Ecosystem multifunctionality means that the ecosystem has the ability to provide multiple functions simultaneously. The study of the ecosystem multifunctionality provides an important basis for the understanding of the ecosystem function and management. Despite the plant community restoration is an important driver of changes in biodiversity and ecosystem multifunctionality, we still little know about the scaling effects the relationship between different dimensions of biodiversity and ecosystem multifunctionality. In this study, we investigated the relative contributions of different dimensions of plant diversity (e.g., species diversity, functional diversity and phylogenetic diversity) changes in ecosystem multifunctionality under different restoration stages (10, 30 and 40 years) in a human-damaged Liaodong oak (Quercus wutaishanica) plant communities in northern China. The results found that (1) ecosystem multifunctionality index was significantly higher in the middle (30 years) and late (40 years) stages of restoration than the early stage (10 years) of restoration. (2) Species richness and phylogenetic diversity were significantly higher in the early stage (10 years) of restoration than in the middle (30 years) and late (40 years) stages of restoration, however, functional dispersion was significantly higher in the later stages (40 years) of restoration than in the early (10 years) and middle stages (30 years) of restoration. (3) Ecosystem multifunctionality is primarily driven by photosynthetic traits of dominant species. The results of this study deepen the under-standing of the relationship between plant diversity and ecosystem multifunctionality in the forests of northern China by considering natural restoration after destruction, and contribute to the conservation of plant diversity and maintenance of ecosystem multifunctionality.

Diamond has superior physical and electronic properties and it is regarded as an ultimate material of power-electronics applications. Numerous studies have been focusing on the diamond-based power devices, especially on diamond metal-oxide-semiconductor field-effect transistors (MOSFETs). Due to the existence of two-dimensional hole gas (2DHG) layer on the hydrogen-terminated (H-) diamond surface, hole carriers with high density can be provided and thus H-diamond MOSFETs have been widely investigated, whereas they generally exhibit normally on properties. In recent years, many researchers have been devoting themselves to the development of diamond MOSFETs with normally off characteristics and good device performances. In this work, we summarized recent advances to achieve normally off features for diamond MOSFETs, including the partial surface modification or using some specific insulator materials on H-diamond surfaces, the oxidation of the silicon terminated (Si-) diamond, and the inversion-type p-channel diamond MOSFETs on hydroxyl terminated (OH-) diamond. In addition, we summarized their interface properties which mainly limited the device performances, and put forward to the possible approaches to improve the device performances and thus to promote their potential for power application.

Mountain apricot (Prunus sibirica) is an important fruit tree variety, and has a wide range of planting and application value in China and even the world. However, the current research on the suitable distribution area of P. sibirica is still inconclusive. In this study, we retrieved distribution data for P. sibirica in China from the Global Biodiversity Information Facility (GBIF), and identified six key environmental factors influencing its distribution through cluster analysis. Using these six selected climate factors and P. sibirica distribution points in China, we applied the maximum entropy model (MaxEnt) to evaluate 1160 candidate models for parameter optimization. The final results predict the potential distribution of P. sibirica under the current climate as well as two future climate scenarios (SSPs126 and SSPs585). This study shows that the model optimized with six key climate factors (AUC = 0.897, TSS = 0.658) outperforms the full model using nineteen climate factors (AUC = 0.894, TSS = 0.592). Under the high-emission scenario (SSPs585), the highly suitable habitat for P. sibirica is expected to gradually shrink towards the southeast and northwest, while expanding in the northeast and southwest. After the 2050s, highly suitable habitats are projected to completely disappear in Shandong, while new suitable areas may emerge in Tibet. Additionally, the total area of suitable habitat is projected to increase in the future, with a more significant expansion under the high-emission scenario (SSPs585) compared to the low-emission scenario (SSPs126) (7.33% vs. 0.16%). Seasonal changes in precipitation are identified as the most influential factor in driving the distribution of P. sibirica.

Beta diversity analysis was conducted to identify factors affecting the beta diversity patterns of grasshoppers and predatory beetles in steppes and the primary environmental drivers of these patterns in Ningxia, northwest China. Data on the insects and environment of 42 steppe monitoring areas in Ningxia were analyzed. A total of 4335 insects of 50 species were captured from 42 monitoring areas, including 743 grasshoppers and 3592 predatory beetles belonging to 27 and 23 species, respectively. Non-metric multidimensional scale (NMDS) indicated a high similarity in the grasshopper communities between temperate meadow steppe (TMS) and temperate steppe (TS). Additionally, there was a high similarity among the predatory beetle communities between temperate desert steppe (TDS) and TS. Redundancy analysis (RDA) showed altitude (AL) and average annual precipitation (PP) could explain the interaction between insects and the environment to a large extent. Among the four steppes, the beta diversity of grasshoppers was predominantly driven by the richness component. In TMS and temperate steppe desertification (TSD), the beta diversity of predatory beetles was dominated by the richness component, whereas in TS and TDS, the beta diversity of predatory beetles was dominated by the turnover component. IntroductionViral diseases have become a vital factor limiting the development of the alfalfa (Medicago sativa) industry. Six viruses infecting alfalfa with a high incidence rate are Alfalfa mosaic virus (AMV), Medicago sativa alphapartitivirus 1 (MsAPV1), Medicago sativa alphapartitivirus 2 (MsAPV2), Medicago sativa deltapartitivirus 1 (MsDPV1), Medicago sativa amalgavirus 1 (MsAV1), and Cnidium vein yellowing virus 1 (CnVYV1). The purpose of this study was to develop preventive measures against these viruses by investigating their transmission through alfalfa seeds.MethodsIn this study, we investigated the transmission rate of alfalfa viruses from seed to seedling by PCR, determined the location of viruses in seed by dissecting seed embryos and seed coat, tracked the changes of viruses in seedlings, and finally discover effective elimination measures for alfalfa viruses from 16 measures.Results and discussionOur results demonstrated that all these six viruses could be transmitted from alfalfa seeds to seedlings with the transmission rate ranging from 44.44% to 88.89%. For AMV, MsAPV2, and MsAV1, the viral load was significantly higher in the seed coats than in the seed embryos; however, it did not show significant differences between these two parts of the seeds for MsAPV1, MsDPV1, and CnVYV1. Dynamic accumulation analysis of AMV and MsAPV2 indicated that the viral load in plants increased continuously in the early growth stage, making it important to inactivate these viruses prior to their seed-to-seedling transmission. Sixteen treatments including physical, chemical, and combinations of physical and chemical measures were compared in terms of their elimination efficiency on AMV and MsAPV2 and impacts on seed germination. The results showed that soaking alfalfa seeds in sterile distilled water for 2h + 2% NaClO for 1h or 2% NaClO for 1h were more promisingly applicable because it could significantly reduce AMV and MsAPV2 particles in both seeds and seedlings. Our data revealed a route of virus transmission in alfalfa and shed light on the discovery of a highly efficient method for the management of alfalfa viral diseases.

