantly related to clarity of instruction in the two countries. In terms of the mediating role of self-efficacy between teacher collaboration and teaching practices, the study revealed the indirect effects of professional collaboration on three sub-scales of teaching practices exist via teacher self-efficacy in both China and England. Theoretical and managerial implications regarding using teacher self-efficacy as a mediating variable to examine the effect of common collaboration forms on specific teaching practices were discussed. Limitations and further research were also offered.

The design of dynamic well killing parameters lacks theoretical support because the U-tube principle is not applicable while drilling bit is not at the well bottom. We present the Y-tube principle and establish a gas and liquid two-phase flow model with the killing fluid backflow and zero net liquid flow theory in wellbore below the drill bit. A dynamic well killing theory is proposed, and the variations of the well killing parameters are revealed. The results were as follows. (1) The Y-tube was composed of an annular part above the bit, a drillstring part, and a wellbore part below the bit. Gas–liquid flow models in those three parts were established to accurately simulate the dynamic well killing process when the drill bit was off-bottom. (2) Based on these models, the dynamic inflow and outflow curves during the killing period were obtained. The variations of the dynamic well killing parameters were analyzed by comparing the two curves. (3) With the increase of pressure, the critical gas velocity of blowout decreases, while the temperature, density and viscosity of the killing fluid do the opposite. With the increase of temperature, density and viscosity, and the decrease of the wellbore diameter, the bottom hole pressure increases. And it decreases with the decrease of the distance between the bit and wellhead. (4) Considering the actual well killing conditions, it is recommended to increase the critical blowout velocity by increasing the killing fluid surface tension and density, and to change the killing fluid density and viscosity so that the dynamic outflow curve is always above the dynamic inflow curve or the dynamic outflow curve is always above the formation pressure. As a result, the dynamic well killing will be successful when the bit is not at the well bottom.

Abstract Endometrioid Endometrial Cancer (EEC) is the main subtype of endometrial cancer. In our study, we demonstrated that SPTBN2 was significantly overexpressed in EEC tissues. Upregulated SPTBN2 expression was positively associated with poor prognosis. In addition, we testified that SPTBN2 knockdown significantly inhibited the proliferation, migration, and invasion of EEC cells. Moreover, we found SPTBN2 could interact with CLDN4 to promote endometrial cancer metastasis via PI3K/AKT pathway. Then we further demonstrated that CLDN4 is upregulated in EEC and promotes EEC metastasis. CLDN4 overexpression could partially reversed the decrease in cell migration and invasion caused by SPTBN2 downregulation. In addition, we confirmed that SPTBN2 was a target of miR-424-5p, which plays a tumor suppressor in endometrial cancer. Rescue experiments showed that inhibition of SPTBN2 could partially reverse the effect of miR-424-5p in EEC. In conclusion, we demonstrated that by acting as a significant target of miR-424-5p, SPTBN2 could interact with CLDN4 to promote endometrial cancer metastasis via PI3K/AKT pathway in EEC. Our study revealed the prognostic and metastatic effects of SPTBN2 in EEC, suggesting that SPTBN2 could serve as a prognostic biomarker and a target for metastasis therapy.

With diversification of yak breeding, it is important to understand the effects of feed type on the rumen, especially microbiota and metabolites. Due to the unique characteristics of yak, research on rumen microbes and metabolites is limited. In this study, the effects of two diet types on rumen eukaryotic microflora and metabolites were evaluated using the Illumina MiSeq platform and liquid chromatography-mass spectrometry (LC-MS). All identified protozoa belonged to Trichostomatia. At the genus level, the relative abundance of Metadinium and Eudiplodinium were significantly (p < 0.05) higher in the roughage group than that of concentrate group, while the concentrate group harbored more Isotricha. Ascomycota, Basidiomycota, and Neocallimastigomycota were the main fungal phyla, and the Wallemia, Chordomyces, Chrysosporium, Cladosporium, Scopulariopsis, and Acremonium genera were significantly (p < 0.05) more abundant in the roughage group than the concentrate group, while the concentrate group harbored more Aspergillus, Neocallimastix, Thermoascus, and Cystofilobasidium (p < 0.05). Metabolomics analysis showed that feed type significantly affected the metabolites of rumen protein digestion and absorption (L-proline, L-phenylalanine, L-tryosine, L-leucine, L-tryptophan, and β-alanine), purine metabolism (hypoxanthine, xanthine, guanine, guanosine, adenosine, and adenine), and other metabolic pathway. Correlation analysis revealed extensive associations between differential microorganisms and important metabolites. The results provide a basis for comprehensively understanding the effects of feed types on rumen microorganisms and metabolites of yaks. The findings also provide a reference and new directions for future research.

