The world is currently faced with a pandemic of coronavirus disease 2019 (COVID-19). Several genome-wide and exome-wide studies (GWAS and EWAS) have been performed to identify the variability in the host genetic constitution that likely underlines the inter-individual variabilities in COVID-19 severity and clinical manifestation. Due to the magnitude of the articles available, creating a list of host-specific genetic variants and genes associated with COVID-19 can be both time-consuming and extremely challenging for COVID-19 researchers. To this end, the COVID-19 Host Genome database was built. This is currently the only dedicated, free-to-use database that deals solely with COVID-19 host-specific genetic variants and genes. HyperText Markup language (HTML), Cascading Style Sheets (CSS), Hypertext Preprocessor (PHP), and My Structured Query Language (MySQL) server (version 5.7.38) were used to develop the website, storage, and extraction of the data. So far, 787 genetic variants from 63 previously published articles were collected. The tabular data are hyperlinked to the original articles and the users can download all data from the database. COVID-19 Host GenomeDB is being revised constantly every month, and can benefit the research community studying the genetic variants to improve COVID-19 treatment and prevention strategies.

In this investigation, new composites of cerium oxide were incorporated in the copper-based metal–organic framework using trimesic acid by the precipitation method. Synthesized Cu-MOF/CeO2 nanocomposite is characterized by numerous analytical methods to interpret their structure, morphology, and thermal behaviour. Surface micrograph analysis revealed that spherical particles and cerium oxide nanoparticles are embedded in the Cu-MOF matrix. The prepared nanocomposite shows a lesser onset potential and overpotential with a high current density (18.6 mA cm−2). The Cu-MOF/CeO2 composite exhibits a small Tafel slope of 54.6 mV dec-1 and suggests that the hydrogen evolution reaction follows the Heyrovsky mechanism. On the other hand, five different MOF/CeO2 nanocomposite concentrations were tested on a human osteosarcoma cell line (MG63). The 50% cell mortality was observed at 97.9 μg/ml and it proved less cytotoxicity effect with better biocompatibility. Thus, overall results showed that the nanocomposite as an efficient electrocatalyst towards hydrogen evolution reaction and targeted drug delivery applications.

The antifungal effects of Citrullus colocynthis extract (Hexane, chloroform, methanol, and water) were tested in vitro on Fusarium oxysporum f. sp. lycopersici (Sacc.) W. C. Snyder & H. N. Hans (FOL), the causal agent of Fusarium wilt. Of these, methanol and water extract at 10% showed the highest inhibition of mycelial growth of FOL by 12.32 and 23.61 mm respectively. The antifungal compounds were identified through Fourier transform infrared (FT-IR) spectroscopy and gas chromatography-mass spectroscopy (GC-MS). The methanol extract was compatible with the biocontrol agent Trichoderma viride. The antagonistic fungi were mass-cultured under laboratory conditions using sorghum seeds. Both T. viride and C. colocynthis methanol extract was also tested alone and together against FOL under both in vitro and in vivo conditions. The combination of T. viride and C. colocynthis showed the highest percentage of antifungal activity (82.92%) against FOL under in vitro conditions. This study revealed that induced systemic resistance (ISR) in enhancing the disease resistance in tomato plants against Fusarium wilt disease. The combined treatment of T. viride and C. colocynthis significantly reduced the disease incidence and index by 21.92 and 27.02% in greenhouse conditions, respectively. Further, the induction of defense enzymes, such as peroxidase (PO), polyphenol oxidase (PPO), β-1,3-glucanase, and chitinase were studied. The accumulation of defense enzyme was greater in plants treated with a combination of T. viride and C. colocynthis compared to the control. Reduction of wilt disease in tomato plants due to the involvement of defense-related enzymes is presumed through this experiment.

Heavy metal contamination harms soil and plant growth, making it a serious global environmental issue. Due to the scarcity of canal water, people have become more reliant on groundwater, but aquifers are expensive and of poor quality. Therefore, wastewater has become one of the essential sources of irrigation in developing countries and rural areas. In rural areas, sanitary, sewage, and municipal wastewater are the most common sources of irrigation. The objective was to analyze and compare the concentration of heavy metals in Spinach (Spinacia oleracea) collected from two different sites, i.e., one field irrigated using sanitary wastewater and another by tube well water through drainage ditches in a rural area of Faisalabad. AAS (Atomic Absorption Spectrophotometry) analyzed heavy metals from plants through irrigated water, soil, and the collected edible parts of plant samples. The samples were digested by using acids and AAS-analyzed metals. The concentration of Cd (0.051 ± 0.029) mg/l found higher than the recommended values by WHO (0.01 mg/l) in sanitary wastewater, as well as Cd concentration (0.478 ± 0.276) mg/kg in edible parts of spinach, also detected beyond the permissible limits (0.2) mg/kg of WHO (2007). These exceeding values of Cd in edible parts of spinach may cause serious human health risks.

Bisphenol A (BPA) is an industrial toxicant that can potentially damage the liver. Tangeretin (TGN) is a natural flavonoid that displays various pharmacological activities. This experiment was carried out to evaluate the protective effects of TGN against BPA-induced hepatic impairment in the male albino rat. Twenty-four male albino rats were equally divided into four different groups: control, BPA (100 mg/kg), BPA + TGN (100 mg/kg + 50 mg/kg) and TGN (50 mg/kg). BPA exposure significantly decreased the activities of catalase (CAT), superoxidase dismutase (SOD), peroxidase (POD), glutathione reductase (GSR), glutathione S-transferase (GST), and glutathione (GSH) content while substantially increasing the thiobarbituric acid reactive substances (TBARS) and hydrogen peroxide (H2O2) levels. A substantial increase in the levels of alanine aminotransferase (ALT), alkaline phosphatase (ALP), aspartate aminotransferase (AST) was also observed in BPA treated rats. Moreover, BPA significantly increased the inflammatory markers, including tumor necrosis factor-α (TNF-α), nuclear factor kappa-B (NF-κB), Interleukin-6 (IL-6), Interleukin-1β (IL-1β)levels, cyclooxygenase-2 (COX-2) activity, and histopathological damages. However, co-treatment with TGN efficiently minimized the BPA-induced biochemical, inflammatory, and histopathological impairments in rat liver. The present study shows that TNG has significant potential to avert BPA-induced liver damage to its antioxidant and anti-inflammatory properties.

The Hepatitis B virus (HBV) infection is one of the most widespread viral infections of humans. HBV causes acute and chronic hepatitis. Chronic hepatitis leads to hepatocellular carcinoma, which is a significant cause of death. DNA-based immunization programs to control the spread of Hepatitis B in developing countries are costly and require special storage and transportation. The alternative way is to express Hepatitis B surface antigen (HBsAg) in plants to develop oral vaccines. In this study, HBsAg gene was isolated, cloned, and then transformed in tomato plants. The transgenic tomato plants were confirmed through RT-qPCR. HBsAg expression was analysed in mature green and red stages of tomato fruit through quantitative real-time PCR. It was observed that expression of HBsAg was high in matured red tomato as compared to mature green. The present study is the first step to developing Solanum lycopersicum as an edible vaccine production system in this world region.