Seed dispersal is an important ecological process and has important implications for plant population expansion and regeneration. Seed dispersal not only reduces the probability of death due to seed density but also facilitates seedling establishment. Many studies have focused on the effect of one or two factors on seed dispersal. However, little is known about studies on the effect of multiple factors and their interactions on seed dispersal. Here, we conducted a field experiment to explore how seed size, soil burial, and seed peeling affect the dispersal and hoarding of seeds of Quercus liaotungensis in dispersal animals. We found that large seeds were preferentially selected by animals, and the predation after dispersal, hoarding after dispersal, predation distance after dispersal, and hoarding distance after dispersal of large seeds were significantly greater than small seeds, which is more beneficial to the plant expansion and regeneration. Soil burial increased the time of seed intact in situ, significantly increased predation in situ, and reduced predation after dispersal, predation distance after dispersal, and hoarding distance after dispersal, which is not beneficial to the plant population expansion and regeneration. Seed peeling reduced the time of seed intact in situ, and the predation after dispersal was significantly greater than that of unpeeled seeds, which is not beneficial to the plant population. We did not find the interactions between seed size, soil burial, and seed peeling on dispersal. The effects of a single factor may be more than their interactions between seed size, soil burial and seed peeling on dispersal. These results implied that seed size, soil burial and seed peeling may affect plant population expansion and regeneration by affecting the dispersal and hoarding of animals.

Corn is a major cereal crop, and accurate monitoring of corn planting areas is crucial for agricultural structural adjustments and ensuring food security. This study proposes an improved HRNet network that utilizes the spectral and spatial features of Sentinel-2 to extract synthetic NDVI time series datasets for identifying corn planting plots. The study involves enhancing the HRNet network by integrating the CBAM attention mechanism and FReLU activation function, processing the 2023 corn planting growth period data in the Loess Plateau region of Pengyang County, Ningxia, China. This is achieved through: 1) preprocessing Sentinel-2A data and constructing smoothed time series data, and 2) conducting field data surveys to create training, validation, and testing sets. Subsequently, the improved HRNet network is utilized to extract corn planting plots in the study area, followed by accuracy assessment. The results demonstrate that the proposed method achieves accuracy (Acc), F1 score, and mean Intersection over Union (mIoU) of 91.06%, 90.82%, and 88.58% respectively, outperforming PSPNet, U-Net, and HRNet networks. Furthermore, it is proven that using the NDVI time series dataset for all months can enhance identification accuracy. This research confirms that the proposed method has high potential and applicability in identifying corn planting areas in the Loess Plateau region.

Abstract Pyroptosis, an inflammatory programmed cell death, distinguishes itself from apoptosis and necroptosis and has drawn increasing attention. Recent studies have revealed a correlation between the expression levels of many pyroptosis-related genes and both tumorigenesis and progression. Despite advancements in cancer treatments such as surgery, radiotherapy, chemotherapy, and immunotherapy, the persistent hallmark of cancer enables malignant cells to elude cell death and develop resistance to therapy. Recent findings indicate that pyroptosis can overcome apoptosis resistance amplify treatment-induced tumor cell death. Moreover, pyroptosis triggers antitumor immunity by releasing pro-inflammatory cytokines, augmenting macrophage phagocytosis, and activating cytotoxic T cells and natural killer cells. Additionally, it transforms “cold” tumors into “hot” tumors, thereby enhancing the antitumor effects of various treatments. Consequently, pyroptosis is intricately linked to tumor development and holds promise as an effective strategy for boosting therapeutic efficacy. As the principal executive protein of pyroptosis, the gasdermin family plays a pivotal role in influencing pyroptosis-associated outcomes in tumors and can serve as a regulatory target. This review provides a comprehensive summary of the relationship between pyroptosis and gasdermin family members, discusses their roles in tumor progression and the tumor immune microenvironment, and analyses the underlying therapeutic strategies for tumor treatment based on pyroptotic cell death.