Background and ObjectivesNeoadjuvant therapy plays an increasingly important role in pancreatic neuroendocrine neoplasms (pNENs), but the systematic evaluation of its efficacy is still lacking. The purpose of this study is to explore the role of neoadjuvant therapy in pNENs.MethodsWe systematically reviewed the literatures published online until October 1, 2021. Meta-analysis was conducted to generate proportion with 95% confidence intervals (95% CI) for tumor response, resection rate, R0 resection rate and survival time.ResultsNine studies with 468 patients were involved in the systematic review. None of these patients met complete response (CR). Furthermore, 43.6% (95% CI [18.1, 69.0]) patients were expected to achieve partial response (PR), 51.3% (95% CI [27.9, 78.3]) to stable disease (SD), and 4.3% (95% CI [0.7, 7.9]) to progressive disease (PD). The estimate resection rate and R0 resection rate after neoadjuvant therapy were 68.2% (95% CI [44.5, 91.9]) and 60.2% (95% CI [53.5, 66.9]), respectively. There was no significant difference in resection rate between different chemotherapy regimens (41.67% vs 33.93%, P=0.453), as well as R0 resection rate (62.50% vs 68.30%, P=0.605). In terms of objective response rate (ORR), there was no significant difference between CAPTEM and FAS (41.67% vs 33.93%, P=0.453), while PRRT showed a higher ORR compared with chemotherapy, although there was also no statistical difference (49.06% vs 36.96%, P=0.154).ConclusionNeoadjuvant therapies could reduce the tumor size and stage of some borderline resectable or unresectable pNENs, and give some patients the chance of radical resection. However, according to the current data, the best treatment regimen for pNENs neoadjuvant therapy is still unknown.

Copper, manganese, and iodine are part of a yak’s required trace elements. However, knowledge about their dietary requirements is scarce. Therefore, an experiment was conducted to evaluate rumen fermentation, blood parameters, and growth performance and screen out the optimum levels of trace elements in yaks’ diet. Here, 18 three-year-old castrated yaks were randomly divided into four groups, which fed with diets containing basal (CON: 4.40, 33.82, and 0 mg/kg) and low-level (LL: 10.00, 40.00, and 0.30 mg/kg), middle-level (ML: 15.00, 50.00, and 0.50 mg/kg), and high-level (HL: 20.00, 60.00, and 0.70 mg/kg) copper, manganese, and iodine for 30 days. With the increase in trace elements, yaks’ daily weight gain (DWG), rumen pH, ammonia nitrogen, microbial protein (MCP), and volatile fatty acids levels and serum triglycerides and urea nitrogen levels showed firstly increasing and then decreasing trends and reached the highest values in ML, and serum ceruloplasmin and total superoxide dismutase (T-SOD) activities showed continuously increasing trends. Yaks’ DWG, rumen MCP, butyrate, and valerate levels and serum triglycerides, urea nitrogen, ceruloplasmin, and T-SOD levels in ML were significantly higher than CON. Therefore, the recommended levels of copper, manganese, and iodine in growing yaks’ diet are 15.00, 50.00, and 0.50 mg/kg (ML), respectively.