Since the epidemic began in November 2019, no viable medicine against SARS-CoV-2 has been discovered. The typical medication discovery strategy requires several years of rigorous research and development as well as a significant financial commitment, which is not feasible in the face of the current epidemic. Through molecular docking and dynamic simulation studies, we used the FDA-approved drug mezonavir against the most important viral targets, including spike (S) glycoprotein, Transmembrane serine protease 2 (TMPRSS2), RNA-dependent RNA polymerase (RdRp), Main protease (Mpro), human angiotensin-converting enzyme 2 (ACE-2), and furin. These targets are critical for viral replication and infection propagation because they play a key role in replication/transcription and host cell recognition. Molecular docking revealed that the antiviral medication mozenavir showed a stronger affinity for SARS-CoV-2 target proteins than reference medicines in this investigation. We discovered that mozenavir increases the complex's stability and validates the molecular docking findings using molecular dynamics modeling. Furin, a target protein of COVID-19, has a greater binding affinity (-12.04 kcal/mol) than other COVID-19 target proteins, forming different hydrogen bonds and polar and hydrophobic interactions, suggesting that it might be used as an antiviral treatment against SARS-CoV-2. Overall, the present in silico results will be valuable in identifying crucial targets for subsequent experimental investigations that might help combat COVID-19 by blocking the protease furin's proteolytic activity.

The purpose of the current research was to determine the impact of various biochar supplements on nutrient digestibility and growth performance of Catla catla fingerlings fed on Moringa oleifera seed meal (MOSM) based diet. An experiment with 90 days of feeding was conducted to investigate the efficacy of biochar obtained from different sources such as parthenium, farmyard manure, poultry waste, vegetable waste, and corncob waste at 2 mg/kg. Spodoptera litura (Fabricius) is an agriculturally significant polyphagous insect pest that has evolved a high level of resistance to conventional insecticides. A dietary assay was used in this work to assess the resilience of field populations of S. litura to λ-cyhalothrin. Analysis of the function and expression of the cytochrome P450 gene was used to test the sensitivity of S. litura larvae to sub-lethal concentrations of the insecticidal plant chemical Precocene 1, both by itself and in combination with λ-cyhalothrin. The activity of esterase enzymes (α and β) was found to decrease 48 h post treatment with Precocene 1. The activity of GST enzyme and cytochrome P450 increased with Precocene 1 treatment post 48 h, however. Expression studies revealed the modulation by Precocene 1 of cytochrome P450 genes, CYP4M16, CYP4M15, CYP4S8V4, CYP4G31, and CYP4L10. While CYP4M16 expression was stimulated the most by the synergistic Precocene 1 + λ–cyhalothrin treatment, expression of CYP4G31 was the most down-regulated by Precocene 1 exposure. Hence, it is evident that λ–cyhalothrin-resistant pest populations are still sensitive to Precocene 1 at a sublethal concentration that is nevertheless capable of hindering their development. Precocene 1 can therefore be considered a potent candidate for the effective management of insecticide-resilient S. litura.

Methotrexate (MTX) is commonly used as a chemotherapeutic drug against cancer but its efficacy is limited due to its side effects. Baicalin is a flavonoid that has potent antioxidant properties. This study aimed to evaluate baicalin’s protective effect on MTX-induced mitochondrial damage in testicular tissues of rats. Twenty-four male Wistar rats were distributed into four groups: control, MTX, MTX + baicalin, and baicalin alone. The activities of enzymes such as catalase, superoxide dismutase, glutathione reductase and glutathione peroxidase was significantly lowered in the MTX treated group, compared to the other groups. Concomitantly, MTX group also showed elevated the levels of reactive oxygen species and thiobarbituric acid reactive substances in the testicular mitochondria. Furthermore, MTX group also showed reduced the activities of tricarboxylic acid (TCA) cycle enzymes, i.e., succinate dehydrogenase, malate dehydrogenase, isocitrate dehydrogenase, and alpha-ketoglutarate dehydrogenase. The actions of electron transport chain (ETC) complexes (I-IV), as well as mitochondrial membrane potential (MMP), were also reduced the MTX group. However, baicalin treatment reversed the damaging effect of MTX in the mitochondria of testicular tissues. The baicalin treatment potentially reduced the damaging impact of MTX in isolated mitochondria from testicular tissues. Thus, the current study revealed that the baicalin has conspicuous potential to attenuate the MTX induced mitochondrial damage in rat’s testicular tissues.

Introduction: Paracetamol (acetaminophen) is a medicine used for the treatment of fever, pain, and inflammation during pneumococcal infection. Objectives: To see how paracetamol affects the transcriptional profile of Streptococcus pneumonia across the genome. Methods: In this study, microarray analysis was performed for transcriptional profiling. Results: Transcriptome data showed differential expression of several genes in S. pneumoniae D39 wild-type incorporated with paracetamol in the growth medium. Furthermore, these genes were categorized using Clusters of Orthologous Groups (COG) functional categorization on the basis of the suspected functions of the respective proteins. The majority of differentially expressed genes are in COG categories E (Amino acid transport and metabolism) and I (Lipid transport and metabolism). Analysis of protein–protein interaction networks exhibited compactly connected networks between fatty acid transport/biosynthesis and antibiotic biosynthesis genes. Moreover, pathways enrichment analysis revealed that fatty acid metabolism and biosynthesis genes were significantly affected under the criteria we've established. Conclusion: These results suggest the fatty acid biosynthesis and metabolism genes to be potential target of paracetamol in S. pneumoniae D39 wild-type.

Chikungunya, Zika, Dengue Viruses, and now Novel Coronavirus are global health challenges that cause human diseases ranging from febrile illnesses to death. Most of these viruses are mainly vectored by Aedes mosquitoes worldwide. Molecular detection of arboviruses was made in female Aedes mosquito pools caught from all the seven districts by using a reliable molecular technique, “RT-PCR.” From 216 collections of Aedes species, arboviruses were detected in 27, including only Alphavirus genus to determine mosquito abundance and evaluate the potential role of Aedes aegypti and Ae. albopictus mosquitoes in arboviruses and nvel Coronavirus transmission. 5322 mosquitoes were collected using aspirators; 35.31% (n = 2049) were identified as female Aedes using morphological keys, pooled into 216 pools, and tested for arboviruses and coronaviruses by using RT-PCR with the help of specific primers. Novel Coronavirus was not detected in this study. Only the Flavivirus genus was detected in twenty-seven pools giving an infection rate of 62.96% (n = 17) for DENV2, while DENV3 was 37.03% (n = 10). Furthermore, our results indicated no role of mosquitoes in the spread of Covid-19. Results showed a higher infection rate in urban sites than in rural ones. The detection of arboviruses indicates possible human health risk due to active role of these mosquitoes in spreading of arbovirus in the study area.