The desert steppe in Ningxia is the largest natural steppe type in the region, characterized by a fragile ecological environment and low carrying capacity. Gravel–sand mulching, a local agricultural practice, involves using a sand and gravel covering layer to maintain soil warmth and moisture. However, this method has led to ecological problems, such as habitat loss and biodiversity loss. Tenebrionidae, one of the most widely distributed beetle families, is commonly used to indicate habitat degradation and is a significant steppe pest in desert steppe areas. This study, conducted in the gravel–sand mulching areas of Shapotou District, Zhongwei City, Ningxia, classified the habitat loss from low to high in these areas into five grades (I-V) and examined the effects of habitat loss on Tenebrionidae. We collected 6565 Tenebrionidae, with Blaps femoralis, Anatolica nureti, and Pterocoma vittata being the dominant species. The findings reveal that habitat loss grade I had the highest abundance, diversity index, and evenness index of Tenebrionidae, significantly higher than those of grades II–IV. Habitat loss had a significant negative effect on Tenebrionidae abundance, a significant positive effect on the richness index, no significant effect on the vegetation diversity index, a significant positive effect on soil available potassium (APP), and a significant negative effect on soil total phosphorus (TP). Redundancy analysis indicated a positive correlation between Tenebrionidae abundance and the vegetation diversity index; a negative correlation between Tenebrionidae richness, the diversity index, and vegetation indices; a positive correlation between Tenebrionidae abundance and soil TP; and a negative correlation between the Tenebrionidae diversity index and soil TP and soil APP. These findings will contribute to biodiversity conservation and ecological restoration and provide a theoretical basis for steppe management, sustainable agricultural development, and pest monitoring in desert steppe environments.

Moisture is the most important environmental factor limiting seed regeneration of shrubs in desert areas. Therefore, understanding the effects of moisture changes on seed germination, morphological and physiological traits of shrubs is essential for vegetation restoration in desert areas. In March to June 2023, in a greenhouse using the potting method, we tested the effects of soil moisture changes (5%, 10%, 15%, 20% and 25%) on seed germination and seedling growth of six desert shrubs (Zygophyllum xanthoxylum, Nitraria sibirica, Calligonum mongolicum, Corethrodendron scoparium, Caragana korshinskii, and Corethrodendron fruticosu). Results showed that (1) seed germination percent and vigor index were significantly higher at 15 and 20% soil moisture content than at 5 and 10%; (2) shoot length, primary root length, specific leaf area and biomass of seedlings were significantly higher in the 15% and 20% soil moisture content treatments than in the 5% and 10% treatments; (3) superoxide dismutase activity (SOD) and soluble protein content (SP) decreased with decreasing soil water content, while peroxidase activity (POD) and catalase activity (CAT) showed a decreasing and then increasing trend with increasing soil water content; (4) the six seeds and seedling of shrubs were ranked in order of their survivability in response to changes in soil moisture: Caragana korshinskii > Zygophyllum xanthoxylum > Calligonum mongolicum > Corethrodendron scoparium > Corethrodendron fruticosu > Nitraria sibirica. Our study shows that shrub seedlings respond to water changes by regulating morphological and physiological traits together. More importantly, we found that C. korshinskii, Z. xanthoxylum and C. mongolicum were more survivable when coping with water deficit or extreme precipitation. The results of the study may provide a reference for the selection and cultivation of similar shrubs in desert areas under frequent extreme droughts in the future.

Arthropods play a crucial role in ecological processes and agricultural productivity. Soil physicochemical properties, indicators of soil health, are closely linked to arthropod communities. Gravel-sand mulching, commonly employed in arid farming, initially enhances water retention and temperature regulation but may contribute to land degradation with prolonged use. This study investigated how varying tillage durations affected soil properties and arthropod diversity under gravel-sand mulching. The analysis employed multiple comparison methods, covariance analysis (ANCOVA), non-metric multidimensional scaling (NMDS), and redundancy analysis (RDA). The results indicated that while soil fertility was better preserved in cultivated fields compared to in the desert grassland, arthropod diversity significantly decreased with longer cultivation periods. A total of 1099 arthropods from 79 species were sampled, by Barber trap. The highest diversity was observed in native grassland (NG), with 305 arthropods from 39 species, while tillage 21 years (GPS-21Y) exhibited the lowest diversity, with only 103 arthropods from 6 species. Dominant species included the carnivores Labidura japonica and Cataglyphis aenes. The analysis revealed low similarity in arthropod communities between GPS-21Y and other fields and high similarity in soil physicochemical properties between NG and the transition zone (STZ). RDA showed available potassium (APP) was negatively correlated with arthropod species diversity and concentration, total Nitrogen (TN) was positively correlated with arthropod species diversity but negatively correlated with species concentration, total phosphorus (TP) was negatively correlated with arthropod species diversity and concentration. This study provides insights into the relationship between maintaining soil fertility and supporting arthropod diversity in grassland agriculture. While soil fertility and arthropod diversity were correlated, continuous cropping practices negatively impacted arthropod diversity, offering valuable information for pest management and sustainable agricultural practices.

This study employs an innovative dynamic switching test system to investigate the dynamic switching characteristics of three p-GaN HEMT devices. The dynamic switching characteristics are different from the previous research on the dynamic resistance characteristics of GaN devices, and the stability of GaN devices can be analyzed from the perspective of switching characteristics. Based on the theory of dynamic changes in threshold opening voltage and capacitance caused by electrical stress, the mechanism of dynamic switching characteristics of GaN HEMT devices is studied and analyzed in detail. The test results have shown that electrical stress induces trap ionization within the device, resulting in fluctuations in electric potential and ultimately leading to alterations in two critical factors of the dynamic switching characteristics of GaN HEMT devices, the parasitic capacitance and the threshold voltage. The dynamic changes in capacitance before and after electrical stress vary among devices, resulting in different dynamic switching characteristics. The test system is capable of extracting the switching waveform for visual comparison and quantitatively calculating the changes in switching parameters before and after electrical stressing. This test provides a prediction for the drift of switch parameters, offering pre-guidance for the robustness of the optimized application scheme.