A relatively stable microbial ecological balance system in the rumen plays an important role in rumen environment stability and ruminant health maintenance. No studies have reported how rumen fluid transplantation (RFT) affects the composition of rumen microorganisms and yak growth performance. In this experiment, we transplanted fresh rumen fluid adapted to house-feeding yaks to yaks transitioned from natural pastures to house-feeding periods to investigate the effects of rumen fluid transplantation on rumen microbial community regulation and production performance. Twenty yaks were randomly divided into the control group (CON; n = 10) and the rumen fluid transplantation group (RT; n = 10). Ten yaks that had been adapted to stall fattening feed in one month were selected as the rumen fluid donor group to provide fresh rumen fluid. Ruminal fluid transplantation trials were conducted on the 1st, 3rd, and 5th. Overall, 1 L of ruminal fluid was transplanted to each yak in the RT and CON group. The formal trial then began with both groups fed the same diet. After this, growth performance was measured, rumen fluid was collected, and rumen microbial composition was compared using 16s rRNA sequencing data. The results showed that rumen fluid transplantation had no significant effect on yak total weight gain or daily weight gain (p > 0.05), and feed efficiency was higher in the RT group than in the CON group at 3 months (treatment × month: p < 0.01). Ruminal fluid transplantation significantly affected rumen alpha diversity (p < 0.05). Up to day 60, the RT group had significantly higher OTU numbers, Shannon diversity, and Simpson homogeneity than the CON group. Principal coordinate analysis showed that the rumen microbiota differed significantly on days 4 and 7 (p < 0.05). Bacteroidota, Firmicutes, Proteobacteria, and Spirochaetes were the most abundant phyla in the rumen. The relative abundances of Bacteroidota, Proteobacteria, and Spirochaetes were lower in the RT group than in the CON group, with a decrease observed in Bacteroidota in the RT group on days 7 and 28 after rumen fluid transplantation (p = 0.013), while Proteobacteria showed a decreasing trend in the CON group and an increasing trend in RT; however, this was only at day 4 (p = 0.019). The relative abundance of Firmicutes was significantly higher in the RT group than in the CON group on days 4, 7, and 28 (p = 0.001). Prevotella and Rikenellaceae_RC9_gut_group were the predominant genera. In conclusion, our findings suggest that rumen fluid transplantation improves yak growth performance and rumen microbial reshaping. The findings of this study provide new insights into yak microbial community transplantation and a reference for improving feed efficiency in the yak industry.

As a widely used material in underground engineering, clay–cement slurry grouting is known for its initial poor anti-seepage and filtration capacity, the low strength of the resulting stone body, and its tendency towards brittle failure. To explore efficient and environmentally friendly grouting materials, industrial-grade graphene oxide (GO) was incorporated into a clay–cement slurry to create a new type of slurry called a GO composite. These GO composites were then utilized to reinforce fractured formations. Uniaxial compression tests, shear strength tests, permeability tests, and electron microscopy scans were conducted to investigate the strength, permeability, and microscopic features of the GO composite-reinforced fractured formations. Furthermore, the optimization effect and application prospects of graphene oxide on clay–cement slurry materials were evaluated. The experimental results demonstrated that the modified slurry effectively improved the compressive strength (increased by 7.2% to 32.5%) and shear strength (increased by 28.6% to 105.3%) of consolidated fractured gravel. By conducting orthogonal experiments with range analysis, variance analysis, and multiple regression analysis, it was shown that there was a strong correlation between the consolidated body and three factors influencing the permeability coefficient. Among these factors, the OPC content had the most significant impact on the permeability coefficient, followed by the GO content. Graphene oxide was found to promote cement hydration reactions, guide the growth of hydration products on the surface of graphene oxide nanosheets, optimize the pore structure in grouting materials, and reduce microcracks between the slurry and the fractured gravel interface. Electron microscopy characterization and fractal analysis revealed that the addition of graphene oxide effectively reduced the degree of microdamage during the sample’s failure process. This ensured the integrity of the sample during the unstable failure process, enhanced the material’s toughness, and improved its ability to resist loads.