The cytotoxicity in freshwater fishes due to different industrial dyes in industrial effluents is a major worldwide issue. Hematoxylin dye has a wide range of uses in textile industries and laboratories. This study was aimed to evaluate the toxic effects of hematoxylin's sublethal effect in vitro in Cirrhinus mrigala. The fish was exposed to different grading concentrations of dye in the aquarium. Fish were sacrificed and dissected to remove the kidney after exposure to hematoxylin dye for specific time intervals. Nephrotoxicity and cytotoxicity induced by this dye were detected through histopathology by using the paraffin wax method. Immediate mortality of fish was noticed against the exposure to 0.08 g/L (LC50) concentration of dye, but at 0.008 mg/L and 0.018 mg/L, it showed tremendous tissue damage in the kidneys, significant reduction in fish growth. This dye induced many alterations in the kidney such as tubular degeneration, vacuolation, shrinkage of a glomerulus, reduced lumen, congestion in the kidney, glomerulonephritis, absence of Bowmen space, necrosis of the hematopoietic interstitial tissues, clogging of tubules, necrosis in the glomerulus and increased space between glomerulus and bowmen's capsule. Although this dye has a wide range of biological and industrial applications, a minute amount of hematoxylin released in effluents is quite toxic to aquatic fauna.

Polyethylene microplastic (PE-MP) is amongst the most manufactured plastics globally. However, emerging studies report its adverse effects on wildlife and humans. Luteolin (LUT) is a flavonoid compound known for its potent antioxidant properties. The present study was designed to estimate the toxic effect of PE-MP on testicles of adult male rats. The therapeutic potential of LUT was also determined in alleviating PE-MP instigated testicular dysfunctions. An experiment was carried on 48 adult male Sprague-Dawley rats, which were randomly classified into four groups: control, PE-MP (1.5 mg/kg), PE-MP + LUT (1.5 mg/kg + 50 mg/kg respectively), and LUT (50 mg/kg). The biochemical, spermatogenic, hormonal, and histopathological parameters were studied after 56 days of treatment. PE-MP exposure disrupted the biochemical profile by reducing catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPx), and glutathione reductase (GSR) activities. In comparison, the concentration of reactive oxygen species (ROS) and malondialdehyde (MDA) levels were increased. PE-MP-intoxication also reduced the sperm motility, viability, the number of hypo-osmotic tails swelled spermatozoa, and epididymal sperm count; additionally, it increased the sperm morphological (head–tail and mid-piece) abnormalities. Furthermore, it lowered luteinizing hormone (LH), follicle-stimulating hormone (FSH), and plasma testosterone. PE-MP exposure led to histopathological damages in testicular tissues. However, LUT inverted all the illustrated impairments in testes. Our results demonstrate that PE-MP could cause testicular damage in male rats, effectively mitigated by LUT treatment.

Small epipelagic and migratory, Euthynnus affinis (kawakawa) is one of the commercially significant tunas of Indo-Pacific tropical and subtropical waters. Unfortunately, the management and stock structure of certain migratory species in the area is not clear. The current study aimed to discriminate the E. affinis through shape and body size variations and to evaluate the variations among seven populations of E. affinis. A total of 114 individuals of E. affinis were collected from two main geographic areas; the Straits of Malacca and the South China Sea. Multivariate analyses, such as discriminant function analyses (DFA) and principal component analyses (PCA) of 11 homologous landmarks and seven morphometric variables were carried out to discriminate seven populations of E. affinis. The results revealed a significant variations among the body shape of the seven populations of E. affinis. Morphological homogenous occurred between populations obtained from the east coast of Peninsular Malaysia (Terengganu, Kelantan, and Johor). However, populations found on the west coast of Peninsular Malaysia (Selangor) were separated and formed another cluster. The variations in the body shape of E. affinis occurred in the body depth and the overall body shape. The percentage of overall correct classification for all seven populations of E. affinis is 88.6%. This present study is the first report using a geometric morphometric method performed on the E. affinis from Peninsular Malaysian waters.

The high spread rate, severe symptoms, psychological and neurological problems, and unavailability of effective medicines are the major factors making Coronavirus disease 2019 (COVID-19) a massive threat to the world. It is thought that COVID-19 causes mild symptoms or mild infectious illness in children. However, we cannot rule out the possibility of serious complications such as the multisystem inflammatory syndrome. COVID-19 induces mild to severe neurological problems in children, such as stroke, encephalopathy, mild shortness of breath, and myalgia. The development of these conditions is associated with pro-inflammatory responses and cytokine storms, which alter the physiology of the blood–brain barrier and allow the virus to enter the brain. Despite the viral entry into the brain, these neurological conditions can also be caused indirectly by severe immune responses. In this article, we describe COVID-19 and the associated neurological and immunological complications in children.

The present work aims to synthesize zinc oxide (ZnO) nanoparticles via green approaches using leaf extract of Parthenium hysterophorus. UV–vis and FT-IR tests confirmed the existence of biomolecules, active materials, and metal oxides. The X-ray diffraction structural study exposes the ZnO nanoparticles formation with hexagonal phase structures. SEM and TEM analysis reveal surface morphologies of ZnO nanoparticles and most of them are spherical with a size range of 10 nm. ZnO nanoparticles were revealed strong antimicrobial activity against both bacterial and fungal strains. The germination of seeds and vegetative growth of Sesamum indicum has been greatly improved.

Reproductive success is attained by various mechanisms in insects. Prolonged post insemination association is one such mechanism to increase the reproductive success. The present study was conducted to assess the role of post insemination association of mating partners on reproductive performance in Chrysomelidae beetle, Zygogramma bicolorata Pallister. The matings were disrupted at different time intervals and fecundity and percent egg viability of the females were recorded. In addition, the mounting attempts, mating attempts, time to commencement of mating and latent period were also recorded. It was hypothesized that: (1) the mounting and mating attempts would not exist, (2) copulation duration, would not affect the reproductive performance, and (3) the beetle would not exhibit the mate guarding behaviour. Interestingly, results revealed that 6.00 ± 1.3 and 6.59 ± 0.93 mounting and mating attempts are needed to establish successful mating. The results revealed that males improved their percent egg viability with a mating duration ranging from nearly 30–50 min. While fecundity increased with a mating duration of above 30 min and up to a duration of 60 min. This result concluded that males of this beetle display post copulatory mate guarding behaviour after 60 min in which male rides on female’s back with his aedeagus inserted in the female genital tract.

