Vaccination against coronavirus disease 2019 (COVID-19), caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), is currently underway across countries worldwide. However, the prevalence and characteristics of prolonged adverse events lasting for several months after receiving the vaccine remain largely unknown. We herein report a 46-year-old woman with prolonged diarrhea and vomiting after receiving the BNT162b2 mRNA vaccine for COVID-19. She had no notable medical history, including that of gastrointestinal diseases. She developed vomiting several hours after receiving the first vaccine dose and further developed severe diarrhea after 7 days. Several days after the second vaccine dose, her condition deteriorated, unrelieved by symptomatic therapies, including anti-diarrheal drugs. Abdominal computed tomography (CT) revealed inflammatory changes in the entire segment of the small intestine with wall thickening. The upper and lower gastrointestinal and capsule endoscopies were unremarkable. The patient's symptoms persisted for more than 6 months after the second vaccine dose. A Vaccine Adverse Event Reporting System (VAERS) database search suggested that diarrhea is observed in approximately 3% of all vaccine recipients, but a literature review indicated that prolonged gastrointestinal symptoms lasting for several months is very rare. In summary, a case of prolonged unexplained gastrointestinal symptoms, possibly based on inflammatory changes in the small intestine, is described. A literature search revealed that this type of manifestation is very rare, and further evidence is needed to determine the causality between vaccination and gastrointestinal symptoms.

Although dexamethasone is an effective treatment in cases of coronavirus disease 2019 (COVID-19) requiring oxygen, the efficacy of methylprednisolone pulse is unclear. We compared the characteristics and outcomes of methylprednisolone pulse to those of dexamethasone.We conducted a retrospective cohort study on adult COVID-19 cases requiring oxygen and no invasive mechanical ventilation treated with methylprednisolone pulse (1 g/day for 3 days) or dexamethasone (6 mg/day orally or 6.6 mg/day intravenously for ≥5 days). The primary outcome was intensive care unit (ICU) admission. The secondary outcomes were hospital mortality, length of hospital stay (LoS), duration of oxygen requirement, and requirement for hospital transfer, vasopressor(s), intubation, extracorporeal membrane oxygenation (ECMO), and continuous renal replacement therapy (CRRT).Twenty two cases of methylprednisolone pulse and 77 cases of dexamethasone were included. Mask ventilation was more common in the methylprednisolone pulse group (P 0.001). The proportion of ICU admissions was similar between both groups (P = 0.635). The secondary outcomes of hospital mortality and the requirement for hospital transfer, vasopressor(s), intubation, and CRRT were similar between groups. No cases received ECMO. Median LoS (P = 0.006) and duration of oxygen requirement (P = 0.004) were longer in the methylprednisolone pulse group.The proportion of ICU admissions was similar between the methylprednisolone pulse and the dexamethasone group. However, more cases in the methylprednisolone pulse group required mask ventilation than in the dexamethasone group, suggesting that some cases benefited from methylprednisolone pulse.

To evaluate the diagnostic performance of ultrafast and standard dynamic contrast-enhanced (DCE)-MRI in evaluating the residual disease after neoadjuvant chemotherapy (NAC) for breast cancer. Sixty-seven consecutive patients underwent MRI after NAC. Visual analysis of enhancement was performed on ultrafast and standard DCE-MRI, and compared between no residual disease and residual disease groups. The lesion diameters measured on the last phase of ultrafast DCE-MRI and early and delayed phases of standard DCE-MRI were compared with pathological diameter of entire residual cancer and residual invasive ductal carcinoma (IDC). The visual analysis in the delayed phase of standard DCE-MRI exhibited the highest sensitivity (90%), whereas ultrafast DCE-MRI revealed the highest positive predictive value (92%). There were no significant differences between the diameters in the delayed phase of the standard DCE-MRI and the pathological entire residual cancer (p = 0.97), and the diameters in ultrafast DCE-MRI and the pathological residual IDC (p = 0.97). The delayed phase of standard DCE-MRI may be effective for detecting the residual disease and evaluating the extension of entire residual cancer. Enhancement in ultrafast DCE-MRI may be strongly suggestive of the presence of residual disease, and effective for evaluating the extension of residual IDC.