The increasing prevalence of obesity worldwide is associated with an increased risk of various diseases, including multiple metabolic diseases, cardiovascular diseases, and malignant tumors. Fatty acid binding proteins (FABPs) are members of the adipokine family of multifunctional proteins that are related to fatty acid metabolism and are divided into 12 types according to their tissue origin. FABP4 is mainly secreted by adipocytes and macrophages. Under obesity, the synthesis of FABP4 increases, and the FABP4 content is higher not only in tissues but also in the blood, which promotes the occurrence and development of various cancers. Here, we comprehensively investigated obesity epidemiology and the biological mechanisms associated with the functions of FABP4 that may explain this effect. In this review, we explore the molecular mechanisms by which FABP4 promotes carcinoma development and the interaction between fat and cancer cells in obese circumstances here. This review leads us to understand how FABP4 signaling is involved in obesity-associated tumors, which could increase the potential for advancing novel therapeutic strategies and molecular targets for the systematic treatment of malignant tumors.

Gel‐polymer electrolytes (GPEs) are expected to play an important role in the next‐generation quasi‐solid‐state lithium batteries (SSLBs), but they suffer from severe thermal and mechanical instability. Herein, a viscoelastic nanofibrous GPE film with high thermomechanical stability for constructing stable quasi‐SSLBs is reported. The GPE is fabricated by in situ polymerization of methyl acrylate in a blockcopolymer (PVDF‐b‐PTFE) nanofiber film followed by being immersed into the commercial liquid electrolytes and then dried. The tough blockcopolymer film shows high polarity that not only enhances the thermomechanical stability, but also promotes the dissociation of Li salts to release more free ions, thereby improving the ionic conductivity of GPE. As a result, the GPE shows high heat resistant to 150 °C, a wide electrochemical window of 5.1 V, and a high Li+‐ion transference number of 0.56. When matching high loading cathodes of LiFePO4 (13.36 mg cm−2) or NCM523 (12.25 mg cm−2), both kinds of batteries present high cycling stability and rate capability at 25 °C. More practically, the GPE can be used to construct soft pack batteries with high capacities, showing appealing application prospects.

Changes in dietary composition affect the rumen microbiota in ruminants. However, information on the effects of dietary concentrate-to-forage ratio changes on yak rumen bacteria and metabolites is limited. This study characterized the effect of three different dietary concentrate-to-forage ratios (50:50, C50 group; 65:35, C65 group; 80:20, C80 group) on yak rumen fluid microbiota and metabolites using 16S rRNA gene sequencing and liquid chromatography-mass spectrometry (LC-MS) analyses. Rumen fermentation parameters and the abundance of rumen bacteria were affected by changes in the dietary concentrate-to-forage ratio, and there was a strong correlation between them. At the genus level, higher relative abundances of norank_f__F082, NK4A214_group, Lachnospiraceae_NK3A20_group, Acetitomaculum, and norank_f__norank_o__Clostridia_UCG-014 were observed with a high dietary concentrate-to-forage ratio (P < 0.05). Combined metabolomic and enrichment analyses showed that changes in the dietary concentrate-to-forage ratio significantly affected rumen metabolites related to amino acid metabolism, protein digestion and absorption, carbohydrate metabolism, lipid metabolism, and purine metabolism. Compared with the C50 group, 3-methylindole, pantothenic acid, D-pantothenic acid, and 20-hydroxy-leukotriene E4 were downregulated in the C65 group, while spermine and ribose 1-phosphate were upregulated. Compared to the C50 group, Xanthurenic acid, tyramine, ascorbic acid, D-glucuronic acid, 6-keto-prostaglandin F1a, lipoxin B4, and deoxyadenosine monophosphate were upregulated in the C80 group, while 3-methylindole and 20-hydroxy-leukotriene E4 were downregulated. All metabolites (Xanthurenic acid, L-Valine, N-Acetyl-L-glutamate 5-semialdehyde, N-Acetyl-L-glutamic acid, Tyramine, 6-Keto-prostaglandin F1a, Lipoxin B4, Xanthosine, Thymine, Deoxyinosine, and Uric acid) were upregulated in the C80 group compared with the C65 group. Correlation analysis of microorganisms and metabolites provided new insights into the function of rumen bacteria, as well as a theoretical basis for formulating more scientifically appropriate feeding strategies for yak.