Grasshoppers are important elements for supporting ecosystem services which make it important to understand their spatial distribution characteristics to reduce outbreaks, implement effective control while minimizing non-target impacts. The spatial software package Geographical Detector (GD) was used to rank the interaction and effects of 19 environmental factors on the distribution of grasshopper species were quantified in four types of steppes in Northwestern, China. Results showed that the distribution of the insect was mainly influenced by elevation (q = 0.44). Among the factor interactions, elevation ∩ average annual precipitation (q = 0.795) was the strongest, followed by elevation ∩ soil available nitrogen (q = 0.722). Temperate meadow steppe and temperate steppe were identified as the areas at greatest risk for grasshopper’s occurrence. This research shows that the Geographical Detector could be provided a useful tool to identify the driving forces for grasshopper distribution. Our research also offers a new method to more broadly understand the interactions between different drivers to explain the distribution of other insects.

Silicon nanowire field effect (SiNW-FET) biosensors have been successfully used in the detection of nucleic acids, proteins and other molecules owing to their advantages of ultra-high sensitivity, high specificity, and label-free and immediate response. However, the presence of the Debye shielding effect in semiconductor devices severely reduces their detection sensitivity. In this paper, a three-dimensional stacked silicon nanosheet FET (3D-SiNS-FET) biosensor was studied for the high-sensitivity detection of nucleic acids. Based on the mainstream Gate-All-Around (GAA) fenestration process, a three-dimensional stacked structure with an 8 nm cavity spacing was designed and prepared, allowing modification of probe molecules within the stacked cavities. Furthermore, the advantage of the three-dimensional space can realize the upper and lower complementary detection, which can overcome the Debye shielding effect and realize high-sensitivity Point of Care Testing (POCT) at high ionic strength. The experimental results show that the minimum detection limit for 12-base DNA (4 nM) at 1 × PBS is less than 10 zM, and at a high concentration of 1 µM DNA, the sensitivity of the 3D-SiNS-FET is approximately 10 times higher than that of the planar devices. This indicates that our device provides distinct advantages for detection, showing promise for future biosensor applications in clinical settings.

Objective To evaluate the clinical outcomes of volumetric modulated arc therapy (VMAT) followed by brachytherapy (BT), combined with chemotherapy, and local hyperthermia (HT) on locally advanced cervical cancer (LACC). Methods In total, 40 patients with FIGO stage IB1-IVB cervical cancer from January 2016 to December 2018 were selectively enrolled in this study. All patients were treated with VMAT (50.4 Gy/1.8 Gy/28 f) concurrent with cisplatin-based chemotherapy (40 mg/m 2 , q1w, 6 cycles) and local HT (40.5-41°C for 60 min, BIW). BT (30-36 y/5-6 f, 2 f/w) was conducted after VMAT. Objective response rate (ORR), local control (LC) time, LC rate, progression-free survival (PFS) rate, cancer-specific survival (CSS) rate, overall survival (OS), median time to tumor progression and treatment-related toxicity were evaluated. Results The median follow-up time was 31 months (8-48). The ORR was 100% at 3 months after treatment and 92.1% at 6 months, respectively. The 1-year, 2-year, and 3-year LC rates were 87.4%, 81.9%, and 70.9%, respectively. The average LC time was 31.50 ± 1.89 months (95% CI 27.79-35.21). The 1-year, 2-year, and 3-year PFS rates were 75.85%, 61.2%, and 51.3%, respectively, while the median PFS was 27.07 months. The 1-year, 2-year, and 3-year OS rates were 95%, 84%, and 79.6%, respectively. In total, 12(30%) patients had grade 3/4 bone marrow suppression. One patient had grade 4 leukopenia. In total, 17 patients had grade 1/2 bone marrow suppression. Two patients had grade 3 nausea and grade 3 vomiting reaction, respectively. No grade 3/4 proctitis and bladder reaction were observed. In the late period of treatment, 1 patient had a rectal hemorrhage. In total, 13 patients had vaginal stenosis. Conclusion VMAT concurrent with chemotherapy, BT, and local HT had a favorable short-term efficacy and acceptable toxicity on cervical cancer, which was an alternative option for LACC.

Abstract Exosomes are highly important in clinical diagnosis due to their high homology with their parental cells. However, conventional exosome detection methods still face the challenges of expensive equipment, low sensitivity, and complex procedures. Field effect transistors (FETs) are not only the most essential electronic component in the modern microelectronics industry but also show great potential for biomolecule detection owing to the advantages of rapid response, high sensitivity, and label-free detection. In this study, we proposed a Si nanowire field-effect transistor (Si-NW Bio-FET) device chemically modified with specific antibodies for the electrical and label-free detection of exosomes. The Si-NW FETs were fabricated by standard microelectronic processes with 45 nm width nanowires and packaged in a polydimethylsiloxane (PDMS) microfluidic channel. The nanowires were further modified with the specific CD63 antibody to form a Si-NW Bio-FET. The use of the developed Si-NW Bio-FET for the electrical and label-free detection of exosomes was successfully demonstrated with a limit of detection (LOD) of 2159 particles/mL. In contrast to other technologies, in this study, Si-NW Bio-FET provides a unique strategy for directly quantifying and real-time detecting exosomes without labeling, indicating its potential as a tool for the early diagnosis of cancer.