IgG4-related disease (IgG4-RD) is an immune-mediated, multiorgan, chronic inflammatory disease. The three-step classification criteria proposed in 2019 by the American College of Rheumatology and the European League Against Rheumatism (ACR/EULAR) encompass a wide range of clinical, radiological, serological, and histopathological findings. The ACR/EULAR proposed a three-step classification process, i.e., entry step, exclusion step, and scoring system. Radiologists need to know that the radiological findings observed in the five domains of the lacrimal and salivary glands, chest, pancreas and biliary ducts, kidney, and retroperitoneum are independently weighted with different points in the scoring system. A total score 20 points indicates that the patient should not be classified as having IgG4-RD; conversely, a total score ≥ 20 points indicates that the patient should be classified as having IgG4-RD. In this review, the 2019 ACR/EULAR classification criteria are discussed, focusing on the interpretation of each radiological item, with the aim of applying them to the diagnosis of IgG4-RD in clinical practice.

To evaluate the correlation of ADC and IVIM-diffusion kurtosis (DK) model parameters with quantitative histological parameters of whole-slide imaging (WSI).This retrospective study (September 2015-July 2016) included 37 consecutive patients (all females; median age: 64 years, range, 41-82 years), each with a single invasive breast ductal carcinoma lesion with a mass appearance on preoperative MRI. DWI with b-values of 0, 50, 100, 300, 550, 850, and 1000 s/mmThe ADC was significantly correlated with the area ratio of interstitium (r = 0.53, p = 0.00082) and entropy (r = -0.58, p = 0.00019). k was significantly correlated with the area ratio of cancer cell nuclei (r = 0.53, p = 0.00079).Since the ADC reflected the area ratio of interstitium and entropy, and diffusion kurtosis reflected the area ratio of cancer cell nuclei, these parameters may be effective in distinguishing between benign and malignant breast tumours and in grading breast cancer.