Introduction: Researchers worldwide with great endeavor searching and repurpose drugs might be potentially useful in fighting newly emerged coronavirus. These drugs show inhibition but also show side effects and complications too. On December 27, 2020, 80,926,235 cases have been reported worldwide. Specifically, in Pakistan, 471,335 has been reported with inconsiderable deaths. Problem statement: Identification of COVID-19 drugs pathway through drug-gene and gene−gene interaction to find out the most important genes involved in the pathway to deal with the actual cause of side effects beyond the beneficent effects of the drugs. Methodology: The medicines used to treat COVID-19 are retrieved from the Drug Bank. The drug-gene interaction was performed using the Drug Gene Interaction Database to check the relation between the genes and the drugs. The networks of genes are developed by Gene MANIA, while Cytoscape is used to check the active functional association of the targeted gene. The developed systems cross-validated using the EnrichNet tool and identify drug genes' concerned pathways using Reactome and STRING. Results: Five drugs Azithromycin, Bevacizumab, CQ, HCQ, and Lopinavir, are retrieved. The drug-gene interaction shows several genes that are targeted by the drug. Gene MANIA interaction network shows the functional association of the genes like co-expression, physical interaction, predicted, genetic interaction, co-localization, and shared protein domains. Conclusion: Our study suggests the pathways for each drug in which targeted genes and medicines play a crucial role, which will help experts in-vitro overcome and deal with the side effects of these drugs, as we find out the in-silico gene analysis for the COVID-19 drugs.

Pollution is severely threatening the wetland habitats. Heavy metals are one among of the major pollutants in wetland habitats. The cadmium (Cd), copper (Cu), chromium (Cr), cobalt (Co), lead (Pb), mercury (Hg), nickel (Ni) and zinc (Zn), were assessed in the water, sediment, benthic species (polychaetes, mollusc and crustaceans), prawn and fishes. The assessment of heavy metals was done by using double-beam Atomic Absorption Spectrophotometer (AAS). The Hg, Cr and Co were greater in sediment, Ni and Cd were higher in polychaetes and molluscs respectively. However, the Cu and Pb greater in crabs and the Zinc was greater in fishes. The concentration of metals showed significant differences among the various sources examined (P 0.05). The inter-correlational analysis among the metals assessed from the various sources showed that the Cr and Pb not correlated among the eight metals examined. However, the Cu and Co were correlated with Hg (r = 0.307) and (r = 0.788) respectively. The nickel was correlated with Hg (r = 0.367), Cu (r = 0.362) and Co (r = 0.432). The Zinc was correlated with the Cd (r = 0.331) and Hg (r = 0.737). However, correlation of metals among the different sources shown that the metals of polychaetes correlated with sediment r = 0.637, the metals of crabs correlated with the sediment and polychaetes r = 0.630 and r = 842 respectively, the metals of molluscs was also correlated with sediment (r = 0.636), polychaetes (r = 0.889) and crabs (r = 0.894). In addition to that the metals of prawns was correlated with the polychaetes (r = 839), crabs (r = 0.628) and molluscs (r = 0.634). The metals of fishes correlated with polychaetes (r = 0.529), crabs (r = 0.710), molluscs (r = 0.493) and prawns (r = 0.593). Indeed the multiple regression model explained that the metals of sediments influence the accumulation of metals in biotic species such as polychaetes, molluscs, crustaceans, prawns and fishes with 84% (F = 21.079; p Pb > Ni > Cr > Zn > Co > Cu > Cd. The study found that the level of heavy metals at various sources in the sanctuary is showing considerable warning and the sanctuary is required intensive assessment on various aspects of pollution since the Point Calimere Wildlife Sanctuary is supporting several species of migratory and endangered shorebirds seasonally.

This study presents ethanol's fabrication by fermenting the golden trumpet flower (Allamanda schottii L) with the yeast strain Saccharomyces cerevisiae. The changes in different parameters during fermentation were studied and optimized while producing the ethanol and the end product was subjected to emission test study by blending petrol and ethanol. The Allamanda floral substrate contains 65% polysaccharides. The strain S. cerevisiae was obtained in the form of baker’s yeast from a domestic shop. For 100 ml of slurry, the highest bioethanol yield recorded was about 18.75 ml via optimization of different culture conditions, including a 1:8 ratio for slurry preparation, maintained under 35 ⁰C, 5.5 pH, 72 h. old inoculum with a quantity of 3.75 g 100 ml−1, fermented for120 h. The highest yield of bioethanol was acquired under the addition of urea. This technique & design is capable of industrial-scale fabrication of bioethanol by using A. schottii floral substrates. This research was conducted to fabricate ethanol by fermentation (A. schottii L) floral substrate with S. cerevisiae. The optimum physiochemical parameters required to obtain the highest yield of bioethanol from A. schottii flower by fermentation was studied. The immobilization strategy with a cheap agricultural substrate and magnetic nanoparticles were also studied. The engine performance and emission studies were done with different blends of petrol and bio-ethanol.

Arsenic (As) is a toxic contaminant present in organic and inorganic forms in the environment. Nobiletin (NOB) is a polymethoxy flavone that has recently gained substantial consideration due to its curative impacts. The present experiment was conducted to assess the hepatoprotective efficiency of NOB on As-generated hepatotoxicity. Twenty-four adult rats were equally distributed into four groups and designated as control, As (50 mg/kg)-treated, As + NOB (50 mg/kg and 25 mg/kg, respectively), and NOB (25 mg/kg)-treated groups. After 30 days, experimental animals were decapitated, then blood and tissue samples were collected for further analysis. The group treated with As showed a significant decrease in the activity of antioxidant enzymes, including catalase (CAT), superoxide dismutase (SOD), peroxidase (POD), glutathione (GSH), glutathione reductase (GSR), and total antioxidant status (TAS), and a substantial increase in the accumulation of As in liver tissues, levels of total oxidant status (TOS), hydrogen peroxide (H2O2), and lipid peroxidation (TBARS). Significant increases in alanine aminotransferase (ALT), alkaline phosphatase (ALP), and aspartate aminotransferase (AST) levels were observed in As-treated rats. Moreover, nuclear factor (NF)-κB, tumor necrosis factor (TNF)-α, interleukin (IL)-1β, interleukin (IL)-6, and cyclo-oxygenase (COX)-2 activity, as well as the levels of pro-apoptotic markers (Bax, Caspase-3, and Caspase-9) were increased on exposure to As. In contrast, the anti-apoptotic marker (Bcl-2) level was significantly decreased. As administration showed a significant disturbance in hepatic tissue histology. However, cotreatment of NOB with As considerably increased the antioxidant enzyme activity, with a noteworthy reduction in the deposition of As in hepatic tissues, TBARS, and H2O2 levels. NOB-administrated rats showed considerable recovery in terms of inflammation, apoptosis, and histological damage. Hence, NOB can be considered a useful curative compound due to its medicinal properties against As-prompted hepatotoxicity.