Recent studies have revealed that tumor-associated macrophages are the most abundant stromal cells in the tumor microenvironment and play an important role in tumor initiation and progression. Furthermore, the proportion of macrophages in the tumor microenvironment is associated with the prognosis of patients with cancer. Tumor-associated macrophages can polarize into anti-tumorigenic phenotype (M1) and pro-tumorigenic phenotype (M2) by the stimulation of T-helper 1 and T-helper 2 cells respectively, and then exert opposite effects on tumor progression. Besides, there also is wide communication between tumor-associated macrophages and other immune compositions, such as cytotoxic T cells, regulatory T cells, cancer-associated fibroblasts, neutrophils and so on. Furthermore, the crosstalk between tumor-associated macrophages and other immune cells greatly influences tumor development and treatment outcomes. Notably, many functional molecules and signaling pathways have been found to participate in the interactions between tumor-associated macrophages and other immune cells and can be targeted to regulate tumor progression. Therefore, regulating these interactions and CAR-M therapy are considered to be novel immunotherapeutic pathways for the treatment of malignant tumors. In this review, we summarized the interactions between tumor-associated macrophages and other immune compositions in the tumor microenvironment and the underlying molecular mechanisms and analyzed the possibility to block or eradicate cancer by regulating tumor-associated macrophage-related tumor immune microenvironment.

Acute kidney injury (AKI) is a frequently occurring severe disease with high mortality. Cystatin C (Cys-C), as a biomarker of early kidney failure, can be used to detect and prevent acute renal injury. In this paper, a biosensor based on a silicon nanowire field-effect transistor (SiNW FET) was studied for the quantitative detection of Cys-C. Based on the spacer image transfer (SIT) processes and channel doping optimization for higher sensitivity, a wafer-scale, highly controllable SiNW FET was designed and fabricated with a 13.5 nm SiNW. In order to improve the specificity, Cys-C antibodies were modified on the oxide layer of the SiNW surface by oxygen plasma treatment and silanization. Furthermore, a polydimethylsiloxane (PDMS) microchannel was involved in improving the effectiveness and stability of detection. The experimental results show that the SiNW FET sensors realize the lower limit of detection (LOD) of 0.25 ag/mL and have a good linear correlation in the range of Cys-C concentration from 1 ag/mL to 10 pg/mL, exhibiting its great potential in the future real-time application.

Background: Understanding the molecular mechanisms driving oncogenic processes in glioma is important in order to develop efficient treatments. Recent studies have proposed gasdermin D (GSDMD) as a newly discovered pyroptosis executive protein associated with tumorigenesis. However, the precise effect of GSDMD on glioma progression remains unknown. Methods: The expression levels of GSDMD in 931 glioma and 1157 normal control tissues were collected. A series of bioinformatic approaches and in vivo and in vitro experiments were used to investigate the roles and mechanisms of GDSMD in glioma. Results: Significant upregulation of GSDMD was detected in glioma tissues compared to normal brain tissues. Patients with glioma and higher GSDMD levels had shorter overall survival, and the Cox regression analysis revealed that GSDMD was an independent risk factor. In addition, upregulation of GSDMD was associated with higher tumor mutation burden and PD-1/PD-L1 expression. Immune infiltration and single-cell analyses indicated that GSDMD was positively associated with an immunosuppressive microenvironment with more infiltrated macrophages and cancer-associated fibroblasts. Furthermore, the in vitro and in vivo experiments revealed that GSDMD knockdown inhibited glioma proliferation, migration, and growth in vivo. Conclusion: Our analyses revealed a relatively comprehensive understanding of the oncogenic role of GSDMD in glioma. GSDMD is a promising prognostic biomarker and a potential target for glioma treatment.

Abstract Regulatory T cells (Tregs) characterized by the expression of the master transcription factor forkhead box protein p3 (Foxp3) suppress anticancer immunity, thereby hindering protective immunosurveillance of tumours and hampering effective antitumour immune responses in tumour-bearing hosts, constitute a current research hotspot in the field. However, Tregs are also essential for the maintenance of the immune tolerance of the body and share many molecular signalling pathways with conventional T cells, including cytotoxic T cells, the primary mediators of tumour immunity. Hence, the inability to specifically target and neutralize Tregs in the tumour microenvironment without globally compromising self-tolerance poses a significant challenge. Here, we review recent advances in characterizing tumour-infiltrating Tregs with a focus on the functional roles of costimulatory and inhibitory receptors in Tregs, evaluate their potential as clinical targets, and systematically summarize their roles in potential treatment strategies. Also, we propose modalities to integrate our increasing knowledge on Tregs phenotype and function for the rational design of checkpoint inhibitor-based combination therapies. Finally, we propose possible treatment strategies that can be used to develop Treg-targeted therapies.

The treatment modality for recurrent cervical cancer (rCC) is limited, and the prognosis of these patients is poor. Seed implantation could be an important component of rCC management in the context of dose boost or salvage therapy after surgery or radiotherapy, which is characterized by a minimally invasive, high local dose, and rapidly does fall, sparing normal tissue. For patients with good performance status and lateral pelvic wall recurrence with an available puncture path, seed implantation was recommended, as well as for selected central pelvic recurrence and extra-pelvic recurrence. The combination of brachytherapy treatment planning system and CT guidance was needed, and three-dimensional printing templates could greatly improve the accuracy, efficiency, and quality of seed implantation to achieve a potential ablative effect and provide an efficient treatment for rCC. However, the recommendations of seed implantation were mainly based on retrospective articles and lack high-quality evidence, and multicenter prospective randomized studies are needed. In this consensus on iodine125 seed implantation for rCC, indication selection, technical process and requirements, dosimetry criteria, radiation protection, combined systemic therapy, and outcomes of seed implantation for rCC are discussed.