Introduction In order to realize carbon neutrality, working toward becoming a hydrogen-based society using renewable energy is essential, and hydrogen fuel cell vehicles will be one of the key devices. For further widespread deployment of fuel cell vehicles, cost reduction and performance enhancement of polymer electrolyte fuel cell (PEFC) systems are urgently needed. Recently, Pt/C-based electrocatalysts using mesoporous carbon as a support material have attracted attention, because well-designed mesopores can act as “accessible pores,” in which direct contact between ionomer and Pt surface is avoided, while the transport of protons, oxygen and generated water is not suppressed. Ordered mesoporous carbon (OMC) with a uniform nanopore structure is one of the ideal candidate materials for a catalyst support with accessible pores. However, with conventional synthesis methods, the typical OMC primary particle size is on the order of micrometers, and mass transport within the nanopores could be a difficult issue. In the present study, we focused on the development of OMC nanoparticles with a network structure. By use of nonionic surfactant micelles with phenol-formaldehyde resole resin as a carbon source, we obtained OMC nanoparticles with a novel hierarchical network structure (ns-OMC). To the best of our knowledge, there are no reports of the synthesis of ordered mesoporous carbon nanoparticles with a rigid network structure. In this study, in addition to the effect of synthesis conditions on the ns-OMC structure, a newly developed technique for the selective deposition of Pt nanoparticles within the ns-OMC nanopores, and the ORR activity of the Pt/ns-OMC catalysts, will be discussed in detail. Experimental Ns-OMC powder was synthesized with resol-nonionic surfactant micelles as a carbon source and structure directing agent. Resol-F127 (nonionic surfactant) micelles were synthesized by mixing phenol, formaldehyde, water and F127 with sodium hydroxide. After hydrothermal treatment of the mixture, the resulting solid was filtered and washed thoroughly and dried under vacuum. The obtained powder was carbonized at 700 ºC followed by annealing at 1000 ºC. Pt was deposited on the ns-OMC powder by the reverse micelle and colloid techniques. Cyclic voltammetry and linear sweep voltammetry were carried out in N2-saturated and O2-saturated 0.1 M HClO4 solution at 25 ºC using the conventional rotating disk electrode (RDE) technique. Potential step cycling measurements simulating load cycling between 0.6 V and 1.0 V vs. RHE were carried out according to the FCCJ protocol. Coating of the catalyst on a glassy carbon disk substrate was carried out by an electrospray (ES) coating technique developed by our group [1]. N2 adsorption measurement at liquid nitrogen temperature was carried out. The morphology of the Pt/ns-OMC was observed by scanning transmission electron microscope (STEM). Results and discussion As seen in the STEM images of the ns-OMC powder (Figure 1), the network structure formed by connection of OMC primary nanoparticles to form developed macropores is clearly observed. The OMC nanoparticles were tightly connected via a thick necking structure. The primary particle size of the OMC and neck thickness were controllable via the resol-F127 micelle concentration and the temperature during the hydrothermal treatment. Highly ordered nanopores were observed on the surface of the OMC particles, with a pore size of ca. 5 nm and an interpore distance of ca. 8 nm. The nanopores and the network structure were stable even after high temperature annealing at 1400 ºC. The BET specific surface area for the ns-OMC annealed at 1000 ºC was 814 m2 g-1, and the BJH pore size distribution curve of the ns-OMC showed a mesopore peak at 4.5 nm, which is in good agreement with the STEM observation. When Pt was deposited on the ns-OMC support, most of the Pt particles, with diameters of 4 to 5 nm, were dispersed on the support and, interestingly, located inside or right above the nanopore entrances. Careful STEM measurements revealed that the Pt particles were not deposited deeply within the OMC nanopores, but most were deposited at relatively shallow positions. RDE measurements revealed that the Pt/ns-OMC catalysts showed higher specific activity and mass activity for the ORR and much higher stability for the ECSA values during potential step cycling measurements in comparison with a commercial Pt/CB catalyst. The higher catalytic activity and durability observed for the Pt/ns-OMC catalyst may be due to a near-ideal distance between neighboring Pt particles deposited on the novel support. Several food constituents augment exercise-induced muscle strength improvement; however, the detailed mechanism underlying these combined effects is unknown because of the lack of a cultured cell model for evaluating the contraction-induced muscle protein synthesis level. Here, we aimed to establish a new in vitro muscle contraction model for analyzing the activation of mammalian target of rapamycin complex 1 (mTORC1) signaling. We adopted the tetanic electric stimulation of 50 V at 100 Hz for 10 min in L6.C11 myotubes. Akt, ERK1/2, and p70S6K phosphorylation increased significantly after electrical pulse stimulation (EPS), compared to untreated cells. Next, we used this model to analyze mTORC1 signaling in combination with exercise and beta-hydroxy-beta-methylbutyrate (HMB), an l-leucine metabolite. p70S6K phosphorylation increased significantly in the EPS+HMB group compared to that in the EPS-alone group. These findings show that our model could be used to analyze mTORC1 signaling and that HMB enhances muscle contraction-activated mTORC1 signaling.

A 66-year-old man was admitted to our hospital because of multiple refractory skin ulcers. Based on his severe systemic arterial calcification and severe calcium-phosphate imbalance due to severe kidney dysfunction, we initially considered calciphylaxis. However, a skin biopsy provided a diagnosis of cholesterol crystal embolization. Although we initiated hemodialysis, steroid treatment, and low-density lipoprotein-cholesterol apheresis, he died of multiple intestinal perforation. An autopsy showed cholesterol crystals occluding multiple organ arterioles. This case suggests that skin ulcers in patients with chronic kidney disease may be an important diagnostic hallmark and may be associated with several serious diseases.