Abstract Dental caries is among the most prevalent dental diseases globally, which arises from the formation of microbial biofilm on teeth. Besides, tooth whitening represents one of the fastest‐growing areas of cosmetic dentistry. It will thus be great if tooth biofilm eradication can be combined with tooth whitening. Herein, a highly efficient photodynamic dental therapy strategy is reported for tooth biofilm eradication and tooth discoloration by employing a photosensitizer (DTTPB) with aggregation‐induced emission characteristics. DTTPB can efficiently inactivate S. mutans, and inhibit biofilm formation by suppressing the expression of genes associated with extracellular polymeric substance synthesis, bacterial adhesion, and superoxide reduction. Its inhibition performance can be further enhanced through combined treatment with chlorhexidine. Besides, DTTPB exhibits an excellent tooth‐discoloration effect on both colored saliva‐coated hydroxyapatite and clinical teeth, with short treatment time (less than 1 h), better tooth‐whitening performance than 30% hydrogen peroxide, and almost no damage to the teeth. DTTPB also demonstrates excellent biocompatibility with neglectable hemolysis effect on mouse red blood cells and almost no killing effect on mammalian cells, which enables its potential applications for simultaneous tooth biofilm eradication and tooth whitening in clinical dentistry.

There is little information available on milk intake and energy and nitrogen requirements of growing yak calves. This study aimed to fill this important gap, as this information could be beneficial in designing a system to wean yak calves earlier than in natural time. We determined the average daily gain and energy and nitrogen balances and requirements of 4-month-old female yak calves (48.8 ± 2.45 kg, n = 8). The calves were allowed to suck once a day and were fed an ad libitum concentrate: hay diet at a ratio of 60:40. Milk intake averaged 540 ± 26 g/d, yielding 2.28 ± 0.112 MJ/d, which was 13% of the gross energy intake (GEI). The digestible energy intake (DEI):GEI ratio was 0.681, metabolizable energy intake (MEI):DEI was 0.913, and MEI:GEI was 0.621. The average daily gain of the calves was 433 ± 153.1 g/d, which consisted of 78.0 ± 8.99 g protein, 52.7 ± 23.74 g fat, and 302.3 ± 95.1 g water, that is, 18.0% protein, 13.0% fat and 69.8% water. There were 130.7 g of body solids and 9.06 MJ of energy in every kg of body mass gain. Of the MEI, 25.17 kJ were required for 1 g of body mass, 83.40 kJ for 1 g of body solids, and 2.62 kJ for 1 kJ of retained energy (RE), and RE was 36.6% of MEI. The maintenance energy requirement was 5.35 MJ/d, the efficiency of utilization of energy for growth (kg) was 0.72, and the heat increment of feeding for growth was 0.28 (1.55 MJ/d). Digestible nitrogen (N) was 0.685 while retained N (RN) was 0.489 of N intake. The N requirement for maintenance was 11.73 g/d or 0.61 g N/kg0.75 per day, while the biological value (BV) of N was 91.1%. The energy and N requirements for yak calves were relatively low, which could be explained, at least in part, by the high efficiency of utilization of energy and high BV of N when compared to other livestock. These findings could be beneficial in designing early weaning systems for the many Himalayan households depending on yak production for their livelihoods.

Dual-gradient drilling technology is being increasingly used in formations with narrow pressure margins. For dual-gradient drilling based on downhole separation, hollow spheres are separated into the annulus at the separator position, resulting in variable mass flow in the wellbore. Thus, existing heat transfer models are no longer suitable for describing wellbore temperature profiles in dual-gradient drilling. This study focused on developing a wellbore heat transfer model that fully considers separated hollow spheres entering the annulus, complex casing programs, and heat sources, for dual-gradient drilling based on downhole separation. The model was solved using an iterative method. Then, the accuracy of the model was verified using temperature data measured from two wells. Finally, the difference in the annular temperature distributions between dual-gradient drilling and conventional single-gradient drilling were investigated, as were the wellbore heat transfer characteristics for dual-gradient drilling. The following major conclusions were drawn: (1) for dual-gradient drilling based on downhole separation, at the separator location, the annular fluid temperature does not decrease, but rather increase in the flow direction because of the inflow of hollow spheres; (2) a clear inflection point exists in the annular fluid temperature curve at the location where the separator would be; (3) the magnitude of the mutation of the temperature curve at the inflection point is considerably affected by the heat capacities of the hollow spheres and the pure drilling fluid; (4) under the same change in separation efficiency, distance between the bit and separator, flow rate, and thermal conductivity of formation, the variation range of the fluid temperature at the bottom hole is greater than that at the wellhead.