Biological invasions represent a natural rapid evolutionary process in which invasive species may present a major threat to biodiversity and ecosystem integrity. Analyzing the genetic structure and demographic history of invaded populations is critical for the effective management of invasive species. The spotted alfalfa aphid (SAA) Therioaphis trifolii is indigenous in the Mediterranean region of Europe and Africa and has invaded China, causing severe damages to the alfalfa industry. However, little is known about its genetic structure and invasion history. In this study, we obtained 167 complete mitochondrial genome sequences from 23 SAA populations across China based on high-throughput sequencing and performed population genetic and phylogenomic analyses. High haplotype diversity and low nucleotide diversity were found in SAA populations in China with distinct genetic structures, i.e., all populations diverged into three phylogenetic lineages. Demographic history analyses showed a recent expansion of the SAA population, consistent with the recent invasion history. Our study indicated that SAA may have invaded through multiple introduction events during commercial trades of alfalfa, although this needs further validation by nuclear markers.

Radioactive iodine-125 (I-125) is the most widely used radioactive sealed source for interstitial permanent brachytherapy (BT). BT has the exceptional ability to deliver extremely high doses that external beam radiotherapy (EBRT) could never achieve within treated lesions, with the added benefit that doses drop off rapidly outside the target lesion by minimizing the exposure of uninvolved surrounding normal tissue. Spurred by multiple biological and technological advances, BT application has experienced substantial alteration over the past few decades. The procedure of I-125 radioactive seed implantation evolved from ultrasound guidance to computed tomography guidance. Compellingly, the creative introduction of 3D-printed individual templates, BT treatment planning systems, and artificial intelligence navigator systems remarkably increased the accuracy of I-125 BT and individualized I-125 ablative radiotherapy. Of note, utilizing I-125 to treat carcinoma in hollow cavity organs was enabled by the utility of self-expandable metal stents (SEMSs). Initially, I-125 BT was only used in the treatment of rare tumors. However, an increasing number of clinical trials upheld the efficacy and safety of I-125 BT in almost all tumors. Therefore, this study aims to summarize the recent advances of I-125 BT in cancer therapy, which cover experimental research to clinical investigations, including the development of novel techniques. This review also raises unanswered questions that may prompt future clinical trials and experimental work.

In this study, the complete mitochondrial genome of Dermestes dimidiatus ab. rosea was characterized using next-generation sequencing, and the phylogenetic relationships of superfamily Bostrichoidea were established. The results showed that the mitochondrial genome of D. dimidiatus ab. rosea was 16,073 bp in size, and it contained thirteen protein-coding genes (PCGs), twenty-two transfer RNA genes (tRNAs), two ribosomal RNA genes (rRNAs), and a control region. The composition of the whole mitochondrial genome of this species was 41.3% A, 13.5% C, 9.3% G, and 35.9% T, which had high A + T content (77.2%). Phylogenetic relationships of the superfamily Bostrichoidea showed that D. dimidiatus ab. rosea and Dermestes tessellatocollis formed in a clade that was a sister group to (Dermestes maculatus + Dermestes frischii), indicating that Dermestidae was a monophyletic group. This is the first report of a complete mitochondrial genome of D. dimidiatus ab. rosea and preliminary study of Bostrichoidea mitochondrial genome, which is of great significance for the molecular identification of this species and the enrichment of mitochondrial genome database.

Alfalfa (Medicago sativa L.) is one of the most important quality forages worldwide and is cultivated throughout China. Alfalfa is susceptible to a variety of viral diseases during its growth, which has caused huge amounts of commercial losses. However, the profile of the alfalfa virus in China remains ambiguous and the viruses transmitted by Odontothrips loti (Haliday), dominant insect pests in alfalfa, are also poorly understood. In the present study, virus diversity was investigated in the primary alfalfa-growing areas in China. A total of 18 alfalfa viruses were identified through RNA-sequencing (RNA-seq) and reverse transcription-polymerase chain reaction (RT-PCR). Two new plant viruses, Medicago sativa virus 1 (MsV1) and Medicago sativa luteovirus 1 (MsLV1), were detected for the first time. Another four viruses, including the Alfalfa ringspot-associated virus (ARaV), Alfalfa virus F (AVF), Alfalfa enamovirus 1 (AEV1), and Alfalfa deltaparitivirus (ADPV), were reported in China for the first time as well. Both Alfalfa mosaic virus (AMV) and Medicago sativa alphapartitivirus 2 (MsAPV2) are the dominant pathogens, with an infection incidence of 91.7–100%, and 74.4–97.2%, respectively. Additionally, O. loti with first- and second-instar nymphs were shown to acquire the AMV within 0.25 h of feeding on a virus-infected alfalfa. Transmission by thrips to healthy alfalfa plants was also demonstrated. Additionally, we clarified the dynamic changes in the AMV in pre-adult stages of O. loti, which indicated that the AMV is propagated in the nymph stage of O. loti. These findings provide valuable information for understanding the alfalfa virome, confirm the role thrips O. loti plays in alfalfa virus transmission, and improve our fundamental knowledge and management of diseases in China.

In this study, we report a pH-responsive hydrogel-modified silicon nanowire field-effect transistor for pH sensing, whose modification is operated by spin coating, and whose performance is characterized by the electrical curve of field-effect transistors. The results show that the hydrogel sensor can measure buffer pH in a repeatable and stable manner in the pH range of 3–13, with a high pH sensitivity of 100 mV/pH. It is considered that the swelling of hydrogel occurring in an aqueous solution varies the dielectric properties of acrylamide hydrogels, causing the abrupt increase in the source-drain current. It is believed that the design of the sensor can provide a promising direction for future biosensing applications utilizing the excellent biocompatibility of hydrogels.