Simple Summary Changes in anti-Müllerian hormone (AMH) levels, a fertility marker in dairy cows, during the peripartum period associated with reproductive recovery have not been fully evaluated. We investigated the relationship between changes in AMH concentration and inflammation markers in serum during the peripartum period in dairy cows. We found a relationship between changes in AMH concentration, especially reflected in the AMH ratio during the perinatal period, and the systemic inflammation status of dairy cows. Excessive inflammation during the early postpartum period may decrease AMH levels and subsequently affect the reproductive prognosis of postpartum cows. Elucidating the mechanism of perinatal AMH changes and the beneficial effects of AMH may improve reproductive efficacy in the dairy industry. Abstract The relationships between changes in anti-Müllerian hormone (AMH) concentration and various traits, including milk somatic cell counts (SCC), were evaluated. Blood samples were collected from 43 Holstein cows 14 days before (D-14) and 10 (D10) and 28 days after (D28) parturition, and vaginal discharge score (VDS) and polymorphonuclear leukocyte (PMNL) percentages were assessed in endometrial samples at D28. Cows were separated into four quartiles (Q1–Q4) based on changes in AMH concentration during the peripartum period (AMH ratio: D28/D-14). Correlations between AMH ratio and each parameter were evaluated and classified into high-AMH (Q4, 1.83 ± 0.12, n = 11) and low-AMH (Q1, 0.83 ± 0.05, n = 11) groups. The AMH ratio was positively correlated with magnesium and non-esterified fatty acids levels, and the albumin/globulin ratio at D10 and D28, but negatively correlated with serum amyloid A (SAA) at D10. SAA and γ-globulin levels were significantly higher in the low-AMH group at D28. There was no significant difference in VDS, PMNL percentage, and milk SCC between the two groups. The decreasing AMH ratio from the prepartum to the postpartum period corresponds to high inflammation biomarker levels. Whether it subsequently affects the reproductive prognosis of postpartum cows needs further investigations.

The collagen gel droplet-embedded drug sensitivity test (CD-DST) was revealed to be useful for predicting the effect of S-1 adjuvant chemotherapy for pancreatic ductal adenocarcinoma (PDAC). However, collection of an adequate number of PDAC cells is difficult due to the surrounding fibroblasts. Thus, the aim of this study was to discover novel biomarkers to predict chemosensitivity based on the CD-DST results. Proteomics analysis was performed using liquid chromatography tandem mass spectrometry (LC–MS/MS). Candidate proteins were validated in patients with 5-FU CD-DST results via immunohistochemistry (IHC). The relationships between the candidate proteins and the effect of the adjuvant S-1 were investigated via IHC. Among the 2696 proteins extracted by LC–MS/MS, C1TC and SAHH could accurately predict the CD-DST results. Recurrence-free survival (RFS) was significantly improved in the IHC-positive group compared with the IHC-negative group in both factors. The negative group did not show a significant difference from the group that did not receive S-1. The double-positive group was associated with significantly prolonged RFS compared to the no adjuvant chemotherapy group. C1TC and SAHH have been shown to be useful biomarkers for predicting 5-FU sensitivity as a substitute for the CD-DST in adjuvant chemotherapy for PDAC.

We applied combination antibiotic therapy to treat vertebral osteomyelitis and a psoas abscess caused by glycopeptide-intermediate (MIC, 2 μg/ml) and daptomycin-nonsusceptible (>2 μg/ml) methicillin-resistant Staphylococcus aureus . The Etest synergy test showed the largest synergistic effects for imipenem/cilastatin and fosfomycin. Whole-gene sequencing showed amino acid changes in SA0802, SA1193 ( mprF ), and SA1531 ( ald ).

Introduction Although the seasonality of infectious diseases has been widely reported, the seasonality of peripheral venous catheter-related bloodstream infection (PV-CRBSI) has not been investigated. This study investigated the seasonality of PV-CRBSI and its relationship with meteorological conditions. Methods A retrospective cohort study of PV-CRBSI at Tokyo Medical University Hospital (Tokyo, Japan), from 2009 to 2019, provided the data for descriptive and time series analyses used to evaluate the number of PV-CRBSI cases per 1000 admissions that occurred each month for each causative organism. By performing univariate and multivariate analyses, the researchers investigated the seasonality of cases and the relationships between meteorological conditions, other external factors, and PV-CRBSIs. Results This study included a total of 184 PV-CRBSI cases. The mean numbers of PV-CRBSI cases per 1000 admissions caused by all organisms, Bacillus cereus, Gram-positive cocci, and Gram-negative rods were 0.67, 0.15, 0.37, and 0.16 per month, respectively, during the study period. The time series analysis showed that the incidences of PV-CRBSI cases associated with B. cereus and Gram-negative rods were significantly different in the winter/spring from those in the summer/autumn (P