Chronic diabetes extensively complicates the glucose metabolism to onset and progress the complication. Concurrently, several contemporary medicines, especially organo-metallic formulations, are emerging to treat hyperglycemia. The current study aims to emphasize the gold nanoparticles (GNPs) potential for glucose metabolism regulation in Streptozotocin (STZ) induced diabetes. Quantitative real-time polymerase chain reaction (RT-PCR) was carried out to detect the mRNA expression of Glucose transporters 2 (GLUT2), Glucokinase (GK) and Glucose 6 Phosphatase (G-6-Pase). The study shows remarkable results such as the prognostic effect of GNPs in reinforcing the repression of enzyme complex G-6-Pase about 13.3-fold when compared to diabetes control. Also, molecular docking studies showed significant inhibition of G-6-Pase by the terpenoid ligands with alpha and beta amyrin from leaf extract of Couroupita guianensis. Thus the study explored the novel mechanism of G-6-Pase downregulated by GNPs intervention that majorly contributes to the regulation of circulatory glucose homeostasis during diabetes.

Abstract The study is intended to deliver the incidence of heavy metals in the feathers of shorebirds from two important Central Asian Flyway (CAF) migratory shorebirds wintering sites such as the Point Calimere Wildlife Sanctuary (PWLS) and Pichavaram Mangrove Forest (PMF), India. Feathers of fifteen species of shorebirds and seven different metals viz., Cu, Cr, Co, Pb, Hg, Ni and Zn were analyzed. Zn was highest in Dunlin, Little-ringed Plover, Marsh Sandpiper, and Common sandpiper, Ni showed highest in Little ringed plover, and Common sandpiper, Co, Cr, and Cu were maximum in Little stint, Marsh sandpiper, and Dunlin, respectively. The Hg was higher in Black-winged stilt, Common redshank, Curlew Sandpiper, Eurasian curlew, Lesser Sand-plover, Temminck’s stint, Kentish plover, Spotted redshank, and Wood sandpiper, the Pb found highest in Kentish plover, Painted stork, Spotted redshank, Wood sandpiper, Eurasian Curlew, and Lesser sand-plover. The concentration of metals showed significant variations among the species of shorebirds studied (P Ni > Co > Cr > Cu > Pb > Hg. Nevertheless, the current study revealed that the level of metals in the shorebirds is alarming; since the PWLS and PMF are located along the CAF routes, it needs intensive studies on various pollutions to manage both the resident as well as migratory shorebirds.

The entomofaunal survey and its toxicity of Blumea mollis (Asteraceae) leaf aqueous extract-mediated (Bm-LAE) silver nanoparticles (AgNPs) were assessed against selected human vector mosquitoes (HVMs). A total of 1800 individuals of 29 species belongs to 7 genera were identified. Month-wise and Genus-wise abundance of HVMs larval diversity were calculated and one-way ANOVA statistically analyzed the average physico-chemical characteristics. The relationship between physicochemical characteristics and HVMs larvae in KWS was interpreted. The total larval density and container index were 23530.18 and 1961.85 examined against 10 different containers. Various spectroscopic and microscopic investigation characterized Bm-AgNPs. The Bm- AgNPs tested against HVMs larvae, the predominant LC50/LC90 values of 18.17/39.56, 23.45/42.49 and 21.82/40.43 μg/mL were observed on An. subpictus Cx. vishnui and Ae. vittatus, respectively. The findings of this investigation, improperly maintained drainages, containers and unused things in study sites, are engaged to HVMs development. This will be essential for designing and implementing HVMs control. The larval toxic potentiality of Bm- AgNPs had a prompt, inexpensive and compelling synthesis of multi-disperse action against HVMs.

Abstract Background Temperature is known to affect living organisms and alter the expression of responsive genes, which affects a series of life processes, such as development, reproduction and metabolism. Several genes and gene families have been involved in high temperature responses, such as heat shock protein (hsp) family, Jumonji family and genes related to cortisol synthesis. Gonad is a vital organ related to the existence of a species. However, the comprehensive understanding of gonadal responses to environmental temperature is limited. Results To explore the effects of environmental temperature on genes and gene networks in gonads, we performed acute heat treatment (48 h) on Chinese tongue sole (Cynoglossus semilaevis). Gonadal transcriptome analysis was conducted on females, pseudomales and males exposed to high (28 °C) and normal (22 °C) temperatures. A total of 1226.24 million clean reads were obtained from 18 libraries. Principal component analysis (PCA) and differentially expressed gene (DEG) analysis revealed different performance of sex responses to heat stress. There were 4565, 790 and 1117 specific genes altered their expression level in females, pseudomales and males, respectively. Of these, genes related to hsp gene family, cortisol synthesis and metabolism and epigenetic regulation were involved in early heat response. Furthermore, a total of 1048 DEGs were shared among females, pesudomales and males, which may represent the inherent difference between high and normal temperatures. Genes, such as eef1akmt3, eef1akmt4, pnmt and hsp family members, were found. Conclusions Our results depicted for the first time the gonadal gene expression under acute high temperature treatment in Chinese tongue sole. The findings may provide a clue for understanding the responses of genes and networks to environmental temperature.

Cytochrome c oxidase I (COI) is one of the mitochondrial genes, an excellent marker used for the DNA barcoding of various organisms. The COI was isolated from twelve different species of waterbirds using the Gene Elute DNA miniprep Kit. The relationship among the waterbirds was assessed by making a phylogenetic tree with the software MEGA-X. The phylogenetic tree's dendrogram showed two main branches in which seven species of water birds aligned one group with four subgroups and the remaining five species aligned with two subgroups based on their similar COI sequences. The Little egret showed 96% similarity with the Cattle egret, Purple heron and Oriental Darter produced 94% similarity with the Grey heron, pond heron shared 90% similarity with the Black-crowned night heron, Asian openbill shared 89% with Pheasant-tailed Jacana and the Common coot 94% similarity with White-breasted waterhen. The pond heron and the Black-crowned night heron showed zero % pairwise distance, but the Cattle egret, Little egret, Oriental Darter, Purple heron and Grey heron showed 12% of the pairwise distance. Twenty-one conserved haplotypes have been shown in their COI sequences based on the multiple sequences alignment. DNA barcoding identifies the species with their genetic property rather than based on their ecology and behvaviour.

Unambiguous and precise identifications of a species are crucial for managing and conserving the species across their dispersal regions. This study analyzed the molecular signatures and phylogenetic relationship of 84 specimens of lutjanids obtained from Malaysia, which covers three marine regions; Straits of Malacca (SM), west South China Sea (west SCS), and east South China Sea (east SCS). The assessment of 613 bp mtDNA COI has markedly classified the samples into five distinct species namely Lutjanus malabaricus, L. argentimaculatus, L. erythtropterus, L. sebae and L. bohar. The intraspecific and interspecific genetic distance ranged between 0.1% − 0.7% and 8.2% − 19.6%, respectively. While genetic homogeneity occurred among population and species across SM and west SCS, significant genetic structuring was detected among species between west and east SCS as evidenced by the occurrence of a single species, L. bohar that can be found only in Sabah (east SCS). The divulged potential loss of gene flow among the regions and was probably caused by oceanic currents. This study detected an ambiguous sequence (from Genbank) probably due to hybridization between species or simply because of an unidentified node. This study proposed that COI is the valid and reliable genetic marker for identifying lutjanids distributed across Malaysian waters. Future revision in distribution pattern and diversity exploration on a large scale with a higher number of samples and different genetic markers is needed to fill this current study's gaps.