According to the 2020 global cancer data released by the World Cancer Research Fund (WCRF) International, gastric cancer (GC) is the fifth most common cancer worldwide, with yearly increasing incidence and the second-highest fatality rate in malignancies. Despite the contemporary ambiguous molecular mechanisms in GC pathogenesis, numerous in-depth studies have demonstrated that zinc finger proteins (ZFPs) are essential for the development and progression of GC. ZFPs are a class of transcription factors with finger-like domains that bind to Zn2+ extensively and participate in gene replication, cell differentiation and tumor development. In this review, we briefly outline the roles, molecular mechanisms and the latest advances in ZFPs in GC, including eight principal aspects, such as cell proliferation, epithelial–mesenchymal transition (EMT), invasion and metastasis, inflammation and immune infiltration, apoptosis, cell cycle, DNA methylation, cancer stem cells (CSCs) and drug resistance. Intriguingly, the myeloid zinc finger 1 (MZF1) possesses reversely dual roles in GC by promoting tumor proliferation or impeding cancer progression via apoptosis. Therefore, a thorough understanding of the molecular mechanism of ZFPs on GC progression will pave the solid way for screening the potentially effective diagnostic indicators, prognostic biomarkers and therapeutic targets of GC.

Abstract Background Ovarian cancer stem cells (OCSC), endowed with tumor-initiating and self-renewal capacity, would account not only for the tumor growth, the peritoneal metastasis, and the relapse, but also for the acquisition of chemotherapy resistance. Nevertheless, figuring out their phenotypical and functional traits has proven quite challenging, mainly because of the heterogeneity of ovarian cancer. A deeper understanding of OCSC mechanisms will shed light on the development of the disease. Therefore, we aim to explore it for the design of innovative treatment regimens which aim at the eradication of ovarian cancer through the elimination of the CSC component. Methods In this study, immunohistochemistry assay and western blot assay were used to detect protein expression in the primary tumor and peritoneal multi-cellular aggregates/spheroids (MCAs/MCSs). OCSCs induced from cell line SKOV3 and HO-8910 were enriched in a serum-free medium (SFM). The effect of EIF5A2 on CSC-like properties was detected by sphere-forming assays, re-differentiation assays, quantitative real-time polymerase chain reaction, western blotting, flow cytometry, cell viability assays, immunofluorescence staining, and in vivo xenograft experiments. RNA-sequencing (RNA-seq) was used to reveal the mechanism by which EIF5A2 positively modulates the stem-like properties of ovarian cancer cells. Results Expression of EIF5A2 was significantly higher in peritoneal MCAs/MCSs compared to matched primary tumors, and EIF5A2 was also unregulated in ovarian cancer cell line-derived spheroids. Knockdown of EIF5A2 reduced the expression of the stem-related markers (ALDH1A1 and OCT-4), inhibited self-renewal ability, improved the sensitivity to chemotherapeutic drugs, and inhibited tumorigenesis in vivo. Mechanistic studies revealed that EIF5A2 knockdown reduced the expression of KLF4, which could partially rescue stem-like properties abolished by EIF5A2 knockdown or strengthened by EIF5A2 overexpression, through the transcription factor E2F1, which directly bind to KLF4 promoter. Conclusion Our results imply that EIF5A2 positively regulates stemness in ovarian cancer cells via E2F1/KLF4 pathway and may serve as a potential target in CSCs-targeted therapy for ovarian cancer.