Oral bacteria may contribute to postoperative infectious complications including postoperative pneumonia or surgical site infection. The aim of this study was to investigate the impact of preoperative dental care on postoperative outcomes among surgical patients under general anesthesia.Retrospective cohort study.We analyzed clinical records of major surgical patients at a university hospital between 2016 and 2018. Subjects were categorized into either the preoperative dental care group, those being referred to dentists by their surgeons based on an individual surgeon's judgment for dental care before surgery, or the control group.The primary outcome was postoperative infectious complications. Secondary outcomes were postoperative inflammation markers (C-reactive protein and fever), and economic outcomes (postoperative length of hospital stay and medical expenses). As the main analysis, the average treatment effects of the preoperative dental care were obtained from the augmented inverse-probability weighting (AIPW) method with consideration of demographics and perioperative risk factors to estimate causal effect of the intervention from the observational data. Then, stratified analyses by age and surgical sites were conducted with the inverse-probability weighting and linear regression methods, respectively.In the AIPW estimation, compared with the control group, the care group saw a significantly lower rate of postoperative infection (average treatment effect -3.02) and shorter fever duration (-2.79 days). The stratified analysis by age revealed significant positive impact of dental care in all age groups, including the highest treatment effects observed among patients younger than 60. Also, treatment effect was observed in wider surgical sites than previously known.This study indicates a significant impact of preoperative dental care on preventing postoperative infection and inflammation. Along with old age or certain types of surgeries in which advantages of dental referral have been already known, preoperative dental referral could be beneficial for broader types of patients.

To investigate effective model composed of features from ultrafast dynamic contrast-enhanced magnetic resonance imaging (UF-MRI) for distinguishing low- from non-low-grade ductal carcinoma in situ (DCIS) lesions or DCIS lesions upgraded to invasive carcinoma (upgrade DCIS lesions) among lesions diagnosed as DCIS on pre-operative biopsy. Eighty-six consecutive women with 86 DCIS lesions diagnosed by biopsy underwent UF-MRI including pre- and 18 post-contrast ultrafast scans (temporal resolution of 3 s/phase). The last phase of UF-MRI was used to perform 3D segmentation. The time point at 6 s after the aorta started to enhance was used to obtain subtracted images. From the 3D segmentation and subtracted images, enhancement, shape, and texture features were calculated and compared between low- and non-low-grade or upgrade DCIS lesions using univariate analysis. Feature selection by least absolute shrinkage and selection operator (LASSO) algorithm and k-fold cross-validation were performed to evaluate the diagnostic performance. Surgical specimens revealed 16 low-grade DCIS lesions, 37 non-low-grade lesions and 33 upgrade DCIS lesions. In univariate analysis, five shape and seven texture features were significantly different between low- and non-low-grade lesions or upgrade DCIS lesions, whereas enhancement features were not. The six features including surface/volume ratio, irregularity, diff variance, uniformity, sum average, and variance were selected using LASSO algorism and the mean area under the receiver operating characteristic curve for training and validation folds were 0.88 and 0.88, respectively. The model with shape and texture features of UF-MRI could effectively distinguish low- from non-low-grade or upgrade DCIS lesions.

Lingual osseous choristoma is a rare benign tumor consisting of normal matured bone tissue. It was first reported in 1913, and less than 100 cases of lingual osseous choristomas, mainly in their twenties and thirties, have been reported in the English literature until now. Here, we report an additional case of lingual osseous choristoma, in an elderly patient, that was incidentally removed by coughing and cured without additional interventions. An 89-year-old male patient was referred to our department for an evaluation of chronic cough. When we examined his oral cavity and pharynx, he expectorated a 10 -mm mass which was histologically diagnosed as an osseous choristoma. We confirmed the well-defined, rounded, high-density mass with a tiny pedicle on the base of the tongue in previous cervical spine CT images. No signs of recurrence were found during the 15-month follow-up examination. Our case serves as a reminder of this rare entity in the diagnosis of tongue masses of the elderly.