This research aimed to evaluate the curative impact of nobiletin (NOB) on paraquat (PQ) induced mitochondrial impairment in rats' hepatic tissues. Twenty-four male rats were divided into control, PQ, PQ + NOB, and NOB treated groups. The alanine aminotransferase levels, aspartate aminotransferase, and alkaline phosphatase were increased in the experimental animals after exposure with PQ. A significant decrease in the level of catalase, glutathione peroxidase, superoxide dismutase, glutathione, thiobarbituric acid reactive substances, and reactive oxygen species were recorded in rats exposed to PQ. The mitochondrial TCA cycle enzyme activities, including isocitrate-dehydrogenase, succinate-dehydrogenase, alpha-ketoglutarate dehydrogenase, and malate-dehydrogenase, were reduced in PQ exposed rats. The activities of mitochondrial enzymes including NADH dehydrogenase, coenzyme Q-cytochrome reductase, succinic-coenzyme Q and cytochrome c-oxidase were significantly decreased after the PQ-exposure. PQ administration also reduced the mitochondrial membrane potential. However, the administration of mitochondria with NOB can decrease the toxic effect of the PQ in isolated mitochondria. NOB treatment potentially reduced the damaging effects of the PQ in the mitochondria isolated from hepatic tissues. Thus, the current study revealed that the NOB could attenuate PQ-induced mitochondrial damage in rats' hepatic tissues.

Anthropogenic activities, urban and industrial wastewaters, and the usual disintegration of rock from physical, chemical, and biological stresses are the main cause of heavy metals in seawater and sediments worldwide. Five different sampling locations were selected after an initial survey of the Dammam region Seawater study area, and sediment samples were procured in triplicate from each location to assess the level of selected heavy metals pollutions in the Arabian Gulf, Dammam, Saudi Arabia. Be, Cr, Ni, Cu, As, Se, Ag, Cd, Pb, Hg, Co, Zn, Fe, Mn, Sr, and Mo were detected using “inductively coupled plasma mass spectrometer (ICP-MS).” The pH ranges from 7.91 ± 0.26–8.22 ± 0.38 in stations 4 and 3, respectively. Surface seawater's electrical conductivity values ranged from 71788 ± 28.52 to 75680 ± 42.92 µS/cm at stations 4 and 3. The minimum and maximum concentration of total dissolved solid was observed as 49524 ± 50.25 and 54786 ± 66.95 mgL-1 in sites 4 and 3, respectively. The level of these metals fluctuated among the sampling sites. The highest concentration of these heavy metals was recorded from sampling site 4 and minimum in location 1. The average level of these metals in water samples were recorded Sr > Cu > Se > Zn > As > Mn > Fe > Ni > Mo > Hg > Co > Cr > Ag > Cd > Pb and Be, respectively. The average level of these metals in sediment samples were recorded Fe > Sr > Ni > Mn > Cu > Cr > Zn > Pb > Ag > As > Mo > Co > Se > Hg > Be and Cd, respectively. It has been concluded that the concentration of As, Hg, Cd, Cr, Cu, Fe, and Sr was high, which is alarming, particularly in the Dammam area.

Aedes mosquitoes are the most important group of vectors that transmit pathogens, including arboviruses, and cause human diseases such as dengue fever, yellow fever, Zika virus, and Chikungunya. Biosynthesis and the use of green silver nanoparticles (AgNPs) is a vital step to identify reliable and eco-friendly controls for these vectors. In this study, Aedes (Ae.) aegypti larvae (2nd and 3rd instar) were exposed to leaf extracts of Ricinus communis (Castor) and AgNPs synthesized from the extract to evaluate their larvicidal potential. Synthesized AgNPs were characterized by UV–Vis spectroscopy, Fourier transform infrared spectroscopy (FTIR), and energy-dispersive X-ray spectroscopy (XRD). Ae. aegypti larvae were treated with different concentrations (50–250 ppm) of the leaf extract and synthesized AgNPs. There were five replicates per treatment, in addition to a positive (temephos) and negative control (dechlorinated water). Mortality was recorded after 12, 24, 36, and 48 h and the data were subjected to Probit analysis. The nanoparticles were more toxic (LC50 = 46.22 ppm and LC90 = 85.30 ppm) than the plant extract (106.24 and 175.73 ppm, respectively). The leaf extracts of Ricinus communis were subjected to HPLC analysis to identify their chemical constituents. This study suggests that plant extracts and synthesized nanoparticles are excellent alternatives to hazardous chemical pesticides used to control vector mosquitoes. This is a potentially useful technique that can reduce aquatic toxicity from insecticide use.

Zygogramma bicolorata Pallister (Coleoptera: Chrysomelidae) is a satisfactory potential candidate for controlling Parthenium hysterophorus L. The study was conducted in order to evaluate the bio-control potential of Z. bicolorata to satisfactory reduced the density of parthenium weed in natural field conditions. Therefore, beetles were released in the twelve study sites @ 12 adults and/or larvae/m2 in their respective sites on 5th, 10th, 15th and 20th day after initial field releases. Observations were made on defoliation by the beetle, perthenium plant height, plant biomass, leaf injury rating and soil seed reservoir. Present study showed that field releases of Z. bicolorata considerably lowering down the parthenium density at all treated sites. At larval release sites by the 60th day of observation a complete defoliation was noted, however at adult release sites it took 80 days for complete defoliation. Moreover, significantly higher number of developmental stages i.e. eggs, larvae and adults were noted in the adults released sites than to larval released sites and the controls. It was also noted that plant height and its density, flowers & leaves production, biomass of parthenium plant and seed reservoir in soil was negatively affected in treated sites as compared to untreated sites. Thus, we have concluded that adults can be utilized in biocontrol program because population build-up was greater in those sites where adults were released.