Macrophages are the most important phagocytes in vivo. However, the tumor microenvironment can affect the function and polarization of macrophages and form tumor-associated macrophages (TAMs). Usually, the abundance of TAMs in tumors is closely associated with poor prognosis. Preclinical studies have identified important pathways regulating the infiltration and polarization of TAMs during tumor progression. Furthermore, potential therapeutic strategies targeting TAMs in tumors have been studied, including inhibition of macrophage recruitment to tumors, functional repolarization of TAMs toward an antitumor phenotype, and other therapeutic strategies that elicit macrophage-mediated extracellular phagocytosis and intracellular destruction of cancer cells. Therefore, with the increasing impact of tumor immunotherapy, new antitumor strategies to target TAMs are now being discussed.

Objective: To evaluate the feasibility and safety of high dose rate interstitial brachytherapy (HDR-IB) assisted with 3-dimensional printing individual template (3D-PIT) for central pelvic recurrent gynecologic cancer (CR-GYN). Methods: Totally 32 patients diagnosed with CR-GYN received iridium-192(192Ir) HDR-IB assisted with 3D-PIT that was classified in 2 types(Type I: transvaginal template/ applicator, and Type II: transvaginal combined transperineal template). The prescribed dose to gross tumor volume (GTV) was 10-36 Gy in 2-6 fractions. We rely on a few dosimetric parameters for quality control. The short-term efficacy was evaluated by RECIST v1.1, and the adverse event was evaluated by CTCAE V4.0. Results: The median V100, D100 and D90 of per fraction among all the patients were 88.9%±9.8%, 3.45Gy±0.54 Gy, and 5.79Gy±0.32 Gy, respectively. Dosimetric comparison between preplan and treatment plan of 20/32 patients with Type II 3D-PIT showed no significant difference in GTV volume, V100, D100, D90, conformation index (CI) and homogeneity index (HI). No severe treatment complications occurred. Grade 3 or 4 late toxicities (fistula) were observed in 3 patients (9%). The local response rate (complete remission, CR + partial remission, PR) was 84.4% (27/32) 1 month after completion of treatment. The median time to progression (TTP) was 15.4 months (95% CI 11.3- 19.6 months), 1-year local control (LC) rate were 51.7%. Conclusions: HDR-IB assisted by 3D-PIT was a reliable modality for CR-GYN due to the clinical feasibility and accepted complications.

It is acknowledged that human activities have initiated and increased steppe habitat loss and fragmentation in Northwest China. Carabid beetles are often used as bioindicators of environmental change because they are extremely sensitive to disturbance. We chose 42 landscapes (18 fragmented and 24 continuous) in two steppe types in Ningxia, Northwest China, to examine the influence of steppe fragmentation on carabid beetles. The results showed that the canonical correlation coefficient between carabid beetles and landscape fragmentation was greatest at a 7-km spatial scale in both desert and typical steppes. Furthermore, the response of carabid beetles to steppe fragmentation was species-specific in both desert and typical steppes. Steppe fragmentation in the desert steppe could significantly enhance the richness and abundance of carabid beetles, which contrasts with the typical steppe. Additionally, steppe fragmentation significantly increased the abundance of two dominant carabid beetles in the desert steppe only. Therefore, the effects of steppe fragmentation on carabid beetles were species-dependent and were also mediated by the plant community and microclimate in the different steppes. The redundancy analysis (RDA) indicated that plant richness and cover were important factors affecting carabid diversity. Finally, these results provide experimental evidence and technical data for biodiversity conservation management in the steppes of Northwest China. High sensitivity and reproducibility are highly desirable to a SERS sensor in diverse detection applications. Moreover, it is a great challenge to determine how to promote the target molecules to be more concentrated on the hotspots of the SERS substrate by engineering a surface with switching interfacial wettability. Along these lines, wafer-scale uniformly hydrophobic silicon nanorods arrays (SiNRs) decorated with Au nanoparticles were designed as the SERS substrate. Typically, the SERS substrate was fabricated by enforcing the polystyrene (PS) sphere self-assembly, as well as the plasma etching and the magnetron sputtering techniques. Consequently, the SERS substrate was treated by soaking within a n-dodecyl mercaptan (NDM) solution at different times in order to obtain adjustable wettabilities. By leveraging the electromagnetic enhancement resulted from the Au nanostructures and enrichment effect induced by the hydrophobicity, the SERS substrate is endowed with efficient SERS capabilities. During the detection of malachite green (MG), an ultralow relative standard deviation (RSD) 4.04–6.14% is achieved and the characteristic signal of 1172 cm−1 can be detected as low as 1 ng/mL. The proposed SiNRs’ structure presents outstanding SERS activity with sensitivity and reproducibility rendering thus an ideal candidate for potential application in analytical detection fields.