RATIONALE AND OBJECTIVES: To evaluate whether parameters from empirical mathematical model (EMM) for ultrafast dynamic contrast-enhanced (DCE) magnetic resonance imaging (MRI) correlate with histological microvessel density (MVD) in invasive breast cancer. MATERIALS AND METHODS: Ninety-eight consecutive patients with invasive breast cancer underwent an institutional review board-approved ultrafast DCE-MRI including a pre- and 18 postcontrast whole breast ultrafast scans (3 seconds) followed by four standard scans (60 seconds) using a 3T system. Region of interest was placed within each lesion where the highest signal increase was observed on ultrafast DCE-MRI, and the increase rate of enhancement was calculated as follows: ΔS = (Slpost – SIpre)/SIpre. The kinetic curve obtained from ultrafast DCE-MRI was analyzed using a truncated EMM: ΔS(t)=A(1 – e(−αt)), where A is the upper limit of the signal intensity, α (min(−1)) is the rate of signal increase. The initial slope of the kinetic curve is given by Aα. Initial area under curve (AUC30) and time of initial enhancement was calculated. From the standard DCE-MRI, the initial enhancement rate (IER) and the signal enhancement ratio (SER) were calculated as follows: IER = (SIearly – SIpre)/SIpre, SER = (SIearly – SIpre)/(SIdelayed – SIpre). The parameters were compared to MVD obtained from surgical specimens. RESULTS: A, α, Aα, AUC30, and time of initial enhancement significantly correlated with MVD (r =0.29, 0.40, 0.51, 0.43, and −0.32 with p = 0.0027, p < 0.0001, p < 0.0001, p < 0.0001, and p = 0.0012, respectively), whereas IER and SER from standard DCE-MRI did not. CONCLUSION: The parameters of the EMM, especially the initial slope or Aα, for ultrafast DCE-MRI correlated with MVD in invasive breast cancer.

We report a case of a 72-year-old woman who was initially diagnosed with ovarian cancer with peritoneal carcinomatosis. Systemic chemotherapy consisting of paclitaxel and carboplatin(TC)was administered. Although a partial response(PR)was achieved after the 4 courses of TC, this regimen was discontinued due to severe adverse events. Ten months after discontinuation of TC, because abdominal CT and colonoscopy showed an intra-tumoral abscess caused by invasion of the tumor to the sigmoid colon, abdominal total hysterectomy, bilateral salpingo-oophorectomy, and a Hartmann's operation were performed to control the disease symptoms. Pathological examination revealed that the tumor was an undifferentiated carcinoma of the sigmoid colon. This case report suggests that the TC regimen may be effective for treating undifferentiated carcinoma of the colon.

We developed a new desiccator system by improving the conventional desiccator method and measured the FA-removal effects of adsorbents using this system. The improved desiccator system consists of: 1) a pump, 2) a flow meter, 3) a FA gas generator, 4) five connected desiccators, and 5) an exhaust. A more stable concentration of FA in the desiccators was obtained when they were connected in series rather than in parallel. The FA removal rate for 3 days for red tea in used tea bags, green tea, oolong tea, or powdered coffee was more than 80%, and the removal rates for 1 day by activated carbon from palm nutshells and silica gel, as commercially available chemical agents, were 100% and 75%, respectively. This improved desiccator system allows direct collection of air in the desiccator, and may have wide applications.

Mini rats, a Wistar-derived transgenic rat strain harboring a rat growth hormone (GH) antisense gene, show lower plasma GH levels than Wistar rats. In our previous study, Mini rats showed a smaller femur size with lower mineral density and a reduction of the metaphyseal and diaphyseal bone mass at 8 and 22 weeks of age. In the present study, we examined the femur and tibia from Mini rats and Wistar rats at 10 to 60 weeks of age, and we performed cancellous and cortical bone histomorphometry on the tibia to examine the status of bone turnover. We found that the bone size, mineral content, and mineral density of the femur and tibia were significantly lower in Mini rats and tended to increase in both strains until 38 or 60 weeks of age. The longitudinal growth rate was significantly higher in Wistar rats until 26 weeks of age, and at 38 weeks of age that in Wistar rats is comparable to that in Mini rats. The periosteal bone formation rate tended to be higher in Wistar rats until 60 weeks of age. The longitudinal growth rate in both strains and the periosteal bone formation rate in Wistar rats tended to decrease with age. In contrast, bone turnover in the secondary spongiosa of the proximal tibial metaphysis showed no apparent difference between the two strains and seemed to remain unchanged until 60 weeks of age. Therefore, it is suggested that the reductions of bone mass in the metaphysis and diaphysis in Mini rats may be attributable to the lower longitudinal growth rate and periosteal bone formation rate, and that GH may influence bone modeling but not bone remodeling.