Diabetes mellitus and hyperglycemia can lead to other health complications. The pathogenesis of diabetes mellitus can affect numerous tissues in the eye structure and cataracts are one of the major causes of visual impairment in diabetes mellitus patients. In this study, we aim to investigate the ability of D-limonene to inhibit aldose reductase hypoglycemic activity in lens organ culture and its anti-glycating properties in STZ-induced cataracts of rats with diabetes. D-limonene repressed aldose reductase in-vitro with IC50 ranges of ~13.9 ± 0.31 µg/mL in an uncompetitive manner. This compound also prevented hyperglycemic induced increases in aldose reductase activity, accumulation of sorbitol, altered crystalline proteins (α, β and γ), and opacification of the rat lens in ex-vivo lens organ culture. Supplementation of D-limonene in STZ-induced diabetic rats restored these changes and delayed the progression of cataracts. This study demonstrates that D-limonene is a pharmaceutically active component that inhibits rat-lens aldose reductase. Therefore, this study showed that D-limonene prevents advanced glycation end product formation contributing to the integrity of α-crystallin chaperone activity and delays cataract development. It has been concluded, D-limonene has a role in inhibiting aldose reductase activity and advanced glycation end products formation in vivo, and in delaying the development of diabetic cataracts. Further studies of D-limonene are needed to assess other complications in DM as they may provide additional evidence for establishing D-limonene as a potential pharmacological target therapy for diabetes mellitus complications. In this study, 104 bacteria were isolated from the gut of Penaeus indicus. The morphologically different bacterial isolates were screened for probiotic properties. The strain D5 showed potent activity against various bacteria. The isolated strain Bacillus subtilis showed 17 mm zone of inhibition against Bacillus sp., 19 mm against Pseudomonas sp., 22 mm against Vibrio sp., and 22 mm against Micrococcus sp. The bacterial growth and bacterocin production in relation with incubation time was evaluated. Bacteriocin production was found to be high at the stationary phase and reduced considerably after fifth day. Bacterial growth and bacterocin production was maximum at pH 7.0 and optimum between pH 6.0 and 8.0. Incubation temperature is one of the important factors which significantly influences on the growth and production of bacteriocin. Bacterial growth and bacterocin production (17 ± 2 mm) were maximum at 30 °C. Glucose stimulated growth and bacterocin production (21 ± 3 mm). Among the supplemented nitrogen sources, glycine positively influenced on growth and metabolite production. In this study, bacteriocin supplemented diet enhanced the growth than control shrimp Bacteriocin was administered at various doses ranged between 10 and 50 mg/100 gm level. Shrimp length was 3.3 ± 0.13 cm and shrimp weight was 9.7 ± 0.12 g in 100 mg. The present finding revealed enhanced growth at high bacteriocin concentration. At 2 × 102 CFU/100 g probiotics in feed, the juvenile shrimp attained 16.8 ± 0.11 g after 40 days. The weight gain was 16.8 ± 0.11 CFU/100 g at 10 × 102 CFU/100 g probiotic concentration. Species identification technology (i.e. DNA barcoding) has been recognized as an important and reliable tools, although the effectiveness of using a single universal barcode marker for plants is still questionable. The present study reports a molecular identification method to discriminate 11 plant species within genus Piper that extensively utilized in traditional healing practices in Malaysia. The plants were sampled from six locations that represents three states in northern Peninsular of Malaysia, namely Perlis, Kedah and Penang. Species discrimination was conducted using BLASTn analysis and phylogenetic inference based on Maximum Likelihood and Neighbour-Joining method for two target genes, rbcL and rpoC1. The reliability of phylogenetic tree regenerations was assessed using Disparity Index analysis. Estimation of evolutionary divergence between all samples was employed based on Maximum Composite Likelihood with Kimura 2-parameter model. The present study revealed that the DNA identification method has successfully discriminated all samples to species level and that the rbcL is the reliable marker for identification. The population genetics study is crucial as it helps in understanding the epidemiological aspects of dengue and help improving a vector control measures. This research aims to investigate the population genetics structure of two common species of Aedes mosquitoes in Penang; Aedes aegypti and Aedes albopictus using Cytochrome Oxidase I (COI) mitochondrial DNA (mtDNA) marker. Molecular investigations were derived from 440 bp and 418 bp mtDNA COI on 125 and 334 larvae of Aedes aegypti and Aedes albopictus respectively, from 32 locations in Penang. All samples were employed in the BLASTn for species identification. The haplotype diversity, nucleotide diversity, neutrality test and mismatch distribution analysis were conducted in DnaSP version 5.10.1. AMOVA analysis was conducted in ARLEQUIN version 3.5 and the phylogenetic reconstructions based on maximum likelihood (ML) and neighbor-joining (NJ) methods were implemented in MEGA X. The relationships among haplotypes were further tested by creating a minimum spanning tree using Network version 4.6.1. All samples were genetically identified and clustered into six distinct species. Among the species, Ae. albopictus was the most abundant (67.2%), followed by Ae. aegypti (25.2%) and the rest were counted for Culex sp. and Toxorhynchites sp. Both Ae. aegypti and Ae. albopictus show low nucleotide diversity (π) and high haplotype diversity (h), while the neutrality test shows a negative value in most of the population for both species. There are a total of 39 and 64 haplotypes recorded for Ae. aegypti and Ae. albopictus respectively. AMOVA analysis revealed that most of the variation occurred within population for both species. Mismatch distribution analysis showed bimodal characteristic of population differentiation for Ae. aegypti but Ae. albopictus showed unimodal characteristics of population differentiation. Genetic distance based on Tamura-Nei parameter showed low genetic divergent within population and high genetic divergent among population for both species. The maximum likelihood tree showed no obvious pattern of population genetic structure for both Ae. aegypti and Ae. albopictus from Penang and a moderate to high bootstrap values has supported this conclusion. The minimum spanning network for Ae. aegypti and Ae. albopictus showed five and three dominant haplotypes respectively, which indicates a mixture of haplotypes from the regions analysed. This study revealed that there is no population genetic structure exhibited by both Ae. aegypti and Ae. albopictus in Penang. Mutation has occurred rapidly in both species and this will be challenging in controlling the populations. However, further analysis needed to confirm this statement. The main objective of the present study is to determine lead toxicity on various sizes of zebra fish in soft and hard water. Lead toxicity analysis was carried out in the laboratory trials for four days at various concentrations for the determination of LC50 value. Zebra fish was grouped into various sizes (G1-G4) and treated with lead for 96 h. Lead induced toxicity and induced various behavioural changes in Zebra fish. In the case of G1 (1.5 ± 0.2 cm length) fish the LC50 values ranged from 27.2 ± 1.8 mg/L to 9.7 ± 1.3 mg/L. In G2 (2.0 ± 0.2 cm length) fish, the LC50 value was 37.01 ± 2.3 mg/L and it reduced as 32.03 ± 3.4 mg/L after 48 h. In G3 group (2.5 ± 0.2 cm length fish), LC50 value was 38.78 ± 2.4 mg/L after 24 h treatment and this value decreased as 33.18 ± 1.1 mg/L after 48 h. After 72 h, LC50 value was 27.2 ± 2.1 mg/L and this value decreased considerably after 96 h (18.3 ± 2.1 mg/L) in G4 (3.0 ± 0.2 cm length) fish. Further, G4 fish was treated