Abstract Background For the recurrence of head and neck cancer after operation and radiotherapy, the local control of radioactive seed implantation is good, and it has a certain palliative effect. This study aims to investigate the acute and late side effects of a three-dimentional printing non co-planar template (3D–PNCT) for computed tomography (CT)-guided radioactive 125I seed (RIS) implantation in recurrent cancer of the head and neck. Methods Between January 2016 and December 2016, forty-two patients with local recurrent malignant tumors of the head and neck received 3D–PNCT-assisted RIS implantation. The prescribed dose was 110–160 Gy. Preoperative planning design, production of individual guide plates, RIS implantation, postoperative dose evaluation, and follow-up were completed for all patients. Side effects in the skin, mucous membranes, blood and spinal cord were evaluated. Results All patients underwent surgery successfully. Duration of follow-up was 4–14 (median, of 8.5) months. The activity of a single RIS was 0.34–0.7 (median, 0.6) mCi. The number of RIS was 10–126 (median, 34). The number of implantation needles was 4–31 (median, 11). The mean D2cc (dose to the most exposed 2-cc volume) and D0.1cc (dose to the most exposed 0.1-cc volume) of the skin were 24.9 (7.1–85.5) and 47.5 (9.4–167.2), respectively, whereas those of the spinal cord were 8.4 (4.5–33.3) and 14.2 (13.6–63.0), mucosa were 35.1 (4.2–82.8) and 87.0 (6.6–214.1), parotid glands were 16.2 (12.8–19.7) and 29.8 (26.1–33.4) and those of the trachea were 17.9 (2.5–45.9) and 32.7 (3.9–83.9), respectively. No case had an acute reaction of grade ≥ 3. Three cases had a grade-1 skin reaction. Blood toxicity did not occur, nor spinal-cord injury. Xerostomia was not aggravated than that of before brachytherapy. One case had a grade-3 nerve response. Conclusions 3D–PNCT-assisted RIS implantation can provide good accuracy for positioning. For local recurrent malignant tumor of head and neck, there were no obvious adverse reactions.

The top-gated graphene field effect transistor (GFET) with electric-field induced doping polarity conversion has been demonstrated. The polarity of channel conductance in GFET can be transition from p-type to n-type through altering the gate electric field scanning range. Further analysis indicates that this complementary doping is attributed to the charge exchange between graphene and interface trap sites. The oxygen vacancies in Al2O3filmare are considered to be the origin of the trap sites. The trapping–detrapping process, which may be tuned by the electric field across the metal/oxide/graphene gate stack, could lead to the changing of the intrinsic electric property of graphene. This study promises to produce the complementary p- and n-type GFET for logic applications.

Thin film deposition is one of the most important processes in IC manufacturing. In this paper, several typical models and numerical simulation methods for thin film deposition and atomic layer deposition are introduced. Several modeling methods based on the characteristics of atomic layer deposition are introduced, it includes geometric method, cellular automata and multi-scale simulation. The principle of each model and simulation method is explained, and their advantages and disadvantages are analyzed. Finally, the development direction of thin film deposition and atomic layer deposition modeling is prospected, and some modeling ideas are also provided.

In this paper, the poly-Si nanowire (NW) field-effect transistor (FET) sensor arrays were fabricated by adopting low-temperature annealing (600 °C/30 s) and feasible spacer image transfer (SIT) processes for future monolithic three-dimensional integrated circuits (3D-ICs) applications. Compared with other fabrication methods of poly-Si NW sensors, the SIT process exhibits the characteristics of highly uniform poly-Si NW arrays with well-controlled morphology (about 25 nm in width and 35 nm in length). Conventional metal silicide and implantation techniques were introduced to reduce the parasitic resistance of source and drain (SD) and improve the conductivity. Therefore, the obtained sensors exhibit >106 switching ratios and 965 mV/dec subthreshold swing (SS), which exhibits similar results compared with that of SOI Si NW sensors. However, the poly-Si NW FET sensors show the Vth shift as high as about 178 ± 1 mV/pH, which is five times larger than that of the SOI Si NW sensors. The fabricated poly-Si NW sensors with 600 °C/30 s processing temperature and good device performance provide feasibility for future monolithic three-dimensional integrated circuit (3D-IC) applications.

This paper summarizes the status of the plasma etch process modeling research. It mainly introduces typical etching models employing the analytical method, geometric method, system identification method, basic principle simulation method, as well as empirical model. Each model’s basic principles, application scopes, advantages and disadvantages are discussed. Based on these, the development history of the etch process modeling is summarized, and the development opportunities of the etch model are prospected. This paper provides a brief view for establishment of the plasma etching process model.

The complete mitochondrial genome (mitogenome) of Therioaphis trifolii is sequenced and annotated based on high-throughput sequencing method in the present study. It is a typical circular DNA molecule of 16,068 bp, with 37 genes and 82.6% A + T content. The gene order is the same as the putative ancestral arrangement of insects. Thirteen protein-coding genes (PCGs) start with an ATN codon and terminate with a TAA codon or a single T residue. All the 22 tRNAs, ranging from 64 to 73 bp, have the typical clover-leaf structure except for trnS1. The length of rrnL and rrnS genes are 1265 bp and 779 bp respectively. The control region is 856 bp in length with 85.2% A + T content. The phylogenetic tree supports the monophyly of Aphidinae and the sister relationship between T. trifolii and seven species of subfamily Aphidinae.

Surface-enhanced Raman spectroscopy (SERS) substrates with high sensitivity and reproducibility are highly desirable for high precision and even molecular-level detection applications. Here, large-scale uniformly hybrid nanoparticle-enhanced Raman spectroscopy (NERS) substrates with high reproducibility and controllability were developed. Using oxygen plasma treatment, large-area and uniformly rough polystyrene sphere (URPS) arrays in conjunction with 20 nm Au films (AuURPS) were fabricated for SERS substrates. Au nanoparticles and clusters covered the surface of the URPS arrays, and this increased the Raman signal. In the detection of malachite green (MG), the fabricated NERS substrates have high reproducibility and sensitivity. The enhancement factor (EF) of Au nanoparticles and clusters was simulated by finite-difference time-domain (FDTD) simulations and the EF was more than 104. The measured EF of our developed substrate was more than 108 with a relative standard deviation as low as 6.64%−13.84% over 15 points on the substrate. The minimum limit for the MG molecules reached 50 ng/mL. Moreover, the Raman signal had a good linear relationship with the logarithmic concentration of MG, as it ranged from 50 ng/mL to 5 μg/mL. The NERS substrates proposed in this work may serve as a promising detection scheme in chemical and biological fields.