Growth hormone (GH) plays an important role in longitudinal bone growth, and hypophysectomized rats have generally been used as a model for GH deficiency in humans. Recently, researchers have bred Mini rats, a Wistar-derived transgenic rat strain harboring a rat GH antisense gene and showing lower plasma GH levels than Wistar rats. In a previous study, Mini rats showed a smaller femur size with lower mineral density and a reduction of the metaphyseal and diaphyseal bone mass. In the present study, Wistar rats were hypophysectomized (HX) and treated with GH, and then their bones were examined and compared with vehicle-treated intact age-matched Wistar rats and Mini rats. GH-treated (HX+GH) rats exhibited an increase in the mineral density of the tibia and increases in the length and mineral content of the femur and tibia. These changes in the HX+GH rats were attributable to an increase in the longitudinal growth and periosteal bone formation of the femur and tibia. When Mini rats and HX rats were compared with Wistar rats, both animals showed similar reductions in the bone length, mineral content, and mineral density of the femur, but the Mini rats showed a greater longitudinal growth rate, cancellous bone mass, and mineralizing surface than the HX rats, as well as less remarkable changes in body weight, relative organ weights, and hematological and blood chemical parameters. Therefore, Mini rats are considered to be a useful model for clarifying features of GH deficiency and examining effects of various treatments on the bone without any specific surgery or drug-administration.

Recently, Mini rats, a Wistar-derived transgenic rat strain harboring a rat GH antisense gene and showing 40% lower plasma GH levels than Wistar rats, have been developed. In a previous study, Mini rats showed a smaller femur size with lower mineral density and a reduction of the metaphyseal and diaphyseal bone mass. In the present study, neonates of Wistar rats were treated subcutaneously with MSG to obtain GH-deficient rats (MSG rats), and their bones were examined and compared with age-matched MSG-untreated Wistar rats and Mini rats. Compared with the Wistar rats, body weights of the MSG rats were comparable, whereas those of the Mini rats were significantly lower. Bone size, bone mineral content and mineral density were significantly lower in the MSG rats and Mini rats than those in the Wistar rats. Histologically, the amounts of the metaphyseal cancellous bone mass and diaphyseal cortical bone mass were less in the MSG rats and much less in the Mini rats. Compared with the Wistar rats, the growth plate width and longitudinal growth rate were similarly lower. However, there were no differences in bone surface-referent parameters in the secondary spongiosa for histomorphometry among the MSG rats, Wistar rats, and Mini rats, indicating that GH may not influence bone remodeling. Thus, Mini rats are considered to be a useful model for clarifying features of GH-deficiency and examining the effects of various treatments on the bone without any specific surgery or drug administration.

Mini rats have been reported to be a Wistar-derived strain of transgenic rats that harbors a rat growth hormone (GH) antisense gene and shows lower plasma GH levels than Wistar rats. Our previous study has revealed that: 1) Mini rats show smaller hind limbs with lower metaphyseal and diaphyseal bone mass than Wistar rats; 2) that the differences of the diaphyseal cortical bone mass and metaphyseal bone mass between Mini rats and Wistar rats are due largely to the differences in the periosteal bone formation and longitudinal growth rate, respectively, and ; 3) that the bone turnover in the secondary spongiosa, an area were bone remodeling may occur, does not differ between the two strains. In the present study, 8-week-old male Mini rats were treated intraperitoneally with GH at doses of 0, 2, 6 or 20 IU/kg/time twice daily for 14 days. After the dosage period, we measured the size, bone mineral content, and mineral density of the femur and tibia in each group, and we performed cancellous and cortical bone histomorphometry on the tibia to examine the effects of GH on the bone modeling and remodeling. In the GH-treated groups there were increases in the metaphyseal cancellous bone mass, growth plate width, longitudinal growth rate, and periosteal bone formation accompanied by increases in bone size, mineral content, and mineral density in a dose-related fashion. However, bone histomorphometric parameters related to bone turnover in the secondary spongiosa of the metaphysis, where lamellar bone formation took place, did not differ between the control and GH-treated groups. These findings indicated that GH may not influence bone remodeling in the latter groups, and that GH replacement may not influence the number of osteoclasts involved in bone resorption in Mini rats as a whole.

Histopathological examination of this tumour revealed a subcutaneous cystic legion formed by ciliated columnar epithelium with smooth muscle bundles and squamous epithelium with sebaceous glands.