with lead in hard water and LC50 value was analyzed. LC50 value was observed as 28.62 ± 2.7 mg/L in hard water and the same fish group (G4) showed 18.3 ± 2.1 mg/L after 96 h. This study utilized genetic and morphometric approaches to assess the molecular and morphometric differentiation among commercially important species of mud crab. Molecular investigations were derived from 542 bp mitochondrial DNA COI on 249 individuals within genus Scylla from nine states in Malaysia represents four marine regions; South China Sea, Sulu Sea, Straits of Singapore and Straits of Malacca. Four specimens were obtained from Indonesia to give a robust analysis in this study. For species delimitation, Automatic Barcode Gap Discovery (ABGD) method on a web interface was employed. Analysis on phylogenetics was implemented utilizing Neighbour joining (NJ) and Maximum Parsimony (MP) methods. The inter- and intraspecies genetic distances (Ds) was computed using Kimura 2-parameter distance and executed in MEGA version 5.05. All samples were genetically and morphologically identified and clustered into four distinct species. Among the species, S. olivacea was the most abundant (n = 111), on the other hand the occurrence of S. paramamosain in Malaysia was very low (n = 29). No single individual of S. serrata from Malaysia was recorded in this study. Both genetic distance and phylogenetic approaches exhibited a correlative monophyletic association among all specimens analysed. This present study is crucial as it reports the reassessment of all species within genus Scylla in Malaysia, eventually could be employed as a reference source for subsequent research mainly on mariculture and other conservation efforts for the species. The study is to scrutinize andrographolides with Indoleamine 2,3-dioxygenase (IDO) inhibitory potential, its molecular mechanism against streptozotocin (STZ) diabetic retinopathy (DR) in Wistar rats. Oxidative stress markers such as Kynurenine metabolites, retinal histopathological changes have been studied. Further, IDO gene expression and docking studies have been performed. Andrographolide treated rats have been reducing the level of thiobarbituric acid reactive substances and protein carbonyls Kynurenine metabolites with an improvement in the level of GSH and expression of IDO as revealed by morphological changes in inner and outer nuclear layer of the retina. The current results of this study have been generated information about an activity of the andrographolide in the essential pocket of IDO. Our results explain, involving IDO and andrographolide would constitute an attempt to identify natural products with therapeutic value and further studies in this direction would be of immense significance in the administration of diabetes and its related problems. Recent trends in anticancer therapy is to use therapeutic agents which not only kill the cancer cell, but are less toxic to surrounding normal cells/tissue. One approach is to cut the nutrient supply to growing tumor cells, by blocking the formation of new blood vessels around the tumor. As the phytochemicals and botanical crude extracts have proven their efficacy as natural antiangiogenic agents with minimum toxicities, there is need to explore varieties of medicinal plants for novel antiangiogenic compounds.Rumex vesicarius L. (Humeidh), is an annual herbal plant with proven medicinal values. The antiangiogenic potential, and developmental toxicity of humeidh in experimental animal models has never been studied before. The crude extracts were prepared from the roots, stems, leaves and flowers of Rumex vesicarius L. in methanol, chloroform, ethyl acetate and n-hexane. The developmental toxicity screening in zebrafish embryos, has revealed that Rumex vesicarius was not toxic to zebrafish embryos. The chloroform stem extract showed significant level of antiangiogenic activity in zebrafish angiogenic assay on a dose dependent manner. Thirty five (35) bioactive compounds were identified by gas chromatography mass spectrophotometry (GC–MS) analysis in the stem extract of Rumex vesicarius. Propanoic acid, 2-[(trimethylsilyl)oxy]-, trimethylsilyl ester, Butane, 1,2,3-tris(trimethylsiloxy), and Butanedioic acid, bis(trimethylsilyl) ester were identified as major compound present in the stem of R. vasicarius.The anticancer activity of roots, stem, leaves and flowers crude extract was evaluated in human breast cancer (MCF7), human colon carcinoma (Lovo, and Caco-2), human hepatocellular carcinoma (HepG2) cell lines. Most of the crude extracts did not show significant level of cytotoxicity in tested cancer cells line, except, chloroform extract of stem which exhibited strong anticancer activity in all tested cancer cells with IC50 values in micro molar range.Based on these results, it is recommended that formulation prepared from R. vesicarius can further be tested in clinical trials in order to explore its therapeutic potential as an effective and safe natural anticancer product. The current study was aimed to evaluate the seasonal variation in bioaccumulation of metals (Pb, Cu and Ni) in the gills, kidney, liver and muscle of Oreochromis niloticus. Sixty fish samples (average wet body weight 460.77 ± 6.22 g) were collected from the freshwater reservoir Wadi Namar Riyadh, Saudi Arabia in winter, summer, spring and autumn seasons. The heavy metals were assessed through atomic absorption. The findings of this study revealed seasonal variation in bioaccumulation of metals. In winter season minimum concentration of Pb, Cu and Ni was recorded while the highest was recorded in summer season. The order of bioaccumulation of Pb and Ni in organs was: liver > kidney > muscle > gills in and liver > gills > kidney > muscle in winter and summer, respectively. The bioaccumulation order of Cu as kidney > liver > gills > muscle and gills > kidney > liver > muscle in winter and summer, respectively. The heavy metals concentration was crossing the permissible limits suggested by WHO. Histopathological examination in kidney exhibited congestion, dilation in bowman capsule space, necrosis. Liver of the fish exhibited cytoplasmic vacuolation, necrosis, sinusoid dilation. It has been concluded that this river is heavily contaminated due to the discharge of untreated waste from industry and domestic sewage from the city and there is a dire need to control this increasing contamination. Limited molecular studies have been documented with ACE gene polymorphism in OA and RA in the major industrial city in the country. The current study aims to investigate the prevalence of the arthritis risks and perform the ACE gene polymorphism in OA and RA in hospital based population. In this hospital-based study, we adopted 1000 arthritis subjects; 539 was suffering with OA and 461 RA in the local hospital. Based on the filled questionnaire and signed consent form, patients were involved in this study. Genotyping was performed in 1000 arthritis subjects. Our results indicated the risk of BMI and smoking status in both OA and RA diseases. Most of the participants involved in this study were considered to opt the non-vegetarian rather than the vegetarian. The arthritis patients were interested in eating the chicken (65.7%) and then red meat (31.5%). Regular walking (22.4%) was found to be higher in the arthritis patients than the regular exercise (4.7%). However, males are more active than the females in both the regular exercise and regular walking. Most of the arthritis patients (37.3%) had the comfortable sleep duration between 9 and 10 h. The prevalence of arthritis has been increased and patients with high BMI should be growing the risks of the various arthritis. Simultaneously, weight gain is connected with both the OA and RA diseases. Physical activity is required to reduce the risks of arthritis. The present study recommends to avoid the smoking and maintain the normal BMI specifically in the females to avoid the arthritis disease in Faisalabad city in the Pakistan. Genetic studies related to Ala54Thr (A54T) polymorphism in FABP2 in the Pakistani population are lacking.
